Instability of purified 9-kDa porcine intestinal calcium-binding protein in the absence of calcium

Purified 9-kDa porcine intestinal calcium-binding protein (ICaBP, Calbindin D 9K) is unstable when stored at 4 °C in the absence of Ca(II). Cleavage of the polypeptide occurs producing ~5.2- and 3.7-kDa fragments. The former dimerizes giving a species which migrates on sodium dodecylsulfate-urea gels with an M r 13,700, in contrast to the observed M r 11,000 for native ICaBP. The fragmentation also results in an irreversible loss of high affinity Ca(II) binding and an alteration of the ultraviolet absorption spectrum. The presence of Ca(II) bound to ICaBP prevents fragmentation. The dimer can be isolated from aged preparations of apo-ICaBP and is stable on further storage with or without Ca(II) present. The observed molecular weights of the fragments along with the amino acid analysis and ultraviolet spectra of the dimer suggest cleavage of the polypeptide chain of ICaBP in the vicinity of residue 49.

Chromoplast-Specific Proteins in Capsicum annuum

Chromoplasts are a common differentiation state of plastids in which the photosynthetic apparatus is absent and carotenoids accumulate to high levels. As a first step toward the isolation of chromoplast-specific genes, we have examined plastids of the bell pepper, Capsicum annuum L., for the presence of chromoplast-specific proteins. Intact chromoplasts were isolated from mature fruits of C. annuum var Emerald Giant, Golden Cal Wonder, and DNAP VS-12 by differential centrifugation followed by isopycnic sedimentation in gradients of silica sols. The plastids were then fractionated into soluble and membrane components and the proteins analyzed by one- and two-dimensional gel electrophoresis using isoelectric focusing, sodium dodecyl sulfate, and sodium dodecyl sulfate-urea gels. Two polypeptides with M(r) of 35,000 and 58,000 accumulate to high levels in membrane fractions of chromoplasts of var Emerald Giant. These polypeptides are either not detectable or barely detectable in chloroplasts from immature fruits. Both polypeptides have been purified to near homogeneity. Yellow chromoplasts from var Golden Cal Wonder and red chromoplasts from var DNAP VS-12 contained the 35-kilodalton polypeptide, but not the 58-kilodalton species.

Subunit VII, the ubiquinone-binding protein, of the cytochrome b-c1 complex of yeast mitochondria is involved in electron transport at center o and faces the matrix side of the membrane.

The functional role and topographical orientation in the inner membrane of subunit VII, the ubiquinone-binding protein, of the cytochrome b-c1 complex of yeast mitochondria has been investigated. The apparent molecular weight of this subunit on sodium dodecyl sulfate-urea gels was calculated to be 15,500, while its amino acid composition was similar to that of the Q-binding proteins present in the cytochrome b-c1 complexes isolated from both beef heart and yeast mitochondria. The specific antibody obtained against subunit VII inhibited 30-47% of the ubiquinol-cytochrome c reductase activity in the isolated cytochrome b-c1 complex and in submitochondrial particles but had no effect on cytochrome c reductase activity in mitoplasts, mitochondria from which the outer membrane has been removed. Furthermore, the antibody against subunit VII strongly inhibited (74%) the reduction of cytochrome b by succinate in the presence of antimycin, an inhibitor of center i, but had no effect on cytochrome b reduction in the presence of myxothiazol, an inhibitor of center o. These results suggest that subunit VII, the Q-binding protein, is involved in electron transport at center o of the cytochrome b-c1 complex of the respiratory chain and that subunit VII is localized facing the matrix side of the inner mitochondrial membrane.

Purification and characterization of chymodenin. A hormone-like peptide from porcine duodenum.

Chymodenin, a hormone-like duodenal peptide which rapidly alters the proportions of secreted pancreatic digestive enzymes to a mixture relatively richer in chymotrypsinogen than that found in basal secretion, has been purified to homogeneity. The starting material was an acidic methanol-soluble, neutral pH-insoluble fraction of an acetic acid extract of porcine duodenum; the purification consisted of cation-exchange chromatography on SP-Sephadex and CM-Sephadex in ammonium bicarbonate gradients, and gel filtration on Sephadex G-75 in dilute acetic acid. The yield of material was followed by radioimmunoassay. Homogeneity was determined from chymodenin's behavior in disc gel electrophoresis in an acidic counter-migration-of-dye system, sodium dodecyl sulfate-urea gels, gel filtration, dansyl-Edman reaction, reversed-phase high pressure liquid chromatography, isotachophoresis, and sedimentation equilibrium ultracentrifugation. The electrophoretic mobility, the molecular weight of 9,000-10,000, and the biological activity differed from those of other gastrointestinal peptide hormones. The amino acid composition was unique. Chymodenin is the first purified hormone-like substance reported capable of altering the composition of the mixture of secreted digestive enzymes, independent of the stimulation of massive pancreatic protein output.

