Extremozyme-Based Biosensors for Environmental Pollution Monitoring: Recent Developments.

Extremozymes combine high specificity and sensitivity with the ability to withstand extreme operational conditions. This work presents an overview of extremozymes that show potential for environmental monitoring devices and outlines the latest advances in biosensors utilizing these unique molecules. The characteristics of various extremozymes described so far are presented, underlining their stability and operational conditions that make them attractive for biosensing. The biosensor design is discussed based on the detection of photosynthesis-inhibiting herbicides as a case study. Several biosensors for the detection of pesticides, heavy metals, and phenols are presented in more detail to highlight interesting substrate specificity, applications or immobilization methods. Compared to mesophilic enzymes, the integration of extremozymes in biosensors faces additional challenges related to lower availability and high production costs. The use of extremozymes in biosensing does not parallel their success in industrial applications. In recent years, the "collection" of recognition elements was enriched by extremozymes with interesting selectivity and by thermostable chimeras. The perspectives for biosensor development are exciting, considering also the progress in genetic editing for the oriented immobilization of enzymes, efficient folding, and better electron transport. Stability, production costs and immobilization at sensing interfaces must be improved to encourage wider applications of extremozymes in biosensors.

Potential Candidate Molecule of Photosystem II Inhibitor Herbicide-Brassicanate A Sulfoxide.

Brassicanate A sulfoxide, a secondary metabolite of broccoli, exhibited the inhibition of weed growth, but its mechanism of action on weeds remains unclear. To elucidate the mechanism by which brassicanate A sulfoxide suppresses weeds, this study explores the interaction between brassicanate A sulfoxide and the photosystem II D1 protein through molecular docking and molecular dynamics simulations. This research demonstrates that brassicanate A sulfoxide interacts with the photosystem II D1 protein by forming hydrogen bonds with Phe-261 and His-214. The successful expression of the photosystem II D1 protein in an insect cell/baculovirus system validated the molecular docking and dynamics simulations. Biolayer interferometry experiments elucidated that the affinity constant of brassicanate A sulfoxide with photosystem II was 2.69 × 10-3 M, suggesting that brassicanate A sulfoxide can stably bind to the photosystem II D1 protein. The findings of this study contribute to the understanding of the mode of action of brassicanate A sulfoxide and also aid in the development of natural-product-based photosynthesis-inhibiting herbicides.

Adaptation responses in C4 photosynthesis of sweet maize (Zea mays L.) exposed to nicosulfuron

Nicosulfuron is an ingredient in photosynthesis-inhibiting herbicides and has been widely used in corn post-emergence weed control. In the current study, a pair of sister lines, HK301 (nicosulfuron-tolerence, NT) and HK320 (nicosulfuron-sensitive, NS), was used to study the effect of nicosulfuron in sweet maize seedlings on C4 photosynthetic enzymes and non-enzymatic substances, expression levels of key enzymes, and chloroplast structure. Nicosulfuron was sprayed at the four-leaf stage, and water was sprayed as a control. After nicosulfuron treatment, phosphoenolpyruvate carboxylase (PEPC), NADP-malic dehydrogenase (NADP-MDH), NADP-malic enzyme (NADP-ME), pyruvate orthophosphate dikinase (PPDK), and ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) activities of NT were significantly higher than those of NS. Compared to NT, malate, oxaloacetic acid, and pyruvic acid significantly decreased as exposure time increased in NS. Compared to NS, nicosulfuron treatment significantly increased the expression levels of PEPC, NADP-MDH, NADP-ME, PPDK, and Rubisco genes in NT. Under nicosulfuron treatment, chloroplast ultrastructure of NS, compared to that of NT, nicosulfuron induced swelling of the chloroplast volume and reduced starch granules in NS. In general, our results indicate that in different resistant sweet maize, C4 photosynthetic enzymes activity and key genes expression play a critical role in enhancing the adaptability of plants to nicosulfuron stress at a photosynthetic physiological level.

Herbicide Exposure and Toxicity to Aquatic Primary Producers.

