Genetic Characterization and Phylogeographic Analysis of the First H13N6 Avian Influenza Virus Isolated from Vega Gull in South Korea.

Avian influenza virus (AIV) is a pathogen with zoonotic and pandemic potential. Migratory birds are natural reservoirs of all known subtypes of AIVs, except for H17N10 and H18N11, and they have been implicated in previous highly pathogenic avian influenza outbreaks worldwide. This study identified and characterized the first isolate of the H13N6 subtype from a Vega gull (Larus vegae mongolicus) in South Korea. The amino acid sequence of hemagglutinin gene showed a low pathogenic AIV subtype and various amino acid substitutions were found in the sequence compared to the reference sequence and known H13 isolates. High sequence homology with other H13N6 isolates was found in HA, NA, PB1, and PA genes, but not for PB2, NP, M, and NS genes. Interestingly, various point amino acid mutations were found on all gene segments, and some are linked to an increased binding to human-type receptors, resistance to antivirals, and virulence. Evolutionary and phylogenetic analyses showed that all gene segments are gull-adapted, with a phylogeographic origin of mostly Eurasian, except for PB2, PA, and M. Findings from this study support the evidence that reassortment of AIVs continuously occurs in nature, and migratory birds are vital in the intercontinental spread of avian influenza viruses.

Pathogenicity of highly pathogenic avian influenza H5N8 subtype for herring gulls (Larus argentatus): impact of homo- and heterosubtypic immunity on the outcome of infection

To improve understanding of the pathobiology of highly pathogenic avian influenza virus (HPAIV) infections in wild birds, pathogenicity and transmissibility of HPAIV H5N8 subtype clade 2.3.4.4b was evaluated in ~ 8-week-old herring gulls (Larus argentatus) divided into 3 groups: naïve birds (group A), birds previously exposed to low pathogenic avian influenza virus (LPAIV) H5N1 (group B) and LPAIV H13N6 (group C). The HPAIV H5N8 virus was highly virulent for naïve gulls, that showed early morbidity, high mortality, a broad spectrum of clinical signs, including violent neurological disorders, systemic distribution of the virus in organs accompanied by high level of shedding and transmission to contact birds. Pre-exposure to homologous and heterologous LPAIV subtypes conferred only partial protection: we observed increased survival rate (statistically significant only in group B), nervous signs, pantropic distribution of virus in organs, shedding (significantly reduced in gulls of group C in the early phase of disease and asymptomatic shedding in the late phase), transmission to contact gulls (more pronounced in group B) and near-complete seroconversion in survivors. Histopathological and immunohistochemical results indicate virus tropism for the neural, respiratory and myocardial tissues. In conclusion, we demonstrate that HPAIV H5N8 clade 2.3.4.4b is highly virulent and lethal for fully susceptible herring gulls and that pre-exposure to homo- and heterosubtypic LPAIV only partially modulates the disease outcome.

Adaptive Heterosubtypic Immunity to Low Pathogenic Avian Influenza Viruses in Experimentally Infected Mallards

Mallards are widely recognized as reservoirs for Influenza A viruses (IAV); however, host factors that might prompt seasonality and trends in subtype diversity of IAV such as adaptive heterosubtypic immunity (HSI) are not well understood. To investigate this, we inoculated mallards with a prevailing H3N8 low pathogenic avian influenza virus (LPAIV) subtype in waterfowl to determine if prior infection with this virus would be protective against heterosubtypic infections with the H4N6, H10N7 and H14N5 LPAIV subtypes after one, two and three months, respectively. Also, we investigated the effect of cumulative immunity after sequential inoculation of mallards with these viruses in one-month intervals. Humoral immunity was assessed by microneutralization assays using a subset of representative LPAIV subtypes as antigens. Our results indicate that prior inoculation with the H3N8 virus confers partial protective immunity against subsequent heterosubtypic infections with the robustness of HSI related to the phylogenetic similarity of the HA protein of the strains used. Furthermore, induced HSI was boosted and followed by repeated exposure to more than one LPAIV subtype. Our findings provide further information on the contributions of HSI and its role in the dynamics of IAV subtype diversity in mallards.

SURVEILLANCE FOR AVIAN INFLUENZA VIRUS IN WILD BIRDS IN POLAND, 2008-15.

