Evolution Analysis of the Land use in the Municipality of Korsimoro, North-Central Burkina Faso: Predictive Modelling for 2050

In the context of the commune of Korsimoro in Burkina Faso, this study looked at the evolution of land use between 1991 and 2021, with projections to 2050. Using satellite data and geospatial processing methods, an analysis of past and projected trends in future land-use scenarios was conducted. The results revealed a significant increase in anthropogenic activities in recent decades, with a 25.69 per cent increase in land dedicated to agriculture and the expansion of urban areas. However, an optimistic outlook for 2050 suggests a regression in anthropogenic activities and an increase in natural areas, with 5.67 per cent increase for wooded savannahs and 78.37 per cent increase for shrub and grassy savannahs, respectively. These findings underscore the importance of rethinking land management policies in order to promote more sustainable development. Sustainable agricultural practices, integrated urban planning and the protection of natural ecosystems appear to be promising avenues for reconciling human needs with environmental preservation.

Prefrontal cortical activity during uneven terrain walking in younger and older adults.

Walking in complex environments increases the cognitive demand of locomotor control; however, our understanding of the neural mechanisms contributing to walking on uneven terrain is limited. We used a novel method for altering terrain unevenness on a treadmill to investigate the association between terrain unevenness and cortical activity in the prefrontal cortex, a region known to be involved in various cognitive functions. Prefrontal cortical activity was measured with functional near infrared spectroscopy while participants walked on a novel custom-made terrain treadmill surface across four different terrains: flat, low, medium, and high levels of unevenness. The assessments were conducted in younger adults, older adults with better mobility function and older adults with worse mobility function. Mobility function was assessed using the Short Physical Performance Battery. The primary hypothesis was that increasing the unevenness of the terrain would result in greater prefrontal cortical activation in all groups. Secondary hypotheses were that heightened prefrontal cortical activation would be observed in the older groups relative to the younger group, and that prefrontal cortical activation would plateau at higher levels of terrain unevenness for the older adults with worse mobility function, as predicted by the Compensation Related Utilization of Neural Circuits Hypothesis. The results revealed a significant main effect of terrain, indicating a significant increase in prefrontal cortical activation with increasing terrain unevenness during walking in all groups. A significant main effect of group revealed that prefrontal cortical activation was higher in older adults with better mobility function compared to younger adults and older adults with worse mobility function in all pooled terrains, but there was no significant difference in prefrontal cortical activation between older adults with worse mobility function and younger adults. Contrary to our hypothesis, the older group with better mobility function displayed a sustained increase in activation but the other groups did not, suggestive of neural compensation. Additional findings were that task-related increases in prefrontal cortical activation during walking were lateralized to the right hemisphere in older adults with better mobility function but were bilateral in older adults with worse mobility function and younger adults. These findings support that compared to walking on a flat surface, walking on uneven terrain surfaces increases demand on cognitive control resources as measured by prefrontal cortical activation.

PLGA nanocapsules as a delivery system for a recombinant LRP-based therapeutic.

Telomerase activity is directly affected by the laminin receptor precursor (LRP) protein, a highly conserved nonintegrin transmembrane receptor, which has been shown to have therapeutic effects in ageing, and age-related diseases. Recently, it has been found that overexpression of LRP-FLAG, by plasmid transfection, leads to a significant increase in telomerase activity in cell culture models. This may indicate that upregulation of LRP can be used to treat various age-related diseases. However, transfection is not a viable treatment strategy for patients. Therefore, we present a nanoencapsulated protein-based drug synthesised using poly(lactic-co-glycolic acid) (PLGA) nanocapsules for delivery of the 37 kDa LRP protein therapeutic. PLGA nanocapsules were synthesised using the double emulsification-solvent evaporation technique. Different purification methods, including filtration and centrifugation, were tested to ensure that the nanocapsules were within the optimal size range, and the BCA assay was used to determine encapsulation efficiency. The completed drug was tested in a HEK-293 cell culture model, to investigate the effect on cell viability, LRP protein levels and telomerase activity. A significant increase in total LRP protein levels with a concomitant increase in cell viability and telomerase activity was observed. Due to the observed increase in telomerase activity, this approach could represent a safer alternative to plasmid transfection for the treatment of age-related diseases.

