EFFECTIVENESS OF COMPARATIVE GOAT MANURE FERTILIZER ON THE GROWTH OF COFFEE SEEDS FROM ARABICA VARIETIES (Coffea arabica L)

Seed quality is determined by good seedling techniques, one of which is the use of planting media. Where the planting media can use soil media with animal waste. The purpose of this study to determine the effectiveness of goat manure seedling media on the growth of Arabica coffee seedlings and to obtain the best comparison of goat manure seedling media on the accretion of Arabica coffee seedlings. This research method used a completely randomized design consisting of one factor and four treatments, namely soil 1:0 (M0), soil + goat manure 1:1 (M1), soil + goat manure 1:2 (M2), and soil + goat Manure 1:3 (M3). Variables observed: percentage, plant height, number of leaves. The data obtained were analyzed using the ANOVA if significantly continued with DMRT at the 5% level. The results showed the effectiveness of goat manure seedling media on the growth of arabica coffee seedlings, with a growth percentage of 97%. The best comparison of goat manure seedling media on the growth parameters of arabica coffee seedlings in the M3 treatment with a mixture of soil and goat manure ratio (1:3), the average plant height (14.19 cm), and the number of leaves (7.0 strands).

Test The Effectiveness Volume Of Hydroton Media On Mustard Plant (Brassica Chinensis Var. Parachinensis) Using Auto Kapiler

This study aims to determine the level of effectiveness the hydroton growing media on mustard plants using the Auto Kapiler hydroponic method with 3 treatments; M1 with hydroton growing media of 2,54 liters (volume 50%), M2 with hydroton growing media of 3,78 liters (75% volume), and M3 with hydroton growing media of 5,04 liters (100% volume). The research was conducted on Jl. Tirtonadi RT. 25 / RW. 04, Jatikerto village, Kromengan sub-district, Malang district which will take place from April 25 to June 14, 2022. Some of the materials and tools used during the research are; hydroton growing media, auto capillary, AB-Mix nutrition, pH meter, and EC meter. Variable observations made consist of; plant height (cm), number of leaves (strands), leaf width (cm), root length (cm), and plant fresh weight (grams). Data analysis in this study used a simple Completely Randomized Design (CRD) with 4 replications and 3 treatments. The data obtained were further analyzed using the least significant Least Significant Different (LSD) α 5% level which was then further analyzed using a linear regression. The results showed that the M3 treatment (100% hydroton growing media volume was 5,04 liters) was better than the M2 treatment (75% hydroton growing media volume 3,78 liters) and M1 (50% hydroton growing media volume 2,52 liter).

Effect of the Variations of Molasses Concentration And Corn Flour on Growing Media for White Oyster Mushroom (Pleurotus Ostreatus) Productivity

Oyster mushroom (Pleurotus ostreatus) can grow well on materials that contain carbohydrates. One of the fungal growth ingredients that contain carbohydrates is corn flour. In addition to corn flour, molasses can also be used to stimulate the growth of white oyster mushrooms. The purpose of this study was to see the effect of variations in the concentration of molasses and corn flour on the growing media on the productivity of white oyster mushroom (Pleurotus ostreatus). The research was conducted in May-July 2022 at the Center for Breeding and Development of Oyster and Ear Mushroom Cultivation on Sunggal District, Deli Serdang Regency. The research method used was a factorial completely randomized design (CRD) with 2 factors, 16 treatment combinations and 2 replications so that there were 32 baglogs. The first factor contained 4 levels of treatment, namely M0: 0 ml/baglog, M1: 7.5 ml/baglog, M2: 15 ml/baglog and M3: 22.5 ml/baglog. The second factor has 4 levels of treatment, namely J0: 0 gr/baglog, J1: 10 gr/baglog, J2: 20 gr/baglog and J3: 30 gr/baglog. Parameters observed were mycelium growth rate (cm), stalk height (cm), hood diameter (cm), hood thickness (mm), number of branches/clumps, wet weight (g). The results of the highest data mycelium growth rate in treatment J0 of 20.75 cm. The result of the highest data stalk height in the M1 treatment was 6.19 cm. The results of the highest data hood diameter in treatment J0 of 10.18 cm. The result of the highest data hood thickness in treatment J2 was 5.09 mm. The result of the highest number of branches/data clusters in the M3 treatment was 9.13 branches. The result of the highest wet weight (g) data in the M3 treatment was 140.75 gr.

