Enhancing rainfed safflower yield, oil content, and fatty acid composition through intercropping with chickpea and stress-modifier biostimulants

This study investigated the impact of stress modifiers in intercropping systems on seed yield and yield components, physiological traits, and antioxidant activity of safflower (Carthamus tinctorius L.) and chickpea (Cicer arietinum L.) under rainfed (water deficit) conditions. The experimental design included three stress modulator levels [control, 1 mM salicylic acid (SA), and 10 mM selenium (Se)] and five planting patterns [intercropping one row of safflower and two rows of chickpeas (1S:2C), two rows of safflower and four rows of chickpeas (2S:4C), and three rows of safflower and five rows of chickpeas (3S:5C), and sole cropping of safflower (Ss) and chickpea (Cs)]. The results revealed that Ss treated with Se produced the highest safflower biological yield (4,905.50 kg ha−1) and seed yield (1,259.50 kg ha−1), while Cs produced the highest chickpea biological yield (2,799.67 kg ha−1) and seed yield (852.44 kg ha−1), followed by Cs treated with SA (2,419.25 kg ha−1 and 764.83 kg ha−1, respectively). Conversely, the 3S:5C intercropping ratio (IR) with Se application recorded the highest safflower oil content (32.08%), while Ss treated with Se produced the highest oil yield (358.62 kg ha−1). The 2S:4C configuration with Se application produced the highest unsaturated fatty acid (oleic and linoleic acids) concentrations in safflower, while 2S:4C and 3S:5C treated with Se produced the highest chlorophyll a and chlorophyll b contents in safflower and chickpea. Furthermore, 1S:2C and 2S:4C treated with SA or Se produced the highest proline and total soluble sugars in safflower and chickpea. The SA and Se treatments in the intercropping systems increased catalase, ascorbate peroxidase, and superoxide dismutase activities compared to the respective control plants (sole cropping) and enhanced oil contents, fatty acid composition, physiological traits, and antioxidant properties. These results underscore the potential of intercropping systems coupled with stress modulator treatments as a sustainable approach for safflower and chickpea cultivation under rainfed conditions.

Effects of food waste hydrolysate as an external carbon source on defoaming in wastewater treatment with activated sludge process

The activated sludge process is the most widely used technology for treating municipal wastewater. However, thick foam often occurs in activated sludge process. Here, we reported for the first time the effect of food waste hydrolysate (FWH) as an external carbon source on defoaming in activated sludge process. The study found that FWH was effective in defoaming at a wide dose range of 50–1600 mg/L total solids, as exhibiting that the foaming tendency of FWH-added foam mixed liquor was reduced to 0 mL-foam/mL-air·min from initial 0.171 mL-foam/mL-air·min in the control without adding FWH with 100 % of defoaming efficiency. Fatty acids, oils, and solid particles in FWH jointly contributed to the deformation. Among these factors, the concentration of long-chain unsaturated fatty acids was mainly responsible for the defoaming. This work provides a cost-effective strategy to solve the foaming problem in activated sludge process as well as providing external carbon sources.

Microbial Biotechnologies to Produce Biodiesel and Biolubricants from Dairy Effluents

The shift from fossil fuels to renewable energy sources is crucial in addressing environmental challenges. Vegetable oils have been focused on as the main potential source for biodiesel and biolubricant production. However, due to their fatty acid (FA) composition they are characterized by low stability to oxidation and variable viscosity. Single-cell oils (SCOs) from oleaginous microorganisms are a possible alternative to vegetable oils: their composition is more suitable, and it can further be improved by controlling the fermentation’s physiological conditions. In the present study, the production of SCOs with targeted technological properties from Lipomyces starkeyi in fermentation under controlled temperatures was assessed. A dairy effluent (scotta) was used as the fermentation substrate to improve the economic sustainability of the process. Batch aerobic fermentations were carried out in a fermenter at two different temperatures (25 °C and 30 °C). The fermentation yields and SCO FA profiles were analyzed. The highest yields of biomass (9.76 g L−1) and microbial oil (1.83 g L−1) were obtained from fermentations carried out at 30 °C. Furthermore, a significantly lower content (46% vs. 55%) of unsaturated FAs and higher content (11% vs. 1.5%) of shorter-chain saturated FAs, with myristic acid almost matching stearic acid, were detected at 30 °C in comparison to 25 °C. Very low peroxide values were also found (0.14 meq O2 kg−1 at 30 °C and 0 meq O2 kg−1 at 25 °C). These results indicate that these SCOs were highly oxidation-resistant, and that a higher fermentation temperature improves their oxidative stability and tribophysical features. The biodiesels’ technological properties, calculated from the FA composition, were within the limits of both U.S. standards and E.U. regulations. Then, SCOs produced from L. starkeyi by fermentation of dairy effluents carried out under controlled temperature can be considered a suitable alternative to vegetable oils to produce biodiesel and biolubricants.

