Exposure of polychlorinated biphenyls via indoor dust particles and their health risks in Dhaka City, Bangladesh

This study is a pioneer evaluation of Polychlorinated Biphenyls (PCBs) within the indoor environment of the megacity Dhaka, Bangladesh. It assessed the occurrence, distribution and associated human health risks of selected 6 PCB congeners in four types of buildings (commercial, industrial, institutional, and residential). Dust samples were collected from the air conditioner filters and subsequently analyzed by Gas Chromatography- Electron Capture Detector (GC-ECD). ∑6 PCB concentrations in commercial, industrial, institutional, and residential buildings ranged from 168 ± 59.42 to 3419 ± 1307, 590 ± 217 to 4669 ± 1731, 244 ± 89.08 to 2538 ± 779, and 161 ± 60.54 to 5413 ± 2136 ng/g, respectively. Considering the total PCB loads from each building type, the industrial category revealed the maximum concentration (19176 ± 1141 ng/g), followed by commercial, residential, and institutional, meaning industrial buildings are more susceptible to PCBs exposure. The heavy PCBs (hexa- and hepta-CBs) were dominant over the light PCBs (di-, tri-, and tetra-CBs). All hazard index (HI) values, except PCB-180, were below 1, suggesting a lower risk of non-carcinogenic effect. The incremental lifetime cancer risk (CR) values were below 10−03, indicating no high carcinogenic risk of the human population from PCB-contaminated dust through the exposure routes.

Occurrence of Polycyclic Aromatic Hydrocarbons and Polychlorinated Biphenyls in Fogwater at Urban, Suburban, and Rural Sites in Northeast France between 2015 and 2021

Polycyclic aromatic hydrocarbons (PAHs) and polychlorinated biphenyls (PCBs) exist in the atmosphere in the vapor and particulate phases, as well as in solubilized form in fog/rain/cloud waters. In the current paper, fogwater samples are collected during 42 events between 2015 and 2021 at four different sites (Strasbourg, Geispolsheim, Erstein, Cronenbourg) in the Alsace region. Organics are extracted using liquid–liquid extraction (LLE) supported on a solid cartridge (XTR Chromabond), and then analyzed by gas chromatography–tandem mass spectrometry (GC-MS/MS). The total PAHs and PCBs concentrations in fog samples vary between 0.58 and 6.7 µg L−1 (average of 2.70 µg L−1), and 0.14 and 15.5 µg L−1 (average of 2.75 µg L−1). Low-molecular-weight (LMW) PAHs are predominant and highly detectable compared to high-molecular-weight (HMW) PAHs, while pentachloro-biphenyls are the dominant PCB congener. The PAHs and PCBs concentrations have increased over the sampling years at all sites, except for a slight decrease in PCBs level at Geispolsheim. A diagnostic ratio (DR) and principal component analysis (PCA) are applied to suggest potential contamination sources at Strasbourg metropolitan. Their results reveal that PAHs derive from a mixture of pyrogenic activities, while PCBs mainly come from industrial activities. The results also reveal, in some cases, inter-site variability for simultaneous and successive fog events.

The association of exposure to polychlorinated biphenyls with lipid profile and liver enzymes in umbilical cord blood samples

Evidence on prenatal exposure to polychlorinated biphenyls (PCBs) and its effects on newborns and potential biological mechanisms is not well defined yet. Therefore, this study aimed to examine whether PCBs are associated with lipid profile and non-invasive markers of hepatocyte injuries in samples of blood obtained from the umbilical cord. This study included 450 mothers-newborn pairs. Umbilical levels of PCBs were measured using Gas Chromatography/Mass Spectrophotometry (GC/MS). Lipid profile including low-density lipoprotein (LDL-C), total cholesterol (TC), triglycerides (TG), and high-density lipoprotein (HDL-C), as well as liver enzymes i.e., alanine amino transferase (ALT), aspartate amino transferase (AST), γ-glutamyl-transferase (GGT) and alkaline phosphatase (ALP) were determined from umbilical cord blood samples. Quantile g-computation analysis was applied to evaluate the collective influence of PCBs on both lipid profiles and liver enzymes, along with the impact of lipid profiles on liver enzymes. Exposure to the mixture of PCBs was significantly associated with increases in ALP, AST, ALT, and GGT levels in cord blood samples, with increments of 90.38 U/L (95%CI: 65.08, 115.70, p < 0.01), 11.88 U/L (95%CI: 9.03, 14.74, p < 0.01), 2.19 U/L (95%CI:1.43, 2.94, p < 0.01), and 50.67 U/L (95%CI: 36.32, 65.03, p < 0.01), respectively. Additionally, combined PCBs exposure was correlated with significant increases in umbilical TG, TC, and LDL-C levels, with values of 3.97 mg/dL (95%CI: 0.86, 7.09, p = 0.01), 6.30 mg/dL (95%CI: 2.98, 9.61, p < 0.01), and 4.63 mg/dL (95%CI: 2.04, 7.23, p < 0.01) respectively. Exposure to the mixture of lipids was linked to elevated levels of AST and GGT in umbilical cord blood samples. Furthermore, a noteworthy mediating role of TC and LDL-C was observed in the association between total PCBs exposure and umbilical cord blood liver enzyme levels. Overall our findings suggested that higher levels of umbilical cord blood PCBs and lipid profile could affect liver function in newborns.