Abstract PO1-26-06: The effect of localized radiotherapy in combination with chemoimmunotherapy in a humanized triple-negative breast cancer mouse model

Abstract Background: Metastatic triple-negative breast cancer (TNBC) is an aggressive sub-type of breast cancer that is challenging to treat. While the addition of immunotherapy (IT) with pembrolizumab (pembro) to chemotherapy prolongs overall survival in patients with programmed death-ligand 1 positive (PD-L1+) TNBC, few patients obtain long-term benefit. Radiotherapy (RT) can stimulate cellular damage, release of tumor antigens and promote the T cell anti-tumor response including at distant sites (abscopal response) when used with IT. The purpose of this study was to investigate the combination of RT with pembro and paclitaxel (pac) in a TNBC humanized mouse model. Methods: Human Immune System BRGS (BALB/c, Rag2-/-, IL2RγC-/-, NODSIRPa ) mice were engrafted with TNBC MDA-MB-231 cells in bilateral flanks. Treatments included: vehicle, pac 10 mg/kg intra-peritoneal (IP) weekly, pembro 15 mg/kg IP weekly, and RT 8 Gy x 3 fractions every other day the first week of treatment to one-sided tumors (contralateral tumors received no RT). Mice were treated and analyzed in the following groups: vehicle, pac + pembro, RT + pac, and RT + pac + pembro. Treatments began when tumors reached 100-300 mm3 and tumors were measured twice weekly. At the end of study, sera, lymph nodes (LNs), spleen, and tumor tissue were collected for immunohistochemistry, single cell suspensions, and flow cytometry analysis for immune cell populations. Results: All treatment arms resulted in a decrease in tumor volume when compared to vehicle with the greatest reduction in the RT + pac and RT + pac + pembro arms. Combination therapy with RT + paclitaxel (p=0.0002) and RT + pac + pembro (p=0.008) resulted in a statistically significant decrease in specific growth rate (SGR) when compared to vehicle. An abscopal effect was not observed in the non-RT tumors compared to RT-tumors in either group. RT + pembro was associated with an increase in hCD45 T cells in LNs and spleens but not in tumors. In mice that received RT, a decrease was noted in HLA-ABC class I and Intermediate Class II major histocompatibility complex (MHC). In addition, RT-tumors had reduced expression of PD-L1 and polio virus receptor (PVR) and increased expression of PD-1. The percentage of interferon gamma (IFNg+), tumor necrosis factor alpha (TNFa+) CD8+ T cells increased with radiation. This percentage was the highest in the triplet therapy with RT+ pembro + pac. Conclusion: Combination treatment with RT + immunochemotherapy resulted in a significant increase in anti-tumor activity when compared to immunochemotherapy alone, however, an abscopal effect was not observed in contralateral non-RT tumors. RT resulted in down-regulation of immune inhibitory receptors and tumor MHC. Establishment of a two-hit mouse model of environmental factor induced autism spectrum disorder

Autism spectrum disorder (ASD) is a group of developmental diseases characterized by social dysfunction and repetitive stereotype behaviors. Besides genetic mutations, environmental factors play important roles in the development of ASD. Valproic acid (VPA) is widely used for modeling environmental factor induced ASD in rodents. However, traditional VPA modeling is low-in-efficiency and the phenotypes often vary among different batches of experiments. To optimize this ASD-modeling method, we tested “two-hit” hypothesis by single or double exposure of VPA and poly:IC at the critical time points of embryonic and postnatal stage. The autistic-like behaviors of mice treated with two-hit schemes (embryonic VPA plus postnatal poly:IC, embryonic poly:IC plus postnatal VPA, embryonic VPA plus poly: IC, or postnatal VPA plus poly:IC) were compared with mice treated with traditional VPA protocol. The results showed that all single-hit and two-hit schemes produced core ASD phenotypes as VPA single treatment did. Only one group, namely, mice double-hit by VPA and poly:IC simultaneously at E12.5 showed severe impairment of social preference, social interaction and ultrasonic communication, as well as significant increase of grooming activity and anxiety-like behaviors, in comparation with mice treated with the traditional VPA protocol. These data demonstrated that embryonic two-hit of VPA and poly:IC is more efficient in producing ASD phenotypes in mice than the single-hit of VPA, indicating this two-hit scheme could be utilized for modeling environmental factors induced ASD.

