Degradation of 2,2′,4,4′-tetrabromodiphenyl ether by Pycnoporus sanguineus in the presence of copper ions

The degradation of 2,2′,4,4′-tetrabromodiphenyl ether (BDE-47) by Pycnoporus sanguineus was investigated in order to explore the impact of the heavy metal Cu2+ on BDE-47 decomposition and the subsequent formation of metabolites, as well as to further elucidate the degradation mechanism of BDE-47. An increase in degradation rate from 18.63% to 49.76% in the first four days and its stabilization at (51.26 ± 0.08)% in the following days of BDE-47 incubation were observed. The presence of Cu2+ at 1 and 2 mg/L was found to promote the degradation rate to 56.41% and 60.79%, respectively, whereas higher level of Cu2+ (≥5 mg/L) inhibited the removal of BDE-47. The similar concentration effects of Cu2+ was also found on contents of fungal protein and amounts of metabolites. Both intracellular and extracellular enzymes played certain roles in BDE-47 transportation with the best degradation rate at 27.90% and 27.67% on the fourth and third day, individually. During the degradation of BDE-47, four types of hydroxylated polybrominated diphenyl ethers (OH-PBDEs), i.e., 6′-OH-BDE-47, 5′-OH-BDE-47, 4′-OH-BDE-17, 2′-OH-BDE-28, and two bromophenols, i.e., 2,4-DBP and 4-BP were detected and considered as degradation products. These metabolites were further removed by P. sanguineus at rates of 22.42%, 23.01%, 27.04%, 27.96%, 64.21%, and 40.62%, respectively.

Sponge-derived polybrominated diphenyl ethers and dibenzo-p-dioxins, irreversible inhibitors of the bacterial α-d-galactosidase.

An integrated in vitro and in silico approach was applied to evaluate the potency of hydroxylated polybrominated diphenyl ethers (OH-PBDEs) and spongiadioxins (OH-PBDDs) isolated from Dysidea sponges on the activity of the recombinant α-d-galactosidase of the GH36 family. It was revealed for the first time that all compounds rapidly and apparently irreversibly inhibited the bacterial α-d-galactosidase. The structure-activity relationship study in the series of OH-PBDEs showed that the presence of an additional hydroxyl group in 5 significantly enhanced the potency (IC50 4.26 μM); the increase of bromination in compounds from 1 to 3 increased their potency (IC50 41.8, 36.0, and 16.0 μM, respectively); the presence of a methoxy group decreased the potency (4, IC50 60.5 μM). Spongiadioxins 6, 7, and 8 (IC50 16.6, 33.1, and 28.6 μM, respectively) exhibited inhibitory action comparable to that of monohydroxylated diphenyl ethers 1-3. Docking analysis revealed that all compounds bind in a pocket close to the catalytic amino acid residues. Molecular docking detected significant compound-enzyme interactions in the binding sites of α-d-galactosidase. Superimposition of the enzyme-substrate and the enzyme-inhibitor complexes showed that their binding sites overlap.

Analysis of hydroxylated polybrominated diphenyl ethers (OH-BDEs) by supercritical fluid chromatography/mass spectrometry

Hydroxylated polybrominated diphenyl ethers (OH-BDEs), which have anthropogenic and natural origins, have exhibited neurotoxic and endocrine disrupting effects in humans and wildlife. Therefore, there is an increased interest in the analysis of these compounds in biological matrices in order to assess their potential toxicological risks. Analysis of OH-BDEs is conventionally completed using liquid chromatography/mass spectrometry (LC/MS), or by gas chromatography/mass spectrometry (GC/MS) after derivatization. Issues with resolution in separating congeners have limited the analysis of OH-BDEs via LC/MS, with published methods only able to include 13 congeners in the analysis. On the other hand, while GC/MS analysis can analyze more OH-BDE congeners, derivatization of OH-BDEs to convert them to GC amenable compounds adds to sample preparation time and limits the column lifetime due to trace residues of highly reactive derivatization agents entering the column. Herein we report the development of a supercritical fluid chromatography/mass spectrometry (SFC/MS) method for the analysis of 22 OH-BDE congeners. Instrument limits of detection for the developed method ranged from 2 to 106fg injected on column, which is lower than previously optimized LC/MS and GC/MS methods. The developed SFC/MS method was successfully applied towards the analysis of in vitro metabolism samples and human serum samples to demonstrate its applicability with different biological matrices.

