Tuning the antioxidant and antibacterial properties of lignin by physicochemical modification during sequential acid precipitation from Kraft black liquor

Turning lignin from black liquor waste into value-added bioactive agents is one of the possible routes for improving the sustainability and profitability of lignocellulosic industry. However, due to chemical and structural variability of lignin, it is necessary to isolate specific lignin fractions from black liquor with the purpose to achieve samples with unique chemical and structural characteristics and therefore, specific biological activities. In this study, poplar lignin fractions isolated from Kraft black liquor by sequential acid precipitation at pH´s 7.5, 5 and 2.5 (denoted as P-7.5, P-5 and P-2.5) were characterized according to their physicochemical, antioxidant and antibacterial properties. In general, lignin fractions displayed a wide elimination of lateral chains (aryl-β ether and C–C) and, therefore a high phenolic content and low molecular weight, as the pH sequential precipitation was decreased from 7.5 to 2.5. Moreover, thermal analysis revealed that the P-7.5 lignin fraction showed higher thermal stability than P-2.5 and P-5. In terms of antioxidant activity, the P-7.5 lignin fraction, with a higher S/G ratio and a less oxidized structure compared to P-5 and P-2.5, exhibited higher antioxidant activity. In addition, lower antibacterial effect was observed for all lignin fractions against Escherichia coli compared to that obtained against Staphylococcus aureus. Among them, the P-2.5 and P-5 fractions, with higher phenolic content and lower molecular weight values than P-7.5, showed a greater antibacterial effect against S. aureus.

Physicochemically modified polymer-based fluidic gates with tunable wetting properties for intelligent liquid manipulations

The creation of a selective flow path and the regulation of a flow rate are of critical importance for polymer-based devices that manipulate liquid at the microscale. The formation of a 3D interconnected network of voids (i.e., physical volumetric modification) and the addition of a nonionic surfactant, Silwet L-77, (i.e., chemical volumetric modification) are expected to affect the wetting properties of polymers, thereby achieving selective gating effect in the polymer-based devices as an appropriate technology. Here, a set of polydimethylsiloxane (PDMS)-based fluidic gates (F-gates) were developed to enable selectivity for liquids and flow-rate control of the liquids by tuning the wetting properties of PDMS with the physicochemical volumetric modifications. For water and oil with different surface tensions (STs), the effects of the physical and chemical volumetric modifications on the fluidic gating of PDMS were quantitatively characterized in terms of contact angle, mass flow rate, and liquid absorption speed. The applicability of PDMS-based F-gates to the selective separation of oil and water even from oil-in-water emulsion was demonstrated by fabricating Janus PDMS-based F-gates. Our physicochemical volumetric modifications were also extensively analyzed to examine whether they satisfy the technological, economic, and ecological requirements of appropriate technology. This is the first effort to tailor the wetting properties of PDMS through physicochemical volumetric modifications, thus configuring a set of PDMS-based F-gates that act both as a separator for liquids of different STs and as a switch for fluid flows.

Gamma irradiation for Cultural Heritage conservation: Comparison of the side effects on new and old paper

Ionizing radiations, commonly applied as diagnostic tools in Cultural Heritage (CH) field, are also proving effective for eliminating biodeteriogens (insects, fungi, bacteria and molds) responsible for the degradation of CH artifacts and often harmful for restorers, archivists and librarians. The use of ionizing radiations, such as gamma rays, for CH treatments is spreading in many countries. However, some CH operators remain resistant due to insufficient knowledge about the potential physico-chemical modifications (secondary effects) induced by radiation. This work aims to investigate and compare the effects of irradiation parameters (such as absorbed dose and dose rate) on old paper samples and new pure-cellulose paper, chosen as a reference model material. Absorbed doses up to 8 kGy have been used, as these values are commonly agreed upon for the preservation treatment of CH artifacts and are generally effective for biodeteriogens removal. Optimizing irradiation conditions helps to minimize secondary effects (such as oxidation, depolymerization or color changes), thereby increasing the reliability of the process and boosting confidence among CH operators. The secondary effects were analyzed using various physico-chemical characterizations (Fourier Transform Infrared spectroscopy, Raman microscopy, viscosimetric and colorimetric analysis) on old and new paper samples. The results indicate varying behaviors, correlated with paper composition, sample age and irradiation parameters, towards gamma radiation. This groundbreaking study not only confirms the efficacy of gamma irradiation treatments but also provides essential data that will aid in the development of optimized best practice protocols and guidelines for non-destructive and minimally destructive methods applied to real case studies and treatments.