Cdc42 Rho GTPase Activity Sustains Epithelial Barrier Function in the Cystic Fibrosis Intestine

Increased paracellular permeability is a common manifestation of cystic fibrosis (CF) intestinal disease in people with CF and in CF mouse models. Increased paracellular permeability has been found in CF airway epithelium cultured in the absence of inflammatory factors, suggesting an inherent defect in CF epithelial barrier function (Weiser et al., Cell Physiol Biochem. 2011;28(2):289-96). CF transmembrane conductance regulator knockout ( Cftr KO) mouse intestinal crypts demonstrate an increase in epithelial cell proliferation, which correlates with an alkaline intracellular pH (pHi), a pHi-dependent increase in lateral cell membrane localization of the Wnt transducer Disheveled (Dvl), and increased Wnt/β-catenin signaling (Strubberg et al., Cell Mol Gastroenterol Hepatol. 2017 Dec 7;5(3):253-271). We hypothesized that the alkaline pHi of Cftr KO crypts also facilitates Dvl-dependent noncanonical Wnt signaling to increase Rho GTPase activity and thereby tight junction remodeling during increased proliferation. In Dvl2-eGFP rescue mice, Dvl2-eGFP intensity near the apical membrane of Cftr KO enteroid crypts was greater versus wild-type (WT) at both the crypt base and the transit-amplifying zone. Immunofluorescence studies of both freshly isolated crypts and early passage enteroids also found increased accumulation of the Rho GTPase Cdc42 at the tight junctions of Cftr KO versus WT crypts. Live imaging revealed a small increase in leak pathway permeability (3kD dextran) in Cftr KO as compared to WT murine enteroids. Increased leak permeability and Cdc42 tight junction localization were normalized in Cftr KO enteroids by suppressing proliferation through inhibition of Wnt/β-catenin signaling. There was no difference in Cdc42 expression in Cftr KO fresh crypts or enteroids, but initial studies show increased Cdc42 GTP-loaded activity in Cftr KO enteroids relative to WT. Similarly, CFTR KO human intestinal epithelial Caco-2 cells, which also show an alkaline pHi, increased Wnt/β-catenin signaling and a hyperproliferative phenotype, demonstrated a significant increase in GTP-loaded Cdc42 activity compared to WT cells. Inhibition of Cdc42 activity with ML141 (R&D Systems) resulted in a significant disruption of the Cftr KO epithelial barrier, whereas WT enteroid permeability was not affected by Cdc42 inhibition, Wnt inhibition or Wnt3a-induced proliferation. We conclude that increased Cdc42 Rho GTPase activity is cell-autonomous and is necessary for maintaining barrier integrity of the Cftr KO crypt epithelium. Systemic administration of a novel Beclin 1-derived peptide significantly upregulates autophagy in the spinal motor neurons of autophagy reporter mice

Autophagy, an intracellular degradation system, plays a vital role in protecting cells by clearing damaged organelles, pathogens, and protein aggregates. Autophagy upregulation through pharmacological interventions has gained significant attention as a potential therapeutic avenue for proteinopathies. Here, we report the development of an autophagy-inducing peptide (BCN4) derived from the Beclin 1 protein, the master regulator of autophagy. To deliver the BCN4 into cells and the central nervous system (CNS), it was conjugated to our previously developed cell and blood–brain barrier-penetrating peptide (CPP). CPP-BCN4 significantly upregulated autophagy and reduced protein aggregates in motor neuron (MN)-like cells. Moreover, its systemic administration in a reporter mouse model of autophagy resulted in a significant increase in autophagy activity in the spinal MNs. Therefore, this novel autophagy-inducing peptide with a demonstrated ability to upregulate autophagy in the CNS has significant potential for the treatment of various neurodegenerative diseases with protein aggregates as a characteristic feature.