A novel eCIRP/TREM-1 pathway inhibitor attenuates acute kidney injury

BackgroundExtracellular cold-inducible RNA-binding protein aggravates acute kidney injury after renal ischemia/reperfusion. Although extracellular cold-inducible RNA-binding protein activates triggering receptor expressed on myeloid cells-1, how this receptor and its antagonism with a novel peptide M3 affects acute kidney injury is poorly understood. We, therefore, hypothesize that inhibiting the extracellular cold-inducible RNA-binding protein/triggering receptor expressed on myeloid cells-1 pathway with M3 attenuates acute kidney injury. MethodsWild-type and triggering receptor expressed on myeloid cells-1-/- mice were subjected to bilateral 30-minute renal hilum clamping followed by reperfusion or sham. After 4 hours, wild-type mice received M3 (10 mg/kg BW) or normal saline intraperitoneally. After 24 hours, renal tissue and serum were collected for analysis. Additionally, wild-type mice were subjected to bilateral renal ischemia for 34 minutes and treated with M3 at 10 mg/kg BW or vehicle at the time of reperfusion. Survival was monitored for 10 days. ResultsAfter renal ischemia/reperfusion, triggering receptor expressed on myeloid cells-1 messenger ribonucleic acid expression increased by 9-fold in wild-type mice compared to sham mice. Wild-type mice also demonstrated significant increases in serum blood urea nitrogen, creatinine, and interleukin-6 and renal tissue levels of interleukin-6 and neutrophil gelatinase-associated lipocalin after renal ischemia/reperfusion compared to sham mice. Triggering receptor expressed on myeloid cells-1-/- mice demonstrated significant reductions in serum blood urea nitrogen, creatinine, and interleukin-6 compared to wild-type mice after renal ischemia/reperfusion. Levels of renal interleukin-6 and neutrophil gelatinase-associated lipocalin were also significantly decreased in the kidneys of triggering receptor expressed on myeloid cells-1-/- mice. Furthermore, treatment with M3 in wild-type mice significantly decreased serum and renal levels of interleukin-6 after renal ischemia/reperfusion. M3 treatment demonstrated significant reductions in renal messenger ribonucleic acid and protein levels of neutrophil gelatinase-associated lipocalin, serum blood urea nitrogen and creatinine, and histologic structural damage as well as apoptosis. Treatment with M3 also increased survival from 35% to 65% in mice with acute kidney injury. ConclusionTriggering receptor expressed on myeloid cells-1 mediates the deleterious effects of extracellular cold-inducible RNA-binding protein in acute kidney injury after renal ischemia/reperfusion. The novel extracellular cold-inducible RNA-binding protein/triggering receptor expressed on myeloid cells-1 pathway antagonist, M3, attenuates acute kidney injury and has the potential to be developed as a therapeutic agent for acute kidney injury.

Artificial intelligence in positioning between mandibular third molar and inferior alveolar nerve on panoramic radiography

Determining the exact positional relationship between mandibular third molar (M3) and inferior alveolar nerve (IAN) is important for surgical extractions. Panoramic radiography is the most common dental imaging test. The purposes of this study were to develop an artificial intelligence (AI) model to determine two positional relationships (true contact and bucco-lingual position) between M3 and IAN when they were overlapped in panoramic radiographs and compare its performance with that of oral and maxillofacial surgery (OMFS) specialists. A total of 571 panoramic images of M3 from 394 patients was used for this study. Among the images, 202 were classified as true contact, 246 as intimate, 61 as IAN buccal position, and 62 as IAN lingual position. A deep convolutional neural network model with ResNet-50 architecture was trained for each task. We randomly split the dataset into 75% for training and validation and 25% for testing. Model performance was superior in bucco-lingual position determination (accuracy 0.76, precision 0.83, recall 0.67, and F1 score 0.73) to true contact position determination (accuracy 0.63, precision 0.62, recall 0.63, and F1 score 0.61). AI exhibited much higher accuracy in both position determinations compared to OMFS specialists. In determining true contact position, OMFS specialists demonstrated an accuracy of 52.68% to 69.64%, while the AI showed an accuracy of 72.32%. In determining bucco-lingual position, OMFS specialists showed an accuracy of 32.26% to 48.39%, and the AI showed an accuracy of 80.65%. Moreover, Cohen’s kappa exhibited a substantial level of agreement for the AI (0.61) and poor agreements for OMFS specialists in bucco-lingual position determination. Determining the position relationship between M3 and IAN is possible using AI, especially in bucco-lingual positioning. The model could be used to support clinicians in the decision-making process for M3 treatment.