We tested wild birds in Poland during 2008-15 for avian influenza virus (AIV). We took 10,312 swabs and feces samples from 6,314 live birds representing 12 orders and 84 bird species, mostly from orders Anseriformes and Charadriiformes, for testing and characterization by various PCR methods. From PCR-positive samples, we attempted to isolate and subtype the virus. The RNA of AIV was detected in 1.8% (95% confidence interval [CI], 1.5-2.1%) of birds represented by 48 Mallards ( Anas platyrhynchos ), 11 Mute Swans ( Cygnus olor ), 48 Common Teals ( Anas crecca ), three Black-headed Gulls (Chroicocephalus ridibundus), one Common Coot ( Fulica atra ), one Garganey (Spatula querquedula), and one unidentified bird species. Overall, the prevalence of AIV detection in Mallards and Mute Swans (the most frequently sampled species) was 2.0% (95% CI, 1.4-2.5%) and 0.5% (95% CI, 0.2-0.8%), respectively; the difference was statistically significant (P=0.000). Hemagglutinin subtypes from H1 to H13 were identified, including H5 and H7 low pathogenic AIV subtypes. Mallards and Common Teals harbored the greatest diversity of subtypes. We observed seasonality of viral detection in Mallards, with higher AIV prevalence in late summer and autumn than in winter and spring. In addition, two peaks in AIV prevalence in summer (August) and autumn (November) were demonstrated for Mallards. The prevalence of AIV in Mute Swans did not show any statistically significant seasonal patterns.

Persistence of low-pathogenic H5N7 and H7N1 avian influenza subtypes in filtered natural waters

Wild aquatic birds are the natural reservoir of avian influenza virus (AIV), and the virus is transmitted among birds through a fecal-oral route. Infected birds excrete significant amounts of AIV into the environment, and thereby sustain the circulation of AIV in the bird populations. Improved knowledge on the influence of environmental factors on the persistence of AIV in natural habitats would be valuable for risk assessments. The presented work investigated the persistence of two low-pathogenic AIV subtypes in natural water samples. The study included two AIVs formerly isolated from wild ducks, which were suspended in filtered natural fresh, brackish or sea water with salinity of 0, 8000 and 20,000 parts per million (ppm), respectively. Also sterilized brackish and sea waters were included in order to examine the influence of microbial flora on virus persistence. All water samples were incubated at temperatures representative for seasonal variation of ambient temperatures in Northern Europe (4, 17 and 25 °C). The results showed a clear correlation between persistence of viral infectivity and temperature, salinity and presence of microbial flora. While independent of virus subtype, the persistence of infectivity was negatively affected by increased temperature, salinity as well as presence of natural microbial flora. The study provides insight on impact of essential physical, chemical and biological parameters on persistence of AIV in aquatic environments. Studies determining the importance of additional environmental parameters and the detailed mechanisms of microbial inactivation of AIV should be encouraged.

PERSISTENCE OF LOW PATHOGENIC AVIAN INFLUENZA VIRUSES IN FILTERED SURFACE WATER FROM WATERFOWL HABITATS IN GEORGIA, USA

The natural reservoirs for avian influenza virus (AIV) are wild bird species of the orders Anseriformes and Charadriiformes. The primary route of transmission for wild birds is through fecally contaminated surface water on shared aquatic habitats. A distilled water model has shown that AIV remains infectious in water for weeks to months with pH, salinity, and temperature affecting stability. To evaluate the effect of pH, salinity, and temperature on AIV persistence in natural surface water, we measured the duration of infectivity for two common low pathogenic AIV subtypes in 15 filtered surface water samples collected from major waterfowl habitats in Georgia, USA. Trials were performed at three incubation temperatures 10, 17, and 28 C. Consistent with previous studies, pH and temperature had a significant effect on the stability of AIV in filtered surface water. Both viruses were less stable at warmer temperatures and in acidic water (pH <5.0). Due to the limited range of salinity of the field water samples, the role of salinity in AIV stability in surface water could not adequately be evaluated. Variations in persistence times between water samples with comparable pH and salinities indicated that other factors affect AIV stability in natural surface water. These results contribute to the current understanding of AIV persistence in aquatic habitats and may help in identifying areas with an increased likelihood of AIV persistence and potential transmission.

Pathogenesis and transmissibility of highly (H7N1) and low (H7N9) pathogenic avian influenza virus infection in red-legged partridge (Alectoris rufa)

An experimental infection with highly pathogenic avian influenza virus (HPAIV) and low pathogenic avian influenza virus (LPAIV) was carried out in red-legged partridges (Alectoris rufa) in order to study clinical signs, gross and microscopic lesions, and viral distribution in tissues and viral shedding. Birds were infected with a HPAIV subtype H7N1 (A/Chicken/Italy/5093/1999) and a LPAIV subtype H7N9 (A/Anas crecca/Spain/1460/2008). Uninoculated birds were included as contacts in both groups. In HPAIV infected birds, the first clinical signs were observed at 3 dpi, and mortality started at 4 dpi, reaching 100% at 8 dpi. The presence of viral antigen in tissues and viral shedding were confirmed by immunohistochemistry and quantitative real time RT-PCR (qRRT-PCR), respectively, in all birds infected with HPAIV. However, neither clinical signs nor histopathological findings were observed in LPAIV infected partridges. In addition, only short-term viral shedding together with seroconversion was detected in some LPAIV inoculated animals. The present study demonstrates that the red-legged partridge is highly susceptible to the H7N1 HPAIV strain, causing severe disease, mortality and abundant viral shedding and thus contributing to the spread of a potential local outbreak of this virus. In contrast, our results concerning H7N9 LPAIV suggest that the red-legged partridge is not a reservoir species for this virus.