Caki-1 Spheroids as a 3D Model for Studying Free Fatty Acid-Induced Proximal Tubule Lipotoxicity

Three-dimensional (3D) multicellular spheroid shows a biological complexity resembling native tissue in comparison to conventional monolayer cultures, making it a better model system to assess drug effcacy and cytotoxicity. The main aim of this study was to examine the usefulness of Caki-1, a human proximal tubule (PT) representative cell line, as a 3D model system for studying free fatty acid-induced PT lipotoxicity. Caki-1 spheroids were generated and maintained on ultra-low attachment plates and characterized regarding time-dependent changes in morphology and functionality. In optimal 3D culture conditions, Caki-1 cells formed well-defined large compact 3D spheroids with uniform morphology, good circularity, and increased diameter from days 2-10. There was no change when the spheroids were incubated with lipid-free bovine serum albumin (BSA, 0.5%) from day 6 to 10. Chronic exposure to palmitate (300 μM) resulted in dispersed and flattened spheroid morphology, with a larger number of dead cells in the peripheral layers and decreased spheroid core. Moreover, palmitate-treated spheroids showed a significant increase in caspase-3 activity at the external layers of the spheroids, suggesting that the detached cells underwent apoptosis. Simultaneous addition of oleate (200 μM) prevents palmitate-induced spheroid disintegration and cell death in Caki-1 3D culture. Mechanistically, palmitate triggered endoplasmic reticulum (ER) stress and autophagy dysfunction, as evidenced by significantly increased protein levels of ER stress markers AFT4 and CHOP, and autophagy substrate p62. Oleate upregulated adipose differentiation-related protein (ADRP, a lipid droplet-associated protein), and promoted lipid droplet formation in palmitate-treated Caki-1 cells. These results indicate that Caki-1 3D spheroids are a simple and reproducible in vitro system for lipotoxicity study. IDO-1 inhibition improves outcome after fluid percussion injury in adult male rats.

The enzyme indoleamine 2,3 dioxygenase 1 (IDO1) catalyzes the rate-limiting step in the kynurenine pathway (KP) which produces both neuroprotective and neurotoxic metabolites. Neuroinflammatory signals produced as a result of pathological conditions can increase production of IDO1 and boost its enzymatic capacity. IDO1 and the KP have been implicated in behavioral recovery after human traumatic brain injury (TBI), but their roles in experimental models of TBI are for the most part unknown. We hypothesized there is an increase in KP activity in the fluid percussion injury (FPI) model of TBI, and that administration of an IDO1 inhibitor will improve neurological recovery. In this study, adult male Sprague Dawley rats were subjected to FPI or sham injury and received twice-daily oral administration of the IDO1 inhibitor PF-06840003 (100 mg/kg) or vehicle control. FPI resulted in a significant increase in KP activity, as demonstrated by an increased ratio of kynurenine: tryptophan, in the perilesional neocortex and ipsilateral hippocampus 3 days postinjury (DPI), which normalized by 7 DPI. The increase in KP activity was prevented by PF-06840003. IDO1 inhibition also improved memory performance as assessed in the Barnes maze and anxiety behaviors as assessed in open field testing in the first 28 DPI. These results suggest increased KP activity after FPI may mediate neurological dysfunction, and IDO1 inhibition should be further investigated as a potential therapeutic target to improve recovery.

Human-Altered Water and Carbon Cycles in the Lake Yangzong Basin since the Yuan Dynasty

Due to the dual influence of climate change and human activities, the water cycle patterns in the lakesheds of the Yunnan karst plateau are undergoing significant changes, leading to increasingly prominent ecological issues. In the history of Lake Yangzong, an artificial water-diversion channel was excavated, altering the lake basin structure. Human activities have intensified, posing severe challenges to water resource supply and water security in recent decades. To investigate the significant increase in human activities, the temporal and phase changes, and the resulting transformation of the water and carbon cycles in the Lake Yangzong basin, we applied X-ray fluorescence spectroscopy (XRF) to scan elements continuously in a 10.2 m sediment core from this lake. By combining correlation analysis, principal component analysis (PCA), core chronology, and total organic carbon (TOC) content, we reconstructed the historical sequence of geochemical element contents in the Lake Yangzong catchment over the past 13,000 years. The results show that PC1 and PC2 contribute 78.4% and 10.3%, respectively, suggesting that erosion intensity is the main factor influencing the lake sedimentation process. From 13,400 to 680 cal a BP (calibrated years before the present), the sedimentation process in Lake Yangzong was mainly controlled by climatic conditions, with vegetation degradation during cold periods and relatively high erosion intensity in the watershed. During the Yuan dynasty, a province was established by the central government in Yunnan, promoting settlement and attracting a large number of immigrants from other provinces to Yunnan. Human activities in the Lake Yangzong basin began to intensify, surpassing natural changes and becoming the dominant force influencing the sedimentation process. In the Ming and Qing dynasties, the population and cultivated land area in Yunnan further increased, resulting in the significant exacerbation of erosion and soil loss in the watershed due to vegetation destruction. In the year 1388, the Tangchi Canal was excavated, transforming Lake Yangzong to an outflow lake, causing Ca2+ to be lost through the Tangchi Canal and preventing the formation of precipitation due to oversaturation. The research results indicate that human activities in the Lake Yangzong area have intensified since the Yuan dynasty, leading to increased erosion intensity. The excavation of the outflow canal transformed Lake Yangzong from an inland lake basin into an outflow state, simultaneously generating a significant transformation in the water and carbon cycling patterns in the watershed.

