Implementation of revenue management in the process industry of North America and Europe

Previous research has extensively studied the application of RM in the service industry but has given little attention to its application in the manufacturing industry. This Chapter provides results of a quantitative study based on 479 firms of the PI in North America and Europe. The results show that the overall importance of RM within the process industry is regarded as high and that the importance is positively correlated with turnover, period of use and the extent of IT integration. The type of RM system employed depends on its period of use: with increasing period of use RM systems shift from capacity to price and capacity control. Barriers to the implementation of RM systems are seen in the absence of a clearly defined pricing strategy, lack of experience and lack of adequate approaches. Comparisons between North America and Europe indicate differences in the application of RM: In North America RM is considered as more important, has been introduced earlier and is more price-based.

Study of Rock-Soil Aggregate’s Shear Strength in Da-Liu Expressway

This study is based on 8 large-scale outdoor tests of rock-soil aggregate in Da-Liu Expressway subgrade construction site.and has got the rock-soil aggregate’s characteristics and strength parameters of deformation. Proposing alternate dry-wet effect should be taken into account in the shakedown analysis of rock-soil aggregate slope. This study provides the necessary basis for the Da-Liu Expressway slope stability analysis and control of Settlement. It also provides an important basis for further research which has been widely used in various projects RM systems approach, especially to determine physical parameters of rock-soil aggregate.

DNA Nanomachines Investigated By Non-Denaturing Mass Spectrometry

We are using a novel approach for the investigation of noncovalent protein-DNA assemblies in vitro, non-denaturing nano-electrospray Q-TOF mass spectrometry combined with ion mobility spectroscopy (IMS-MS/MS), to study assembly, size and shape of protein nanomachines which act on DNA.Specifically we highlight recent results on the BcgI restriction-modification system, which consists of two types of subunits: BcgIA contains both endonuclease and methyltransferase motifs; BcgIB is homologous to the HsdS subunits of Type I RM systems that mediate DNA sequence recognition. Together they form an A2B protomer which is active only when bound to two copies of its site, and then cuts eight phosphodiester bonds, those on both sides of both sites, before dissociating from the DNA.The data is discussed in the context of functional assays and additional biophysical characterization by Analytical Ultracentrifugation.We are also presenting preliminary results on the assembly of other DNA-protein complexes with particular emphasis on natively unstructured proteins which adopt a defined conformation only when interacting with specific DNA sequences.

Slow water dynamics at the surface of macromolecular assemblies of different morphologies

In this contribution we, for the first time, explore the slow dynamical states of confined water molecules in lamellar structures of AOT with various degrees of hydration using a picosecond resolved fluorescence spectroscopic technique using coumarin-500 as the fluorophore. A comparison of slow dynamics between AOT lamellar structures and AOT RMs have been made by preparing RMs that have a diameter the same as the interplanar water layer thickness of lamellar structures and the same number of water molecules in lamellar structures in order to understand the effect of morphology and hydration on the relaxation dynamics of water molecules in these nanoconfining systems. The relaxation time scales obtained in the lamellar systems differ to those of the RM systems and the difference of the timescales has been explained as a interplay between two opposing factors arising out of the morphology and interlayer distance, respectively. The geometrical restriction of the probe at the lamellar interface is determined by measuring time-resolved rotational anisotropy. The hydrogen bond energy of the water molecules residing at the lamellar interface is measured applying the Arrhenius type barrier crossing model.

On the Formation of New Reverse Micelles: A Comparative Study of Benzene/Surfactants/Ionic Liquids Systems Using UV−Visible Absorption Spectroscopy and Dynamic Light Scattering

The microenvironment of the polar core generated in different ionic liquid reverse micelle (IL RM) systems were investigated using the solvatochromic behavior of 1-methyl-8-oxyquinolinium betaine (QB) as an absorption probe and dynamic light scattering (DLS) technique. The novel RM systems consist of two different ILs--1-butyl-3-methylimidazolium tetrafluoroborate (bmimBF4) and 1-butyl-3-methylimidazolium bis(trifluoromethylsulfonyl)imide (bmimTf2N)--sequestrated by two different surfactants--Triton X-100 (TX-100) and benzyl-n-hexadecyldimethylammonium chloride (BHDC)--in order to make IL/surfactant/benzene RMs. The effect of the variation of Ws (Ws=[IL]/[surfactant]) on the QB spectroscopy was used to characterize these nonaqueous RMs. DLS results confirm the formation of these IL RM systems because increasing Ws increases the droplet sizes. Moreover it is demonstrated that the structure of the sequestrated ILs depends strongly on the type of surfactant use to create the RMs.

Time, Price, and Advanced Booking of Hotel Rooms

ABSTRACT The effectiveness of revenue management systems has diminished in recent years due to the systems' inability to address the increasing complication of online deal-seeking behavior. To restore their efficiency, one must first understand the changes in advanced-booking behavior and their implications. This study expands the consumer booking model by addressing the impact of time-before-the-date-of-stay and exploring the implication for the hotel's pricing/marketing strategies. The findings underscore the urgent need for empirical research on timing by showing that the predictions of the advanced-booking model, and consequently the effectiveness of RM systems, depend on the actual patterns over time.

Identification of a new subfamily of HNH nucleases and experimental characterization of a representative member, HphI restriction endonuclease

The restriction endonuclease (REase) R. HphI is a Type IIS enzyme that recognizes the asymmetric target DNA sequence 5'-GGTGA-3' and in the presence of Mg(2+) hydrolyzes phosphodiester bonds in both strands of the DNA at a distance of 8 nucleotides towards the 3' side of the target, producing a 1 nucleotide 3'-staggered cut in an unspecified sequence at this position. REases are typically ORFans that exhibit little similarity to each other and to any proteins in the database. However, bioinformatics analyses revealed that R.HphI is a member of a relatively big sequence family with a conserved C-terminal domain and a variable N-terminal domain. We predict that the C-terminal domains of proteins from this family correspond to the nuclease domain of the HNH superfamily rather than to the most common PD-(D/E)XK superfamily of nucleases. We constructed a three-dimensional model of the R.HphI catalytic domain and validated our predictions by site-directed mutagenesis and studies of DNA-binding and catalytic activities of the mutant proteins. We also analyzed the genomic neighborhood of R.HphI homologs and found that putative nucleases accompanied by a DNA methyltransferase (i.e. predicted REases) do not form a single group on a phylogenetic tree, but are dispersed among free-standing putative nucleases. This suggests that nucleases from the HNH superfamily were independently recruited to become REases in the context of RM systems multiple times in the evolution and that members of the HNH superfamily may be much more frequent among the so far unassigned REase sequences than previously thought.

The physiology of a lepidopteran muscle receptor. II. The function of the receptor muscle.

ABSTRACT Stimulation of the pupal receptor muscle via its nerve resulted in an increased sensory discharge frequency from the MRO. The extent of this excitation was similar in haemolymph and in pupal saline. Stimulation of an MRO whose connexions with the C.N.s. were intact resulted in a transient reflex inhibition of the tonic discharge in the RM of the stimulated receptor. Cessation of sensory stimulation resulted in a transient elevation of RM tone in the stimulated receptor. These changes would tend to protect the sense organ during rapid stretching and would also ‘take up the slack’ when it was released. A slight crossed intrasegmental RM reflex is described which would normally reinforce the ipsilateral reflex. No RM reflexes were found in adjacent segments. These results are discussed in relation to the nature of the deformation occurring in the sensory dendrites, and in relation to the function of RM systems generally.
