The ability to regulate voltage-gated K+-permeable channels in the mature root epidermis is essential for waterlogging tolerance in barley.

Oxygen depletion under waterlogged conditions results in a compromised operation of H+-ATPase, with strong implications for membrane potential maintenance, cytosolic pH homeostasis, and transport of all nutrients across membranes. The above effects, however, are highly tissue specific and time dependent, and the causal link between hypoxia-induced changes to the cell's ionome and plant adaptive responses to hypoxia is not well established. This work aimed to fill this gap and investigate the effects of oxygen deprivation on K+ signalling and homeostasis in plants, and potential roles of GORK (depolarization-activated outward-rectifying potassium) channels in adaptation to oxygen-deprived conditions in barley. A significant K+ loss was observed in roots exposed to hypoxic conditions; this loss correlated with the cell's viability. Stress-induced K+ loss was stronger in the root apex immediately after stress onset, but became more pronounced in the root base as the stress progressed. The amount of K+ in shoots of plants grown in waterlogged soil correlated strongly with K+ flux under hypoxia measured in laboratory experiments. Hypoxia induced membrane depolarization; the severity of this depolarization was less pronounced in the tolerant group of cultivars. The expression of GORK was down-regulated by 1.5-fold in mature root but it was up-regulated by 10-fold in the apex after 48 h hypoxia stress. Taken together, our results suggest that the GORK channel plays a central role in K+ retention and signalling under hypoxia stress, and measuring hypoxia-induced K+ fluxes from the mature root zone may be used as a physiological marker to select waterlogging-tolerant varieties in breeding programmes.

Anti-effects of cordycepin to hypoxia-induced membrane depolarization on hippocampal CA1 pyramidal neuron

Cordycepin has important neuroprotective effects in hypoxic or ischemic insult. However, the direct electrophysiological evidence of cordycepin's neuroprotection from hypoxic or ischemic insult remains unknown. Hence, in this study, the electrophysiological mechanism by which cordycepin protects against ischemic and hypoxic damages has been studied using an energy-deprivation injury model through whole-cell patch clamp techniques. Results revealed that cordycepin (80µM) significantly delayed hypoxia-induced membrane depolarization, including cordycepin reduced slope, and extended the duration of slow depolarization, prolonged the ability to generate spontaneous action potential (AP) firing, delayed the onset of rapid depolarization, and maintained the more hyperpolarized membrane potential after rapid depolarization. Additionally, cordycepin also delayed the hypoxia-induced decrease in the evoked AP amplitude. Furthermore, cordycepin can rescue the neuronal electrophysiological function after the 5min hypoxia pretreatment insult as seen the recovery on the evoked spike amplitude, membrane potential, and evoked AP latency during reoxygenation of hippocampal slices with cordycepin. Collectively, the results in this study provide direct evidence demonstrating the important neuroprotective effects of cordycepin against the hypoxic insult via improvement of the neuronal electrophysiological function, and the mechanism underlying the anti hypoxia-induced membrane depolarization is strongly recommended.

Abstract 16945: Role of Thromboxane A 2 -Activated Nonselective Cation Channels in Hypoxic Pulmonary Vasoconstriction of Rat

Hypoxia-induced pulmonary vasoconstriction (HPV) is a physiological mechanism responsible for controlling the ventilation-perfusion ratio in the lungs. Maintenance of the ventilation-perfusion ratio at 0.8-1 is critical for the optimization and maximization of gas exchange in the lung. Inhibition of K + channels has been shown to be a mechanism for hypoxia-induced membrane depolarization in pulmonary arterial smooth muscle cells (PASMCs). Pharmacological inhibition of K + channels, however, does not induce significant contraction in isolated rat pulmonary arteries, but significantly enhances HPV. These results suggest that, in addition to K + channel inhibition, other mechanisms are involved in hypoxia-induced membrane depolarization. We recently found that a partial contraction by thromboxane A 2 (TXA 2 ) is required to induce HPV. Therefore we hypothesize that TXA 2 receptor (TP) stimulation is involved in eliciting membrane polarization by activating nonselective cation (NSC) channels. Consistently, we found that 5-10 nM U46619, a stable agonist of TP, was essential for the contraction of isolated rat pulmonary arteries by 4-aminopyridine, a blocker of voltage-gated K + channel (Kv). Whole-cell voltage clamp experiments with rat PASMCs revealed that U46619 induced a weakly outward-rectifying NSC current (I NSC,TXA2 ), which was blocked by ruthenium red (RR), an antagonist of the transient receptor potential vanilloid (TRPV) subfamily. In addition, 2-aminoethoxydiphenyl borate, an agonist for TRPV1-3, activated NSC channels. In contrast, agonists for TRPV1 (capsaicin), TRPV3 (camphor), or TRPV4 (4α-PDD) rarely increased the membrane conductance. RT-PCR analysis showed the expression of transcripts for TRPV2 and TRPV4 in rat PASMCs. Finally, it was confirmed that the pretreatment with RR and tranilast (a TRPV2 inhibitor) both significantly inhibited HPV in the presence of U46619. Taken together, these data suggest that the concerted effects of I NSC,TXA2 increase and Kv inhibition during under hypoxia are required to induce membrane depolarization sufficient for eliciting HPV. Our results also imply that TXA 2 -activated NSC might be formed by TRPV2 in rat PASMCs and contribute to the hypoxia-induced membrane depolarization in PASMCs.

