Carrier Based Liquid Bioformulation of Salt-Tolerant Pgpr Bacillus Species for Prolonged Survivability

Salinity has emerged as one of the agricultural plants' most severe environmental stresses. Recently, a plant growth-promoting rhizobacteria (PGPR) is being touted as a means of solving yield and environmental issues worldwide. However, multi-strain salt-tolerant rhizobacteria have a short shelf life due to their structural and cellular components, therefore, they need to be supplemented with a liquid carrier material to serve as a shelter and energy source for the bacteria for longer survival. The present study has been undertaken to develop a liquid biofertilizer formulation from multi-strain salt-tolerant PGPR – UPMR, UPMRE6, and a mixed strain of UPMRB9 and UPMRE6 using an optimum amount of cell protectants, namely glycerol (5 mM), trehalose (10 mM), and polyvinyl pyrrolidone (PVP) at 1%. The shelf-life was assessed through measurements of optical density and bacterial biomass to determine the bacterial population and growth trend at monthly intervals. After three months of incubation, the optical density was the highest in the mixed strain treatment supplemented with trehalose with 1.3% and 2.2% increase relative to the UPMRE6 and UPMRB9, respectively, using the same cell protectants. Similarly, bacterial biomass production was the highest in the mixed strains treatment amended with trehalose (0.025 g/mL), with 13.64% and 38.89% increment followed by UPMRE6 and UPMRB9, respectively. Irrespective of the type of protectants used and PGPR type, the optical density and bacterial biomass generally decreased long incubation period. The results demonstrated that the use of 10 mM trehalose has the potential to extend the bacterial shelf life with the slightest cell loss.

A REVIEW: HARNESSING SALT-TOLERANT PLANT GROWTH-PROMOTING RHIZOBACTERIA FOR SUSTAINABLE AGRICULTURE

The salinity of the soil is becoming a major concern for the worlds food security. it is essential to create environmentally friendly sustainable techniques that boost saline soil productivity. Certain microorganisms have a dependency on salt as well as tolerance to it. Microbes that are resistant to salt may withstand ionic and osmotic stress. From very alkaline, saline, and sodic soils, many genera of salt-tolerant plant growth-promoting rhizobacteria (ST-PGPR) have been discovered. Numerous them are also known to help plants cope with a range of biotic and abiotic stressors. The possibility of using ST-PGPR to improve the productivity of plants under salt stress has been studied in the past several years, indicating that saline agro-ecosystems can be restored through its use. This review will focus on ST-PGPR and how they might boost saline agro-ecosystem production. Additionally, it sheds light on future research directions on the use of ST-PGPR for saline soil reclamation and PGPR-mediated mechanisms of salt tolerance in various crop plants. Plant growth-promoting rhizobacteria Pseudomonas aeruginosa HG28-5 improves salt tolerance by regulating Na+/K+ homeostasis and ABA signaling pathway in tomato