Production of trans-fat-free margarine prepared using catfish oil and palm stearin with the addition of avocado waste oil for tocopherol enrichment

Catfish oil (CFO) is rich in unsaturated fatty acids (UFA) which are potentially produced into margarine through the blending method with palm stearin (PS) addition. The high UFA content of CFO causes margarine to be susceptible to oxidative damage, which can also trigger the formation of trans fatty acids (TFA). An antioxidant compound in the form of avocado waste oil (AWO) that contains tocopherol is added during margarine production to inhibit oxidative damage and TFA formation in the final product. Thus, the temperature and duration of CFO extraction, avocado peel and seed ratio of AWO extraction, and CFO:PS:AWO ratio of margarine production were analyzed for their effect on CFO, AWO, and margarine characteristics. This study was conducted by several stages; CFO extraction with temperature (50, 70 and 90oC) and duration (1 and 2 hrs) treatment, AWO extraction with avocado peel and seed ratio treatment (100:0, 75:25, 50:50, 25:75, 0:100), and margarine production with CFO:PS:AWO ratio treatment (41:40:0, 36:40:5, 31:40:10, 26:40:15, 21:40:20). The results showed that the increasing extraction temperature and duration caused the decrease of CFO hydrolytic-oxidative stability. The best CFO extraction treatment was 50oC and 1 hr of extraction. The increasing avocado peel ratio in AWO extraction led to the increase of AWO antioxidant potential. The best AWO extraction treatment was the ratio of peel:seed = 75:25. The addition of AWO in margarine production for tocopherol enrichment was proven to suppress oxidative damage and TFA formation. The best margarine production treatment was the 3rd formulation (10% of AWO) with UFA-rich and trans-fat-free margarine characteristics. It was selected as the most preferred color, taste and flavor of margarine. These studies’ results are beneficial for the product development of UFA-rich and transfat-free margarine with good oxidative stability prepared using freshwater fish oil and plant byproduct oil.

In aquaculture, the use of Artemia as an optimal nutrient resource for fish fry is an object of growing interest. The advantages of using Artemia include the possibility of mass cultivation under controlled conditions, the possibility of long-term storage in frozen form and resistance to external environmental conditions. Artemia, belonging to crustaceans, is a rich source of protein, fat, vitamins and minerals necessary for the healthy development of fish larvae. Due to the high content of unsaturated fatty acids, omega-3 and omega-6, fry contribute to the development of a healthy immune system, which reduces the risk of many diseases. Their small size and mobility make them available for cultivation, which ensures a high level of quality and reduces feed consumption. The results of the study show that the use of crustaceans as the main food source for fish farming contributes to improving the life, growth and health of fry. This makes them a successful type of feed in aquaculture and an optimal resource for fish farming. Further research in this area can lead to increased efficiency of fish farming, development of practices and improvement of the overall potential of the aquaculture industry. This article discusses the advantages of using Artemia, its biological value, ease of cultivation and versatility in the nutrition of various fish larvae. Various methods and technologies of growing Artemisia are analyzed, as well as their application in breeding species of fish larvae. Special attention is paid to optimizing the maximum growth of fish larvae and feeding conditions.