Multifocal animated imaging of changes in cellular oxygen and calcium concentrations and membrane potential within the intact adult mouse carotid body ex vivo

Carotid body (CB) type I cell hypoxia-sensing function is assumed to be based on potassium channel inhibition. Subsequent membrane depolarization initiates an intracellular calcium increase followed by transmitter release for excitation of synapses with linked nerve endings. Several reports, however, contradict this generally accepted concept by showing that type I cell oxygen-sensing properties vary significantly depending on the method of their isolation. We report therefore for the first time noninvasive mapping of the oxygen-sensing properties of type I cells within the intact adult mouse CB ex vivo by using multifocal Nipkow disk-based imaging of oxygen-, calcium- and potential-sensitive cellular dyes. Characteristic type I cell clusters were identified in the compact tissue by immunohistochemistry because of their large cell nuclei combined with positive tyrosine hydroxylase staining. The cellular calcium concentrations in these cell clusters either increased or decreased in response to reduced tissue oxygen concentrations. Under control conditions, cellular potential oscillations were uniform at ∼0.02 Hz. Under hypoxia-induced membrane depolarization, these oscillations ceased. Simultaneous increases and decreases in potential of these cell clusters resulted from spontaneous burstlike activities lasting ∼1.5 s. type I cells, identified during the experiments by cluster formation in combination with large cell nuclei, seem to respond to hypoxia with heterogeneous kinetics.

Role of oxygen sensitive potassium channels in pulmonary arteries smooth muscle cells in hypoxic pulmonary vasoconstriction

Mechanically induced potentials in atrial fibroblasts from rat hearts are sensitive to hypoxia/reoxygenation.

Membrane potential changes of atrial fibroblasts in response to mechanical stress have been considered to modulate the rhythmic electrical activity of healthy hearts. Our recent findings suggest that cardiac arrhythmia after infarction is related to enhanced susceptibility of the fibroblasts to physical stretch. In this study, we analysed the effect of hypoxia/reoxygenation, which are major components of tissue ischemia/reperfusion, on the membrane potential of atrial fibroblasts. Intracellular microelectrode recordings were performed together with isometric force measurements on isometrically contracting right atrial tissue preparations from adult rats. Lowering the oxygen tension in the perfusate from 80 kPa to 3.5 kPa reduced active force development and decreased the resting membrane potential of the cardiac fibroblasts from -23+/-5 mV to -5+/-2 mV ( n=35). Application of gadolinium (40 microM) to inhibit non-selective cation channels prevented hypoxia-induced membrane depolarization of the fibroblasts. Reoxygenation of the myocardial tissue resulted in a transient increase of the resting membrane potential to maximally -60+/-8 mV. These findings indicate that transmembrane currents in atrial fibroblasts are sensitive to changes in tissue oxygenation. In conclusion, altered electro-mechanical function of the ischemic heart may possibly involve changes of the membrane potential of the cardiac fibroblasts.

Potassium channels underlying the resting potential of pulmonary artery smooth muscle cells.

1. The molecular identity of the K channels giving rise to the negative membrane potential of pulmonary artery smooth muscle cells has yet to be determined. 2. To date, most studies have focused on voltage-gated, delayed rectifier channels and their roles in mediating hypoxia-induced membrane depolarization. There is, however, strong evidence that an outwardly rectifying K+ conductance distinct from the classical delayed rectifier is involved. 3. Growing evidence that TASK-like channels can sense hypoxia and are present in pulmonary artery smooth muscle cells suggests that they may be responsible for the resting K+ conductance and resting potential. 4. The present review considers the evidence that particular K channels maintain the resting membrane potential of pulmonary artery smooth muscle cells and mediate the depolarizing response to hypoxia.

Molecular basis of hypoxia-induced pulmonary vasoconstriction: role of voltage-gated K+ channels.

The hypoxia-induced membrane depolarization and subsequent constriction of small resistance pulmonary arteries occurs, in part, via inhibition of vascular smooth muscle cell voltage-gated K+ (KV) channels open at the resting membrane potential. Pulmonary arterial smooth muscle cell KV channel expression, antibody-based dissection of the pulmonary arterial smooth muscle cell K+ current, and the O2 sensitivity of cloned KV channels expressed in heterologous expression systems have all been examined to identify the molecular components of the pulmonary arterial O2-sensitive KV current. Likely components include Kv2.1/Kv9.3 and Kv1.2/Kv1.5 heteromeric channels and the Kv3.1b alpha-subunit. Although the mechanism of KV channel inhibition by hypoxia is unknown, it appears that KV alpha-subunits do not sense O2 directly. Rather, they are most likely inhibited through interaction with an unidentified O2 sensor and/or beta-subunit. This review summarizes the role of KV channels in hypoxic pulmonary vasoconstriction, the recent progress toward the identification of KV channel subunits involved in this response, and the possible mechanisms of KV channel regulation by hypoxia.

