A Review and Bibliometric Analysis of Studies on Advances in Peripheral Nerve Regeneration.

Peripheral nerve injuries (PNIs) pose significant clinical challenges due to their complex healing processes and the often incomplete functional recovery. This review and bibliometric analysis aimed to provide a comprehensive overview of advancements in peripheral nerve regeneration research, focusing on trends, influential studies, and emerging areas. By analyzing 2921 publications from the Web of Science Core Collection, key themes such as nerve regeneration, repair, and the critical role of Schwann cells were identified. The study highlights a notable increase in research output since the early 2000s, with China and the United States leading in publication volume and citations. The analysis also underscores the importance of collaborative networks, which are driving innovation in this field. Despite significant progress, the challenge of achieving complete functional recovery from PNIs persists, emphasizing the need for continued research into novel therapeutic strategies. This review synthesizes current knowledge on the mechanisms of nerve regeneration, including the roles of cellular and molecular processes, neurotrophic factors, and emerging therapeutic approaches such as gene therapy and stem cell applications. Additionally, the study revealed the use of nanotechnology, biomaterials, and advanced imaging techniques, which hold promise for improving the outcomes of nerve repair. This bibliometric analysis not only maps the landscape of peripheral nerve regeneration research but also identifies opportunities for future investigation. This study has some limitations, including reliance on the Web of Science Core Collection, which may exclude relevant research from other databases. The analysis is predominantly English-based, potentially overlooking significant non-English studies. Citation trends might be influenced by shifting research priorities and accessibility issues, affecting the visibility of older work. Additionally, geographical disparities and limited collaboration networks may restrict the global applicability and knowledge exchange in this field.

Hydrogels for Neural Regeneration: Exploring New Horizons.

Nerve injury can significantly impair motor, sensory, and autonomic functions. Understanding nerve degeneration, particularly Wallerian degeneration, and the mechanisms of nerve regeneration is crucial for developing effective treatments. This manuscript reviews the use of advanced hydrogels that have been researched to enhance nerve regeneration. Hydrogels, due to their biocompatibility, tunable properties, and ability to create a supportive microenvironment, are being explored for their effectiveness in nerve repair. Various types of hydrogels, such as chitosan-, alginate-, collagen-, hyaluronic acid-, and peptide-based hydrogels, are discussed for their roles in promoting axonal growth, functional recovery, and myelination. Advanced formulations incorporating growth factors, bioactive molecules, and stem cells show significant promise in overcoming the limitations of traditional therapies. Despite these advancements, challenges in achieving robust and reliable nerve regeneration remain, necessitating ongoing research to optimize hydrogel-based interventions for neural regeneration.

Co-contraction in patients with obstetric palsy (literature review)

The frequency of obstetric palsy ranges from 0.5 to 3.0 cases per 1000 live births. The functional outcome disorders are determined by the nature of the damage and the number of damaged roots of the brachial plexus. There are 50 % of patients have complete recovery, 15 % have severe functional disorders of the upper limb, and 35 % have some limitation of the shoulder joint function. There are 3 variants of residual phenomena in children with obstetric palsy: paresis, paralysis, and co‑contraction. The article describes the mechanisms of nerve regeneration in children and adults, the pathogenesis of contractions and motor disorders in general in children with obstetric palsy, and modern methods of treatment of co‑contraction. This article will be useful for neurologists, rehabilitation specialists, and orthopedists.

Amniotic Fluid Stem Cells Improve Rat Bladder Dysfunction After Pelvic Nerve Transection

The effects of human amniotic fluid stem cells (hAFSCs) transplantation on bladder dysfunction after pelvic nerve transection (PNT) remain to be clarified. Five groups of female Sprague-Dawley rats were studied including sham operation, unilateral PNT alone or plus hAFSCs transplantation, and bilateral PNT alone or plus hAFSCs transplantation. hAFSCs were injected at the site of PNT. Cystometries, neurofilament density within bladder nerves, and the expressions of bladder protein gene-product 9.5 (PGP9.5), growth-associated protein 43 (GAP-43), nerve growth factor (NGF), p75 (NGF receptor), CXCL12, CCL7, and enkephalin were studied. Compared to sham-operation group, bladder weight increased and neurofilament density decreased at 10 and 28 days after unilateral and bilateral PNT, but all improved after hAFSCs transplantation. Unilateral PNT could increase bladder capacity, residual volume, and number of nonvoiding contractions but decrease peak voiding pressure and leak point pressure. Bilateral PNT caused overflow incontinence and increased the number of nonvoiding contractions. These cystometric parameters improved after hAFSCs transplantation. After PNT, bladder PGP9.5 mRNA and immunoreactivities decreased at 10 and 28 days, GAP-43 mRNA and immunoreactivities increased at 10 days and decreased at 28 days, both NGF and p75 mRNAs and immunoreactivities increased at 10 and/or 28 days, and enkephalin immunoreactivities decreased at 10 and 28 days, but these were all improved after hAFSCs transplantation. Our results showed that bladder dysfunction induced by PNT could be improved by hAFSCs transplantation, and PGP9.5, GAP-43, and neurotrophins could be involved in the mechanisms of nerve regeneration after hAFSCs transplantation.

