Characterization of a novel root-associated diazotrophic rare PGPR taxa, Aquabacter pokkalii sp. nov., isolated from pokkali rice: new insights into the plant-associated lifestyle and brackish adaptation

Salinity impacts crop growth and productivity and lowers the activities of rhizosphere microbiota. The identification and utilization of habitat-specific salinity-adapted plant growth-promoting rhizobacteria (PGPR) are considered alternative strategies to improve the growth and yields of crops in salinity-affected coastal agricultural fields. In this study, we characterize strain L1I39T, the first Aquabacter species with PGPR traits isolated from a salt-tolerant pokkali rice cultivated in brackish environments. L1I39T is positive for 1-aminocyclopropane-1-carboxylate deaminase activity and nitrogen fixation and can promote pokkali rice growth by supplying fixed nitrogen under a nitrogen-deficient seawater condition. Importantly, enhanced plant growth and efficient root colonization were evident in L1I39T-inoculated plants grown under 20% seawater but not in zero-seawater conditions, identifying brackish conditions as a key local environmental factor critical for L1I39T-pokkali rice symbiosis. Detailed physiological studies revealed that L1I39T is well-adapted to brackish environments. In-depth genome analysis of L1I39T identified multiple gene systems contributing to its plant-associated lifestyle and brackish adaptations. The 16S rRNA-based metagenomic study identified L1I39T as an important rare PGPR taxon. Based on the polyphasic taxonomy analysis, we established strain L1I39T as a novel Aquabacter species and proposed Aquabacter pokkalii sp nov. Overall, this study provides a better understanding of a marine-adapted PGPR strain L1I39T that may perform a substantial role in host growth and health in nitrogen-poor brackish environments.

Potential of Drought Tolerant Rhizobacteria Amended with Biochar on Growth Promotion in Wheat.

Drought stress is the prime obstacle for worldwide agricultural production and necessitates innovative strategies for enhancing crop resilience. This study explores the efficacy of plant growth-promoting rhizobacteria (PGPR) and biochar (BC) as sustainable amendments for mitigating the effects of drought on wheat growth. Multiple experiments were carried out on isolated strains to assess their drought tolerance potential and multiple plant growth-promoting attributes. Experiments in the laboratory and natural environment were conducted to assess the impact of plant growth-promoting rhizobacteria, biochar, and their synergistic application on various growth parameters of wheat. The results revealed that the drought-tolerant PGPR strains (Bacillus subtilis and Bacillus tequilensis), alongside biochar (rice husk), alleviated the phytotoxic impact of drought by increasing the root length from 17.0% to 70.0% and shoot length from 30.0% to 82.0% as compared to un-inoculated stressed controls. The total chlorophyll and carotenoid contents of the plants were substantially increased to 477% and 423%, respectively, when biochar and PGPR were applied synergistically. Significant enhancements in membrane stability index, relative water content, proline, and sugar level were achieved by combining biochar and bacterial strains, resulting in increases of 19.5%, 37.9%, 219%, and 300%, respectively. The yield of wheat in terms of plant height, spike length, number of spikelets per spike, and number of grains per spike was enhanced from 26.7% to 44.6%, 23.5% to 62.7%, 91.5% to 154%, and 137% to 182%, respectively. It was concluded that the biochar-based application of PGPR induced drought tolerance in wheat under water deficit conditions, ultimately improving the production and yield of wheat.

The Impact of Inoculation of Two Strains of Rhizobacteria on Radionuclide Transfer in Sesbania Grandiflora.

Transfer factors of some naturally-occurring and artificial radionuclides from an agricultural soil to rhizobacteria-treated Sesbania grandiflora, a small leguminous tree, were studied. Two plant growth promoting rhizobacteria (PGPR) strains (SCR17 and PCE2) were used to carry out an agricultural experiment in pots in semi-arid region (Syria). The results showed the bacterial strain (SCR17) increased the transfer and accumulation of 238U and 40K in Sesbania grandiflora, while both bacterial strains showed no effect on the accumulation of 234Th, 226Ra, 210Po and 210Pb in the treated plants. The transfer factor of 137Cs from soil to rhizobacteria-treated Sesbania grandiflora was negligible. The values of the transfer factors of 234Th, 226Ra, 210Po and 40K were found to be within the global values, while the values of the transfer factors of 238U and 210Pb were found to be relatively higher. This study highlights the importance of using Phytoremediation by PGPR strains for radionuclides-contaminated soils. Therefore, this method is a promising technique for the restoration and rehabilitation of contaminated sites with radionuclides, as it is low cost, easy to apply, and environmentally friendly.

