Nanobody-Dependent Delocalization of Endocytic Machinery in Arabidopsis Root Cells Dampens Their Internalization Capacity.

Plant cells perceive and adapt to an ever-changing environment by modifying their plasma membrane (PM) proteome. Whereas secretion deposits new integral membrane proteins, internalization by endocytosis removes membrane proteins and associated ligands, largely with the aid of adaptor protein (AP) complexes and the scaffolding molecule clathrin. Two AP complexes function in clathrin-mediated endocytosis at the PM in plant cells, the heterotetrameric AP-2 complex and the hetero-octameric TPLATE complex (TPC). Whereas single subunit mutants in AP-2 develop into viable plants, genetic mutation of a single TPC subunit causes fully penetrant male sterility and silencing single subunits leads to seedling lethality. To address TPC function in somatic root cells, while minimizing indirect effects on plant growth, we employed nanobody-dependent delocalization of a functional, GFP-tagged TPC subunit, TML, in its respective homozygous genetic mutant background. In order to decrease the amount of functional TPC at the PM, we targeted our nanobody construct to the mitochondria and fused it to TagBFP2 to visualize it independently of its bait. We furthermore limited the effect of our delocalization to those tissues that are easily accessible for live-cell imaging by expressing it from the PIN2 promoter, which is active in root epidermal and cortex cells. With this approach, we successfully delocalized TML from the PM. Moreover, we also show co-recruitment of TML-GFP and AP2A1-TagRFP to the mitochondria, suggesting that our approach delocalized complexes, rather than individual adaptor complex subunits. In line with the specific expression domain, we only observed minor effects on root growth, yet realized a clear reduction of endocytic flux in epidermal root cells. Nanobody-dependent delocalization in plants, here exemplified using a TPC subunit, has the potential to be widely applicable to achieve specific loss-of-function analysis of otherwise lethal mutants.

Biochemical and biophysical pH clamp controlling Net H+ efflux across the plasma membrane of plant cells.

P-type H+ ATPases mediate active H+ efflux from plant cells. They generate a proton motive force across the plasma membrane, providing the free energy to drive the transport of other solutes, partly by coupling to H+ influx. Wegner & Shabala (2020) recently suggested that passive H+ influx can exceed pump-driven efflux due to 'active buffering', that is, cytosolic H+ scavenging and apoplastic H+ generation by metabolism ('biochemical pH clamp'). Charge balance is provided by K+ efflux or anion influx. Here, this hypothesis is extended to net H+ efflux: even though H+ pumping is faster than backflow via symporters and antiporters, a progressive increase in the transmembrane pH gradient is avoided. Cytosolic H+ release is associated with bicarbonate formation from CO2 . Bicarbonate serves as substrate for the PEPCase, catalyzing the reaction from phosphoenolpyruvate to oxaloacetate, which is subsequently reduced to malate. Organic anions such as malate and citrate are released across the plasma membrane and are (partly) protonated in the apoplast, thus limiting pump-induced acidification. Moreover, a 'biophysical pH clamp' is introduced, that is, adjustment of apoplastic/cytosolic pH involving net H+ fluxes across the plasma membrane, while the gradient between compartments is maintained. The clamps are not mutually exclusive but are likely to coexist.

The root response to gravity: from the macro to the nanoscale.

Plants are sessile organisms which adapt to their everchanging environment. The root is buried is the soil and continuously explores its surroundings. Indeed, while growing downwards to anchor the plants in the ground, it has to avoid obstacles and seek for nutrients and water. This seeking mechanism depends on the root perception of gravity. Through differential growth, the root is able to align according to the gravity vector. The growth is regulated at the cellular level by an increase of the plant hormone auxin, which activates the small Rho Guanine triphosphatase (Rho GTPase) of plant 6 (ROP6) at the plasma membrane to inhibit endocytosis and trigger cytoskeleton reorganization. Through a collaborative work, four French laboratories addressed the question of ROP6 membrane dynamics upon gravistimulation. Based on cellular biology, biochemistry and super resolution imaging approaches, they discovered that ROP6 is organized into nanoclusters at the plasma membrane of plant cells in response to auxin. The stabilization of ROP6 in these nanoclusters is required for signaling and thus the regulation of gravitropic bending. The formation of these nanoclusters is dependent upon the membrane lipid phosphatidylserine, which directly interact with ROP6. Using a genetic toolkit, the authors uncovered that phosphatidylserine is rate limiting for the ROP6-dependent nanocluster formation, which in turn tunes the cellular read outs. This work, not only explain the fine mechanism of the root response to gravity from the developmental level to the nanoscale but also provide a valuable insight towards the understanding of small GTPase signaling in eukaryotic system.