The effect of mycorrhizal and phosphorous fertilizer applications on soybean plant production (Glycine max L. Meriil)

This study aims to determine the production of soybeans with the application of mycorrhizal biological fertilizers and P fertilizers on the production of soybeans (Glycine max L. Meriil). This experiment was carried out at the Experimental Garden of the Faculty of Agriculture, Indonesian Methodist University, from August–October 2018. This study used a Randomized Block Design (RAK) consisting of Mycorrhiza (M) namely M0: 0 g/plant (control); M1: 5 g/plant; M2: 10 g/plant; M3: 15g/plant and Fertilizer (P), namely P0: 0 g/plant (control); P1:0.25 g/plant; P2: 0.5 g/plant; P3: 0.75 g/plant. All treatment combinations were 12 combinations and each treatment combination was repeated 3 times. Variables observed in this study include: flowering age (days); seed production per sample (g); seed production per plot (g); dry weight of 100 seeds (g). The experimental results showed that the interaction between mycorrhizal application and application of NPK fertilizer had a significant effect on seed production per sample, seed production per plot, dry weight of 100 seeds and not significantly different with flowering age. The highest seed production per sample was found in the M3P3 treatment of 26.32 g, the highest seed production per plot was found in the M3P2 treatment of 161.80 g and the highest dry weight of 100 seeds was found in the M3P3 treatment of 25.60 g. The mycorrhizal biofertilizer application treatment showed that the fastest flowering time was found in the M3 treatment, namely 36.58 days and 36.92 days in the P (P3) fertilizer treatment.

Inhibition of the Interaction of TREM-1 and eCIRP Attenuates Inflammation and Improves Survival in Hepatic Ischemia/Reperfusion.

Triggering receptor expressed on myeloid cells-1 (TREM-1) has important implications in sepsis and inflammation and is a novel receptor for extracellular cold-inducible RNA-binding protein (eCIRP). We hypothesize that the inhibition of TREM-1 via its interaction with eCIRP by novel peptide inhibitor M3 or knockout gene will attenuate the inflammation and injury associated with severe hepatic ischemia/reperfusion (I/R). Wild-type (WT) C57BL/6 and TREM-1-/- mice underwent 60 min of 70% hepatic ischemia, with 24 h of reperfusion. Additionally, WT mice underwent hepatic I/R and were treated with M3 (10 mg/kg body weight) or vehicle (normal saline) at the start of reperfusion. Blood and ischemic liver tissues were collected, and analysis was performed using enzymatic assays, enzyme-linked immunosorbent assay, reverse-transcription quantitative polymerase chain reaction, and pathohistology techniques. For survival surgery, mice additionally underwent resection of non-ischemic lobes of the liver and survival was monitored for 10 days. There was an increase in serum levels of tissue markers including aspartate aminotransferase, alanine aminotransferase, and lactate dehydrogenase as well as cytokine levels (IL-6) and histological scoring of hematoxylin and eosin sections in WT I/R mice. These markers decreased substantially in TREM-1-/- mice. Additionally, neutrophil infiltration markers and markers of local inflammation (myeloperoxidase, macrophage inflammatory protein-2, cyclooxygenase-2) were attenuated in TREM-1-/- mice. Similarly, we show a significant decrease in injury and inflammation markers with M3 treatment. Additionally, we demonstrate decreased apoptosis with TREM-1 inhibition. Finally, M3 treatment improved the survival rate from 42% to 75% after hepatic I/R. TREM-1 is an important eCIRP receptor in the inflammatory response of hepatic I/R, and deficiency of TREM-1 via knockout gene or peptide inhibition attenuated liver injury and inflammation, and improved survival. Inhibition of the TREM-1 and eCIRP interaction in hepatic I/R may have important therapeutic potential.