Omega-3 long chain highly unsaturated fatty acid content in tissue correlates with escape response efficiency in golden gray mullet

Omega-3 long-chain highly unsaturated fatty acids (n-3 HUFA) are poorly synthesized de novo by marine fish, but are necessary to maintain their metabolic and physiological performance. Consequently, fish must find them in their diet. However, the availability of these lipid compounds is predicted to decrease in relation to global changes in ocean waters. The aim of this study was to experimentally determine the effects of an n-3 HUFA-deficient diet on the escape performance of Chelon auratus, a microphytobenthos grazer of high ecological importance. Fish were fed either a standard n-3 HUFA diet or an n-3 HUFA-deficient diet for 2 months. At the end of this conditioning period, several variables (latency, responsiveness, initial direction of escape, response duration, distance covered, maximum acceleration and velocity, and turning rate) related to escape performance were characterized in the tested fish. The results showed that a lower dietary n-3 HUFA content caused a significant reduction in the proportion of these fatty acids in fish muscle, both in membranes and in reserves. While the FA composition of the brain also reflected that of the diet in reserves, the membrane fatty acid composition of the brain remained similar between the two dietary groups, suggesting the preservation of neural and cognitive functions of the fish. Neither growth rate nor escape response variables were significantly different between the two diets. However, some escape response variables (e.g., latency, duration, maximum velocity and acceleration, and turning rate) were highly correlated with fish n-3 HUFA content in brain or muscle, and these correlations were tissue specific. While response duration and latency seemed to be more correlated with n-3 HUFA content in the brain membrane, other escape response variables such as turning rate, maximum velocity, cumulative distance, and maximum acceleration seemed to be correlated with n-3 HUFA content in the lipid reserve, both in the brain and in the muscle. Our results indicate that the reduction of n-3 HUFA in the dietary sources of marine fish appears to reduce the performance of the escape response, suggesting some impact of n-3 HUFA dietary changes with global change on predator-prey relationships within the marine ecosystem.

35 Role and mechanism of supplemental methionine in metabolic stress of broilers raised at high ambient temperature or stock density

Abstract Current poultry production practices may render fast-growing broilers prone to metabolic stresses induced by high ambient temperatures and(or) high stocking densities. As the first limiting amino acid of broilers fed corn-soy diets, methionine also serves as a free radical scavenger. Our laboratory has explored metabolic effects and biochemical mechanisms of supplemental methionine in grower and finisher diets for broilers raised at high temperature or high stock density. In Experiment 1, a total of 360 male, 1 d-old Cornish cockerel chicks were fed four diets (10 pens/treatment, 9 chicks/pen): 100 or 130% required methionine in the diets as DL-methionine (DL-MET) or 2-hydroxy-4-(methylthio)butanoate (HMTBA). The room was maintained at 4 to 13oC above the suggested thermoneutral temperature. The 130% DL-MET and HMTBA diets enhanced (P < 0.05) hepatic GSH concentrations of growers and plasma ferric reducing ability of finishers than the 100% diets. The DL-MET-fed growers had greater (P < 0.05%) muscle GSH and hepatic unsaturated fatty acid concentrations than those fed HMTBA. The methionine form and concentration exerted an interaction effect (P < 0.05) on the expression of inflammation-related genes in the liver of finishers. In Experiment 2, we determined if 30% extra DL-MET alleviated adverse effects of a greater stocking density on broilers. A total of 560 male, 1 d-old Cornish cockerels were housed at two stocking densities (9 and 12 birds/m2) and fed two concentrations of DL-MET (grower: 2.90 or 3.77 g/kg and finisher: 2.60 or 3.38 g/kg). The high stocking density decreased (P < 0.05) body weight and growth performance of growers and(or) finishers. Those differences were largely removed or attenuated by the 130% DL-MET diet. There were interaction effects (P < 0.05) between dietary DL-MET concentrations and stocking densities on activities of glutathione S-transferase in the liver of growers and the breast, thigh, and adipose tissue of finishers as well as on activities of glutathione peroxidase and superoxide dismutase in the thigh of growers. The 130% DL-MET diet decreased (P < 0.05) hepatic gene expression of heat shock protein 90 (18%) and malondialdehyde concentrations in the thigh and breast of finishers (35%). In summary, the high ambient temperature-induced metabolic responses of broilers were mediated by the extra methionine supplementation. That function varied with the chicken age and(or) tissue and the methionine form. Meanwhile, the 30% extra DL-MET addition partially reversed adverse effects caused by the greater stocking density, partially via improving tissue redox status. Microencapsulation of fish oil by spray drying, spray freeze-drying, freeze-drying, and microwave freeze-drying: Microcapsule characterization and storage stability.

