Primary liver cancer (PLC) is among the most common malignant tumors of the digestive tract, with high incidence and mortality. Hepatocellular carcinoma (HCC) the predominant PLC form has complex pathogenesis. Traditional Chinese medicine (TCM) has gained widespread use as supportive treatment, demonstrating clear efficacy in HCC management. This article summarizes signaling pathways regulating ferroptosis in HCC cells and systematically reviews current research on pathway modulation by TCM active constituents and compound formulas, emphasizing their cytotoxic effects. HCC cellular survival strategies that evade ferroptosis, including hepatic stellate cell-mediated protective mechanisms, are discussed. Clinical limitations of ferroptosis-based treatments and potential solutions are addressed. A comprehensive literature search using PubMed, Embase, Web of Science, and Scopus examined signaling pathways in TCM-mediated ferroptosis in HCC cells. Search terms included "hepatocellular carcinoma," "ferroptosis," "signaling pathway," "traditional Chinese medicine," "TCM monomer," "TCM formula," and "TCM extract," limited to studies from January 2021 to October 2025. Thirty-nine studies were identified: 33 on TCM active ingredients, 1 on TCM extract, and 5 on herbal formulas. TCM active ingredients and formulas promote ferroptosis in HCC cells through modulation of signaling pathways. Pathway modulation inhibits HCC cell proliferation, invasion, and metastasis; promotes apoptosis; increases chemotherapeutic sensitivity; improves glucose metabolism; and reduces drug resistance. TCM can induce ferroptosis in HCC cells through multiple pathways, targets, and mechanisms,highlighting its therapeutic advantages and providing guidance for developing novel ferroptosis inducers.

Adiponectin and irisin regulate energy homeostasis and interact with peroxisome proliferator-activated receptor coactivator 1α (PGC-1α). However, whether they establish a signal connection via PGC-1α is unclear. In the current study, the expression of irisin was significantly decreased in the skeletal muscle of adiponectin knockout (KO) mice, accompanied by a de crease in APPL1/p38 mitogen-activated protein kinase (MAPK)/PGC-1α. However, adiponectin administration reversed this effect. In vitro, the p38 MAPK/PGC-1α signalling pathway mediated adiponectin-induced FNDC5 expression and irisin release in mouse-derived C2C12 myotube cells. Moreover, obesity caused dysregulation of the adiponectin/APPL1/p38 MAPK/PGC-1α signalling pathway in murine skeletal muscle, ultimately inhibiting irisin synthesis and secretion; meanwhile, prolonged exercise or exogenous recombinant adiponectin intervention activated this pathway in mouse skeletal muscle. This corresponded with an apparent improvement in high-fat diet-induced insulin resistance. The effect of mechanically stretching C2C12 myotube cells was consistent with in vivo findings. Hence, adiponectin upregulates irisin through the APPL1/p38MAPK/PGC-1α signalling pathway in murine skeletal muscle, which may enhance insulin sensitivity.

Hypertension is a major public health concern, affecting more than one billion adults worldwide. The etiology of hypertension is associated with various genetic and epigenetic interactions. The candidate genes from various signaling pathways, such as RAAS, KKS and endothelin system, play a significant role in the progression of hypertension. The research aimed to develop a comprehensive knowledge base pertaining to hypertension, which would facilitate the integration of information related to SNPs associated with hypertension, pertinent biological pathways, risk assessment factors, molecular mechanisms, pharmacogenomic aspects of hypertension, a blood pressure calculator, and the existing literature on hypertension. This database has been implemented using HTML and Java programming languages. The deployment of the HTSNPedia database is expected to enhance the efficiency of identifying novel pharmacological drug candidates for the treatment of hypertension. A comprehensive understanding of the interrelationships among signaling pathways, molecular mechanisms, and risk-associated SNPs related to hypertension may significantly contribute to advancements in human health, particularly for individuals affected by this condition.