Potassium and Water-Deficient Conditions Influence the Growth, Yield and Quality of Ratoon Sugarcane (Saccharum officinarum L.) in a Semi-Arid Agroecosystem

Groundwater and soil potassium deficiencies are present in northern India. Sugarcane is a vital crop in the Indian Punjab; it is grown on approximately 91,000 hectares with an average yield of 80 tonnes ha−1 and a sugar recovery rate of 9.59%. The role of potassium (K) fertilizer under both sufficient and deficient irrigation in ratoon sugarcane crops is not well documented. We conducted a split-plot ratoon cane experiment during 2020–2021 at the Gurdaspur Regional Research Station of Punjab Agricultural University, India, on K-deficient soils. Main treatments were fully irrigated (I1) and water stressed (I0) conditions, with sub-treatments reflecting K fertilizer application rates of 0 (M1), 67 (M2), 133 (M3), and 200 (M4) kg K ha−1. The ratoon sugarcane performance was assessed in terms of growth, productivity, sugar quality and incidence of key insect pests. At harvest, trends in the growth and yield parameters in I1 were improved over the I0 treatment, with cane height (+12.2%), diameter (+3.3%), number of internodes (+5.4%), biomass yield (+7.6%) and cane yield (+5.9%) all higher, although little significant difference was observed between treatments. Ratoon cane yield under irrigation was 57.1 tonnes ha−1; in water-stressed conditions, it was 54.7 tonnes ha−1. In terms of sugarcane quality parameters, measured 12 months after harvesting the initial seed crop, values of Brix (+3.6%), pol (+3.9%), commercial cane sugar percentage (+4.0%) and extractable sugar percentage (+2.8%) were all higher in the irrigated treatments than the water-stressed plot. Irrigated treatments also had a significantly lower incidence of two key insect pests: top borer (Scirpophaga excerptalis) was reduced by 18.5% and stalk borer (Chilo auricilius) by 21.7%. The M3 and M4 treatments resulted in the highest cane yield and lowest incidence of insect pests compared to other K-fertilizer treatments. Economic return on K-fertilizer application increased with increasing fertilizer dosage. Under the potassium-deficient water-stressed conditions of the region of north India, a fertilizer application rate of 133 kg K ha−1 is recommended to improve ratoon sugarcane growth, yield, and quality parameters and economic returns for sugarcane farmers.

Effectiveness of biochar and compost on improving soil hydro-physical properties, crop yield and monetary returns in inceptisol subtropics.

Organic manures in combination with biochar might improve efficacy of biochar in improving soil functions related to hydro-physical properties and a field experiment was conducted over the course of two years with two levels of biochar @ 0 and 2tha−1 and four levels of compost (100% recommended dose of N through farm yard manure, 100% recommended dose of N through vermicompost, 50% recommended dose of N through farm yard manure, and vermicompost each, and unfertilized control). Each treatment was replicated three times in factorial randomized block design (RBD). The objective of this research was to determine the effects of biochar and compost on soil hydro-physical properties, water use efficiency, monetary returns and yield of knolkhol (Brassica oleracea var. gongyloides L.) under sub-tropics of North West India. Compared with no-biochar, application of biochar significantly increased knolkhol yield by 7.8% and soil properties (infiltration rate, aggregate stability, maximum water holding capacity and hydraulic conductivity). Similarly, integration of compost significantly enhanced the soil water retention, aggregate stability, hydraulic conductivity and crop yield and gave highest infiltration rate, water retention, hydraulic conductivity and crop yield under M3 (50 % N through farm yard manure, +50 % N through vermicompost) treatment. Furthermore, synergetic positive effect of biochar and compost were noted for soil infiltration rate (4–38%), water retention (0.9–13.7%), aggregate stability (6–10.7%) and yield (6–11.9%) over the sole application of compost. Combined use of farm yard manure, and vermicompost accompanied by biochar resulted in highest net returns and B:C ratio. Biochar in combination with farm yard manure, and vermicompost can enhance soil hydraulic properties resulting in increased crop yield and maximum monetary returns under subtropical conditions.

Potensi <i>Murraya keonigii</i> sebagai Herbisida Alami

Various kinds of agricultural products are needed to support organic farms to produce superior quality products, one of it was natural herbicides. Some plants have the potential to become natural herbicides by producing chemical compounds, namely allelopathy which can inhibit the growth of surrounding plants and Murraya keonigii is one of it. This study aims to see the potential of M. keonigii to become a natural herbicide. Grass seeds were given M keonigii’s extract at a dose of 0 gr / l (control / M0), 50 gr / lt (M1), 60 gr / lt (M2), and 70 gr / lt (M3). The results showed that the aromatic or allelopathic compounds in M ​​keonigii affected the germination. Germination was slower at doses of 60 gr / l (M2) and 70 gr / lt (M3) compared to controls (M0) and 50 gr / lt (M1). Radicle length did not increase rapidly in M2 and M3 treatment. Germination power of grass seeds in control and M0 had a value of 100% or all germinated, while M1 and M2 had 67% germination. The higher the dose of M keonigii extract, the slower the process of grass seed germination.