The objective of this paper was to evaluate the effect of spray drying (SD), spray freeze-drying (SFD), freeze-drying (FD), and microwave freeze-drying (MFD) on the characteristics of fish oil (FO) microcapsules. The physicochemical properties, morphology, fatty acid composition, and stability of the microcapsules were analyzed. The encapsulation efficiencies of microcapsules dried by SD, SFD, FD, and MFD were 86.98%, 77.79%, 63.29%, and 57.89%, respectively. SD microcapsules exhibited superior properties in terms of effective loading capacity, color, and flowability. Conversely, SFD microcapsules demonstrated improved solubility. Microencapsulation positively affected the thermal stability of FO, but the content of unsaturated fatty acids decreased. The findings from the storage experiment indicated that the oxidative stability of SD fish oil microcapsules was marginally lower compared to microcapsules produced through three alternative drying techniques, all of which were based on the FD concept. The comparison of various drying methods and their effects on the quality of FO microcapsules offers valuable insights that can serve as a foundation for the industrial production of high-quality microcapsules.

Plasma activated water (PAW), Ozonated water (OW) and Slightly acidic electrolytic water (SAEW) treated tilapia fillets during chilled storage: A comparative study on quality changes and related mechanisms

Changes in protein degradation and lipid oxidation in tilapia fillets treated with plasma-activated water (PAW), ozonated water (OW), and slightly acidic electrolyzed water (SAEW) were investigated during refrigeration (12 d) at 4 °C. Compared to the control group, PAW, OW, and SAEW effectively delayed the formation of trichloroacetic acid (TCA)-soluble peptides and free amino acids, and significantly reduced the drip loss rate by 1.52–3.60% after 12 d (P < 0.05). However, the total content of unsaturated fatty acids in all treatment groups decreased after 12 d. Compared to PAW and OW, SAEW better-maintained protein secondary and tertiary structure stability and preserved the most polyunsaturated fatty acids (27.32%). UHPLC-Q-TOF/MS analysis showed that accumulation of dipeptides/tripeptides (such as Trp-Trp-Pro, Tyr-Thr-Trp, Leu-Gln-Phe) enhanced the bioavailability and potential bioactivity of the proteins in all treatment groups. The 4-hydroxy-octanoic acid and 4-amino-pentanoic acid from fatty acid oxidation were significantly (P < 0.05) down-regulated in PAW and SAEW. Overall, PAW, OW, and SAEW may be feasible techniques for improving the quality of tilapia fillets during cold storage.

Effect of feeding Moringa oleifera based complete pellet feed on milk yield, composition, fatty acids, somatic cell count and cell mediated immunityin lactating goats

Lactation cum feeding trial was conducted on 12 female Barbari goats (Age approx. 2-3 years and mean body weight 34.72 ±1.28 kg) for 120 days after approximately one month of kidding and effect on milk production, milk constituents, somatic cell count, milk fatty acid profile and cell mediated immunity was studied. Animals were divided into two groups (Gr G and Gr M) of six each as per completely randomized design. Two types of complete pellet (Moringa oleifera and Cicer arietinum) based was formulated containing sundried Moringa biomass and Gram straw along with conventional feed ingredients. Lactating goats of Gr G was fed with gram straw based complete pellet feed while goats of Gr M was fed with Moringa based complete pellet feed. Both the complete pellet feeds were iso nitrogenous. No significant difference in body weight changes was recorded between groups. The milk production (g/day) was higher (Avg. 14.54%) in Gr M as compared to Gr G at different fortnight. No significant difference was reported in the milk composition. Milk fat, SNF, Protein and lactose was similar in both the groups. Milk Somatic cell count (×10 3/ml) was significantly (P&lt;0.05) lower in lactating goats fed with Moringa based feed. Cell-mediated immune response assessed through delayed-type hypersensitivity (DTH) reaction against phyto haemagglutinin (PHA-P) was more pronounced in Gr M at 72 and 96 hrs.Higher concentration of unsaturated fatty acids (about 16.19%) and lower saturated fatty acids (about 5.49%) was reported in Gr M. Milk fatty acid profile indicators like Index of atherogenicity (IA) was lower while hypo/hyper cholesteremic ratio and health promoting index was higher in milk of moringa based pellet fed goats. Present study concluded that Barbari goats fed with Moringa based complete pellet feed produced higher milk yield with healthier fatty acid profile with low somatic cell counts.

Study on Ways to Improve the Quality of Black Goat Meat Jerky and Reduce Goaty Flavor through Various Spices.