The aim of the present review was to give an overview of the current state of science concerning herbicide exposure and toxicity to aquatic primary producers. To this end we assessed the open literature, revealing the widespread presence of (mixtures of) herbicides, inevitably leading to the exposure of non-target primary producers. Yet, herbicide concentrations show strong temporal and spatial variations. Concerning herbicide toxicity, it was concluded that the most sensitive as well as the least sensitive species differed per herbicide and that the observed effect concentrations for some herbicides were rather independent from the exposure time. More extensive ecotoxicity testing is required, especially considering macrophytes and marine herbicide toxicity. Hence, it was concluded that the largest knowledge gap concerns the effects of sediment-associated herbicides on primary producers in the marine/estuarine environment. Generally, there is no actual risk of waterborne herbicides to aquatic primary producers. Still, median concentrations of atrazine and especially of diuron measured in China, the USA and Europe represented moderate risks for primary producers. Maximum concentrations due to misuse and accidents may even cause the exceedance of almost 60% of the effect concentrations plotted in SSDs. Using bioassays to determine the effect of contaminated water and sediment and to identify the herbicides of concern is a promising addition to chemical analysis, especially for the photosynthesis-inhibiting herbicides using photosynthesis as endpoint in the bioassays. This review concluded that to come to a reliable herbicide hazard and risk assessment, an extensive catch-up must be made concerning macrophytes, the marine environment and especially sediment as overlooked and understudied environmental compartments.

Microbial amperometric biosensor for online herbicide detection: Photocurrent inhibition of Anabaena variabilis

A microbial biosensor based on the cyanobacteria Anabaena variabilis was developed for online herbicide detection through inhibition of generated photocurrent. Specifically, atrazine and diuron were employed as model photosynthesis-inhibiting herbicides. To fabricate the biosensor, bacterial cells were immobilized on a carbon felt electrode using alginate as an entrapping polymer and p-benzoquinone (BQ) as the redox mediator to sustain the electron transfer. The current generated by the photo-bioelectrocatalytic oxidation of water was evaluated. During the calibration, an immediate concentration-dependent decrease of the photocurrent was observed after the injection of the tested herbicides. The biosensor showed a sensitivity of −24.6 μA μM−1 cm−2 towards atrazine up to 0.56 μM. As diuron is a much stronger photosynthesis inhibitor, the biosensor allows only turn on/turn off detection for this compound.In order to avoid the release of the mediator in solution, the encapsulation of BQ in the polymer was tested. Addition of activated carbon was necessary to create a conductive network inside the alginate matrix, enabling to achieve a sensitivity of −7.7 μA μM−1 cm−2 towards atrazine up to 131 μM (lower limit of detection 64 nM). Therefore, not only the entrapment prevented the release of BQ in solution, but improved the operating range of the sensor, with a limited decrease of sensitivity that does not hinder its application. These characteristics make the biosensor suitable for environmental analysis, opening for the on-line monitoring of herbicides.

Responses of photosynthesis-related parameters and chloroplast ultrastructure to atrazine in alfalfa (Medicago sativa L.) inoculated with arbuscular mycorrhizal fungi

Atrazine is an ingredient in photosynthesis-inhibiting herbicides and has been widely used to combat weeds in farmland. However, most atrazine that is applied fails to degrade in the soil and subsequently affects non-target plants. In this study, we investigated the influence of arbuscular mycorrhizal fungi (AMF), Funneliformis mosseae on the photosynthesis-related parameters, chlorophyll content, and chloroplast ultrastructure in alfalfa plants, some of which had been exposed to atrazine. Our results showed that the percentage of AMF hyphal colonization reached 91.23% 35 days after the alfalfa was planted, which suggests a symbiotic relationship between F. mosseae and alfalfa roots. F. mosseae alleviated the inhibition of net photosynthesis and stomatal function significantly in alfalfa exposed to atrazine for 24 h. A chlorophyll fluorescence analysis revealed that F. mosseae prevented a major reduction in the performance of photosystem II (PSII) photochemistry in the presence of atrazine, such as the relative decrease of Fv/Fm between the non-mycorrhizal and F. mosseae mycorrhizal treatments was 4.4% and 5.8% after 24 and 48 h of atrazine exposure time. However, F. mosseae has no significant alleviation on a sharp reduction in the chlorophyll a, chlorophyll b and carotenoid content in alfalfa exposed to atrazine. For the chloroplast ultrastructure in alfalfa exposed to atrazine, the number of both plastoglobules and partial granal stacks was greater in the presence of F. mosseae. In general, our results indicate that the F. mosseae inoculation was beneficial to sustain photosynthesis-related performance, such as net photosynthesis, stomatal conductance, the maximum quantum yield (Fv/Fm) and effective quantum yield (ΦPSII) of PSII photochemistry in alfalfa after exposure to atrazine, because the mycorrhizal alfalfa had a greater number of plastoglobules and granal stacks in the chloroplast, thereby enhancing its resistance to the oxidative damage induced by atrazine.

