Infection of turnip protoplasts with radish mosaic virus and its RNA.

Conditions for infection of turnip protoplasts with radish mosaic virus (RaMV) and its RNA were investigated. Up to 90% of protoplasts were infected with RaMV in the absence of poly-L-ornithine (PLO) when protoplasts were inoculated with 10μg/ml RaMV in 50mM K-citrate-phosphate buffer, pH5.0. About 35% of protoplasts were infected with 0.5μg/ml RaMV RNA together with 1μg/ml PLO in 25mM phosphate buffer, pH5.8. Few protoplasts, however, were infected with RaMV RNA in the absence of PLO, and the efficiency of infection was greatly enhanced by the addition of PLO. RaMV antigen was detectable 16hr after inoculation and the percentage of fluorescing protoplasts continued to increase up to 36hr. The number of virus particles was estimated to be 1.5×105 per infected protoplast at 48hr after inoculation.

In Vitro Replication of Cowpea Mosaic Virus RNA III. Template Recognition by Cowpea Mosaic Virus RNA Replicase

Cowpea mosaic virus (CPMV) RNA replicase has been purified about 200-fold from CPMV-infected Vigna unguiculata leaves. Optimal reaction conditions for replicase activity have been established that allow RNA synthesis to proceed for at least 15 h. Using a polymerase assay under conditions optimal for CPMV RNA-directed RNA synthesis, all natural RNA species tested appeared to be able to direct the incorporation of labeled ribonucleotides, whereas synthetic homoribopolymers were either inactive or only slightly active. Using a nitrocellulose membrane filter assay to measure complex formation between the replicase preparation and various RNA species, all natural RNA species tested, except that of the comovirus radish mosaic virus, appeared to be unable to compete with (32)P-labeled CPMV RNA in binding to replicase. We propose that CPMV replicase is actually template specific but does not display this property in a polymerase assay, since labile complexes between heterologous templates and replicase become stabilized by the formation of phosphodiester bonds. From homoribopolymer competition binding experiments we conclude that the polyadenylic acid on the CPMV genome might be a part of the replicase binding site.

Replication of pea enation mosaic virus RNA in isolated pea nuclei

Isolated nuclei from healthy pea plants were primed with pea enation mosaic virus (PEMV), southern bean mosaic virus (SBMV), radish mosaic virus (RdMV), tobacco mosaic virus (TMV), PEMV RNA, SBMV RNA, RdMV RNA, or TMV RNA. RNA replication occurred only with PEMV RNA and not with intact PEMV or any of the other viruses or RNAs, as judged by ensuing actinomycin D-insensitive polymerase activity. Molecular hybridization experiments showed that some of the product of the polymerase was PEMV-specific (-)RNA. The substrate and ionic requirements of this polymerase were the same as those for the RNA-dependent RNA polymerase present in nuclei isolated from PEMV-infected pea plants. No virus particles could be recovered from nuclei primed with PEMV RNA. These results are discussed in relation to the possible mechanism for in vivo infection of pea cells.

Genetic Complementation between Middle and Bottom Components of Two Strains of Radish Mosaic Virus

SUMMARY Kale virus (KV), turnip virus (HZ) and radish mosaic virus (RMV) are slightly related serologically to cowpea mosaic virus. Purified preparations comprize, top, middle and bottom components, of which only the last two contain nucleic acid. The nucleoprotein components of each virus were separated by centrifuging twice through a sucrose gradient in an MSE BXIV zonal rotor. Sometimes this was followed by centrifuging to equilibrium in a density gradient of rubidium bromide. Neither nucleoprotein component was infective alone, but infectivity was restored when each was mixed with the other component. Mixtures in which the two components came from different viruses were infective only when they were made with KV and HZ. Serologically these two viruses are more closely related than either is to RMV. KV differed from HZ by two characters related to the coat protein. KV particles regularly formed aggregates of 12 particles and contained antigens that were not present in HZ. Single lesion isolates from mixtures of bottom component from one strain and middle component from the other did not form aggregates and lacked the antigens present in KV. Like cowpea mosaic virus, KV and HZ each have two coat proteins, and our results can be explained if each of the four virus components codes for a different coat protein. Therefore, the coat protein of the virus is coded by the nucleic acid of both middle and bottom components.

SUMMARY Strains KV and HZ of radish mosaic virus when centrifuged to equilibrium in caesium chloride gave four bands, although they have only two nucleoprotein components. However, when centrifuged to equilibrium in rubidium bromide, HZ gave only two major bands and KV gave these and several extra small bands, presumably because this strain contains aggregates of 12 particles derived from all 3 components and therefore having different densities from the nucleoprotein components.

A strain of radish mosaic virus occurring in turnip in Yugoslavia

SUMMARYFields of turnip (Brassica rapa L. var. rapa) in Yugoslavia often contained plants infected with radish mosaic virus (RaMV). Yugoslav isolates resemble the type strain in host range, symptoms, physical properties, particle morphology and flea‐beetle transmission and are serologically closely related to the type strain, but did not infect radish and are designated as the European strain. Light microscopy of infected turnip and some other cruciferous species revealed the characteristic inclusion bodies in the cytoplasm.

SUMMARY Turnip cells infected with radish mosaic virus contain characteristic large vesiculated cytoplasmic inclusion bodies, visible by light and electron microscopy. They consist of spherical aggregates of virus particles, sometimes in a crystalline array, embedded in a membranous mass. The presence of active dictyosomes and abundant endoplasmic reticulum in the outer zone of young inclusions suggests that these organelles are involved in the formation of inclusion bodies.

Inhibitory action of the pressed juice from some plants on which aphids were fed against the infectivity of Tobacco mosaic virus.

An attempt was made to show the inhibitory effect of substances secreted by aphids on feeding plants, upon the infectivity of tobacco mosaic virus (TMV).Pressed juice of tobacco plants affected by TMV, was mixed with pressed sap from Japanese radish, turnip or tobacco, on which a number of aphid individuals belonging to Myzus persicae and Rhopalosiphum pseudobrassicae, had been fed for 24 hours. The virus juice was tested for infectivity by local lesion method using Nicotiana glutinosa. The juice produced local lesions on the half leaves of N. glutinosa far less in number than the control, viz., the virus juice mixed with sap from aphid free plants. When pressed sap from radish infected by radish mosaic virus, was mixed with juice of aphid-infested plants and inoculated immediately to radish seedlings, however, no inhibitory effect upon the virus was observed.
