Dynamics of Nakaseomyces glabratus biofilm formation

Formation of biofilms of microorganisms, including those of Nakaseomyces glabratus, is responsible for the development of local and systemic pathologies in humans and animals. The system of gene expression coordination (quorum sensing) in the representation of signaling molecules allows regulation of the amount and composition of biofilm populations thus expanding the adaptive capacity of microorganisms. In the presence of gingivitis and odontolithiasis clinical signs in dogs, excessive growth of gram-positive yeast microorganisms is a differential sign of the decreased resistance of the digestive system mucous membranes to colonization. Examination of the densitometric and morphometric parameters revealed general patterns of biofilm formation, regardless of the source of Nakaseomyces glabratus isolates. Depending on the time of cultivation of the microorganisms, a gradual increase in the optic density absolute values was established. Intercellular communications were achieved by coaggregation of the heteromorphic structures, which formed clusters with rounded liquid-containing formations detected among them. The population immobilization of the architectonics of the mature three-dimensional biofilm, as consistent with cultivation conditions, was accompanied by the differentiation of numerous cells of different sizes and shapes depending on the stage of the cell cycle. Results of the examination of the general patterns of the heterogeneous micromycete population development are promising for expanding the boundaries of knowledge of the adaptation mechanisms of ubiquitous microorganisms to long-term in vivo and in vitro persistence. Methods for studying morphometric and densitometric indicators avoiding interfering into the natural biofilm architectonics are recommended to optimize the long-term and retrospective mycological studies, as well as to develop effective mycosis treatment and prevention regimens.

Use of echinocandin outpatient parenteral antimicrobial therapy for the treatment of infection caused by Candida spp.: utilization, outcomes and impact of a change to weekly dosing.

Outpatient parenteral antimicrobial therapy (OPAT) can deliver extended parenteral treatment of fungal infections in an ambulatory setting, whilst minimizing treatment burden and cost. The extended dosing interval of rezafungin may potentiate the benefits of OPAT. This retrospective cohort study includes all adult patients who received echinocandin therapy in a large OPAT programme between 2012 and 2022. Patient characteristics, treatment and outcomes were studied. Data were analysed to determine the effects of replacing daily dosing with weekly dosing of echinocandin. Across the study period, 11% (44/386) of all patients in our Health Service treated with ≥7 days of echinocandin were managed via OPAT. All were Candida and related 'yeast-like' species infections. Nakaseomyces glabrata (20/41; 49%) was the most common pathogen, fungaemia the most common presentation (17/41; 41%) and azole resistance the most frequent indication for echinocandin use (21/41; 51%).In total, 633 days of echinocandin were administered as OPAT. Thirteen patients (13/41; 32%) received concurrent parenteral antibacterials. Treatment success was achieved in 30/41 (73%) patients. If daily echinocandin dosing was replaced with weekly dosing, a potential 52% (633 to 326) reduction in the total number of treatments (for any therapy) delivered by the OPAT team is possible. The ongoing need for daily antibacterial administration mitigated the benefit in some of this cohort. Echinocandin therapy can be safely delivered via OPAT with outcomes equivalent to bed-based care. The extended dosing interval of rezafungin will allow for a substantial reduction in the number of treatments required across the patient cohort.

Mining for antifungal agents to inhibit biofilm formation of Candida albicans: A study on green synthesis, antibiofilm, cytotoxicity, and in silico ADME analysis of 2-amino-4H-pyran-3-carbonitrile derivatives