Effect of adding various supplements on physicochemical properties and starch digestibility of cooked rice

This study investigated the physicochemical modifications of cooked rice caused by adding various supplements (rapeseed oil, dried wasabi powder, and dried chili pepper powder). The physicochemical and digestive properties of treated cooked rice were analyzed using multiple techniques to determine the impact of supplements on the rice quality, including its starch digestibility. All samples with added supplements showed an increase in surface firmness (0.77–0.95 kg·m/s2 (N)) and a decrease in thickness (2.23–2.35 mm) and surface adhesiveness (1.43–7.22 J/m3). Compared to the control group, two absorption peaks at 2856 and 1748 cm−1 and new signals at 1683 and 1435 cm−1 appeared in the Fourier transform infrared (FTIR) spectroscopy. Analysis of FTIR results revealed that the interaction force was mainly through noncovalent interactions. Moreover, adding supplements increased the resistant starch (RS) levels in all samples. Scanning electron microscopy (SEM) suggested that oil-enriched phases, proteins, and polyphenols could cause large agglomeration and loose gel structure. These results suggested the formation of amylose-guest molecule complexes, which may influence starch functionality. Our work could provide insight into the starch–supplement interactions and the key factors affecting starch digestibility.

Study on the performance and mechanism of physicochemically modified asphalt based on spatial crosslinking by the crumb rubber and polyurethane precursor

ABSTRACT Crumb rubber (CR)-modified asphalt shows positive effects on engineering performance and environmental conservation. However, its limited compatibility with asphalt due to unstable interactions poses challenges for its application. The incorporation of polyurethane precursor-based reactive modifier (PRM) facilitates the establishment of a three-dimensional crosslinked network structure within the asphalt, thereby enhancing pavement performance and compatibility. In this study, a novel compound-modified asphalt (CPMA) was prepared with CR and PRM. The performance of CPMA was assessed using the Dynamic Shear Rheometer (DSR), Blend Beam Rheology (BBR) and storage stability tests. Microstructure and modification mechanism were revealed by Atomic Force Microscopy (AFM) and Fluorescence Microscopy (FM) analyses. The results show that CPMA produced using an optimized process has better high-temperature performance and fatigue life than CR-modified asphalt (CRMA) and PRM-modified asphalt (PMA). Polar molecules, such as asphaltenes and resins, are crosslinked by the PRM to form a three-dimensional network, which enhances the thermal storage stability of CPMA. Furthermore, the flexibility and resilience of CR mitigated the low-temperature hardening of PMA. Physicochemical compound modifications provide significant advantages in terms of performance enhancement and system stability and demonstrate the potential of CR and PRM to complement and enhance their respective applications.

Drug Delivery Systems for Infectious Eye Diseases: Advancements and Prospects

Infectious ocular diseases like keratitis, conjunctivitis, and endophthalmitis pose significant clinical challenges due to the complexities of delivering drugs to the eye. Recent advancements in drug delivery systems offer promising improvements for treating these conditions. Key strategies include targeted delivery through physicochemical modifications, magnetic nanoparticles, and ligand-receptor interactions. This review explores the safety and biocompatibility of ocular drug delivery systems through in vivo ocular toxicity studies, in vitro cytotoxicity assays, hemocompatibility studies, ocular tolerance tests, and genotoxicity assays. It also examines combination therapies and stimuli-responsive delivery systems for their potential to enhance therapeutic efficacy. Furthermore, we discuss tailored and optimized drug delivery approaches for infectious ocular diseases, outlining current challenges and future directions for developing effective ocular drug delivery systems.