Screening and optimization of interpolation methods for mapping soil-borne polychlorinated biphenyls

There is yet no scientific consensus, and for now, on how to choose the optimal interpolation method and its parameters for mapping soil-borne organic pollutants. Take the polychlorinated biphenyls (PCBs) for instance, we present the comparison of some classic interpolation methods using a high-resolution soil monitoring database. The results showed that empirical Bayesian kriging (EBK) has the highest accuracy for predicting the total PCB concentration, while root mean squared error (RMSE) in inverse distance weighting (IDW) is among the highest in these interpolation methods. The logarithmic transformation of non-normally distributed data contributed to enhance considerably the semivariogram for modeling in kriging interpolation. The increasing of search neighborhood reduced IDW's RMSE, but slightly affected in ordinary kriging (OK), while both of them resulted in over smooth of prediction map. The existence of outliers made the difference between two points increase sharply, and thereby weakening spatial autocorrelation and decreasing the accuracy. As predicted error increased continuously, the prediction accuracy of different interpolation methods reached unanimity gradually. The attempt of the assisted interpolation algorithm did not significantly improve the prediction accuracy of the IDW method. This study constructed a standardized workflow for interpolation, which could reduce human error to reach higher interpolation accuracy for mapping soil-borne PCBs.

Polychlorinated Biphenyls and Semen Quality in Healthy Young Men Living in a Contaminated Area.

Polychlorinated biphenyls (PCBs) are persistent organic pollutants and endocrine disruptors that have been implicated in potential damage to human semen. However, the studies conducted so far provide contrasting results. Our study aimed to investigate the associations between PCB serum and semen levels and semen quality in high school and university students living in a highly PCB-polluted area of Italy. Subjects with a normal body mass index who did not make daily use of tobacco, alcohol, drugs, or medication were selected. All participants provided a fasting blood and a semen sample. Gas chromatography-mass spectrometry was used to determine the concentrations of 26 PCB congeners. The concentrations of PCB functional groups and total PCBs were also computed. A total of 143 subjects (median age 20, range 18-22 years) were enrolled. The median total PCB concentrations were 3.85 ng/mL (range 3.43-4.56 ng/mL) and 0.29 ng/mL (range 0.26-0.32 ng/mL) in serum and semen, respectively. The analysis of the associations between sperm PCB concentration and semen parameters showed (a) negative associations between some PCB congeners, functional groups and total PCBs and sperm total motility; (b) negative associations of total PCBs with sperm normal morphology; and (c) no association of PCBs with sperm concentration. Subjects at the highest quartile of semen total PCB concentration had 19% and 23% mean reductions in total motility and normal morphology, respectively, compared to those at the lowest quartile. The analysis of the associations of serum PCB levels with sperm parameters yielded null or mixed (some positive, other negative) results. In conclusion, the present study provides evidence of a negative effect of some PCB congeners and total PCBs in semen on sperm motility and normal morphology. However, the associations between the concentration of serum and semen PCB congeners and functional groups and sperm quality parameters were inconsistent.

