Induction of systemic resistance in rice plants against Burkholderia glumae infection by bioformulation of Streptomyces spp. under greenhouse conditions

The ability of plant growth-promoting Streptomyces (PGPS) against bacterial panicle blight (BPB) disease caused by Burkholderia glumae has been previously examined under laboratory and greenhouse conditions. To further harness their potential, the development of bioformulations containing PGPS is needed. This study investigates the efficacy of Streptomyces bioformulations in suppressing the growth of B. glumae under greenhouse conditions. The powder-based bioformulations of Streptomyces were prepared and characterized based on potential phytotoxic effect and shelf life stability. The efficacy of Streptomyces bioformulations in suppressing BPB disease was assessed according to the expression of Gns1 (β-1,3-glucanase), OsPR1a (pathogenesis-related class 1), Oswrky45 (SA-responsive) and OsJAMYB (JA-responsive) using RT-qPCR. The talc-kaolin-Streptomyces bioformulations (BioSTKR8, BioSJAS2 and BioSTBS5 containing Streptomyces corchorusii TKR8, Streptomyces corchorusii JAS2 and Streptomyces misionensis TBS5, respectively) showed no phytotoxic effect on rice plants. Bioformulations were more stable when stored under refrigeration than at room temperature, with more than 50 % spore viability at 180 days after storage. Treatment with BioSTKR8 (7 %) and BioSTBS5 (10 %) was able to reduce BPB disease severity while increasing rice yield by up to 301 % (BioSTKR8) and 170 % (BioSTBS5) relative to control (100 %). Transcript levels of Gns1, OsPR1a, Oswrky45 and OsJAMYB were up-regulated by the treatment of BioSTKR8 and BioSTBS5 at 7 DPI compared to the control. Results suggest that BioSTKR8 and BioSTBS5 are very promising bioformulations to be utilized as potential plant growth promoters and biological control agents (BCA) for sustainable rice production and BPB disease management.

Streptomyces-mediated growth enhancement and Bacterial Panicle Blight disease suppression in rice plants under greenhouse conditions

Streptomyces corchorusii TKR8, Streptomyces corchorusii JAS2 and Streptomyces misionensis TBS5 were previously obtained from rice fields and have been studied as a biocontrol agent against the causal agent of Bacterial Panicle Blight (BPB) disease on rice, Burkholderia glumae, and rice plant growth promoter. This study evaluated the potential of plant growth-promoting Streptomyces (PGPS) to control B. glumae and promote rice plants’ growth under greenhouse conditions. PGPS were further characterized based on their phenotypic and biochemical differences. Multilocus sequence analysis (MLSA) by amplifying gyrB, rpoB and trpB using PCR was conducted to identify the PGPS further. The antimicrobial activity of PGPS against B. glumae was investigated using a survival assay and microscopic analysis. Result indicates that JAS2 (61.2 %) utilized the highest number of carbohydrates tested, followed by TKR8 (53.1 %) and TBS5 (40.8 %) as analyzed using API 50 CH. Based on MLSA analysis of the concatenated partial sequences (1520 bp) from three housekeeping genes, the neighbor-joining tree identified JAS2 and TKR8 as S. corchorusii. Meanwhile, TBS5 as S. misionensis. Antimicrobial activity of PGPS against B. glumae has found that the supernatant of Streptomyces reduced the survival viability of B. glumae up to 50.7–70.3 %. SEM images showed that substantial morphological changes happened in cell membranes of B. glumae after the Streptomyces treatment. The highest vigor index of inoculated seedlings was determined when rice seeds were treated with a spore suspension of 1 × 107 spore/mL (for JAS2 and TKR8) and 1 × 106 spore/mL (for TBS5). Under greenhouse conditions, Streptomyces-treated plants showed improvement in rice plants' growth and grain yield and reduced the BPB disease severity. Results suggest that the S. corchorusii TKR8, S. corchorusii JAS2 and S. misionensis TBS5 should be promoted as biocontrol agents against B. glumae and bioformulations for rice crops.

Root Morphogenesis of Arabidopsis thaliana Tuned by Plant Growth-Promoting Streptomyces Isolated From Root-Associated Soil of Artemisia annua.