Study on Plant Growth Promoting Traits of PGPR Isolates From Semi-Arid Kachchh, Gujarat Under Salt Stress Conditions

Salinity is a major problem in the agricultural sector, as it turns productive agronomical land to become unproductive. Therefore, Plant-growth-promoting rhizobacteria (PGPR), that live in the plant root zone named the rhizosphere, is one of the prominent solutions to overcome this problem in an eco-friendly manner as Rhizobacteria responds to osmotic stress and support plant development. Thus, the present study was aimed to characterize various traits of the PGPR strains isolated from saline soils of Kachchh. The characterization of the traits in the presence and absence of sodium chloride was assessed including IAA production (76.97±1.68 mg/l), Ammonia production (38.59±0.19 mg/l), Siderophore production (49.21±1.83%), Phosphate solubilization (4897.73±25.53 mg/l). When assessed for the salt tolerance of the strains in the presence of NaCl between 20-50 gm/L, the strain D6 exhibited a better growth even at 5% NaCl concentration (2.047OD). Further, the effect of PGPR on the growth of V. radiata was 100% in all the experimental setup, whereas in case of B. juncea, the highest germination of 96.67% was observed only in T2 and T1+T2+T3. Further, the molecular sequencing of the strains revealed the identification of the strains as Klebsiella quasipneumoniae, Bacillus paralicheniformis and Klebsiella pneumoniae.

Plant growth-promoting rhizobacteria Pseudomonas aeruginosa HG28-5 improves salt tolerance by regulating Na+/K+ homeostasis and ABA signaling pathway in tomato

Salinity stress badly restricts the growth, yield and quality of vegetable crops. Plant growth-promoting rhizobacteria (PGPR) is a friendly and effective mean to enhance plant growth and salt tolerance. However, information on the regulatory mechanism of PGPR on vegetable crops in response to salt stress is still incomplete. Here, we screened a novel salt-tolerant PGPR strain Pseudomonas aeruginosa HG28–5 by evaluating the tomatoes growth performance, chlorophyll fluorescence index, and relative electrolyte leakage (REL) under normal and salinity conditions. Results showed that HG28–5 colonization improved seedling growth parameters by increasing the plant height (23.7%), stem diameter (14.6%), fresh and dry weight in the shoot (60.3%, 91.1%) and root (70.1%, 92.5%), compared to salt-stressed plants without colonization. Likewise, HG28–5 increased levels of maximum photochemical efficiency of PSII (Fv/Fm) (99.3%), the antioxidant enzyme activities as superoxide dismutase (SOD, 85.5%), peroxidase (POD, 35.2%), catalase (CAT, 20.6%), and reduced the REL (48.2%), MDA content (41.3%) and ROS accumulation in leaves of WT tomatoes under salt stress in comparison with the plants treated with NaCl alone. Importantly, Na+ content of HG28–5 colonized salt-stressed WT plants were decreased by15.5% in the leaves and 26.6% in the roots in the corresponding non-colonized salt-stressed plants, which may be attributed to the higher K+ concentration and SOS1, SOS2, HKT1;2, NHX1 transcript levels in leaves of colonized plants under saline condition. Interestingly, increased abscisic acid (ABA) content and upregulation of ABA pathway genes (ABA synthesis-related genes NCED1, NCED2, NCED4, NECD6 and signal genes ABF4, ABI5, and AREB) were observed in HG28–5 inoculated salt-stressed WT plants. ABA-deficient mutant (not) with NCED1 deficiency abolishes the effect of HG28–5 on alleviating salt stress in tomato, as exhibited by the substantial rise of REL and ROS accumulation and sharp drop of Fv/Fm in the leaves of not mutant plants. Notably, HG28–5 colonization enhances tomatoes fruit yield by 54.9% and 52.4% under normal and saline water irrigation, respectively. Overall, our study shows that HG28–5 colonization can significantly enhance salt tolerance and improved fruit yield by a variety of plant protection mechanism, including reducing oxidative stress, regulating plant growth, Na+/K+ homeostasis and ABA signaling pathways in tomato. The findings not only deepen our understanding of PGPR regulation plant growth and salt tolerance but also allow us to apply HG28–5 as a microbial fertilizer for agricultural production in high-salinity areas.

