Foliar Resistance to a Natural Population of Plasmopara viticola in Some Grapevine Varieties Grown in Türkiye

ABSTRACTIt is critical to use a population of the pathogen that contains different strains to determine the resistance of grapevine varieties to the downy mildew pathogen Plasmopara viticola in a specific region. This study tested six local grape varieties (Çavuş, Kalecik Karası, Müşküle, Papaz Karası, Sultana and Yapıncak), eight Turkish registered varieties (Barış, Bozbey, Güzgülü, Özer Karası, Reçel Üzümü, Tekirdağ Çekirdeksizi, Trakya İlkeren and Yalova İncisi) and seven varieties of foreign origin (Cabernet Sauvignon, Cardinal, Chardonnay, Cinsault, Isabella, Semillon and Gamay) using detached leaf and leaf disc assays with pathogen populations collected from vineyards. There was a significant correlation for disease severity between the two assay methods. No sporulation was observed on the varieties Çavuş and Isabella in both assay methods. The variety Yapıncak had very low disease severity (2.00%) in the leaf disc assay, and the pathogen did not sporulate on this variety in the detached leaf assay. Cluster analysis was performed to determine the resistance level of the varieties against the pathogen, as different disease severities were observed in some varieties depending on the test method used, and the 21 varieties were classified into four clusters. Barış, Bozbey, Cardinal, Çavuş, Isabella, Semillon and Yapıncak in Cluster I exhibited the lowest disease severity mean (2.88%), while those in Cluster 3, including Cabernet Sauvignon, Güzgülü, Papaz Karası and Yalova Incisi, had the highest disease severity mean (56.75%). These varieties in Clusters 1 and 3 were considered highly resistant and highly susceptible, respectively. Leaf hair density, as reported in official variety descriptions, did not associate with reduced disease severity in the two assays. This study assessed the response of grapevine varieties to the P. viticola population in the region for the first time. The highly resistant and resistant varieties identified will provide new material for breeders and contribute to the improvement of organic viticulture in the area.

A Phytocomplex Obtained from Salvia officinalis by Cell Culture Technology Effectively Controls the Grapevine Downy Mildew Pathogen Plasmopara viticola.

The negative impact of using conventional fungicides in plant disease protection has increased the interest in safer alternatives such as plant secondary metabolites, generally having a better toxicological profile. However, cultivation conditions and plant material strongly affect the quality and quantity of secondary metabolites obtained from field grown plants, limiting the standardization needed for industrial production. Plant cell culture technology can provide highly homogeneous biomasses with specific chemical characteristics. A phytocomplex with high rosmarinic acid content (10.12% w/w) was obtained from a selected cell line of Salvia officinalis and was tested against the grapevine downy mildew pathogen, Plasmopara viticola. Grapevine leaf discs were sprayed with the phytocomplex at 5 g/L and then inoculated with P. viticola sporangia. Sporulation level on each disc was assessed after 7 days with an image processing software. The phytocomplex reduced by 95% the sporulation level compared to the control and was also more effective than rosmarinic acid alone, used at the same concentration found in the phytocomplex. Persistence of the phytocomplex was also assessed: when applied 5 days before inoculation, it reduced by 90% the sporulation level compared to the control. These results highlight the possibility to take advantage of cell culture techniques to produce safer pesticides with high quality standards.

Activity and Mechanism of Action of the Bioceramic Silicon Nitride as an Environmentally Friendly Alternative for the Control of the Grapevine Downy Mildew Pathogen Plasmopara viticola.

Downy mildew of grapevine, caused by Plasmopara viticola (Berk. and Curt.) Berl. and de Toni, is one of the most devastating diseases of grapevine, severely affecting grape and wine production and quality worldwide. Infections are usually controlled by the intensive application of synthetic fungicides or by copper-based products in organic farming, rising problems for soil contamination and adverse impacts on environment and human health. While strict regulations attempt to minimize their harmful consequences, the situation calls for the development of alternative fungicidal strategies. This study presents the unprecedented case of a bioceramic, silicon nitride, with antimicrobial properties against P. viticola, but without adverse effects on human cells and environment, opening the way to the possible extension of silicon nitride applications in agriculture. Raman spectroscopic assessments of treated sporangia in conjunction with microscopic observations mechanistically showed that the nitrogen-chemistry of the bioceramic surface affects pathogen’s biochemical components and cell viability, thus presenting a high potential for host protection from P. viticola infections.

Identification and Characterization of Nep1-Like Proteins From the Grapevine Downy Mildew Pathogen Plasmopara viticola.

