Assessing ecological responses of exposure to the pyrethroid insecticide lambda-cyhalothrin in sub-tropical freshwater ecosystems

Pyrethroid insecticides are widely detected in aquatic ecosystems due to their extensive use in agriculture and horticulture, which could pose a potential risk to aquatic non-target organisms. While previous ecotoxicological studies have been conducted mainly with standard tests and local species under temperate conditions, scarce information is available on the effects of pyrethroid insecticides on communities and ecosystems under (sub-)tropical conditions. A single application of lambda-cyhalothrin at concentrations of 0, 9, 30, and 100 ng/L was evaluated in outdoor mesocosms under sub-tropical conditions. Lambda-cyhalothrin was found to dissipate rapidly in the water column, with only 11 % and 7 % of the remaining dose measured at 1 and 3 days after application, respectively. Lambda-cyhalothrin concentrations disappeared considerably faster from the water compartment compared to temperate conditions. Consistent decreases in abundance were observed for Lecane lunaris at the medium and higher treatments (NOEC = 9 ng/L) and at the highest treatment (NOEC = 30 ng/L) for Keratella tropica. On the contrary, two taxa belonging to Cladocera (i.e., Ceriodaphnia sp. and Diaphanosoma sp.) showed the most prominent increase in abundance related to the lambda-cyhalothrin treatments. At the community level, a consistent no observed effect concentrations (NOECs) of 9 ng/L could be calculated for the zooplankton community. A marginal significant overall treatment related effect was observed for the macroinvertebrate community. The results of species sensitivity distribution (SSD) analysis based on results of acute toxicity experiments conducted alongside the mesocosm experiment and obtained from the literature indicated that macroinvertebrates from temperate regions may be generally more sensitive than their counterparts in (sub-)tropical regions. Overall, these findings suggest that environmentally relevant concentrations of the pyrethroid insecticide lambda-cyhalothrin may lead to different ecological outcomes in freshwater ecosystems in the (sub-)tropics relative to temperate regions.

Voltage-gated sodium channel gene mutation and P450 gene expression are associated with the resistance of Aphis spiraecola Patch (Hemiptera: Aphididae) to lambda-cyhalothrin.

Aphis spiraecola Patch is one of the most economically important tree fruit pests worldwide. The pyrethroid insecticide lambda-cyhalothrin is commonly used to control A. spiraecola. In this 2-year study, we quantified the resistance level of A. spiraecola to lambda-cyhalothrin in different regions of the Shaanxi province, China. The results showed that A. spiraecola had reached extremely high resistance levels with a 174-fold resistance ratio (RR) found in the Xunyi region. In addition, we compared the enzymatic activity and expression level of P450 genes among eight A. spiraecola populations. The P450 activity of A. spiraecola was significantly increased in five regions (Xunyi, Liquan, Fengxiang, Luochuan, and Xinping) compared to susceptible strain (SS). The expression levels of CYP6CY7, CYP6CY14, CYP6CY22, P4504C1-like, P4506a13, CYP4CZ1, CYP380C47, and CYP4CJ2 genes were significantly increased under lambda-cyhalothrin treatment and in the resistant field populations. A L1014F mutation in the sodium channel gene was found and the mutation rate was positively correlated with the LC50 of lambda-cyhalothrin. In conclusion, the levels of lambda-cyhalothrin resistance of A. spiraecola field populations were associated with P450s and L1014F mutations. Our combined findings provide evidence on the resistance mechanism of A. spiraecola to lambda-cyhalothrin and give a theoretical basis for rational and effective control of this pest species.

Cytotoxic and genotoxic profiles of the pyrethroid insecticide lambda-cyhalothrin and its microformulation Karate® in CHO-K1 cells.

Lambda-cyhalothrin (LCT) and its microformulation Karate® (25 % a.i.) were analysed for its genotoxicity and cytotoxicity on Chinese hamster ovary (CHO-K1) cells. Cytokinesis-block micronucleus cytome (CBMN-cyt) and alkaline single-cell gel electrophoresis (SCGE) bioassays were selected to test genotoxicity. Neutral red uptake (NRU), succinic dehydrogenase activity (MTT) and apoptogenic induction were employed for estimating cytotoxicity. Both compounds were analysed within a concentration range of 0.1-100µg/mL. Only LCT produced a significant augment in the frequency of micronuclei (MNs) when the cultures were exposed to highest concentrations of 10 and 100µg LCT/mL. A noticeable decrease in NDI was observed for cultures treated with LCT at 10 and 100µg/mL. Karate® induced the inhibition of both the proportion of viable cells and succinic dehydrogenase activity and triggered apoptosis 24h of exposition. Whilst an increased GDI in CHO-K1 cells was observed in the treatments with 1-100µg Karate®/mL, the GDI was not modified in the treatments employing LCT at equivalent doses. SCGE showed that Karate® was more prone to induce genotoxic effects than LCT. Only 50µg/mL of Karate® was able to increase apoptosis. Our results demonstrate the genomic instability and cytotoxic effects induced by this pyrethroid insecticide, confirming that LCT exposure can result in a severe drawback for the ecological equilibrium of the environment.