Phosphoenolpyruvate-sugar phosphotransferase transport system of Streptococcus mutans: purification of HPr and enzyme I and determination of their intracellular concentrations by rocket immunoelectrophoresis.

Enzyme I and HPr, the general proteins of the phosphoenolpyruvate-sugar phosphotransferase system, play a pivotal role in the control of sugar utilization in gram-negative and gram-positive bacteria. To determine whether growth conditions could modify the rate of biosynthesis of these proteins in Streptococcus mutans, we first purified to homogeneity enzyme I and HPr from S. mutans ATCC 27352. Using specific antibodies obtained against these proteins, we determined by rocket electrophoresis the intracellular levels of enzyme I and HPr in cells of S. mutans 27352 grown under various batch culture conditions and in a number of glucose-grown cells of other strains of S. mutans. HPr was purified by the procedure reported by Gauthier et al. (L. Gauthier, D. Mayrand, and C. Vadeboncoeur, J. Bacteriol. 160:755-763, 1984) and displayed a single band with a molecular weight of 6,650 when analyzed by sodium dodecyl sulfate-urea gel electrophoresis. Enzyme I was purified by DEAE-cellulose chromatography, affinity chromatography on an anti-Streptococcus salivarius column, and preparative electrophoresis. The protein migrated as a single band in native and denaturating gel electrophoresis. The subunit molecular weight of enzyme I determined by electrophoresis under denaturating conditions was 68,000. In gel filtration chromatography at 4 degrees C, the enzyme migrated as a 135,000- to 160,000-molecular-weight species, suggesting that enzyme I is a dimer. In double immunodiffusion experiments, antibodies against HPr reacted with several oral streptococci, Streptococcus lactis, Streptococcus faecium, and Lactobacillus casei, but not with Bacillus subtilis, Staphylococcus aureus, and Escherichia coli. Antibodies against enzyme I of S. mutans 27352 cross-reacted with enzyme I from all the other oral streptococci tested. No cross-reaction was observed with other gram-positive and gram-negative bacteria. The levels of enzyme I and HPr determined by rocket electrophoresis in S. mutans 27352 varied at the most by twofold, depending on the growth conditions. Glucose-grown cells of other S. mutans strains contained levels of enzyme I and HPr which were similar to those found in S. mutans 27352.

Purification and partial amino acid sequence of the chloroplast cytochrome b-559.

The hydrophobic cytochrome b-559, purified from unstacked, ethanol-washed spinach thylakoid membranes, using extraction with 2% Triton X-100 in 4 M urea and three chromatographic steps in the presence of protease inhibitors, has a dominant band on sodium dodecyl sulfate-urea gels corresponding to Mr = 10,000. The yield of this preparation is 30-50% (5-10 mg) starting with 600 mg of chlorophyll. The heme content yields a calculated molecular weight of no more than 17,500/heme, and perhaps somewhat smaller after correction for impurities. The Mr = 10,000 band is stained by the tetramethylbenzidine-H2O2 heme reagent on lithium dodecyl sulfate gels run at 0 degrees C. The Mr = 10,000 protein, further separated by high performance liquid chromatography, contains a unique NH2 terminus that is not blocked, and the amino acid sequence for the first 27 residues is NH2-Ser-Gly-Ser-Thr-Gly-Glu-Arg-Ser-Phe-Ala-Asp-Ile-Ile-Thr-Ser-Ile-Arg-Tyr-Trp -Val-Ile-X-Ser-Ile-Thr-Ile-Pro. . . COOH. Approximately 55% of the amino acids are hydrophobic, based on amino acid analysis of the Mr = 10,000 peptide, which also indicated the presence of at least one histidine. Only one cytochrome b-559 component could be identified, whose yield indicated that it arises from a single b-559 protein in chloroplasts corresponding to the in situ high potential cytochrome of the chloroplast photosystem II.

Partial purification and characterization of the multiple molecular forms of staphylococcal clotting activity (coagulase).

The clotting activity of Staphylococcus aureus strain 104 was purified 46,000-fold, but absolute purity was not achieved. Carbohydrate content of the purified material was not more than 5%. Elution of clotting activity from denaturing and nondenaturing polyacrylamide gels revealed the presence of four distinct molecular forms. Molecular weights of the forms were approximately 31,500, 34,800, 44,800, and 56,800 as determined by gel filtration in 8 M urea, by sodium dodecyl sulfate-urea polyacrylamide gel electrophoresis, and by calculation with determined values for the Stokes radius and sedimentation coefficient. Molecular weights determined on sodium dodecyl sulfate-urea gels were found to decrease as the gel concentration increased, suggesting that the amount of sodium dodecyl sulfate bound was less than normal. Estimated frictional ratios for the forms showed that they differ in shape from one another and that they are all highly asymmetrical. Each of the forms had an isoelectric point between pH 5.44 and 5.47 when focused in 6% polyacrylamide gels for 9 h; however, prolonged focusing altered the isoelectric point of the forms to within the range of pH 4.35 to 4.65. The multiple clotting forms were not artifacts of the purification procedure and did not appear to be products of the proteolytic degradation of a larger protein.