The cellulose synthase (CesA) gene family in four Gossypium species: phylogenetics, sequence variation and gene expression in relation to fiber quality in Upland cotton.

Cellulose synthases (CesAs) are multi-subunit enzymes found on the plasma membrane of plant cells and play a pivotal role in cellulose production. The cotton fiber is mainly composed of cellulose, and the genetic relationships between CesA genes and cotton fiber yield and quality are not fully understood. Through a phylogenetic analysis, the CesA gene family in diploid Gossypium arboreum and Gossypium raimondii, as well as tetraploid Gossypium hirsutum ('TM-1') and Gossypium barbadense ('Hai-7124' and '3-79'), was divided into 6 groups and 15 sub-groups, with each group containing two to five homologous genes. Most CesA genes in the four species are highly collinear. Among the five cotton genomes, 440 and 1929 single nucleotide polymorphisms (SNPs) in the CesA gene family were identified in exons and introns, respectively, including 174 SNPs resulting in amino acid changes. In total, 484 homeologous SNPs between the A and D genomes were identified in diploids, while 142 SNPs were detected between the two tetraploids, with 32 and 82 SNPs existing within G. hirsutum and G. barbadense, respectively. Additionally, 74 quantitative trait loci near 18 GhCesA genes were associated with fiber quality. One to four GhCesA genes were differentially expressed (DE) in ovules at 0 and 3days post anthesis (DPA) between two backcross inbred lines having different fiber lengths, but no DE genes were identified between these lines in developing fibers at 10 DPA. Twenty-seven SNPs in above DE CesA genes were detected among seven cotton lines, including one SNP in Ghi_A08G03061 that was detected in four G. hirsutum genotypes. This study provides the first comprehensive characterization of the cotton CesA gene family, which may play important roles in determining cotton fiber quality.

Exogenous Abscisic Acid Can Influence Photosynthetic Processes in Peas through a Decrease in Activity of H+-ATP-ase in the Plasma Membrane.

Simple SummaryNumerous stressors (drought, low and high temperatures, mechanical damages, etc.) act on plants under environmental conditions, suppressing their physiological processes (in particular, photosynthesis). Abscisic acid (ABA) is an important hormone, which participates in increasing plant tolerance to the action of stressors; as a result, treatment by exogenous ABA is a perspective way of regulating the tolerance in agriculture. We investigated the influence of ABA spraying on photosynthetic processes, as well as on their heat tolerance and their regulation by electrical signals propagating after local burning and modifying photosynthesis. It was shown that ABA spraying decreased photosynthetic activity and increased photosynthetic heat tolerance; additionally, the ABA treatment weakened the influence of electrical signals on photosynthesis. We revealed that these responses could be caused by a decrease in activity of H+-ATP-ase, which is an important ion transporter in plant cell plasma membrane that supports efflux of H+ from cytoplasm. As a whole, our results show the potential influence of the ABA treatment on photosynthetic processes, which is related to a decrease in activity of H+-ATP-ase. The aim of our work was to investigate the participation of inactivation of the plasma membrane H+-ATP-ase on the influence of ABA treatment on photosynthetic processes and their regulation by electrical signals in peas. The ABA treatment of seedlings was performed by spraying them with aqueous solutions (10−5 M). The combination of a Dual-PAM-100 PAM fluorometer and GFS-3000 infrared gas analyzer was used for photosynthetic measurements; the patch clamp system on the basis of a SliceScope Pro 2000 microscope was used for measurements of electrical activity. It was shown that the ABA treatment stimulated the cyclic electron flow around photosystem I and decreased the photosynthetic CO2 assimilation, the amplitude of burning-induced electrical signals (variation potentials), and the magnitude of photosynthetic responses relating to these signals; in contrast, treatment with exogenous ABA increased the heat tolerance of photosynthesis. An investigation of the influence of ABA treatment on the metabolic component of the resting potential showed that this treatment decreased the activity of the H+-ATP-ase in the plasma membrane. Inhibitor analysis using sodium orthovanadate demonstrated that this decrease may be a mechanism of the ABA treatment-induced changes in photosynthetic processes, their heat tolerance, and regulation by electrical signals.