Salinity stress badly restricts the growth, yield and quality of vegetable crops. Plant growth-promoting rhizobacteria (PGPR) is a friendly and effective mean to enhance plant growth and salt tolerance. However, information on the regulatory mechanism of PGPR on vegetable crops in response to salt stress is still incomplete. Here, we screened a novel salt-tolerant PGPR strain Pseudomonas aeruginosa HG28–5 by evaluating the tomatoes growth performance, chlorophyll fluorescence index, and relative electrolyte leakage (REL) under normal and salinity conditions. Results showed that HG28–5 colonization improved seedling growth parameters by increasing the plant height (23.7%), stem diameter (14.6%), fresh and dry weight in the shoot (60.3%, 91.1%) and root (70.1%, 92.5%), compared to salt-stressed plants without colonization. Likewise, HG28–5 increased levels of maximum photochemical efficiency of PSII (Fv/Fm) (99.3%), the antioxidant enzyme activities as superoxide dismutase (SOD, 85.5%), peroxidase (POD, 35.2%), catalase (CAT, 20.6%), and reduced the REL (48.2%), MDA content (41.3%) and ROS accumulation in leaves of WT tomatoes under salt stress in comparison with the plants treated with NaCl alone. Importantly, Na+ content of HG28–5 colonized salt-stressed WT plants were decreased by15.5% in the leaves and 26.6% in the roots in the corresponding non-colonized salt-stressed plants, which may be attributed to the higher K+ concentration and SOS1, SOS2, HKT1;2, NHX1 transcript levels in leaves of colonized plants under saline condition. Interestingly, increased abscisic acid (ABA) content and upregulation of ABA pathway genes (ABA synthesis-related genes NCED1, NCED2, NCED4, NECD6 and signal genes ABF4, ABI5, and AREB) were observed in HG28–5 inoculated salt-stressed WT plants. ABA-deficient mutant (not) with NCED1 deficiency abolishes the effect of HG28–5 on alleviating salt stress in tomato, as exhibited by the substantial rise of REL and ROS accumulation and sharp drop of Fv/Fm in the leaves of not mutant plants. Notably, HG28–5 colonization enhances tomatoes fruit yield by 54.9% and 52.4% under normal and saline water irrigation, respectively. Overall, our study shows that HG28–5 colonization can significantly enhance salt tolerance and improved fruit yield by a variety of plant protection mechanism, including reducing oxidative stress, regulating plant growth, Na+/K+ homeostasis and ABA signaling pathways in tomato. The findings not only deepen our understanding of PGPR regulation plant growth and salt tolerance but also allow us to apply HG28–5 as a microbial fertilizer for agricultural production in high-salinity areas.

Comparative Analysis of Plant Growth-Promoting Rhizobacteria's Effects on Alfalfa Growth at the Seedling and Flowering Stages under Salt Stress.

Alfalfa (Medicago sativa L.), a forage legume known for its moderate salt-alkali tolerance, offers notable economic and ecological benefits and aids in soil amelioration when cultivated in saline-alkaline soils. Nonetheless, the limited stress resistance of alfalfa could curtail its productivity. This study investigated the salt tolerance and growth-promoting characteristics (in vitro) of four strains of plant growth-promoting rhizobacteria (PGPR) that were pre-selected, as well as their effects on alfalfa at different growth stages (a pot experiment). The results showed that the selected strains belonged to the genera Priestia (HL3), Bacillus (HL6 and HG12), and Paenibacillus (HG24). All four strains exhibited the ability to solubilize phosphate and produce indole-3-acetic acid (IAA) and 1-aminocyclopropane-1-carboxylate (ACC) deaminase. Among them, except for strain HG24, the other strains could tolerate 9% NaCl stress. Treatment with 100 mM NaCl consistently decreased the IAA production levels of the selected strains, but inconsistent changes (either enhanced or reduced) in terms of phosphate solubilization, ACC deaminase, and exopolysaccharides (EPS) production were observed among the strains. During the various growth stages of alfalfa, PGPR exhibited different growth-promoting effects: at the seedling stage, they enhanced salt tolerance through the induction of physiological changes; at the flowering stage, they promoted growth through nutrient acquisition. The current findings suggest that strains HL3, HL6, and HG12 are effective microbial inoculants for alleviating salt stress in alfalfa plants in arid and semi-arid regions. This study not only reveals the potential of indigenous salt-tolerant PGPR in enhancing the salt tolerance of alfalfa but also provides new insights into the mechanisms of action of PGPR.