One Health assessment of persistent organic chemicals and PFAS for consumption of restored anadromous fish

Abstract Background Restoration efforts have led to the return of anadromous fish, potential source of food for the Penobscot Indian Nation, to the previously dammed Penobscot River, Maine. Objective U.S. Environmental Protection Agency (EPA), Penobscot Indian Nation’s Department of Natural Resources (PINDNR), and Agency for Toxic Substances and Disease Registry (ATSDR), measured contaminants in six species of anadromous fish. Fish tissue concentrations were then used, along with exposure parameters, to evaluate potential human and aquatic-dependent wildlife risk. Methods PINDNR collected, filleted, froze, and shipped fish for analysis of polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs), dioxins/furans, and per- and polyfluoroalkyl substances (PFAS). Contaminant levels were compared to reference doses (where possible) and wildlife values (WVs). Results Chemical concentrations ranged from 6.37 nanogram per gram (ng/g) wet weight (ww) in American Shad roe to 100 ng/g ww in Striped Bass for total PCBs; 0.851 ng/g ww in American Shad roe to 5.92 ng/g ww in large Rainbow Smelt for total PBDEs; and 0.037 ng/g ww in American Shad roe to 0.221 ng/g ww in Striped Bass for total dioxin/furans. PFAS concentrations ranged between 0.38 ng/g ww of PFBA in Alewife to 7.86 ng/g ww of PFUnA in Sea Lamprey. Dioxin/furans and PFOS levels indicated that there are potential human health risks. The WV for mink for total PCBs (72 ng/g) was exceeded in Striped Bass and the WV for Kestrel for PBDEs (8.7 ng/g) was exceeded in large Rainbow Smelt. Mammalian wildlife consuming Blueback Herring, Striped Bass, and Sea Lamprey may be at risk based on PFOS WVs from Canada. Impact Anadromous fish returning to the Penobscot River potentially could represent the restoration of a major component of tribal traditional diet. However, information about contaminant levels in these fish is needed to guide the tribe about consumption safety. Analysis of select species of fish and risk calculations demonstrated the need for a protective approach to consumption for both humans and wildlife. This project demonstrates that wildlife can also be impacted by contamination of fish and their risks can be as great or greater than those of humans. A One Health approach addresses this discrepancy and will lead to a healthier ecosystem.

Concentrations and risks of PCBs in soil contaminated with transformer oil in selected locations in Warri South Local Government Area, Delta State, Nigeria

According to the list of persistent organic pollutants (POP), polychlorinated biphenyls (PCBs) are a class of organic substances whose usage has been outlawed in the majority of nations. In addition to been used in paint, flame retardants, plasticizers, and lubricants, they were principally employed in transformers, capacitors, and other industrial purpose. PCBs have been found everywhere over the world because they are capable of being released into the environment through primary and secondary sources, travel over great distances in the atmosphere and are thus persistent. PCBs were examined in soil from area around transformer installations at several sites in Warri South Local Government Area (LGA), Delta State, Nigeria. Using Agilent 6975 GC-MS at three different locations, the evaluation was conducted for both total PCBs and congeners. The soil samples were cleaned with column chromatography using silica gel and hexane as the eluting solvent and extracted with a 1:1 hexane-acetone solution in an ultrasonic bath. With the following results recorded - Ubeji (0.148 ± 0.019 mg/kg), Ogunu (0.531 ± 0.063 mg/kg) and Edjeba (0.415 ± 0.34 mg/kg), the assessment demonstrate that all locations were polluted with PCBs that were below the maximum limit of 2.0 mg/kg as allowed by the United States Toxic Substance Control Act (USTSCA). The total cancer risk calculated in these chosen locations corresponds to Ubeji (5.41×10−7), Ogunu (1.95×10−6) and Edjeba (1.52×10−6). This result showed that all the locations assessed have very low to low cancer risks compared with the value advised by USTSCA.

Polyurethane Foam Emission Samplers to Identify Sources of Airborne Polychlorinated Biphenyls from Glass-Block Windows and Other Room Surfaces in a Vermont School.

We hypothesized that emissions of polychlorinated biphenyls (PCBs) from Aroclor mixtures present in building materials explain their concentrations in school air. Here, we report a study of airborne concentrations and gas-phase emissions in three elementary school rooms constructed in 1958. We collected airborne PCBs using polyurethane foam passive air samplers (PUF-PAS, n = 6) and PCB emissions from building materials using polyurethane foam passive emission samplers (PUF-PES, n = 17) placed over flat surfaces in school rooms, including vinyl tile floors, carpets, painted bricks, painted drywall, and glass-block windows. We analyzed all 209 congeners represented in 173 chromatographic separations and found that the congener distribution in PUF-PES strongly resembled the predicted diffusive release of gas-phase PCBs from a solid material containing Aroclor 1254. Concentrations of airborne total PCBs ranged from 38 to 180 ng m-3, a range confirmed by an independent laboratory in the same school. These levels exceed action levels for all aged children set by the State of Vermont and exceed guidance levels set by the U.S. EPA for children under age 3. Emissions of PCBs from the glass-block windows (30,000 ng m-2 d-1) greatly exceeded those of all other surfaces, which ranged from 35 to 2700 ng m-2 d-1. This study illustrates the benefit of the direct measurement of PCB emissions to identify the most important building remediation needed to reduce airborne PCB concentrations in schools.

