Rapid Down‐Slope Transport of Fresh Dissolved Organic Matter to the Deep Ocean in the Eastern North Atlantic

AbstractIntense convective mixing in the central North Atlantic is a major gateway for dissolved organic matter (DOM) into the deep ocean, sustaining elevated dissolved organic carbon (DOC) concentrations. Rapid down‐slope transport on adjacent Irish and Hebrides Margins represents another, less‐explored mechanism contributing to the deep‐sea DOM reservoir. Our analyses of solid‐phase extractable DOM (SPE‐DOM) in bottom waters in this region showed 7–11 μM higher DOC concentration and 190–330 years youngerSPE‐DOM radiocarbon ages compared to similar depths in the open eastern North Atlantic. We estimated a down‐slope DOC flux of 43 Tg C yr−1 from the Irish and Hebrides shelves. During transport, conservative mixing, dominated by physical rather than biological/chemical processes, determined the molecular DOM composition, while minor particulate organic matter degradation introduced less‐refractory DOM with terrigenous characteristics. Thus, rapid down‐slope transport emerges as an efficient conduit for delivering fresh DOM into the deep ocean.

Combined Quantification and Characterization of Dissolved Organic Matter by Liquid Chromatography-Mass Spectrometry Using Charged Aerosol Detection.

Dissolved organic matter (DOM) is a complex mixture of thousands of molecular formulas comprised of an unknown number of chemical compounds, the concentration and composition of which are critical to ecosystem function and biogeochemical cycling. Despite its importance, our understanding of the DOM composition is lacking. This is principally due to its molecular complexity, which means that no single method is capable of describing DOM in its entirety. Quantification is typically done by proxy (e.g., relative to carbon content) and does not necessarily match well to compositional data, due to incomplete analytical windows and selectivity of different analytical methods. We present an integrated liquid chromatography (LC)-diode array detector (DAD)-charged aerosol detector (CAD)-mass spectrometry (MS) pipeline designed to both characterize and quantify solid-phase extractable DOM (SPE-DOM) in a single analysis. We applied this method to a set of eight Swedish water bodies sampled in the summer and winter. Chromophoric SPE-DOM was proportionally higher in samples with higher SPE-DOM concentrations but remained relatively consistent between sampling occasions. Ionizable SPE-DOM was relatively consistent across sites but was proportionally higher in summer. Overall, the carbon content of DOM was very consistently ∼40% across sites in both summer and winter. These findings suggest that SPE-DOM concentration at these sites is driven by (presumably allochthonous) chromophoric inputs, with an increased relative contribution in summer of material that is more ionizable and less chromophoric and may be either autochthonous or selectively enriched from allochthonous sources. Thus, with minimal additional effort, this method provided further compositional insights not attained by any single analysis in isolation.

Beyond Hydrology: Exploring the Factors Influencing the Seasonal Variation of the Molecular Composition of Riverine Dissolved Organic Matter

AbstractThe export of dissolved organic matter (DOM) from rivers is essential for linking terrestrial and marine carbon reservoirs in the global carbon cycle. However, there is limited knowledge regarding how the molecular composition of riverine DOM changes under different hydrological conditions, especially during extreme rainfall events. Moreover, the factors beyond hydrology that impact DOM composition have not been well defined. To address these gaps, samples were collected from a human‐impacted medium‐sized subtropical monsoonal river across various hydrological conditions throughout a complete hydrological cycle. Utilizing high‐resolution mass spectrometry, it was discovered that the solid‐phase extractable DOM (SPE‐DOM) during the high‐flow (1 < runoff (Q): annual mean runoff (Qm) < 3) and extreme‐rain (Q:Qm > 3) periods exhibited a higher number of molecular formulae, lower H/C, higher O/C, and a higher proportion of carboxylic‐rich alicyclic molecules compared to the low‐flow period (LFP) (Q:Qm < 1). These alterations were attributed to input from more diverse sources, particularly a greater input from soil organic matter with higher oxidation degrees. Additionally, the P‐containing formulae were more enriched during the extreme‐rain period, likely from agricultural lands and sediment release. Conversely, the fraction of S‐containing formulae was significantly higher during the LFP, possibly due to the amplified influence of anthropogenic input. Furthermore, the DOM aromaticity did not fluctuate with runoff but was significantly associated with temperature. In summary, the study indicated that the composition of DOM varied significantly under different hydrological conditions, with temperature and anthropogenic activities identified as crucial factors influencing riverine DOM export.

