Nitrate Uptake and Primary Production Along the Amazon River Plume Continuum

Abstract The Amazon River plume (ARP) has been shown to support high rates of nitrogen fixation and primary production. However, nitrogen fixation alone cannot account for total primary production determined in the region, hinting that other nitrogen uptake processes might play a role. For the first time, we measured nitrate uptake rates in the ARP during three cruises in May 2018, June 2019 and April/May 2021, along with primary production rates and an analysis of phytoplankton community composition via high performance liquid chromatography. Based on a classification according to the salt content the region was divided into estuarine (ES), mesohaline (MH) and oceanic (OC) stations. Primary production was light limited near the river mouth at ES stations and was maximal off the coasts of French Guiana and Suriname, where also nitrate uptake was highest with rates of 11.4 mmol m−2 d−1. The role of eddies pinching off a deflecting plume are discussed as possible reason for higher nutrient concentrations at the MH stations. Surprisingly, at most MH stations north of 5°N, nitrate uptake rates were low despite the presence of sufficient substrate concentration (up to 1.44 μM nitrate). Diatoms, dinoflagellates or Synechococcus sp. dominated phytoplankton communities. OC stations showed lowest productivity rates in accordance with oligotrophic conditions. However, rates seem to be sufficient to completely deplete the remaining riverine nitrate, preventing its export to the open ocean.

Phosphate Influx and Dust Deposition Create Zonal and Meridional Biogeochemical Gradients in Trichodesmium Abundance

Abstract Trichodesmium plays a key role in the biogeochemical cycling of carbon, nitrogen and phosphorus in the Tropical Atlantic Ocean. A complex interplay of physicochemical factors control the growth of Trichodesmium. However, owing to the large spatial and temporal variability, the relative influence of these factors in controlling Trichodesmium distribution and abundance remains unclear. In this study, we examined the basin‐scale distribution pattern of Trichodesmium in the upper 200 m water column of the Atlantic Ocean (25°N–30°S and 70°W–20°E) using a large data set (n = 33,235) and tried to constrain the distribution based on various physicochemical parameters. We suggest that the combined effect of warm temperatures and phosphate (PO43−) availability determines the zonal spatial extent and the abundance of Trichodesmium in the Tropical North Atlantic Ocean. However, the availability of dissolved iron, along with high sea surface temperatures and meteorological parameters such as the wind direction and precipitation, likely govern the meridional distribution of Trichodesmium across the Atlantic Ocean. Excess PO43− at the surface rules out the possibility of PO43− limitation in regulating the meridional distribution of the Trichodesmium. Depth‐integrated nitrogen fixation rates, based on a multiple linear regression, vary from 0.07 to 306 μmol N m−2 d−1. The presence of Trichodesmium colonies down to a depth of 200 m and the depth‐integrated nitrogen fixation rates reflect the pivotal role of Trichodesmium in the nitrogen budget of this region.

Nitrogen fixation rate and phosphorus enrichment effects on diazotrophic cyanobacteria in the Gulf of Riga

In eutrophied marine systems such as the Baltic Sea, diazotrophic cyanobacteria have the potential to add additional bioavailable nitrogen (N) to the system through fixation of atmospheric dinitrogen (N2). However, their growth is regarded to be limited by phosphorus availability (P). This study investigates the response of two cyanobacteria species, Aphanizomenon flosaquae and Nodularia spumigena, collected from the Gulf of Riga under different environmental conditions to a short-period dissolved inorganic phosphorus (DIP) enrichment. The samples were collected during the summer cyanobacterial bloom of 2022 in the central region of the Gulf of Riga. Contrary to expectations, neither species demonstrated a significant increase in biomass. The study also established that N2-fixation rates did not correlate directly with the total diazotrophic cyanobacteria biomass, but showed a significant correlation with heterocyst presence in both species addressed during this study. The findings suggest the influence of additional factors beyond DIP availability on the N2-fixing cyanobacteria growth in the Gulf of Riga.

Integrated Proteomics and Metabolomics Reveal Altered Metabolic Regulation of Xanthobacter autotrophicus under Electrochemical Water-Splitting Conditions.