Bacillus cereus: an effective bio-inoculant for promoting salt stress tolerance of rice seedlings under saline soil conditions

Plant growth-promoting rhizobacteria (PGPR) are a powerful tool to maintain sustainable agriculture and promote plant resistance to biotic and abiotic types of stress. Salinity, a major abiotic stress hampers plant growth, development, and yield. Salt-tolerant PGPR are effective agents for ameliorating salinity effects on rice plants. The present study endeavored to isolate, determine halotolerant ability, characterize Plant Growth Promoting (PGP) traits, and finally observe the effect of PGPR strain on plant growth promotion of rice plants under saline and non-saline conditions. Based on the 16S rRNA gene sequencing technique, the rhizobacterial strain DB2 was identified as Bacillus cereus ATCC 14579(T) from NCMR, NCCS Pune. To check the growth-promoting ability, the strain was inoculated with two rice genotypes named Chinsurah Nona I (salt tolerant-non aromatic) and Badshabhog (aromatic) under polyhouse conditions. Results showed a significant increment in relative water content (RWC), total chlorophyll content (TCC), root length (RL), and shoot length (SL) in both rice genotypes inoculated with DB2 under both saline and non-saline conditions. Under non-saline conditions enhancement of RWC, TCC, RL, and SL was better in DB2 inoculated Chinsurah Nona I than in Badshabhog inoculated with DB2. Whereas, DB2-inoculated Badshabhog showed more recovery of RWC, TCC, RL, and SL than DB2-inoculated Chinsurah Nona I under saline conditions. Under salt stress conditions, inoculation with the rhizobacterial strain showed a significant reduction in electrolytic leakage (EL) in rhizobacteria inoculated with both rice genotypes. Moreover, DB2 inoculation showed a significant reduction in Na+ content in the roots of Chinsurah Nona I (44.6%) and Badshabhog (24.5%) rice genotypes. The present study has indicated that the application of salt-tolerant PGPR may be an effective and sustainable solution for rice cultivation under salt-stress conditions.

Enhancing quinoa (Chenopodium quinoa) growth in saline environments through salt-tolerant rhizobacteria from halophyte biotope.

The use of plant growth-promoting rhizobacteria (PGPR) in agriculture is one of the most promising approaches to improve plants' growth under salt stress and to support sustainable agriculture under climate change. In this context, our goal was to grow and enhance quinoa growth using native rhizobacteria that can withstand salt stress. To achieve this objective, we isolated rhizobacteria from three saline localities in a semi-arid region in Tunisia, which are characterized by different halophyte species and tested their plant growth-promoting (PGP) activities. Then, we inoculated quinoa seedlings cultivated on 300 mM NaCl with the three most efficient rhizobacteria. A positive effect of the three-salt tolerant rhizobacteria on the growth of quinoa under salinity was observed. In fact, the results of principal component analysis indicated that the inoculation of quinoa by salt-tolerant PGPR under high salinity had a prominent beneficial effect on various growth and physiological parameters of stressed plant, such as the biomass production, the roots length, the secondary roots number, proline content and photosynthesis activities. Three rhizobacteria were utilized in this investigation, and the molecular identification revealed that strain 1 is related to the Bacillus inaquosorum species, strain 2 to Bacillus thuringiensis species and strain 3 to Bacillus proteolyticus species. We can conclude that the saline soil, especially the halophytic rhizosphere, is a potential source of salt-tolerant plant growth-promoting rhizobacteria (ST-PGPR), which stimulate the growth of quinoa and improve its tolerance to salinity.

Co-application of biochar and salt tolerant PGPR to improve soil quality and wheat production in a naturally saline soil