From swamp to sea: Quantifying terrestrial dissolved organic carbon in a tropical shelf sea using hydrogen isotope ratios

Dissolved organic carbon (DOC) is a key component of coastal biogeochemical cycles, but its composition and reactivity depend on the relative contribution of autochthonous aquatic versus allochthonous terrigenous DOC (tDOC). In complex coastal waters, tDOC is commonly quantified using the bulk DOC stable carbon isotope ratio (δ13CDOC). However, several limitations can hamper the use of δ13CDOC in marine ecosystems, such as (1) the narrow and often overlapping separation of the autochthonous and allochthonous endmembers, and (2) mineralization of tDOC to dissolved inorganic carbon creates a reservoir effect such that autochthonous DOC can acquire a terrigenous-like δ13CDOC. The stable isotope ratio of non-exchangeable hydrogen in the DOC (δ2Hn) has emerged as a new tool that can potentially overcome these limitations: (1) δ2Hn has a large separation between aquatic and terrigenous endmembers (>50‰) and (2) it is not subject to reservoir effects caused by tDOC mineralization. Here, we evaluate the potential of δ2Hn obtained from solid phase-extracted dissolved organic matter (SPE-DOM), by comparing it to δ13CDOC and chromophoric DOM (CDOM) optical properties. We collected samples at a site in Southeast Asia’s Sunda Shelf that experiences substantial seasonal variation in tDOC input, driven primarily by the monsoon-induced physical advection of peatland-derived tDOC. Over a 1-year monthly time series, the terrigenous fraction of DOC (fterr) determined using δ2Hn of SPE-DOM and δ13CDOC of bulk DOC was well correlated (r2 = 0.42), and there was no significant difference in fterr between the two isotope systems. In fact, δ2Hn displayed slightly stronger correlations with salinity and CDOM optical properties compared to δ13CDOC. Our results indicate that δ2Hn of SPE-DOM is effective for quantifying tDOC across coastal gradients, potentially offering greater sensitivity than δ13CDOC, and is a viable alternative in settings where δ13CDOC is inadequate.

Expanding the Limits of Structural Characterization of Marine Dissolved Organic Matter Using Nonuniform Sampling Frequency-Reversed Edited HSQC NMR.

The multiplicity-edited heteronuclear single quantum correlation (ME-HSQC) NMR method is widely used for the structural characterization of marine dissolved organic matter (DOM), which is a complex molecular mixture comprising millions of individual compounds. However, the standard ME-HSQC suffers from significant signal cancellation and subsequent loss of crucial structural information due to the overlap between CH3/CH (positive) and CH2 (negative) cross-peaks in overcrowded regions. This study introduces nonuniform sampling in frequency-reversed ME-HSQC (NUS FR-ME-HSQC), highlighting its remarkable potential for the comprehensive structural characterization of marine DOM. By reversing the frequency of CH2 cross-peaks into an empty region, the FR-ME-HSQC method effectively simplifies the spectra and eliminates signal cancellation. We demonstrate that nonuniform sampling enables the acquisition of comparable spectra in half the time or significantly enhances the sensitivity in time-equivalent spectra. Comparative analysis also identifies vulnerable CH2 cross-peaks in the standard ME-HSQC that coincide with CH3 and CH cross-peaks, resulting in the loss of critical structural details. In contrast, the NUS FR-ME-HSQC retains these missing correlations, enabling in-depth characterization of marine DOM. These findings highlight the potential of NUS FR-ME-HSQC as an advanced NMR technique that effectively addresses challenges such as signal overcrowding and prolonged experimental times, enabling the thorough investigation of complex mixtures with implications in several fields, including chemistry, metabolomics, and environmental sciences. The advantages of NUS FR-ME-HSQC are experimentally demonstrated on two solid-phase-extracted DOM (SPE-DOM) samples from the surface and deep ocean. With this new technology, differences in the composition of DOM from various aquatic environments can be assigned to individual molecules.

