Genetic parameters of colostrum and calf serum antibodies in Swedish dairy cattle

BackgroundA sufficient IgG content in the colostrum is essential for the newborn calf, as it provides passive immunity which substantially affects the probability of survival during rearing. Failure of passive transfer (FPT) occurs when a calf does not absorb enough antibodies from the colostrum and is defined by an IgG concentration in calf serum lower than 10 g/L. Apart from delayed access to colostrum, FPT can be due to a low production of IgG in the mother or poor IgG absorption by the calf. The aim of this study was to estimate the genetic background of antibody levels and indicator traits for antibodies in the colostrum and calf serum, and their correlation with milk production.ResultsColostrum data were available for 1340 dairy cows with at least one calving and calf serum data were available for 886 calves from these cows. Indicator traits for antibody concentrations were estimated using refractometry (a digital Brix refractometer for colostrum and an optical refractometer for serum), and enzyme-linked immunosorbent assays (ELISA) were used to determine the levels of total IgG and natural antibodies (NAb) of various antibody isotypes in the colostrum and calf serum. Colostrum traits had heritabilities ranging from 0.16 to 0.31 with repeatabilities ranging from 0.21 to 0.55. Brix percentages had positive genetic correlations with all colostrum antibody traits including total IgG (0.68). Calf serum antibody concentrations had heritabilities ranging from 0.25 to 0.59, with a significant maternal effect accounting for 17 to 27% of the variance. When later in life calves produced their first lactation, the lactation average somatic cell score was found to be negatively correlated with NAb levels in calf serum.ConclusionsOur results suggest that antibody levels in the colostrum and calf serum can be increased by means of selection.

Identification of candidate genes affecting chronic subclinical mastitis in Norwegian Red cattle: combining genome-wide association study, topologically associated domains and pathway enrichment analysis.

The aim of this study was to identify genes associated with chronic subclinical mastitis (SCM) in Norwegian Red (NR) cattle. Twelve SCM traits defined based on fixed threshold for test-day somatic cell count (SCC) were, together with lactation-average somatic cell score (LSCS) used for association and pathway enrichment analyses. A GWAS was performed on 3795 genotyped NR bulls with 777K SNP data and phenotypic information from 7300847 test-day SCC observations from 3543764 cows. At 5% chromosome-wide significance level 36 unique SNP were detected to be associated with one or more of the traits. These SNPs were analysed for linked genes using genomic positions of topologically associated domains (TAD). For the SCM traits with SCC >50000 and >100000 cells/ml on two test-days in a row and LSCS, the same top significant genes were identified - checkpoint clamp loader component (RAD17) and cyclin B1 (CCNB1). The SCM traits with SCC >250000, 300000, 350000 or 400000 cells/ml on two test-days in a row and D400 (number of days before the first case with SCC >400000 cells/ml) displayed similar top significant genes: acyl-CoA thioesterase 2 and 4 (ACOT2; ACOT4). For the traits SCM200_3 (SCC >200000 cells/ml on three test-days in a row) and SCM150, SCM200 (SCC >150000; 200000 cells/ml on two test-days in a row) a group of chemokine (C-X-C motif) ligand genes and the Fos proto-oncogene, AP-1 transcription factor subunit (FOS) gene, were identified. Further functional studies of these identified candidate genes are necessary to clarify their actual role in development of chronic SCM in NR cattle.

Alternative subclinical mastitis traits for genetic evaluation in dairy cattle

Chronic subclinical mastitis (SCM), characterized by changes in milk composition and high somatic cell count (SCC) in milk for a prolonged period of time, is often caused by a bacterial infection. Different levels of SCC have been suggested and used as threshold to identify subclinical infection. The aim of this study was to examine different definitions of SCM based on test-day SCC and estimate genetic parameters for these traits and their genetic correlation to milk production. Test-day SCC records from 1,209,128 Norwegian Red cows in lactation 1 to 3 were analyzed. Twelve SCM traits were defined as binary with 2 test-day SCC in a row above SCC thresholds from 50,000 to 400,000 cells/mL (SCM50, SCM100, SCM150, SCM200, SCM250, SCM300, SCM350, and SCM400), with 3 test-day SCC in a row above 200,000 and 400,000 cells/mL (SCM200_3 and SCM400_3), and the number of days before the first case with SCM50 (D50) or SCM400 (D400). The heritability and genetic correlations were estimated for SCM traits and the mean lactation-average somatic cell score (LSCS) using linear animal repeatability models. The total mean frequency of SCM ranged from 1.2% to 51.8%, for different trait definitions, high for low SCC threshold (SCM50) and low for the highest SCC threshold (SCM400_3). For the 2 traits based on number of days, the mean values were 104 (D50) and 117 (D400) days. The mean LSCS was 4.4 (equivalent to around 82,000 SCC). Heritabilities for the 12 alternative SCM traits were low and varied from 0.01 (SCM400_3) to 0.1 (SCM100), whereas for LSCS the estimated heritability was 0.3 and standard error varied from 0.001 to 0.003. Genetic correlations among the SCM traits ranged from 0.7 (D50 and SCM400) to 1 (SCM350 and SCM400), whereas between SCM traits and milk production the correlation ranged from 0.07 (LSCS) to 0.3 (D400). The standard error for genetic correlations varied from 0.001 to 0.06. The heritability was low and the genetic correlations were strong among SCM traits. Genetic correlations lower than 1 suggest that the alternative SCM traits are genetically different from LSCS, the trait currently used in genetic evaluations for Norwegian Red. Hence, the alternative traits will add information and improve breeding for better udder health.