Chitinase Gene Positively Regulates Hypersensitive and Defense Responses of Pepper to Colletotrichum acutatum Infection.

Anthracnose caused by Colletotrichum acutatum is one of the most devastating fungal diseases of pepper (Capsicum annuum L.). The utilization of chitin-binding proteins or chitinase genes is the best option to control this disease. A chitin-binding domain (CBD) has been shown to be crucial for the innate immunity of plants and activates the hypersensitive response (HR). The CaChiIII7 chitinase gene has been identified and isolated from pepper plants. CaChiIII7 has repeated CBDs that encode a chitinase enzyme that is transcriptionally stimulated by C. acutatum infection. The knockdown of CaChiIII7 in pepper plants confers increased hypersensitivity to C. acutatum, resulting in its proliferation in infected leaves and an attenuation of the defense response genes CaPR1, CaPR5, and SAR8.2 in the CaChiIII7-silenced pepper plants. Additionally, H2O2 accumulation, conductivity, proline biosynthesis, and root activity were distinctly reduced in CaChiIII7-silenced plants. Subcellular localization analyses indicated that the CaChiIII7 protein is located in the plasma membrane and cytoplasm of plant cells. The transient expression of CaChiIII7 increases the basal resistance to C. acutatum by significantly expressing several defense response genes and the HR in pepper leaves, accompanied by an induction of H2O2 biosynthesis. These findings demonstrate that CaChiIII7 plays a prominent role in plant defense in response to pathogen infection.

Complete microviscosity maps of living plant cells and tissues with a toolbox of targeting mechanoprobes

Mechanical patterns control a variety of biological processes in plants. The microviscosity of cellular structures effects the diffusion rate of molecules and organelles, thereby affecting processes such as metabolism and signaling. Spatial variations in local viscosity are also generated during fundamental events in the cell life cycle. While crucial to a complete understanding of plant mechanobiology, resolving subcellular microviscosity patterns in plants has remained an unsolved challenge. We present an imaging microviscosimetry toolbox of molecular rotors that yield complete microviscosity maps of cells and tissues, specifically targeting the cytosol, vacuole, plasma membrane, and wall of plant cells. These boron-dipyrromethene (BODIPY)-based molecular rotors are rigidochromic by means of coupling the rate of an intramolecular rotation, which depends on the mechanics of their direct surroundings, with their fluorescence lifetime. This enables the optical mapping of fluidity and porosity patterns in targeted cellular compartments. We show how apparent viscosity relates to cell function in the root, how the growth of cellular protrusions induces local tension, and how the cell wall is adapted to perform actuation surrounding leaf pores. These results pave the way to the noninvasive micromechanical mapping of complex tissues.

The auxin‐inducible phosphate transporter AsPT5 mediates phosphate transport and is indispensable for arbuscule formation in Chinese milk vetch at moderately high phosphate supply

Phosphorus is a macronutrient that is essential for plant survival. Most land plants have evolved the ability to form a mutualistic symbiosis with arbuscular mycorrhizal (AM) fungi, which enhances phosphate (Pi) acquisition. Modulation of Pi transporter systems is the master strategy used by mycorrhizal plants to adapt to ambient Pi concentrations. However, the specific functions of PHOSPHATE TRANSPORTER 1 (PHT1) genes, which are Pi transporters that are responsive to high Pi availability, are largely unknown. Here, we report that AsPT5, an Astragalus sinicus (Chinese milk vetch) member of the PHT1 gene family, is conserved across dicotyledons and is constitutively expressed in a broad range of tissues independently of Pi supply, but is remarkably induced by indole-3-acetic acid (auxin) treatment under moderately high Pi conditions. Subcellular localization experiments indicated that AsPT5 localizes to the plasma membrane of plant cells. Using reverse genetics, we showed that AsPT5 not only mediates Pi transport and remodels root system architecture but is also essential for arbuscule formation in A. sinicus under moderately high Pi concentrations. Overall, our study provides insight into the function of AsPT5 in Pi transport, AM development and the cross-talk between Pi nutrition and auxin signalling in mycorrhizal plants.