Concealed by darkness: Combination of NMR and HRMS reveal the molecular nature of dissolved organic matter in fractured-rock groundwater and connected surface waters.

Detailed molecular composition of solid phase extracted dissolved organic matter (SPEDOM) collected from fractured-rock groundwater was compared to connected surface river water at two different watersheds in the unconfined chalk aquifer of Champagne in France using full scan ultrahigh resolution electrospray and photoionization Fourier transform ion cyclotron mass spectrometry (FT-ICR MS), Orbitrap tandem MS (MS/MS) and 1H magnetic resonance spectroscopy (NMR). 1H NMR spectroscopy indicated that groundwater SPEDOM carried a higher contribution of aliphatic compounds while surface river waters SPEDOM were enriched in carboxyl-rich alicyclic molecules (CRAM), acetate derivatives and oxygenated units. Furthermore, we show here that use of photoionization (APPI(+)) in aquifer studies is key, ionizing about eight times more compounds than ESI in surface river water samples, specifically targeting the dissolved organic nitrogen pool, accounting for more than 50% of the total molecular space, as well as a non-polar, more aromatic fraction; with little overlap with compounds detected by ESI(-) FT-ICR MS. On the other hand, groundwater SPEDOM samples did not show similar selectivity as less molecular diversity was observed in APPI compared to ESI. Mass-difference transformation networks (MDiNs) applied to ESI(-) and APPI(+) FT-ICR MS datasets provided an overview of the biogeochemical relationships within the aquifer, revealing chemical diversity and microbial/abiotic reactions. Finally, the combination of ESI(-) FT-ICR MS and detailed Orbitrap MS/MS analysis revealed a pool of polar, anthropogenic sulfur-containing surfactants in the groundwaters, likely originating from agricultural runoff. Overall, our study shows that in this aquifer, groundwater SPEDOM contains a significantly reduced pool of organic compounds compared to surface river waters, possibly related to a combination of lack of sunlight and adsorption of high O/C formulas to mineral surfaces.

Experimental Insight into the Enigmatic Persistence of Marine Refractory Dissolved Organic Matter.

More than 90% of marine dissolved organic matter (DOM) is biologically recalcitrant. This recalcitrance has been attributed to intrinsically refractory molecules or to low concentrations of molecules, but their relative contributions are a long-standing debate. Characterizing the molecular composition of marine DOM and its bioavailability is critical for understanding this uncertainty. Here, using different sorbents, DOM was solid-phase extracted from coastal, epipelagic, and deep-sea water samples for molecular characterization and was subjected to a 180-day incubation. 1H nuclear magnetic resonance spectroscopy and ultra-high-resolution mass spectrometry (UHRMS) analyses revealed that all of the DOM extracts contained refractory carboxyl-rich alicyclic molecules, accompanied with minor bio-labile components, for example, carbohydrates. Furthermore, dissolved organic carbon concentration analysis showed that a considerable fraction of the extracted DOM (86-95%) amended in the three seawater samples resisted microbial decomposition throughout the 180-day heterotrophic incubation, even when concentrated threefold. UHRMS analysis revealed that DOM composition remained mostly invariant in the 180-day deep-sea incubations. These results underlined that the dilution and intrinsic recalcitrance hypotheses are not mutually exclusive in explaining the recalcitrance of oceanic DOM, and that the intrinsically refractory DOM likely has a relatively high contribution to the solid-phase extractable DOM in the ocean.