Inoculation with Stutzerimonas stutzeri strains decreases N₂O emissions from vegetable soil by altering microbial community composition and diversity.

Plant growth-promoting rhizobacteria (PGPR) have been applied to mitigate nitrous oxide (N₂O) emissions from agricultural soils, but the microbial ecological mechanisms underlying N₂O mitigation are poorly understood. That is why only limited PGPR strains can mitigate N₂O emissions from agricultural soils. Therefore, it is of substantial significance to reveal soil ecological mechanisms of PGPR strains to achieve efficient and reliable N₂O-mitigating effect after inoculation. Inoculation with Stutzerimonas stutzeri strains decreased N₂O emissions from two soils with contrasting textures probably by altering soil microbial community composition and gene abundance involved in nitrification and denitrification. Our findings provide detailed insight into soil ecological mechanisms of PGPR strains to mitigate N₂O emissions from vegetable agricultural soils.

Comparative Analysis of Plant Growth-Promoting Rhizobacteria's Effects on Alfalfa Growth at the Seedling and Flowering Stages under Salt Stress.

Alfalfa (Medicago sativa L.), a forage legume known for its moderate salt-alkali tolerance, offers notable economic and ecological benefits and aids in soil amelioration when cultivated in saline-alkaline soils. Nonetheless, the limited stress resistance of alfalfa could curtail its productivity. This study investigated the salt tolerance and growth-promoting characteristics (in vitro) of four strains of plant growth-promoting rhizobacteria (PGPR) that were pre-selected, as well as their effects on alfalfa at different growth stages (a pot experiment). The results showed that the selected strains belonged to the genera Priestia (HL3), Bacillus (HL6 and HG12), and Paenibacillus (HG24). All four strains exhibited the ability to solubilize phosphate and produce indole-3-acetic acid (IAA) and 1-aminocyclopropane-1-carboxylate (ACC) deaminase. Among them, except for strain HG24, the other strains could tolerate 9% NaCl stress. Treatment with 100 mM NaCl consistently decreased the IAA production levels of the selected strains, but inconsistent changes (either enhanced or reduced) in terms of phosphate solubilization, ACC deaminase, and exopolysaccharides (EPS) production were observed among the strains. During the various growth stages of alfalfa, PGPR exhibited different growth-promoting effects: at the seedling stage, they enhanced salt tolerance through the induction of physiological changes; at the flowering stage, they promoted growth through nutrient acquisition. The current findings suggest that strains HL3, HL6, and HG12 are effective microbial inoculants for alleviating salt stress in alfalfa plants in arid and semi-arid regions. This study not only reveals the potential of indigenous salt-tolerant PGPR in enhancing the salt tolerance of alfalfa but also provides new insights into the mechanisms of action of PGPR.

Beneficial rhizobacterium Bacillus velezensis SQR9 regulates plant nitrogen uptake via endogenous signaling pathway.

Nitrogen fertilizer is widely used in agriculture to boost crop yields, plant growth-promoting rhizobacteria (PGPRs) can increase plant nitrogen use efficiency through nitrogen fixation and organic nitrogen mineralization. However, it is not known if they can activate the plant uptake of nitrogen. In this study, we investigated the effects of a PGPR strain Bacillus velezensis SQR9-emitted volatile compounds (VCs) on plant nitrogen uptake. Strain SQR9 VCs promoted nitrogen accumulation in both rice and Arabidopsis. In addition, isotope labeling experiments showed that strain SQR9 VCs promoted the absorption of nitrate and ammonium. Several key nitrogen uptake genes were up-regulated by strain SQR9 VCs, such as AtNRT2.1 in Arabidopsis and OsNAR2.1, OsNRT2.3a and OsAMT1 family members in rice, and the deletion of these genes compromised the promoting effect of SQR9 VCs on plant nitrogen absorption. Furthermore, the calcium (Ca2+) and transcription factor NIN-LIKE PROTEIN 7 play an important role in strain SQR9 VCs-promoted nitrate uptake. Taken together, our results suggest that PGPRs can promote nitrogen uptake through regulating the plant's endogenous signaling and nitrogen transport pathways.