Transformation of hydroxylated and methoxylated 2,2′,4,4′,5-brominated diphenyl ether (BDE-99) in plants

The occurrence and fate of hydroxylated polybrominated diphenyl ethers (OH-PBDEs) and methoxylated polybrominated diphenyl ethers (MeO-PBDEs) have received significant attention. However, there is limited information on the metabolism relationship between OH-pentaBDEs and MeO-pentaBDEs that were frequently detected with relatively high concentrations in the environment. In this study, the biotransformation between OH-BDE-99 and MeO-BDE-99 was investigated in rice, wheat, and soybean plants. All the three plants can metabolize OH-BDE-99 to corresponding homologous methoxylated metabolites, while the transformation from MeO-BDE-99 to OH-BDE-99 could only be found in soybean. The conversion of parent compounds was the highest in soybean, followed by wheat and rice. Transformation products were found mainly in the roots, with few metabolites being translocated to the shoots and solution after exposure. The results of this study provide valuable information for a better understanding of the accumulation and transformation of OH-PBDEs and MeO-PBDEs in different plants.

Quantification of Hydroxylated Polybrominated Diphenyl Ethers (OH-BDEs), Triclosan, and Related Compounds in Freshwater and Coastal Systems.

Hydroxylated polybrominated diphenyl ethers (OH-BDEs) are a new class of contaminants of emerging concern, but the relative roles of natural and anthropogenic sources remain uncertain. Polybrominated diphenyl ethers (PBDEs) are used as brominated flame retardants, and they are a potential source of OH-BDEs via oxidative transformations. OH-BDEs are also natural products in marine systems. In this study, OH-BDEs were measured in water and sediment of freshwater and coastal systems along with the anthropogenic wastewater-marker compound triclosan and its photoproduct dioxin, 2,8-dichlorodibenzo-p-dioxin. The 6-OH-BDE 47 congener and its brominated dioxin (1,3,7-tribromodibenzo-p-dioxin) photoproduct were the only OH-BDE and brominated dioxin detected in surface sediments from San Francisco Bay, the anthropogenically impacted coastal site, where levels increased along a north-south gradient. Triclosan, 6-OH-BDE 47, 6-OH-BDE 90, 6-OH-BDE 99, and (only once) 6’-OH-BDE 100 were detected in two sediment cores from San Francisco Bay. The occurrence of 6-OH-BDE 47 and 1,3,7-tribromodibenzo-p-dioxin sediments in Point Reyes National Seashore, a marine system with limited anthropogenic impact, was generally lower than in San Francisco Bay surface sediments. OH-BDEs were not detected in freshwater lakes. The spatial and temporal trends of triclosan, 2,8-dichlorodibenzo-p-dioxin, OH-BDEs, and brominated dioxins observed in this study suggest that the dominant source of OH-BDEs in these systems is likely natural production, but their occurrence may be enhanced in San Francisco Bay by anthropogenic activities.

Photochemical Formation of Hydroxylated Polybrominated Diphenyl Ethers (OH-PBDEs) from Polybrominated Diphenyl Ethers (PBDEs) in Aqueous Solution under Simulated Solar Light Irradiation.

Hydroxylated polybrominated diphenyl ethers (OH-PBDEs) are of great concern due to their higher toxicity compared to PBDEs. However, the abiologic process whereby PBDEs are converted to OH-PBDEs in the aquatic environment is not well understood. To explore the possibility of OH-PBDEs photoformation in natural water, the photohydroxylation of BDE-47 has been investigated in aqueous Fe(III) and/or fulvic acid (FA) solutions and in natural lake water under simulated solar light irradiation. The results showed that 6-OH-BDE-47 and 2'-OH-BDE-68 were generated from BDE-47 under these conditions. Based on the identification of derivatives and reactive radicals, OH-PBDEs formation can be ascribed to an addition reaction of ortho-tetra-BDE radical and hydroxyl radical ((•)OH), with or without a subsequent Smiles rearrangement reaction. Since the ortho-tetra-BDE radical could be readily produced by the photolysis of BDE-47, even in pure water, (•)OH production was considered as critical for the photoformation of OH-PBDEs. Thus, it is reasonable to deduce that the photoreactive components (Fe(III), FA) in aqueous solution played an important role through influencing (•)OH generation. Although the yields of OH-PBDEs did not increase regularly with increasing concentration of these photoreactive components in solution, this study suggests a possible abiotic origin of OH-PBDEs formation in the aquatic environment.