Effects of Methyl Jasmonate on Quality and Phytochemical Contents of Postharvest ‘Kamphaeng Saen 42’ Mulberry Fruit

The ‘Kamphaeng Saen 42’ mulberry fruit is consumed fresh and is rich in nutrients. However, it has a short shelf life of only 1-2 days under room temperature conditions. This research aimed to study the effects of methyl jasmonate (MeJA) on quality and phytochemical content of postharvest 'Kamphaeng Saen 42' mulberry fruit. The fruits were fumigated with 0 (control), 10, and 20 µmolL-1 MeJA at 20 °C for 6 h, then stored at 10 ± 2 °C, 90 ± 2 relative humidity (RH) for 8 days. The results showed that fruits treated with 10 µmolL-1 MeJA were most effective in retarding weight loss, and all treated fruits tended to exhibit reduced spoilage compared to the control, although the differences were not significant. Additionally, fruit treated with 10 µmolL-1MeJA showed a significant increase in phytochemicals and antioxidant activities; their total ascorbic acid and total phenolic contents were higher than in other treatments (p<0.01). Fruit treated with 20 µmolL-1 MeJA had a significantly increased skin color change (p< 0.05), resulting in the highest anthocyanin content. These results suggested that MeJA treatment effectively maintains quality and boosts phytochemical content in postharvest 'Kamphaeng saen 42' mulberry fruit.

Effect of <i>Polyalthia Longifolia</i> Leaf Extract on Uterine Function Indices of Female Wistar Rats

The bark of Polyalthia longifolia plant is commonly used in the treatment of various diseases like diabetes, malaria and many others. In this research, phytochemical screening and effects of the aqueous extract of P. longifolia leaves on the uterine function indices of female Wistar rats were investigated. Phytochemical analysis of the extract was performed using standard methods. The animals were randomized into four groups (Control A, B, C and D) of five rats each. Animals in the control group were given 1ml of distilled water daily, for 30days. While those in groups B, C and D were administered with the same volume of 50, 100 and 200mg/kg body weight of the plant extract, respectively, for 30days as well. At the end of the administration period, the animals were sacrificed 24 hours after the last day of administration. Uterine function indices were evaluated afterwards. Phytochemical screening indicated the presence of saponins, flavonoids, tannins, phenolics, cardiac glycosides, steroids, anthraquinones, phlobatannins and alkaloids while terpenoids were not detected. The extract at a moderate dose of 100mg/kg body weight gave a significant increase and decrease (P<0.05) in uterine glucose and uterine cholesterol respectively; it also revealed a significant increase and decrease(P<0.05) in alkaline phosphatase activity and total protein, when compared with the control. In general, it could be inferred that the aqueous extract of P. longifolia leaves possesses fertility enhancing potentials with regards to the uterus and uterine function indices.

Effects of Etching Time and Ethanol Wet Bonding on Bond Strength and Metalloproteinase Activity in Radicular Dentin.