Factors affecting ewe performance in a crossbred dairy sheep research flock in the United States.

The Spooner Agricultural Research Station operated the only dairy sheep research flock in North America through 2016. The original nondairy ewe flock was "bred up" to a crossbred dairy flock through the use of rams and semen of the East Friesian (EF) and Lacaune (LA) dairy breeds. The objective of this study was to determine the environmental and nonadditive genetic effects that influence performance of dairy ewes. The traits analyzed were 180 d adjusted milk (180d MY), fat (180d FY), and protein (180d PY) yields, percentage fat (%F) and protein (%P) in milk, lactation average somatic cell score (LSCS), and number of lambs born per ewe lambing (NLB). The univariate repeatability models included the fixed effects of year of lambing, age, weaning system (except for the trait of NLB), individual breed composition, and individual retained heterosis along with the random additive genetic, permanent environmental, and residual effects. Estimates of heritability were moderate for 180d MY (0.32 ± 0.04), 180d FY (0.26 ± 0.04), and 180d PY (0.29 ± 0.04), high for %F (0.54 ± 0.04) and %P (0.61 ± 0.04), and low for LSCS (0.12 ± 0.03) and NLB (0.08 ± 0.02). Ewes that reared their lambs had lower ( < 0.01) 180d MY, 180d FY, 180d PY, %F, and %P and higher ( < 0.001) LSCS than ewes that had their lambs removed shortly after parturition. Relative to nondairy breeding, EF and LA breeding had positive ( < 0.001) effects on 180d MY, 180d FY, and 180d PY, but a negative ( < 0.03) effect on %P. Purebred EF ewes were predicted to have lower ( < 0.001) %F than purebred LA or nondairy ewes. Purebred LA ewes were predicted to have a higher ( < 0.001) LSCS than purebred EF or nondairy ewes. Purebred EF ewes were expected to be more ( < 0.001) prolific than purebred LA or nondairy ewes. Individual retained heterosis had a favorable ( < 0.01) effect on 180d MY, 180d FY, 180d PY, and NLB. Knowledge of the factors affecting dairy ewe performance are important for dairy sheep producers to make more informed husbandry and breeding decisions.

Genetic associations of in-line recorded milkability traits and udder conformation with udder health

ABSTRACTMilkability and udder conformation traits of Swedish Holstein (SH) and Swedish Red (SR) cows from 93 herds with automatic milking systems or conventional milking parlors were used to study genetic relationships to lactation average somatic cell score (LSCS) and incidence of clinical mastitis (CM). Estimated genetic correlations between measures of milking speed (average flow rate, milking time and box time) and LSCS ranged between 0.29 and 0.57 and showed that high milking speed is associated with increasing LSCS. Regressions indicated a curvilinear relationship. Genetic correlations between milking speed and CM showed similar values as for LSCS in SH cows, but were inconsistent in SR cows. Shallow udder and strong fore udder attachment were consistently correlated with good udder health. The unfavorable relationships between milking speed and udder health traits should be considered together with a few udder conformation traits when selecting for better milkability.