Pengaruh Waktu Pengomposan Media Tanam terhadap Pertumbuhan dan Hasil Jamur Tiram Putih (Pleorotus ostreatus)

The cultivation of consumption mushrooms in Indonesia is showing an encouraging development. Currently, Indonesia is included as one of the world's major mushroom suppliers. Mushrooms are a source of vegetable protein that does not contain cholesterol and can prevent high blood pressure, heart disease, diabetes, and can reduce body weight. The research objective was to determine the effect of composting time on planting media on the growth and yield of white oyster mushrooms. The research was conducted in the Mapin Kebak Village, sub-district Alas Barat star from May to June 2020. The research was conducted using an experimental method and using a completely randomized design consisting of 5 treatments, where each treatment consisted of three replications. The research treatments consisted of M1 = composting for 6 hours; M2 = composting for 12 hours; M3 = composting for 24 hours; M4 = composting for 36 hours and M5 = composting for 48 hours. Furthermore, the data from the observations were analyzed by ANOVA at the real level of 5%. If the results of the data analysis show a significant difference, then a further test is carried out using the Least Significant Difference Test at the 5% real level. The results of the analysis showed that the fastest time to spread mycelium was in the M5 treatment, the composting time was 48 hours. The fastest primordia emergence time was 24 hours composting M3 treatment. The heaviest fruit weight was in treatment M3 and the lowest fruit weight was in treatment M1.

Targeting the eCIRP/TREM-1 interaction with a small molecule inhibitor improves cardiac dysfunction in neonatal sepsis

BackgroundNeonatal sepsis and the associated myocardial dysfunction remain a leading cause of infant mortality. Extracellular cold-inducible RNA-binding protein (eCIRP) acts as a ligand of triggering receptor expressed on myeloid cells-1 (TREM-1). M3 is a small CIRP-derived peptide that inhibits the eCIRP/TREM-1 interaction. We hypothesize that the eCIRP/TREM-1 interaction in cardiomyocytes contributes to sepsis-induced cardiac dysfunction in neonatal sepsis, while M3 is cardioprotective.MethodsSerum was collected from neonates in the Neonatal Intensive Care Unit (NICU). 5–7-day old C57BL/6 mouse pups were used in this study. Primary murine neonatal cardiomyocytes were stimulated with recombinant murine (rm) CIRP with M3. TREM-1 mRNA and supernatant cytokine levels were assayed. Mitochondrial oxidative stress, ROS, and membrane potential were assayed. Neonatal mice were injected with rmCIRP and speckle-tracking echocardiography was conducted to measure cardiac strain. Sepsis was induced by i.p. cecal slurry. Mouse pups were treated with M3 or vehicle. After 16 h, echocardiography was performed followed by euthanasia for tissue analysis. A 7-day survival study was conducted.ResultsSerum eCIRP levels were elevated in septic human neonates. rmCIRP stimulation of cardiomyocytes increased TREM-1 gene expression. Stimulation of cardiomyocytes with rmCIRP upregulated TNF-α and IL-6 in the supernatants, while this upregulation was inhibited by M3. Stimulation of cardiomyocytes with rmCIRP resulted in a reduction in mitochondrial membrane potential (MMP) while M3 treatment returned MMP to near baseline. rmCIRP caused mitochondrial calcium overload; this was inhibited by M3. rmCIRP injection impaired longitudinal and radial cardiac strain. Sepsis resulted in cardiac dysfunction with a reduction in cardiac output and left ventricular end diastolic diameter. Both were improved by M3 treatment. Treatment with M3 attenuated serum, cardiac, and pulmonary levels of pro-inflammatory cytokines compared to vehicle-treated septic neonates. M3 dramatically increased sepsis survival.ConclusionsInhibition of eCIRP/TREM-1 interaction with M3 is cardioprotective, decreases inflammation, and improves survival in neonatal sepsis.Trial registration Retrospectively registered.

PENGARUH WAKTU PEMBERIAN MIKRO ORGANISME LOKAL TERHADAP PERTUMBUHAN DAN HASIL TANAMAN KUBIS (Brasicca oleraceae L.)