Soil salinity is a global problem that drastically reduces crop productivity. Therefore, there is a need to make saline soils productive. Plant growth promoting rhizobacteria (PGPR) and biochar have been shown to increase crop productivity and soil fertility when applied separately. However, their co-application in this regard has not been studied extensively. In this study, a biochar was co-applied with Bacillus thuringiensis or Bacillus tropicus, characterized as P-solubilizing and salt-tolerant PGPR, to wheat grown in a naturally saline soil (EC 8.12 dS m−1). Co-application of biochar with PGPR increased the number of tillers & spikes, straw yield, and 100-grain weight by 16–28.57%, 26.9–36.6%, 18–21.8%, and 34–37.9% respectively. This led to a 37% and 34% increase in grain yield in the case of co-application with B. thuringiensis and B. tropicus respectively. The higher growth and yield were thanks to better plant nutrition and mitigated salinity stress under co-application. Briefly, the co-application increased phosphorus content in grain, straw, and roots by up to 9–43%, 45–69%, and 5–21.5% respectively, which was higher than that in individual applications of biochar or any PGPR. In addition, co-application increased microbial biomass by 61–77% and soil available phosphorus by 37–53% across two PGPR. Evidently, the co-application contributed to increased P nutrition in two ways. Firstly, the biochar itself served as a source of available P. Secondly, the PGPR solubilized it more abundantly when biochar was there. As far the salinity stress, biochar, its co-application with B. thuringiensis, and with B. tropicus increased soil K+/Na+ by 36%, 72% and 44% respectively leading to 40%, 63%, and 55% higher K+/Na+ in grain, straw, and roots respectively. Overall, our results indicate that biochar and PGPR worked in tandem to substantially improve soil quality and wheat production compared to their sole applications. We conclude that co-application of salt tolerant PGPR and biochar is a more sustainable and better technology to produce wheat in a naturally saline soil compared to their sole applications.

Rhizoactinobacteria Enhance Growth and Antioxidant Activity in Thai Jasmine Rice (Oryza sativa) KDML105 Seedlings under Salt Stress.

Salinity is one of the most devastating abiotic stresses hampering the growth and production of rice. Nine indole-3-acetic acid (IAA)-producing salt-tolerant plant-growth-promoting rhizobacteria (ST-PGPR) were inoculated into Thai jasmine rice (Oryza sativa L.) variety Khao Dawk Mali 105 (KDML105) seedlings grown under different concentrations of NaCl (0, 50, 100, and 150 mM). The ST-PGPR strains significantly promoted the growth parameters, chlorophyll content, nutrient uptake (N, P, K, Ca, and Mg), antioxidant activity, and proline accumulation in the seedlings under both normal and saline conditions compared to the respective controls. The K+/Na+ ratio of the inoculated seedlings was much higher than that of the controls, indicating greater salt tolerance. The most salt-tolerant and IAA-producing strain, Sinomonas sp. ORF15-23, yielded the highest values for all the parameters, particularly at 50 mM NaCl. The percentage increases in these parameters relative to the controls ranged from >90% to 306%. Therefore, Sinomonas sp. ORF15-23 was considered a promising ST-PGPR to be developed as a bioinoculant for enhancing the growth, salt tolerance, and aroma of KDML105 rice in salt-affected areas. Environmentally friendly technologies such as ST-PGPR bioinoculants could also support the sustainability of KDML105 geographical indication (GI) products. However, the efficiency of Sinomonas sp. ORF15-23 should be evaluated under field conditions for its effect on rice nutrient uptake and growth, including the 2AP level.

Survival of beneficial microbes in liquid bioformulation and optimization of different carrier materials using RSM technique.

Soil salinity in rice cultivation areas is considered a severely limiting factor that adversely affects the quantity and quality of rice production in wetlands. Recently, the alternative use of salt-tolerant plant growth-promoting rhizobacteria (PGPR) inhabiting extreme saline conditions has gained remarkable attention and had positive effects on soil and crops. Therefore, a study has been initiated to develop a liquid biofertilizer formulation from locally isolated multi-strain salt-tolerant PGPR strains such as Bacillus tequilensis and Bacillus aryabhattai, using glycerol (5 mM), trehalose (10 mM), and polyvinylpyrrolidone (PVP) at 1% as additives to prolong the shelf-life of the bacteria. After 3 months of incubation, the bacterial population in the trehalose-supplemented mixed strain was highest at 9.73×107 CFU/mL, followed by UPMRE6 and UPMRB9 at 9.40×107 CFU/mL and 8.50×107 CFU/mL respectively. The results showed that the optimal trehalose concentration successfully prolonged the shelf-life of bacteria with minimal cell loss. Validation of quadratic optimization by response surface methodology revealed that the cell density of the mixed strain was 4.278×107 log CFU/mL after 24 h. The precision ratio was 99.7% higher than the predicted value in the minimized medium formulation: 0.267 g/mL trehalose, 1% glycerol, at 120 rpm agitation using the data analysis tools of Design Expert software. The population study confirmed the better and longer survival of salt-tolerant PGPR fortified with 10 mM trehalose, which was considered the best liquid biofertilizer formulation. Moreover, the optimized trehalose-glycerol liquid formulation can be used commercially as it is cost-effective.