Candida albicans (C. albicans) biofilm infections are quite difficult to manage due to their resistance against conventional antifungal drugs. To address this issue, there is a desperate need for new therapeutic drugs. In the present study, a green and efficient protocol has been developed for the synthesis of 2-amino-4H-pyran-3-carbonitrile scaffolds 4a-i, 6a-j, and 8a-g by Knoevenagel-Michael-cyclocondensation reaction between aldehydes, malononitrile, and diverse enolizable C-H activated acidic compounds using guanidinium carbonate as a catalyst either under grinding conditions or by stirring at room temperature. This protocol is operationally simple, rapid, inexpensive, has easy workup and column-free purification. A further investigation of the synthesized compounds was conducted to examine their antifungal potential and their ability to inhibit the growth and development of biofilm-forming yeasts like fungus C. albicans. According to our findings, 4b, 4d, 4e, 6e, 6f, 6g, 6i, 8c, 8d, and 8g were found to be active and potential inhibitors for biofilm infection causing C. albicans. The inhibition of biofilm by active compounds were observed using field emission scanning electron microscopy (FESEM). Biofilm inhibiting compounds were also tested for in vitro toxicity by using 3T3-L1 cell line, and 4b, 6e, 6f, 6g, 6i, 8c, and 8d were found to be biocompatible. Furthermore, the in silico ADME descriptors revealed drug-like properties with no violation of Lipinski's rule of five. Hence, the result suggested that synthesized derivatives could serve as a useful aid in the development of novel antifungal compounds for the treatment of fungal infections and virulence in C. albicans.

A Review on Nanosponges: An Idiosyncratic Approach for Delivery of Proactive Molecules

Background: A lot of work has been done by many researchers to develop a system that is targeted at a specific site. Nanosponges are one of the systems that serve this purpose and have several advantages over other methods. Objective: The article’s strategy is to provide access to data regarding the nanosponges system, including its preparation, evaluation, and advantages in various fields, such as the transportation of proteins, vaccines, biocatalysts, and drugs with enhanced bioavailability. Nanosponges also contribute significantly to the treatment of breast cancer, lung cancer, fungal infection, water treatment, and topical application than other approaches. Method: Nanosponges contain a solid, porous system in which hydrophilic or lipophilic drugs are loaded at their core in the nanocavity. All the information provided in this article was collected after analysing various reports on nanosponges available on Google Scholar, PubMed, Scopus, Web of Science, and Science Direct. It is concluded that the nanosponges have excellent aqueous solubility properties. Therefore, they can be used as carriers for drugs with poor water solubility. Moreover, greater bioavailability can be achieved by using the nanosponge approach. Conclusion: Nanosponges possess various outstanding properties that form the unique system of this approach. Nanosponges provide an effective carrier system for enzymes, proteins, vaccines, and therapeutic fields. This review provides a broad overview of the development of nanosponges, their evaluation, and the uses of nanosponges based on cyclodextrin for drug delivery.

Potentializing Partially Purified Bioactive Phytochemical for the Therapeutics of Fungal Dermatophytosis

Fungal infection are the most common causes of superficial or cutaneous infections around the world and remain a major public health problem in spite of the presence of various antifungal drugs. The difficulties associated with the existing prolonged treatment of fungal infection and antifungal drugs resistance remain challenges to select an effective antifungal agent. Plants have medicinal properties due to bioactive phytochemicals that can be potentially explored for the curing of fungal skin infections. Their use as herbal formulations against fungal infection is one of the safe alternative treatments instead of synthetic drugs. Phytochemicals as secondary constituents contain alkaloid, flavonoid, saponin, phenol, and tannin are more potent with respect to their crude counterpart. The present study aims to explore two medicinal plants as Catharanthus roseus, and Tagetes erecta locally available in Krishi Vigyan Kendra (KVK) Banasthali Vidyapith using solvents as water and methanol.

Individualized regimen of Posaconazole oral suspension in Chinese HSCT patients based on population pharmacokinetic model

To establish a population pharmacokinetic (PopPK) model of posaconazole suspension in Chinese hematopoietic stem cell transplantation (HSCT) patients and to recommend an optimal dosing regimen. A single-center, retrospective, model-based study was conducted in 62 Chinese patients, including 103 with posaconazole plasma concentrations. PopPK analysis using NONMEM software. A one-compartment model of first-order elimination and absorption was in good agreement with the experimental data. Analysis of covariance showed that body weight (WT), creatinine clearance (CCR), and proton pump inhibitor (PPI) had a significant effect on the pharmacokinetics of posaconazole. The dose simulation results show that patients with CCR ≥ 90 mL/min require at least 3 mg/kg TID and 7 mg/kg BID dosing regimens for prevention and treatment, respectively. However, when combined with PPI, at least 5 mg/kg BID and 5 mg/kg TID dosing regimens are required for prevention and treatment, respectively. Regardless of whether it is used in combination with PPI or not, patients with a CCR of 60–90 mL/min can achieve PTA goals by using a 4 mg/kg BID and 4 mg/kg TID regimen for prevention and treatment, respectively. A dosing regimen of 3 mg/kg BID in patients with a CCR of 30–60 mL/min is sufficient to meet the PTA goal of prophylaxis, and the dose needs to be elevated to 4 mg/kg BID for the treatment of fungal infections, and there is no need to change the dose according to the coadministration of PPI. When the patient's CCR is less than 30 mL/min, whether or not combined with PPI, the administration regimen of 2 mg/kg BID and 3 mg/kg BID can meet the PTA goals for prevention and treatment, respectively.