The obligate biotrophic oomycete Plasmopara viticola causes tremendous problems in viticulture by evoking grapevine downy mildew. P. viticola, like other plant pathogens, achieves infection by suppression of plant innate immunity by secretion of effector molecules into its host plant. An ever-expanding family of proteins with effector-like characteristics is formed by the “Necrosis and Ethylene inducing peptide 1 (Nep1)-like proteins” (NLPs). NLPs can be divided into two groups by their ability to induce necrosis. While cytotoxic NLPs may act as virulence factors for a necrotrophic or hemibiotrophic plant pathogen, the role of non-cytotoxic NLPs is so far unknown. In this study, we identified eight independent NLPs in P. viticola and selected three for functional analysis. While one was identified as a putative pseudo gene, two contain all so far described critical key elements for necrosis formation except for an N-terminal signal peptide. Further characterization revealed that none of the putative necrosis elicitors was able to actually induce necrosis, neither in several susceptible or resistant Vitis species nor in the dicot model plant Nicotiana benthamiana. This inability exists independently of the presence or absence of a signal peptide. However, any possible mechanism for the suppression of the ability to induce necrosis in planta was not detected. Interestingly, expression analysis of the presumed pseudo gene revealed remarkable differences between pure sporangia solution and sporangia in the presence of leaf material. To our knowledge, this is the first report of this kind of regulation that suggests an important function of so far nonfunctional “pseudo” NLP genes during the first hours of infection.

Rpv3\u20131 mediated resistance to grapevine downy mildew is associated with specific host transcriptional responses and the accumulation of stilbenes

BackgroundEuropean grapevine cultivars (Vitis vinifera spp.) are highly susceptible to the downy mildew pathogen Plasmopara viticola. Breeding of resistant V. vinifera cultivars is a promising strategy to reduce the impact of disease management. Most cultivars that have been bred for resistance to downy mildew, rely on resistance mediated by the Rpv3 (Resistance toP.viticola) locus. However, despite the extensive use of this locus, little is known about the mechanism of Rpv3-mediated resistance.ResultsIn this study, Rpv3-mediated defense responses were investigated in Rpv3+ and Rpv3ˉ grapevine cultivars following inoculation with two distinct P. viticola isolates avrRpv3+ and avrRpv3ˉ, with the latter being able to overcome Rpv3 resistance. Based on comparative microscopic, metabolomic and transcriptomic analyses, our results show that the Rpv3–1-mediated resistance is associated with a defense mechanism that triggers synthesis of fungi-toxic stilbenes and programmed cell death (PCD), resulting in reduced but not suppressed pathogen growth and development. Functional annotation of the encoded protein sequence of genes significantly upregulated during the Rpv3–1-mediated defense response revealed putative roles in pathogen recognition, signal transduction and defense responses.ConclusionThis study used histochemical, transcriptomic and metabolomic analyses of Rpv3+ and susceptible cultivars inoculated with avirulent and virulent P. viticola isolates to investigate mechanism underlying the Rpv3–1-mediated resistance response. We demonstrated a strong correlation between the expressions of stilbene biosynthesis related genes, the accumulation of fungi-toxic stilbenes, pathogen growth inhibition and PCD.

Plasmopara viticola effector PvRXLR131 suppresses plant immunity by targeting plant receptor-like kinase inhibitor BKI1.

SummaryThe grapevine downy mildew pathogen Plasmopara viticola secretes a set of RXLR effectors (PvRXLRs) to overcome host immunity and facilitate infection, but how these effectors function is unclear. Here, the biological function of PvRXLR131 was investigated via heterologous expression. Constitutive expression of PvRXLR131 in Colletotrichum gloeosporioides significantly enhanced its pathogenicity on grapevine leaves. Constitutive expression of PvRXLR131 in Arabidopsis promoted Pseudomonas syringae DC3000 and P. syringae DC3000 (hrcC‐) growth as well as suppressed defence‐related callose deposition. Transient expression of PvRXLR131 in Nicotiana benthamiana leaves could also suppress different elicitor‐triggered cell death and inhibit plant resistance to Phytophthora capsici. Further analysis revealed that PvRXLR131 interacted with host Vitis vinifera BRI1 kinase inhibitor 1 (VvBKI1), and its homologues in N. benthamiana (NbBKI1) and Arabidopsis (AtBKI1). Moreover, bimolecular fluorescence complementation analysis revealed that PvRXLR131 interacted with VvBKI1 in the plasma membrane. Deletion assays showed that the C‐terminus of PvRXLR131 was responsible for the interaction and mutation assays showed that phosphorylation of a conserved tyrosine residue in BKI1s disrupted the interaction. BKI1 was a receptor inhibitor of growth‐ and defence‐related brassinosteroid (BR) and ERECTA (ER) signalling. When silencing of NbBKI1 in N. benthamiana, the virulence function of PvRXLR131 was eliminated, demonstrating that the effector activity is mediated by BKI1. Moreover, PvRXLR131‐transgenic plants displayed BKI1‐overexpression dwarf phenotypes and suppressed BR and ER signalling. These physiological and genetic data clearly demonstrate that BKI1 is a virulence target of PvRXLR131. We propose that P. viticola secretes PvRXLR131 to target BKI1 as a strategy for promoting infection.