Short-chain fatty acids facilitated long-term dechlorination of PCBs in Taihu Lake sediment microcosms: Evidence from PCB congener and microbial community analyses

Microbial reductive dechlorination hosts great promise as an in situ bioremediation strategy for polychlorinated biphenyls (PCBs) contamination. However, the slow dechlorination in sediments limits natural attenuation. Short-chain fatty acids, as preferred carbon sources and electron donors for dechlorinating microorganisms, might stimulate PCB dechlorination. Herein, two sets of short-chain fatty acids, sole acetate and a fatty acid mixture (acetate, propionate, and butyrate), were amended periodically into Taihu Lake (China) sediment microcosms containing nine PCB congeners (PCB5, 12, 64, 71, 105, 114, 149, 153, and 170) after 24 weeks of incubation. Short-chain fatty acids facilitated the long-term PCB dechlorination and the promoting effect of the fatty acid mixture compared favorably with that of sole acetate. By the end of 108 weeks, the total PCB mass concentrations in acetate amended and fatty acid mixture amended microcosms significantly declined by 7.6% and 10.3% compared with non-amended microcosms (P < 0.05), respectively. Short-chain fatty acids selectively favored the removal of flanked meta and single-flanked para chlorines. Notably, a rare ortho dechlorination pathway, PCB25 (24-3-CB) to PCB13 (3-4-CB), was enhanced. Supplementary fatty acids significantly increased reductive dehalogenases (RDase) gene pcbA5 instead of improving the growth of Dehalococcoides. These findings highlight the merits of low cost short-chain fatty acids on in situ biostimulation in treating PCBs contamination.

Effects of mixtures of polychlorinated biphenyls (PCBs) and three organochlorine pesticides on cognitive function differ between older Mohawks at Akwesasne and older adults in NHANES

BackgroundAkwesasne Mohawks has been exposed to high concentrations of polychlorinated biphenyls (PCBs) and background levels of organochlorine pesticides, hexachlorobenzene (HCB), dichlorodiphenyl dichloroethylene (DDE), and mirex. We have previously reported relative contributions to the mixture of low- and high-chlorinated PCBs, HCB, and DDE on cognitive decrements in Mohawks of various ages. ObjectiveThis study examines differences in the mixture effects of PCB congener groups, HCB, DDE, and mirex on cognitive function in older Mohawks and less PCB-exposed older adults from the National Health and Nutrition Examination Survey (NHANES) 1999–2002 cycles. MethodsWe used Bayesian kernel machine regression (BKMR) to evaluate the mixture effects of different PCB congener groups, HCB, DDE, and mirex on cognitive function in both populations. Models were adjusted for age, sex, education levels, and race/ethnicity focusing on individuals 60 years and older. ResultsOlder Mohawks had 3-fold higher mean total PCB concentrations and 1.8-fold higher mirex, but slightly lower mean DDE and HCB levels than NHANES older adults. Higher mixture concentrations were significantly associated with greater cognitive decline. In older Mohawks, low- and high-chlorinated PCBs, HCB, and DDE contributed to the cognitive score decline. In contrast, score decline in older NHANES adults were primarily from high-chlorinated PCBs and DDE with a threshold dose of approximately 2.08–2.27 ng/g and 2.02–2.40 ng/g, respectively. ConclusionMixtures of PCBs and organochlorine pesticides increase the risk of cognitive decline in both older Mohawks and NHANES older adults. However, contributions to these mixture effects show significant differences. In older Mohawks, high- and low-chlorinated PCBs, DDE, and HCB are the primary contributors, while high-chlorinated PCBs and DDE are important contributors in NHANES older adults. Due to chronic heavy exposures to PCBs, older Mohawks had a significantly increased risk of cognitive decline compared to general older adults from NHANES.