(1) Background: The objective of this in vitro study was to evaluate the impact of different etching times and ethanol pre-treatments on the immediate bond strength of a hydrophilic multi-mode universal adhesive (Clearfil Universal Bond Quick, Kuraray, UBQ) and on the consequent gelatinolytic activity of metalloproteinases (MMPs) on radicular dentin. (2) Methods: Sixty single-root teeth were selected and divided into four groups according to the adhesive protocol applied for fiber post cementation: (G1) 15 s H3PO4 application + UBQ; (G2) 30 s H3PO4 application + UBQ; (G3) 15 s H3PO4 application + ethanol pre-treatment + UBQ; (G4) 30 s H3PO4 + ethanol pre-treatment + UBQ. After adhesive procedures, fiber posts were luted into the post space with a dual-curing cement (DC Core, Kuraray) and light-cured for 40 s. To perform the push-out test and nanoleakage analyses for both coronal end apical areas, 1 mm slices were prepared, following a 24 h storage period in artificial saliva. Additionally, an in situ zymographic assay was conducted to explore endogenous MMP activity within the radicular layer. Results were statistically analyzed with ANOVA and Tukey post hoc tests. Statistical significance was set at p < 0.05. (3) Result: ANOVA revealed a statistically significant difference in push-out bond strength related to the pre-treatment variable but did not highlight any significance of etching time. Specimens pre-treated with ethanol wet bond application showed higher bond strength (p < 0.01). In situ zymography quantification analyses revealed that all tested groups, independently of etching time end ethanol pre-treatment, activated MMP gelatinolytic activity. A significant increase in MMP activity was detected for the 30 s etching time. However, ETOH pre-treatment significantly reduced MMP activity within the adhesive interface (p < 0.01). (4) Conclusions: The tested adhesive showed similar results regardless of the etching time protocol. The gelatinolytic activity of MMPs was observed in all the groups. Further investigations and extended follow-ups are required to validate the results of the present study in vivo.

Integrated evaluation of the biological response of the earthworm Eisenia fetida using two glyphosate exposure strategies: soil enriched and soils collected from crops in Southeastern Mexico.

Under laboratory conditions, the toxicological effects of pesticides tend to be less variable and realistic than under field conditions, limiting their usefulness in environmental risk assessment. In the current study, the earthworm Eisenia fetida was selected as a bioindicator for assessing glyphosate toxic effects in two different trials to solve this dilemma. In Trial 1, the worms were exposed for 7 and 14days to concentrations of a commercial glyphosate formulation (1 to 500mg a.i. kg-1) currently used in the field. In Trial 2, the worms were kept in nine soils collected from different plots with crops for 14days of exposure. In both experiments, glutathione S-transferase (GST), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), and acetylcholinesterase (AChE) activities and contents of lipid peroxidation (LPO) were evaluated. In T1, the glyphosate formulation produced a 40% inhibition of AChE activity and a significant increase in GST, SOD, CAT, and GPx activities and LPO contents in E. fetida on day 7. In T2, higher concentrations of glyphosate were detected in the soils of soybean, papaya, and corn (0.92, 0.87, and 0.85mgkg-1), which induced a positive correlation between the levels of glyphosate residues with GST, SOD, CAT, GPx, and LPO and a negative correlation with AChE. These findings indicate that crop soils polluted with glyphosate elicited higher oxidative stress than under laboratory conditions, confirmed by IBRv2, PCA, and AHC analyses.

One becomes aware of the evidence of cognitive insights following attending the research-based learning program with contemplative education, electroencephalography (EEG), and electrodermal activity (EDA) were used to track the improvement of emotional and cognitive states. Research-based learning (RBL) and teaching models were developed to enhance research characteristics and cognitive training for children in the 21st century. To date, there have been no reliable cognitive tools to monitor the cognitive insight of the child who attends the course. The EEG, EDA, and cognitive scores of thirty healthy students participating in a research-based program for an hour per week were evaluated prior to the course, 3 months, and 6 months after the program started. The cognitive tests included a standard progressive matric (SPM) test, arithmetic test, Eriksen flanker task, and biofeedback sessions. The same procedures were determined in the comparative students’ group. After six months of taking the program, students in the RBL group showed a significant improvement in their SPM and arithmetic test scores. A significant increase in beta and gamma activity was detected in the temporal cortex of the RBL group during the SPM test. A more significant enhancement of frontal theta power was observed during the arithmetic test. This study shows that contemplative education and research-based learning improve cognitive abilities, indicating a significant increase in EEG quantity for cognitive engagement and the ability to confer individual differences in cognitive abilities.

