Rapid formulation screening with a Multipart Microscale Fluid bed Powder processor

The aim of this study was to investigate early formulation screening in small scale with a miniaturized fluid bed device. Altogether eight different batches were granulated in a Multipart Microscale Fluid bed Powder processor (MMFP) with constant process conditions using electrostatic atomization. Atomization voltage and granulation liquid flow rate were kept constant. Acid acetylsalicylic was used as model active pharmaceutical ingredient (API), lactose monohydrate, microcrystalline cellulose and polyvinylpyrrolidone were used as excipients. Granule size distributions were measured with spatial filtering technique. Friability test was performed by spinning granules in the mixer with glass beads. Compressibility of the granules was evaluated by tableting and the breaking force of the tablets was measured. Multivariate analysis, namely partial least squares regression and multilinear regression were applied to the data. It was possible to generate granules of different compositions rapidly employing MMFP with electrostatic atomization fast and acquire reliable and logical results with only small amount of material. However, a major challenge was to find suitable analytical methods for such small batches.

Development of a scalable procedure for fine calcium alginate particle preparation

A simple and scalable two step method for the preparation of loaded calcium alginate fine powder was developed. Spray-dryer was used to prepare sodium alginate particles and all the spraying parameters were studied to optimize the powder characteristics. The particles were gelified successively by external gelation in a calcium chloride solution. Two model active pharmaceutical ingredients were encapsulated in alginate particles (i.e., bacitracin and bovine serum albumin). The obtained powders show the suitable characteristics for pulmonary drug delivery.

Quantitative HPLC Analysis of Active Pharmaceutical Ingredients in Syrup Vehicle Using Centrifugal Filter Devices and Determination of Xanthan Gum in Syrup Vehicle Using Rheometry

Using rapid centrifugal filtration (<or= 30 min) of diluted samples, the filter membrane prevented compounds with molecular weight higher than the nominal molecular weight limit (NMWL) from transporting through the membrane, thus separating them from compounds with molecular weight smaller than NMWL, which would pass through the membrane. The purpose of this study aims to remove high molecular weight matrix (such as xanthan gum) interferences while achieving a quantitative analysis of the active pharmaceutical ingradients of interest. Two model active pharmaceutical ingredients, L-arginine and amphotericin B, were quantitatively recovered from the diluted syrup vehicle after centrifugation with the filter devices. The reproducibility [% relative standard deviation (RSD), peak area] of the filtered samples was less than 0.5%. For amphotericin B samples. The linear range was 0.28 microg/mL to 28.2 microg/mL. The limit of detection was 0.06 microg/mL. The limit of quantification was 0.28 microg/mL. The viscosity of a syrup vehicle changed linearly with the concentration of xanthan gum. A method was thus developed to determine xanthan gum in the syrup vehicle. The accuracy was within 95.0% to 105.0% at different concentration levels.

Continuous Crystallization of Pharmaceuticals Using a Continuous Oscillatory Baffled Crystallizer

In this paper, we report an investigation into the continuous crystallization of a model active pharmaceutical ingredient (API) using a continuous oscillatory baffled crystallizer (COBC). The results show that continuous crystallization offers significant advantages in terms of process, operation and costs, and delivers the isolation of the model API in just over 12 min compared to the 9 h and 40 min in a batch process.

Scanning White-Light Interferometry as a Novel Technique to Quantify the Surface Roughness of Micron-Sized Particles for Inhalation

A novel approach of measuring the surface roughness of spherical and flat micron-sized drug particles using scanning white-light interferometry was applied to investigate the surface morphology of micron-sized active pharmaceutical ingredients (APIs) and excipient particles used for inhalation aerosols. Bovine serum albumin (BSA) and alpha-lactose monohydrate particles were chosen as model API and excipient particles, respectively. Both BSA and lactose particles were prepared with different degrees of surface corrugation using either controlled spray drying (four samples of BSA) or decantation (two samples of lactose). Particle size distributions were characterized by laser diffraction, and particles were imaged by scanning electron microscopy (SEM). Surface roughness of the BSA and lactose particles was quantified by white-light optical profilometry using vertical scanning interferometry (VSI) at full resolution using a 50x objective lens with 2.0x and 0.5x fields of view for BSA and lactose, respectively. Data were analyzed using Vision software (version 32, WYKO), and surface roughness values are expressed as root-mean-square roughness ( Rrms). Furthermore, data were compared to topographical measurements made using conventional atomic force microscopy. Analysis of the optical profilometry data showed significant variation in BSA roughness ranging from 18.58 +/- 3.80 nm to 110.90 +/- 13.16 nm for the smoothest and roughest BSA particles, respectively, and from 81.20 +/- 15.90 nm to 229.20 +/- 68.20 nm for decanted and normal lactose, respectively. The Rrms values were in good agreement with the AFM-derived values. The particle morphology was similar to SEM and AFM images. In conclusion, scanning white-light interferometry provides a useful complementary tool for rapid evaluation of surface morphology and roughness in particles used for dry powder inhalation formulation.

