Phytochemical Screening and Antimicrobial Activity of Various Extracts of Aerial Parts of Rhanterium epapposum

Rhanterium epapposum, native to the Arabian Peninsula, is traditionally used to cure skin infections. The objective is to screen the phytochemical content and antimicrobial activity of aqueous, methanol and 80% methanol extracts of aerial parts of R. epapposum. The phytochemical screening of aqueous, methanolic, and 80% methanol extracts of R. epapposum was conducted using gas chromatographymass spectrometry. The antimicrobial activities of the extracts were assessed by well diffusion and microdilution methods. Qualitative phytochemical analysis revealed the presence of 2-methoxy-4-vinylphenol in all three extracts, whereas ethanol, 2-methoxy-, acetate; n-hexadecanoic acid; and 2,3-butanediol are present in higher amount exclusively in the methanol, 80% methanol and aqueous extracts of the aerial parts of R. epapposum, respectively. The highest antibacterial activity was shown by the aqueous extract S. aureus, P. aeruginosa, E. cloacae, and K. pneumoniae, methanolic extract against S. aureus, methicillin-resistant S. aureus, and E. coli, and 80% methanol extract against S. epidermidis, and S. paucimobilis. Interestingly, 80% methanol extracts showed the highest antifungal activity against C. albicans, C. guillermondii, C. vaginalis, C. utilis, and C. tropicalis. The aerial parts of R. epapposum showed broad-spread antimicrobial activity against bacteria and fungi. Especially, the 80% methanol extract showed potent antifungal activity against all the tested fungal strains.

Alpha-glucosidase inhibitory and antioxidant activities of various extracts of aerial parts of Fagonia indica Burm. F.

Purpose: To investigate in vitro antioxidant and anti-diabetic potentials of Fagonia indica Burm.f. Var. indica in order to provide scientific evidence for its traditional use.
 Methods: Cold maceration method was employed for the preparation of extracts of aerial parts of the plant using chloroform, n-hexane, methanol and water as solvents. Total flavonoid and polyphenolic content of various extracts were determined by standard methods. The antioxidant and anti-diabetic activities of the extracts were determined using 1, 1-diphenyl-2-picrylhydrazyl (DPPH) and in vitro αglucosidase (maltase) inhibitory assays, respectively.
 Results: Preliminary results indicated the presence of various phytochemicals in the extracts. The chloroform extract exhibited the highest contents of total flavonoids (65.98 ± 1.63 mg QE/g) and polyphenols (26.75 ± 1.09 mg GAE/g). This extract also showed the highest free radical scavenging (64.74 ± 1.43 %) with IC50 value of 34.18 ± 5.57 µg/mL while the methanol extract exerted the highest αglucosidase (maltase) inhibitory activity (45.22 ± 0.46 %) with half-maximal concentration (IC50) of 220.4 ± 0.41 µg/mL.
 Conclusion: The extracts of the aerial parts of Fagonia indica possess significant anti-diabetic and antioxidant effect, thus justifying the traditional use of the plant for treatment of diabetes.

Estimation of flavonoids and screening of in vitro antioxidant activity of various extracts of aerial parts of Blepharis maderaspatensis by hydroxyl radical and total antioxidant activity

Antioxidants are substances that hamper or inhibit the oxidation of oxidizable substrates in the cells. The aerial parts of Blepharis maderaspatensis dry powder was extracted with various solvents (PE, EA and methanol) through Soxhlet extractor. The aerial parts of different concentrates (Pet.ether, ethyl acetate and methanol) of Blepharis maderaspatensis was evaluated for its in-vitro antioxidant potential by hydroxyl radical, total antioxidant activity taking ascorbate used as standard for the both methods and total flavonoids content was estimated as equivalent to rutin. The methanolic concentrates of Blepharis maderaspatensis & standard exhibited antioxidant potential possessing IC50 210µg/ml & 62µg/ml (hydroxyl radical) 200µg/ml & 57µg/ml (Total antioxidant activity) respectively. An IC50 value was originate that methanolic concentrates of Blepharis maderaspatensis more efficient in hydroxyl radical, total antioxidant activity compared EA & PE concentrates. The methanolic and EA concentrates of Blepharis maderaspatensis showed the total flavonoids content (11.230±0.22 and 2.858±0.56 mg of rutin equivalent/g of powder) respectively. The difference in scavenging potential of the extracts can be due to variation in the percentage of bioactive compound flavonoids present in methanolic extracts. Invitro antioxidant studies obviously show methanolic concentrates of Blepharis maderaspatensis have better antioxidant activity due to the presence of total flavonoids content. The findings from the study reveal that the antioxidative potentials of methanolic concentrates Blepharis maderaspatensis could serve as free radical inhibitors, possibly acting as natural antioxidants.