Production of Hydroxylated Polybrominated Diphenyl Ethers (OH-PBDEs) from Bromophenols by Manganese Dioxide

Hydroxylated polybrominated diphenyl ethers (OH-PBDEs) are of significant concern because of their enhanced toxicological effects compared to PBDEs. Research to date has attributed the origin of OH-PBDEs to biological metabolism of PBDEs and natural production in the environment. However, it is unclear how OH-PBDEs are formed naturally. In this study, we explored the formation of OH-PBDEs via the oxidative transformation of simple bromophenols (BPs, e.g., 4-BP, 2,4-DBP, and 2,4,6-TBP) by birnessite (δ-MnO2). Results showed that OH-PBDEs were readily produced by δ-MnO2 with BPs as precursors. For example, oxidation of 2,4-DBP by δ-MnO2 yielded 2'-OH-BDE-68 and 2',5'-OH-BDE-25. Other OH-PBDEs, such as 6-OH-BDE-13, 2',5'-OH-BDE-3, 4'-OH-BDE-121, and 2',5'-OH-BDE-69, were detected from the reaction with 4-BP and 2,4,6-TBP. The formation of OH-PBDEs likely resulted from the oxidative coupling of bromophenoxy radicals. Mild acidic conditions enhanced while coexisting cations (e.g., Na(+), Mg(2+), and Ca(2+)) suppressed the transformation. Given the ubiquity of BPs and δ-MnO2, oxidation of BPs by δ-MnO2 and other metal oxides is likely an abiotic route for the formation of OH-PBDEs in the environment.

Halogenated phenolic compound determination in plasma and serum by solid phase extraction, dansylation derivatization and liquid chromatography–positive electrospray ionization–tandem quadrupole mass spectrometry

A robust, sensitive and accurate method was developed for the simultaneous determination in plasma and serum of suite a halogenated phenolic compounds (HPCs) for which several are known to persist in the environment and analytically pure standards are available. Namely, 14 congeners of hydroxylated polybrominated diphenyl ethers (OH-PBDEs), six congeners of hydroxylated polychlorinated biphenyls (OH-PCBs), pentachlorophenol, pentabromophenol and the flame retardant tetrabromobisphenol A (TBBPA). Solid phase extraction (SPE) enriched the target compounds and cleaned up the samples as a result of efficient adsorption on a strong anion-exchange solid phase SPE cartridge (Oasis MAX). After final clean-up the HPCs were derivatized with dansyl chloride and analyzed by liquid chromatography–tandem mass spectrometry with electrospray ionization in positive mode (ESI(+)). Chromatographic separation was achieved on a Luna PFP(2) column (2mm×100mm, 3μm particle size) with mobile phases of water and acetonitrile (both containing 0.1% formic acid). The addition of the dansyl moiety to the HPCs greatly improved analyte sensitivity as the electrospray ionization efficiency was enhanced. Instrument limits of detection (ILODs) for LC–ESI(+)–MS/MS analysis of the HPCs were in the range of 0.01–0.07ng/mL and the method limits of quantification (MLOQs) were in the range of 0.02–0.15ng/g. Recovery efficiencies of the suite of HPCs ranged from 64% to 118% with relative standard deviations from 2% to 12% from fortified bovine serum samples. The method was successfully applied for HPC determination in representative polar bear and snapping turtle plasma samples.

Improved 3D-QSPR analysis of the predictive octanol–air partition coefficients of hydroxylated and methoxylated polybrominated diphenyl ethers

The octanol/air partition coefficient (KOA) is a key physicochemical parameter for describing the partition of organic pollutants between air and environment organic phase. The development of appropriate method to estimate KOA is of great importance. In the present study, the steric, electrostatic, hydrophobic, hydrogen bond donor and acceptor descriptors were computed by comparative molecular field analysis (CoMFA) and comparative molecular similarity indices analysis (CoMSIA). On the basis of these parameters, the statistically quantitative structure–property relationship (QSPR) models for logKOA of hydroxylated polybrominated diphenyl ethers (OH-PBDEs) and methoxylated polybrominated diphenyl ethers (MeO-PBDEs) congeners were developed using partial least-squares (PLS) analysis, of which the R2 is about 0.980, 0.952 respectively. The electrostatic field was found to be main factors governing the logKOA. The results of validation indicate the models of this study exhibit optimum stability, and thus it is feasible to predict logKOA.