Blinded by the light: Increased chlorophyll fluorescence of herbicide-exposed periphyton masks unfavorable structural responses during exposure and recovery

In surface waters within agricultural catchments, periphyton – i.e., biofilms containing algae, heterotrophs, and associated detritus – is subjected to multiple stressors including herbicides. Although herbicide effects on periphyton are frequently studied, the focus has been on photosynthesis-inhibiting herbicides while other modes of toxic action have received little attention. Against this background, a 21-days-lasting bioassay was conducted, during which mature periphytic communities were exposed to the carotenoid-biosynthesis-inhibiting herbicide diflufenican for 12 days (up to 10 μg/L; n = 4), followed by a 9-days-lasting recovery phase in herbicide-free medium. Variables related to periphytic functioning (photosynthetic efficiency and non-photochemical quenching) and structure (pigment concentrations, biomass, and algal community structure) were quantified every third day during both experimental phases. Exposure to ≥ 0.2 μg diflufenican/L resulted in 20–25% and 25–30% lowered carotenoid and chlorophyll a concentrations, respectively, likely explained by a reduced algal biovolume as well as diflufenican’s mode of toxic action and thus a shift towards a higher heterotrophy of the communities. Despite these adverse effects on the photosynthetic apparatus, the photosynthetic efficiency increased by up to ∼15% under diflufenican exposure judged on higher chlorophyll fluorescence. This may be explained by an up to ∼60% reduced non-photochemical quenching as well as binding of diflufenican to the pigment-protein membrane complex of the photosystem II, two processes causing higher chlorophyll fluorescence. Additionally, phototrophs may have actively increased energy assimilation to cope with higher energy demands under chemical stress. Although periphyton showed some recovery potential following the exposure phase, observed as increasing chlorophyll a concentrations and non-photochemical quenching, periphyton may not be able to quickly recover from stress given the persistent increase in the photosynthetic efficiency. While the processes underlying the observed effects yet remain speculative, the results suggest a shift towards a higher degree of heterotrophy in periphytic communities ultimately increasing the importance of heterotrophic ecosystem functions at impacted sites over the long term.

Advances in Enzyme-Based Biosensors for Pesticide Detection.

The intensive use of toxic and remanent pesticides in agriculture has prompted research into novel performant, yet cost-effective and fast analytical tools to control the pesticide residue levels in the environment and food. In this context, biosensors based on enzyme inhibition have been proposed as adequate analytical devices with the added advantage of using the toxicity of pesticides for detection purposes, being more “biologically relevant” than standard chromatographic methods. This review proposes an overview of recent advances in the development of biosensors exploiting the inhibition of cholinesterases, photosynthetic system II, alkaline phosphatase, cytochrome P450A1, peroxidase, tyrosinase, laccase, urease, and aldehyde dehydrogenase. While various strategies have been employed to detect pesticides from different classes (organophosphates, carbamates, dithiocarbamates, triazines, phenylureas, diazines, or phenols), the number of practical applications and the variety of environmental and food samples tested remains limited. Recent advances focus on enhancing the sensitivity and selectivity by using nanomaterials in the sensor assembly and novel mutant enzymes in array-type sensor formats in combination with chemometric methods for data analysis. The progress in the development of solar cells enriched the possibilities for efficient wiring of photosynthetic enzymes on different surfaces, opening new avenues for development of biosensors for photosynthesis-inhibiting herbicides.