Demonstration of protease activity for epidermal growth factor-binding protein

The epidermal growth factor can be isolated from the male mouse submaxillary gland as part of a high molecular weight complex. The complex is composed of two molecules of epidermal growth factor and two molecules of epidermal growth-factor binding protein ( J.M. Taylor, W.M. Mitchell, and S. Cohen, 1974, J. Biol. Chem. 249, 3198–3203). The proteolytic activity of epidermal growth-factor binding protein was demonstrated by its self-proteolysis in moderate (3–7 m) concentrations of urea, and, its inhibition by formation of a complex with pancreatic trypsin inhibitor. This complex was characterized by its p I and by its ability to yield pancreatic trypsin inhibitor and epidermal growth factor-binding protein in sodium dodecyl sulfate-urea gel electrophoresis. The association equilibrium constant was determined to be 3.6 × 10 7 m −1 by inhibition studies of the esteropeptidase. These results, which indicate that epidermal growth factor-binding protein is capable of autodigestion and of forming a stable complex with a macromolecular inhibitor of trypsin, lend strong support to the hypothesis that epidermal growth factor-binding protein is capable of cleaving a larger precursor by its proteolytic action.

Purification of a lymphokine: Osteoclast activating factor from human tonsil lymphocytes

Osteoclast activating factor is a lymphokine produced by mitogen-stimulated human lymphocytes. The current studies describe purification to essential homogeneity of the major form of osteoclast activating factor present in supernatants of phytohemagglutinin stimulated lymphocyte cultures. Preliminary chemical and biological characterization of the purified material was carried out. The active factor is a peptide which migrates in polyacrylamide gel electrophoresis as an α-2 fraction in native gels and as a 9,000-dalton species in sodium dodecyl sulfate-urea gels. The purified fraction stimulates bone resorption in vitro at doses between 0.1 and 500 ng/ml, with half-maximal stimulation at approximately 1 ng/ml.

Purification and properties of the NAD +-dependent (type D) and O 2-dependent (type O) forms of rat liver xanthine dehydrogenase

The xanthine-oxidizing enzyme of rat liver has been purified as an NAD +-dependent dehydrogenase (type D) and as the O 2-dependent oxidase (type O). The purified D and O variants are nearly homogenous as judged by polyacrylamide discontinuous gel electrophoresis and are indistinguishable on sodium dodecyl sulfate-urea gels. The absorption spectrum of the type D enzyme is indistinguishable from that of the type O enzyme and closely resembles the spectra of xanthine-oxidizing enzymes from other sources. The types D and O enzymes have essentially the same cofactor composition. Oxidation of xanthine by type D is stimulated by NAD + with concomitant NADH formation. Type D is able to utilize NADH as well as xanthine as electron donor to various acceptors, in contrast to type O that is unable to oxidize NADH. Arsenite, cyanide and methanol completely abolish xanthine oxidation by the type D enzyme while affecting the activities with NADH to varying extents. In these respects rat liver xanthine dehydrogenase closely resembles chicken liver xanthine dehydrogenase. However, in contrast to the avian enzyme, the purified rat liver enzyme is unstable as a dehydrogenase and is gradually converted to an oxidase. This conversion is accompanied by an increase in the aerobic xanthine → cytochrome c activity. The native type D enzyme in rat liver extracts is precipitable with antibody prepared against purified type O. The K m for xanthine is not significantly different for the two forms.

Purification and characterization of an inhibitor of phospholipase A1 in Bacillus subtilis.

The protoplasts of a mutant of Bacillus subtilis 168 (B. subtilis CMK33) are osmotically fragile when compared to protoplasts of the parent organism and contain an active, membrane-associated phospholipase A1. A protein found in the parent organism specifically inhibits the phospholipase A1 (Kent, C., and Lennarz, W.J. (1972) Proc. Nat. Acad. Sci. U.S.A. 69, 2793-2797). The inhibitor exists in both a soluble and particulate form. The soluble inhibitor is not found in the cytoplasm, but rather in a "periplasmic" fraction released from the cell during incubation with lysozyme. The soluble inhibitor has been purified to homogeneity by diethylaminoethyl-cellulose and hydroxylapatite chromatography. Its molecular weight is 28,000 to 32,000 as determined by gel filtration chromatography and 36,000 to 37,000 as determined by sodium dodecyl sulfate-urea gel electrophoresis. The inhibitor appears to inactivate the membrane bound phospholipase A1 by an enzymatic process that is dependent on time and protein concentration. Binding of the inhbitor to the membrane-associated phospholipase cannot be detected. When purified inhibitor is added to cells of B. subtilis CMK33 during treatment with lysozyme, the osmotic stability of the resultant protoplasts is similar to that of protoplasts of the wild type of organism.