Evaluation of the antioxidant activity of various extracts of aerial parts of Cassia absus: An in-vitro techniques

Cassia absus (Linn)Cassia absus(Linn) (family Fabaceae ) is generally known as “chaksu ” inan ayurvedic traditional system.The current study,aerial parts of different concentrates(Pet.ether, ethyl acetate and methanol) of Cassia absus, was evaluated for its in-vitro antioxidant potential byDiphenylpicrylhydrazyl radical,nitric oxide activity andtotal antioxidant activitytaking ascorbate as the standardfor all the three methods. The IC50 value was originated that methanolic concentrates of Cassia absusmore efficient inDiphenylpicrylhydrazyl radical,nitric oxide activity, total antioxidant activitycompared EA&PEconcentrates.The methanolic concentrates of Cassia absus& ascorbic acid exhibited antioxidant potential possessing IC50230µg/ml &130µg/ml (Nitric oxide). 205µg/ml &57µg/ml (total antioxidant),195µg/ml & 66µg/ml (Diphenylpicrylhydrazyl radical)respectively. The difference in the scavenging potential of the extracts can be due to variation in the percentage of bioactive compounds present in different solvents. Invitroantioxidant studiesobviouslyshow the methanolic concentrates of Cassia absushave better antioxidant activity. This result indicates that aerial parts of methanolic concentratesCassia absuscould serve as a natural antioxidant, which may be useful in preventfree radical-induced diseases.

A PHYTOCHEMICAL STUDY ON EUPATORIUM GLANDULOSUM

Objective: Eupatorium glandulosum is a useful medicinal plant belongs to the family Asteraceae. It is traditionally used to treat various diseases such as wound healing, antioxidant, and antiproliferative. The present study was aimed to investigate the physicochemical and phytochemical properties of various extracts of aerial parts of E. glandulosum. 
 Methods: The E. glandulosum plant materials were extracted using the solvents alcohol, ethyl acetate, and chloroform by Soxhlet method. The extracts were screened for physiochemical constants, preliminary phytochemical analysis for carbohydrates, glycosides, alkaloids, flavonoids, phenols, tannins, and saponins. The quantitative phytochemical analysis was carried out for total flavonoid and total phenols using standard procedures.
 Results: The physiochemical constituents such as total ash, acid-insoluble ash, and water-soluble ash were found to be 14.25% (w/w), 5% (w/w), and 7.30% (w/w), respectively. The preliminary phytochemical screening confirmed the presence of carbohydrates, glycosides, alkaloids, flavonoids, phenols, tannins, and saponins. The flavonoid content of the plant extracts was found to be in the descending order ethyl acetate ˃alcohol ˃chloroform and the phenolic content was found to be alcohol ˃ethyl acetate ˃chloroform.
 Conclusion: The result showed the presence of phytochemical constituents and higher values of phenolic and flavonoid content make the plant useful for the formulation of the different drugs for human uses for treating various diseases.