Physicochemical and bioactive modifications of lotus root polysaccharides associated with probiotic fermentation

The physicochemical properties and bioactivities of lotus root polysaccharides (LRP) were investigated by examining the effects of probiotic fermentation. Five polysaccharides, namely LRP, LRP-Pl, LRPRh, LRPPa, and LRPAc, were extracted from unfermented and fermented lotus roots using Lactobacillus plantarum, Lactobacillus rhamnosus, Lactobacillus paracasei and Lactobacillus acidophilus, respectively. The results revealed that the yield of LRP-Ac (3.28 %) was 3.34 times higher than that of LRP (0.98 %), with its total sugar content and glucose content in the monosaccharide composition being the highest among the five polysaccharides. Probiotic fermentation led to an increase in Mw, as well as alterations in surface morphology and glycosidic bonds of LRPs, depending on the bacterial strains. Moreover, the five polysaccharides showed differences in antioxidant, α-amylase/α-glucosidase inhibitory, prebiotic, and macrophage-stimulating activities. Specifically, LRP-Pl exhibited the strongest DPPH radical scavenging ability; LRP-Rh showed the highest ABTS radical scavenging ability along with FRAP capacities; LRP-Ac possessed the greatest α-glucosidase inhibitory as well as prebiotic activities. However, the fermented polysaccharides showed relatively weak macrophage-stimulating ability compared to unfermented ones. Finally, the structure-activity relationship analysis revealed a close correlation between the chemical composition, Mw, and monosaccharide composition of LRPs and their bioactivities. These results are useful for the preparation of high-value LRPs.

A Review of Modified Gelatin: Physicochemical Properties, Modification Methods, and Applications in the Food Field.

Gelatin is a significant multifunctional biopolymer that is widely utilized as a component in food, pharmaceuticals, and cosmetics. Numerous functional qualities are displayed by gelatin, such as its exceptional film-forming ability, gelling qualities, foaming and emulsifying qualities, biocompatibility and biodegradable qualities. Due to its unique structural, physicochemical, and biochemical characteristics, which enhance nutritional content and health benefits as well as the stability, consistency, and elasticity of food products, gelatin is utilized extensively in the food business. Additionally, gelatin has demonstrated excellent performance in encapsulating, delivering, and releasing active ingredients. Gelatin's various modifications, such as chemical, enzymatic, and physical processes, were analyzed to assess their impact on gelatin structures and characteristics. Hopefully, gelatin will be more widely used in various applications after modification using suitable methods.

Modern topical drugs-new developments in galenics and pharmacology

The importance of topically applied pharmaceuticals, especially for the treatment of dermatological diseases, is still essential. Thanks to detailed knowledge of the organisation and function of the physicochemical barrier of the skin, new galenic concepts have increasingly been developed to market maturity in recent years. Colloidal drug carrier systems in particular, but also targeted physicochemical modifications of matrices are used to optimise the cutaneous bioavailability of topically applied drugs. In addition, new molecules are increasingly becoming available for the development of these preparations or drugs known from systemic therapy are being formulated into topicals. Against this background, dermatopharmacology is also developing more and more into ahighly specialised science in this area, which is producing innovative concepts for the therapy of special indications. In addition to the classic small-molecule drugs, biotechnological molecules are increasingly finding their way into application, so that amodern age of dermatopharmacology has dawned, which will open up previously unimagined possibilities for topical therapy now and in the coming years.

Utilizing Nano-Adsorbents and Electrostatic Field Treatment for Sustainable Refinement of Crude Canola Oil.