End-to-side neurorrhaphy for nerve repair and function rehabilitation

BackgroundEnd-to-side neurorrhaphy is a promising procedure for nerve repair in peripheral nerve injury. However, in previous studies, this technique was limited to somatic nerves. The present study was designed to investigate the feasibility of nerve regeneration after end-to-side neurorrhaphy between autonomic nerve and somatic nerve. Materials and methodsThirty adult male Sprague–Dawley rats were randomly divided into the following three groups (n = 10 per group) for different treatments: (1) end-to-side neurorrhaphy group, the left L6 and S1 spinal nerves were transected in the dura, and the distal stump of L6 ventral root (L6VR) was sutured to the lateral face of L4 ventral root (L4VR) through end-to-side coaptation; (2) no repair group, the rats received the same operation as the end-to-side neurorrhaphy group but without coaptation; (3) control group, the rats received the same operation as the end-to-side neurorrhaphy group but the L6VR was preserved. After 4 month, the origin and mechanism of nerve regeneration were evaluated by retrograde nerve tracing. Morphologic and functional properties of the regenerated nerve were investigated by morphologic examination and intravesical pressure measurement. ResultsRetrograde nerve tracing indicated that the new neural reflex pathway was successfully established, and the main regeneration mechanism was axon collateral sprouting. Morphologic examination and intravesical pressure measurement indicated prominent axonal regeneration and good bladder functional rehabilitation in the neurorrhaphy group. Wet weight and morphology of left extensor digitorum longus muscles appeared no detrimental effect on the donor nerve. ConclusionsThese results indicated that the somatic motor axons growth into autonomic nerve may be achieved through axon collateral sprouting for nerve repair and function rehabilitation after end-to-side neurorrhaphy of autonomic nerve and somatic nerve without apparent impairment of the donor somatic nerve.

Re-Innervation of the Bladder through End-to-Side Neurorrhaphy of Autonomic Nerve and Somatic Nerve in Rats

End-to-side neurorrhaphy is widely used in the peripheral nervous system for nerve repair; however, the application of this technique has been limited to somatic nerves. The feasibility of nerve regeneration through end-to-side neurorrhaphy between autonomic and somatic nerves with different characteristics in the peripheral nervous system is still undetermined. In this study, rats were divided into three groups for different treatments (n=10 per group). In the end-to-side neurorrhaphy group, left L6 and S1 were transected in the dura, and the distal stump of L6 ventral root was sutured to the lateral face of L4 ventral root through end-to-side coaptation. In the no repair group, the rats did not undergo neurorrhaphy. In the control group, the left L6 dorsal root and S1 roots were transected, respectively, but the L6 ventral root was kept intact. After 16 weeks, the origin and mechanism of nerve regeneration was evaluated by retrograde double labeling technique as well as histological examination and intravesical pressure measurement. Retrograde double labeling indicated that the reconstructed reflex pathway was successfully established and the primary regeneration mechanism involved axon collateral sprouting. Morphological examination and intravesical pressure measurement indicated prominent nerve regeneration and successful re-innervation of the bladder in the neurorrhaphy group, compared with the "no repair" group (p<0.05). No significant changes were observed in the histology of the donor nerve and the bilateral extensor digitorum longus muscles in the neurorrhaphy group. Nerve regeneration may be achievable for nerve repair through end-to-side neurorrhaphy between autonomic and somatic nerves without apparent impairment of donor somatic nerve.

Neurotoxins and neurotoxicity mechanisms. an overview

Neurotoxins represent unique chemical tools, providing a means to 1) gain insight into cellular mechanisms of apopotosis and necrosis, 2) achieve a morphological template for studies otherwise unattainable, 3) specifically produce a singular phenotype of denervation, and 4) provide the starting point to delve into processes and mechanisms of nerve regeneration and sprouting. There are many other notable uses of neurotoxins in neuroscience research, and ever more being discovered each year. The objective of this review paper is to highlight the broad areas of neuroscience in which neurotoxins and neurotoxicity mechanism come into play. This shifts the focus away from neurotoxins per se, and onto the major problems under study today. Neurotoxins broadly defined are used to explore neurodegenerative disorders, psychiatric disorders and substance use disorders. Neurotoxic mechanisms relating to protein aggregates are indigenous to Alzheimer disease, Parkinson's disease. NeuroAIDS is a disorder in which microglia and macrophages have enormous import. The gap between the immune system and nervous system has been bridged, as neuroinflammation is now considered to be part of the neurodegenerative process. Related mechanisms now arise in the process of neurogenesis. Accordingly, the entire spectrum of neuroscience is within the purview of neurotoxins and neurotoxicity mechanisms. Highlights on discoveries in the areas noted, and on selective neurotoxins, are included, mainly from the past 2 to 3 years.

Neurophysiological and behavioral evidence for selective reinnervation in skin-grafted Rana pipiens

Rana pipiens with skin dorosoventrally reversed can respond to stimulation of the back with forelimb wipes to the belly and to stimulation of the belly with hindlimb wipes to the back. These "misdirected wiping responses" have been explained in terms of two alternative hypotheses of nerve regeneration: nerve respecification or selective reinnervation. Experimental behavioral and neurophysiological experiments reported here support the selective reinnervation hypothesis. Severing ventral nerves, which normally innervate the belly, greatly reduced the percentage of misdirected responses on stimulation of belly skin grafted to the back, while severing dorsal nerves, which normally innervate the back, increased the percentage of misdirected responses elicited under the same circumstances. Moreover, neurophysiological recordings of grafted animals showed three effects of skin grafting on nerve distributions: (i) termination of dorsal and ventral nerve receptive field at graft edges; (ii) overlap of nonadjacent ventral nerve receptive fields; and (iii) dorsal coursing of ventral nerves to reinnervate target belly skin displaced to the back. These neurophysiological observations, and particularly the third effect, also support selective reinnervation as the mechanism of nerve regeneration in skin-grafted Rana pipiens.