Preparation and Characterization of Electrospun Ketoconazole Loaded Nanofibers as Dermal Patch

Objective: This study aims to formulate a ketoconazole patch containing electrospun nanofibres and enhance the solubility due to theincreased particle surface using the electrospinning method. Methods: The Design-Expert version13 software was used in experimental designing, to ensure an efficient nanofiber preparation process. Several nanofiber formulations were prepared with different concentrations of drug and polymers. In this study, Ketoconazole was selected as the model drug, ketoconazole is widely used as an antifungal drug in the treatment of fungal infections. Eudragit and Polyethylene glycol polymers were used with ketoconazole to formulate electrospun nanofibers. Then Ketoconazole nanofibers were investigated and evaluated chemically and morphologically by Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM). Results: Utilizing the electrospinning method, Ketoconazole electrospun nanofiber was successfully fabricated. The producedketoconazole-loaded nanofibers mat was formulated as a dermal patch. The Eudragit and polyethylene glycol polymers revealed goodcharacteristics that led to the production of uniform ketoconazole nanofibers. The prepared patch showed good physical and mechanicalproperties. The SEM results Images for the produced nanofiber mat showed good nanofiber distribution and no beads formation withseveral fiber diameter sizes, while Fourier Transform Infrared Spectroscopy findings revealed the conjugation between polymers andketoconazole pure powder, the major characteristic peaks of ketoconazole appeared in the FTIR spectrum. Formula 7 showed minimumnanofibers diameter with maximum entrapment efficiency. Conclusions: In this study, the use of the electrospinning method completed the formulation of a dermal patch containing antifungalnanofibers successfully. Using design expert software to ensure maximum optimization, two biocompatible polymers were used, Eudragit E100 and PEG 600 to formulate ketoconazole nanofibers. The formulated dermal patch could be promising for pharmaceutical antifungal applications.

Fungal Enzyme-Responsive Hydrogel Drug Delivery Platform for Triggered Antifungal Release.

Fungal infections can lead to debilitating consequences if they are not treated effectively. Antifungal drugs used to treat these infections can be toxic and overuse contributes to growing antifungal resistance. Candida spp., particularly C. albicans, are implicated in a majority of these infections. Virulent C. albicans produce secreted aspartic proteases (Saps) that aid in pathogen tissue invasion and proliferation at an infected site. Here, fungi-responsive hydrogels are developed that degrade in the presence of Saps to provide a triggered release of encapsulated liposomal antifungals. The hydrogel backbone incorporates a Sap-cleavable peptide sequence enabling Sap-responsive degradation. Hydrogels are found to effectively degrade in the presence of Saps extracted from C. albicans. Encapsulated liposomal antifungals show similar release kinetics as hydrogel degradation products in the presence of Saps, supporting a degradation-dependent release mechanism. Antifungal liposome-loaded responsive hydrogels exhibit successful eradication of C. albicans cultures and remain stable in sterile murine wound fluid. Finally, no significant cytotoxicity is observed for murine fibroblast cells and red blood cells exposed to hydrogel degradation products. These fungi-responsive hydrogels have the potential to be used for local, on-demand delivery of antifungal drugs, for effective treatment of fungal infections while helping to limit unnecessary exposure to thesetherapeutics.

Chiral Fluorescent Antifungal Azole Probes Detect Resistance, Uptake Dynamics, and Subcellular Distribution in Candida Species.

