Protein Typing of Fish, Pork, and Beef by Disc Electrophoresis

Abstract Techniques and apparatus are described for the identification of species of protein by disc electrophoresis. The technique is more convenient to use than starch gel zone electrophoresis and takes less time.

Studies of the plasma proteins in pregnancy using starch gel zone electrophoresis

Serum proteins in 126 serum samples from 100 pregnant women were analyzed by starch gel zone electrophoresis. An elevation of the slow α 2-globulin ( α 2-macroglobulin) was noted in 59% of the samples, an increase of β-globulin (transfenin) in 29%, and an increase of fast α 2-globulin and decrease of albumin and γ-globulin in approximately 12%. Samples from only 3 patients showed the unusual finding known as the “pregnancy band” migrating in front of the slow α 2 fraction. Changes of the pattern during the course of pregnancy have been discussed.

The expressed sap was separated into its components by agar gel zone electrophoresis. p-Fluorophenylalanine was shown to be incorporated into TMV at a reduced rate as compared with the natural amino acid phenylalanine. The analogue inhibited incorporation of phosphorus, a reversal probably due to the increase in phenylalanine observed in the soluble amino-acid-free fraction.

Cell Bound ^|^alpha;-Amylase in Aspergillus oryzae

α-Amylase in Aspergillus oryzae is generally known to be present in mycelium as well as in medium, whether it is grown with or without shaking. The interesting fact is reported in this paper, when A. oryzae was grown in a phosphate deficient medium, however, α-amylase accumulation occurred only within the cell. From the results of pH-activity curve, starch gel zone electrophoresis and differential centrifugation, there seemed to be no distinctions between exo and endocellular α-amylase. The liberation of α-amylase from the cell was observed to be dependent on pH and the concentrations of anions. The significance of the α-amylase accumulation and liberation in this system is discussed.

Studies on corneal polysaccharides: I. Separation

A method for the isolation of the polysaccharides of bovine corneal stroma has been developed. A procedure based on chromatography on ECTEOLA columns readily removed the peptides from proteolytic digests of corneal tissue and made possible a nearly quantitative recovery of an essentially protein-free product. A detailed investigation of the general applicability and limitations of ECTEOLA chromatography revealed that stepwise elution with salt solutions of increasing concentration effected partial separation of the individual polysaccharides. Gel filtration and zone electrophoresis studies showed that complete fractionation was not achieved because of the charge and molecular weight polydispersity within each species. Alcohol fractionation of ECTEOLA eluates, however, obviated these difficulties and proved to be highly effective in rendering a virtually complete separation of the glucosamino-glycans and the galaotosaminoglycans. The total hexosamine content of bovine corneal stroma was found to be 1.6% of the dry tissue, and at least 90% of the hexosamine appeared to be part of the polysaccharide fractions.

SummaryTo determine the time of appearance of the haptoglobin serum proteins in the developing human being, starch gel zone electrophoresis was performed on sera from 5 fetuses, 63 umbilical cord bloods, and 86 infants. Plasma cells and serum proteins in marine fish.

Summary1) Serum proteins of 18 species of fish have been analyzed by means of starch gel zone electrophoresis. In 10 of these species, observations were also made on occurrence of plasma cells in spleens. 2) Elasmobranchs had typical plasma cells in their spleens and proteins in their serum migrating like gamma globulins upon electrophoresis. Teleosts, however, generally had very few plasma cells in their spleens and no demonstrable gamma globulin in the serum. These observations suggest that among marine fish, there may be an association between plasma cells and gamma globulins.