ACC-Deaminase producing Pseudomonas putida RT12 inoculation: A promising strategy for improving Brassica juncea tolerance to salinity stress

An important abiotic stressor that hinders plant growth, nutrient uptake, and global agricultural productivity is soil salinity. Among the different strategies to overcome the issue of salinity in agriculture sector, plant growth-promoting rhizobacteria (PGPR) have gained recognition as promising beneficial microbes that can improve plants’ response to biotic and abiotic stressors. The salinity tolerance and traits that promote plant growth of eight PGPR strains (RT1, RT2, RT3, RT4, RT5, RT7, and RT12) were evaluated in this study. During screening, one strain, RT12, had the highest plant growth-promoting activity and salt tolerance in the group. The strain when subjected to NaCl stress showed quantitative ACC-deaminase activity, in the presence of NaCl at various concentrations, demonstrating extraordinary tolerance to salt stress by withstanding doses of up to 3M NaCl. In order to further investigate the effects of salt stress on Brassica juncea (mustard), RT12, which was identified as Pseudomonas putida using 16s RNA sequencing, was inoculated. Two salt treatments (100 and 150 mM) were applied to the mustard variety ‘Mingora’ in a greenhouse. The results revealed that through ACC utilization, PGPR directly induced plant growth in salt-stressed mustard plants by lowering excess ethylene production. All plant parameters were negatively impacted by an increase in NaCl concentration in uninoculated plants. However, P. putida RT12 inoculation enhanced all growth parameters, antioxidant production, total soluble sugar (TSS), total protein (TP), proline, relative water content (RWC), chlorophyll contents, and nutrient uptake in salt-treated plants. The inoculation with P. putida also caused a marked decline in Na+ uptake and an increase in K+ uptake in the shoot. By maintaining a greater K+/Na+ ratio in the tissues of RT12-inoculated plants compared to controls, this change in ion uptake helped to maintain nutritional balance of the plants. The findings suggest that inoculating plants with ACC deaminase-producing PGPR, such as P. putida RT12, may boost growth and stress resistance.

Integrated physiological, biochemical and transcriptomic analyses reveal the mechanism of salt tolerance induced by a halotolerant Serratia sp. NTN6 in maize

Salt stress is a serious environmental challenge that hinders crop growth and yield. Plant growth-promoting rhizobacteria (PGPR) can successfully and environmentally friendly boost plant development in saline soils. In this study, a halotolerant Serratia sp. NTN6 was isolated from the rhizosphere soil of halophyte Puccinellia tenuiflora and its plant growth-promoting properties were determined. NTN6 inoculation significantly promoted seed germination and seedling growth in maize under salt stress. Shoot height, stem thickness, root fresh weight, shoot fresh weight, root dry weight, and shoot dry weight of salt-stressed seedlings increased by 36.3%, 32.5%, 146.4%, 193.6%, 25.0%, and 109.6%, respectively, after inoculation with NTN6. NTN6-mediated maize salt tolerance was revealed via transcriptomic, physiological, and biochemical analyses. Under salt stress, NTN6 regulated the transcript levels of genes related to chlorophyll synthesis, photosynthesis apparatus, and carbon assimilation pathways, and thus increasing chlorophyll content, Fv/Fm, and net photosynthetic rate; NTN6 reduced the content of O2•–, H2O2, and MDA by regulating the transcripts and activities of antioxidant enzymes; NTN6 affected the transcriptional response of genes involved in ABA synthesis and signaling to salt stress and reduced ABA level. Finally, NTN6 boosted photosynthetic capacity in salt-stressed maize by modulating photochemical activity, C4 pathway and Calvin cycle as well as mitigating reactive oxygen species damage to the photosystem reaction center. This study first investigated the mechanism of Serratia sp.-induced salt tolerance in maize and these findings demonstrate the valuable contribution of halotolerant PGPR strain from halophytes in improving salinity tolerance in non-halophytic crops.

