Cell walls of group D streptococci. II. Chemical studies on the type 1 antigen purified from the autolytic digest of cell walls.

Autolysis of group D, type I cell walls by an indigenous autolytic enzyme results in the solubilization of the type I antigen. The antigen was purified from the autolytic digest by diethylaminoethyl cellulose chromatography. Chemical analysis of the purified type 1 antigen revealed glucose, glucosamine, galactosamine, rhamnose, ribitol, phosphorus, and residual peptoglycan components. This analysis suggested that the type I antigen is a heteropolymer which may contain a ribitol teichoic acid. The antigen purified by the procedure outlined here is higher in phosphorus content, lower in peptidoglycan, and more reactive serologically than the antigen prepared by methods previously described.

Immunisation with staphylococcal cell walls against mastitis

Three groups of goats were inoculated with purified staphylococcal cell walls from strains chosen on the basis of the configuration of their ribitol teichoic acids; strain 3528 (α-ribitol teichoic acid), 845 Glaxo strain (β-ribitol teichoic acid) and Oxford strain (α+β-ribitol teichoic acids). All three groups, plus an untreated control group were challenged with the Compton BB strain by intramammary perfusion of the right gland. The temperature was measured, and the yield, pH, Whiteside values, bacterial and neurophil counts were determined on the milk samples before and after challenge. Ribitol teichoic acid serum antibody levels were determined by haemagglutination titration up to the time of challenge. The goats in the group receiving 3528 walls (α-ribitol teichoic acid) were protected as shown by their mild reactions to challenge; this correlated with the highest overall ribitol teichoic acid antibody levels. Some protection was observed with the Oxford strain walls (α+β-ribitol teichoic acid), but the goats which received 845 Glaxo strain walls (β-ribitol teichoic acid) were not protected and developed gangrenous mastitis. When all the vaccinated animals are compared as a group with the controls, no protection could be claimed. Further experiments with larger groups of goats are required to determine the role of ribitol teichoic acid antigen in conferring immunity.

Effect of teichoic acid on resistance to the membrane-lytic agent of Streptococcus zymogenes.

Davie, Joseph M. (Indiana University, Bloomington), and Thomas D. Brock. Effect of teichoic acid on resistance to the membrane-lytic agent of Streptococcus zymogenes. J. Bacteriol. 92:1623-1631. 1966.-The resistance of Streptococcus zymogenes to its own lytic agent has been shown to be due to the production of a specific, inhibitory teichoic acid. A survey of streptococcal strains showed that only strains resistant to the lytic agent produced the specific inhibitor. In addition, the inhibitor can be removed from spheroplasts of resistant strains, thereby making them sensitive to the lysin. Throughout the early part of the growth cycle, the inhibitor is associated with the cell and cannot be found in the medium. During late logarithmic phase, however, the inhibitor is released into the medium by the cells, and therefore is a contributing factor to the apparent lability of the lytic agent. The purified, inhibitory teichoic acid contains ribitol, phosphate, glucose, and d-alanine. The alkaline lability of the biological activity of the teichoic acid was correlated with the hydrolysis of the d-alanine. A streptococcal strain which is sensitive to the membrane-lytic agent produced an inactive ribitol teichoic acid which lacks the ester-linked d-alanine, whereas a lysin-resistant mutant of this strain produces a teichoic acid which contains d-alanine and which has inhibitory activity.

Synthesis of teichoic acids: VI. The formation of multiple wall polymers in Bacillus subtilis W-23

The existence of two ribitol teichoic acids (polyribitol phosphate and glucosyl polyribitol phosphate) in the wall of Bacillus subtilis W-23 has been demonstrated by immunological means. Some of the properties of the UDP- d-glucose polyribitol phosphate glucosyl transferase of this organism have been described, and in particular it has been shown that this enzyme will use the polyribitol phosphate from the cell wall as a glucosyl acceptor. These data have suggested the presence of two distinct teichoic acid-synthesizing systems in this organism.

Cell-wall deficiencies in L-forms of Staphylococcus aureus.

SUMMARY: Three strains of penicillin-induced and one strain of D-cycloserine-induced L-forms of Staphylococcus aureus were investigated for components of the normal bacterial cell wall. None of the 4 strains was found to contain more than 0.008% of its dry weight as muramic acid, indicating the lack of mucopeptide. These forms also lacked the cell-wall ribitol teichoic acid as shown by the failure to yield any material containing ribitol. Neither muramic acid nor ribitol was detected as acid-soluble nucleotides in the L-forms, and teichoic acid was not released into the supernatant medium. Serological evidence for polysaccharide A was not found, but there was serological identity between extracts of membranes obtained from staphylococci and extracts of membranes isolated from the L-forms.