Azoles are essential for fungal infection treatment, yet the increasing resistance highlights the need for innovative diagnostic tools and strategies to revitalize this class of antifungals. We developed two enantiomers of a fluorescent antifungal azole probe (1 S and 1 R ), analyzing 60 Candida strains via live-cell microscopy. A database of azole distribution images in strains of Candida albicans, Candida glabrata, and Candida parapsilosis, among the most important pathogenic Candida species, was established and analyzed. This analysis revealed distinct populations of yeast cells based on the correlation between fluorescent probe uptake and cell diameter. Varied uptake levels and subcellular distribution patterns were observed in C. albicans, C. glabrata, and C. parapsilosis, with the latter displaying increased localization to lipid droplets. Comparison of the more potent fluorescent antifungal azole probe enantiomer 1 S with the moderately potent enantiomer 1 R highlighted time-dependent differences in the uptake profiles. The former displayed a marked elevation in uptake after approximately 150 min, indicating the time required for significant cell permeabilization to occur and its association with the azole's antifungal activity potency. Divergent uptake levels between susceptible and high efflux-based azole-resistant strains were detected, offering a rapid diagnostic approach for identifying azole resistance. This study highlights unique insights achievable through fluorescent antifungal azole probes, unraveling the complexities of azole resistance, subcellular dynamics, and uptake within fungal pathogens.

Risk Factors and Outcomes of Mucorales Infection in a Modern Cohort of Solid Organ Transplant, Hematopoietic Cell Transplant, and Chimeric Antigen Receptor T-cell Therapy Recipients

Mucorales infections continue to cause significant morbidity and mortality in immunocompromised hosts despite the advent of new approaches for diagnosis and treatment of fungal infections. We aimed to evaluate risk factors and outcomes of Mucorales infection in solid organ transplant, hematopoietic cell transplant, and chimeric antigen receptor T-cell therapy recipients. This single-center retrospective study included solid organ transplant, hematopoietic cell transplant, and chimeric antigen receptor T-cell patients with cultures positive for Mucorales. Forty-three patients were included for analysis; 34 solid organ transplant (79%) and 9 hematopoietic stem cell transplant or chimeric antigen receptor T-cell (21%). Infection with Mucorales occurred a median of 184 days after transplant. At the time of diagnosis, 36 patients were on antifungal prophylaxis with the majority receiving posaconazole (53%). Thirty-three had clinically significant disease; 30 received definitive anti-Mucorales therapy and 3 empiric antifungal therapy. Isavuconazole was the most common azole used for treatment in monotherapy recipients. All-cause mortality was 64% and, of these deaths, 18 (75%) were directly related to Mucormycosis. The highest mortality was seen in disseminated and intra-abdominal disease (100%), followed by pulmonary disease (50%). There was no significant association with mortality and transplant type or number of immunosuppressive agents. Mucormycosis is an important cause of morbidity and mortality in immunocompromised patients. Breakthrough infection was not uncommon in this study. Data regarding the incidence of infection at approximately 6 months after transplantation can inform prophylaxis and treatment regimens. The spectrum of antifungal regimens used reflects the lack of consensus on ideal regimens for these organisms and a need for more studies.

Revolutionizing treatment for topical fungal infections: evaluating penetration-enhancer-containing vesicles as a fluconazole delivery system: Ex-vivo and in-vivo dermal testing

Fungal infections pose a significant challenge in numerous developing nations and worldwide, necessitating urgent solutions. Oral administration of antifungal medications often leads to severe adverse reactions. Hence, employing topical delivery systems is preferred to ensure efficient dermal delivery of antifungal agents while minimizing side effects. Furthermore, the incorporation of penetration enhancers into nanocarriers loaded with antifungal agents has demonstrated enhanced efficacy in combating mycotic infections. Consequently, ultra-deformable penetration enhancer-containing vesicles (PEVs) were developed to explore this promising approach. In this study, Labrasol® and Transcutol® were used as penetration enhancers in formulating ultra-deformable PEVs containing the antifungal agent Fluconazole (FCZ). The PEVs underwent comprehensive characterization, including measurements of particle size (PS), charge, and entrapment efficiency (EE%). The results revealed that the size of tested PEVs ranged from 100 to 762 nm. All particles exhibited a negative charge, with a minimum zeta potential (ZP) of −38.26 mV, and an intermediate entrapment efficiency (EE%) that reached approximately 40%w/w. Ex-vivo studies demonstrated the ability of PEVs to deliver FCZ to the dermis while minimizing transdermal delivery. The selected formula was tested in-vivo using candidiasis-induced rat model and showed a superiority in its antifungal effect against Candida Albicans compared to the drug control. Stability studies were executed for the selected formula, and revealed good stability shown by the insignificant change in the PS, ZP& EE% over a six-month period.

