Dendritic cells induce an immunological response depending on the maturational status from the interaction with different receptors at the cell surface as toll-like-receptors (TLR). We wanted to evaluate the different TLR expression on monocyte-derived dendritic cells (moDC) and the influence in these expressions of specific agonists and amoxicillin in patients with allergic reaction to this drug. We studied 5 patients with an allergic reaction to amoxicillin. MoDC were cultured with amoxicillin with or without TLR2, 4 and 8 agonists and TLR4 antagonist. From these, we studied TLR1-9 gene expression by RT-qPCR, and DC maturation and lymphocyte proliferation (by culturing lymphocytes with moDC) by flow cytometry. All studied TLR genes were expressed in inmature moDC. Amoxicillin induced a trend to increase the expression of TLR1, 3, 4, 7 and 8 being higher for TLR9. Amoxicillin induced DC maturation in 2 patients, in combination with LPS (TLR4 agonist) induced maturation in 4 patients and with PAM (TLR2 agonist) plus LPS resulted in maturation in 3 patients. Proliferation assays with amoxicillin showed positive results in 2 patients, together with PAM in 4 patients and in combination with LPS and PAM in 4 patients. Mo-derived-DC express TLR form 1 to 9 and can be increased their induction with amoxicillin in patients allergic to this drug. The proliferative response increased when amoxicillin was combined with TLR2 and TLR4 agonists in patients allergic to this drug.