Serum levels of polychlorinated biphenyls and polybrominated diphenyl ethers in early pregnancy and their associations with gestational diabetes mellitus

Polychlorinated Biphenyls (PCBs) and Polybrominated Diphenyl Ethers (PBDEs) are extensively present in humans and may disturb glucose metabolism during pregnancy. However, previous reports on the associations between PCBs/PBDEs levels and gestational diabetes mellitus (GDM) have been inconsistent. We performed a nested case-control study to measure the serum levels of 6 PCB and 7 PBDE congeners in early pregnancy, and to assess their associations with GDM risk and blood glucose levels. Totally, 208 serum samples (104 GDM cases and 104 controls) were included based on a prospective cohort which was carried out in Jiangsu province, China, from 2020 to 2022. The results showed that PCB-153 was the major PCB congener, whereas PBDE-47 was the predominant PBDE congener. The continuous concentrations of PCB-153, PBDE-28, and total PCB were significantly related to an increased risk of GDM, with adjusted ORs (95%CI) of 1.25 (1.04–1.50), 1.19 (1.02–1.39), and 1.37 (1.05–1.79), respectively. Potential dose-response relationships were also observed between serum levels of PCB-153 (P = 0.011), PBDE-28 (P = 0.028), total PCB (P = 0.048), and total PCB/PBDE (P = 0.010) and GDM risk. Moreover, PCB-153, PBDE-28 and total PCB levels were positively related to 1-h OGTT blood glucose (adjusted βPCB-153: 0.14, 95%CI: 0.00–0.28; adjusted βPBDE-28: 0.20, 95%CI: 0.08–0.32; adjusted βtotal PCB: 0.30, 95%CI: 0.09–0.50), whereas none of the PCBs/PBDEs were statistically related to fasting blood glucose and 2-h OGTT blood glucose (all P > 0.05). Further meta-analysis also supported the association of PCBs exposure with GDM risk. Our study provides further evidence that PCBs/PBDEs exposure may increase GDM risk during pregnancy.

Organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs) in adipose tissue of women from Grand Tunis and their association with demographic factors and dietary habits

Dichlorodiphenyl trichloroethane and its metabolites (DDTs), hexachlorocyclohexane isomers (HCHs), hexachlorobenzene (HCB) and polychloronated biphenyls (PCBs) were measured in 25 woman adipose tissues collected in 2016 from Grand Tunis, Tunisia. p,p’-DDE, p,p’-DDT, HCB and β-HCH were the dominant organochlorine pesticides (OCPs) in decreasing order in all samples. The total OCP levels varied from 79 to 343 ng g−1 lipid with a median value of 189 ng g−1 lipid and DDTs contributed approximately 88% to sum OCP. The ratio of p,p’-DDT/p,p’-DDE across all samples is below one, which suggests mainly historic exposure but may indicate some recent exposure to the banned pesticide. The median concentration of PCBs was 109 ng g−1 lipid and ranged between 27 and 204 ng g−1 lipid. PCB-153, PCB-180, PCB-138 and PCB-170 were the most abundant congeners, which contributed about 78% of the total PCBs. Spearman analysis showed that dominant organochlorine compounds (OCs) are highly positive correlated except for PCB-28/31, indicating that women from Tunis are exposed via similar routes. Inhalation exposure could be a possible pathway for the uptake of the less chlorinated congeners. We found positive and statistically significant association with subjects age for HCB (r = 0.517; p = 0.009) and PCBs (r = 0.65; p = 0.001) levels and a weak age-dependent accumulation was found for HCHs (r = 0.375; p = 0.065) and DDTs (r = 0.388; p = 0.056). The concentrations of OC subgroups were not associated with BMI, parity and residence. No association was observed between fish, red/white meat, milk and dairy products consumption and levels of HCB, HCHs and PCBs. DDTs levels were significantly correlated only with milk (p = 0.048) and milk products (p = 0.047) intake.

Caged mussels and passive samplers used to identify Niagara River organic contaminant sources and shoreline water quality impacts (2012–2018)

Since 1983 the Ontario Ministry of Environment (MECP) monitored and identified Niagara River contaminant sources using caged mussels, and recently (2012–2018), passive samplers. Source identification was consistent among monitoring tools. The advantage of passive samplers was the capability to estimate dissolved water contaminants concentrations enabling comparisons with Water Quality Criteria (WQC) and ambient river concentrations. Dieldrin, p, p’-DDE, p, p’-DDD, hexachlorobenzene, and total PCBs exceeded WQC throughout the river. The highest concentrations of mirex (1.4 ng/L), octachlorostyrene (2.8 ng/L), and hexachlorobutadiene (16 ng/L) were at Occidental Chemical Co. Buffalo Ave. sewer 003, and the highest concentrations of α-HCH were at Cayuga Creek (36 ng/L) and Gill Creek (68 ng/L): all were >WQC. The Pettit Flume cove, Little Niagara River downstream of the 102nd St. Waste Site, Bloody Run Creek (associated with the Hyde Park Hazardous Waste site) and Occidental sewer 003 had the highest concentrations of chlorinated benzenes. Sources of dioxins to the Niagara River included; Pettit Flume cove: (water: 3.9–10 pg TEQ/L; sediment: up to 989,400 pg/g), and Bloody Run Creek: (water: 1.2 pg TEQ/L; sediment: 35,098 pg/g–101,113 pg/g). Total PCBs remain high at known and newly identified sources along the Tonawanda Channel (USA): concentrations ranged up to 387 ng/L with most sites between 5 and 19 ng/L compared with the Chippawa Channel (Canada) (<1 ng/L). Generally, contaminant concentrations on the Canadian side were <0.1 ng/L. Continued contaminant track-downs of storm sewers, sediment and groundwater contributions from historical waste sites is needed to determine required remedial actions.