Substantial Biogeochemical and Biomolecular Processing of Dissolved Organic Matter in an Anticyclonic Eddy in the Northern South China Sea Down to Bathypelagic Depths

Solid-phase extracted dissolved organic matter (SPE-DOM) was isolated from two depth profiles at the core and at the edge of an anticyclonic eddy (ACE) in the northern South China Sea. Non-target nuclear magnetic resonance (NMR) spectroscopy and Fourier transform ion cyclotron mass spectrometry (FTICR/MS) of SPE-DOM revealed a higher uniformity of DOM molecules within the ACE than at the edge of the ACE. Small-scale upwelling of external nutrients may have contributed to higher productivity and production of fresher DOM, with higher proportions of CHNO and CHNOS compounds and low molecular weight species at the edge of the eddy. Common SPE-DOM molecules of supposedly biological origin such as carbohydrates and olefins were most abundant in the chlorophyll maximum layer in both stations. An unusual suite of ~10 abundant and ~35 less abundant tert-butyl benzene derivatives with potential to act as endocrine disruptors within a marine food chain and ~two dozen ketones of putative bacterial origin was recognized at meso- and bathypelagic depths in single-digit micromolar concentrations, with a distinct maximum at 1000 m depth at the edge of ACE. Downwelling might bring temporary large volumes of productive marine waters into deep waters, with micromolar concentration of abundant, microbial food web-specific metabolites (e.g. 2,4-di-tert-butylphenol et al.). In our study, these eventually added up to one quarter of common background biogeochemical marine organic matter even at bathypelagic depths and beneath and are significant food and energy sources for marine biota. Mesoscale chemical heterogeneity of marine water columns might extend to larger depths than currently anticipated and may create activity hotspots influencing biota, processing of DOM, and cycling of nutrients and trace elements.

Seasonal Variations in Proton Binding Characteristics of Dissolved Organic Matter Isolated from the Southwest Baltic Sea

The physicochemical characteristics of dissolved organic matter (DOM) strongly influence its interactions with inorganic species such as protons and trace elements in natural waters. We collected water samples at Boknis Eck, a time series station in the Baltic Sea with a low exposure to freshwater inputs, to investigate how seasonal fluctuations impact the proton binding properties of the isolated DOM. We used potentiometric titrations to assess the binding properties of solid-phase extracted DOM (SPE–DOM) over a seasonal cycle. We report and critically analyze the first NICA parameters estimates of carboxylic-like and phenolic-like sites for brackish water SPE–DOM. The total amount of functional groups (QmaxH,tot) showed no seasonal fluctuations and an average value of 136 ± 5.2 mmol·mol C–1. The average proton affinity (logKH) and binding site heterogeneity (m) showed a relatively minor variability for samples obtained between April and September, when the water remained stratified. These results contribute to a better understanding of the ion binding characteristics of DOM in natural brackish waters.

Dissolved organic matter in a tropical saline-alkaline lake of the East African Rift Valley.

Saline-alkaline lakes of the East African Rift are known to have an extremely high primary production supporting a potent carbon cycle. To date, a full description of carbon pools in these lakes is still missing. More specifically, there is not detailed information on the quality of dissolved organic matter (DOM), the main carbon energy source for heterotrophs prokaryotes. We report the first exhaustive description of DOM molecular properties in the water column of a meromictic saline-alkaline lake of the East African Rift. DOM availability, fate and origin were studied either quantitatively, in terms of dissolved organic carbon (DOC) and nitrogen (DON) or qualitatively, in terms of optical properties (absorbance) and molecular characterization of solid-phase extracted DOM (SPE-DOM) through negative electrospray ionization Fourier-transform ion cyclotron resonance mass spectrometry (FT-ICR-MS). DOM availability was high (DOC ∼ 8.1 mM in surface waters) and meromixis imprinted a severe quantitative and qualitative change on DOM pool. At the surface, DOM was rich in aliphatic and moderately in aromatic molecules and thus mirroring autochthonous microbial production together with photodegradation. At the bottom changes were extreme: DOC increased up to 5 times (up to 50 mM) and, molecular signature drifted to saturated, reduced and non-aromatic DOM suggesting intense microbial activity within organic sediments. At the chemocline, DOC was retained indicating that this interface is a highly reactive layer in terms of DOM processing. These findings underline that saline-alkaline lakes of the East African Rift are carbon processing hot spots and their investigation may broaden our understanding of carbon cycling in inland waters at large.