Structure-dependent activities of hydroxylated polybrominated diphenyl ethers on human estrogen receptor

Polybrominated diphenyl ethers (PBDEs) have been shown to affect the estrogen receptor (ER) signaling pathway, and one of the proposed disruption mechanisms is direct binding of hydroxylated PBDE (OH-PBDE) to ER. In this paper, the binding affinity of 22 OH-PBDEs with different degrees of bromination to ER was assessed quantitatively using a surface plasmon resonance biosensor technique. Seven OH-PBDEs were found to bind directly with ER with KD ranging from 1.46x10−7M to 7.90x10−6M, and the affinity is in the order of 6-OH-BDE-047≧4′-OH-BDE-049>4′-OH-BDE-017>6′-OH-BDE-099≧5′-OH-BDE-099>2′-OH-BDE-007>3′-OH-BDE-028. In MVLN luciferase gene reporter assays, 10 low-brominated OH-PBDEs induced luciferase activity alone, but are 105 to 107 fold less potent than E2. Their estrogenic activity is in the order of 4′-OH-BDE-049>4′-OH-BDE-017>2′-OH-BDE-007>3′-OH-BDE-028>3-OH-BDE-047≧3′-OH-BDE-007. The good correlation between estrogenic activity and ER binding affinity of the low-brominated OH-PBDEs strongly suggest that these compounds induce ER transcriptional activity by binding directly with ER. The other 12 high-brominated OH-PBDEs inhibited luciferase activity of E2 to various degrees, demonstrating their antagonistic activity. Molecular docking analysis of the ER/OH-PBDE complexes revealed two distinctive binding modes between low- and high-brominated OH-PBDEs which provided rationale for the difference in their ER activity.

Hydroxylated polybrominated diphenyl ethers exhibit different activities on thyroid hormone receptors depending on their degree of bromination

Polybrominated diphenyl ethers (PBDEs) have been shown to disrupt thyroid hormone (TH) functions in experimental animals, and one of the proposed disruption mechanisms is direct binding of hydroxylated PBDE (OH-PBDE) to TH receptors (TRs). However, previous data on TH receptor binding and TH activity of OH-PBDEs were very limited and sometimes inconsistent. In the present paper, we examined the binding potency of ten OH-PBDEs with different degrees of bromination to TR using a fluorescence competitive binding assay. The results showed that the ten OH-PBDEs bound to TR with potency that correlated to their bromination level. We further examined their effect on TR using a coactivator binding assay and GH3 cell proliferation assay. Different TR activities of OH-PBDEs were observed depending on their degree of bromination. Four low-brominated OH-PBDEs (2′-OH-BDE-28, 3′-OH-BDE-28, 5-OH-BDE-47, 6-OH-BDE-47) were found to be TR agonists, which recruited the coactivator peptide and enhanced GH3 cell proliferation. However, three high-brominated OH-PBDEs (3-OH-BDE-100, 3′-OH-BDE-154, 4-OH-BDE-188) were tested to be antagonists. Molecular docking was employed to simulate the interactions of OH-PBDEs with TR and identify the structural determinants for TR binding and activity. According to the docking results, low-brominated OH-PBDEs, which are weak binders but TR agonists, bind with TR at the inner side of its binding pocket, whereas high-brominated compounds, which are potent binders but TR antagonists, reside at the outer region. These results indicate that OH-PBDEs have different activities on TR (agonistic or antagonistic), possibly due to their different binding geometries with the receptor.

Anthropogenic and naturally occurring polybrominated phenolic compounds in the blood of cetaceans stranded along Japanese coastal waters

We determined the residue levels and patterns of hydroxylated polybrominated diphenyl ethers (OH-PBDEs), and related compounds, such as PBDEs, methoxylated PBDEs (MeO-PBDEs), and bromophenols (BPhs) in the blood of eleven cetacean species stranded along the Japanese coasts. The dominant OH- and MeO-PBDE isomers found in all cetaceans were 6OH-BDE47 and 6MeO-BDE47. Additionally, 2,4,6-triBPh was dominant isomer in all cetaceans. In contrast, specific differences in the distribution of para- and meta- OH-PBDE isomers and some BPhs (potential PBDEs metabolites) were found among the cetaceans. Residue levels of ΣMeO-PBDEs and 6OH-BDE47 + 2′OH-BDE68, and 2,4,6-triBPh and 6OH-BDE47 + 2′OH-BDE68 showed a significant positive correlation. These results may suggest that the large percentages of OH-PBDEs, MeO-PBDEs and 2,4,6-triBPh might share common source (i.e. biosynthesis by marine organisms), or metabolic pathway in cetacean species. Significant correlations were found between the concentrations of BDE99 and 2,4,5-triBPh. This result suggested that 2,4,5-triBPh in cetaceans could be a metabolite of BDE99.

