Influence of Education Level and Cigarette Consumption on Poverty in RW 1 RT 1 Ngaglik Batu, East Java

This research aims to determine the effect of cigarette consumption and community education level on income in Ngaglik Village RT.01 RW.01 Batu City. This analysis uses independent variables, namely cigarette consumption and education level. The dependent variable was people’s income. This research uses primary data obtained from distributing questionnaires to 48 family cards occupying Ngaglik Subdistrict RT.01 RW.01 Batu City. The analysis was done using the Slovin Method. Then a normality test, two variable linear regression test, multicollinearity test, heteroscedasticity test, and autocorrelation test were carried out with the help of SPSS tools. Firstly, the results of this study showed that cigarette consumption had no significant effect on income, while education level did. Second, the head of the families in this study were mostly men. Third, it is very important to educate about the dangers of smoking so that people have knowledge about not smoking and alleviating poverty. Keywords: cigarette consumption, education level, income

NLRC5 promotes tumorigenesis by regulating the PI3K/AKT signaling pathway in cervical cancer

Cervical cancer (CC) is the fourth most common cancer among women worldwide. NLR Family CARD Domain Containing 5 (NLRC5) plays an important role in tumorigenesis. However, its effect and mechanism in CC remains unclear. In this study, we aimed to investigate the function of NLRC5 in CC. NLRC5 was found to be down-regulated in CC tissues compared with normal cervical tissues. However, patients with higher NLRC5 expression had better prognosis, patients with higher age, HPV infection, lymph node metastasis, recurrence and histological grade had worse prognosis. Univariate and multivariate analyses showed NLRC5 to be a potential prognostic indicator for CC. Pearson correlation analysis showed that NLRC5 might exert its function in CC through autophagy related proteins, especially LC3. In vitro experiments demonstrated that NLRC5 inhibited LC3 levels and promoted the proliferation, migration, and invasion of CC cells by activating the PI3K/AKT signaling pathway. Treatment with LY294002 reversed the above phenotype. Taken together, our finding suggested that NLRC5 would participate in cervical tumorigenesis and progression by regulating PI3K/AKT signaling pathway. In addition, NLRC5 and LC3 combined as possible predictors in CC.

Counseling and assistance in verification and validation of housing data for surveyors

The data collection of housing database in Jembatan Besi Village conducted by surveyors from the Faculty of Civil Engineering and Planning, Trisakti University often encounters data errors and obstacles during its implementation. These errors can be in the form of incorrect writing of addresses, family card numbers, head of family NIKs, or geotagging (location points) of buildings. Therefore, in the middle of the data collection, a workshop was held to analyze and evaluate the data that had been entered and to correct data errors. The method used is through data checking by field coordinators and repairs are carried out simultaneously. Workshop activities include presentation of the progress of the 50% survey results, data validation and verification, and data correction. During the workshop, the survey results were obtained as much as 60.55% with a total of 2,704 buildings and data correction facilities including geotagging, RT and RW data, Family Card Numbers, NIK, address writing, and deleting duplicate data.

NLRC5 promotes endometrial carcinoma progression by regulating NF-κB pathway-mediated mismatch repair gene deficiency

The innate immune molecule NLR family CARD domain-containing 5 (NLRC5) plays a significant role in endometrial carcinoma (EC) immunosurveillance. However, NLRC5 also plays a protumor role in EC cells. Mismatch repair gene deficiency (dMMR) can enable tumors to grow faster and also can exhibit high sensitivity to immune checkpoint inhibitors. In this study, we attempted to determine whether NLRC5-mediated protumor role in EC is via the regulation of dMMR. Our findings revealed that NLRC5 promoted the proliferation, migration, and invasion abilities of EC cells and induced the dMMR status of EC in vivo and in vitro. Furthermore, the mechanism underlying NLRC5 regulated dMMR was also verified. We first found NLRC5 could suppress nuclear factor-kappaB (NF-κB) pathway in EC cells. Then we validated that the positive effect of NLRC5 in dMMR was restricted when NF-κB was activated by lipopolysaccharides in NLRC5-overexpression EC cell lines. In conclusion, our present study confirmed the novel NLRC5/NF-κB/MMR regulatory mechanism of the protumor effect of NLRC5 on EC cells, thereby suggesting that the NLRC5-mediated protumor in EC was depend on the function of MMR.