Dissolved Organic Matter Cycling in the Coastal Upwelling System Off Central Peru During an “El Niño” Year

The Peruvian upwelling system (PUS) is among the most productive regions in the ocean, with high rates of primary production and an intense oxygen minimum zone (OMZ). The main perturbation of this system is associated to “El Nino” (EN), which affects water mass distribution and reduces primary production. Previous studies in the PUS provided first insights into the dynamics of dissolved organic matter (DOM), but high-resolution studies involving the molecular characterization of the DOM pool to reveal the processes that affect the carbon cycle in this highly productive system are lacking. We characterized the molecular composition of solid-phase extractable DOM (SPE-DOM) in the coastal upwelling system off Central Peru and linked it to specific processes that affect DOM cycling. Seasonal sampling (April, August, and December) was carried out off Central Peru (12°S) during 2015, a low productivity year marked by EN conditions. The DOM molecular composition was obtained via Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR-MS). Solid-phase extractable dissolved organic carbon (SPE-DOC) concentrations showed significant differences (p < 0.05) between the water masses present off central Peru. In order to explore if changes in SPE-DOC concentrations were the result of water mass mixing, we applied a conservative mixing (CM) model. The model revealed a non-conservative behavior of SPE-DOC and allowed us to identify two distinct groups of samples with increased and decreased SPE-DOC concentrations, respectively, and one group of samples inside the CM range. Differences in environmental parameters characterizing these groups were in accordance with respective processes associated to production and degradation of SPE-DOC. The trends observed for molecular parameters revealed the imprint of processes related to DOM production and DOM degradation, both biotic (microbial degradation) and abiotic (photodegradation). Our study suggests that even under low productivity conditions like EN, there is an active cycling of the DOM pool off central Peru.

Lignin Dimers as Potential Markers for 14C-young Terrestrial Dissolved Organic Matter in the Critical Zone

The origin, molecular composition and fate of dissolved organic matter (DOM) provides essential information that links surface and subsurface processes and explores the functioning of the Critical Zone. Therefore it is important to identify specific marker compounds that provide information on the temporal and spatial linkages in the Critical Zone. Here we used ultra-high resolution mass spectrometry and accelerator mass spectrometry in order to identify markers for fast transport of surface-derived DOM through the Critical Zone. We assessed the molecular composition and radiocarbon age of solid phase extracted DOM (SPE-DOM) from forest top soils. The 14C ages of SPE-DOM in our study were similar to that of bulk DOM. Calibrated ages ranged from 0 to 49 years and the within-site variability was larger than between sampling sites. Spearman rank correlation between the 14C ages and the DOM composition identified 129 sum formulae that were significantly correlated to 14C age. We found that molecular entities with younger 14C ages had lower molecular weight, higher unsaturation and less oxygen and heteroatoms than those associated with older 14C ages. A chemical library search suggested that phenylpropanoids and compounds, which are known to be lignin derived, are key molecular species for terrestrial DOM with young 14C ages. Among them, lignin dimers emerged as prominent surface-derived compounds that can potentially be used as markers for fast transport of water and DOM into the subsurface and groundwater.