Microbial Interactions as a Sustainable Tool for Enhancing PGPR Antagonism against Phytopathogenic Fungi

Microbial interactions, which regulate the dynamics of eco- and agrosystems, can be harnessed to enhance antagonism against phytopathogenic fungi in agriculture. This study tests the hypothesis that plant growth-promoting rhizobacteria (PGPR) can also be potential biological control agents (BCAs). Antifungal activity assays against potentially phytopathogenic fungi were caried out using cultures and cell-free filtrates of nine PGPR strains previously isolated from agricultural soils. Cultures of Bacillus sp. BS36 inhibited the growth of Alternaria sp. AF12 and Fusarium sp. AF68 by 74 and 65%, respectively. Cell-free filtrates of the same strain also inhibited the growth of both fungi by 54 and 14%, respectively. Furthermore, the co-cultivation of Bacillus sp. BS36 with Pseudomonas sp. BS95 and the target fungi improved their antifungal activity. A subsequent metabolomic analysis using Fourier-transform ion cyclotron resonance mass spectrometry (FTICR-MS) identified fengycin- and surfactin-like lipopeptides (LPs) in the Bacillus sp. BS36 cell-free filtrates, which could explain their antifungal activity. The co-production of multiple families of LPs by Bacillus sp. BS36 is an interesting feature with potential practical applications. These results highlight the potential of the PGPR strain Bacillus sp. BS36 to work as a BCA and the need for more integrative approaches to develop biocontrol tools more accessible and adoptable by farmers.

Effects of plant growth-promoting rhizobacteria on blueberry growth and rhizosphere soil microenvironment.

Plant growth-promoting rhizobacteria (PGPR) have a specific symbiotic relationship with plants and rhizosphere soil. The purpose of this study was to evaluate the effects of PGPR on blueberry plant growth, rhizospheric soil nutrients and the microbial community. In this study, nine PGPR strains, belonging to the genera Pseudomonas and Buttiauxella, were selected and added into the soil in which the blueberry cuttings were planted. All the physiological indexes of the cuttings and all rhizospheric soil element contents were determined on day 6 after the quartic root irrigation experiments were completed. The microbial diversity in the soil was determined using high-throughput amplicon sequencing technology. The correlations between phosphorus solubilization, the auxin production of PGPR strains, and the physiological indexes of blueberry plants, and the correlation between rhizospheric microbial diversity and soil element contents were determined using the Pearson's correlation, Kendall's tau correlation and Spearman's rank correlation analysis methods. The branch number, leaf number, chlorophyllcontentand plant height of the treated blueberry group were significantly higher than those of the control group. The rhizospheric soil element contents also increased after PGPR root irrigation. The rhizospheric microbial community structure changed significantly under the PGPR of root irrigation. The dominant phyla, except Actinomycetota, in the soil samples had the greatest correlation with phosphorus solubilization and the auxin production of PGPR strains. The branch number, leaf number, and chlorophyllcontent had a positive correlation with the phosphorus solubilization and auxin production of PGPR strains and soil element contents. In conclusion, plant growth could be promoted by the root irrigation of PGPR to improve rhizospheric soil nutrients and the microenvironment, with modification of the rhizospheric soil microbial community. Bacillus cereus: an effective bio-inoculant for promoting salt stress tolerance of rice seedlings under saline soil conditions

Plant growth-promoting rhizobacteria (PGPR) are a powerful tool to maintain sustainable agriculture and promote plant resistance to biotic and abiotic types of stress. Salinity, a major abiotic stress hampers plant growth, development, and yield. Salt-tolerant PGPR are effective agents for ameliorating salinity effects on rice plants. The present study endeavored to isolate, determine halotolerant ability, characterize Plant Growth Promoting (PGP) traits, and finally observe the effect of PGPR strain on plant growth promotion of rice plants under saline and non-saline conditions. Based on the 16S rRNA gene sequencing technique, the rhizobacterial strain DB2 was identified as Bacillus cereus ATCC 14579(T) from NCMR, NCCS Pune. To check the growth-promoting ability, the strain was inoculated with two rice genotypes named Chinsurah Nona I (salt tolerant-non aromatic) and Badshabhog (aromatic) under polyhouse conditions. Results showed a significant increment in relative water content (RWC), total chlorophyll content (TCC), root length (RL), and shoot length (SL) in both rice genotypes inoculated with DB2 under both saline and non-saline conditions. Under non-saline conditions enhancement of RWC, TCC, RL, and SL was better in DB2 inoculated Chinsurah Nona I than in Badshabhog inoculated with DB2. Whereas, DB2-inoculated Badshabhog showed more recovery of RWC, TCC, RL, and SL than DB2-inoculated Chinsurah Nona I under saline conditions. Under salt stress conditions, inoculation with the rhizobacterial strain showed a significant reduction in electrolytic leakage (EL) in rhizobacteria inoculated with both rice genotypes. Moreover, DB2 inoculation showed a significant reduction in Na+ content in the roots of Chinsurah Nona I (44.6%) and Badshabhog (24.5%) rice genotypes. The present study has indicated that the application of salt-tolerant PGPR may be an effective and sustainable solution for rice cultivation under salt-stress conditions.