Removal of polar impurities, such as phospholipids, free fatty acids (FFA), and peroxides, can be challenging during the refining of crude canola oil. Current conventional refining methods are energy-intensive (e.g., hot water washes) and can generate significant waste (e.g., wastewater effluent) and neutral oil loss. This study investigated the joint use of nano-adsorbents and electrostatic field (E-field) treatment as a potential and sustainable alternative in removing these impurities during the oil refining process. Specifically, aluminum oxide (Al2O3) nanoparticles were employed to neutralize FFAs, achieving a 62.4% reduction in acid value while preserving the fatty acid profile of the oil. After refining, E-field treatment was successful in removing the spent nano-adsorbent from solution (up to 72.3% by weight), demonstrating enhanced efficiency compared to conventional methods (e.g., gravitational settling, filtration, and centrifugation). The neutral oil loss using Al2O3 nano-adsorbents was also comparable to conventional refining methods, with a 4.38% (by weight) loss. After E-field treatment, the Al2O3 nano-adsorbent was then calcined to assess reusability. The Al2O3 nano-adsorbent was effectively recycled for three refining cycles. the methods do not use of large amounts of water and generate minimal waste byproducts (e.g., effluent). Nonetheless, while the nano-adsorbents demonstrated promising results in FFA removal, they were less effective in eliminating peroxides and pigments. E-field techniques were also effective in removing spent nano-adsorbent; although, optimization of E-field parameters could further improve its binding capacity. Finally, future studies could potentially focus on the physicochemical modifications of the nano-adsorbent material to enhance their refining capacity and reusability. Overall, this study presents a sustainable alternative or addition to conventional refining methods and lays the groundwork for future research.

Increased imaging ligand hydrophilicity and improved pharmacokinetic properties provides enhanced in vivo targeting of fibroblast activation protein

In this work, the impact of physicochemical modifications on pharmacokinetics and in vivo targeting of a small molecule fibroblast activation protein inhibitor (FAPI) imaging ligand in a murine model of rheumatoid arthritis was evaluated. While similar ligands have been well-reported in oncology for molecular imaging and radiotherapy, there are limited reports of FAPI derivatives in targeted applications in immunology. As inflammation may increase both specific and non-specific delivery of targeted agents in general, we sought to identify the optimal targeted molecular imaging probe characteristics for efficient cell surface engagement. A series of FAPI derivatives were synthesized and their physicochemical properties modified via conjugation of fluorescent dyes and/or an albumin-binding small molecule. The impact of these modifications on cell surface binding affinity was assessed using an overexpressing cell line. Additionally, a thorough mechanistic characterization of fibroblast activation protein (FAP) cell surface internalization was evaluated in both overexpressing and endogenously expressing cells. Lastly, the pharmacokinetics and in vivo uptake in inflamed arthritic paws were characterized via near-infrared (NIR) imaging. All targeted molecular imaging agents tested maintained strong nanomolar binding affinity to cell surface FAP independent of chemical modification. The murine fibroblast-like synoviocytes expressed lower absolute cell-surface FAP compared to a transfected line, and the net internalization half-life measured for the transfected cells via flow cytometry was 7.2 h. The unmodified FAPI ligand exhibited the poorest in vivo targeting, likely resulting from its large apparent volume of distribution (62.7 ml) and rapid systemic clearance (t1/2 = 0.5 h). Conjugation of a charged, hydrophilic AF647 fluorophore decreased systemic clearance (t1/2 = 2.1 h) and demonstrated a 2-fold improvement in blocking FAPI-800CW engagement of FAP in vivo when compared to blocking of FAPI-800CW with FAPI with up to 2.8-fold improvements noted for the equivalent albumin binding construct comparison.

Optimization of Yucca filamentosa fiber based graft copolymer through response surface methodology and evaluation of physico-chemical properties

The study of the physico-chemical modification of Yucca filamentosa (Yf) natural fiber by graft copolymerization with ethylmethacrylate using ferrous ammonium sulfate-potassium persulfate as a redox initiator has been reported in the article. Initially, six process parameters; reaction duration, reaction temperature, solvent amount, pH, FAS:KPS ratio, and monomer concentration were used in the study in a sequential experimental design technique, and the significant process variables affecting the yield of the graft copolymer were identified. The Resolution-V design method identified the significant parameters as the reaction temperature, amount of solvent, and the concentration of monomer. In second phase of the study, the screened variables were utilized in the development of a model through the technique of response surface methodology (RSM) for the prediction of the yields, and its optimization. The developed RSM model fitted well with the experimental data, and predicted for the optimal conditions of reactions as temperature 50 °C, solvent 100 ml, and the monomer 3.05 × 10−3 mol/L; at which the highest graft yield percentage obtained was 124.2%. The techniques of FTIR, SEM, and XRD were used for the characterization graft copolymers. Studies of the various physico-chemical properties showed that the produced graft copolymers were more resistant than the natural fibers.

