Abstract

Antigen of CTLV could be detected from diseased citrus leaves by ELISA using horseradish peroxidase-conjugated IgG (HRP-ELISA). In the HRP-ELISA procedure, sample and conjugate were mixed and incubated in the plate for 16hr at 4C. The relative sensitivity of antigen detection was 1:16, when the sample of the infected citrus leaf extract was diluted with healthy leaf extract. Citrus scions from post-entry quarantine and scions taken from citrus groves in Japan were tested by modified HRP-ELISA and by an inoculation test to Chenopodium quinoa. The results showed that ELISA was reliable method for detecting CTLV in citrus trees.