Assessing the potential of biopesticides to control the cabbage stem flea beetle Psylliodes chrysocephala.

Cabbage stem flea beetle (CSFB) is an economically important pest of oilseed rape crops in Europe that was effectively controlled by neonicotinoid insecticide seed treatments until they were banned by the European Union in 2013. Since then, CSFB has been a difficult pest to control effectively, in part due to many populations having developed resistance to pyrethroids, the only authorized insecticides used to control this pest in many countries. Alternative solutions are therefore necessary, such as biopesticides. We tested an entomopathogenic fungus, three entomopathogenic bacteria isolates, two fatty acids and azadirachtin against CSFB adults under laboratory conditions. We also tested the efficacy of the pyrethroid insecticide lambda-cyhalothrin. Fatty acids were effective, with up to 100% CSFB mortality after 24 h. The entomopathogenic fungus Beauveria bassiana resulted in up to 56% mortality 14 days after treatment. Entomopathogenic bacteria formulations and azadirachtin were not effective (<50% and <40% mortality, respectively). Results from a bioassay using lambda-cyhalothrin indicated that the CSFB used in this study were resistant to this insecticide. Entomopathogenic fungi and fatty acids could potentially be used to control CSFB as part of an integrated pest management programme. This study is the first to investigate the efficacy of different biopesticides to control CSFB under laboratory conditions. As such, these biopesticides require further testing to optimise the formulation and application methods, and to assess the impact on nontarget organisms. Finally, efficacy under field conditions must be determined to understand the influence of environmental variables. © 2023 The Authors. Midgut Cell Damage and Oxidative Stress in Partamona helleri (Hymenoptera: Apidae) Workers Caused by the Insecticide Lambda-Cyhalothrin.

The stingless bee Partamona helleri plays a role in pollinating both native and cultivated plants in the Neotropics. However, its populations can be reduced by the pyrethroid insecticide lambda-cyhalothrin. This compound may cross the intestinal barrier and circulate through the hemolymph, affecting various non-target bee organs. The aim of the present study was to assess the extent of cellular damage in the midgut and the resulting oxidative stress caused by lambda-cyhalothrin in P. helleri workers. Bees were orally exposed to lambda-cyhalothrin. The lethal concentration at which 50% of the bees died (LC50) was 0.043 mg a.i. L-1. The P. helleri workers were fed this concentration of lambda-cyhalothrin and their midguts were evaluated. The results revealed signs of damage in the midgut epithelium, including pyknotic nuclei, cytoplasm vacuolization, changes in the striated border, and the release of cell fragments, indicating that the midgut was compromised. Furthermore, the ingestion of lambda-cyhalothrin led to an increase in the activity of the detoxification enzyme superoxide dismutase and the levels of the NO2/NO3 markers, indicating oxidative stress. Conversely, the activities of the catalase and glutathione S-transferase enzymes decreased, supporting the occurrence of oxidative stress. In conclusion, the ingestion of lambda-cyhalothrin by P. helleri workers resulted in damage to their midguts and induced oxidative stress.

Efficacy of ethanolic seed extracts of Annona spp. against Aphis glycines

Soybean aphid, Aphis glycines Matsumura (Hemiptera: Aphididae), is an important pest that can cause significant economic damage to soybean. Synthetic insecticides are currently employed as the main management strategy to control A. glycines; however, over-use of these products has stimulated research in the development of alternative management methods, including botanical derivatives. In this study, we evaluated the bioactivity of Annona mucosa and A. muricata seed extracts and a formulation based on acetogenins (Anosom® 1 EC, 10,000 ppm) compared to the pyrethroid insecticide lambda-cyhalothrin (Warrior II®) on A. glycines. Initially, the insecticidal effect was assessed in vial bioassays and the concentration-response curves were estimated (LC50/LC90 for A. muricata 305.721 ppm/1290.3 ppm and A. mucosa 134.229 ppm/487.890 ppm, respectively). In the systemic effect bioassay, the same extracts were used (at 100, 500, 1,000, 5000 and 10,000 ppm), showing a reduction in the number of insects on the trifoliates. Both extracts at LC50 and LC90 were used in a behavior test, indicating that they are repellent and Warrior II® was attractive for the insects. The LC90 rates were also used in lethal time (LT) and in greenhouse bioassays. The LT50 for A. mucosa and A. muricata extracts was around 20 h. All treatments showed insect mortality rates above 65%, reaching 100% for Warrior II®, followed by the LC90 of A. mucosa (92.1%). The results demonstrate the potential of Annona spp. derivatives for the management of A. glycines in soybean, and these Annonaceae-derived seed extracts could likely serve as an additional IPM tool to improve A. glycines control strategies in organic farming systems.