BIOCHEMICAL RESPONSES IN AGROBACTERIUM-INFECTED OILSEED RAPE EXPLANTS DURING EARLY STAGES OF REGENERATION IN THE PRESENCE OF DITHIOTHREITOL

We investigated the impact of exogenous dithiothreitol on the activities of enzymatic antioxidants ascorbate peroxidase and catalase, and the levels of non-enzymatic antioxidants and osmoprotectant proline, and total soluble sugars in Agrobacterium-infected and non-infected (control) explants of oilseed rape (Brassica napus L.) after the 3rd and 17th day of in vitro culture. This investigation was conducted with or without dithiothreitol at a concentration of 1 mg/L. Dithiothreitol is a sulphur-containing compound that act as a reducing agent, impacting the redox environment within plant cells. Explants were obtained from six-day-old hypocotyls of the cultivars Ability and Lancia, and infected with Agrobacterium tumefaciens GV3101 carrying the binary vector. The application of dithiothreitol into co-cultivation and regeneration media led to a significant decrease in the activities of ascorbate peroxidase and catalase in both infected and non-infected explants of both cultivars. In contrast, explants regenerated without dithiothreitol exhibited a significant increase in ascorbate peroxidase activity and a decrease in catalase activity, although the decrease in catalase activity was not as pronounced. The proline content increased significantly in all analysed explants, with the highest increase observed in explants regenerated in the presence of dithiothreitol. This suggests that dithiothreitol might indirectly contribute to the reduced formation of reactive oxygen species, which is generally negatively correlated with transformation efficiency.

The multiple enhancements of deep eutectic solvent on cellulase significantly improve the saccharification efficiency of lignocellulosic biomass

The introduction of specific choline chloride (ChCl)- and betaine (Bet)-based deep eutectic solvents (DESs) into the conventional acetic acid–sodium acetate solution led to a significant increase in cellulase activity and stability. In particular, a higher enhancement in β-glucosidase activity was obtained, which plays a key role in improving cellulase function. The DESs prolonged the half-life of cellulase. The addition of 10 % (v/v) ChCl/1,3-propanediol (1:2) and 5 % (v/v) Bet/glycerol (1:2) resulted in the most significant improvement, with the half-life of cellulase reaching three times the initial half-life value. The above both DESs allowed the cellulase to maintain an excellent level of activity over time, even after 72 h. The DESs tightened the structure of the enzyme by changing its secondary structure, which had a stabilizing effect on the overall conformation. The density functional theory calculations of DESs revealed that the DES with a more stable structure more effectively protected the active conformation. The molecular dynamics simulation on β-glucosidase indicated that the hydrogen bond donor end of the DES primarily formed hydrogen bonds with the enzyme, which induced a spatially compact conformation by increasing the α-helix content while reducing the β-sheet content. It also restricted the movement of amino acids, particularly stabilizing the active site, and increased the hydrophilic region, thereby enhancing enzyme activity and stability. In addition, the DES formed a protective layer that prevented the unproductive adsorption of lignin on the enzymes during the hydrolysis of lignocellulosic feedstocks. These multiple improvements provided by DESs significantly increased the enzymatic saccharification efficiency of corn stover and poplar.

Neuroprotective effect of Withania somnifera leaves extract nanoemulsion against penconazole-induced neurotoxicity in albino rats via modulating TGF-β1/Smad2 signaling pathway

Penconazole (PEN) is a systemic triazole fungicide used to control various fungal diseases on grapes, stone fruits, cucurbits, and strawberries. Still, it leaves residues on treated crops after collection with many hazardous effects on population including neurotoxicity. Withania somnifera leaves extract (WSLE) is known for its memory and brain function enhancing ability. To evoke such action efficiently, WSLE bioactive metabolites are needed to cross the blood–brain barrier, that could limit the availability of such compounds to be localized within the brain. Therefore, in the present study, the association between PEN exposure and neurotoxicity was evaluated, and formulated WSLE nanoemulsion was investigated for improving the permeability of the plant extract across the blood–brain barrier. The rats were divided into five groups (n = 6). The control group was administered distilled water, group II was treated with W. somnifera leaves extract nanoemulsion (WSLE NE), group III received PEN, group IV received PEN and WSLE, and group V received PEN and WSLE NE. All rats were gavaged daily for 6 weeks. Characterization of compounds in WSLE using LC–MS/MS analysis was estimated. Neurobehavioral disorders were evaluated in all groups. Oxidative stress biomarkers, antioxidant enzyme activities, and inflammatory cytokines were measured in brain tissue. Furthermore, the gene expression patterns of GFAP, APP, vimentin, TGF-β1, Smad2 and Bax were measured. Histopathological changes and immunohistochemical expression in the peripheral sciatic nerve and cerebral cortex were evaluated. A total of 91 compounds of different chemo-types were detected and identified in WSLE in both ionization modes. Our data showed behavioral impairment in the PEN-treated group, with significant elevation of oxidative stress biomarkers, proinflammatory cytokines, neuronal damage, and apoptosis. In contrast, the PEN-treated group with WSLE NE showed marked improvement in behavioral performance and histopathological alteration with a significant increase in antioxidant enzyme activity and anti-inflammatory cytokines compared to the group administered WSLE alone. The PEN-treated group with WSLE NE in turn significantly downregulated the expression levels of GFAP, APP, vimentin, TGF-β1, Smad2 and Bax in brain tissue. In conclusion, WSLE NE markedly enhanced the permeability of plant extract constituents through the blood brain barrier to boost its neuroprotective effect against PEN-induced neurotoxicity.