Stoichiometry, polarity, and organometallics in solid-phase extracted dissolved organic matter of the Elbe-Weser estuary.

Dissolved organic matter (DOM) is ubiquitous in natural waters and plays a central role in the biogeochemistry in riverine, estuarine and marine environments. This study quantifies and characterizes solid-phase extractable DOM and trace element complexation at different salinities in the Weser and Elbe River, northern Germany, and the North Sea. Dissolved organic carbon (DOC), total dissolved nitrogen (TDN), Co and Cu concentrations were analyzed in original water samples. Solid-phase extracted (SPE) water samples were analyzed for DOC (DOCSPE), dissolved organic nitrogen (DONSPE), sulfur (DOSSPE) and trace metal (51V, 52Cr, 59Co, 60Ni, 63Cu, 75As) concentrations. Additionally, different pre-treatment conditions (acidification vs. non-acidification prior to SPE) were tested. In agreement with previous studies, acidification led to generally higher recoveries for DOM and trace metals. Overall, higher DOM and trace metal concentrations and subsequently higher complexation of trace metals with carbon and sulfur-containing organic complexes were found in riverine compared to marine samples. With increasing salinity, the concentrations of DOM decreased due to estuarine mixing. However, the slightly lower relative decrease of both, DOCSPE and DONSPE (~77%) compared to DOSSPE (~86%) suggests slightly faster removal processes for DOSSPE. A similar distribution of trace metal and carbon and sulfur containing DOM concentrations with salinity indicates complexation of trace metals with organic ligands. This is further supported by an increase in Co and Cu concentration after oxidation of organic complexes by UV treatment. Additionally, the complexation of metals with organic ligands (analyzed by comparing metal/DOCSPE and metal/DOSSPE ratios) decreased in the order Cu > As > Ni > Cr > Co and thus followed the Irving-Williams order. Differences in riverine and marine trace metal containing DOMSPE are summarized by their average molar ratios of (C107N4P0.013S1)1000V0.05Cr0.33Co0.19Ni0.39Cu3.41As0.47 in the riverine endmember and (C163N7P0.055S1)1000V0.05Cr0.47Co0.16Ni0.07Cu4.05As0.58 in the marine endmember.

Environmental Drivers of Dissolved Organic Matter Molecular Composition in the Delaware Estuary

Estuaries as connectors of freshwater and marine aquatic systems are hotspots of biogeochemical element cycling. In one of the best studied temperate estuaries, the Delaware Estuary (USA), we investigated the variability of dissolved organic matter (DOM) over five sampling cruises along the salinity gradient in August and November of 3 consecutive years. Dissolved organic carbon (DOC) concentrations were more variable in the upper reaches of the estuary (245±49 µmol L-1) than at the mouth of the estuary (129±14 µmol L-1). Bulk DOC decreased conservatively along the transect in November but was non-conservative with increased DOC concentrations mid-estuary in August. Detailed analysis of the solid-phase extractable DOM pool via ultrahigh resolution mass spectrometry (Fourier-transform ion cyclotron resonance mass spectrometry, FT-ICR-MS) revealed compositional differences at the molecular level that were not reflected in changes in concentration. Besides the mixing of terrestrial and marine endmember signatures, river discharge levels and biological activity were found to impact DOM molecular composition. DOM composition changed less between August and November than along the salinity gradient. Relative contributions of presumed photolabile DOM compounds did not reveal non-conservative behavior indicative of photochemical processing; suggesting that on the timescales of estuarine mixing photochemical removal of molecules plays a minor role in the turbid Delaware Bay. Overall, a large portion of molecular formulae overlapped between sampling campaigns and persisted during estuarine passage. Extending the analysis to the structural level via the fragmentation of molecular masses in the FT-ICR-MS cell, we found that the relative abundance of isomers along the salinity gradient did not change, indicating a high structural similarity of aquatic DOM independent of the origin. These results point towards a recalcitrant character of the DOM supplied by the Delaware River. We demonstrate that in addition to bulk DOC quantification, detailed information on molecular composition is essential for constraining sources of DOM and to identify the processes that impact estuarine DOM, thereby controlling amount and composition of DOM eventually discharged to the ocean through estuaries.