Inhibitory Effects of Macrotetrolides from Streptomyces spp. On Zoosporogenesis and Motility of Peronosporomycete Zoospores Are Likely Linked with Enhanced ATPase Activity in Mitochondria.

The release of zoospores from sporangia and motility of the released zoospores are critical in the disease cycle of the Peronosporomycetes that cause devastating diseases in plants, fishes, animals and humans. Disruption of any of these asexual life stages eliminates the possibility of pathogenesis. In the course of screening novel bioactive secondary metabolites, we found that extracts of some strains of marine Streptomyces spp. rapidly impaired motility and caused subsequent lysis of zoospores of the grapevine downy mildew pathogen Plasmopara viticola at 10 μg/ml. We tested a number of secondary metabolites previously isolated from these strains and found that macrotetrolide antibiotics such as nonactin, monactin, dinactin and trinactin, and nactic acids such as (+)-nonactic acid, (+)-homonactic acid, nonactic acid methyl ester, homonactic acid methyl ester, bonactin and feigrisolide C impaired motility and caused subsequent lysis of P. viticola zoospores in a dose- and time-dependent manners with dinactin being the most active compound (MIC 0.3 μg/ml). A cation channel-forming compound, gramicidin, and a carrier of monovalent cations, nigericin also showed similar biological activities. Among all 12 compounds tested, gramicidin most potently arrested the motility of zoospores at concentrations starting from 0.1 μg/ml. All macrotetrolide antibiotics also displayed similar motility impairing activities against P. viticola, Phytophthora capsici, and Aphanomyces cochlioides zoospores indicating non-specific biological effects of these compounds toward peronosporomyctes. Furthermore, macrotetrolide antibiotics and gramicidin also markedly suppressed the release of zoospores from sporangia of P. viticola in a dose-dependent manner. As macrotetrolide antibiotics and gramicidin are known as enhancers of mitochondrial ATPase activity, inhibition of zoosporogenesis and motility of zoospores by these compounds are likely linked with hydrolysis of ATP through enhanced ATPase activity in mitochondria. This is the first report on motility inhibitory and lytic activities of macrotetrolide antibiotics and nactic acids against the zoospores of peronosporomycete phytopathogens.

Plant-pest Interactions under Elevated CO2 Concentration in the System Grapevine (Vitis vinifera) – Downy Mildew (Plasmopara Viticola) – Grape Berry Moth (Lobesia Botrana)

Plants interact with a wide range of organisms, some of which are harmful, like pathogens and herbivorous insects. These very complex and dynamic biotic interactions can be affected by changes in environmental conditions, such as an elevated concentration of atmospheric CO2 (eCO2) as a consequence of anthropogenic global change. We hypothesize that future eCO2 alters both morphological and genetic features of grapevine plants as well as aggressiveness and pathogenicity of pest organisms, which may result in changes in future pest infestation levels in vineyards. For studying the effects of eCO2 on interactions between grapevine plants and two of their economically most significant pests – the downy mildew pathogen Plasmopara viticola and larvae of the grape berry moth Lobesia botrana – a free-air carbon dioxide enrichment (FACE) system as well as indoor climate chambers are used. The study focuses on possible changes in the biology of P. viticola (e.g. duration of incubation period, vitality of zoospores, germination of soilborne oospores) and in the developmental biology of L. botrana (e.g. hatching rate, duration of development stages, pupal weight, longevity and survival rate, fertility of females). Microscopic studies of pest- relevant morphological characteristics of grapevines (e.g. thickness of berry skin, stomatal density, stomatal aperture) are supposed to provide explanations for possible changes in pest infestation levels under eCO2. Furthermore, the expression levels of relevant defense genes in grapevines are investigated via qPCR to record possible changes in defense reactions of the host plant towards the respective pest organism. The data obtained will contribute to a better understanding of mechanisms in plant- pest interactions under altered environmental conditions.