In this study, we analyzed the physicochemical and sensory properties of black goat jerky marinated with various spices (non-spice, control; rosemary, RO; basil, BA; ginger, GI; turmeric, TU; and garlic, GA). The physicochemical properties of black goat jerky analyzed were pH, water holding capacity, color, cooking yield, shear force, and fatty acid composition. The sensory characteristics were analyzed through the aroma profile (electronic nose), taste profile (electronic tongue), and sensory evaluation. The pH and water holding capacity of the GI showed higher values than the other samples. GI and GA showed similar values of CIE L* and CIE a* to that of the control. The shear force of the GI and TU was significantly lower than that of other samples (p<0.05). Regarding fatty acid composition, GI showed high unsaturated and low saturated fatty acid contents compared with that of the other samples except for RO (p<0.05). In the aroma profile, the peak area of hexanal, which is responsible for a faintly rancid odor, was lower in all treatment groups than in the control. In the taste profile, the umami of spice samples was higher than that of the control, and among the samples, GI had the highest score. In the sensory evaluation, the GI sample showed significantly higher scores than the control in terms of flavor, aroma, goaty flavor, and overall acceptability (p<0.05). Therefore, marinating black goat jerky with ginger powder enhanced the overall flavor and reduced the goat odor.

Impaired Long-Chain Fatty Acid Metabolism in Carnitine Palmitoyltransferase II Knockout Mouse Leads to Chronic Pancreatitis

Background: To support high rates of protein synthesis, pancreatic acinar cells must generate large quantities of ATP. Mitochondrial dysfunction and impaired energy metabolism play a significant role in the development of pancreatitis. Patients with elevated plasma triglycerides are at increased risk for development of severe acute pancreatitis. Carnitine palmitoyltransferase II (CPT2) is required for importing and thus metabolizing long-chain fatty acids (LCFAs) in mitochondria. To inhibit LCFA metabolism, we utilized the CPT2acinarKO mouse to probe the consequences of impaired LCFA import on pancreatic function. Hypothesis: Loss of CPT2 causes an accumulation of long-chain acylcarnitines and disruption of the tissue acylcarnitine/free carnitine ratio which is thought to alter cellular function. Given acinar cells’ high metabolic requirement, impaired LCFA utilization would reduce energy production, secretory function, and alter pancreatic lipid composition. Methods: We generated mice lacking CPT2 specifically in acinar cells (CPT2acinarKO) by crossing tamoxifen-inducible, Elastase-Cre mice with CPT2 floxed mice. Mice were fed a high fat (60% kcal fat), low fat (10% kcal fat), or chow (14% kcal fat) diet for 12 weeks. Analysis of fat accumulation, edema, and fibrosis was performed with H&amp;E and picrosirius staining. Mass Spec. lipid profiling was performed in pancreas and serum. Acinar cells were isolated from wildtype, CPT2 floxed, and CPT2acinarKO mice to assess secretory function, ATP levels, and necrosis via lactate dehydrogenase (LDH) release. Results: Histologic examination revealed CPT2acinarKO mice on all three diets displayed pancreatic fat accumulation. Strikingly, CPT2acinarKO mice fed a chow or low fat diet developed chronic pancreatitis with a 25% loss in pancreatic weight, immune cell infiltration, edema, and fibrosis. As anticipated, long-chain acylcarnitines were significantly elevated in CPT2acinarKO pancreas. Assessment of secretion in 2 weeks post tamoxifen cells revealed significantly reduced maximal (10 pM) and supramaximal (1 nM) cholecystokinin (CCK) stimulated amylase release as a percent of total cellular amylase. Acini isolated at 4 weeks post tamoxifen revealed significantly increased basal secretion and reduced maximal and supramaximal CCK stimulated amylase release. When normalized to cellular DNA, secretion was similar between CPT2acinarKO and CPT2 floxed acinar cells indicating CPT2acinarKO cells had reduced amylase levels. Wildtype acinar cells treated with unsaturated and saturated free fatty acid concentrations as low as 100 μM for 3 hours caused significant necrosis. CPT2acinarKO pancreas had significantly decreased ATP levels indicative of mitochondrial dysfunction. Conclusion: Loss of CPT2 in the pancreatic acinar cells impairs LCFA import, reduces ATP levels, disrupts the secretory pathway, promoting chronic pancreatitis. In contrast to other CPT2 knockout models, pancreatitis occurs independent of high fat feeding in CPT2acinarKO mice. Further investigation is needed to identify metabolic alterations leading to acinar damage and mitochondrial dysfunction following the loss of CPT2. Insights into the effects of saline forage on the meat quality of Tibetan sheep by metabolome and multivariate analysis