Anti-biofilm and anti-quorum sensing activities of extract, fractions and compounds from the leaves of Cassia alata L. against yeast pathogens

BackgroundCassia alata or Senna alata, also known as “ringworm bush” because of its very effective fungicidal properties, is commonly used in African traditional medicine to treat fungal infections. Despite extensive phytochemical and pharmacological studies previously reported on C. alata, the antibiofilm activity against pathogenic yeast as well as the related anti-quorum sensing mechanism of some active constituents has not yet been elucidated. The aim of the study was to isolate the bioactive constituents from the methanol extract of the leaves of C. alata (CAExt) using antibiofilm-guided fractionation against yeast fungal pathogens and then to investigate the anti-quorum sensing activity of the active constituents by assessing their ability to inhibit violacein production in Chromobacterium violaceum. MethodsChromatographic methods were used to isolate the constituents of CAExt, and spectroscopic methods were used to elucidate the chemical structures of the isolated compounds. The broth microdilution assay was used to evaluate the antifungal activity against Candida albicans and C. parapsilosis, while crystal violet staining was used for the inhibition of biofilm formation and the disruption of preformed biofilm. The biosensor strain C. violaceum ATCC 12472 was used to investigate the anti-quorum sensing activity of the most active constituents. ResultsThe crude extract exhibited biofilm inhibition and eradication activities against the tested pathogenic yeast. The biofilm inhibition percentages ranged from 53.22 % to 75.38 %, while the biofilm eradication percentages ranged from 23.21 % to 64.25 %. The ethyl acetate fraction demonstrated high biofilm inhibition and eradication activities against the tested microorganisms. The biofilm inhibition percentages ranged from 58.19 % to 79.30 %, while the biofilm eradication percentages ranged from 34.105 % to 69.54 %. The purification of subfractions led to the identification of six compounds: stigmasterol (1), sitosterol (2), lupeol (3), emodin (4), kaempferol (5) and stigmasterol glycoside (6), two of which (4 and 5) showed potent biofilm inhibition and eradication activities. Both compounds demonstrated significantly lower MBIC50 values of 70.81 µg/mL and 65.65 µg/mL against Candida albicans and MBEC50 values of 63.65 µg/mL and 82.66, respectively, against C. albicans and C. parapsilosis. The crude extract and compounds (4) and (5) also demonstrated quorum sensing inhibitory activity, as indicated by the MQSIC value of 1024 µg/mL for the crude extract and 128 µg/mL for the two compounds. Moreover, compounds (4) and (5) displayed significant inhibitory effects on violacein production, as indicated by their low IC50 values of 28.08 µg/mL and 26.44 µg/mL, respectively. ConclusionsData obtained in this study not only support the traditional use of C. alata in the treatment of fungal infections but also reveal C. alata extract, as well as the two isolated bioactive compounds emodin (4) and kaempferol (5), as a potential source for developing antibiofilm alternative agents against biofilm-associated yeast infections. Quality by design driven development of lipid nanoparticles for cutaneous targeting: a preliminary approach.