This research aims at finding out the effect of the treatment period of the local microorganism towards the growth and yield of cabbage plants (Brassica oleraceae L.) and was conducted in the experiment garden of Faculty of Agriculture Flores University in Lokoboko District, East Ndona Subdistrict, Ende Regency. The design used in this experiment is Randomized Block Design (RBD), with treatment used is M0 (without LMO treatment), M1 (once in 3 days LMO treatment), M2 (once in 5 days LMO treatment), and M3 (once in 7 days LMO treatment). The variable of the observation in this experiment is total leaves, fresh weight of crops per plant, fresh weight of crops per hectare, the diameter of the crops, fresh weight of residue, and the index of harvest. The result indicates that with the treatment of the period giving MOL 3 times a day shows the growth increase of plants’ total leaf in every period of observation 14hst, 21hst, 28hst, 35hst, 42hst). In all result variable showing the good increase to the result, M1 treatment fresh weight crop per plant is 1700 gr, fresh weight crop per ha are 68.00 tons, diameter crop is 19.52 cm, fresh weight of residue per plan are 1.94 kg, and harvest index is 46.70, M2 treatment fresh weight crop per plant is 1660 gr, fresh weight crops per ha are 1.88 kg, and harvest index are 46.89, M3 treatment fresh weight crop per plant is (1680 gr), fresh weight crop per ha are 67.2 tons, diameter crop is 18.92 cm, fresh weight residue per plant is (1,88 kg), and harvest index is 47.19.

Spatial-temporal distribution of winter wheat (Triticum aestivum L.) roots and water use efficiency under ridge–furrow dual mulching

In dryland agricultural production areas, knowledge on the growth and variation of winter wheat root systems under different mulching methods is important for guiding winter wheat production. We conducted a two-year field trial to explore the effects of different mulching practices on the spatial-temporal distribution of root growth, soil water storage, and grain yield. Four cultivation practices were tested: (i) traditional flat planting (CK), (ii) flat planting with half-film mulching (M1), (iii) ridge–furrow planting with film mulching over ridges only (M2), and (iv) ridge–furrow planting with film mulching over ridges and wheat straw mulching in furrows (M3). The results showed that the root diameter (RD), root length density (RLD), and root surface area density (RSD) of wheat under the four treatments were mainly concentrated in the 0–40 cm soil layer. The mean RLD in the 0–40 cm soil layer accounted for 62.2 %, 61.0 %, 59.3 %, and 55.8 % of the total root length density under M3, M2, M1, and CK, respectively, at maturity in both years. The M3 cultivation method produced the maximum values for RD, RLD, and RSD among all cultivation practices and the highest water storage efficiency after the three precipitation events. The M3 treatment had 11.1 %, 15.4 %, and 38.4 % higher mean grain yields than M2, M1, and CK, respectively, over the two-year study. Soil water storage had positive correlations with RD, RLD, and RSD. The RD, RLD, and RSD in the 0–40 cm soil layer had strong positive correlations with yield components, yield, aboveground biomass, and water use efficiency (WUE). Based on this study, ridge–furrow planting with film mulching over ridges and wheat straw mulching in furrows is an effective, sustainable cultivation method for wheat production in rainfed regions, which can increase the spatial-temporal distribution of root systems and soil water content across the root zones to increase crop production and WUE.

Methyl Jasmonate Applications From Flowering to Ripe Fruit Stages of Strawberry (Fragaria × ananassa 'Camarosa') Reinforce the Fruit Antioxidant Response at Post-harvest.