This work aimed to investigate how two different types of forage (saline and alkaline) impact the meat quality and muscle metabolism of Tibetan sheep. An integrative multi-omics analysis of meat quality and different metabolites was performed using untargeted and targeted metabolomics approaches. The research results indicated that GG grass (saline and alkaline forage) possessed superior characteristics in terms of apparent quality and secondary metabolite content compared with HG grass (Non saline alkali forage), regardless of the targeted metabolites or non-targeted ones. Simultaneously, under stress conditions, the carbohydrates-rich salt-alkali grass play a significant role in slowing down the decline in pH, increasing the unsaturated fatty acid content and reducing the thawing loss in Tibetan sheep. This study provides an understanding of the impact of different salt-alkali grass on the quality of Tibetan sheep meat, while providing a scientific basis for the future development of salt-alkali livestock industry.

Influence of autonomic nervous system tone on the content of unsaturated fatty acids in blood lipids in goats

The relevance of this study lies in the need to determine the role of the autonomic nervous system in the metabolism of unsaturated fatty acids, especially essential ones, in the body of goats, which will improve the understanding of individual metabolic characteristics in these animals. The use of established blood markers will make it possible to analyse the state of lipid metabolism in the body of goats and ensure an increase in the effectiveness of methods of its correction. The purpose of this study was to determine the specific features of the autonomic nervous system influence on the relative content of unsaturated fatty acids in goat plasma lipids. The experiment involved goats of the Zaanenska breed, of which, using electrocardiographic analysis according to the Baevsky method and depending on the tone of the autonomic nervous system, three experimental groups were formed: normotonics, sympathotonics, and vagotonics. The method of gas-liquid chromatography was used to determine the percentage of unsaturated fatty acids in goat plasma lipids. Thus, in goats with sympathotonics, which have a predominance of sympathetic nervous system activity, a high percentage of the following lipids in blood plasma was noted: linolenic (P &lt; 0.01), cis-4, 7, 10, 13, 16, 19-docosahexaenoic (P &lt; 0.05), docosahexapentaenoic (P &lt; 0.001), and arachidonic acids (P &lt; 0.001) compared to normotonics. These animals also had a lower content of palmitoleic (P &lt; 0.05) and oleic acids (P &lt; 0.01) in plasma lipids against the background of normotonics. In vagotonic goats, in which the influence of the parasympathetic nervous system predominates, the relative content of cis-4, 7, 10, 13, 16, 19-docosahexaenoic (P &lt; 0.05), linoleic (P &lt; 0.001) and arachidonic acids (P &lt; 0.001) and high cis11-eicosenoic acid (P &lt; 0.01) in blood plasma lipids was found to be low compared to normotonic goats. The established features suggest a substantial influence of the tone of the autonomic nervous system on the quantitative redistribution of unsaturated fatty acids in the composition of goat blood plasma lipids and the need to consider them when analysing the state of the corresponding metabolism in the body, which will substantially improve not only the effectiveness of scientific research, but also its reliability

A telomere-to-telomere gap-free reference genome assembly of avocado provides useful resources for identifying genes related to fatty acid biosynthesis and disease resistance.

Avocado (Persea americana Mill.) is an economically valuable plant because of the high fatty acid content and unique flavor of its fruits. Its fatty acid content, especially the relatively high unsaturated fatty acid content, provides significant health benefits. We herein present a telomere-to-telomere gapless genome assembly (841.6Mb) of West Indian avocado. The genome contains 40 629 predicted protein-coding genes. Repeat sequences account for 57.9% of the genome. Notably, all telomeres, centromeres, and a nucleolar organizing region are included in this genome. Fragments from these three regions were observed via fluorescence in situ hybridization. We identified 376 potential disease resistance-related nucleotide-binding leucine-rich repeat genes. These genes, which are typically clustered on chromosomes, may be derived from gene duplication events. Five NLR genes (Pa11g0262, Pa02g4855, Pa07g3139, Pa07g0383, and Pa02g3196) were highly expressed in leaves, stems, and fruits, indicating they may be involved in avocado disease responses in multiple tissues. We also identified 128 genes associated with fatty acid biosynthesis and analyzed their expression patterns in leaves, stems, and fruits. Pa02g0113, which encodes one of 11 stearoyl-acyl carrier protein desaturases mediating C18 unsaturated fatty acid synthesis, was more highly expressed in the leaves than in the stems and fruits. These findings provide valuable insights that enhance our understanding of fatty acid biosynthesis in avocado.