Screening of heavy metal-resistant rhizobial and non-rhizobial microflora isolated from Trifolium sp. growing in mining areas.

Plant growth-promoting rhizobacteria (PGPR) can promote plant growth and development with several beneficial effects, especially in challenging environmental conditions, such as the presence of toxic contaminants. In this study, 49 isolates obtained from Trifolium sp. nodules growing on a Pb/Zn mine site were characterized for PGP traits including siderophores production, phosphate solubilization, extracellular enzymes production, and antifungal activity. The isolates were also screened for their ability to grow at increasing concentrations of NaCl and heavy metals, including lead, zinc, cobalt, copper, nickel, cadmium, and chromium. The findings of our study indicated that isolates Cupriavidus paucula RSCup01-RSCup08, Providencia rettgeri RSPro01, Pseudomonas putida RSPs01, Pseudomonas thivervalensis RSPs03-RSPs09, and Acinetobacter beijerinckii RSAci01 showed several key traits crucial for promoting plant growth, thus demonstrating the greatest potential. Most isolates displayed resistance to salt and heavy metals. Notably, Staphylococcus xylosus RSSta01, Pseudomonas sp. RSPs02, Micrococcus yunnanensis RSMicc01, and Kocuria dechangensis RSKoc01 demonstrated a significant capacity to grow at salt concentrations ranging from 10 to 20%, and isolates including Cupravidus paucula RSCup01-RSCup08 exhibited resistance to high levels of heavy metals, up to 1300 mg/L Pb++, 1200 mg/L Zn++, 1000 mg/L Ni++, 1000 mg/L Cd++, 500 mg/L Cu++, 400 mg/L Co++, and 50 mg/L CrVI+. Additionally, the analysis revealed that metal-resistant genes pbrA, czcD, and nccA were exclusively detected in the Cupriavidus paucula RSCup01 strain. The results of this study provide insights into the potential of plant growth-promoting rhizobacteria strains that might be used as inoculants to improve phytoremediation in heavy metal-contaminated soils.

A novel PGPR strain, Streptomyces lasalocidi JCM 3373T, alleviates salt stress and shapes root architecture in soybean by secreting indole-3-carboxaldehyde.

While soybean (Glycine max L.) provides the most important source of vegetable oil and protein, it is sensitive to salinity, which seriously endangers the yield and quality during soybean production. The application of Plant Growth-Promoting Rhizobacteria (PGPR) to improve salt tolerance for plant is currently gaining increasing attention. Streptomycetes are a major group of PGPR. However, to date, few streptomycetes has been successfully developed and applied to promote salt tolerance in soybean. Here, we discovered a novel PGPR strain, Streptomyces lasalocidi JCM 3373T, from 36 strains of streptomycetes via assays of their capacity to alleviate salt stress in soybean. Microscopic observation showed that S. lasalocidi JCM 3373T does not colonise soybean roots. Chemical analysis confirmed that S. lasalocidi JCM 3373T secretes indole-3-carboxaldehyde (ICA1d). Importantly, IAC1d inoculation alleviates salt stress in soybean and modulates its root architecture by regulating the expression of stress-responsive genes GmVSP, GmPHD2 and GmWRKY54 and root growth-related genes GmPIN1a, GmPIN2a, GmYUCCA5 and GmYUCCA6. Taken together, the novel PGPR strain, S. lasalocidi JCM 3373T, alleviates salt stress and improves root architecture in soybean by secreting ICA1d. Our findings provide novel clues for the development of new microbial inoculant and the improvement of crop productivity under salt stress.