Study of in-vitro antioxidant activity of various extracts of aerial parts of Olax scandens

Olax scandensRoxb. (family Olacaceae) Available in throughout tropical India. The current study, aerial parts of different concentrates of Olax scandenswas evaluated for its in-vitro antioxidant potential by Diphenylpicrylhydrazyl activity, superoxide scavenging activity, iron chelating activity taking ascorbate, quercetin & Ethylenediamine tetraacetate as the standard correspondingly. An IC50 value was originated that methanolic concentrates of Olax scandensis more efficient in Diphenylpicrylhydrazyl radical, superoxide radical activity, Iron chelating capacity compared EA & PE concentrates. The methanolic concentrates of Olax scandens & ascorbate exhibited antioxidant potential possessing IC50 226µg/ml & 66µg/ml (DPPH). 185µg/ml & 60µg/ml (Superoxide) , 287µg/ml & 65µg/ml (iron-chelating Activity) respectively. Invitro antioxidant studies obviously show methanolic concentrates of Olax scandens have better antioxidant activity. These results indicate that aerial parts of methanolic concentrates Olax scandenscould serve as a natural antioxidant, which may be useful in preventing free radical-induced diseases.

Antinociceptive effect of extracts of Marrubium astracanicum Jacq. aerial parts.

The genus Marrubium is used for treatment of joint pain, gout, stomach-ache and colic in Iranian Traditional Medicine. Marrubium astracanicum Jacq. (M. astracanicum) is a native species in the flora of Iran. The aim of this study was to evaluate the antinociceptive properties of various extracts of aerial parts of M. astracanicum. Antinociceptive activities of total hydroalcoholic extract (THE) and its n-hexane (non-polar) and residual partition (polar) fractions were analyzed using formalin test in mice. Morphine (5 mg/kg) and normal saline were used as positive and negative controls, respectively. Intraperitoneal administration of THE (50, 100 and 200 mg/kg), non-polar fraction (200 mg/kg) and polar fraction (100 and 200 mg/kg), 30 min before formalin injection, caused significant analgesic activity in acute phase (0-5 min after formalin injection) of formalin test (p<0.05 as compared to control and p>0.05 in comparison with morphine). In chronic phase (15-60 min after formalin injection), non-polar and polar fractions (50, 100 and 200 mg/kg) showed significant analgesic activity (p<0.001 as compared to control and p>0.05 in comparison with morphine). Different extracts of M. astracanicum demonstrated antinociceptive activity that support the traditional usage of Marrubium genus for the treatment of arthritis, gout and other inflammatory diseases.

Bioactive compounds from Hypericum humifusum and Hypericum perfoliatum: inhibition potential of polyphenols with acetylcholinesterase and key enzymes linked to type-2 diabetes

Context: Natural products are reported to have a wide spectrum of pharmacological properties such as antimicrobial, anti-inflammatory and anti-cholinesterase. The genus Hypericum (Hypericaceae) is a source of a variety of molecules with different biological activities, notably hypericin and various phenolics.Objectives: The goals of the present work were the determination of total phenolic and flavonoid content, hypericin and hyperforin concentration as well as the evaluation of biological of Hypericum humifusum L. (Hhu) and Hypericum perfoliatum L. (Hper).Materials and methods: The various extracts of aerial parts were powdered, and then extracted with methanol. Antibacterial activity was performed according to minimum inhibitory concentration (MIC) and minimum bactericidal (MBC) methods against four Gram-positive bacteria, four Gram-negative bacteria and yeast.Results: The results revealed that H. humifusum, bear the highest total phenolic and flavonoid content (48–113 mg GAE/g and 8–41 mg RE/g, respectively) as well as hypericin (60–90 mg/g) and hyperforin (8–30 mg/g) concentration. Both species showed significant antioxidant activity as revealed by DPPH, FRAP, ABTS, and metal chelating assays. H. humifusum exhibited a strong acetylcholinesterase (3.86–4.57 mg GALAEs/g), α-glucosidase (0.73–2.55 mmol ACEs/g) and α-amylase (3–8 mmol ACEs/g) inhibitory activity. The extract of H. humifusum exhibited strong antibacterial activity mainly against Staphylococcus epidermidis, Staphylococus aureus, and Enterococcus faecium (MIC values ranging from 200 to 250 μg/mL). The highest antifungal activity was showed for H. perfoliatum extract (MIC value = 250 μg/mL).Conclusion: The data suggest that H. humifusum could be used as valuable new natural agents with functional properties for pharmacology industries.