Glufosinate-ammonium 저항성 형질전환벼의 저항성 수준과 적응성에 관한 연구

본 연구의 목적은 bar 유전자를 발현한 형질전환벼에 대한 글루타민 합성효소 저해제인 glufosinate-ammonium, L-metionine sulfoximine과 광합성저해제인 paraquat에 저항성 수준과 형질전환벼와 비형질전환벼의 암모늄 축적량, 저온반응 및 수량을 비교하는데 있다. 형질전환벼 계통은 비형질전환벼에 비해 glufosinate-ammonium에 45~95배 높은 저항성을 보였다. 한편 형질전환벼 계통은 L-metionine sulfoximine을 처리했을 때도 형질전환벼 계통이 비형질전환벼에 비해 18배 이상의 저항성을 보여 교차저항성이 있음을 확인할 수 있었다. 그러나 glufosinate-ammonium과 작용기작이 다른 제초제인 paraquat에 대한 다중저항성은 없었다. Glufosinate-ammonium 처리에 의해 ammonium 축적은 비형질전환벼에서 증가하였으나 모든 형질전환벼의 계통에서는 적거나 효과가 없었다. 형질전환벼 계통 258, 411, 607 및 608을 제외한 계통은 비형질전환벼에 비해 저온 내성 및 저온 후 회복정도가 유의적으로 낮은 경향을 보였다. 형질전환벼 계통 142, 144, 258 및 608의 수량은 비형질전환벼와 유사하거나 높았다. The objectives of this research were to quantify resistance levels of transgenic rice expressing the bar gene to glutamine synthetase (GS)-inhibiting, and methionine sulfoximine and photosynthesis-inhibiting herbicide, paraquat, and compare the ammonium accumulation, chilling injury, and yield between transgenic and non-transgenic rice. The transgenic rice lines were 45-96-fold more resistant to glufosinate-ammonium than non-transgenic rice. The transgenic rice lines were also 18-fold more resistant to methionine sulfoximine, but was not resistant to paraquat, which has different target site. Glufosinate-ammonium increased the ammonium accumulation in leaves of non-transgenic rice plants, but had minimal or no effect on leaves of transgenic lines. The transgenic lines except for 258, 411, 607 and 608 were more susceptible during chilling and recovery than non-transgenic rice plants. The yield of transgenic lines 142, 144, 258 and 608 was similar or higher than that of non-transgenic rice in pot conditions.

Resistance levels and fitness of glufosinate-resistant transgenic sweet potato in field experiments

Transgenic herbicide-resistant sweet potato plants [ Ipomoea batatas (L.) Lam.] produced through a biolistic transformation were used in this study. The objectives of this research were to (a) quantify resistance levels of transgenic sweet potato expressing the bar gene to glutamine synthetase (GS)-inhibitors, glufosinate and methionine sulfoximine, protoporphyrinogen oxidase (PPO)-inhibiting herbicide, oxyfluorfen and photosynthesis-inhibiting herbicide, paraquat; (b) compare the relative chlorophyll content, quantum yield, growth, and tuberous root yield between transgenic and wild-type sweet potato; (c) compare the response of transgenic and wild-type sweet potato plants to chilling and high temperature, and (d) compare amino acids, minerals and sugar contents of transgenic and wild-type sweet potato tubers. The transgenic sweet potato lines were 20–82-fold more resistant to glufosinate than the wild-type. The representative transgenic line 7171 also showed resistance to methionine sulfoximine, but was not resistant to oxyfluorfen and paraquat, which have different target sites. The stem length, number of leaves, petiole length, shoot fresh weight, and storage roots of transgenic lines were lower than those of the wild-type in field experiments. Reduced growth and root yield were not due to reduced quantum yield and relative chlorophyll contents. Glufosinate increased the ammonium accumulation in leaves of wild-type plants, but had minimal or no effect on leaves of transgenic line 7171. There were differences in mineral contents, free sugars, total amino acids, and free amino acids in the petiole and storage roots between wild-type and line 7171. There was no difference in drought tolerance between wild-type and line 7171, but the transgenic line was more tolerant to chilling temperature than wild-type plants. Follow-up research is required to elucidate the mechanisms underlying the growth and yield differences, different nutritional values, and differential response to low temperature between wild-type and transgenic sweet potato.