Mengatasi Permasalahan Administrasi Pernikahan dan Kependudukan Melalui Inovasi Pelayanan Publik Jalan Berdua di Kota Pasuruan

The marriage registration process is one of the important stages in validating a marriage legally. This process often requires complex efforts, takes time and costs significant money, especially for couples who come from areas far from population administration offices or religious courts. To overcome this challenge, the Pasuruan City Government has introduced an innovative Double Road service. This service allows couples to register their marriage as well as change their marital status on their Family Card and Electronic KTP documents. This second road innovation has the impact of improving population administration services regarding changes in marital status on Family Cards and Electronic KTPs. This research aims to describe the problems that arise in the Innovation program and the efforts made to overcome the problems that arise in the Dual Road Innovation program in Pasuruan City. This research uses a qualitative descriptive approach with primary and secondary data sources. Primary data was obtained through interviews, observation and documentation related to Jalan Berdua innovation. Secondary data is in the form of documents related to research. Data collection techniques involve observation, open interviews, and documentation. This research will provide an overview of the problems faced in implementing the Double Road Innovation in Pasuruan City and the efforts that have been made to overcome them. The results of this research are: Overall, the Jalan Berdua program in Pasuruan City is a successful example of how innovation in population administration services can improve efficiency, accessibility, community satisfaction, inter-agency cooperation, and effective use of data.

IMPLEMENTASI PELAYANAN DINAS KEPENDUDUKAN DAN PENCATATAN SIPIL KABUPATEN LAMPUNG SELATAN OLEH TIM KECAMATAN TANJUNG BINTANG

The problem in this study is that people in Tanjung Bintang District take approximately 2 hours to take care of population documents such as ID cards, family cards, birth certificates, and others. The mileage experienced by the community when handling documents is very difficult, time-consuming, and inefficient. This is a problem for today's society. The research method used is qualitative descriptive research, which is an analysis based on data obtained through interviews with informants, observation, and documentation. The results of the study, namely the Quality of Service of the Population and Civil Registration Office of South Lampung Regency by the Tanjung Bintang District Team, measured by five dimensions from Zeithhanl, Parasurama, and Berry, namely physical evidence (tangible), reliability (reliability), responsibility, assurance, and empathy (emphaty), are good enough, but there are still shortcomings. The existence of the Tanjung Bintang District Team is very helpful for the village community in terms of taking care of population documents that no longer need to go far to Disdukcapil Kalianda, so that it is more effective and efficient. In addition, findings regarding inhibiting factors in service by the Tanjung Bintang District Team are: limited space, officers who have never been given training in public services, lack of human resources, tools that depend on each other, and internet network disruption. Acacetin Attenuates Sepsis-induced Acute Lung Injury via NLRC3-NF-κB Pathway.

Acacetin, a flavonoid derived compound has been recognized for its diverse biological activities, such as anti-oxidative and anti-inflammatory effects. Acute lung injury (ALI) is a severe condition characterized by respiratory insufficiency and tissue damage, commonly triggered by pneumonia and severe sepsis. These conditions induce an inflammatory response via Toll-like receptor 4 (TLR4) signaling activation. This study explored acacetin's therapeutic potential against lipopolysaccharide (LPS) induced ALI in mice, focusing on its ability to modulate the NF-κB pathway via regulation of the Nod-like receptor family CARD domain containing 3 (NLRC3), a signal sensor that plays an important role in the regulation of inflammation and the maintenance of homeostasis. Our findings revealed that high-dose acacetin reduced the mortality rate of ALI mice, significantly ameliorated LPS-induced lung pathological changes, reduced lung edema, and decreased the expression of inflammatory mediators in lung tissues. This protective impact of acacetin appears to stem form its capacity to enhance NLRC3 expression, which, intern, can inhibit the activation of NF-κB and subsequently inhibit the production of inflammatory mediators. NLRC3 deficiency inhibits the protective effect of acacetin on ALI mice. Molecular docking also verified that acacetin tightly bound acacetin to NLRC3. Additionally, acacetin was found to influence macrophage recruitment dynamics via NLRC3, inhibiting the overactivation of NLRC3-NF-κB related pathways. Taken together, our results indicate that acacetin inhibited LPS-induced acute lung injury and macrophage overrecruitment to the lungs in mice by upregulating NLRC3.