Rhizobacterial Isolates from Prosopis limensis Promote the Growth of Raphanus sativus L. Under Salt Stress

Microbial biotechnology employes techniques that rely based on the natural interactions that occur in ecosystems. Bacteria, including rhizobacteria, play an important role in plant growth, providing agricultural crops with an alternative that can mitigate the negative effects of abiotic stress, such as those caused by saline environments. In this study, bacterial isolates were obtained from soil and roots of Prosopis limensis Bentham from the department of Lambayeque, Peru. This region has high salinity levels, therefore, the collected samples were used to isolate plant growth-promoting rhizobacteria (PGPR), which were identified through morphological, and physical-biochemical characteristics. These salt tolerant bacteria were screened phosphate solubilization, indole acetic acid, deaminase activity and molecular characterization by 16S rDNA sequencing. Eighteen samples from saline soils of the Prosopis limensis plants in the northern coastal desert of San Jose district, Lambayeque, Peru. The bacterial isolates were screened for salt tolerance ranging from 2 to 10%, a total of 78 isolates were found. Isolates 03, 13 and 31 showed maximum salt tolerance at 10%, in vitro ACC production, phosphate solubilization and IAA production. The three isolates were identified by sequencing the amplified 16S rRNA gene and were found to be Pseudomonas sp. 03 (MW604823), Pseudomonas sp. 13 (MW604824) and Bordetella sp. 31 (MW604826). These microorganisms promoted the germination of radish plants and increased the germination rates for treatments T2, T3 and T4 by 129, 124 and 118% respectively. The beneficial effects of salt tolerant PGPR isolates isolated from saline environments can be new species, used to overcome the detrimental effects of salt stress on plants. The biochemical response and inoculation of the three isolates prove the potential of using these strains as a source of products that can be employed for the development of new compounds proving their potential as biofertilizers for saline environments.

Coping with salt stress-interaction of halotolerant bacteria in crop plants: A mini review.

Salinity is one of the major environmental abiotic stress factors that limit the growth and yield of crop plants worldwide. It is crucial to understand the importance of several adaptive mechanisms in plants toward salt stress so as to increase agricultural productivity. Plant resilience toward salinity stress is improved by cohabiting with diverse microorganisms, especially bacteria. In the last few decades, increasing attention of researchers has focused on bacterial communities for promoting plant growth and fitness. The biotechnological applications of salt-tolerant plant growth-promoting rhizobacteria (PGPR) gained widespread interest for their numerous metabolites. This review provides novel insights into the importance of halotolerant (HT) bacteria associated with crop plants in enhancing plant tolerance toward salinity stress. Furthermore, the present review highlights several challenges of using HT-PGPR in the agricultural field and possible solutions to overcome those challenges for sustainable agriculture development in the future.

The Rhizosphere of Petrosimonia Triandra may Possess Growth-Inducing and Salinity-Tolerance Potential

The root microbiome of halotolerant plants can represent a great source of salt-tolerant plant growth-promoting rhizobacteria (ST-PGPR) with a wide range of beneficial effects on plants growth, development and productivity. The better understanding of the variety of complex salt tolerance and adaptive mechanisms of halophytes can contribute to improve salt tolerance in crops, and as a result, a better and increased use of saline areas worldwide. In addition, reclaiming saline areas for agricultural use may be able to alleviate the global threat of progressive soil salinization.