Genomic associations with somatic cell score in first-lactation Holstein cows

This genome-wide association study aimed to identify loci associated with lactation-average somatic cell score (LASCS) and the standard deviation of test-day somatic cell score (SCS-SD). It is one of the first studies to combine detailed phenotypic and genotypic cow data from research dairy herds located in different countries. The combined data set contained up to 52 individual test-days per lactation and thereby aimed to capture temporary increases in somatic cell score associated with infection. Phenotypic data for analysis consisted of 46,882 test-day records on 1,484 cows, and genotypic data consisted of 37,590 single nucleotide polymorphisms (SNP). Using an animal model, the associations between each individual SNP and the phenotypic data were estimated. To account for the risk of false positives, a false discovery rate threshold of 0.20 was set. The analyses showed that LASCS was significantly associated with a SNP on Bos taurus autosome (BTA) 4 and a SNP on BTA18. Likewise, SCS-SD was associated with this SNP on BTA18. In addition, SCS-SD significantly associated with a SNP on BTA6. Relatively few associations were found, suggesting that LASCS and SCS-SD are controlled by multiple loci distributed across the genome, each with a relatively small effect. Increased knowledge on genetic regulation of LASCS and SCS-SD may aid in identification of genes that play a role in mastitis resistance. Such knowledge helps us understand the genetic mechanisms leading to mastitis and in discovery of targets for mastitis therapeutics.

Genetic analysis of clinical mastitis and somatic cell count traits in Austrian Fleckvieh cows

The objectives of this study were to investigate genetic associations between clinical mastitis (CM) and different somatic cell count traits, and to examine their relationships, in terms of estimated breeding values, with other traits that are routinely evaluated in Austrian Fleckvieh dual-purpose cows. Records on veterinary treatments of CM were available from the Austrian health-monitoring project. For CM, 3 intervals in early lactation were considered: −10 to 50 d, 51 to 150 d, and −10 to 150 d after calving. Within each interval, absence or presence of CM was scored as 1 or 0 based on whether or not the cow had recorded at least one veterinary treatment of CM. The average somatic cell score of the first 2 test-days after calving was defined as early lactation average somatic cell score, and lactation mean somatic cell score was the average of all test-day somatic cell scores from 8 to 305 d after calving. Subclinical mastitis was expressed as a binary trait based on prolonged elevated somatic cell counts. If somatic cell counts on 3 consecutive test-days in the interval from 8 to 305 d after calving were above 200,000 cells/mL, the binary variable subclinical mastitis was defined as 1 and otherwise 0. Records of Austrian Fleckvieh cows, with calving from January 1, 2007, to February 28, 2009, were analyzed using univariate and bivariate sire models. Threshold liability models were applied for binary traits, and Gaussian models were used for early lactation average somatic cell score and lactation mean somatic cell score. A Bayesian approach using Gibbs sampling was applied for genetic analyses. Posterior means of heritability of liability to CM were 0.06 and 0.02 in the first and second interval, respectively, and 0.05 in the full period (−10 to 150 d). Heritability estimates of somatic cell count traits were higher (0.09 to 0.13). The posterior mean of the genetic correlation between CM in lactation period 1 (−10 to 50 d after calving) and 2 (51 to 150 d after calving) was close to unity. Posterior means of genetic correlations between CM and somatic cell count traits ranged from 0.64 to 0.77. Because CM and somatic cell count describe different aspects of udder health, information on both traits should be considered for selection of bulls. Correlations of sire breeding values revealed that especially the udder conformation trait udder depth may be useful as additional information to reduce both CM and somatic cell count.

Heterosis effects in a black and white dairy cattle population under different production environments

The effect of the environmental level of production (ENV) on the expression of heterosis for 305-day milk, fat, protein, and fat plus protein (FP) yields, lactation average somatic cell score (LSCS), and age at first calving (AFC) was investigated in first lactation Black and White dairy cows in the Netherlands, and officially enrolled in the Dutch herd-book. Holstein Friesian (HF), Dutch Friesian (DF), and first generation (F 1) crosses obtained from the mating of HF sires and DF dams (HD) were involved in the study, and data from animals with a calving date between 1990 and 2000 were used. A total of 22,930 cows with production and AFC information distributed in 3549 herds and 11,055 cows with LSCS information distributed in 2071 herds, were available. Adjusted lactation yield of milk for each herd was obtained using a model that accounted for fixed effects of herd, year and month of calving, genotype, and AFC. The overall mean of all adjusted data was computed, and 3 ENV were defined on the basis of the overall mean ± 0.5 standard deviations. Once ENV was defined, traits were analysed with a model that included fixed effects of ENV, herd nested within ENV, AFC (only production traits and LSCS), year and month of calving, genotype, and the interaction between ENV and genotype. Least squares means for the interaction effect were used to estimate heterosis and to evaluate its magnitude across ENV. Holstein Friesian achieved higher productions than DF. First generation crosses showed productions close to HF, especially in the low ENV. Estimates of heterosis for yield traits ranged from 2.4% (milk) in the high to 5.3% (fat) in the low ENV, and reduced with increasing ENV. Estimates for LSCS and AFC were low, with the exception of LSCS in the high ENV. Results suggest that the highest non-additive genetic effects for yield traits and LSCS were expressed in the most stressful ENV, i.e., the low one for production and the high one for LSCS.