The Antioxidant, Anti-Inflammatory and Moisturizing Effects of Camellia oleifera Oil and Its Potential Applications.

Camellia oleifera oil (CO oil) extracted from C. oleifera seeds has a 2300-year consumption history in China. However, there is relatively little research regarding its non-edible uses. This study determined the physicochemical properties of CO oil extracted via direct pressing, identified its main components using GC-MS, and evaluated its antioxidant, moisturizing, and anti-inflammatory activities. The results revealed that CO oil's acid, peroxide, iodine, and saponification values were 1.06 ± 0.031 mg/g, 0.24 ± 0.01 g/100 g, 65.14 ± 8.22 g/100 g, and 180.41 ± 5.60 mg/g, respectively. CO oil's tocopherol, polyphenol, and squalene contents were 82.21 ± 9.07 mg/kg, 181.37 ± 3.76 mg/kg, and 53.39 ± 6.58 mg/kg, respectively; its unsaturated fatty acid (UFA) content was 87.44%, and its saturated fatty acid (SFA) content was 12.56%. CO oil also demonstrated excellent moisture retention properties, anti-inflammatory effects, and certain free radical scavenging. A highly stable CO oil emulsion with competent microbiological detection was developed using formulation optimization. Using CO oil in the emulsion significantly improved the formulation's antioxidant and moisturizing properties compared with those of the emulsion formulation that did not include CO oil. The prepared emulsion was not cytotoxic to cells and could reduce cells' NO content; therefore, it may have potential nutritional value in medicine and cosmetics.

Effects of Supplementing Tributyrin on Meat Quality Characteristics of Foreshank Muscle of Weaned Small-Tailed Han Sheep Lambs.

This experiment aimed to evaluate the effects of supplementing tributyrin (TB) on the meat quality characteristics of foreshank muscle of weaned lambs. A total of 30 healthy weaned Small-Tailed Han female lambs with body weights ranging from 23.4 to 31.6 kg were selected and randomly divided into five groups, and each group consisted of 6 lambs. The control group was fed a basic total mixed ration, while other groups were fed the same ration supplemented with 0.5, 1.0, 2.0, and 4.0 g/kg TB, respectively. The experiment lasted 75 d, including 15 d of adaptation. Foreshank muscle obtained at the same position from each lamb was used for chemical analysis and sensory evaluation. The results showed that supplementing TB increased the muscle contents of ether extract (p = 0.029), calcium (p = 0.030), phosphorus (p = 0.007), and intermuscular fat length (p = 0.022). Besides, TB increased the muscle pH (p = 0.001) and redness (p < 0.001) but reduced the lightness (p < 0.001), drip loss (p = 0.029), cooking loss (p < 0.001), shear force (p = 0.001), hardness (p < 0.001), cohesiveness (p < 0.001), springiness (p < 0.001), gumminess (p < 0.001), and chewiness (p < 0.001). In addition, TB increased the muscle content of inosine-5'-phosphate (p = 0.004). Most importantly, TB increased the muscle contents of essential amino acids (p < 0.001). Furthermore, TB increased the saturated fatty acids level in the muscle (p < 0.001) while decreasing the unsaturated fatty acids content (p < 0.001). In conclusion, supplementing TB could influence the meat quality of foreshank muscle of weaned lambs by modifying the amino acid and fatty acid levels.

Effect of three unsaturated fatty acids on the protein oxidation and structure of myofibrillar proteins from rainbow trout (Oncorhynchus mykiss)

In this study, the impact of three unsaturated fatty acids (Oleic acid: OA, Eicosapentaenoic acid: EPA, Docosahexaenoic acid: DHA) on the oxidation and structure of rainbow trout myofibrillar protein (MP) was explored. The findings revealed a notable increase in carbonyl content (P < 0.05) and a significant decrease in total sulfhydryl content (P < 0.05) of MP with the concentration increase of the three unsaturated fatty acids. Endogenous fluorescence spectroscopy and surface hydrophobicity analyses showed that unsaturated fatty acids can cause unfolding and exposure of hydrophobic groups in MP. In addition, SDS-PAGE showed that disulfide bonds were associated with MP cross-linking and aggregate size induced by unsaturated fatty acids. Overall, three unsaturated fatty acid treatments facilitated the oxidation of myofibrillar proteins, and the extent of protein oxidation was closely associated with the concentration of unsaturated fatty acids.