Y2SK2- and SK3-type dehydrins from Agapanthus praecox act as protectants to improve plant cell viability during cryopreservation

The cryopreservation of plants through vitrification prevents ice damage by bringing the plants to a glassy state for cryogenic storage. Most angiosperm seeds are dehydrated to extremely low levels of their original water content, accumulate late embryogenesis abundant (LEA) proteins, and greatly increase their cytoplasmic viscosity, thereby reaching a glassy state during maturation. In this study, two LEA family recombinant dehydrin (DHN) proteins (Y2SK2 and SK3) from Agapanthus praecox, when added to the plant vitrification solution, were found to double the survival rate of Arabidopsis thaliana seedlings after cryopreservation. Evans blue staining revealed that the Y2SK2 and SK3 proteins decreased the damage to the plasma membranes of plant cells during cryopreservation. Furthermore, the Y2SK2 and SK3 proteins significantly decreased the malondialdehyde and H2O2 contents, increased the glutathione (GSH) content, and downregulated NADPH oxidase (RbohA) in A. thaliana seedlings. Thus, these DHNs may reduce excessive reactive oxygen species production and membrane lipid peroxidation damage under the complex stresses of cryopreservation. Additionally, Y2SK2 can effectively enhance peroxidase activity and catalase2 (CAT2) expression levels; SK3 obviously improved ascorbic acid (AsA) content, as well as Cu/Zn superoxide dismutase (Cu/Zn-SOD) and ascorbate peroxidase 5 (APX5) expression in plant cells and enhanced H2O2 scavenging capacity by promoting the activity of the AsA-GSH cycle. These findings suggested that Y2SK2 and SK3, the DHNs added at the dehydration step, can relieve cell cryoinjury by inducing high antioxidant levels and positive oxidative stress responses and act as protectants, improving plant cell viability after cryopreservation. Application of dehydrins optimization plant cryopreservation protocol, dehydrins can significantly improve complex stress damage and enhance the cell viability from cryopreservation.

Molecular Tuning of Styryl Dyes Leads to Versatile and Efficient Plasma Membrane Probes for Cell and Tissue Imaging.

The plasma membrane (PM) plays a major role in many biological processes; therefore, its proper fluorescence staining is required in bioimaging. Among the commercially available PM probes, styryl dye FM1-43 is one of the most widely used. In this work, we demonstrated that fine chemical modifications of FM1-43 can dramatically improve the PM staining. The newly developed probes, SP-468 and SQ-535, were found to display enhanced photophysical properties (reduced cross-talk, higher brightness, improved photostability) and, unlike FM1-43, provided excellent and immediate PM staining in 5 different mammalian cell types including neurons (primary culture and tissue imaging). Taking advantage of these features, we successfully used SP-468 in STED super resolution neuronal imaging. Additionally, we showed that the new probes displayed differences in their internalization pathways compared to their parent FM1-43. Finally, we showed that the new probes kept the ability to stain the PM of plant cells. Overall, this work presents new useful probes for PM imaging in cells and tissues and provides insights on the molecular design of new PM targeting molecules.

Assessing Extrinsic Membrane Protein Dependency to PI4P Using a Plasma Membrane to Endosome Relocalization Transient Assay in Nicotiana benthamiana.

Phosphoinositides are key players from which the various membranes of the cells acquire their identity. The relative accumulation of these low-abundant anionic phospholipids in the cytosolic leaflet of the plasma membrane and of various organelles generates a landmark code, responsible for the selective recruitment of extrinsic proteins at given membranes. One of the key players in the protein/lipid interaction at the plasma membrane in plant cells, is phosphatidylinositol 4-phosphate (PI4P), which patterns the recruitment of effector proteins from the plasma membrane to organelles along the endocytic pathway. Here we describe a fast assay to assess the requirement of PI4P for membrane localization of extrinsic membrane proteins in vivo. This system relies on perturbing PI4P distribution in plant cells via the action of a PI4P phosphatase that depletes the pool of PI4P at a given membrane. This system efficiently decreases PI4P levels, and can therefore be easily used to assess requirement of PI4P (and electrostatics) for the targeting of extrinsic membrane proteins to the plasma membrane or endosomes. Ultimately, this system could also be extended to test the phosphatase activity in planta of enzymes putatively involved in PI4P metabolism.