The Importance of Considering Levels of P and N Fertilization to Promote Beneficial Interaction between Rapeseed and Phosphate-Solubilizing Bacteria

Biointrants constitute a promising opportunity to lower mineral input on rapeseed, characterized by high nutrient requirements. As bio-inoculants, phosphate-solubilizing bacteria (PSB) could increase the amount of available P in a soil solution. However, the deployment of these bio-inoculants in fields is not always successful. Disentangling the factors conditioning their reliability is necessary. Because the activities of microorganisms are particularly subject to nutrient availability, the N fertilization level could represent a key factor for the success of PSB inoculation in the early stages of plant growth. In this study, Pfaba (Pseudomonas sp.), a promising plant growth-promoting rhizobacteria (PGPR) strain isolated from soil, was inoculated on rapeseed grown in rhizotrons under two N fertilization levels (N160 or N80) in P labile or P complexed conditions. Pfaba confirmed its PSB potential to solubilize recalcitrant P complexed forms for the benefit of plant growth, but only when the N supply is adequate (N80). In a P complexed environment, Pfaba tended to increase root and shoot biomass (respectively, from 2.17 ± 0.47 g for control modality to 2.88 ± 0.85 g, and from 6.06 ± 1.67 g for control modality to 8.33 ± 1.70 g), increase the P and N contents in roots (respectively, from 0.15 ± 0.09 mg for control modality to 0.70 ± 0.51 mg, and from 37.90 ± 11.09 mg for control modality to 41.34 ± 14.16 mg), and restore root length at a comparable level than plants supplemented with labile P. Conversely, these positive effects were inhibited with lower levels of N fertilization. Our results highlight the importance of nutrient availability to promote beneficial interaction between plants and microorganisms. These findings could also contribute to ensuring the successful deployment of microbial biointrants.

Characterization of Rhizosphere Microbial Diversity and Selection of Plant-Growth-Promoting Bacteria at the Flowering and Fruiting Stages of Rapeseed.

Plant rhizosphere microorganisms play an important role in modulating plant growth and productivity. This study aimed to elucidate the diversity of rhizosphere microorganisms at the flowering and fruiting stages of rapeseed (Brassica napus). Microbial communities in rhizosphere soils were analyzed via high-throughput sequencing of 16S rRNA for bacteria and internal transcribed spacer (ITS) DNA regions for fungi. A total of 401 species of bacteria and 49 species of fungi in the rhizosphere soil samples were found in three different samples. The composition and diversity of rhizosphere microbial communities were significantly different at different stages of rapeseed growth. Plant-growth-promoting rhizobacteria (PGPRs) have been widely applied to improve plant growth, health, and production. Thirty-four and thirty-one PGPR strains were isolated from the rhizosphere soil samples collected at the flowering and fruiting stages of rapeseed, respectively. Different inorganic phosphorus- and silicate-solubilizing and auxin-producing capabilities were found in different strains, in addition to different heavy-metal resistances. This study deepens the understanding of the microbial diversity in the rapeseed rhizosphere and provides a microbial perspective of sustainable rapeseed cultivation.

Utilization of Stenotrophomonas koreensis and Bacillus amyloliquefaciens for Improving Growth, Reducing Nitrogen Fertilization and Controlling Bipolaris sorokiniana in Wheat

Wheat (<em>Triticum aestivum</em> L.), a vital cereal, faces significant challenges from common root rot and spot blotch diseases caused by <em>Bipolaris sorokiniana</em>. This study aimed to explore the potential of plant growth promoting rhizobacteria (PGPR) to enhance wheat growth, reduce fertilizer input, and combat <em>Bipolaris </em>diseases. Two PGPR isolates, selected for their superior antagonistic properties, were identified as <em>Stenotrophomonas koreensis</em> RB11 and <em>Bacillus amyloliquefaciens </em>RB12. These PGPR strains displayed multiple plant growth promoting and biocontrol attributes, including phosphate solubilization, indole-3-acetic acid production, nitrogen fixation and antagonism against <em>B. sorokiniana</em> and other fungi. Wheat seed priming with the PGPR significantly improved germination, plant growth, nutrient content and biomass carbon accumulation in the rhizosphere soil. Importantly, the application of RB11 and RB12 allowed for a 25% and 50% reduction in nitrogen fertilizer usage, respectively, without compromising the yield. RB11 and RB12 also demonstrated potent inhibitory effects on <em>B. sorokiniana</em> conidial germination and significantly controlled common root rot and spot blotch in wheat, similar to those observed with the fungicide Protaf 250EC. Overall, this study underscores the multifaceted roles of <em>S. koreensis</em> RB11 and <em>B. amyloliquefaciens</em> RB12 in promoting wheat growth, reducing fertilizer inputs and effectively suppressing wheat pathogens. These findings contribute to the development of PGPR-based strategies for sustainable crop production and disease control.

