What can we learn from 28 years of monitoring of fish tissue polychlorinated biphenyls in Michigan's rivers?

Polychlorinated biphenyls (PCBs) are an important part of chemical legacies in the Laurentian Great Lakes basin. Used in industrial products worldwide, PCBs are now extensively monitored because of their potential toxicity to humans. Fish consumption is a major pathway for exposure. Edible portion (i.e., fish fillet) data from Michigan's fish tissue PCB monitoring program were evaluated using regression statistics, principal component analysis, and t-tests to answer three questions: (1) How do fish tissue total PCB concentrations vary across Michigan's rivers? (2) Are the PCB congener patterns uniformly distributed among tested sites and species? (3) Do monitoring methods limit our ability to discern trends in fish tissue PCB concentrations? Our results indicate that although contaminated sites have been successfully identified, based on higher PCB concentrations in samples from Areas of Concern (AOCs) compared to non-AOC sites, 77% of fish samples from 2010 to 2015 exceeded the safe fish tissue PCB concentration for unrestricted consumption (97 g/day) by sensitive populations. The PCB congener profiles vary among species and locations. Results demonstrate that these data are not useful for supplementing ongoing spatial and temporal trend analysis. Only 15 of the 83 species + waterbody pairs had adequate data for evaluating temporal trends with more than three data points. In general, the trends at each location varied based on the analytical method. Polychlorinated biphenyl (PCB) contamination of subsistence species on Unalaska Island in the Aleutian Archipelago

Polychlorinated biphenyls (PCBs) are a group of synthetic, lipophilic organochlorines that were banned due to their impacts on human and wildlife health and environmental persistence. Although banned, the continued release from pre-banned products allows them to persist at toxic levels in the environment. This is especially the case in lipid rich food webs of the Arctic, where PCBs accumulate due to both long-range atmospheric transport and locally contaminated sites such as formerly used defense (FUD) sites. At the request of the leadership of the Qawalangin Tribe of Unalaska Island in the Aleutian Archipelago, we analyzed PCB concentrations in samples of threespine stickleback (Gasterosteus aculeatus) and subsistence foods (i.e., salmonid species and blue mussels [Mytilus edulis]) collected at both FUD and non-FUD sites. PCBs were extracted from samples using a QuEChERS method. The mean PCB concentrations across all mussel samples was 6.1 ppb; mussels from FUD sites had nearly double the PCB concentrations (7.6 ppb) compared to non-military sites (3.9 ppb), and at two FUD sites the PCB concentrations exceeded safe consumption guidelines. The mean total PCB concentration for fish was 2.8 ppb; fish PCB concentrations were higher at FUD sites (3.2 ppb) compared to non-military sites (1.2 ppb). These results support the need to remediate the FUD sites of “Building 551/T Dock to Airport” and “Delta Western”. More generally, these results provide further evidence of the continued problem of PCB contamination at FUD sites in the Arctic, many of which are co-located with indigenous communities.

Profiles of polychlorinated biphenyls (PCBs) in cement kilns co-processing solid waste

Co-incineration of sewage sludge in cement kilns can be used for its disposal. In the present study, samples were collected from three cement production runs where sewage sludge and other wastes (e.g. municipal solid waste, waste acid and wet sewage sludge) were co-processed. The samples were analyzed for polychlorinated biphenyls (PCBs). The dioxin-like (dl)-PCB concentrations in the stack gases from run 1, 2, and 3 were 344.6, 548.7, and 104.3 pg m−3, respectively. The toxic equivalency (TEQs) values for runs 1, 2, and 3 were 5.6, 8.9, and 0.7 pg TEQ Nm−3, respectively. Calculation of net emissions for the three runs indicated that the co-incineration of other waste in addition to sewage sludge in cement kilns would not increase emission of the dl-PCBs. PCB concentrations in samples from the suspension boiler and humidifier tower, kiln-end bag filter, and cyclone preheater were much higher than those in samples from the kiln head area, indicating that these stages will be important for controlling PCB formation. Chlorinated biphenyl (CB)-77, CB-105 and CB-118 were the major dl-PCB congeners, CB-52, CB-101 were the major indicator PCB congeners, and tetra-CB to hexa-CB were the major homologues for the total input or output materials.