Bioavailability and molecular composition of dissolved organic matter from a diffuse hydrothermal system

Fluids from the diffusive hydrothermal vent Woody located at the Menez Gwen hydrothermal field were collected directly from a fissure using a Kiel pumping system (KIPS). The fluids were incubated in a biodegradation experiment with the resident microbial community at in situ temperature (55°C). In addition to the initial fluid (T0), sub-samples were collected at three time points: 12.5h (T1), 38h (T2) and 180h (Tfinal). Whole water samples were characterized by spectrophotometry and for dissolved organic carbon (DOC) and total dissolved nitrogen (TDN) concentrations. Solid-phase extracted dissolved organic matter (SPE-DOM) was molecularly characterized using a 15Tesla Fourier-transform ion cyclotron resonance mass spectrometer (FT-ICR-MS). Based on Mg2+ concentrations, vent fluids were diluted ca. 1:99% with seawater. SPE-DOC concentrations remained constantly low (30μM) over the course of incubation, while DOC and TDN (1180μM and 16μM, respectively, at T0) decreased sharply (to 80μM and 9μM, respectively, at Tfinal). This biolabile DOC is likely composed of volatiles and short-chain hydrocarbons, and the reason behind their extraordinarily high concentration remains unknown. Molecular characterization of SPE-DOM in fluids and seawater identified over 3000 molecular formulae, which were separated into different groups according to their reactivity. The half-life of the more reactive groups ranged between 6–7h, 18–21h and 5–106days, and represented 7.3, 2.0 and 7.5% of the total formulae present in T0, while the majority (83%) did not exhibit any significant trend during the experiment. Despite the small molecular changes in SPE-DOM, a decrease in the number of formulae and average molecular mass was observed, the latter consistent with an increase in the spectral slope over the wavelength range 275–295nm (S275–295) during the experiment. An increase in aromatic molecular formulae during the incubation was also corroborated by a concurrent increase in the absorption coefficient at 254nm normalized to DOC concentrations (SUVA254), an optical indicator of aromatic content. Thus both FT-ICR-MS and optical properties over the course of the incubation corroborated a decrease in molecular mass and increase in aromaticity. This study is the first to report the optical and molecular transformations of bio-labile DOM from diffusive hydrothermal systems.

Novel insights into the molecular structure of non-volatile marine dissolved organic sulfur

Abiotic sulfurization likely contributes to the preservation of organic matter in fossil deposits. It is unknown whether this mechanism also stabilizes dissolved organic matter (DOM) in the oceanic water column. Knowledge of the structure of the sulfur-containing compounds in DOM could help to understand the observed stability of at least a fraction of refractory DOM in the ocean. Structural analysis of dissolved organic sulfur (DOS) is complicated by it's great molecular diversity and the low concentration of each compound. Two contrasting marine DOM samples, one deep-sea sample from the North Pacific Ocean and one surface sample from the marginal North Sea (Germany), were examined in this study. Selective alteration experiments targeting different sulfur-containing functional groups were applied prior to Fourier-transform ion cyclotron resonance mass spectrometry (FT-ICR-MS). All treatments were also carried out with corresponding model compounds, in order to test for the applicability of the chosen reactions and for matrix effects, and to confirm the types of changes caused by the treatments. Alteration experiments were composed of harsh hydrolysis, selective derivatization of thiols, oxidation, and deoxygenation. None of these treatments induced detectable changes to DOS, indicating the absence of thioesters, sulfonic acid esters, alkylsulfates, thiols, non-aromatic thioethers, and sulfoxides in solid-phase-extractable DOM (SPE-DOM). Collision-induced fragmentation of isolated nominal masses in the FT-ICR-MS confirmed these findings and revealed the presence of sulfonic acids in both samples. Additionally, the presence of thiophenes and possibly also sulfones is likely, while the presence of any other form of sulfur in SPE-DOM can be ruled out based on our results. In conclusion, only unreactive (fully oxidized and hydrolyzed) sulfur compounds resist in the water column as part of refractory DOM. There was no detectable difference in molecular structure between DOS of the marginal North Sea and the deep North Pacific, indicating that the chemical inertness and the presence of sulfonic acids are global features of DOS residing in the oxic water column.

