The accurate and non-invasive detection of dopamine (DA) in sweat is crucial for the early diagnosis of neurological diseases, but the selective and sensitive detection of DA in complex biological matrices still remains a challenge. This study presents a novel screen-printed electrode based on a ternary nanocomposite of gold nanobipyramid@copper selenide@MXene nanosheets (Au NBP@Cu2-xSe@MXene) for the highly sensitive and selective detection of DA. For the first time, Au NBPs with localized surface plasmon resonance (LSPR) in the near-infrared second window (NIR-II) are employed to enable a one-step, pretreatment-free detection strategy by leveraging the negligible background absorption of biological matrices. Under NIR-II irradiation, the Au NBP core generates an LSPR-induced photothermal effect, which can be harnessed by the Cu2-xSe shell to generate a thermoelectric field that significantly accelerates the interfacial electrocatalytic oxidation of DA. Besides, Au NBPs can generate LSPR hot carriers, which are injected into Cu2-xSe to participate in the redox process. Furthermore, the MXene substrate ensures efficient charge transport and structural stability. As a result, this ternary nanocomposite-based sensor exhibits a wide linear detection range for DA from 0.1 to 1000 μM. It achieves a detection limit of 0.107 μM under standard conditions, which is further reduced to 0.068 μM under NIR-II laser irradiation, demonstrating the effective signal amplification via photothermal-thermoelectric coupling. This work provides a robust and innovative material platform that integrates photothermal, thermoelectric, and electrochemical mechanisms, paving the way for the development of next-generation, high-performance wearable sensors for non-invasive health monitoring.

Programmed Cell Death-1 receptors on the surface of monocytes and T and B cells modulate various immune responses and are key immunotherapy targets. Immunotherapeutic monoclonal antibodies, including Nivolumab, block the activity of this receptor and are a mainstay of cancer therapy. Use of these drugs has been associated with side effects, including local inflammation and systemic fever in some cancer patients. To understand the role of physiologic (37°C), pathophysiologic (39°C) and standard (25°C) temperatures on the formation of the immune complexes between PD-1 and Nivolumab, we used Surface Plasmon Resonance and Molecular Dynamics simulations, at appropriate temperatures. The mean lives obtained from the dissociation curves of Nivolumab:PD-1 complexes at 37°C or 25°C are ~16 times and ~125 times longer, respectively, than those at 39°C (~7 s). These discrepancies are due to N-terminal loop from PD-1 moving away from Nivolumab only at the higher temperatures, as well as due to changes in the epitope:paratope interface.

Major histocompatibility complex class II (MHC II) protein triggers adaptive immune responses by presenting antigens on antigen-presenting cells (APCs) to be recognised by CD4+ T cells. Recombinant MHC II proteins are effective for the analysis of peptide-binding interaction, detection of antigen-specific T-cells and clinical testing for donor-specific anti-human leukocyte antigen (HLA) antibody for organ transplantation. However, Escherichia coli-based production is limited due to difficulty in obtaining properly folded protein complexes. In this study, we utilised the expression machinery of Nicotiana benthamiana via the Tsukuba System to produce the soluble extracellular domains of MHC II proteins. Thirteen different MHC II proteins of the three isotypes (HLA-DR, DQ and DP) conjugated with peptides (pMHC II) were successfully expressed. The HLA-DRA*01:01/DRB1*01:01 covalently linked to a class II-associated invariant chain peptide (CLIP) or influenza hemagglutinin (HA, 306-318) was further analysed as a model. The resultant proteins were detectable in soluble fractions and purified using metal affinity purification and ion exchange chromatography, yielding 4.37 and 2.81 μg/gFW, respectively. pMHC II were also biotinylated invitro using commercial BirA, enabling future construction of tetramers or multimers. The purified proteins were confirmed through enzyme-linked immunosorbent assay (ELISA) using anti-HLA antibodies. Furthermore, Surface plasmon resonance biosensor (Biacore) verified glycosylation-dependent recognition of MHC II-HA complex by cognate T-cell receptor (HA1.7 TCR), resulting in an equilibrium dissociation constant of KD = 3.62 ± 1.59 μM. Overall, this plant-based system offers a promising scalable and economical alternative for large-scale production of functional MHC II proteins for basic and clinical applications.

Tris(2-chloroethyl) phosphate (TCEP) is a ubiquitous organophosphorus flame retardant and an emerging environmental contaminant of concern; however, the specific molecular mechanisms underlying its respiratory toxicity remain poorly understood. This study aimed to systematically elucidate the respiratory toxicity mechanisms of TCEP using an integrated toxicological approach. An integrated strategy combining network toxicology, molecular docking, surface plasmon resonance (SPR), and in vitro experimental validation was adopted to systematically explore the respiratory toxicity mechanisms of TCEP in human bronchial epithelial BEAS-2B cells. Network analysis revealed that potential TCEP targets were significantly enriched in pathways associated with inflammation and oxidative stress. Molecular docking identified MMP-9, MMP-2, and PTGS2 as core targets with strong binding affinities (-5.1 to -5.4 kcal/mol), and these results were further verified by SPR assays, which confirmed the direct binding of TCEP to these targets with micromolar affinities (KD = 42.4-92.0 μM). In vitro experiments revealed that TCEP exposure triggered concentration-dependent cytotoxicity and marked oxidative stress in BEAS-2B cells, characterized by glutathione depletion, catalase inhibition, increased lipid peroxidation, and compensatory upregulation of superoxide dismutase. Additionally, TCEP significantly upregulated the protein expression levels of MMP-9, MMP-2, and PTGS2, while pretreatment with the ROS scavenger N-acetylcysteine (NAC) significantly mitigated this upregulation, indicating that oxidative stress acts as a key upstream mediator responsible for the activation of these targets. These findings collectively demonstrate that TCEP impairs respiratory epithelial homeostasis via a synergistic multi-target mechanism involving oxidative stress, inflammation, and tissue remodeling, which provides important theoretical support for environmental risk assessment and the development of targeted therapeutic interventions.