Temperature-dependent sensitivity of growth and photosynthesis of Scenedesmus obliquus, Navicula pelliculosa and two strains of Microcystis aeruginosa to the herbicide atrazine

The temperature-dependent sensitivities of two algal species and two strains of cyanobacteria to the photosynthesis-inhibiting herbicide atrazine were evaluated in order to understand how the interaction between acclimation temperature and herbicide will affect growth and photosynthesis of aquatic microorganisms. The green alga Scenedesmus obliquus, the diatom Navicula pelliculosa and a toxic and non-toxic strain of Microcystis aeruginosa were acclimated to three different temperatures (10, 15 and 25 °C) and exposed to five concentrations of the herbicide atrazine (0–0.15 μM) for 72 h. Growth, photosynthetic yields, energy fluxes within photosystem II and pigment content were then measured as potential responses to each treatment. With the exception of N. pelliculosa, the toxicity of atrazine was higher when microorganisms were acclimated to lower temperatures. N. pelliculosa was not only the most tolerant to atrazine, but also had a similar sensitivity to this herbicide at every temperature. The observed differences in growth sensitivity to atrazine at low temperature are associated with the ability of algae and cyanobacteria to cope with high excitation pressure, by increasing its protective carotenoid content and non-photochemical energy dissipation. Our results demonstrate that future guidelines for the protection of aquatic life should consider water temperature as an important factor influencing the toxicity of atrazine to aquatic microorganisms.

Effect of 24-Epibrassinolide on Chlorophyll Fluorescence and Photosynthetic CO2 Assimilation in Vicia faba Plants Treated with the Photosynthesis-Inhibiting Herbicide Terbutryn

Simultaneous measurements of chlorophyll (Chl) fluorescence and CO2 assimilation (A) in Vicia faba leaves were taken during the first weeks of growth to evaluate the protective effect of 24-epibrassinolide (EBR) against damage caused by the application of the herbicide terbutryn (Terb) at pre-emergence. V. faba seeds were incubated for 24 h in EBR solutions (2 × 10−6 or 2 × 10−5 mM) and immediately sown. Terb was applied at recommended doses (1.47 or 1.96 kg ha−1) at pre-emergence. The highest dose of Terb strongly decreased CO2 assimilation, the maximum quantum yield of PSII photochemistry in the dark-adapted state (F V/F M), the nonphotochemical quenching (NPQ), and the effective quantum yield (ΔF/F′M) during the first 3–4 weeks after plant emergence. Moreover, Terb increased the basal quantum yield of nonphotochemical processes (F 0/F M), the degree of reaction center closure (1 − q p), and the fraction of light absorbed in PSII antennae that was dissipated via thermal energy dissipation in the antennae (1 − F′V/F′M). The herbicide also significantly reduced plant growth at the end of the experiment as well as plant length, dry weight, and number of leaves. The application of EBR to V. faba seeds before sowing strongly diminished the effect of Terb on fluorescence parameters and CO2 assimilation, which recovered 13 days after plant emergence and showed values similar to those of control plants. The protective effect of EBR on CO2 assimilation was detected at a photosynthetic photon flux density (PFD) of 650 μmol m−2 s−1 and the effect on ΔF/F′M and photosynthetic electron transport (J) was detected under actinic lightings up to 1750 μmol m−2 s−1. The highest dose of EBR also counteracted the decrease in plant growth caused by Terb, and plants registered the same growth values as controls.

Response of Phytoplankton Community to Low-Dose Atrazine Exposure Combined with Phosphorus Fluctuations

The effects of atrazine on a controlled phytoplankton community derived from a natural freshwater wetland exposed to low doses of this photosynthesis-inhibiting herbicide were examined. The community was exposed for 7 weeks to doses of 0.1, 1, and 10 microg L(-1) atrazine, combined with changes in nutrient concentration, and the photosynthetic activity, biomass, and community structure were noted during the experiment. Responses of the phytoplankton community were examined in terms of photosynthetic activity, biomass, and community structure. Significant effects of atrazine on the phytoplankton assemblage, in terms of primary production and community structure, were highlighted, even at doses as low as 1 and 0.1 microg L(-1), when associated with phosphorus fluctuations. The most abundant Chlorophyceae decreased in concentration with increasing atrazine dose, whereas cyanobacteria were more tolerant to atrazine, particularly with increased nutrient supply. The subinhibitory doses of atrazine used in the present study confirmed the higher sensitivity of long-term exposure of multispecies assemblages under resource competition. Our study supports the emerging hypothesis that the increasing prevalence of cyanobacterial blooms in European aquatic systems may result from a combination of unbalanced nutrient enrichment and selective pressures from multiple toxicants.