Biological-inorganic hybrid systems are a growing class of technologies that combine microorganisms with materials for a variety of purposes, including chemical synthesis, environmental remediation, and energy generation. These systems typically consider microorganisms as simple catalysts for the reaction of interest; however, other metabolic activity is likely to have a large influence on the system performance. The investigation of biological responses to the hybrid environment is thus critical to the future development and optimization. The present study investigates this phenomenon in a recently reported hybrid system that uses electrochemical water splitting to provide reducing equivalents to the nitrogen-fixing bacteria Xanthobacter autotrophicus for efficient reduction of N2 to biomass that may be used as fertilizer. Using integrated proteomic and metabolomic methods, we find a pattern of differentiated metabolic regulation under electrochemical water-splitting (hybrid) conditions with an increase in carbon fixation products glycerate-3-phosphate and acetyl-CoA that suggests a high energy availability. We further report an increased expression of proteins of interest, namely, those responsible for nitrogen fixation and assimilation, which indicate increased rates of nitrogen fixation and support previous observations of faster biomass accumulation in the hybrid system compared to typical planktonic growth conditions. This work complicates the inert catalyst view of biological-inorganic hybrids while demonstrating the power of multiomics analysis as a tool for deeper understanding of those systems.

More diverse rhizobial communities can lead to higher symbiotic nitrogen fixation rates, even in nitrogen-rich soils.

Symbiotic nitrogen (N) fixation (SNF) by legumes and their rhizobial partners is one of the most important sources of bioavailable N to terrestrial ecosystems. While most work on the regulation of SNF has focussed on abiotic drivers such as light, water and soil nutrients, the diversity of rhizobia with which individual legume partners may play an important but under-recognized role in regulating N inputs from SNF. By experimentally manipulating the diversity of rhizobia available to legumes, we demonstrate that rhizobial diversity can increase average SNF rates by more than 90%, and that high rhizobial diversity can induce increased SNF even under conditions of high soil N fertilization. However, the effects of rhizobial diversity, the conditions under which diversity effects were the strongest, and the likely mechanisms driving these diversity effects differed between the two legume species we assessed. These results provide evidence that biodiversity-ecosystem function relationships can occur at the scales of an individual plant and that the effects of rhizobial diversity may be as important as long-established abiotic factors, such as N availability, in driving terrestrial N inputs via SNF.

Heterotrophic activity, primarily driven by sulfate-reducing prokaryotes, has traditionally been linked to nitrogen fixation in the root zone of coastal marine plants, leaving the role of chemolithoautotrophy in this process unexplored. Here, we show that sulfur oxidation coupled to nitrogen fixation is a previously overlooked process providing nitrogen to coastal marine macrophytes. In this study, we recovered 239 metagenome-assembled genomes from a salt marsh dominated by the foundation plant Spartina alterniflora, including diazotrophic sulfate-reducing and sulfur-oxidizing bacteria. Abundant sulfur-oxidizing bacteria encode and highly express genes for carbon fixation (RuBisCO), nitrogen fixation (nifHDK) and sulfur oxidation (oxidative-dsrAB), especially in roots stressed by sulfidic and reduced sediment conditions. Stressed roots exhibited the highest rates of nitrogen fixation and expression level of sulfur oxidation and sulfate reduction genes. Close relatives of marine symbionts from the Candidatus Thiodiazotropha genus contributed ~30% and ~20% of all sulfur-oxidizing dsrA and nitrogen-fixing nifK transcripts in stressed roots, respectively. Based on these findings, we propose that the symbiosis between S. alterniflora and sulfur-oxidizing bacteria is key to ecosystem functioning of coastal salt marshes.

Short-term fate of nitrogen fixed by moss-cyanobacteria associations under different rainfall regimes

Nitrogen (N) fixation by moss-cyanobacteria associations has been recognized as an important N input pathway in many ecosystems from arctic tundra to tropical forests. However, the transfer of fixed N2 from mosses to the soil as well as the effects of rainfall frequency and volume on this N transfer has hardly been studied – even though mosses can leach nutrients upon rewetting. In this study, we investigated the transfer of fixed N2 by moss-cyanobacteria associations in one month under four watering regimes with a combination of high and low volume and frequency. For this, we used two morphologically similar moss species collected from ecosystems with different climate and N availability (subarctic - Hylocomium splendens; and tropical - Thuidium delicatulum). Acetylene reduction assays were conducted as a measure of N2 fixation rates in mosses, and 15N-N2 tracing was used to follow the fixed N2 from moss to the underlying substrate. Nitrogen fixation rates were higher in T. delicatulum than in H. splendens, but rainfall volume and frequency did not show strong effects on N2 fixation rates. Nonetheless, the extent of N leached from mosses was more sensitive to an increase in rainfall volume than to an increase in frequency, and more N was lost from T. delicatulum under high volume precipitation than from H. splendens. Both total nitrogen and 15N enrichment results demonstrate that the fixed N2 was mostly stored in moss tissues with less than 1 % leached to the substrate. Our results show that both moss species retain almost all fixed N2 within their tissues under small rainfall disturbances within one month, while increased N availability under higher precipitation volume renders some moss species an important N source for the soil.