The Maslahah of State Policy in Responding to Unregistered Marriage: Inclusion of Unregistered Marriage on The Family Card

Based on Minister of Home Affairs regulations No. 108 and No. 109 of 2019, unregistered (siri) marriage couples can now record their marriage in the Family Card. This article aims to analyze the policy of issuing Family Cards for unregistered marriage couples using the Islamic jurisprudence (ushul fiqh) approach. This article is a literature study, with data sourced from the Minister of Home Affairs regulations and various related literatures. The obtained data is then analyzed using a normative-philosophical approach, specifically ushul fiqh theories such as the concepts of public interest (maslahah) and preventing harm (adz-dzari�ah). The article argues that in every policy, there will be potential benefits (maslahah) and harms (mudharat). In the policy of recording unregistered marriages, the government needs to close the means that cause harm (saddu al-dhariah) to ensure this policy is not interpreted as a form of legalizing unregistered marriages. The government also needs to open the means that lead to benefits (fathu al-dhariah) so that the goal of fulfilling children's civil rights with the basis of having a birth certificate, and the purpose of the Marriage Law, namely the validity of marriage, can be achieved through the use of unregistered marriage data for expediting the marriage legalization process (itsbat nikah) in Religious Courts. The conclusion of this article is that to achieve the ideal policy objectives, aspects of benefits and harms need to be considered. This article contributes to understanding state policies from the perspective of ushul fiqh and indicates that it remains a relevant approach.

Normalisasi Data Kependudukan Dengan Model Min Max Dan Algoritma K-Means Untuk Pengelompokkan Tingkat Ekonomi Masyarakat

Desidential data at Disdukcapil Deli Serdang has only been used for administrative purposes such as making certificates, ID cards, family cards, and others. Meanwhile, the data should be able to increase its usefulness functions such as classifying the economic level of the Deli Serdang Regency community. So far, many people feel disadvantaged because the distribution of government assistance to low-level economic communities is often misdirected, there are people who should get assistance but instead cannot, but there are people who are less deserving of assistance who can get assistance, this can be influenced by various factors such as kinship, closeness or other things. techniques are needed in classifying the economic level of the Deli Serdang Regency community based on certain criteria. The K-Means algorithm is one of the algorithms in data mining to classify certain data, for that it is very suitable to use the K-Means algorithm in classifying the economic level of the community.

Faktor-Faktor yang Berhubungan dengan Perilaku Rumah Tangga dalam Membuang Sampah di RT.08/RW.08 Kp. Pulo Timaha Kecamatan Babelan Kabupaten Bekasi Tahun 2024

Waste is any material or object that is solid and is no longer used by humans and has been used to carry out an activity. This research aims to determine the factors related to household behavior in disposing of waste in Kp. Pulo Timaha RT.08/RW.08 Bekasi Regency 2023. This research is a type of quantitative analytical research using a cross sectional design. Data collection was carried out by means of interviews using questionnaires and observation. The sampling method uses Random Sampling Technique. The sample in this study consisted of 71 Family Cards. The analysis used is univariate and bivariate analysis (ChiSquare Test). Univariat results of respondents with unfavorable behavior were 53 (74.6%), education >9 years as many as 45 (63.4%), poor knowledge as many as 57 (80.3%), negative attitudes as many as 63 (88.7%), socialization of health workers who were not socialized was 48 (67.6%), and infrastructure was not available as many as 52 (73.2%). The results of bivariate research show that there is a relationship with waste disposal behavior, namely household knowledge (Pvalue <0.001 PR=6.389 CI=3.035-13.488), household attitude (Pvalue 0.003 PR=3.938 CI=2.061-7.523), and socialization of health workers (Pvalue <0.001 PR=10.435 CI=3.325-32.483). Meanwhile, there is no relationship between education and infrastructure on waste disposal behavior. Therefore, there is a need for socialization about proper and correct waste disposal to increase household knowledge, attitudes and behavior in disposing of waste.