Comparison of fate and ecological effects of the herbicide linuron in freshwater model ecosystems between tropical and temperate regions

This paper compares the fate and effects of linuron in an outdoor plankton-dominated microcosm study carried out in Thailand with those reported in temperate model ecosystem studies evaluating linuron and other photosynthesis-inhibiting herbicides. Lower linuron concentrations disappeared slightly faster from the water compartment compared to temperate conditions, which appears to be related with the experimental design rather than differences in climatic conditions. Sensitivity of primary producers and zooplankton were similar for the climatic regions, whereas effects on ecosystem functioning were less pronounced in tropical microcosms. Recovery potential of affected endpoints appears higher for tropical ecosystems compared to their temperate counterparts. These findings support the use of toxicity data generated in temperate countries in the tropics. Recommendations for the methodology of tropical model ecosystem experiments are discussed.

Ecological effects of the herbicide linuron in tropical freshwater microcosms

Effects of a single application of the photosynthesis-inhibiting herbicide linuron (0, 15, 50, 150, and 500 μg/L) on the ecology of outdoor plankton-dominated microcosms were studied in Thailand. As a result of the decreased photosynthesis, DO and pH decreased while EC, alkalinity and nutrient concentrations increased. Chlorophytes belonging to the genera Scenedesmus, Coelastrum and Pediastrum were the most sensitive taxa, whereas several other chlorophytes, diatoms, and cryptophytes increased in abundance. Tolerant taxa appeared to be less digestable for several zooplankton taxa, which subsequently decreased in abundances. Chamaesiphon sp. (Cyanobacteria) was the most susceptible periphyton species. As a consequence of functional redundancy, effects of the herbicide on the chlorophyll- a content of periphyton and especially phytoplankton did not always reflect the effects noted on community level.

Virtual imprinting as a tool to design efficient MIPs for photosynthesis-inhibiting herbicides

Molecular modelling and computational screening were used to identify functional monomers capable of interacting with several different photosynthesis-inhibiting herbicides. The process involved the design of a virtual library of molecular models of functional monomers containing polymerizable residues and residues able to interact with the template through electrostatic, hydrophobic, Van der Waals forces and dipole–dipole interactions. Each of the entries in the virtual library was probed for its possible interactions with molecular models of the template molecules. It was anticipated that the monomers giving the highest binding score would represent good candidates for the preparation of affinity polymers. Strong interactions were computationally determined between acidic functional monomers like methacrylic acid (MAA) or itaconic acid (IA) with triazines, and between vinylimidazole with bentazone and bromoxynil. Nevertheless, weaker interactions were seen with phenylureas. The corresponding blank polymers were prepared using the selected monomers and tested in the solid phase extraction (SPE) of herbicides from chloroform solutions. A good correlation was found between the binding score of the monomers and the affinities of the corresponding polymers. The use of computationally designed blanks can potentially eliminate the need for molecular imprinting, (adding a template to the monomer mixture to create specific binding sites). Data also showed that some monomers have a natural selectivity for some herbicides, which can be further enhanced by imprinting. Thus, in regard to retention on the blank polymer, we can estimate if the resulting imprinted polymer will be effective or not.

Integration of photosynthetic biosensor with molecularly imprinted polymer-based solid phase extraction cartridge

A simple method for the detection of individual groups of photosynthesis-inhibiting herbicides was proposed as a practical approach for environmental monitoring. The detection method is based on fluorescent measurement of the inhibition of spinach thylakoidal activity by herbicides. The sensor selectivity was improved by combining biosensor with a sample preconcentration based on a SPE cartridge packed with a polymer, imprinted with cyanazine. The combination of biosensor with molecularly imprinted polymer (MIP) allows selective measurement of triazines herbicides (cyanazine, atrazine) and their discrimination from phenylurea (diuron) or triazinone (metribuzin) herbicides. This combination also helps improving the sensitivity of the measurement allowing the detection of photosynthesis-inhibiting herbicides in water at the level required by European Union regulations.