Short-term nitrogen addition reduces soil microbial nitrogen fixation rate in subtropical Pinus taiwanensis and Castanopsis faberi forests

Biological nitrogen (N) fixation is an important source of N in terrestrial ecosystems, but the response of soil microbial N fixation rate to N deposition in different forest ecosystems still remains uncertain. We conducted a field N addition experiment to simulate atmosphere N deposition in subtropical Pinus taiwanensis and Castanopsis faberi forests. We set up three levels of nitrogen addition using urea as the N source: 0 (control), 40 (low N), and 80 g N·hm-2·a-1(high N) to examine the chemical properties, microbial biomass C, enzyme activities, and nifH gene copies of top soils (0-10 cm). We also measured the microbial N fixation rate using the 15N labeling method. Results showed that N addition significantly reduced the soil microbial N fixation rate in the P. taiwanensis and C. faberi forests by 29%-33% and 10%-18%, respectively. Nitrogen addition significantly reduced N-acquiring enzyme (i.e., β-1, 4-N-acetylglucosaminidase) activity and nifH gene copies in both forest soils. There was a significant positive correlation between the microbial N fixation rate and soil dissolved organic C content in the P. taiwanensis forest, but a significant negative relationship between the rate of soil microbial nitrogen fixation and NH4+-N content in the C. faberi forest. Overall, soil microbial N fixation function in the P. taiwanensis forest was more sensitive to N addition than that in the C. faberi forest, and the factors affecting microbial N fixation varied between the two forest soils. The study could provide insights into the effects of N addition on biological N fixation in forest ecosystems, and a theoretical basis for forest management.

Nitrogen-fixing bacterial communities differ between perennial agroecosystem crops.

Biocrusts, common in natural ecosystems, are specific assemblages of microorganisms at or on the soil surface with associated microorganisms extending into the top centimeter of soil. Agroecosystem biocrusts have similar rates of nitrogen (N) fixation as those in natural ecosystems, but it is unclear how agricultural management influences their composition and function. This study examined the total bacterial and diazotrophic communities of biocrusts in a citrus orchard and a vineyard that shared a similar climate and soil type but differed in management. To contrast climate and soil type, these biocrusts were also compared with those from an apple orchard. Unlike natural ecosystem biocrusts, these agroecosystem biocrusts were dominated by proteobacteria and had a lower abundance of cyanobacteria. All of the examined agroecosystem biocrust diazotroph communities were dominated by N-fixing cyanobacteria from the Nostocales order, similar to natural ecosystem cyanobacterial biocrusts. Lower irrigation and fertilizer in the vineyard compared with the citrus orchard could have contributed to biocrust microbial composition, whereas soil type and climate could have differentiated the apple orchard biocrust. Season did not influence the bacterial and diazotrophic community composition of any of these agroecosystem biocrusts. Overall, agricultural management and climatic and edaphic factors potentially influenced the community composition and function of these biocrusts.

Polyglycerol citrate: A novel coating and inoculation material for soybean seeds

The microbial inoculation of legumes such as soybeans is crucial for thriving plant growth due to symbiotic nitrogen (N) fixation and biological plant N fertilization. Soybean requires microbial pre-inoculation before sowing using the rhizobia strain Bradyrhizobium japonicum. Peat is typical for this purpose, although not sustainable since it is a finite resource. Here, we propose a straight-forwarding route to prepare and apply polyglycerol-citrate polymer (PGC), a biodegradable and fully renewable polymer, as a carrier for Bradyrhizobium japonicum inoculants for soybeans. This novel eco-friendly polymer combines the advantages of a polymeric, water-soluble structure based on biopolymers, which can protect the inoculant cells during the seed inoculation process, with protective properties of glycerol for bacterial cells and the contribution of citric acid for metabolic processes. A greenhouse study was conducted using soybean seeds coated with three different proportions of PGC with B. japonicum planted in a sand substrate free of external interference. Comparative results of N content and δ15N signature in soybean plant parts calculated from the natural abundance method associated with viability tests showed equal or superior symbiotic performance and nitrogen fixation rates to peat-based inoculants, considered the gold-standard carrier for inoculants. It ensured the shelf life of the inoculant formulations, offering convenience for farmers and environmental benefits through reduced fertilization.