Identification of 14 glutathione S-transferase genes from Lasioderma serricorne and characterization of LsGSTe1 involved in lambda-cyhalothrin detoxification

In insects, glutathione S-transferases (GSTs) play a pivotal role in the detoxification of a wide range of pesticides. The cigarette beetle, Lasioderma serricorne, is an economically important pest insect of stored products. Recently, pyrethroid insecticides have been used to control this pest. However, little is known concerning the responses and functions of GSTs in L. serricorne under pyrethroid exposure. In this study, transcriptome sequencing was performed on L. serricorne, and a total of 14 GSTs were identified by retrieving the unigene dataset. Of these, 13 predicted GSTs fell into six cytosolic classes, namely, delta, epsilon, omega, sigma, theta, and zeta, and one was assigned to an “unclassified” group. The GST genes were differentially expressed in various larval tissues and at different developmental stages. Exposure to the pyrethroid insecticide lambda-cyhalothrin (LCT) caused oxidative stress in L. serricorne larvae and led to significantly elevated expression levels of six genes, among which LsGSTe1 was the most upregulated. Recombinant LsGSTE1 protein displayed LCT-metabolizing activity. Furthermore, LsGSTE1 protects cells against oxidative stress. Moreover, knockdown of LsGSTe1 by RNA interference dramatically increased the susceptibility of L. serricorne larvae to LCT treatment. The results from this study provide sequence resources and expression data for GST genes in L. serricorne. Our findings indicate that LsGSTE1 plays a dual role in LCT detoxification by metabolizing the pesticide and by preventing LCT-induced oxidative stress. Thus, the LsGSTe1 gene could be used as a potential target for sustainable management of the cigarette beetle.

Joint toxic effects of phoxim and lambda-cyhalothrin on the small yellow croaker (Larimichthys polyactis)

Although pesticides commonly exist as combinations in real-life situations of the aquatic ecosystem, the impact of the toxicity of their mixtures has remained largely unclear. In this study, we investigated the combined effects of two neurotoxic pesticides, including one organophosphate insecticide phoxim (PHO) and one pyrethroid insecticide lambda-cyhalothrin (LCY), on the embryos of the small yellow croaker (Larimichthys polyactis), and their potential pathways. LCY exhibited higher toxicity relative to PHO, with a 72-h LC50 value of 0.0074 mg a.i. L−1, while the corresponding value for PHO was 0.12 mg a.i. L−1. The mixture of PHO and LCY exerted a synergistic effect on the embryos of L. polyactis. The activities of antioxidant enzyme CAT and apoptotic enzyme caspase 3 were substantially changed in most single and combined exposure groups relative to the baseline value. Under both single and combined exposures, more significant changes were found in the mRNA expression of five genes, including the immunosuppression gene ngln2, the apoptosis gene P53, the endocrine system gene cyp19a1b, as well as neurodevelopment genes of ap and acp2, relative to the baseline value. Furthermore, the non-target metabolomic analysis demonstrated that hundreds of differential metabolites, including two bile acids (taurodeoxycholic acid and tauroursodeoxycholic acid), were significantly increased in the exposure groups. The bile acids were closely associated with the gut microbiota, and 16S rRNA sequencing results demonstrated dysfunction of the gut microbiota after exposure, especially in the combined exposure group. Our findings indicated that there might be a potential risk connected to the co-occurrence of these two pesticides in aquatic vertebrates. Consequently, future ecological risk assessments should incorporate synergistic mixtures because the current risk assessments do not consider them.