Human NLRC4 expression promotes cancer survival and associates with type I interferon signaling and immune infiltration.

The immune system can control cancer progression. However, even though some innate immune sensors of cellular stress are expressed intrinsically in epithelial cells, their potential role in cancer aggressiveness and subsequent overall survival in humans is mainly unknown. Here, we show that nucleotide-binding oligomerization domain-like receptor (NLR) family CARD domain-containing 4 (NLRC4) is downregulated in epithelial tumor cells of patients with colorectal cancer (CRC) by using spatial tissue imaging. Strikingly, only the loss of tumor NLRC4, but not stromal NLRC4, was associated with poor immune infiltration (mainly DCs and CD4+ and CD8+ T cells) and accurately predicted progression to metastatic stage IV and decrease in overall survival. By combining multiomics approaches, we show that restoring NLRC4 expression in human CRC cells triggered a broad inflammasome-independent immune reprogramming consisting of type I interferon (IFN) signaling genes and the release of chemokines and myeloid growth factors involved in the tumor infiltration and activation of DCs and T cells. Consistently, such reprogramming in cancer cells was sufficient to directly induce maturation of human DCs toward a Th1 antitumor immune response through IL-12 production in vitro. In multiple human carcinomas (colorectal, lung, and skin), we confirmed that NLRC4 expression in patient tumors was strongly associated with type I IFN genes, immune infiltrates, and high microsatellite instability. Thus, we shed light on the epithelial innate immune sensor NLRC4 as a therapeutic target to promote an efficient antitumor immune response against the aggressiveness of various carcinomas.

The Fulfilment of Legal Identity: A Case Study of Residents Without Population Identification Numbers

Legal identity is crucial for individuals as it establishes their rights and obligations as legal subjects. Despite its importance, many residents in Yogyakarta lack a legal identity. This research aims to analyse the regulations governing the provision of legal identity for residents without a population identification number (NIK) and to identify the causes behind this issue in Yogyakarta. This study employs non-doctrinal legal research, combining normative and empirical methods. The findings reveal that obtaining a NIK is essential for fulfilling human rights and constitutional rights. This process involves population registration, which begins with recording the individual’s biodata. Changes in the population administration information system have led to the exclusion of residents who have not completed biometric recording from the population database, resulting in a lack of legal identity. In Yogyakarta, two main factors contribute to the absence of NIKs: First, impoverished individuals with domicile issues caused by the costs and time required for domicile relocation, potential trafficking, homelessness, and loss of family card data. Second, individuals with disabilities who are unable to complete the registration process.

Analisis Realiasi Penerimaan Program Bantuan Pemerintah terhadap Kemiskinan di Desa Lumban Tobing, Kecamatan Doloksanggul

Lumban Tobing Village, Doloksanggul District is located in North Sumatra Province which is the capital of Humbang Hasundutan Regency in Indonesia. The variations quality of life conditions in Lumbang Tobing Village causes variations in poverty conditions in this area. This research aims to determine the condition of poverty in Lumban Tobing Village and formulate a comprehensive poverty program hatching program in Lumban Tobing Village. The method used in this research is descriptive research using a qualitative approach. By using as many as 19 family cards, subjects or as many as 254 families receiving aid were recorded. Research conditions show that poverty conditions in Lumban Tobing Village vary due to different livelihoods, age levels of household heads and limited human resources. The method of poverty in question is the provision of subsidy program assistance in the form of BLT, BST, BPNT, PHK and basic food supplies.