Fungal infections are the fourth common cause of infection affecting around 50 million populations across the globe. Dermatophytes contribute to the majority of superficial fungal infections. Clotrimazole (CTZ), an imidazole derivative is widely preferred for the treatment of topical fungal infections. Conventional topical formulations enable effective penetration of CTZ into the stratum corneum, however, its low solubility results in poor dermal bioavailability, and variable drug levels limit the efficacy. The aim was to increase dermal bioavailability and sustain drug release, thereby potentially enhancing drug retention and reducing its side effects. This work evaluated the CTZ loaded solid lipid nanoparticles (SLN) consisting of precirol and polysorbate-80 developed using high pressure homogenization and optimized with QbD approach. Prior to release studies, CTZ-SLNs were characterized by different analytical techniques. The laser diffractometry and field emission scanning electron microscopy indicated that SLNs were spherical in shape with mean diameter of 450 ± 3.45nm. DSC and XRD results revealed that the drug remained molecularly dispersed in the lipid matrix. The CTZ-SLNs showed no physicochemical instability during 6months of storage at different temperatures. Further, the Carbopol with its pseudoplastic behavior showed a crucial role in forming homogenous and stable network for imbibing the CTZ-SLN dispersion for effective retention in skin. As examined, in-vitro drug release was sustained up to 24h while ex-vivo skin retention and drug permeation studies showed the highest accumulation and lowest permeation with nanogel in comparison to pure drug and Candid® cream. Further, the in-vivo antifungal efficacy of nanogel suggested once-a-day application for 10days, supported by histopathological analysis for complete eradication infection. In summary, the findings suggest, that nanogel-loaded with CTZ-SLNs has great potential for the management of fungal infections caused by Candida albicans.

AMPs are a class of naturally occurring microbicidal compounds that are recognized for their wide range of inhibitory effects and distinctive modes of operation. The utilization of AuNPs in diverse biomedical applications is facilitated through the application of nanotechnology. AMP‐AuNP nanostructures exhibit diverse features through the utilization of molecular linkage techniques. The aforementioned qualities encompass the ability to expedite the process of wound healing, facilitate precise targeting through the incorporation of DNA aptamers, serve as a means for bacterial imaging, enable the use of photothermal therapy, and contribute to the advancement of prospective treatments for fungal infections. The utilization of these synthesized antimicrobial gold nanostructures in many biomedical applications holds significant promise for advancing treatments and enhancing disease management.

A Real-World Data Observational Analysis of the Impact of Liposomal Amphotericin B on Renal Function Using Machine Learning in Critically Ill Patients.

Liposomal amphotericin B (L-AmB) has become the mainstay of treatment for severe invasive fungal infections. However, the potential for renal toxicity must be considered. To evaluate the incidence of acute kidney injury (AKI) in critically ill patients receiving L-AmB for more than 48 h. Retrospective, observational, single-center study. Clinical, demographic and laboratory variables were obtained automatically from the electronic medical record. AKI incidence was analyzed in the entire population and in patients with a "low" or "high" risk of AKI based on their creatinine levels at the outset of the study. Factors associated with the development of AKI were studied using random forest models. Finally, 67 patients with a median age of 61 (53-71) years, 67% male, a median SOFA of 4 (3-6.5) and a crude mortality of 34.3% were included. No variations in serum creatinine were observed during the observation period, except for a decrease in the high-risk subgroup. A total of 26.8% (total population), 25% (low risk) and 13% (high risk) of patients developed AKI. Norepinephrine, the SOFA score, furosemide (general model), potassium, C-reactive protein and procalcitonin (low-risk subgroup) were the variables identified by the random forest models as important contributing factors to the development of AKI other than L-AmB administration. The development of AKI is multifactorial and the administration of L-AmB appears to be safe in this group of patients.

Vidarabine as a novel antifungal agent against Candida albicans: insights on mechanism of action.

Around 1.5 million mortality cases due to fungal infection are reported annually, posing a massive threat to global health. However, the effectiveness of current antifungal therapies in the treatment of invasive fungal infections is limited. Repurposing existing antifungal drugs is an advisable alternative approach for enhancing their effectiveness. This study evaluated the antifungal efficacy of the antiviral drug vidarabine against Candida albicans ATCC 90028. Antifungal susceptibility testing was performed by microbroth dilution assay and further processed to find the minimum fungicidal concentration. Investigation on probable mode of vidarabine action against C. albicans was assessed by using the ergosterol reduction assay, reactive oxygen species (ROS) accumulation, nuclear condensation, and apoptosis assay. Results revealed that C. albicans was susceptible to vidarabine action and exhibited minimum inhibitory concentration at 150µg/ml. At a concentration of 300µg/ml, vidarabine had fungicidal activity against C. albicans. 300µg/ml vidarabine-treated C. albicans cells demonstrated 91% reduced ergosterol content. Annexin/FITC/PI assay showed that vidarabine (150µg/ml) had increased late apoptotic cells up to 31%. As per the fractional inhibitory concentration index, vidarabine had synergistic activity with fluconazole and caspofungin against this fungus. The mechanism underlying fungicidal action of vidarabine was evaluated at the intracellular level, and probably because of increased nuclear condensation, enhanced ROS generation, and cell cycle arrest. In conclusion, this data is the first to report that vidarabine has potential to be used as a repurposed antifungal agent alone or in combination with standard antifungal drugs, and could be a quick and safe addition to existing therapies for treating fungal infections.