Banchromene and other secondary metabolites from the endophytic fungus Fusarium sp. obtained from Piper guineense inhibit the motility of phytopathogenic Plasmopara viticola zoospores

A new chromene, (S)-banchromene (1), together with seven known compounds, ergosterol, beauvericin (2), fusaproliferin (3), radicinin (4), poly(3-hydroxybutyric acid) (PHB, 5), N-methylpyrrolidone and an inseparable mixture of isochromene derivatives 6a, 6b, were isolated from a culture of Fusarium sp. strain CAMKT24b1, an endophytic fungus from the leaves and twigs of Piper guineense (Piperaceae). The structures of these metabolites were elucidated on the basis of their spectroscopic data; the absolute configuration of 1 was determined by ab initio-calculation of the optical rotation. In tests with the zoospores of the grapevine downy mildew pathogen Plasmopara viticola, compounds 1–4 showed moderate to high levels of motility-impairing activity at concentrations as low as 2.5μg/mL. Compound 2 was the most active, exhibiting both motility-halting and lytic activities. Furthermore, compounds 2 and 3 displayed significant cytotoxic activity against brine shrimp larvae (Artemia salina) at 10μg/mL. This is the first report on motility inhibitory and lytic activities of metabolites from an endophytic Fusarium species against the zoospores of the downy mildew pathogen P. viticola.

Zoosporicidal metabolites from an endophytic fungus Cryptosporiopsis sp. of Zanthoxylum leprieurii

Two polyketides, cryptosporiopsin A (1) and hydroxypropan-2',3'-diol orsellinate (3), and a natural cyclic pentapeptide (4), together with two known compounds were isolated from the culture of Cryptosporiopsis sp., an endophytic fungus from leaves and branches of Zanthoxylum leprieurii (Rutaceae). The structures of these metabolites were elucidated on the basis of their spectroscopic and spectrometric data. Cryptosporiopsin A and the other metabolites exhibited motility inhibitory and lytic activities against zoospores of the grapevine downy mildew pathogen Plasmopara viticola at 10-25μg/mL. In addition, the isolated compounds displayed potent inhibitory activity against mycelial growth of two other peronosporomycete phytopathogens, Pythium ultimum, Aphanomyces cochlioides and a basidiomycetous fungus Rhizoctonia solani. Weak cytotoxic activity on brine shrimp larvae was observed.

Depsidones and other constituents from Phomopsis sp. CAFT69 and its host plant Endodesmia calophylloides with potent inhibitory effect on motility of zoospores of grapevine pathogen Plasmopara viticola

In our search for secondary metabolites regulating the motility behavior of zoospores of the grapevine downy mildew pathogen Plasmopara viticola, we found that extracts from an endophytic fungus Phomopsis sp. CAFT69 and its host plant Endodesmia calophylloides remarkably impaired motility of zoospores followed by lysis. The active principles in the extracts were isolated and identified as two new compounds, namely excelsional (1a) and 9-hydroxyphomopsidin (2a), together with excelsione (1b), phomopsidin (2b), alternariol (3a), alternariol-5-O-methyl ether (3b), the hitherto undescribed 5′-hydroxyalternariol (3c), altenusin (4) from the fungus, xanthochymol (5) and 1,5-dihydroxy-3-methoxyxanthone (mesuaxanthone, 6) from the plant. Bioassays revealed that compounds 1a/b, 2a/b, and 3a–6 displayed motility inhibition and lytic activities against zoospores of the grapevine downy mildew pathogen P. viticola in a dose- and time-dependent manner from 1 to 10μg/mL. Their structures were elucidated by extensive spectroscopic analyses including 2D NMR techniques. This is the first report of an endophyte and its natural products from E. calophylloides and the first isolation of compounds 5 and 6 from this plant.

Proteomic analysis of the compatible interaction between Vitis vinifera and Plasmopara viticola

We analyzed the proteome of grapevine (Vitis vinifera) leaves 24, 48 and 96 h post infection (hpi) with the downy mildew pathogen Plasmopara viticola. Total proteins were separated on 2-DE gels. By MS analysis, we identified 82 unique grapevine proteins differentially expressed after infection. Upregulated proteins were often included in the functional categories of general metabolism and stress response, while proteins related to photosynthesis and energy production were mostly downregulated. As expected, the activation of a defense reaction was observed more often at the late time point, consistent with the establishment of a compatible interaction. Most proteins involved in resistance were isoforms of different PR-10 pathogenesis-related proteins. Although >50 differentially expressed protein isoforms were observed at 24 and 96 hpi, only 18 were detected at 48 hpi and no defense-related proteins were among this group. This profile suggests a transient breakdown in defense responses accompanying the onset of disease, further supported by gene expression analyses and by a western blot analysis of a PR-10 protein. Our data reveal the complex modulation of plant metabolism and defense responses during compatible interactions, and provide insight into the underlying molecular processes which may eventually yield novel strategies for pathogen control in the field.