Genetic analysis of somatic cell score and udder type traits in South African Holstein cows

Selection accuracy for resistance to mastitis may be increased by combining somatic cell score (SCS) and udder type into an udder health index, using genetic parameter estimates among them. A multi-trait animal model was used to estimate genetic parameters among lactation average SCS and udder type traits in South African Holstein cattle, through REML procedures. Data comprised records on 22 999 Holstein cows in 722 herds, collected through the National Milk Recording Scheme from 1996 to 2002. Average SCS in the first three lactations (SCS1, SCS2, SCS3) were considered as different traits and the udder type traits were fore udder attachment (FUA), rear udder height (RUH), udder cleft (UC), udder depth (UD), fore teat length (FTL) and fore teat placement (FTP). Heritability estimates for SCS were 0.19 ± 0.02, 0.17 ± 0.02 and 0.19 ± 0.02, respectively for SCS1, SCS2 and SCS3. Udder type traits had heritability estimates ranging from 0.13 ± 0.01 for UC to 0.34 ± 0.01 for FTL. The genetic correlations between lactation SCS ranged from 0.82 ± 0.04 to 0.99 ± 0.03 for correlations of SCS3 with SCS1 and SCS2, respectively. Genetic correlations between SCS and udder type traits were in the range -0.01 ± 0.07 between FUA and SCS3 to -0.38 ± 0.04 between UD and SCS1 and SCS2. Slow genetic progress is expected when selection is applied independently on SCS and udder type traits, due to the generally low heritability estimates. Low, shallow udders with narrowly placed teats are associated with low SCS in the South African Holstein population. Genetic parameters and single versus multi-trait evaluation of udder health traits

Genetic parameters were estimated for lactation average somatic cell score (SCS) and clinical mastitis (CM) for the first three lactations of multiparous Finnish Ayrshire cows. A multi-trait linear sire model was used for estimation of covariance components, and the efficiencies of single- versus multi-trait multi-lactation (MT) sire evaluations were compared. Heritability of SCS and CM in the first three lactations ranged from 0.11 to 0.13 and 0.02 to 0.03, respectively. Within lactation, genetic correlations between SCS and CM ranged from 0.68 to 0.72. Within both traits, across-lactation genetic correlations were lowest between 1 and 3, and highest between 2 and 3, with estimates ranging from 0.75 to 0.86 and from 0.81 to 0.98 for CM and SCS, respectively. Residual and phenotypic correlations were low and ranged from 0.09 to 0.13 and from 0.10 to 0.13, respectively. The absolute difference between genetic and residual correlations was from 0.5 to 0.6. Within-lactation genetic correlations between traits that are much less than unity suggest a multi-trait model for genetic evaluation of mastitis resistance. Comparison of model prediction performance between single-trait (ST) and MT models using a data splitting method showed that the MT model was more stable in predicting breeding values in future records of animals. Especially, for young sires and CM, the SD of EBVs from the MT model was 14 to 23% higher than the ST model, indicating more effective use of information in terms of revealing more genetic variation.

Genetic Parameters for Clinical Mastitis, Somatic Cell Score, and Production in the First Three Lactations of Swedish Holstein Cows

Using a mixed linear animal model, genetic parameters were estimated for clinical mastitis (MAST), lactation average somatic cell score (LSCS), and milk production traits in the first 3 lactations of more than 200,000 Swedish Holstein cows with first calving from 1995 to 2000. Heritability estimates for MAST (0.01 to 0.03) were distinctly lower than those for LSCS (0.10 to 0.14) and production traits (0.23 to 0.36). The genetic correlation between MAST and LSCS was high for all lactations (mean 0.70), implying that selection for low LSCS will reduce the incidence of mastitis. Undesirable genetic relationships with production were found for both MAST and LSCS with genetic correlations ranging from 0.01 to 0.45. This emphasizes the need for including udder health traits in the breeding goal. Genetic correlations across lactations for the same trait were positive and high for both MAST (>0.7), LSCS (>0.8), and production traits (>0.9), with the strongest correlations between second and third parity for all traits (>0.9 for udder health traits and close to unity for production traits).