New Control Methods Against the Cabbage Stem Flea Beetle in Oilseed Rape Crops

Cabbage stem flea beetle (CSFB) is one of the most damaging pests of oilseed rape (OSR) grown in the UK. An intensive research effort is underway in the UK and throughout OSR growing regions of continental Europe to develop new control methods for this pest. In the first article of a series of three, we review this research and consider approaches to this pest problem that may be available immediately and in the short-term. Agronomic practices (sowing date, seed rate, crop defoliation, companion crops, etc) are being investigated and several of these approaches are already being used on farm. The use of physically acting biopesticides such as fatty acids, while not yet being used may also provide an effective solution and could be available in the short-term. Each of these approaches has the potential to form part of future Integrated Pest Management (IPM) programmes but importantly none should be seen as simple replacements for conventional synthetic insecticides. Solutions that may be available in the medium and longer-term will be discussed in the next articles. The cabbage stem flea beetle (CSFB), Psylliodes chrysocephala (Coleoptera: Chrysomelidae) is one of the most important pests of winter OSR crops in central and northern European countries. The adult beetle causes 'shot holing' feeding damage on cotyledons and true leaves of young winter OSR plants in late summer-early autumn. The larvae feed inside petioles and stems of the plants, causing additional damage. The focus on making food production more environmentally friendly means reduced chemical inputs, protecting nontarget species and overall biodiversity. This led to the withdrawal of systemic synthetic neonicotinoid pesticide seed treatments in many crops (oilseed rape included) in 2013. Since the withdrawal of neonicotinoid seed treatments, there has been a reliance on pyrethroid insecticides. This has been seen with a shift in the use of insecticides in oilseed rape, with CSFB the target for 28% of insecticide applications in 2012 (before neonicotinoid seed treatments were withdrawn) to 75% in 2020. The result of this shift has been the development of resistance to this group of insecticides in CSFB population. This situation has become so severe that some UK populations of CSFB are now 100% resistant to the pyrethroid insecticide lambda-cyhalothrin. These changes have led to the area of oilseed rape grown in the UK being halved (756,000 hectares in 2012 versus 307,000 hectares in 2021), with growers citing CSFB as one of the main reasons for this decrease. At the same time, the number of insecticide spray rounds has increased from two to three between 2012 and 2020, with these applications dominated by the use of pyrethroids. The most widely used pyrethroid insecticide, lambda-cyhalothrin, has also increased as a proportion of the insecticide-treated area from 32% in 2012 to 76% in 2020. Additional surveys by Fera, completed as part of the CropMonitor information service, between 2009 and 2020 show that the number of CSFB larvae per plant increased from an average of 0.5 larvae per plant in the years before the withdrawal of neonicotinoids, to more than 2 larvae per plant the following year in 2014 in East England, and almost 5 larvae per plant in 2019 in the South West of England. Here we present solutions that are currently available or that could be made available in the short-term. We consider how each of these approaches could be used in Integrated Pest Management (IPM) programmes.

Monitoring Resistance of Euschistus heros (Fabricius) (Hemiptera: Pentatomidae) to Insecticides by Using Encapsulated Artificial Diet Bioassay.

The novel ingestion bioassay method was developed for detecting and monitoring resistance of Euschistus heros by encapsulating an artificial liquid diet using Parafilm®. This methodology was compared with the tarsal contact (vial test) and topical application methods for thiamethoxam, imidacloprid, and lambda-cyhalothrin. The best bioassay method for the neonicotinoid insecticides thiamethoxam and imidacloprid was ingestion. For pyrethroid insecticide lambda-cyhalothrin, the best result was obtained by topical application. Using the best bioassay method for each insecticide, the susceptibility to these insecticides was monitored in 30 populations of E. heros collected from soybean crops in Brazil from 2018 to 2020. High variations in susceptibility to thiamethoxam (resistance ratios, 1.6-22 times), imidacloprid (resistance ratios, 1.6-22 times), and lambda-cyhalothrin (resistance ratios, 5-40 times) were detected among the evaluated E. heros populations. In order to monitor the susceptibility of E. heros to insecticides, diagnostic concentrations were defined based on the LC99 of the susceptible reference population: 5.65 µL of a.i./mL for thiamethoxam, 12.45 µL of a.i./mL for imidacloprid, and 0.20 µg of a.i./insect for lambda-cyhalothrin. Subsequently, we select an E. heros strain resistant to neonicotinoid insecticides and another to lambda-cyhalothrin. The resistance ratios obtained after seven selection cycles were 66, 41 and 44 times for thiamethoxam, imidacloprid and lambda-cyhalothrin, respectively.