Molecular evidence for rapid dissolved organic matter turnover in Arctic fjords

Dissolved organic matter (DOM) in the ocean comprises one of the largest active carbon pools on earth. Deep water formation at high latitudes carries DOM from the active surface layers to the deep ocean. However, information on sources and fate of DOM in the Arctic Ocean is limited. To reveal the relevance of autochthonous DOM production and transformation in Arctic fjord systems to the global deep ocean DOM pool, we performed a comprehensive study on the molecular composition of DOM and the composition of the associated microbial communities in four selected fjords of Svalbard. At various water depths, a total of 34 samples were taken in fall 2010 for the determination of bulk concentrations of dissolved organic carbon (DOC) and total dissolved nitrogen (TDN), for the molecular characterization of solid-phase extractable DOM as well as for microbial community fingerprinting.While TDN concentration and the composition of the microbial community showed a clear distinction between surface and bottom water samples, bulk DOC (~60μmol C L−1) and dissolved black carbon (~1.8% of DOC) as a marker for terrestrial input were uniformly distributed. In-depth molecular-level analyses of the DOM composition using ultrahigh resolution mass spectrometry via Fourier-transform ion cyclotron resonance mass spectrometry (FT-ICR-MS) revealed insignificant variation of the relative abundance of 11630 molecular masses that were detected in the water samples.From these findings we conclude that DOM produced during the spring/summer bloom is rapidly transformed within the short, but productive warm season by the specialized resident microbial community. Thus, in fall the DOM pool mainly consists of semi-refractory and refractory material, most of which has been introduced from Arctic Ocean water inflow. Assuming that our findings are representative for high latitude marine systems in general, the contribution of autochthonous seasonal DOC production in plankton bloom situations to the DOC pool in regions of deep water formation might be marginal.

Characterization of dissolved organic matter across the Subtropical Convergence off the South Island, New Zealand

Dissolved organic matter (DOM) was investigated along a gradient across the Subtropical Convergence (STC) off the South Island, New Zealand. Ultrahigh resolution electrospray ionization Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR-MS), excitation emission matrix fluorescence (EEM) spectroscopy, and molecular lignin analysis techniques were used to study this DOM. The analysis revealed a group of compounds found only in the coastal DOM samples, which were also characterized by an elevated terrestrial DOM fluorescence pattern and elevated lignin content if compared to the offshore samples. This group exhibited a high degree of carbon unsaturation, as evident from high double bond equivalence minus oxygen values (DBE-O > 9) and maximum fluorescence intensity. Sulfur-containing molecular formulae for summer DOM samples were much more abundant across the entire transect of the STC compared to winter and exhibited distinctly different hydrogen:carbon and oxygen:carbon elemental ratios, suggesting a possible correlation between biological activity and sulfur compounds. The molecular formulae common to all STC samples were compared with those calculated for DOM extracted from freshwater collected from a stream discharging into Doubtful Sound (West Coast, New Zealand). ESI-FT-ICR-MS measurements undertaken in negative electrospray ionization (ESI−) mode indicated that 30% of the molecular formulae were present in both types of DOM, while in positive ESI mode (ESI+) over 90% of the formulae were present in all samples. Hence, a significant portion of the molecular formulae assigned to the solid-phase extractable DOM pool (SPE-DOM) appear to be identical in oceanic and freshwater samples.