Khatmiamycin, a motility inhibitor and zoosporicide against the grapevine downy mildew pathogen Plasmopara viticola from Streptomyces sp. ANK313

In the course of our screening for anti-peronosporomycetal agents, we isolated a new compound khatmiamycin (1), together with five known metabolites, GTRI-02 (3), 4-ethyl-5-methyl-heptanamide (4), aloesaponarin II (5), LL-C10037α (6) and LL-C10037β (7) from the culture broth of a terrestrial Streptomyces sp. ANK313. The structures of these metabolites were assigned on the basis of their spectroscopic data. Khatmiamycin (1) exhibited potent motility inhibitory (100%) and lytic (83±7%) activities against zoospores of the grapevine downy mildew pathogen Plasmopara viticola at 10 μg ml(-1), followed by compounds 5 (MIC 25 μg ml(-1)), 7, 6, 3 in the order of decreasing activity. Khatmiamycin (1) also showed potent antibacterial activity against Staphylococcus aureus and Streptomyces viridochromogenes (Tü57) by causing inhibition zones of 11 and 14 mm diameter, respectively, at the dose of 40 μg per disk. This is the first report on motility inhibitory and lytic activities of metabolites from a terrestrial Streptomyces species against the zoospores of downy mildew pathogen P. viticola.

Protein Kinase C Is Likely to be Involved in Zoosporogenesis and Maintenance of Flagellar Motility in the Peronosporomycete Zoospores

The motility of zoospores is critical in the disease cycles of Peronosporomycetes that cause devastating diseases in plants, fishes, vertebrates, and microbes. In the course of screening for secondary metabolites, we found that ethyl acetate extracts of a marine Streptomyces sp. strain B5136 rapidly impaired the motility of zoospores of the grapevine downy mildew pathogen Plasmopara viticola at 0.1 μg/ml. The active principle in the extracts was identified as staurosporine, a known broad-spectrum inhibitor of protein kinases, including protein kinase C (PKC). In the presence of staurosporine (2 nM), zoospores moved very slowly in their axis or spun in tight circles, instead of displaying straight swimming in a helical fashion. Compounds such as K-252a, K-252b, and K-252c structurally related to staurosporine also impaired the motility of zoospores in a similar manner but at varying doses. Among the 22 known kinase inhibitors tested, the PKC inhibitor chelerythrine was the most potent to arrest the motility of zoospores at concentrations starting from 5 nM. Inhibitors that targeted kinase pathways other than PKC pathways did not practically show any activity in impairing zoospore motility. Interestingly, both staurosporine (5 nM) and chelerythrine (10 nM) also inhibited the release of zoospores from the P. viticola sporangia in a dose-dependent manner. In addition, staurosporine completely suppressed downy mildew disease in grapevine leaves at 2 μM, suggesting the potential of small-molecule PKC inhibitors for the control of peronosporomycete phytopathogens. Taken together, these results suggest that PKC is likely to be a key signaling mediator associated with zoosporogenesis and the maintenance of flagellar motility in peronosporomycete zoospores.

Exploiting medicinal plants and phylloplane microflora for the management of grapevine downy mildew

Grapevine downy mildew is the most devastating disease throughout the world causing huge monetary losses. Twenty medicinal plant extracts and six phylloplane microfloras were evaluated for their efficacy against sporangial germination of grapevine downy mildew pathogen Plasmopara viticola in vitro. The results revealed that the Neem Seed Kernel Extract (NSKE) at 5% significantly inhibited the sporangial germination (75.36%) of P. viticola. Among the phylloplane microflora Pseudomonas fluorescens was highly effective in reducing the sporangial germination (64.26%). Post inoculation spraying of NSKE (5%) and P. fluorescens (0.2%) effectively inhibited the disease development in the greenhouse. Three sprays with NSKE (5%) and phylloplane P. fluorescens (0.2%): first spray after initial appearance of disease and the second and third at 10 day intervals were found to be promising in reducing disease incidence in the field.