Wheat plant –Plant Growth Promoting Rhizobacteria (PGPR) interaction to alleviate the salt stress in vitro and in vivo

The current work studied the mechanistic basis of salt tolerant PGPR bacteria in alleviating salinity stress in wheat.Two experiments were conducted at the Agricultural Research Center (ARC), 2021-2022, to evaluate the interaction between wheat var. Misr1 and two salt tolerant PGPR bacterial strains, Azospirillum (A.) brasilense NO40 and Bacillus thuringiensis (Bt) under saline conditions. The staining technique used to detect the colonization effectiveness of applied bacteria. Images of 30-day wheat seedlings were captured and subjected to the following measurements: root area, leaf area, root length, shoot length, and dark green color index (DGCI). TEM was used to look for bacterial colonization in wheat seedling. Chlorophyll, proline, peroxidase activity, rhizosphere dehydrogenase activity and electrical conductivity (EC) were all quantified. Results revealed that the wheat seedling roots inoculated with the salt tolerant bacterial strains produced a highly intense red colour. The colonized bacteria appeared on the outer surface of the root and intercellular space of root tissue. The data showed a significant rise in all measured parameters in the inoculated wheat seedlings with the salt tolerant bacterial strains compared to the non-inoculated wheat seedlings. Additionally, the wheat seedlings inoculated with a mixture of NO40 and Bt showed the best results as compared to that inoculated with only NO40 or with only Bt. Finally, EC values before and after planting with the inoculated wheat seeds revealed a decrease in the salinity of the soil rhizosphere (9.27 mmhos/cm vs. 5.1 mmhos/cm). In conclusion: Salt stress in wheat saline soils was alleviated by bacterial strains studied. Optimum results were obtained from the mixture of A. brasilense and Bt. Results must later be verified in field trials.

The PGPR Mechanisms of Salt Stress Adaptation and Plant Growth Promotion

Worldwide crop productivity hampers severely due to the adverse effects of salinity. Global warming causes a rapid escalation of the salt-affected area, and new agricultural land is affected through saltwater intrusion. The ever-growing human population impulses to utilize the saline area for crop cultivation to ensure food security. Salinity resistance crops could be a promising substitute but with minor success because inappropriate tactics on saline soil management resulted in unsatisfactory yield. Salt-tolerant plant growth-promoting rhizobacteria (ST-PGPR) is considered an alternate way towards enhancing crop growth in saline ecosystems. It is reported that PGPR is enabled to produce exopolysaccharides which lead to biofilm formation and generate osmoprotectants and antioxidant enzymes that can significantly contribute to stimulating plant growth in the saline ecosystem. In addition, several plant growth-promoting characteristics of PGPR such as the acquisition of essential nutrients and upsurge hormone production could enhance plant growth simultaneously. In this review, we will explore the survival mechanisms of ST-PGPR and their influence on plant growth promotion in saline ecosystems.

A transcriptomic study reveals salt stress alleviation in cotton plants upon salt tolerant PGPR inoculation

Soil salinity is a major constraint for reducing crop productivity worldwide. To combat this situation, the current project was aimed to examine the effect of plant growth-promoting rhizobacteria (PGPR) inoculation on the growth of cotton ( Gossypium hirsutum, var. Jin668) plants during salt stress and to identify salt stress-responsive genes in cotton plants. For this purpose, two bacteria, Bacillus subtilis and Bacillus pumilus were selected among the 20 strains isolated from the cotton rhizosphere under the salt stress (200 mM NaCl) and identified by 16 S rRNA sequencing. B. subtilis and B. pumilus were applied to Jin668 plants under salt stress which enhanced resistance to salt stress (leaf and root growth) compared to only salt-treated plants and control plants. Transcriptomic analysis revealed 556 differentially expressed genes (481 up-regulated and 75 down-regulated) in the B. subtilis + Salt versus Salt treatments and 943 (536 up-regulated and 407 down-regulated) genes in the B. pumilus + Salt versus Salt treatments. KEGG analysis of B. pumilus + Salt versus Salt and B. pumilus + Salt versus Salt revealed the pathways plant-pathogen interaction and plant hormone signal transduction were expressed in both treatments, while ascorbate and aldarate metabolism pathways and glyoxylate and dicarboxylate metabolism pathways were uniquely expressed in the B. pumilus + Salt versus Salt comparison, and the pentose and glucuronate interconversions pathway was uniquely expressed in the B. pumilus + Salt treatment versus Salt comparison. These data showed that B. subtilis and B. pumilus significantly enhance salt stress tolerance in cotton plants during salt stress conditions. • Two Bacteria, Bacillus subtilis and Bacillus pumilus were selected from the cotton rhizosphere under the salt stress. • B. subtilis and B. pumilus were applied to cottonplants under salt stress which enhanced resistance to salt stress. • Transcriptomic analysis revealed 556 DEGs in the B.subtilis vs Salt and 943 DEGS in the B.pumilus vs Salt treatments. • KEGG analysis of B. subtilis and B. pumilus versus Salt revealed the variouspathways were expressed in both treatments. • In conclusion, B. subtilis and B. pumilus significantly enhancetolerance in cotton plants during salt stress conditions.