Integrated analysis of metabolome, transcriptome, and bioclimatic factors of Acer truncatum seeds reveals key candidate genes related to unsaturated fatty acid biosynthesis, and potentially optimal production area

BackgroundLipids found in plant seeds are essential for controlling seed dormancy, dispersal, and defenses against biotic and abiotic stress. Additionally, these lipids provide nutrition and energy and are therefore important to the human diet as edible oils. Acer truncatum, which belongs to the Aceaceae family, is widely cultivated around the world for its ornamental value. Further because its seed oil is rich in unsaturated fatty acids (UFAs)- i.e. α-linolenic acid (ALA) and nervonic acid (NA)- and because it has been validated as a new food resource in China, the importance of A. truncatum has greatly risen. However, it remains unknown how UFAs are biosynthesized during the growth season, to what extent environmental factors impact their content, and what areas are potentially optimal for their production.ResultsIn this study, transcriptome and metabolome of A. truncatum seeds at three representative developmental stages was used to find the accumulation patterns of all major FAs. Cumulatively, 966 metabolites and 87,343 unigenes were detected; the differential expressed unigenes and metabolites were compared between stages as follows: stage 1 vs. 2, stage 1 vs. 3, and stage 2 vs. 3 seeds, respectively. Moreover, 13 fatty acid desaturases (FADs) and 20 β-ketoacyl-CoA synthases (KCSs) were identified, among which the expression level of FAD3 (Cluster-7222.41455) and KCS20 (Cluster-7222.40643) were consistent with the metabolic results of ALA and NA, respectively. Upon analysis of the geographical origin-affected diversity from 17 various locations, we found significant variation in phenotypes and UFA content. Notably, in this study we found that 7 bioclimatic variables showed considerable influence on FAs contents in A. truncatum seeds oil, suggesting their significance as critical environmental parameters. Ultimately, we developed a model for potentially ecological suitable regions in China.ConclusionThis study provides a comprehensive understanding of the relationship between metabolome and transcriptome in A. truncatum at various developmental stages of seeds and a new strategy to enhance seed FA content, especially ALA and NA. This is particularly significant in meeting the increasing demands for high-quality edible oil for human consumption. The study offers a scientific basis for A. truncatum’s novel utilization as a woody vegetable oil rather than an ornamental plant, potentially expanding its cultivation worldwide.

Genetic evidence for functions of Chloroplast CA in Pyropia yezoensis: decreased CCM but increased starch accumulation

In response to the changing intertidal environment, intertidal macroalgae have evolved complicated Ci utilization mechanisms. However, our knowledge regarding the CO2 concentrating mechanism (CCM) of macroalgae is limited. Carbonic anhydrase (CA), a key component of CCM, plays essential roles in many physiological reactions in various organisms. While many genes encode CA in the Pyropia yezoensis genome, the exact function of specific CA in P. yezoensis remains elusive. To explore the particular function of chloroplast CA in intertidal macroalgae, we produced chloroplast-localized βCA1 knockdown mutants of P. yezoensis through RNA interference, and Pyβca1i mutants (hereinafter referred to as ca1i) showed a notable decrease in leaf area and overall biomass, as well as decreased soluble protein and unsaturated fatty acid content under different DIC conditions. However, ca1i mutants showed relatively higher starch content compared to the wild-type. The activity of enzymes involved in the Calvin cycle, photorespiration, Pentose-phosphate pathway, and floridean starch synthesis of P. yezoensis indicated an effective starch accumulation pathway after the interference of βCA1. All results suggest that the decreased activity of PyβCA1 impaired the CCM and development of thalli of P. yezoensis, but stimulated starch accumulation in the cytoplasm through feedback to the photorespiration pathway and pentose phosphate pathway to replenish intermediates for the Calvin cycle. This study is the first to explore the specific function of chloroplast CA in intertidal macroalgae using genomic technology. The results provide valuable insights into the adaption mechanisms of intertidal macroalgae to their environment.