1. Ribitol teichoic acids prepared by fractional precipitation of trichloroacetic acid extracts of bacterial cell walls are essentially undegraded and have similar chain length to the teichoic acid originally present in the walls. 2. The chain length of teichoic acid can be determined directly, without prior extraction from the wall. Accurate values have been obtained by measurement of the formaldehyde produced by oxidation of walls with periodate. Less accurate values have been derived from the amount of inorganic phosphate formed by heating walls at pH4. 3. The relative amounts of N-acetylglucosaminylribitol and its mono- and di-phosphates produced by heating walls of Staphylococcus aureus with alkali agree with the amounts calculated for the hydrolysis of teichoic acid having the chain length determined by other methods. 4. Chemical considerations indicate that the linkage between teichoic acid and the wall may involve a phosphoramidate bond between the terminal phosphate of the teichoic acid and one of the amino groups in the glycosaminopeptide.

The Heat-Labile Serum Factor Associated with Intracellular Killing of Staphylococcus Aureus

Abstract The presence of fresh serum has been shown to be required for rapid phagocytosis and intracellular killing of virulent staphylococci by human blood leukocytes (1, 2). When human serum is subjected to heating, it no longer promotes killing but still causes demonstrable phagocytosis, though slower in rate and reduced in amount. The phagocytosis-promoting ability of human serum is apparently dependent upon the presence of antibody against the staphylococcal surface antigen or antigens and is removed by adsorption with ribitol teichoic acid isolated from the staphylococcal cell wall (3). The present report deals with the results of our study on the heat-labile serum factor responsible for rapid phagocytosis and intracellular killing of staphylococci. MATERIALS AND METHODS. Human serum and leukocytes. Fresh samples of adult human blood were obtained from healthy subjects by venipuncture. Leukocytes were prepared by dextran sedimentation and were washed and resuspended in Hanks' solution.

STUDIES ON THE CHEMISTRY AND IMMUNOCHEMISTRY OF CELL WALLS OF STAPHYLOCOCCUS AUREUS

The cell walls of an 80/81 strain of Staphylococcus aureus (NYH-6) contain alanine, glycine, glutamic acid, lysine, muramic acid, glucosamine, and ribitol phosphate. 94 per cent of the phosphorus and 41 per cent of the glucosamine are removed by extraction of the cell walls with hot 5 per cent TCA, but significant amounts of the other constituents are not extracted by this procedure. The residue after hot TCA extraction (mucopeptide) is susceptible to lysozyme whereas the intact cell walls are resistant. Staphylococcus aureus cell walls are agglutinated by S. aureus antisera. Agglutination of the cell walls of one S. aureus strain is inhibited by absorption of antisera with cell walls of other S. aureus strains but not by absorption with S. albus cell walls. The ribitol teichoic acid can be isolated from cold TCA extracts of the cell walls. This compound consists almost entirely of ribitol phosphate and glucosamine. The isolated teichoic acid of strain NYH-6 is readily fixed to tanned sheep erythrocytes and these sensitized cells are agglutinated by S. aureus antisera. Cold TCA extracts of cell walls of other strains of S. aureus inhibit hemagglutination whereas extracts of S. albus walls do not. Studies on the inhibition of both hemagglutination and precipitation indicate that the antigenic determinant of S. aureus NYH-6 teichoic acid is beta-N-acetylglucosamine.

The occurrence and location of teichoic acids in lactobacilli.

SUMMARY: The walls and cell contents of a number of lactobacilli were prepared by mechanical disruption of the organisms followed by differential centrifugation. The nature of the teichoic acid present in the separated fractions was determined by extracting it with dilute trichloroacetic acid, precipitating with ethanol and identifying the precipitated polymer by hydrolysis to ribitol or glycerol phosphates and other recognizable degradation products. All the organisms contained a glycerol teichoic acid within the cell. This and similar observations with other bacteria indicates that glycerol teichoic acids play an important part in cellular metabolism. Both glycerol and ribitol teichoic acids occur in walls. The presence and type of teichoic acid in the walls correlate with the serological behaviour of lactobacilli and provide a useful means of classification. It is suggested that teichoic acids themselves may possess antigenic properties.