Salt-Tolerant PGPR Confer Salt Tolerance to Maize Through Enhanced Soil Biological Health, Enzymatic Activities, Nutrient Uptake and Antioxidant Defense.

Salt-tolerant plant growth-promoting rhizobacteria (PGPR) can improve soil enzyme activities, which are indicators of the biological health of the soil, and can overcome the nutritional imbalance in plants. A pot trial was executed to evaluate the effect of inoculation of different salt-tolerant PGPR strains in improving soil enzyme activities. Three different salinity levels (original, 5, and 10 dS m–1) were used and maize seeds were coated with the freshly prepared inocula of ten different PGPR strains. Among different strains, inoculation of SUA-14 (Acinetobacter johnsonii) caused a maximum increment in urease (1.58-fold), acid (1.38-fold), and alkaline phosphatase (3.04-fold) and dehydrogenase (72%) activities as compared to their respective uninoculated control. Acid phosphatase activities were found to be positively correlated with P contents in maize straw (r = 0.96) and grains (r = 0.94). Similarly, a positive correlation was found between alkaline phosphatase activities and P contents in straw (r = 0.77) and grains (r = 0.75). In addition, urease activities also exhibited positive correlation with N contents in maize straw (r = 0.92) and grains (r = 0.91). Moreover, inoculation of Acinetobacter johnsonii caused a significant decline in catalase (39%), superoxide dismutase (26%) activities, and malondialdehyde contents (27%). The PGPR inoculation improved the soil’s biological health and increased the uptake of essential nutrients and conferred salinity tolerance in maize. We conclude that the inoculation of salt-tolerant PGPR improves soil enzyme activities and soil biological health, overcomes nutritional imbalance, and thereby improves nutrient acquisition by the plant under salt stress.

A Salt-Tolerant Streptomyces paradoxus D2-8 from Rhizosphere Soil of Phragmites communis Augments Soybean Tolerance to Soda Saline-Alkali Stress.

Soil salinity and alkalization limit plant growth and agricultural productivity worldwide. The application of salt-tolerant plant growth-promoting rhizobacteria (PGPR) effectively improved plant tolerance to saline-alkali stress. To obtain the beneficial actinomyces resources with salt tolerance, thirteen isolates were isolated from rhizosphere saline and alkaline soil of Phragmites communis. Among these isolates, D2-8 was moderately halophilic to NaCl and showed 120 mmol soda saline-alkali solution tolerance. Moreover, the plant growth-promoting test demonstrated that D2-8 produced siderophore, IAA, 1-aminocyclopropane-1-carboxylate deaminase (ACCD), and organic acids. D2-8 showed 99.4% homology with the type strain Streptomyces paradoxus NBRC 14887T and shared the same branch, and, therefore, it was designated S. paradoxus D2-8. Its genome was sequenced to gain insight into the mechanism of growth-promoting and saline-alkali tolerance of D2-8. IAA and siderophore biosynthesis pathway, genes encoding ACC deaminase, together with six antibiotics biosynthesis gene clusters with antifungal or antibacterial activity, were identified. The compatible solute ectoine biosynthesis gene cluster, production, and uptake of choline and glycine betaine cluster in the D2-8 genome may contribute to the saline-alkali tolerance of the strain. Furthermore, D2-8 significantly promoted the seedling growth even under soda saline-alkali stress, and seed coating with D2-8 isolate increased by 5.88% of the soybean yield in the field. These results imply its significant potential to improve soybean soda saline-alkali tolerance and promote crop health in alkaline soil.

