Effects of Stilbestrol and Testosterone Pellets Implanted into Growing-Fattening Pigs

The effects of implanting pellets of stilbestrol and testosterone subcutaneously were studied with 42 growing-fattening pigs. Two lots of seven pigs each were assigned to each experimental group—control, testosterone implanted, and stilbestrol implanted. The implantations were made at the beginning of the experiment and 12 weeks later. The pigs averaged 43 pounds at the start of the experiment and were slaughtered as they reached 210 pounds. The following characteristics were studied: average daily gain, feed per 100 pounds gain, daily feed consumption, dressing percentage, length of carcass, weight of regular ham, percent of external fat in the regular ham, eye muscle measurement, fat back thickness, length of femur, specific gravity of the femur, weight of leaf fat, and seediness of the fresh belly. Of these characteristics, seediness of the belly in the stilbestrol group is the only one that showed a significant deviation from the control. The reproductive organs of the treated pigs showed definite evidence of hormonal stimulation.

STUDIES ON RECOVERY OF THE REPRODUCTIVE SYSTEM OF THE MALE RAT FROM REGRESSIVE CHANGES INDUCED BY STILBESTROL1

IN A RECENT PUBLICATION (i) the deleterious effects of stilbestrol on the male reproductive system were discussed. Since considerable regressive changes were found in the testes, seminal vesicles and prostate it was obviously desirable to ascertain not only if these organs could recover a functional level but also to determine the length of time necessary for this recovery after the withdrawal of treatment. With these objectives in mind experiments designed to elucidate the problem were executed. Data collected from these studies are presented in this communication. STUDIES ON STILBESTROL III. SOME EFFECTS OF CONTINUOUS INJECTIONS OF STILBESTROL IN NORMAL AND CASTRATE ADULT RATS

HAVING STUDIED THE EFFECTS of stilbestrol in the female rat, normal castrate and immature (1, 2) and found them to be typical of the natural estrogens we investigated its effect in male rats. This study was undertaken to see if stilbestrol produced the same histological changes as the natural estrogens in the pituitary, thyroid, adrenals, gonads and liver and to determine if it were any more toxic in the male than in the female. Corn oil solutions of both estrogens were used. The natural estrogens2 and stilbestrol3 employed were from the same lot as was used in the female rats. Stilbestrol was given at 5 different dose levels, namely, 250, 100, 50, 5 and 1γ daily. The natural estrogens were given at 4 different dose levels in the normal male rats.

STUDIES ON STILBESTROL II. THE EFFECT OF MASSIVE DOSES ON NORMAL IMMATURE FEMALE RATS

IN THE PREVIOUS PAPER (I) in this series we were unable to find toxic effects of stilbestrol in doses 500 times the physiologic dose. Accordingly we decided to determine the effect of much larger doses over a long period of treatment. Young immature female rats weighing 55 to 65 gm. at the start of treatment were used. In the first group designated as series A injections of 1.25 mg. of stilbestrol were given three times weekly. A second group, series B, were given 2.5 mg. of stilbestrol three times weekly. A third group, series C, served as controls. The animals were weighed at the time injections were given and at autopsy. The body length from tip of nose to tip of tail and tail length were measured at the same times. Very little difficulty was en-countered in making these measurements as the young rats quickly became condi-oned to the handling and no real restraint was necessary.

CHRONIC TOXICITY STUDIES OF DIETHYLSTILBESTROL I. ORAL ADMINISTRATION BY STOMACH TUBE TO RATS

THIS STUDY was made because of the conflicting reports in the literature regarding the toxicity of stilbestrol in man and animals. In many of these reports there is no evidence of controlled studies. The doses of stilbestrol are frequently excessive and so far above the calculated therapeutic dose range that the pathological changes reported are of little significance when considering the drug for clinical use. For these reasons this chronic toxicity study of the effects of stilbestrol in various dose ranges and in two different media was conducted. Four oral dose levels of stilbestrol were established for this study on rats as follows. (a) Fifty 7 3 times weekly (7507 weekly/kg. body weight of rat) which is more than 5 times as great as the average human therapeutic dose (1 mg. daily for a 50 kg. person) or corresponds to a weekly dose of 37.5 mg. of stilbestrol for the human being.

TREATMENT OF THE MENO-PAUSE WITH STILBESTROL

SINCE DODDS (1, 2) first reported the estrogenic effects of stilbestrol there have been numerous reports of clinical trials with this interesting drug. Bishop (3) reported that in 17 of 25 women it controlled the symptoms of the menopause. He ad-ministered stilbestrol to a total of 46 patients of all types, only 3 of whom developed nausea and vomiting. A breast biopsy, made on one patient after she had received 280 mg. of stilbestrol, showed increased epithelial activity but no evidence of carcinoma. Winterton and MacGregor (4) observed nausea in 6 of 51 patients treated for amenorrhea, dysmenorrhea or the menopause. In 5 this symptom was slight and in one, severe enough to require omission of the drug. No other toxic reactions were noted. Others have reported similar results (5-14). Moricard and Saulnier (15), on the other hand, reported nausea in 7 of 12 patients. In 3 it was severe enough to make omission of the drug necessary. A CLINICAL STUDY IN ESTROGENIC THERAPY1

AN ESTROGENIC PREPARATION that can be administered orally has long been desired. The wish seems to have been fulfilled by the discovery of stilbestrol. Favorable reports of the estrogenie efficiency of this compound are numerous, but a variety of opinions concerning its toxicity have been expressed. During the past year my associates have collaborated with me to observe the effects of stilbestrol administered to 250 patients. We have been interested to test the efficiency, toxicity, optimum dosage and indication for the use of stilbestrol. In order to avoid prejudice each observer selected his own cases, chose the dosage and recorded his results. All other complaints and symptoms of the patients as well as the estrogenic deficiency have been considered and treated concurrently with the stilbestrol therapy.