Early Peri-Implant Bone Healing on Laser-Modified Surfaces with and without Hydroxyapatite Coating: An In Vivo Study.

(1) Objective: The aim of this study was to assess the biological behavior of bone tissue on a machined surface (MS) and modifications made by a laser beam (LS) and by a laser beam incorporated with hydroxyapatite (HA) using a biomimetic method without thermic treatment (LHS). (2) Methods: Scanning electron microscopy coupled with energy-dispersive X-ray spectrometry (SEM/EDX) was performed before and after installation in the rabbit tibiae. A total of 20 Albinus rabbits randomly received 30 implants of 3.75 × 10 mm in the right and left tibias, with two implants on each surface in each tibia. In the animals belonging to the 4-week euthanasia period group, intramuscular application of the fluorochromes calcein and alizarin was performed. In implants placed mesially in the tibiofemoral joint, biomechanical analysis was performed by means of a removal torque (N/cm). The tibias with the implants located distally to the joint were submitted for analysis by confocal laser microscopy (mineral apposition rate) and for histometric analysis by bone contact implant (%BIC) and newly formed bone area (%NBA). (3) Results: The SEM showed differences between the surfaces. The biomechanical analysis revealed significant differences in removal torque values between the MSs and LHSs over a 2-week period. Over a 4-week period, both the LSs and LHSs demonstrated removal torque values statistically higher than the MSs. BIC of the LHS implants were statistically superior to MS at the 2-week period and LHS and LS surfaces were statistically superior to MS at the 4-week period. Statistical analysis of the NBA of the implants showed difference between the LHS and MS in the period of 2 weeks. (4) Conclusions: The modifications of the LSs and LHSs provided important physicochemical modifications that favored the deposition of bone tissue on the surface of the implants.

Role of charge exchange collision on generation of active species for cold plasma food processing

Charge exchange collision (CXC) is well known in solar and space plasmas. In this work, we present how the CXC between N2+ and N2 can be exploited to overcome major challenges in cold plasma food processing (CPFP). CPFP is an emerging application of glow discharge plasmas for physicochemical modifications to achieve shelf-life enhancement, preservation, surface activation for germination, antimicrobial treatment, surface cleaning, etc. The commercial application of CPFP is in its infancy and it faces two major challenges. The first challenge is the difficulty in generating the desired active species for the required modification, and the second is the very high processing cost. In this paper, with the help of numerical modeling for nitrogen discharge, we show that the CXC between N2+ and N2 can be utilized to generate active species selectively, enhance energy efficiency, and possibly eliminate the processing gas cost. The modeling is followed by experimental demonstration and validation of the proposed concept. This work may lead to a new direction of transdisciplinary research towards the commercial application of CPFP.

Polycaprolactone scaffold surface modification with soft X-ray/extreme ultraviolet (SXR/EUV) radiation and low-temperature oxygen and nitrogen plasma for biomedical applications

Modification of the surfaces of polymeric scaffolds is often required to make the material suitable for specific tissue engineering applications. Physico-chemical properties of scaffolds can be altered using various methods, such as plasma treatment, laser processing, chemical modifications, grafting with nanoparticles, or surface coating. In this paper physico-chemical modification of polycaprolactone (PCL) surface fibers was performed by exposing PCL samples to simultaneous soft X-ray/extreme ultraviolet (SXR/EUV) radiation and low-temperature, SXR/EUV-induced, nitrogen, and oxygen plasmas. The physical and chemical changes on modified PCL surfaces were examined using a scanning electron microscope and X-ray photoelectron spectroscopy, respectively. The effects of physico-chemical scaffold surface changes were verified with biological tests, i.e., MTT assay and immunofluorescence on murine osteoblast cell line (7F2). It was found that exposure of scaffolds to ionizing radiation and low-temperature plasmas induced strong chemical changes on their surface, i.e., appearance of various new chemical groups. Also, smoothing of the surface of PCL fibers, i.e., disappearance or significant reduction of the size of micropores on their fibers was also observed. Increased viability and adhesion of 7F2 osteoblasts on modified PCL samples after 24 h cell culture compared to non-treated PCL was also confirmed.Graphical abstract