The Potential Use of Plant Growth Promoting Rhizobacteria (PGPR) for Medicinal Plant Cultivation in Meghalaya: A Review

Beneficial PGPR have been identified in a range of plant species, including those with medicinal properties. These PGPR have been shown to enhance the growth and development of medicinal plants through the improvement of nutrient availability in rhizospheric root systems. Some medicinal plants have been found to harbour specific strains of PGPR that are associated with increased medicinal properties and enhanced bioactive compound production. So, the use of PGPR can minimise the application of chemical fertilizer for the production of rare species of medicinal plants. This review highlights the future research works which are needed in many areas of Meghalaya by the use of various beneficial strains of PGPR to reduce the use of pesticide for commercial application in the cultivation of medicinal plants. The PGPR has the ability to increase the soil fertility be decreasing the concentration of inorganic fertilizer and thereby they are regarded as the potential strains to be used as biofertilizer which in turn will also improve the cultural practices of Meghalaya to conserve a large number of rare and endangered medicinal plants.

Plant growth-promoting rhizobacteria: Peribacillus frigoritolerans 2RO30 and Pseudomonas sivasensis 2RO45 for their effect on canola growth under controlled as well as natural conditions.

Even though canola is one of the most important industrial crops worldwide, it has high nutrient requirements and is susceptible to pests and diseases. Therefore, natural methods are sought to support the development of these plants. One of those methods could be a plant growth-promoting rhizobacteria (PGPR) that have a beneficial effect on plant development. The aim of this study was a genomic comparison of two PGPR strains chosen based on their effect on canola growth: Peribacillus frigoritolerans 2RO30, which stimulated canola growth only in sterile conditions, and Pseudomonas sivasensis 2RO45, which promoted canola growth in both sterile and non-sterile conditions. First of all, six bacterial strains: RO33 (Pseudomonas sp.), RO37 (Pseudomonas poae), RO45 (Pseudomonas kairouanensis), 2RO30 (Peribacillus frigoritolerans), 2RO45 (Pseudomonas sivasensis), and 3RO30 (Pseudomonas migulae), demonstrating best PGP traits in vitro, were studied for their stimulating effect on canola growth under sterile conditions. P. frigoritolerans 2RO30 and P. sivasensis 2RO45 showed the best promoting effect, significantly improving chlorophyll content index (CCI) and roots length compared to the non-inoculated control and to other inoculated seedlings. Under non-sterile conditions, only P. sivasensis 2RO45 promoted the canola growth, significantly increasing CCI compared to the untreated control and to other inoculants. Genome comparison revealed that the genome of P. sivasensis 2RO45 was enriched with additional genes responsible for ACC deaminase (acdA), IAA (trpF, trpG), and siderophores production (fbpA, mbtH, and acrB) compared to 2RO30. Moreover, P. sivasensis 2RO45 showed antifungal effect against all the tested phytopathogens and harbored six more biosynthetic gene clusters (BGC), namely, syringomycin, pyoverdin, viscosin, arylpolyene, lankacidin C, and enterobactin, than P. frigoritolerans 2RO30. These BGCs are well known as antifungal agents; therefore, it can be assumed that these BGCs were responsible for the antifungal activity of P. sivasensis 2RO45 against all plant pathogens. This study is the first report describing P. sivasensis 2RO45 as a canola growth promoter, both under controlled and natural conditions, thus suggesting its application in improving canola yield, by improving nutrient availability, enhancing stress tolerance, and reducing environmental impact of farming practices.