Effect of a prior exposure to a low pathogenic avian influenza virus in the outcome of a heterosubtypic low pathogenic avian influenza infection in mallards (Anas platyrhynchos).

Wild birds, particularly Anseriformes and Charadriiformes, are considered the natural reservoir of low pathogenic avian influenza (LPAI) viruses. The high prevalence and subtype diversity of avian influenza viruses at premigrational staging areas provide the perfect opportunity for multiple exposures to different LPAI virus subtypes. Natural consecutive and concurrent infections of sentinel ducks with different LPAI virus subtypes have been reported. The protective immune response from different LPAI virus infections is not understood nor is the effect of such repeated exposures. This study experimentally evaluated the effect of a prior exposure to a LPAI virus on the outcome of a heterosubtypic LPAI virus infection in mallards (Anas platyrhynchos). The results of this investigation suggest that recent prior exposure to a LPAI virus may affect the outcome of a subsequent heterosubtypic LPAI infection in mallards by reducing the duration of cloacal and oropharyngeal viral shedding as well as the viral load excreted via the cloaca. Wild mallards are likely exposed to multiple subtypes of LPAI virus during the periods of peak viral circulation, and the results of this study suggest that the duration of viral shedding in subsequent exposures might be reduced.

The Polymerase Acidic Protein Gene of Influenza A Virus Contributes to Pathogenicity in a Mouse Model

Adaptation of influenza A viruses to a new host species usually involves the mutation of one or more of the eight viral gene segments, and the molecular basis for host range restriction is still poorly understood. To investigate the molecular changes that occur during adaptation of a low-pathogenic avian influenza virus subtype commonly isolated from migratory birds to a mammalian host, we serially passaged the avirulent wild-bird H5N2 strain A/Aquatic bird/Korea/W81/05 (W81) in the lungs of mice. The resulting mouse-adapted strain (ma81) was highly virulent (50% mouse lethal dose = 2.6 log(10) 50% tissue culture infective dose) and highly lethal. Nonconserved mutations were observed in six viral genes (those for PB2, PB1, PA, HA, NA, and M). Reverse genetic experiments substituting viral genes and mutations demonstrated that the PA gene was a determinant of the enhanced virulence in mice and that a Thr-to-Iso substitution at position 97 of PA played a key role. In growth kinetics studies, ma81 showed enhanced replication in mammalian but not avian cell lines; the PA(97I) mutation in strain W81 increased its replicative fitness in mice but not in chickens. The high virulence associated with the PA(97I) mutation in mice corresponded to considerably enhanced polymerase activity in mammalian cells. Furthermore, this characteristic mutation is not conserved among avian influenza viruses but is prevalent among mouse-adapted strains, indicating a host-dependent mutation. To our knowledge, this is the first study that the isoleucine residue at position 97 in PA plays a key role in enhanced virulence in mice and is implicated in the adaptation of avian influenza viruses to mammalian hosts.

Australian surveillance for avian influenza viruses in wild birds between July 2005 and June 2007

To identify and gain an understanding of the influenza viruses circulating in wild birds in Australia. A total of 16,303 swabs and 3782 blood samples were collected and analysed for avian influenza (AI) viruses from 16,420 wild birds in Australia between July 2005 and June 2007. Anseriformes and Charadriiformes were primarily targeted. Cloacal, oropharyngeal and faecal (environmental) swabs were tested using polymerase chain reaction (PCR) for the AI type A matrix gene. Positive samples underwent virus culture and subtyping. Serum samples were analysed using a blocking enzyme-linked immunosorbent assay for influenza A virus nucleoprotein. No highly pathogenic AI viruses were identified. However, 164 PCR tests were positive for the AI type A matrix gene, 46 of which were identified to subtype. A total of five viruses were isolated, three of which had a corresponding positive PCR and subtype identification (H3N8, H4N6, H7N6). Low pathogenic AI H5 and/or H7 was present in wild birds in New South Wales, Tasmania, Victoria and Western Australia. Antibodies to influenza A were also detected in 15.0% of the birds sampled. Although low pathogenic AI virus subtypes are currently circulating in Australia, their prevalence is low (1.0% positive PCR). Surveillance activities for AI in wild birds should be continued to provide further epidemiological information about circulating viruses and to identify any changes in subtype prevalence.