In this study, the capacity to tune root morphogenesis by a plant growth-promoting rhizobacterium, Streptomyces lincolnensis L4, was investigated from various aspects including microbial physiology, root development, and root endophytic microbial community. Strain L4 was isolated from the root-associated soil of 7-year plantation of Artemisia annua. Aiming at revealing the promotion mechanism of Streptomyces on root growth and development, this study first evaluated the growth promotion characters of S. lincolnensis L4, followed by investigation in the effect of L4 inoculation on root morphology, endophytic microbiota of root system, and expression of genes involved in root development in Arabidopsis thaliana. Streptomyces lincolnensis L4 is able to hydrolyze organic and inorganic phosphorus, fix nitrogen, and produce IAA, ACC deaminase, and siderophore, which shaped specific structure of endophytic bacterial community with dominant Streptomyces in roots and promoted the development of roots. From the observation of root development characteristics, root length, root diameter, and the number of root hairs were increased by inoculation of strain L4, which were verified by the differential expression of root development-related genes in A. thaliana. Genomic traits of S. lincolnensis L4 which further revealed its capacity for plant growth promotion in which genes involved in phosphorus solubilization, ACC deamination, iron transportation, and IAA production were identified. This root growth-promoting strain has the potential to develop green method for regulating plant development. These findings provide us ecological knowledge of microenvironment around root system and a new approach for regulating root development.

The Impact of Growth-Promoting Streptomycetes Isolated from Rhizosphere and Bulk Soil on Oilseed Rape (Brassica napus L.) Growth Parameters

Inoculation of Streptomyces to improve oilseed rape (Brassica napus L.) yields and minimise the use of chemical fertilisers is a promising sustainable strategy. In this study, we isolated 72 actinobacterial strains from rhizosphere of oilseed rape and maize and from bulk soil for screening and characterising their antimicrobial activity. Nine promising strains, identified as Streptomyces sp. by morphology, physiological characteristics, and 16S rRNA gene sequencing, were selected for their plant growth-promoting traits and in planta experiments. The actinobacterial strains were positive for IAA production, siderophore production, and HCN production. In planta experiments were conducted by soaking the oilseed rape seeds in the actinobacterial suspension, followed by plant growth under controlled conditions in a cultivate chamber (22–28 °C, 8 h dark/16 h light, constant humidity 80%). We recorded root and shoot length (cm) and seedling fresh weight (g). For most of the abovementioned parameters, a significant enhancement was observed with strain KmiRC20A118 treatment. The length of the root increased by 53.14%, the shoot length increased by 65.6%, and the weight of the fresh plant by 60% compared to the control. The integrated application of PGPS (Plant Growth Promoting Streptomyces) from the rhizosphere of oilseed rape is a promising strategy to improve the growth of oilseed rape.

Plant growth promoting Streptomyces strains are selectively interacting with the wheat cultivars especially in saline conditions

Plant growth promoting (PGP) effect of Streptomyces on wheat growth in different conditions has been mostly reported although mechanisms which caused wheat cultivars differently response to a PGP Streptomyces has been less studied. In this study, the effect of two Streptomyces strains, previously reported as PGPR, on the growth of four salt-sensitive commercial wheat cultivars under normal and saline conditions was investigated. Strain C-2012 differently affected the growth of the cultivars in the normal and stress conditions. Cultivars Gonbad with the highest (63%) and Zarin without increased dry biomass upon C-2012 treatments were selected for further study. Salinity significantly decreased seedling fresh and dry weight, K+ and chlorophyll content and glutathione S-transferase activity. Moreover, the stress increased proline and Na+ content and peroxidase (POX) and ascorbate peroxidase (APX) activity in both cultivars. Strain C-2012, generally, ameliorated the negative effect of the stress with increased chlorophyll and carotenoid and reduced Na+ content and APX and SOD activity in both cultivars, however, its effect on biomass was different. Increase in SOD, APX and POX activities in bacterial inoculated-Zarin, but not Gonbad, under normal conditions suggested that this cultivar may recognize strain C-2012 as a gentle stressor and not as a PGPR. These results showed that the responses of the wheat cultivars to a defined PGPR is different in the physiological, phenotypic and molecular level. Based on the results, the evaluation of the effect of a bio-fertilizer on each wheat cultivar is necessary prior to use in a commercial field.

First insights into salt tolerance improvement of Stevia by plant growth-promoting Streptomyces species.