Variability in PCB and OH-PCB serum levels in children and their mothers in urban and rural U.S. communities.

Environmental exposures that affect accumulation of polychlorinated biphenyls (PCBs) in humans are complex and not fully understood. One challenge in linking environmental exposure to accumulation is determining variability of PCB concentrations in samples collected from the same person at different times. We hypothesized that PCBs in human blood serum are consistent from year to year in people who live in the same environment between sampling. We analyzed blood serum from children and their mothers from urban and rural U.S. communities (n = 200) for all 209 PCBs (median ∑PCBs = 45 ng/g lw) and 12 hydroxylated PCBs (median ∑OH-PCBs = 0.09 ng/g fw). A subset of these participants (n = 155) also had blood PCB and OH-PCB concentrations analyzed during the previous calendar year. Although many participants had similar levels of PCBs and OH-PCBs in their blood from one year to the next, some participants had surprisingly different levels. Year-to-year variability in ∑PCBs ranged from −87% to 567% and in ∑OH-PCBs ranged from −51 to 358% (5th–95th percentile). This is the first study to report variability of all PCBs and major metabolites in two generations of people and suggests short-term exposures to PCBs may be a significant component of what is measured in human serum.

Chemical contaminants, health indicators, and reproductive biomarker responses in fish from rivers in the Southeastern United States

Largemouth bass ( Micropterus salmoides) and common carp ( Cyprinus carpio) were collected from 13 sites located in the Mobile (MRB), Apalachicola–Flint–Chattahoochee (ARB), Savannah (SRB), and Pee Dee (PRB) River Basins to document spatial trends in accumulative chemical contaminants, health indicators, and reproductive biomarkers. Organochlorine residues, 2,3,7,8-tetrachlorodibenzo- p-dioxin-like activity (TCDD-EQ), and elemental contaminants were measured in composite samples of whole fish, grouped by species and gender, from each site. Mercury (Hg) and polychlorinated biphenyls (PCBs) were the primary contaminants of concern. Concentrations of Hg in bass samples from all basins exceeded toxicity thresholds for piscivorous mammals (> 0.1 μg/g ww), juvenile and adult fish (> 0.2 μg/g ww), and piscivorous birds (> 0.3 μg/g ww). Total PCB concentrations in samples from the MRB, ARB, and PRB were > 480 ng/g ww and may be a risk to piscivorous wildlife. Selenium concentrations also exceeded toxicity thresholds (> 0.75 μg/g ww) in MRB and ARB fish. Concentrations of other formerly used (total chlordanes, dieldrin, endrin, aldrin, mirex, and hexachlorobenzene) and currently used (pentachlorobenzene, pentachloroanisole, dacthal, endosulfan, γ-hexachlorocyclohexane, and methoxychlor) organochlorine residues were generally low or did not exceed toxicity thresholds for fish and piscivorous wildlife. TCDD-EQs exceeded wildlife dietary guidelines (> 5 pg/g ww) in MRB and PRB fish. Hepatic ethoxyresorufin O-deethylase (EROD) activity was generally greatest in MRB bass and carp. Altered fish health indicators and reproductive biomarker were noted in individual fish, but mean responses were similar among basins. The field necropsy and histopathological examination determined that MRB fish were generally in poorer health than those from the other basins, primarily due to parasitic infestations. Tumors were found in few fish ( n = 5; 0.01%); ovarian tumors of smooth muscle origin were found in two ARB carp from the same site. Intersex gonads were identified in 47 male bass (42%) representing 12 sites and may indicate exposure to potential endocrine disrupting compounds. Comparatively high vitellogenin concentrations (> 0.35 mg/mL) in male fish from the MRB, SRB, and PRB indicate exposure to estrogenic or anti-androgenic chemicals.