Comprehensive Two-Dimensional Separation of Hydroxylated Polybrominated Diphenyl Ethers by Ultra-Performance Liquid Chromatography Coupled with Ion Mobility-Mass Spectrometry

A comprehensive two-dimensional system coupling ultra-performance liquid chromatography (UPLC) and ion mobility-mass spectrometry (IM-MS) has been applied for the separation and analysis of hydroxylated polybrominated diphenyl ethers (OH-PBDEs). A complex mixture containing 23 OH-PBDE congeners ranging from hydroxylated monobromodiphenyl ether (OH-monoBDE) to hydroxylated octabromodiphenyl ether (OH-octaBDE) was satisfactorily separated within 16 min of analysis time. The first-dimensional reversed-phase UPLC was performed on a sub-2 μm BEH C(18) chromatographic column using acetonitrile-water gradient elution program with a flow rate ramp. It enabled excellent chromatographic separation for both between-class and within-class OH-PBDEs based on their differences in hydrophobicity. Following the pre-ionization resolution in the first dimension, the second-dimensional IM-MS employed a hybrid electrospray quadrupole ion mobility time-of-flight mass spectrometer and added an extra post-ionization separation for between-class OH-PBDE congeners on account of their relative mobility disparity during a very short period of 8.80 ms. The orthogonality of the developed two-dimensional system was evaluated with the correlation coefficient of 0.9665 and peak spreading angle of 14.87°. The peak capacity of the system was calculated to be approximately 2 and 15 times higher than that of the two dimensions used alone, respectively. The two-dimensional separation plane also contributed to the removal of background interference ions and the enhanced confidence in the characterization of OH-PBDEs of interest.

PBDEs and methoxylated analogues in sediment cores from two Michigan, USA, inland lakes

Polybrominated diphenyl ethers (PBDEs) have been widely studied in sediments from the North American Great Lakes; however, no studies have been conducted of occurrences of methoxylated (MeO-) PBDEs in abiotic compartments in this region. In the present study, 23 tri- to hepta-PBDEs and 12 MeO-PBDEs were analyzed in dated sediment cores collected from two inland lakes (White Lake and Muskegon Lake) in Michigan, USA. Concentrations of Σ(23) PBDEs ranged from 3.9 × 10(-1) to 2.4 × 10(0) and from 9.8 × 10(-1) to 3.9 × 10(0) ng/g dry weight in White Lake and Muskegon Lake, respectively. The historical trends of tri- to hepta-PBDEs in the two lakes were different, possibly because of different input and remediation histories. The tri- to hepta-PBDE profiles were similar in the two lakes, with BDE-47 as the predominant congener, followed by BDE-99 and BDE-183. A different temporal trend for BDE-183 was found compared with other PBDEs, which is consistent with debromination of high-brominated PBDEs during sedimentation and aging. Methoxylated-PBDEs were detected only in Muskegon Lake (3.6 × 10(-3) to 1.2 × 10(-1) ng/g dry wt). Methoxylated PBDEs showed different temporal trends compared with tri- to hepta-PBDEs. The differences in patterns of concentrations of MeO-PBDEs in the two lakes might be due to different aquatic communities in each lake. The occurrences of MeO-PBDEs could be the major source of hydroxylated-polybrominated diphenyl ethers (OH-PBDEs) observed in organisms collected in these freshwater systems.

Structure-based investigation on the binding interaction of hydroxylated polybrominated diphenyl ethers with thyroxine transport proteins

Polybrominated diphenyl ethers (PBDEs) have been shown to alter thyroid hormone level in experimental animals. One of the possible mechanisms for hormone disruption is the competitive binding of hydroxylated PBDEs (OH-PBDEs) with hormone transport proteins. In this study, binding interaction of 14 diversely structured OH-PBDEs with two thyroxine transport proteins was investigated by fluorescence displacement assay, circular dichroism, and molecular docking. Binding affinity of the 14 OH-PBDEs with transthyretin (TTR) and thyroxine-binding globulin (TBG) was measured by competitive fluorescence displacement assay. The binding constant was found to fall in the range of 1.4 × 10 7 M −1 and 6.9 × 10 8 M −1 for TTR, and between 6.5 × 10 6 M −1 and 2.2 × 10 8 M −1 for TBG. Binding affinity increased significantly with bromination number from 1 to 4, whereas 5- and 6-brominated diphenyl ethers did not show any further increase. Protein secondary structural change of TTR and TBG upon binding with 5-OH-BDE-047 was investigated by circular dichroism. The spectral change displayed a pattern similar to the one with thyroxine, suggesting that the environmental chemical binds to the two proteins at the same sites as the hormone. In molecular docking analysis, a ligand-binding channel in TTR was revealed for OH-PBDEs binding, which appeared to be mostly hydrophobic inside but guarded by positively charged residue Lys15 at the entrance. Binding affinity of the 14 OH-PBDEs with TTR could be rationalized reasonably well by their pocket binding mode and hydrophobic characteristics. Based on the binding constant obtained in this work, possibility of in vitro competitive displacement of thyroid hormones from the transport proteins by OH-PBDEs was evaluated.