On the way to cyanobacterial blooms: Impact of the herbicide metribuzin on the competition between a green alga ( Scenedesmus) and a cyanobacterium ( Microcystis)

The hypothesis that exposure to a common and widely applied photosynthesis-inhibiting herbicide, metribuzin, would alter the outcome of the competitive battle between susceptible green algae ( Scenedesmus obliquus) and tolerant cyanobacteria ( Microcystis aeruginosa) was tested. In a long-term (17 d) experiment, Scenedesmus and Microcystis populations as well as mixtures that started with different inoculum composition (i.e. 3:1, 1:1 and 1:3 Scenedesmus: Microcystis) were grown in the absence or presence of metribuzin (100 μg l −1). In the absence of metribuzin, Scenedesmus was competitively superior and out-competed Microcystis regardless the initial composition of the mixed communities. However, this competitive outcome was reversed completely in the presence of metribuzin, where despite growth inhibition Microcystis became dominant. Hence, photosynthesis-inhibiting herbicides may not only affect algal community structure, but also provide cyanobacteria founder populations a window for dominance and thus play an important role in promoting cyanobacteria blooms.

Comparing aquatic risk assessment methods for the photosynthesis-inhibiting herbicides metribuzin and metamitron

Three different risk assessment procedures are described that aim to protect freshwater habitats from risks of the photosynthesis-inhibiting herbicides metribuzin and metamitron. These procedures are (1) the first-tier approach, based on standard toxicity tests and the application of an assessment factor, (2) the Species Sensitivity Distribution (SSD) approach, based on laboratory tests with a wider array of species and the application of a statistical model to calculate the HCx (the Hazardous Concentration for x% of the species), and (3) the model ecosystem approach, based on the evaluation of treatment-related effects in field enclosures. A comparison of the risk assessment procedures reveals that the first-tier approach is the most conservative for metamitron and metribuzin, and that HC5 values (and even HC10 values) based on acute EC50 values of algae and aquatic vascular plants may be used to derive maximum permissible concentrations for single applications. For both compounds these HC5 values were very similar to the ecological threshold concentrations in the enclosure studies. In contrast to model ecosystem experiments, however, HCx values based on lab toxicity tests do not provide information on the recovery potential of sensitive endpoints and on indirect effects, which may be important for regulatory decision-making. In the enclosure study, indirect effects of metribuzin on invertebrate populations were observed at an exposure concentration that was approximately 20 times lower than the corresponding HC5 value based on lab toxicity data for aquatic invertebrates.

The F684/F735 chlorophyll fluorescence ratio: a potential tool for rapid detection and determination of herbicide phytotoxicity in algae

The use of herbicides constitutes the principal method of weed control but the introduction of these compounds into the aquatic environment (primarily through runoff) may have severe consequences for non-target plants. In this study, we describe a sensitive and inexpensive method for detection of photosynthesis-inhibiting herbicides, based on chlorophyll (Chl) fluorescence emission. Algae exhibited a Chl fluorescence signature with two maxima around 684 and 735 nm, correlated with the total Chl content of the algal suspension. The ratio of these two maxima (i.e. F684/F735) can be used as an indicator of stress in the photosynthetic apparatus, and thus represents a very simple method for in vivo evaluation of the health status of algae. Determination of the F684/F735 fluorescence ratio revealed the presence and phytotoxicity of atrazine, metribuzin, terbuthylazine, diuron, DCPMU, DCPU and paraquat. The toxic effect of these pollutants was estimated by monitoring the increase in the F684/F735 value, which reflects photosystem II and photosystem I photochemistry. We observed a drastic increase in the magnitude of this ratio, correlating quantitatively with herbicide concentration and corresponding to a decline in algal photosynthetic activity. For the tested herbicides affecting photosynthetic electron transport, the magnitude of the effect was as follows: diuron=DCPMU>metribuzin>atrazine>terbuthylazine>paraquat>DCPU. The F684/F735 Chl fluorescence ratio thus gives toxicity responses which compare favourably with tests such as the algal growth inhibition test, and could therefore be used to detect the presence and phytotoxicity of herbicides in aquatic environments.