Cell stage appears critical for control of plasma membrane order in plant cells

ABSTRACTLipids and proteins modulate both the global order of plasma membrane (PM) and its organization in distinct domains. This raises the question of the influence on PM-ordered domain formation of PM composition, which is finely controlled during cell differentiation. Labeling of plant cell PM with an environment-sensitive probe demonstrated that the level of PM order is regulated during anisotropic expansion observed during both cell regeneration from protoplasts and cell differentiation along the growing root. Indeed, PM order progressively decreased during the polarized growth of regenerated tobacco cells, without observed correlation between this parameter and the kinetics of either cell wall regeneration or cell morphology. This suggests that the dynamics of PM formation and renewal could control the PM organization, maybe by involving the secretory pathway.

The expression of a tubby-like protein from Malus domestica (MdTLP7) enhances abiotic stress tolerance in Arabidopsis

BackgroundTubby-like proteins (TLPs), characterized by a signature tubby domain, are widespread in plants and animals. To date, only plant TLPs involved in multifarious stress responses and male gametophyte development have been identified. However, studies on the molecular functions of plant TLPs are largely unknown.ResultsIn this investigation, the roles of a TLP from Malus domestica (MdTLP7) in response to abiotic stresses were characterized by expressing it in Arabidopsis. The expression of wild-type full-length MdTLP7 (FL) significantly increased the stress tolerance of Arabidopsis seedlings to osmotic, salt, cold and heat stress, while the expression of truncated MdTLP7 containing only the tubby domain (Tub) also showed some function. Located on a central α helix surrounded by 12 anti-parallel β strands in the tubby domain, the K190/R192 site may be involved in fixation to the plasma membrane, as shown by 3D homology modelling with animal TLPs. This site might play a crucial role in anti-stress functions since site-directed mutagenesis of MdTLP7 reduced stress tolerance. Subcellular localization showed that MdTLP7 was mainly localized in the plasma membrane in plant cells, suggesting that it might participate in the transduction of stress signals.ConclusionsThe results of this study showed that MdTLP7 could improve abiotic stress tolerance not only in bacteria but also in plants. The K190/R192 residues in the tubby domain were not only the plasma membrane binding site of MdTLP7 but also played a key role in stress tolerance. These results may provide a basis for further exploring the mechanism of anti-stress functioning and downstream target genes of plant TLPs.

The Crotalaria juncea metal transporter CjNRAMP1 has a high Fe uptake activity, even in an environment with high Cd contamination

Large quantities of Fe and Cd accumulate in the leaves of the metal-accumulating leguminous plant, Crotalaria juncea. A member of the metal transporter NRAMP family was cloned from C. juncea. The amino acid sequence of this clone, designated CjNRAMP1, was similar to the sequence of Arabidopsis AtNRAMP1, which is involved in Fe and Cd transport. Organ-specific analysis showed that CjNRAMP1 mRNA was expressed mainly in the leaves of C. juncea plants, as well as in stems and roots. Use of green fluorescent protein fused to CjNRAMP1 suggested its localization to the plasma membranes of plant cells. Complementation experiments using yeast strains with impaired metal transport systems showed that CjNRAMP1 transported both Fe and Cd in an inward direction within the cells. Transgenic Arabidopsis plants overexpressing CjNRAMP1 showed high tolerance to Cd, with Cd translocation from roots to leaves being substantially greater in transgenic than in wild-type plants. Overexpression of CjNRAMP1 resulted in a greater accumulation of Fe in shoots and roots, suggesting that CjNRAMP1 recognizes Fe and Cd as substrates and that the high Cd tolerance of CjNRAMP1 is due to its strong Fe uptake activity, even in the presence of high Cd concentrations in the rhizosphere.