Evolution of insecticide resistance and its mechanisms in Anopheles stephensi in the WHO Eastern Mediterranean Region

BackgroundWhile Iran is on the path to eliminating malaria, the disease with 4.9 million estimated cases and 9300 estimated deaths in 2018 remains a serious health problem in the World Health Organization (WHO) Eastern Mediterranean Region. Anopheles stephensi is the main malaria vector in Iran and its range extends from Iraq to western China. Recently, the vector invaded new territories in Sri Lanka and countries in the Horn of Africa. Insecticide resistance in An. stephensi is a potential issue in controlling the spread of this vector.MethodsData were collated from national and international databases, including PubMed, Google Scholar, Scopus, ScienceDirect, SID, and IranMedex using appropriate search terms.ResultsIndoor residual spaying (IRS) with DDT was piloted in Iran in 1945 and subsequently used in the malaria eradication programme. Resistance to DDT in An. stephensi was detected in Iran, Iraq, Pakistan, and Saudi Arabia in the late 1960s. Malathion was used for malaria control in Iran in 1967, then propoxur in 1978, followed by pirimiphos-methyl from 1992 to 1994. The pyrethroid insecticide lambda-cyhalothrin was used from 1994 to 2003 followed by deltamethrin IRS and long-lasting insecticidal nets (LLINs). Some of these insecticides with the same sequence were used in other malaria-endemic countries of the region. Pyrethroid resistance was detected in An. stephensi in Afghanistan in 2010, in 2011 in India and in 2012 in Iran. The newly invaded population of An. stephensi in Ethiopia was resistant to insecticides of all four major insecticide classes. Different mechanisms of insecticide resistance, including metabolic and insecticide target site insensitivity, have been developed in An. stephensi. Resistance to DDT was initially glutathione S-transferase based. Target site knockdown resistance was later selected by pyrethroids. Esterases and altered acetylcholinesterase are the underlying cause of organophosphate resistance and cytochrome p450s were involved in pyrethroid metabolic resistance.ConclusionsAnopheles stephensi is a major malaria vector in Iran and many countries in the region and beyond. The species is leading in terms of development of insecticide resistance as well as developing a variety of resistance mechanisms. Knowledge of the evolution of insecticide resistance and their underlying mechanisms, in particular, are important to Iran, considering the final steps the country is taking towards malaria elimination, but also to other countries in the region for their battle against malaria. This systematic review may also be of value to countries and territories newly invaded by this species, especially in the Horn of Africa, where the malaria situation is already dire.

The investigation of the pyrethroid insecticide lambda-cyhalothrin (LCT)-affected Ca2+ homeostasis and -activated Ca2+-associated mitochondrial apoptotic pathway in normal human astrocytes: The evaluation of protective effects of BAPTA-AM (a selective Ca2+ chelator)

Exposure to insecticides has been found to have deleterious effects on human health. Lambda-cyhalothrin (LCT), a mixture of isomers of cyhalothrin, is a pyrethroid insecticide routinely used in pest control programs. LCT was reported to cause neurotoxic effects in various models. However, the mechanism of underlying effect of LCT on cytotoxicity in normal human brain cells is still elusive. This study examined whether LCT affected Ca2+ homeostasis and Ca2+-related physiology in Gibco® Human Astrocytes (GHA cells), and explored whether BAPTA-AM (1,2-bis(2-aminophenoxy)ethane-N,N,N'N'-tetraacetic acid), a selective Ca2+ chelator, has protective effects on LCT-treated GHA cells. The data show that LCT (10–15 μM) concentration-dependently induced cytotoxicity in both GHA cells and DI TNC1 normal rat astrocytes but only induced intracellular Ca2+ concentration ([Ca2+]i) rises in GHA cells. In terms of Ca2+ signaling in GHA cells, LCT-induced [Ca2+]i rises were reduced by removing extracellular Ca2+ and were inhibited by store-operated Ca2+ channel modulators (2-APB, econazole or SKF96365). In Ca2+-free medium, pretreatment with the endoplasmic reticulum Ca2+ pump inhibitor thapsigargin abolished LCT-induced [Ca2+]i rises. Conversely, incubation with LCT abolished thapsigargin-induced [Ca2+]i rises. Regarding cytotoxicity, LCT evoked apoptosis by regulating apoptotic protein expressions (Bax, BCl-2, cleaved caspase-9/-3). This apoptotic response was significantly inhibited by prechelating cytosolic Ca2+ with BAPTA-AM. Together, in GHA cells, LCT induced [Ca2+]i rises by inducing Ca2+ entry via store-operated Ca2+ channels and Ca2+ release from the endoplasmic reticulum. Moreover, BAPTA-AM has a protective effect on inhibiting LCT-activated mitochondrial apoptotic pathway. This study provided new insights into the molecular protective mechanism of LCT-induced cytotoxicity in normal human astrocytes.