Pelayanan Dinas Kependudukan dan Pencatatan Sipil terhadap Kepemilikan Kartu Tanda Penduduk, Kartu Keluarga dan Akta Kelahiran bagi Warga Masyarakat Kabupaten Timor Tengah Utara

The purpose of this research is to analyze the services of the population and civil registration offices towards ownership of identity cards, family cards and birth certificates for citizens of North Central Timor District. This research method is empirical juridical legal research, which is field research that examines the applicable legal provisions and the reality that occurs in the community. The aspects to be researched in this research are conducted through interviews, observation and documentation. The results showed that the service of DISDUKCAPIL TTU Regency was good enough, so that there was an increase in residents who wanted to take care of population documents, by looking at indicators of the five dimensions of public services, namely Tangible (Physical Evidence), Reliability, Responsiveness, Assurance, Empathy. Factors that hinder, namely: public awareness, distance, power outages resulting in disrupted networks, damage to tools/machines. Efforts of DISDUKCAPIL TTU Regency; conducting socialization related to the importance of ownership of population documents, conducting online ball pick-up activities, providing special services to people with disabilities, coordinating with parties related to public services to be able to get services from DISDUKCAPIL.

Implementation of Bekasi City Regional Health Insurance Program Implementation Policy

The main problem raised in this study is the ineffectiveness of policies and programs in Bekasi City based on Family Cards and Identity Numbers. Qualitative research strategies combined with descriptive analysis are research methods used by researchers. The expectation that original data could be collected and research subjects could be thoroughly studied to provide the desired findings led to the selection of this methodology and approach. As a result of various factors, including standard factors, policy size and objectives, resources, implementing organization characteristics, implementing attitudes, communication between related organizations and implementation activities, as well as the social, economic, and political environment, the findings of this study can be used to explain why the Bekasi City Health Insurance policy has not been implemented effectively. However, these variables, the characteristic factors of implementing agents and policy resource factors, have not been implemented properly in their implementation, so there are still obstacles in the implementation of the Regional Health Insurance policy. Of course, this is not the best approach to providing quality public services, especially in terms of the Regional Health Insurance Program for People Based on Family Cards and Identity Numbers.

NLRC4 methylation and its response to intravenous immunoglobulin therapy in Kawasaki disease: a case control study

BackgroundKawasaki disease (KD) is a systemic vasculitis accompanied by many systemic physiological and biochemical changes. Elucidating its molecular mechanisms is crucial for diagnosing and developing effective treatments. NLR Family CARD Domain Containing 4 (NLRC4) encodes the key components of inflammasomes that function as pattern recognition receptors. The purpose of this study was to investigate the potential of NLRC4 methylation as a biomarker for KD.MethodsIn this study, pyrosequencing was utilized to analyze NLRC4 promoter methylation in blood samples from 44 children with initial complete KD and 51 matched healthy controls. Methylation at five CpG sites within the NLRC4 promoter region was evaluated.ResultsCompared to controls, NLRC4 methylation significantly decreased in KD patients (CpG1: p = 2.93E-06; CpG2: p = 2.35E-05; CpG3: p = 6.46E-06; CpG4: p = 2.47E-06; CpG5: p = 1.26E-05; average methylation: p = 5.42E-06). These changes were significantly reversed after intravenous immunoglobulin (IVIG) treatment. ROC curve analysis demonstrated remarkable diagnostic capability of mean NLRC4 gene methylation for KD (areas under ROC curve = 0.844, sensitivity = 0.75, p = 9.61E-06, 95% confidence intervals were 0.762–0.926 for mean NLRC4 methylation). In addition, NLRC4 promoter methylation was shown to be significantly negatively correlated with the levels of central granulocyte percentage, age, mean haemoglobin quantity and mean erythrocyte volume. Besides, NLRC4 promoter methylation was positively correlated with lymphocyte percentage, lymphocyte absolute value.ConclusionsOur work revealed the role of peripheral NLRC4 hypomethylation in KD pathogenesis and IVIG treatment response, could potentially serve as a treatment monitoring biomarker, although its precise functions remain to be elucidated.