Preharvest applications of methyl jasmonate (MeJA) have been shown to improve post-harvest fruit quality in strawberry fruit. However, the effectiveness of consecutive field applications at different phenological stages on the reinforcement of the antioxidant capacity remains to be analyzed. To determine the best antioxidant response of strawberry (Fragaria × ananassa ‘Camarosa’) fruit to different numbers and timing of MeJA applications, we performed three differential preharvest treatments (M1, M2, and M3) consisted of successive field applications of 250 μmol L–1 MeJA at flowering (M3), large green (M2 and M3), and ripe fruit stages (M1, M2, and M3). Then, we analyzed their effects on fruit quality parameters [firmness, skin color, soluble solids content/titratable acidity (SSC/TA) ratio, fruit weight at harvest, and weight loss] along with anthocyanin and proanthocyanidin (PA) accumulation; the antioxidant-related enzymatic activity of catalase (CAT), guaiacol peroxidase (POX), and ascorbate peroxidase (APX); the total flavonoid and phenolic contents, antioxidant capacity, and ascorbic acid content (AAC) during post-harvest storage (0, 24, 48, and 72 h). We also evaluated the effect on lignin, total carbon and nitrogen (%C and N), lipid peroxidation, and C and N isotopes signatures on fruits. Remarkably, the results indicated that MeJA treatment increases anthocyanin and PA contents as well as CAT activity in post-harvest storage, depending on the number of preharvest MeJA applications. Also, M3 fruit showed a higher AAC compared to control at 48 and 72 h. Noticeably, the anthocyanin content and CAT activity were more elevated in M3 treatment comparing with control at all post-harvest times. In turn, APX activity was found higher on all MeJA-treated fruit independent of the number of applications. Unlike, MeJA applications did not generate variations on fruit firmness and weight, lignin contents,% C and N, and in lipid peroxidation and water/nitrogen use efficiency according to C and N isotope discrimination. Finally, we concluded that an increasing number of MeJA applications (M3 treatment) improve anthocyanin, PA, AAC, and CAT activity that could play an essential role against reactive oxygen species, which cause stress that affects fruits during post-harvest storage.

Priming of Defense Systems and Upregulation of MYC2 and JAZ1 Genes after Botrytis cinerea Inoculation in Methyl Jasmonate-Treated Strawberry Fruits.

Several attempts have been made to study the effects of methyl jasmonate (MeJA) on plants in the past years. However, the comparative effects of the number and phenological time of MeJA applications on the activation of defense systems is currently unknown in strawberries. In the present research, we performed three field treatments during strawberry (Fragaria × ananassa ‘Camarosa’) fruit development and ripening which consisted of differential MeJA applications at flowering (M3), and the large green (M2 and M3) and red ripe (M1, M2, and M3) fruit stages. We also checked changes in gene expression related to plant defense against Botrytis cinerea inoculation post-harvest. In M3 treatment, we observed an upregulation of the anthocyanin and lignin contents and the defense-related genes, encoding for chitinases, β-1,3-glucanases and polygalacturonase-inhibiting proteins, after harvest (0 hpi), along with the jasmonate signaling-related genes FaMYC2 and FaJAZ1 at 48 h after B. cinerea inoculation (48 hpi) during postharvest storage. Although we did not find differences in gray mold incidence between the MeJA treatments and control, these results suggest that preharvest MeJA treatment from the flowering stage onwards (M3) primes defense responses mediated by the upregulation of different defense-related genes and retains the upregulation of MYC2 and JAZ1 at 48 hpi.

An extracellular cold-inducible RNA-binding protein-derived small peptide targeting triggering receptor expressed on myeloid cells-1 attenuates hemorrhagic shock.

Extracellular cold-inducible RNA-binding protein (eCIRP) is a damage-associated molecular pattern, which is released into the circulation after hemorrhagic shock (HS). Recently, we discovered that triggering receptor expressed on myeloid cells-1 (TREM-1) serves as a new receptor of eCIRP to exaggerate inflammation. Here, we hypothesize that by inhibiting the interaction between eCIRP and TREM-1 with the use of a novel short peptide derived from human eCIRP known as M3, we can inhibit the inflammatory response and acute lung injury in HS. Hemorrhagic shock was induced using C57BL/6 mice by cannulating both femoral arteries. One femoral artery was used for removal of blood while the other was used for continuous monitoring of mean arterial blood pressure. The mean arterial pressure of 25 mm Hg to 30 mm Hg was maintained for 90 minutes, followed by a resuscitation phase of 30 minutes with 1 mL of normal saline. The treatment group was given 10 mg/kg of M3 during the resuscitation phase. Four hours after resuscitation, serum and lungs were collected and analyzed for various injury and inflammatory markers by using colorimetry, real-time polymerase chain reaction, and enzyme-linked immunosorbent assay. There was an increase in the serum levels of tissue injury markers (alanine aminotransferase, aspartate aminotransferase, and lactate dehydrogenase) as well as cytokines (TNF-α and IL-6) when comparing the vehicle group versus the sham group. This increase was significantly inhibited in the M3-treated group. The mRNA expression of proinflammatory cytokines TNF-α, IL-6, and IL-1β and the chemokines MIP-2 and KC in lungs was significantly increased in the vehicle-treated HS mice, while their expression was significantly decreased in M3-treated HS mice. Finally, M3 treatment significantly decreased the lung injury score compared with vehicle-treated HS mice. The novel eCIRP-derived TREM-1 antagonist (M3) can be a potential therapeutic adjunct in the management of hemorrhagic shock.