Human Blood Monitoring Program in Japan: Contamination and Bioaccumulation of Persistent Organochlorines in Japanese Residents

Concentrations of persistent organochlorines (OCs)-such as polychlorinated biphenyls (PCBs), 1,1,1-trichloro-2,2-bis(p-chlorophenyl)ethane (DDT) and its metabolites (DDTs), hexachlorocyclohexane isomers (HCHs), chlordane compounds (CHLs), hexachlorobenzene, and tris(4-chlorophenyl)methane [TCPMe]-were determined in plasma samples from residents of three sub-metropolitan locations in Japan (Miyako, Saku, and Tottori) for the purpose of studying the geographic variation and specific accumulation of OCs. Residue concentrations of PCBs and DDTs were the highest in samples collected in Saku (400 and 370 ng/g lipid wt, respectively) whereas samples from Miyako contained greater CHL residues (70 ng/g lipid wt) than those from the other two locations. This contamination pattern reflects the historic use of OCs in each area. For the first time, tris (4-chlorophenyl) methane (TCPMe) concentrations were detected in most of the plasma sample analyzed. Concentrations of TCPMe which ranged from <0.1 to 8.1 ng/g lipid wt eight, were lower than those previouly reported in other human tissue. Larger geographic differences in OC accumulation were observed for PCBs and CHLs, whereas DDTs and HCHs exhibited little variability. PCB concentrations in samples from Saku residents were higher than those from residents of countries in the circumpolar Arctic region but lower than those reported for some populations in the United States and Western European countries. Interestingly, CHL residue concentrations in human blood from Japan are among the highest values reported for the countries examined, suggesting continued increased exposure to CHLs of the Japanese population. Time-trend analysis of CHLs in human blood samples from Miyako (Okinawa prefecture) showed that CHL residues have decreased substantially during the last decade, indicating the effect of the official ban of CHLs in 1986 in Japan. Isomer-specific analysis of PCBs revealed lower proportions of higher chlorinated congeners such as hepta- and octachlorobiphenyls in women than in men, suggesting the possibility of preferential elimination of higher chlorinated biphenyls in women. The difference in sex-dependent accumulation of OC compounds in healthy and ill persons was suggested. To our knowledge, this is the first report on the specific accumulation of persistent QCs, including TCPMe, in human blood samples from Japan.

PCBs and organochlorine pesticides (OCPs) in edible fish and shellfish from China

A wide variety of fish and shellfish were collected from local supermarkets of Dalian, Tianjin and Shanghai in China and analyzed for the concentrations of polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs) such as hexachlorobenzene (HCB), hexachlorocyclohexane isomers (HCHs), chlordane compounds (CHLs, including trans-chlordane, cis-chlordane, trans-nonachlor and cis-nonachlor) and dichlorodiphenyltrichloroethane (DDT) and its metabolites (DDTs, including o, p′-DDE, p, p′-DDE, o, p′-DDD, p, p′-DDD, o, p′-DDT and p, p′-DDT). Concentrations of PCBs in samples collected from Dalian, Tianjin and Shanghai ranged from 1.11 to 8.04 ng/g, 1.26 to 5.60 ng/g and 0.83 to 11.4 ng/g on wet weight basis, respectively. The concentrations were lower compared to those in developed countries such as Japan and Italy. Average concentrations of HCB, HCHs, CHLs and DDTs were 0.38, 0.92, 0.47 and 28.9 ng/g on wet weight basis, respectively. The daily intake of PCBs and organochlorine pesticides ingested by people living in these cities in China through fish and shellfish was estimated and compared with those observed in other areas.

Polychlorinated biphenyls in the Mediterranean Sea

Although concentrations of polychlorinated biphenyls (PCBs) in biota, sediment and water of the Mediterranean Sea have been determined, most available data are for samples collected within the narrow coastal zone at relatively few locations. PCB concentrations in samples from the open Mediterranean Sea have not previously been reported. We report here the concentrations of PCBs in surface and sub-surface water from 36 locations. Analysis of our data indicate that there may be some correlation between PCB concentration in Mediterranean seawater and certain demographic and oceanographic features of the region.