The present study aimed to investigate the potential of plant growth-promoting rhizobacteria (PGPR) to improve the salt stress and alleviate its impact on Stevia crop plant under different levels of salt concentration. Two Streptomyces spp. isolated from the rhizosphere of halophytic plants (Cucumis sativus L. and Salicornia europaea L.) have shown potential for plant growth promotion in Stevia plant. The streptomycetes isolates were identified by classical microbiological techniques and partial sequencing of 16S rRNA gene as Streptomyces variabilis (4NC) and S. fradiae (8PK). The results have shown that inoculation of Stevia plant by these isolates has enhanced plant growth parameters under applied salt stress. Moreover, total cellular proteins were extracted from the two Streptomyces isolates and SDS-PAGE technique was conducted. Mass spectrometric analysis has identified unique polypeptide of the elongation factor thermos unstable (EF-Tu) indicating the elevation of ribosomal RNA and ribosomal protein genes transcription. On the same context, alleviation of salt stress in Stevia plants inoculated with the two Streptomyces isolates has potentially promoted the accumulation of the major pronounced RuBisCO large subunit protein band detected approximately at 53kDa. These results may give novel insights and accretion our understanding of salinity tolerance mechanisms using PGP streptomycetes to develop resistant sugar crops of highly important economic value. This study has presented the integration of microbiological, biochemical, and molecular techniques to evaluate the effect of salt stress and to assess the level of stress amelioration using PGPR on proteostasis of sugar crops in Egypt.

Biological Control of Potato Common Scab With Rare Isatropolone C Compound Produced by Plant Growth Promoting Streptomyces A1RT.

Potato is prone to many drastic diseases like potato common scab (CS). As no highly effective methods exist for managing CS, this study explored the possibility of using biological control. Ten bacterial strains were isolated from CS-infected potato tubers from four different locations of Punjab, Pakistan, and identified based on biochemical and molecular analysis. Analysis of 16s rDNA sequences amplified by PCR revealed the isolated bacterial strains to be Streptomyces scabies, S. turgidiscabies and S. stelliscabiei. Pathogenic islands were also confirmed among the isolates after identification of txtAB, nec1, and tomA genes with PCR amplification. One strain isolated from soil was antagonistic to the pathogenic Streptomyces spp., and determined to be Streptomyces A1RT on the basis of 16s rRNA sequencing. A methanolic extract of Streptomyces A1RT contained Isatropolone C, which was purified and structurally determined by 1H- and 13C-NMR, 1H/1H-COSY, HMQC, and HMBC techniques. Streptomyces A1RT also produced the plant growth hormone indole-3-acetic acid (IAA) with a titer of 26 μg ml-1 as confirmed by spectrophotometry and HPLC. In a greenhouse assay, disease severity index was established from 0 to 500. Average disease severity indexes were recorded as 63, 130.5, and 78 for Streptomyces scabies, S. turgidiscabies and S. stelliscabiei, respectively. When Streptomyces A1RT was applied in soil that contained one of these pathogenic isolates, the average disease severity indexes were significantly (P < 0.05) reduced to 11.1, 5.6 and 8.4, respectively. A significant increase in tuber weight and shoot development was also observed with the tubers treated with Streptomyces A1RT. The use of the plant growth-promoting Streptomyces A1RT against potato CS thus provides an alternative strategy to control the disease without affecting environmental, plants, animals and human health.

Effect of plant growth-promoting Streptomyces sp. on growth promotion and grain yield in chickpea (Cicer arietinum L).

The physiological and molecular responses of six strains of Streptomyces sp. (CAI-13, CAI-85, CAI-93, CAI-140, CAI-155 and KAI-180), with their proven potential for plant growth-promotion (PGP) in rice were studied to understand the mechanisms causing the beneficial effects. In this investigation, those six strains were evaluated for their PGP capabilities in chickpea in the 2012–13 and 2013–14 post-rainy seasons. All of the Streptomyces sp. strains exhibited enhanced nodule number, nodule weight, root weight and shoot weight at 30 days after sowing (DAS) and pod number, pod weight, leaf area, leaf weight and stem weight at 60 DAS in both seasons over the un-inoculated control. At chickpea crop maturity, the Streptomyces strains had enhanced stover yield, grain yield, total dry matter, pod weight, seed number and seed weight in both seasons over the un-inoculated control. In the rhizosphere, at crop maturity, the Streptomyces strains also significantly enhanced soil biological and mineral nutrient traits including microbial biomass carbon, dehydrogenase activity, total nitrogen, available phosphorous and organic carbon in both seasons over the un-inoculated control. Of the six strains of Streptomyces sp., CAI-85, CAI-93 and KAI-180 were found superior to CAI-155, CAI-140 and CAI-13, in terms of their effects on root and shoot development, nodule formation and crop productivity. Scanning electron microscopy micrographs had revealed the success in colonization of the chickpea roots by all six strains. This investigation further confirms the broad-spectrum of PGP activities by the selected Streptomyces sp.