Subcellular localisation of an endoplasmic reticulum-plasma membrane tethering factor, SYNAPTOTAGMIN 1, is affected by fluorescent protein fusion

ABSTRACTMembrane contact sites (MCS) have increasingly received attention because of their general role in a number of important cellular processes. SYNAPTOTAGMIN 1 (SYT1) is a tethering factor connecting the endoplasmic reticulum (ER) and the plasma membrane (PM) in plant cells. Confocal microscopy using fluorescent protein fusion is an indispensable tool for studying protein localisation and functions. However, several studies have reported that fluorescent protein dimerisation affects the subcellular localisation of proteins tagged by the fluorescent protein. Here, we investigate the effects of fluorescent protein dimerisation by comparing the subcellular localisation of SYT1 fused with a synthetic GFP (SYT1-sGFP) and SYT1 fused with a monomeric GFP (SYT1-mGFP). SYT1-mGFP was confined to specific domains in the ER, whereas SYT1-sGFP spread along the ER when transiently overexpressed. SYT1-localised regions were suggested to correspond to ER-PM contact sites because of its immobility. Similar results were obtained in the transgenic Arabidopsis, even though SYT1-sGFP and SYT1-mGFP were expressed at comparable levels. It is suggested that SYT1-mGFP more accurately reproduced SYT1 localisation in intact cells because the proportion of persistent area in the ER was more similar between the wild type and the plant expressing SYT1-mGFP than between the wild type and the plant expressing SYT1-sGFP. Taken together, these results suggest that the fusion of sGFP makes SYT1-sGFP form excessive ER-PM contact sites in the ER.

Silicon as a Beneficial Element and as an Essential Plant Nutrient: An Outlook (Review)

Silicon application has been known to enhance the plant tolerance against several biotic and abiotic stresses. The mechanisms of alleviation of UV-radiation, drought, salt, and heavy metal stresses have been investigated but still definitely unknown. As a consequence, the needs for understanding the role of Si in the physiological and biochemical processes in plants are of a major importance. In most cases, the published data supported the role of Si in regulation and alleviation the adverse effects of several environmental stresses under specific conditions and for specific plant species. The physiological functions of Si in plants have been studied and showed that Si influx and efflux is regulated by specific genes in plasma membrane of plant cells. The role of these genes in Si transport and alleviation several stress conditions is still poorly understood. The occurrence of high Si content in tissues of graminaceous plants (rice, barley and maize) has positively activated physiological, physical and biochemical defense mechanisms for increasing stress balance, such as decreased lipid peroxidation, reduced membrane permeability, and stimulation of defense enzymes activities. The activities of stress defense enzymes, also called antioxidative enzymes (ascorbate peroxidase, catalase, peroxidase, and superoxide dismutase), have been found to be linked to Si nutrition especially under abiotic stress conditions. There are genotypic differences controls the accumulation of Si within a specific plant species, such as rice. These differences are originated from variations in abundance of certain transporters of Si in roots or/and shoot of plant. These genotypic differences allow the plant to facilitate high growth under abiotic stress conditions. However, the responsible genes for Si accumulation in plant varieties have not yet been well characterized. Harvesting cultivated high Si – accumulator plant species results in removal of large quantity of available Si from soil. In order to replenish the needed amount of available Si from such soil, application of Si – fertilizer would improve its status in soil and maintain appreciable amount of available Si in this soil. Fertilization of soil by the required and safe rate of Si has been found to improve the chemical, physical and biological properties of plant and soil. Tests for available Si in soil and for Si content in specific plant tissue in combination with evaluation of the quantitative relationship between both would be reliable guides for Si–fertilization. In this concern, whether Si is known as a beneficial element or as an essential plant nutrient, the use of Si compounds as a fertilizer or as an amendment has been proved to be of a major importance in the present and near future.

Ectopic Expression of SjCBL1, Calcineurin B-Like 1 Gene From Sedirea japonica, Rescues the Salt and Osmotic Stress Hypersensitivity in Arabidopsis cbl1 Mutant.

Extensive studies with Arabidopsis thaliana suggested that calcineurin B-like (CBL) proteins constitute a unique family of calcium sensors in plants, which mediate a variety of abiotic stress responses. However, little is known about their function in most plants that do not have available genome sequences. In this study, we have developed a pair of universal primers that make it possible to isolate CBL1-like genes from various plants without sequence information. Using these primers, we successfully cloned a full-length cDNA of CBL1-like gene in Sedirea japonica (SjCBL1). Bimolecular fluorescence complementation (BiFC) and pull-down assays demonstrated that like Arabidopsis CBL1 (AtCBL1), SjCBL1 can interacts physically with Arabidopsis CBL-interacting protein kinase 1 (AtCIPK1) at the plasma membrane of plant cells in a Ca2+-dependent manner. In addition, overexpression of SjCBL1 in the Arabidopsis cbl1 mutant resulted in not only rescuing the hypersensitive phenotype toward salt and osmotic stresses, but also substantially enhancing the tolerance to them. Taken together, these results strongly suggest that SjCBL1 is a functional ortholog of AtCBL1 in Sedirea japonica, which can play a critical role in response to salt and osmotic stresses. Therefore, it is clear that our findings should significantly contribute to broadening and deepening our understanding of the CBL1-mediated Ca2+ signaling networks in the plant kingdom.