Pyrethroid insecticide lambda-cyhalothrin induces hepatic cytochrome P450 enzymes, oxidative stress and apoptosis in rats

This study aimed to examine in rats the effects of the Type II pyrethroid lambda-cyhalothrin on hepatic microsomal cytochrome P450 (CYP) isoform activities, oxidative stress markers, gene expression of proinflammatory, oxidative stress and apoptosis mediators, and CYP isoform gene expression and metabolism phase I enzyme PCR array analysis. Lambda-cyhalothrin, at oral doses of 1, 2, 4 and 8mg/kg bw for 6days, increased, in a dose-dependent manner, hepatic activities of ethoxyresorufin O-deethylase (CYP1A1), methoxyresorufin O-demethylase (CYP1A2), pentoxyresorufin O-depentylase (CYP2B1/2), testosterone 7α- (CYP2A1), 16β- (CYP2B1), and 6β-hydroxylase (CYP3A1/2), and lauric acid 11- and 12-hydroxylase (CYP4A1/2). Similarly, lambda-cyhalothrin (4 and 8mg/kg bw, for 6days), in a dose-dependent manner, increased significantly hepatic CYP1A1, 1A2, 2A1, 2B1, 2B2, 2E1, 3A1, 3A2 and 4A1 mRNA levels and IL-1β, NFκB, Nrf2, p53, caspase-3 and Bax gene expressions. PCR array analysis showed from 84 genes examined (P<0.05; fold change>1.5), changes in mRNA levels in 18 genes: 13 up-regulated and 5 down-regulated. A greater fold change reversion than 3-fold was observed on the up-regulated ALDH1A1, CYP2B2, CYP2C80 and CYP2D4 genes. Ingenuity Pathway Analysis (IPA) groups the expressed genes into biological mechanisms that are mainly related to drug metabolism. In the top canonical pathways, Oxidative ethanol degradation III together with Fatty Acid α-oxidation may be significant pathways for lambda-cyhalothrin. Our results may provide further understanding of molecular aspects involved in lambda-cyhalothrin-induced liver injury.

Lambda-cyhalothrin exposure, mating behavior and reproductive output of pyrethroid-susceptible and resistant lady beetles (Eriopis connexa)

Insecticide resistance is a ubiquitous consequence in arthropod pest species subjected to insecticide use in agricultural fields. The widespread use of insecticides also allows selection of insecticide resistance among non-target arthropod species, such as natural enemies. Nonetheless, the potential consequences of sublethal insecticide exposure in resistant natural enemies are frequently neglected. The detected pyrethroid resistance in the lady beetle Eriopis connexa (Germar) in Brazilian agricultural fields afford the opportunity of assessing the consequences of the sublethal exposure of the broadly used pyrethroid insecticide lambda-cyhalothrin in the mating behavior and reproductive output of susceptible and resistant populations of this species. Survival bioassays with lambda-cyhalothrin allowed estimation of sublethal exposure times at the maximum labeled rate to assess the reproductive consequences of such exposure in pyrethroid-susceptible and resistant strains of E. connexa. Such sublethal exposures led to significant difficulties in female mounting by male of both populations. Pyrethroid exposure also extended the duration of female body tremulation and of coupling, while latency to mate, tremulation, coupling and female shaking to dislodge the males after coupling differed between strains with interacting effect of insecticide exposure only for the latter behavior. As a consequence mainly of latency to mate, progeny production was significantly smaller among pyrethroid-resistant females where lambda-cyhalothrin exposure exhibited a negligible effect. Thus, population rather than exposure itself prevailed in determining reproductive output of E. connexa and pyrethroid resistance incurred in reproductive costs in this species that may counterweight the benefits of its survival.

Pyrethroid insecticide lambda-cyhalothrin and its metabolites induce liver injury through the activation of oxidative stress and proinflammatory gene expression in rats following acute and subchronic exposure.