The manufacture and characterisation of hot-melt extruded enteric tablets

The aim of this highly novel study was to use hot-melt extrusion technology as an alternative process to enteric coating. In so doing, oral dosage forms displaying enteric properties may be produced in a continuous, rapid process, providing significant advantages over traditional pharmaceutical coating technology. Eudragit L100-55, an enteric polymer, was pre-plasticized with triethyl citrate (TEC) and citric acid and subsequently dry-mixed with 5-aminosalicylic acid, a model active pharmaceutical ingredient (API), and an optional gelling agent (PVP K30 or Carbopol 971P). Powder blends were hot-melt extruded as cylinders, cut into tablets and characterised using powder X-ray diffraction (PXRD), differential scanning calorimetry (DSC) and dissolution testing conducted in both pH 1.2 and pH 6.8 buffers. Increasing the concentration of TEC significantly lowered the glass transition temperature (Tg) of Eudragit L100-55 and reduced temperatures necessary for extrusion as well as the die pressure. Moreover, citric acid (17% w/w) was shown to act as a solid-state plasticizer. HME tablets showed excellent gastro-resistance, whereas milled extrudates compressed into tablets released more than 10% w/w of the API in acidic media. Drug release from HME tablets was dependent upon the concentration of TEC, the presence of citric acid, PVP K30, and Carbopol 971P in the matrix, and pH of the dissolution media. The inclusion of an optional gelling agent significantly reduced the erosion of the matrix and drug release rate at pH 6.8; however, the enteric properties of the matrix were lost due to the formation of channels within the tablet. Consequently this work is both timely and highly innovative and identifies for the first time a method of producing an enteric matrix tablet using a continuous hot-melt extrusion process.

Powder X-ray Diffraction as an Emerging Method to Structurally Characterize Organic Solids

The current level of laboratory instrumentation and computational resources allows X-ray powder diffraction to be implemented into the toolbox of organic chemists, providing a means for rapid (i.e., within a day) structural characterization of organic solids, without the need for single crystals. We illustrate such use of powder diffraction using two case studies of molecular cocrystals of trifluoroacetic acid and malonic acid, involving theobromine, a model active pharmaceutical ingredient. We also report on a previously unobserved conformation of malonic acid in the solid state.

Effect of Formulation and Processing Variables on the Granulation Kinetics of Hot Melt Granulation (HMG) Process

The objective of the study was to evaluate the effect of formulation factors, such as type of drug and particulate properties of a drug, and processing variables, i.e. jacket temperature, impeller speed, and scale, on granulation kinetics the of hot-melt granulation (HMG) process. Two model active pharmaceutical ingredients (API) Ro-A and indomethacin were selected for this evaluation using poloxamer 188 as a meltable binder. The effect of solid-state properties of API was investigated for Ro-A, whereas the binder properties were maintained constant. General factorial design was used to investigate the effect of independent process variables, impeller speed and jacket temperature using impeller motor power consumption as response variable. Consistent granulation could be developed for Ro-A by optimizing the binder level and impeller speed, however, the addition of third excipient was necessary for indomethacin. The granulation rate was related to the bulk density and the surface area of the drug. The jacket temperature affected overall granulation time but had no significant effect on the granulation kinetics, suggesting that faster heating rate is desirable for optimal productivity. A significant increase in the granulation rate was observed with increase in impeller speed. The effect of impeller speed was further confirmed at 5 L and 25 L scale. From the formulation prospective, the critical factors were the level of binder, inherent binding properties of the API, the solid-state properties of API and binder. From processing perspectives, the impeller speed had a significant effect on the granulation kinetics.

Modeling the impact of direct phototransformation on predicted environmental concentrations (PECs) of propranolol hydrochloride in UK and US rivers

There is currently uncertainty on the persistence of active pharmaceutical ingredients (APIs) and on their depletion mechanisms in natural surface waters such as rivers, and hence predictions of their fate are often poor. In this study, a β-adrenergic receptor, propranolol hydrochloride, was selected as a model API to explore the relative significance of direct phototransformation as a potential removal process of hydrophilic APIs in rivers. Phototransformation kinetics of propranolol was measured under simulated solar irradiation in the laboratory, which were then converted to the kinetics applicable in UK and US rivers. The effects of light intensity, light penetration, river size and flow were examined. The extrapolated phototransformation half-lives were applied in the river catchment models of GREAT-ER and P hATE TM. Results demonstrated that direct phototransformation significantly reduced the predicted environmental concentrations of propranolol in the water phase. Predicted reductions of mean concentrations in the River Aire (UK) were 27% in summer and 3% in winter; and for the US rivers simulated, reductions were 28–68% in summer and 11–41% in winter. The highest reductions were predicted for long rivers with low turbidity and low flow conditions.

Nano- and micro-particulate formulations of poorly water-soluble drugs by using a novel optimized technique

A novel technique for the production of nano- and micro-particulate formulations of poorly water-soluble drugs has been developed. This technique involves the use of static mixer elements to provide fast precipitation by continuous turbulent mixing of two liquid flows, an aqueous phase and an organic phase, respectively. The objective of this study was to develop the mixer technique by investigating the influence of the element number on the particle size of the resulting dispersions. Four model active pharmaceutical ingredients (APIs) with a variety of polymers, lipids or surfactants underwent intensive mixing and the final suspensions showed a narrow size distribution. Parameters such as the flow rate and the temperature of the precipitated organic–aqueous phases were also significant in the reduction of particle size. Further development of the mixing technique led to reproducible and stable formulations with minimal excipient amounts. These formulations were spray- or freeze-dried to improve stability.