Phosphoregulation of the Plant Cellulose Synthase Complex and Cellulose Synthase-Like Proteins.

Cellulose, the most abundant biopolymer on the planet, is synthesized at the plasma membrane of plant cells by the cellulose synthase complex (CSC). Cellulose is the primary load-bearing polysaccharide of plant cell walls and enables cell walls to maintain cellular shape and rigidity. The CSC is comprised of functionally distinct cellulose synthase A (CESA) proteins, which are responsible for synthesizing cellulose, and additional accessory proteins. Moreover, CESA-like (CSL) proteins are proposed to synthesize other essential non-cellulosic polysaccharides that comprise plant cell walls. The deposition of cell-wall polysaccharides is dynamically regulated in response to a variety of developmental and environmental stimuli, and post-translational phosphorylation has been proposed as one mechanism to mediate this dynamic regulation. In this review, we discuss CSC composition, the dynamics of CSCs in vivo, critical studies that highlight the post-translational control of CESAs and CSLs, and the receptor kinases implicated in plant cell-wall biosynthesis. Furthermore, we highlight the emerging importance of post-translational phosphorylation-based regulation of CSCs on the basis of current knowledge in the field.

New substituted imidazo[1,5-a]pyridine and imidazo[5,1-a]isoquinoline derivatives and their application in fluorescence cell imaging

A series of new dyes based on the fluorescent imidazo[1,5-a]pyridine moiety has been designed and synthesized specifically for fluorescence cell imaging application. The obtained molecules contain a carboxylic functional group inserted to increase the water solubility and to provide an effective conjugation site. Their structural and optical properties have been deeply investigated and compared with theoretical calculations. The suitability of the products as fluorescent labels for cell imaging applications has been investigated by laser scanning confocal microscopy, both in vivo on Arabidopsis thaliana seedlings and in vitro, on mouse fibroblast cells. The compounds displayed good permeability to the plant cell wall and plasma membrane, they were efficiently internalized in the inner plant tissues, and retained a strong fluorescence in aqueous media.

The ctenidium of the giant clam, Tridacna squamosa, expresses an ammonium transporter 1 that displays light-suppressed gene and protein expression and may be involved in ammonia excretion

Ammonium transporters (AMTs) can participate in ammonia uptake or excretion across the plasma membrane of prokaryotic, plant and invertebrate cells. The giant clam, Tridacna squamosa, harbors nitrogen-deficient symbiotic zooxanthellae, and normally conducts light-enhanced ammonia absorption to benefit the symbionts. Nonetheless, it can excrete ammonia when there is a supply of exogenous nitrogen or exposed to continuous darkness. This study aimed to elucidate the role of AMT1 in the ctenidium of T. squamosa by cloning and characterizing the AMT1/AMT1, determining its subcellular localization, and examining changes in its transcript and protein expression levels in response to light exposure. The cDNA coding sequence of AMT1 from T. squamosa consisted of 1527bp and encoded 508 amino acids of 54.6kDa. AMT1-immunofluorescence was detected mainly at the apical epithelium of ctenidial filaments, and it decreased significantly after 12h of exposure to light. By contrast, the epithelial cells surrounding the tertiary water channels in the ctentidium, which are known to exhibit light-enhanced glutamine synthetase expression and take part in the assimilation of exogenous ammonia in light, did not display any AMT1-immunolabelling. Furthermore, the transcript level and protein abundance of ctenidial AMT1/AMT1 decreased significantly at the 6th and 12th h of light exposure. Taken together, these results indicate that AMT1 might participate in ammonia excretion instead of ammonia absorption and assimilation in T. squamosa. It is probable that the expression levels of AMT1/AMT1 need to be down-regulated during light exposure to achieve light-enhanced ammonia uptake.