Occurrence of Persistent Organochlorine Pollutants in Sediments from Lake Piediluco, Italy

In surficial sediments of Lake Piediluco, a small, riverine, regulated and heavily modified water body in the Central Italian Apennines, contamination by persistent organochlorine pollutants (POPs) was assessed. During spring and autumn, six representative points were investigated. Reflecting a substantial zonation of pollution within this aquatic ecosystem, concentrations were found to vary from site to site. OCPs, particularly DDT and its metabolites DDE and DDD, were detected at varying frequencies and concentrations from 0.301 to 8.185 ng/g d.w., whilst total PCBs (Σ50 congeners) were from 0.570 to 10.206 ng/g d.w. Although both PCB congener-specific and homolog patterns suggest a prevalent presence of Aroclors 1254 and 1260, a more limited likely contribution of 1248 technical mixture is not to be excluded. In the western area of the lake, affected daily by continuous basin water remixing for hydroelectricity production, major seasonal differences in OCP concentrations were found. Conversely, the eastern area was typified by low seasonal fluctuation and small variation among sites. Regardless of either collection site or seasonality, polychlorinated-p-dioxin and furan (PCDD/Fs) contamination were below 8.3 pg WHO-TEQ/g. In contrast to the present study, POP pollution from PCBs, PCDD/Fs and some DDT-derived pollutants has not been investigated so far. Nevertheless, the concentrations are typical of low-polluted or pristine area lakes and are consistently lower than those of other Italian lakes of greater size and depth (e.g., Como, Garda and Maggiore). The physical and chemical properties of investigated analytes are more consistent with probable diffuse point source contamination originating from the catchment area, rather than from atmospheric depositions by regional or long-range transports. Finally, according to the international sediment quality guidelines (SQGs), Piediluco sediments pose a low risk to sediment-dwelling organisms and, more generally, to the wildlife of such aquatic ecosystems.

Association of polychlorinated biphenyls with vitamin D in female subjects

IntroductionPolychlorinated biphenyls (PCBs) are known endocrine disrupters. A potentially causal association of PCBs with vitamin D has been reported. Higher body mass index (BMI) is associated with lower PCB levels whilst the strongest association of PCBs with BMI is in non-obese individuals. Therefore, this study examined the association of PCBs with vitamin D3 (25(OH)D3) and the active 1,25-dihydrovitamin D3 (1,25(OH)2D3) in a cohort of non-obese women. Methods58 female participants (age 31.9 ± 4.6 years; BMI 25.7 ± 3.7 kg/m2) had seven indicator PCBs [PCB28, PCB52, PCB101, PCB118, PCB138, PCB153 and PCB180] measured using high resolution gas chromatography, with total PCB level calculated. 25(OH)D3 and 1,25(OH)2D3 levels were determined by isotope-dilution liquid chromatography tandem mass spectrometry. ResultsIn this cohort, vitamin D3 (25(OH)D3) and 1,25(OH)2D3 levels were 50.7 ± 25.3 nmol/L and 0.05 ± 0.02 ng/ml, respectively. Of those, 28 had vitamin D deficiency [25(OH)D3 level <20 ng/ml (<50nmol/)]. Total PCBs correlated positively with total group 25(OH)D3 (r = 0.22, p = 0.04) as did PCB118 (r = 0.25, p = 0.03). Total PCBs did not correlate with total group 1,25(OH)2D3; however, PCB180 did correlate positively with 1,25(OH)2D3 (r = 0.34, p = 0.03) as did PCB153 (r = 0.33, p < 0.03), with PCB 28 correlating negatively (r = −0.29, p < 0.04). In the vitamin D deficient subgroup, total PCBs, PCB153 and PCB180 positively correlated with 25(OH)D3 (p < 0.05).Multilinear regression analysis indicated all associations could be accounted for by BMI. ConclusionThough certain PCBs associated with 25(OH)D3 and 1,25(OH)2D3, all associations could be accounted for by BMI. This study therefore indicates that the deleterious effects from PCB accumulation are not mediated by effects on 25(OH)D3 or 1,25(OH)2D3.