Lambda-cyhalothrin (LTC) [α-cyano-3-phenoxybenzyl-3-(2-chloro-3,3,3-trifluoro-1-propenyl)-2,2-dimethylcyclo-propanecarboxylate] is a synthetic type II pyrethroid insecticide commonly used in residential and agricultural areas. The potential hepatotoxicity of pyrethroids remains unclear and could easily be assessed by measuring common clinical indicators of liver disease. To understand more about the potential risks for humans associated with LTC exposure, male adult rats were orally exposed to 6.2 and 31.1mg/kg bw of LTC for 7, 30, 45, and 60days. Histopathological changes and alterations of main parameters related to oxidative stress and inflammatory responses in the liver were evaluated. Further, lambda-cyhalothrin metabolites [3-(2-chloro-3,3,3-trifluoroprop-1-enyl)-2,2-dimethyl-cyclopropane carboxylic acid (CFMP), 4-hydroxyphenoxybenzoic acid (4-OH-3-PBA), and 3-phenoxybenzoic acid (3-PBA)] in the liver tissues were identified and quantified by ultra-high-performance liquid chromatography coupled to quadripole time-of-flight mass spectrometry (UHPLC-MS-Q-ToF). Results revealed that LTC exposure significantly increased markers of hepatic oxidative stress in a time-dependent and dose-dependent manner, and this was associated with an accumulation of CFMP and 3-PBA in the liver tissues. In addition, the levels of tumor necrosis factor-α (TNF-α) and interleukin (IL-6 and IL-1β) gene expressions were significantly increased in the liver of exposed rats compared to controls. Correlation analyses revealed that CFMP and 3-PBA metabolite levels in the liver tissues were significantly correlated with the indexes of oxidative stress, redox status, and inflammatory markers in rats exposed to lambda-cyhalothin. Overall, this study provided novel evidence that hepatic damage is likely due to increased oxidative stress and inflammation under the condition of acute and subchronic exposure to lambda-cyhalothrin and that LTC metabolites (CFMP and 3-PBA) could be used as potential biomarker in human biomonitoring studies.

Multiple exposure routes of a pesticide exacerbate effects on a grazing mayfly

Hydrophobic pesticides such as pyrethroid insecticides tend to occur in their soluble form mainly as transient pulses in streams. In addition, they are regularly detected in significant quantities adsorbed to stream sediments and other organic in-stream structures. Consequently, stream biota is likely subjected to pesticide exposure via multiple routes. In this study we aimed at investigating the influence of exposure routes for the pyrethroid insecticide lambda-cyhalothrin on the grazing mayfly Heptagenia sulphurea. Therefore, H. sulphurea was exposed to lambda-cyhalothrin via single- (water or biofilm) or biphasic exposure (water and biofilm) at environmentally realistic concentrations (0, 0.1, 1μgL−1) and exposure duration (2h) in a full factorial design (n=5). Mortality, moulting frequency, and biofilm accrual (proxy for feeding rate) were recorded subsequent to a 7 d post exposure period. Mortality significantly increased and moulting frequency significantly decreased with increasing concentrations of lambda-cyhalothrin in the water phase whereas exposure via biofilm prompted no significant effects on these endpoints (α=0.05). Effect predictions systematically underestimated and overestimated effects for mortality and moulting frequency, respectively. Similarly, mayfly feeding rate was significantly reduced by water phase exposure whereas pre-exposed biofilm did not significantly affect this variable. However, we found a significant but non-systematic interaction between water phase and biofilm exposure on mayfly feeding rate. Our results show that exposure to the same pesticide via multiple exposure routes may increase the magnitude of effects beyond the level predicted from single phase exposures which has clear implications for the aquatic risk assessment of hydrophobic pesticides. However, our results additionally reveal that interactions between pesticide exposure routes may vary between selected dependent variables. We emphasize that unravelling the underlying mechanisms causing these discrepancies in interactive effects between exposure routes is a major aspect that should receive further attention in future research.

Identification of an alternative knockdown resistance (kdr)-like mutation, M918L, and a novel mutation, V1010A, in theThrips tabacivoltage-gated sodium channel gene

Knockdown resistance (kdr) has been identified as a main mechanism against pyrethroid insecticides in many arthropod pests including in the onion thrips, Thrips tabaci. To characterize and identify pyrethroid-resistance in onion thrips in Washington state, we conducted insecticide bioassays and sequenced a region of the voltage gated sodium channel gene from several different T. tabaci populations. Field collected Thrips tabaci were found to have large variations in resistance to the pyrethroid insecticide lambda-cyhalothrin. We identified two single nucleotide substitutions in our analysis of a partial sequence of the T. tabaci voltage-gated sodium channel gene. One mutation resulted in the non-synonymous substitution of methionine with leucine (M918L), which is well known to be responsible for super knockdown resistance in some pest species. Another non-synonymous substitution, a valine (GTT) to alanine (GCT) replacement at amino acid 1010 (V1010A) was identified in our study and was associated with lambda-cyhalothrin resistance. We have characterized a known kdr mutation and identified a novel mutation in the voltage-gated sodium channel gene of Thrips tabaci associated with resistance to lambda-cyhalothrin. This gene region and these mutations are expected to be useful in the development of a diagnostic test to detect kdr resistance in many onion thrips populations.