Comparison of permeability of topical Cream drug through polymer synthetic membranes of different structures using Franz Cell diffusion test

In this study, in-vitro release experiments were performed using a Franz Diffusion Cell system for Terbinafine Hydrochloride, which is used in the treatment of various fungal infections in the skin caused by many susceptible dermatophytes. In order to evaluate TH transdermally, it was aimed to compare the permeability between polymeric membranes synthesized with polyvinyl alcohol and chitosan mixtures and commercial membranes. In the transdermal drug delivery system calculations, the Fick Diffusion Law was modified and the obtained data were compared. The study was evaluated according to the USP <1724> topical drug products in semisolid form - performance tests. A new high-performance liquid chromatography method was validated for the assay of TH released into the receptor medium in accordance with the ICH guideline. According to the results obtained during the validation process, the limit of detection value was calculated as 0.005 µg/mL and the limit of quantification value was 0.013 µg/mL. Since polymer-based synthetic membranes provide permeability according to molecular size, properties and the ideal environment for measuring and analyzing the diffusion rate of a compound, in experiments conducted using membranes of different structures, the permeability profiles of selected polymer synthetic membranes were statistically evaluated. It has been noted that the hydroxyl groups in PVA and the hydrophilic CS-g-PNDMAAm graft copolymer act at varying rates to enhance or reduce the permeability across the membrane by promoting its diffusion. NaY zeolite has been shown to function as an adsorbent against the medication, lessening the impact of hydrophilic PVA without supplying permeability.

New Benzylidene‐Triazole Tethered Coumarin Molecular Hybrids: Synthesis, SAR, Molecular Modelling Simulations Targeting Tubulin α/β Dimer and its Antimicrobial Evaluation

AbstractA series of new 7‐{3‐[3‐(Benzylidene‐amino)‐[1,2,4]triazole‐1‐yl]‐propoxy}‐4‐methyl‐chromen‐2‐one molecular hybrids 7(a–k) were synthesized using molecular hybridization approach. The synthesized new benzylidene‐triazole tethered coumarin molecular hybrids were characterized by spectroscopic studies and elemental analysis. The coumarin molecular hybrids have been explored for their antifungal efficacy in terms of tubulin inhibitors using molecular modelling studies against tubulin alphabeta hetero dimer protein. The results clearly demonstrated strong binding affinity of coumarin molecular hybrids with indole substituted and 2,4‐dihydroxy substituted as antagonists with respective binding scores of −10.5 and −10.7 kcal/mol towards tubulin alphabeta hetero dimer protein which was higher in comparison to standards griseofulvin (−8.2 kcal/mol) and fluconazole (−8.1 kcal/mol). In addition, broth dilution method was used to conduct in‐vitro antifungal studies against three fungal strains namely Candida albicans, Aspergillus niger and Aspergillus fumigatus. The results obtained clearly illustrate the antifungal activity of indole and 2,4‐dihydroxy substituted compounds which are consistent with docking simulations. Out of all the synthesized molecular hybrids, indole and 2,4‐dihydroxy substituted compounds depict outstanding antifungal activity with Minimum Inhibitory Concentration (MIC) value of 31.25 μg/ml and 62.5 μg/ml towards C.albicans and with MIC value of 31.25 for both these molecules towards A.niger in each case which is quite higher than griseofulvin and fluconazole respectively. Present work clearly demonstrates the potential efficacy of synthesized indole and 2,4‐dihydroxy substituted coumarin molecular hybrids to be considered as lead compounds for the development of new agents against resistant strains for the treatment of fungal infection.