Nitrogen fixation‐driven carbon sequestration: Brief communication

Abstract Prior to the commercial nitrogen fertilizer production in 1919, all the world's terrestrial and aquatic carbon was supported by nitrogen fixation. Annual N deposition to semi‐arid lands and temperate forests is less than 5 kg/ha‐year and 10 kg/ha, respectively. Plant and soil C/N ratios range from 9.9 to 29.8 and 9 to 14, respectively. In an equilibrium, sustainable ecosystem where N is not removed from soil pools and is only dependent on annual N inputs, maximum C sequestration rates are approximately 3.25 kg to 46 kg/ha for arid ecosystems and 23 to 101 kg/ha for forest ecosystems. Commercial N applications range from approximately 70 to 160 kg/ha‐year. Managed nitrogen fixation rates range from approximately 50 to 130 kg N/ha‐year. For each additional kg N‐entering forests, the additional C is approximately 13 kg. N‐fixing plants range from alder and lupines in the arctic, to Prosopis and Acacias in semi‐arid lands and the large trees Inga and Pentaclethra in tropical rainforests. If N‐fixing plants achieving 50 kg N/ha‐year were planted on the equivalent of a 447 km square, on six continents, the IEA target of 1.7Gt CO2 (0.72 Gt of carbon) capture capacity by 2030 https://www.ief.org/news/whats‐the‐target‐for‐carbon‐sequestration‐and‐how‐do‐we‐get‐there could be achieved.

Effect of Water Sources on the Growth Performance and Chlorophyll Content of Azolla pinnata R.Br.


 
 
 Azolla pinnata R.Br. is an aquatic fern that grows on the surface of water bodies. The unique quality of A. pinnata is its symbiotic relationship with a prokaryotic cyanobacterium (Anabaena azollae), which confers a high rate of nitrogen fixation. It is important to find the growing conditions of Azolla in different water sources for its optimum biomass production for different usages such as biofertilizers, animal feeds, water purification and biofuel production. This preliminary study was conducted to investigate the effect of water sources on the growth performance (shoot biomass, root biomass, root length), and the chlorophyll content of Azolla. Presence of Anabaena in Azolla fronds was monitored by light microscope and number of filaments were counted per leaflet. The experiment utilized three types of water sources (tap water as control (T1), lake water (T2) and cattle manure mixed tap water (T3), in plastic buckets with 40 cm in diameter and 25 cm in height. Each treatment was triplicated in a completely randomized design and total biomass was collected weekly. Data were statistically analyzed using Minitab 17. The highest biomass of Azolla was obtained in T3 from the first week until 8th week and it was higher from T1 and T2. The highest chlorophyll content was obtained in T3 (6.4 SPAD value) showed a significant difference from T1 and T2 (p<0.05). The dry weight of roots and root length in T1 showed a significant difference from T2 and T3 (p<0.05). The highest root length was observed as 5 cm in T1 indicating that roots may be in search of nutrients and the lowest root length was observed as 0.8 cm in T3 with high nutrient concentrations in water. The highest number of Anabaena was observed in the leaflet of Azolla fronds in T3 which in turn increase N content in Azolla plants. It reveals that application of cattle manure in tap water significantly affects the plant biomass and the chlorophyll content of Azolla. Further studies are needed to conduct in large scale growing facilities with different nutrient concentrations of water sources to find sustainable and optimum growing conditions for Azolla and the nutrient concentrations of the plants.

Insights into Genetic and Physiological Characteristics of Clover Rhizobia in Afghanistan Soils

Livestock production in Afghanistan highly relies on grazing and clover feed, which is a key component of pastures and forage crops. This study elucidated the genetic diversity of clover-nodulating rhizobia in different ecological regions and their effects on clover growth. A total of 57 rhizobia were isolated and their genetic diversities were studied through 16S rRNA and nifD genes. The isolates were inoculated to clover (Afghan local variety), to investigate the potential of nitrogen fixation and influences of clover growth. The 16S rRNA gene analysis showed two distinct groups of Rhizobium (94.7%) and Ensifer (5.3%) species. The nifD phylogenetic relationship revealed a high similarity to Rhizobium and a novel lineage group close to Rhizobium leguminosarum species. In the plant test, different genotypes significantly (p < 0.01) exhibited an increase in plant biomass production, compared to the un-inoculated plants. Among genotypes, the highest plant biomass was recorded in PC8 (1769.0 mg/plant) and PC9 (1409.2 mg/plant) isolates as compared to un-inoculated plants (144.0 mg/plant). Moreover, these isolates showed maximum nitrogen fixation rates of 8.2 and 6.5 µM/plant, respectively. These isolates were identified as the most promising rhizobial strains for developing biofertilizers in the context of Afghanistan.