Antioxidant and antibacterial activities and polyphenolic constituents of Helianthemum sessiliflorum Pers.

In this study, the various extracts of aerial parts of Helianthemum sessiliflorum Pers. were examined in vitro for possible source of antioxidants and for antibacterial activity. The antioxidant activity was performed by DPPH radical scavenging method which showed that ethyl acetate extract possessed the best antioxidant potential (IC50 = 32.75 ± 2.07 μg/mL). The significant linear correlation was realised between the values of the total phenolic/flavonoid content and antioxidant activity of plant extracts. The ethyl acetate and n-butanol extracts showed moderate antibacterial activity. In addition, the phytochemical study of n-butanol extract afforded nine known phenolic compounds (1–9). This is the first report of six of them (1, 3, 5–8) in Cistaceae family. The structural identification of the isolated compounds was achieved using several spectroscopic methods.

Antioxidant, Antimicrobial, and Free Radical Scavenging Potential of Aerial Parts ofPeriploca aphyllaandRicinus communis

Context. Many diseases are associated with oxidative stress caused by free radicals. Objective. The present study evaluated the in vitro antioxidant and antibacterial activities of various extracts of aerial parts of Periploca aphylla and Ricinus communis. Materials and Methods. In vitro antioxidant activities of the plant extract were determined by DPPH and NO scavenging method. Superoxide anion radical activity was measured by the reduction of nitro blue tetrazolium as compared with standard antioxidants. Total phenolic contents and antibacterial activities of these plants were determined by gallic acid equivalent (GAE) and serial tube dilution method, respectively. Results. Plants showed significant radical scavenging activity. The results were expressed as IC50. n-Propyl gallate and 3-t-butyl-4-hydroxyanisole were used as standards for antioxidant assay. All the extracts of both plants showed comparable IC50 to those of standards. Plants extract exhibited high phenolic contents and antibacterial activities were comparable with standard drug, Ciprofloxacin. Discussion and Conclusion. The present study provides evidence that Periploca aphylla and Ricinus communis prove to be potent natural antioxidants and could replace synthetic antioxidants. Plants can also be used against pathogenic bacterial strains.

Antioxidant and α-Amylase Inhibitory Compounds from Aerial Parts ofVarthemia iphionoidesBoiss

Various extracts of aerial parts of Varthemia (Varthemia iphionoides Boiss) were investigated for radical-scavenging activity, antioxidative activity, and porcine pancreas alpha-amylase inhibitory activity. The ethanol and water extracts showed a pronounced 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging activity, with inhibition of about 90% at a concentration of 100 microg/ml, and alpha-amylase inhibitory activity of about 70% at a concentration of 200 microg/ml by the 2-chloro-4-nitrophenyl alpha-maltotrioside (CNP-G3) degradation method. The ethanol extract was purified by column chromatography to give seven 3-methoxyflavones (1-7) and eudesmane sesquiterpene, selina-4,11(13)-dien-3-on-12-oic acid (8). The structures of these compounds were established by NMR, MS, and UV spectroscopy. Of 3-methoxyflavones, 5,7,4'-trihydroxy-3,6-dimethoxyflavone (1), 5,7,4'-trihydroxy-3,3'-dimethoxyflavone (2), and 5,4'-dihydroxy-3,7,3'-trimethoxyflavone (3,7,3'-tri-O-methyl-quercetin) (7) exhibited pronounced radical-scavenging activity. The antioxidative activity in the linoleic acid system was considerable in compounds 1, 2, and 5,4'-dihydroxy-3,6,7-trimethoxyflavone (4). Compounds 1, 2, 4, 5 (5,7,4'-trihydroxy-3-methoxyflavone), and 6 (5,4'-dihydroxy-3,7-dimethoxyflavone) showed markedly high inhibitory activity against porcine pancreas alpha-amylase. Eudesmane sesquiterpene did not show any activity.