Provision of micoriza arbuscula in some types of grass to the degree of infection and phosporus uptake

This study was conducted in Faculty of Agriculture North Sumatera University. This study used a factorial completely randomized design (FCRD) consisting of two factors. The first factor was type of grass and the second factor is the microfer level with 5 treatments and 3 replications. The results of the diversity analysis showed the use of MVA significantly (P <0.05) in the degree of root infection and phospor uptake. This study use five types of grass (S) with MVA levels (M). The ratio of the degree of root is found in the M3 treatment 47.53% and the ratio of phosporus uptake is found in the M3 treatment 110.10% and the ratio of phosporus uptake is found in the S1 treatment 91.85%. From this study, it can be concluded that the best treatment is found in M3 treatment.

Effect of rhizobacteria treatments on nutrient content and organic and amino acid composition in raspberry plants

Plant growth-promoting rhizobacteria (PGPR) have been found to be beneficial to plant growth, yield, crop quality, the environment, and sustainable agricultural production. Therefore, six bacterial strains were tested to determine their effects on raspberry's nutrient content and organic and amino acid composition. The experiment was performed from 2015 to 2017. Two-year-old raspberry plants were inoculated with bacterial suspensions by a dipping method and were planted in 30-L pots. The mineral content and organic acid and amino acid composition of the leaf and root were compared in the Alcaligenes 637Ca, Staphylococcus MFDCa1 and MFDCa2, Agrobacterium A18, Pantoea FF1, and Bacillus M3 bacterial strains. Nitrogen (N) content of the leaf was 2.55% in the A18 treatment, while N content of the root was 1.61% in MFDCa2. The leaf's iron (Fe) content was highest in the M3 treatment with 91.76 mg kg-1, while 637Ca gave the highest root's Fe content with 107.80 mg kg-1. The content of malonic acid (16.78 ng µL-1), malic acid (4.59 ng µL-1), citric acid (16.88 ng µL-1), and fumaric acid (4.94 ng µL-1) in leaves was higher in MFDCa2 than in the other treatments. In addition, 637Ca treatment had the highest root organic acid content in tartaric acid (5.94 ng µL-1), butyric acid (15.19 ng µL-1), and maleic acid (5.13 ng µL-1). FF1 treatment was more effective than the other treatments for increasing the leaf?s amino acid content, while the 637Ca, MFDCa1 FF1, and M3 treatments were more effective in increasing the root?s amino acid content. As a result, it was determined that PGPR treatments play a significant role in mineral nutrient uptake and the organic acid and amino acid composition of the raspberry plant.

M3, a natural lignan xyloside, exhibits potent anticancer activity in HCT116 cells.

4-O-(2′,3′,4′-tri-O-methyl-β-D-xylopyranosyl) diphyllin (M3) is a cytotoxic compound that was first isolated from the aerial parts of Phyllanthus taxodiifolius. The current study demonstrated that M3, a lignan xyloside, exhibits anticancer effects in a number of cancer cell lines by MTT test, including HCT116 cells. An association was identified between M3 treatment and the reduced proliferation of cancer cells; the half maximal inhibitory concentration (IC50) value of M3 ranged from 0.08 to 1.12 µM. Furthermore, M3 was revealed to exhibit a stronger antiproliferative effect by Annexin-V-FLUOS test compared with VP-16, another natural lignan used in cancer treatment. Notably, the IC50 value of M3 with MTT test in HCT116 cells was 0.08 µM. In addition, it was revealed that M3 could induce apoptosis in HCT116 cells in a caspase-3-dependent manner at a lower concentration compared with VP-16. Further analysis identified that the antiproliferation effect of M3 was associated with the promotion of microtubule depolymerization by CytoDYNAMIX Screen 03 Tubulin Polymerization assay. In summary, the current study demonstrated that M3 could inhibit proliferation and induce apoptosis in HCT116 cells, which supports a potential therapeutic application of M3 in cancer treatment, particularly in colon cancer.