Continuous Grinding Kinetics of Ethenzamide Particles by Fluidized-Bed Jet-Milling

ABSTRACTContinuous grinding kinetics of Ethenzamide powder, as a model active pharmaceutical ingredient (API) was investigated by fluidized-bed jet-milling. Because the oversize fractions after the classification were well fitted by a modified Rosin-Rammler distribution function, an equation of grade efficiency curve was obtained, which was also characteristic of API. A continuous grinding model was developed on the basis of a batch model by using 1st Kapur function relating grinding rate, the grade efficiency curve, and the overall process flow model consisting of grinding, classification, and mixing zones. The residual ratio obtained was well fitted to the experimental results except for the particle size range smaller than 4 μm and larger than 100 μm. Furthermore, because the volume of the active grinding zone adopted as the fitting parameter was found to be 5 cm3 in all experiments and the value was considered to be appropriate dimensionally, this result supports the reliability of the model.

Deliquescence in Binary Mixtures

Deliquescence is a first-order phase transformation from solid to solution and occurs at a specific relative humidity (RH) that is characteristic to the solid. The goal of this research was to investigate the reduction in critical relative humidity (RH0) in binary solid mixtures of deliquescent active pharmaceutical ingredients (APIs) and deliquescent excipients. The RH where deliquescence is induced, RH0, was measured using a gravimetric water vapor sorption balance. Values were obtained for model deliquescent APIs and excipients as well as for their binary mixtures. Model APIs included ranitidine HCl and diphenhydramine HCI, and sucrose and sodium chloride were examples of excipients investigated. To probe the thermodynamics of this phenomenon, water activity (a(w)) of various saturated solutions of API and excipient systems was measured using a water activity meter. Optical microscopy was used to observe visually the phenomenon under investigation. The Ross equation was used to estimate RH0mix, and predicted values were compared with experimental results. There was close agreement between RH0 measurements (single-component RH0, and RH0mix) and a(w) measurements of the corresponding saturated solutions. In addition, RH0mix values were always lower than RH0 for the API and excipients alone. In general, experimentally observed RH0mix values were higher than those predicted using the Ross equation. Mixtures of deliquescent API and deliquescent excipient are more hygroscopic (i.e., have lower RH0) than either the API or the excipient alone. This might have significant effects on API and drug product stability.

Improvement of dissolution rates of poorly water soluble APIs using novel spray freezing into liquid technology.

To develop and demonstrate a novel particle engineering technology, spray freezing into liquid (SFL), to enhance the dissolution rates of poorly water-soluble active pharmaceutical ingredients (APIs). Model APIs, danazol or carbamazepine with or without excipients, were dissolved in a tetrahydrofuran/water cosolvent system and atomized through a nozzle beneath the surface of liquid nitrogen to produce small frozen droplets, which were subsequently lyophilized. The physicochemical properties of the SFL powders and controls were characterized by X-ray diffraction, scanning electron microscopy (SEM), particle size distribution, surface area analysis, contact angle measurement, and dissolution. The X-ray diffraction pattern indicated that SFL powders containing either danazol or carbamazepine were amorphous. SEM micrographs indicated that SFL particles were highly porous. The mean particle diameter of SFL carbamazepine/SLS powder was about 7 microm. The surface area of SFL danazol/poloxamer 407 powder was 11.04 m2/g. The dissolution of SFL danazol/poloxamer 407 powder at 10 min was about 99%. The SFL powders were free flowing and had good physical and chemical stability after being stored at 25 degrees C/60%RH for 2 months. The novel SFL technology was demonstrated to produce nanostructured amorphous highly porous particles of poorly water soluble APIs with significantly enhanced wetting and dissolution rates.

The transformation of color-mixture equations from one system to another

Evaporative crystallization is widely applied in several industrial processes, including the pharmaceutical industry. Microwave irradiation can significantly speed up solvent evaporation in these crystallization processes, resulting in reduced particle size due to rapid crystallization. A single-mode microwave setup was used for evaporative crystallization of the model active pharmaceutical ingredient, niflumic acid, and the polymer, polyvinylpirrolidone (PVP). Production of crystals by microwave irradiation offers a modern way for drug formulation, and by reducing the particle size the dissolution rate and bioavailability of the active pharmaceutical ingredient can be enhanced. In this study, a 2.5-fold increase in the dissolution rate of the produced niflumic acid crystals was observed compared to the dissolution rate of the original drug in 120 min. When niflumic acid was produced together with the PVP in the microwave system, an amorphous solid dispersion was created with particles in the nano-size range, which showed a 5-fold increase in dissolution rate in 120 min compared to the dissolution of the crystalline niflumic acid samples created by the microwave irradiation in the absence of PVP.