Dietary exposure and risk assessment of polychlorinated biphenyls to artisanal fishermen and fish vendors: A case study of Dar es Salaam

AbstractBackgroundFish is one of the main sources of food and income in Tanzania, with 25% of the population depending on fishing activities as a source of food, income, and employment. But fish consumers are vulnerable to exposure to toxic pollutants including PCBs due to the lipophilic nature of the pollutants which accumulate in fish.ResultsThe estimated mean NDL PCBs concentration level was 0.84 ng/g w/w and the mean fish consumption rate among fishermen was 0.4 kg/day. Mean exposure of the fishermen and fish vendors to total PCBs ranged between 9.62 and 36.12 ng/kg bw/day depending on the estimation scenario (lower bound/upper bound) for both normal and higher fish consumers. The general population exposure to total PCBs was 0.4 and 0.8 ng/kg bw/day in lower‐bound and upper‐bound estimation scenarios respectively. The estimated lifetime carcinogenic and non‐carcinogenic hazard indices from intake of estimated total PCBs by fishermen and fish vendors exceeded the recommended tolerable daily intake, TDI, raising serious concerns for adverse health effects to fishermen and fish vendors. However, for the general population, there was no concern for any potential health effects as the estimated hazard indices were far below the recommended levels for intake of total PCBs.ConclusionThe exposure levels of fishermen and fish vendors to PCBs via consumption of Rastrelliger kanagutra (Indian mackerel) in Dar es Salaam are high enough to course serious health concerns, especially for high fish consumers and efforts should be taken to encourage diversification of fish species intake.

Investigation of polychlorinated biphenyls in breast milk from two regions in Bulgaria.

Human breast milk is an optimally balanced infant food and a suitable tool for assessing the burden of humans with lipophilic persistent organic pollutants. The aim of this study was to investigate the accumulation profile of polychlorinated biphenyls in breast milk of women living in Bulgaria and to assess the health risk to infants. Breast milk samples were obtained from 72 healthy primiparae and multiparae mothers, living in two regions in northeastern Bulgaria - Varna region and Dobrich region, in the period October 2019-July 2021. Important information for the study, such as age, body mass, smoking and dietary habits, was collected through a questionnaire. Fifteen congeners of PCBs, including six indicator congeners, were determined by capillary gas chromatography system with mass spectrometry detection. The lipid content of the tested samples was in the range from 0.5% to 6.7%, with average value 3.25%. The six indicator PCBs in human milk samples formed up to 89% of the total PCBs levels. The most abundant congener was PCB 153, followed by PCB 138 and PCB 180. Five of the 15 PCB congeners (77, 126, 128, 156, 169) were not detected in any of the milk samples. The arithmetic mean PCB levels in milk samples from Varna (32.7ng/g lw) were found higher than PCB levels in breast milk of mothers from Dobrich (22.5ng/g lw). The highest PCB levels were found in milk samples from primiparae mothers in 36-40 age group (for both regions). Infant exposure to PCBs present in human milk was estimated using toxic equivalents (TEQ). The health risk to infants was assessed and was compared to the tolerable daily intake (TDI). Positive correlation was found between the arithmetic mean PCBs levels and two important factors - the age and body mass index of the primiparae group. The mean values of the analyzed PCB congeners in breast milk samples from multiparae were lower than in those from primiparae mothers. The regional differences in PCB concentrations were small, suggesting similar exposures in the studied regions. The levels of PCBs in breast milk were found lower than levels from studies in other European countries. Statistical data does not show any association between PCB levels in milk and dietary habits. The results showed that infants are not at risk of any adverse effects caused by PCBs through breast milk.