Continual selection for increased milk yield for more than 40 yr, combined with the antagonistic relationship between increasing yield, somatic cell count, and fertility, have resulted in sires that may not be optimal for producing daughters for grazing systems where seasonal calving is very important. The objective of this study was to investigate the possible existence of a genotype x environment interaction (G x E) in grazing vs. confinement herds within the United States for lactation average somatic cell score (LSCS), days open (DO), days to first service (DFS), and number of services per conception (SPC). Grazing herds were defined as those that utilized grazing for at least 6 mo each year and were enrolled in Dairy Herd Improvement (DHI). Control herds were confinement DHI herds of similar size in the same states. For LSCS, the performance of daughters in grazing and control herds was examined using linear regression of LSCS on the November 2000 USDA-DHIA sire predicted transmitting abilities (PTA) for SCS. Genetic parameters for all traits were estimated using REML in a bivariate animal model that treated the same trait in different environments as different traits. Rank correlations were calculated between sires' estimated breeding values for LSCS, calculated separately for sires in both environments. The coefficient of regression of daughter LSCS on sire PTA was less in grazing herds than in control herds. The coefficient of regression for control herds was closer to expectation. Estimates of heritability were approximately 0.12 for LSCS, and less than 0.05 for the reproduction traits. Heritabilities for DO, DFS, and SPC were slightly higher for control herds. Estimates of genetic correlation for each reproductive trait between the 2 environments were high and not significantly different from unity. Generally, these traits appear to be under similar genetic control, but a lower coefficient of regression of LSCS on sire PTA for SCS in grazing herds suggests differences in daughter performance in grazing herds may be overstated based on current PTA for SCS.

Relationships of Sire Predicted Transmitting Ability for Somatic Cell Score with Measures of Daughter Performance

The relationship between sire predicted transmitting ability (PTA) for somatic cell score (SCS) and occurrence of mastitis in daughters was characterized with 304 Holsteins first freshening since 1991 in the Virginia Tech dairy herd. No direct sire selection for PTA SCS was practiced in the herd. Linear regressions of first lactation average SCS (1.3), number of cases of mastitis (0.80), days clinical (7.0), and number of treatments for mastitis (2.0) on PTA SCS were significant. Linear regression coefficients for second and third lactations were generally positive, but like quadratic and cubic coefficients in this study, were not significant. Relationship of sire PTA SCS and measures of lifetime (84 mo opportunity) performance were determined for 2,494,195 Holsteins born between 1979 and 1987. Linear coefficients from regression on PTA SCS for number of lactations (−0.31), days of productive life (−87.0), total days in milk (−72.2), lifetime milk (−1609.1), fat (−80.7), and protein (−48.2) production, relative net income adjusted for opportunity cost for fluid markets (+13.47), first lactation average SCS (+1.00), and first lactation mature equivalent milk (+429.9) were different from zero. Partial regressions holding PTA milk or PTA protein constant were substantially larger than linear coefficients. When PTA productive life was held constant, the relationship between PTA SCS and measures of herd life and lifetime yield became positive, and relative net income adjusted for opportunity cost variables became moderately positive. One of the major expenses associated with higher susceptibility (higher PTA SCS) may be shorter herd life.

Method R Estimates of Additive Genetic, Dominance Genetic, and Permanent Environmental Fraction of Variance for Yield and Health Traits of Holsteins

Fractions of variance accounted for by additive genetic, dominance genetic, and permanent environmental effects for milk, fat, and protein yields; somatic cell score; and productive life were estimated from Holstein data used for national genetic evaluations. Contemporary group assignments were determined using the national procedure. Data included 1,973,317 milk and fat records for 812,659 cows, 1,019,421 protein records for 462,067 cows, 468,374 lactation average somatic cell score (SCS) records for 232,909 cows, and 735,256 cows with productive-life records. Variance components were estimated with the JAADOM program, which uses iteration on data and second-order Jacobi iteration for obtaining solutions to the mixed-model equations and Method R for estimation of variance components. Ten different random data subsets were used to estimate parameters for each trait. Estimated additive genetic, dominance genetic, and permanent environmental fractions of variance were 0.34, 0.05, and 0.10 for milk yield; 0.34, 0.05, and 0.11 for fat yield; 0.31, 0.05, and 0.10 for protein yield; and 0.17, 0.01, and 0.16 for lactation average SCS. Estimated additive genetic and dominance genetic fractions of variance were 0.12 and 0.06 for productive life. Mean empirical standard errors of additive genetic, dominance genetic, and permanent environmental variance fractions were 0.003, 0.006, and 0.006.