The Mechanisms of Sodium Chloride Stress Mitigation by Salt-Tolerant Plant Growth Promoting Rhizobacteria in Wheat

We investigated the growth-promoting mechanism of salt-tolerant plant growth promoting rhizobacteria (ST-PGPR) in wheat under sodium chloride (NaCl) stress by measuring the growth and physiological and biochemical responses of wheat plants inoculated with ST-PGPR under 0–400 mM NaCl. The results showed that ST-PGPR plays a significant role in the growth of wheat under NaCl stress. Under 300 mM NaCl, wheat plants inoculated with the three ST-PGPR strains increased in plant height, root length, dry weight, and fresh weight by 71.21%, 89.19%, 140.94%, and 36.31%, respectively, compared to the control group. The proline and soluble sugar contents of wheat inoculated with Bacillus thuringiensis increased by 38.8% and 21.4%, respectively. The average content of antioxidant enzymes increased by 13.89%, and compared with the control, in wheat inoculated with the three species of ST-PGPR, the average content of ethylene decreased 2.16-fold. In addition, a mathematical model based on the “interaction equation” revealed that the best results of mixed inoculation were due to the complementary strengths of the strains. The analysis of experimental phenomena and data revealed the mechanisms by which Brevibacterium frigoritolerans, Bacillus thuringiensis, and Bacillus velezensis alleviate NaCl stress in wheat: (1) by lowering of osmotic stress, oxidative stress, and ethylene stress in wheat and (2) by using root secretions to provide substances needed for wheat. This study provides a new approach for the comprehensive understanding and evaluation of ST-PGPR as a biological inoculant for crops under salt stress.

Bacillus mycoides PM35 Reinforces Photosynthetic Efficiency, Antioxidant Defense, Expression of Stress-Responsive Genes, and Ameliorates the Effects of Salinity Stress in Maize.

Soil salinity is one of the abiotic constraints that imbalance nutrient acquisition, hampers plant growth, and leads to potential loss in agricultural productivity. Salt-tolerant plant growth-promoting rhizobacteria (PGPR) can alleviate the adverse impacts of salt stress by mediating molecular, biochemical, and physiological status. In the present study, the bacterium Bacillus mycoides PM35 showed resistance up to 3 M NaCl stress and exhibited plant growth-promoting features. Under salinity stress, the halo-tolerant bacterium B. mycoides PM35 showed significant plant growth-promoting traits, such as the production of indole acetic acid, siderophore, ACC deaminase, and exopolysaccharides. Inoculation of B. mycoides PM35 alleviated salt stress in plants and enhanced shoot and root length under salinity stress (0, 300, 600, and 900 mM). The B. mycoides PM35 alleviated salinity stress by enhancing the photosynthetic pigments, carotenoids, radical scavenging capacity, soluble sugars, and protein content in inoculated maize plants compared to non-inoculated plants. In addition, B. mycoides PM35 significantly boosted antioxidant activities, relative water content, flavonoid, phenolic content, and osmolytes while reducing electrolyte leakage, H2O2, and MDA in maize compared to control plants. Genes conferring abiotic stress tolerance (CzcD, sfp, and srfAA genes) were amplified in B. mycoides PM35. Moreover, all reactions are accompanied by the upregulation of stress-related genes (APX and SOD). Our study reveals that B. mycoides PM35 is capable of promoting plant growth and increasing agricultural productivity.