Electrophysiological actions of N-[1-[4-(4-fluorophenoxy)butyl]-4-piperidinyl]- N-methyl-2-benzothiazolamine (R56865) on CA1 neurons of the rat hippocampal slice during hypoxia

The electrophysiological effects of N-[1-[4-(4-fluorophenoxy)butyl]-4-piperidinyl]- N-methyl-2-benzothiazolamine (R56865), a drug which protects heart cells from ischemia-induced arrhythmias, was studied on intracellularly-recorded CA1 neurons of the rat hippocampal slice under normal or hypoxic conditions. On normoxic cells R56865 (1 μM) reduced firing accommodation without changing passive membrane properties, spike characteristics or synaptic transmission. On hypoxic cells R56865 selectively reduced the amplitude of hypoxia-induced membrane depolarization and partly counteracted the depression of synaptic transmission evoked by Schaffers collateral stimulation. Despite its influence on repetitive firing properties, R56865 might be useful to limit the extent of cellular depolarizing responses to hypoxia.

Adenosine modulates hypoxia-induced responses in rat PC12 cells via the A2A receptor.

1. The present study was undertaken to determine the role of adenosine in mediating the cellular responses to hypoxia in rat phaeochromocytoma (PC12) cells, an oxygen-sensitive clonal cell line. 2. Reverse transcriptase polymerase chain reaction studies revealed that PC12 cells express adenosine deaminase (the first catalysing enzyme of adenosine degradation) and the A2A and A2B adenosine receptors, but not the A1 or A3 adenosine receptors. 3. Whole-cell current- and voltage-clamp experiments showed that adenosine attenuated the hypoxia-induced membrane depolarization. The hypoxia-induced suppression of the voltage-sensitive potassium current (IK(V)) was markedly reduced by adenosine. Furthermore, extracellularly applied adenosine increased the peak amplitudes of IK(V) in a concentration-dependent manner. This increase was blocked by pretreatment not only with a non-specific adenosine receptor antagonist, 8-phenyltheophylline (8-PT), but also with a selective A2A receptor antagonist, ZM241385. 4. Ca2+ imaging studies using fura-2 acetoxymethyl ester (fura-2 AM) revealed that the increase in intracellular free Ca2+ during hypoxic exposure was attenuated significantly by adenosine. Voltage-clamp studies showed that adenosine inhibited the voltage-dependent Ca2+ currents (ICa) in a concentration-dependent fashion. This inhibition was also abolished by both 8-PT and ZM241385. 5. The modulation of both IK(V) and ICa by adenosine was prevented by intracellular application of an inhibitor of protein kinase A (PKA), PKA inhibitor fragment (6-22) amide. In addition, the effect of adenosine on either IK(V) or ICa was absent in PKA-deficient PC12 cells. 6. These results indicate that the modulatory effects of adenosine on the hypoxia-induced membrane responses of PC12 cells are likely to be mediated via activation of the A2A receptor, and that the PKA pathway is required for these modulatory actions. We propose that this modulation serves to regulate membrane excitability in PC12 cells and possibly other oxygen-sensitive cells during hypoxia.

On the mechanisms underlying hypoxia-induced membrane depolarization in striatal neurons.

Clinical and experimental evidence has shown that the striatal neurons are particularly vulnerable to hypoxia and ischaemia. An excessive excitatory action of glutamate, released by the corticostriatal terminals, has been implicated in this peculiar vulnerability of striatal neurons. We have studied the effects of hypoxia on the membrane properties of striatal neurons intracellularly recorded from a corticostriatal slice preparation. Brief (2-10 min) periods of hypoxia produced reversible membrane depolarizations. During the initial phase of the hypoxia-induced depolarization the frequency of action potential discharge was transiently increased; 2-3 min after the onset of hypoxia the firing activity was fully abolished. Brief periods of hypoxia also caused a reversible reduction of the amplitude of the excitatory postsynaptic potentials (EPSPs) evoked by cortical stimulation. Longer period of hypoxia (12-20 min) produced irreversible membrane depolarizations. In voltage-clamp experiments hypoxia caused an inward current coupled with an increased membrane conductance. Tetrodotoxin (TTX) or low calcium (Ca2+)-high magnesium containing solutions blocked synaptic transmission, but they did not reduce the hypoxia-induced electrical changes. Antagonists of excitatory amino acid receptors failed to affect the electrical effects caused by oxygen deprivation. Hypoxia-induced inward currents were reduced either by the potassium (K+) channel blockers, barium and tetraethyl ammonium (TEA) cations, or by lowering external sodium (Na+) concentration. Blockade of ATP-dependent Na(+)-K+ pump by both ouabain and strophanthidin enhanced hypoxia-induced membrane depolarization/inward current. Our findings indicate that the release of excitatory amino acids does not seem to be required for the acute hypoxia-induced electrical changes in striatal neurons. Moreover, TTX-resistant Na+ influx and K+ currents seem to play an important role in the generation of hypoxia-induced electrical changes. These data also suggest that the selective vulnerability of striatal neurons to oxygen deprivation may be caused by their peculiar sensitivity to energy metabolism failure.