Deep learning-powered multimodal biometric authentication: integrating dynamic signatures and facial data for enhanced online security

The significant increase in online activities in the wake of recent global events has underlined the importance of biometric person authentication on digital platforms. Although many biometric devices may be used for precise biometric authentication, acquiring the necessary technology, such as 3D sensors or fingerprint scanners, can be prohibitively expensive and logistically challenging. Addressing the demands of online environments, where access to specialized hardware is limited, this paper introduces an innovative approach. In this work, by fusing static and dynamic signature data with facial data captured through regular computer cameras, a dataset of 1750 samples from 25 individuals is constructed. Deep learning models, including convolutional neural networks (CNN), long short-term memory (LSTM), gated recurrent unit (GRU), and temporal convolutional networks (TCN), are employed to craft a robust multi-classification model. This integration of various deep learning algorithms has demonstrated remarkable performance enhancements in biometric authentication. This research also underscores the potential of merging dynamic and static biometric features, derived from readily available sources, to yield a high-performance recognition framework. As online interactions continue to expand, the combination of various biometric modalities holds potential for enhancing the security and usability of virtual environments.

THE USE OF LABORATORY CRITERIA IN THE RISK ASSESSMENT OF POSTOPERATIVE COMPLICATIONS AFTER TRANSPEDICULAR FIXATION OF THE LUMBAR SPINE

Objective. To investigate the peculiarities of the development of postoperative complications after surgical treatment of degenerative diseases of the lumbar spine with transpedicular fixation of the vertebrae and their relationship with changes in preoperative laboratory parameters. Methods. A retrospective analysis of the biochemical examination data of 35 patients with degenerative diseases of the lumbar spine who underwent discectomy with transpedicular fixation was performed. In four groups of patients with postoperative complications: 10 patients — inflammation of the what fabrics around metal structures and 5 patients — instability of the metal structure. The results of the examination of 20 people without specified complications served as a comparison group. Comparison was made with a control group (20 practically healthy people). Results. Patients have a significant increase in serum ALT activity by 42.29 and AST by 63.55 %, by 38.37 % of the level of CS content, by 37.21 % of total glycoproteins compared to those in the control group. In patients with soft tissue inflammation around the serum instrument zone, the content of LDLP was recorded by 29.27 %, by 23.53 % of the UA, by 12.34 % of TP results higher by 32.01 % ALT activity and 83 %, 49 % AST, 54.96 % AlP activity, 31.40 % — AlP/AcP, 60.47 % — concentration of CS, 95.57 % — total glycoproteins than in persons in the control group. In the serum of patients with the development of instability of metal structures, 28.55 % was observed by activity of the ALT, AlP — by 38.47 %, AcP — by 48.84 %, less by 58.35 % of AlP/AcP, the CHOL content of 13 88 %, higher by 25.43 % of the LDLP content than such persons in the control group. Content of the P was lower by 31.51 % and the Ca/P ratio was higher by 38.95 %, the content of CS by 83.72 %, total glycoproteins — by 48.83 %. Conclusions. Comparative analysis of the results of preoperative clinical and laboratory examination of patients with degenerative spinal diseases with transpedicular fixation which is reflected in the deviations of their preoperative laboratory parameters from such persons with uncomplicated course of the postoperative period after similar surgery.