Prenatal organochlorine pollutant exposure and risk of schizophrenia in a national birth cohort

Non-genetic prenatal exposures have been associated with schizophrenia risk. However, the role of prenatal exposure to environmental neurotoxicants in offspring schizophrenia risk has been studied in only limited instances. Polychlorinated biphenyls (PCBs) and the pesticide metabolite p,p′-dichlorodiphenyl dichloroethylene (DDE) have been linked to neurodevelopmental outcomes, including impairments implicated in schizophrenia. To determine whether prenatal maternal levels of organochlorine pollutants including PCBs or DDE are associated with schizophrenia in the offspring, an investigation was conducted in the Finnish Prenatal Study of Schizophrenia (FIPS-S), a case-control study nested in a national birth cohort. Cases were born in 1987–1991 and had at least two diagnoses of schizophrenia (ICD-10 F20; ICD-9 295) or schizoaffective disorder (ICD-10 F25; ICD-9 295.7) recorded in the national Care Register for Health Care. Each case was individually matched to a control on sex, date of birth, and residence in Finland on the date of case diagnosis. In 500 case-control pairs, PCB congeners 74, 99, 118, 138, 153, 156, 170, 180, 183, 187, and some widespread organochlorine pesticides or their metabolites including DDE were measured in archived prenatal maternal sera using gas chromatography - high triple quadrupole mass spectrometry. Maternal total PCBs were quantified as the sum of concentrations of the measured congeners. Associations with schizophrenia were examined using conditional logistic regression. Maternal PCB or DDE levels greater than the 75th percentiles of the control distributions showed no evidence of association with offspring schizophrenia (PCBs: adjusted odds ratio (aOR) = 1.13, 95 % CI = 0.85–1.50), p = 0.41; DDE: aOR = 1.08, 95 % CI = 0.80–1.45; p = 0.63). Maternal levels of either pollutant dichotomized at the 90th percentile or considered as a continuous variable also did not show evidence for association with offspring schizophrenia. This study found a lack of evidence that prenatal maternal levels of the organochlorine pollutants DDE and PCBs are associated with offspring risk of schizophrenia.

Occurrence, distribution, and risk assessment of halogenated organic pollutants (HOPs) in marine fish muscle: The case study of Vietnam

Halogenated organic pollutants (HOPs), including polychlorinated biphenyls (PCBs), organochlorine pesticides (OCPs), and chlorophenols (CPs), were identified in three marine fish species in Vietnam. Total PCBs, OCPs, and CPs concentrations ranged from 4.5 to 711.6 ng g−1 lipid weight (lw), 69.9–2360 ng g−1 lw, and 208.1–3941.2 ng g−1 lw, respectively. CPs were the most frequently detected pollutants in the marine environment of Vietnam of the three HOPs studied, followed by OCPs and PCBs. There are significant differences in HOPs between three types of seafood in Vietnam, including yellowstripe scad, Indian mackerel, and silver pomfret in this study. Notably, the types and amounts of HOPs found in the fish were differently influenced by the economic and industrial activities of the sampled areas. Despite these findings, the consumption of HOP-contaminated fish from the study areas was found not to pose any significant health risks to Vietnam's coastal population.

High quality diet attenuated the positive association between polychlorinated biphenyls and premature mortality among middle-aged and older adults

ObjectivePolychlorinated biphenyls (PCBs) have been reported to be a risk factor for premature death, while a high diet quality is thought to lower mortality risk. We aimed to examine whether PCBs were associated with higher all-cause and cause-specific mortality risk and whether such associations could be modified by the diet quality among US middle-aged and older adults. MethodsIncluded were 1259 participants aged 40 years or older from the 1999–2004 National Health and Nutrition Examination surveys. Exposure to PCBs was assessed in non-fasting serum samples, and mortality status was ascertained through December 31, 2019 using the public-use, linked mortality files. Diet quality was assessed using the Healthy Eating Index-2015 based on 24-h dietary recalls. Cox proportional hazard regression was applied to assess the associations of different PCB congener groups with mortality and the modifying effect by the diet quality. ResultsDuring a median follow-up of 17.75 years, 419 deaths occurred, including 131 from cardiovascular disease (CVD) and 102 from cancer. Serum concentrations of dioxin-like PCBs and non-dioxin-like PCBs were significantly associated with all-cause mortality, with hazard ratios (HRs) of 1.84 (95% confidence interval [CI], 1.10, 2.99) and 1.82 (1.09, 3.03) for extreme-tertile comparisons. A significant interaction was noted between dioxin-like PCBs and diet quality (P for interaction: 0.012), with a substantially more pronounced association among participants with a low diet quality (HR, 3.47; 95% CI: 1.29, 9.32), compared to those with a high diet quality (HR, 0.98; 95% CI: 0.40, 2.43). A similar weaker association was observed for total PCBs in participants with a high diet quality (P for interaction: 0.032). However, effect modifications by diet quality were not noted for the associations between different PCB groups and CVD mortality. ConclusionsWhile our findings need to be validated in other populations and mechanistic studies, they may suggest that a high quality diet could potentially attenuate the harmful effects of chronic PCB exposure.