Tandem hydroformylation-amine condensation and reduction of canola fatty acid methyl esters for production of diesel renewable antioxidant additives

A canola fatty acid methyl esters (FAME) mixture was synthesized through transesterification and modified by rhodium homogeneous tandem hydroformylation-amine condensation, with primary amines, yielding imine products. The obtained imines were reduced by maceration with sodium borohydride and alumina, yielding secondary amine products. The synthesized fatty imines and amines were evaluated as additives for commercial diesel and biodiesel on concentrations of 0.5 and 1.0 %, while tested for oxidative stability fallowing the EN14112 standard method. The tested amino products exhibited higher oxidative stability, which justifies its application as antioxidant additives, while the diesel blends did not show expressive physicochemical modifications. The modified amino-FAME antioxidant performance, justifies a renewable biorefinery application, extend the biodiesel shelf-life prior to diesel blending, complies with UN’s Sustainable Development Goals (SDGs) and if prepared from residues, enhance the circular economy appeal.

Development of sustainable sugarcane bagasse biorefinery using a greener deep eutectic solvent from hemicellulose-derived acids

Acid-based deep eutectic solvents (ADES) have recently been tested as an emerging technology in biomass deconstruction. In particular, the use of hemicellulose-derived carboxylic acids has attracted great interest as this type of ADES can enable a sustainable closed-loop biorefinery, due to their efficiency and recyclability. Therefore, this study proposed the pretreatment of sugarcane bagasse (SCB) with acetic acid, as a biomass-derived chemical, and choline chloride [ChCl]:AA to perform a complete biomass fractionation to enhance enzymatic digestibility. Thus, this work analyzed the effect of different temperatures and reaction time during pretreatment, studying the impact on biomass composition and digestibility during enzymatic hydrolysis. In addition, the reuse of ADES to produce an environmentally friendly and low-cost SCB biorefinery was explored by studying the physicochemical modifications in polysaccharide-rich material (PRM) and lignin-rich material (LRM) by ATR-FTIR and SEM photography. Furthermore, the ADES content was also characterized by HPLC after each recycling cycle. The results demonstrated efficient polysaccharide conversion, promoting an enrichment in glucan (>60%) and xylan (>20%) content under mild operating conditions (130°C and 90 min) supported by ATR-FTIR and SEM. Furthermore, SCB pretreated under these selected conditions achieved higher digestibility, increasing by 89% for glucan and 73% for xylan, using a simple and competitive process. In contrast, increasing the temperature above 130°C did not improve the enzymatic digestibility of SCB. Thus, ChCl:AA could be recycled and reused. Therefore, this study demonstrated the potential of AA in DES pretreatments as an environmentally friendly and efficient method for the valorization of SCB components.

A review on sustainable management of biomass: physicochemical modification and its application for the removal of recalcitrant pollutants-challenges, opportunities, and future directions.

Adsorption is one of the most efficient methods for remediating industrial recalcitrant wastewater due to its simple design and low investment cost. However, the conventional adsorbents used in adsorption have several limitations, including high cost, low removal rates, secondary waste generation, and low regeneration ability. Hence, the focus of the research has shifted to developing alternative low-cost green adsorbents from renewable resources such as biomass. In this regard, the recent progress in the modification of biomass-derived adsorbents, which are rich in cellulosic content, through a variety of techniques, including chemical, physical, and thermal processes, has been critically reviewed in this paper. In addition, the practical applications of raw and modified biomass-based adsorbents for the treatment of industrial wastewater are discussed extensively. In a nutshell, the adsorption mechanism, particularly for real wastewater, and the effects of various modifications on biomass-based adsorbents have yet to be thoroughly studied, despite the extensive research efforts devoted to their innovation.