Survival and behavioural responses of the predatory ladybird beetle,Eriopis connexapopulations susceptible and resistant to a pyrethroid insecticide

The ladybird beetle, Eriopis connexa (Germar) (Coleoptera: Coccinellidae), is one of the commonest predators of aphids (Hemiptera: Aphididae) in the cotton agroecosystem and in many other row and fruit crops in Brazil, and has been introduced into other countries such as the USA for purposes of aphid control. In addition, the boll weevil, Anthonomus grandis Boheman (Coleoptera: Curculionidae) is the most serious cotton pest where it occurs, including Brazil. Controlling boll weevils and other pests such as cotton defoliators still tends to involve the intense application of insecticides to secure cotton production. The pyrethroid insecticide lambda-cyhalothrin (LCT) is commonly used, but this compound is not effective against aphids; hence, a desirable strategy would be to maintain E. connexa populations in cotton fields where LCT is applied. Using populations of E. connexa resistant (Res) and susceptible (Sus) to LCT, we compared behavioural responses on treated cotton plants and under confinement on partially and fully treated surfaces, and assessed the insects' survival on treated plants compared with that of the boll weevil. The E. connexa resistant population caged on treated plants with 15 and 75g a.i. ha-1 exhibited ≫82% survival for both insecticide concentrations compared with ≪3% and ≪17% survival for susceptible E. connexa populations and boll weevils, respectively. The response of E. connexa Res and Sus populations when released, either on the soil or on the plant canopy, indicated avoidance towards treated plants, as measured by elapsed time to assess the plant. When compared with susceptible individuals, resistant ones took longer time to suffer insecticide knockdown, had a higher recovery rate after suffering knockdown, and spent more time in the plant canopy. Based on behavioural parameters evaluated in treated arenas, no ladybird beetles exhibited repellency. However, irritability was evident, with the susceptible population exhibiting greater irritability compared with the resistant population and a subgroup comprising resistant individuals that had recovered from knockdown. The outcomes for the E. connexa Res population indicate a promising strategy for its maintenance when using the insecticide LCT in integrated pest management schemes to control boll weevil or other non-target pest of ladybird beetles in cotton fields.

Combined Effect of Invertebrate Predation and Sublethal Pesticide Exposure on the Behavior and Survival of Asellus aquaticus (Crustacea; Isopoda)

Invertebrate communities of lentic habitats comprise, amongst others, the crustacean Asellus aquaticus (Isopoda) and the turbellarian Dendrocoelum lacteum (Tricladida). Because D. lacteum preferentially preys on A. aquaticus, contaminants introduced into the aquatic environment may affect this predator-prey interaction, finally influencing the performance of the predator. However, no studies investigating implications of organic pollutants on this food web subsystem currently exist. Hence, the present study assessed short-term implications of pesticides with different modes of action, namely, the triazole fungicide tebuconazole and the pyrethroid insecticide lambda-cyhalothrin, during a 72 h trial. The experiments for tebuconazole showed a statistically significant decrease in predatory success of D. lacteum. Lambda-cyhalothrin, in contrast, increased predation success by 40%, which is, however, not statistically significant. Both the decrease and the increase in predation seemed to be primarily driven by an altered activity of the prey A. aquaticus. This may be hypothesized because any shift in the prey's activity influenced its probability to stick to mucus, a viscous substance released by D. lacteum, or to encounter the predator directly.

Induction of micronuclei and nuclear abnormalities in erythrocytes of mosquito fish (Gambusia affinis) following exposure to the pyrethroid insecticide lambda-cyhalothrin

In the present study the induction of micronuclei (MN) and nuclear abnormalities (NA) in erythrocytes of mosquitofish (Gambusia affinis) (Baird & Girard 1853) was studied. Fish were exposed to three different concentrations of lambda-cyhalothrin (LCT) (1×10−4μg/l, 2×10−4μg/l, 4×10−4μg/l) for periods of 6, 12, 24, and 48h. NA (notched, lobed, blebbed nuclei), MN, bi-nucleated cells, and the ratio of polychromatic erythrocytes (PCEs) to normochromatic erythrocytes (NCEs) were evaluated to assess genotoxicity and cytotoxicity. LCT significantly induced MN and NA in erythrocytes of G. affinis. The PCE/NCE ratio was also decreased after 24- and 48-h treatments of 4×10−4μg/l LCT. The results show that LCT has genotoxic and cytotoxic potential on G. affinis.
