Glucosinolate Accumulation and Hydrolysis in Leafy Brassica Vegetables Are Influenced by Leaf Age.

The health-beneficial effects of Brassica vegetables are mainly attributed to their high contents of glucosinolates and the products of their hydrolysis, especially isothiocyanates. Distribution of glucosinolates across plant organs can strongly vary. Here, we investigated the effect of leaf age on glucosinolate accumulation and hydrolysis in two leafy Brassica vegetables, pak choi and giant red mustard. We also evaluated the activity of the hydrolyzing enzyme myrosinase across the leaves. Finally, we assessed whether glucosinolates are transported from older leaves to younger leaves. Young leaves of both species contained more than 3-fold more glucosinolates than older ones. Accordingly, more isothiocyanates were released in the young leaves. Myrosinases fully hydrolyzed all of the amounts of glucosinolates regardless of the leaf age. Moreover, older leaves were observed to supply younger leaves with glucosinolates. Thus, this study suggests that consumers can improve the nutritional value of food by incorporating young leaves of leafy Brassicas in their diet.

Quantification and Diversity Analyses of Major Glucosinolates in Conserved Chinese Cabbage (Brassica rapa L. ssp. pekinensis) Germplasms

The genebank at the National Agrobiodiversity Center (RDA-Genebank, Jeonju, Republic of Korea), conserves approximately 8000 germplasms of Brassica spp., of which Chinese cabbage (Brassica rapa L. ssp. pekinensis) is one of the major crops actively used as food in Northeast Asia, including Korea, as the main ingredient for kimchi. Glucosinolates are a major class of compounds in Chinese cabbage that are responsible for their distinctive flavor, and RDA-Genebank is constantly building a related database (DB) to select suitable germplasms required by consumers and provide resources for breeding programs. In this study, ten glucosinolates were analyzed in sixty Chinese cabbage germplasms. Six aliphatic glucosinolates were the major components, accounting for 85.00% to 91.98% of total glucosinolates in each germplasm. Among them, gluconapin (333.26 to 23,501.58 μmol∙kg-1 DW) was highly represented, followed by glucobrassicanapin (545.60 to 10,344.70 μmol∙kg-1 DW) and progoitrin (155.28 to 8536.51 μmol∙kg-1 DW). In addition, we selected germplasms with a high content of each studied glucosinolate. To analyze the diversity and distribution of glucosinolates among the studied germplasms, Pearson's correlation was performed, and the related results were interpreted through their biosynthetic pathways. The k-means clustering indicated four optimal clusters, which were confirmed through principal component analysis. Orthogonal projection to latent structure discriminant analysis (OPLS-DA) was also performed on the status (landrace and cultivar) and origin (Korea, China, Taiwan, and Japan) passport data of the germplasms, followed by the calculation of variable importance in the projection (VIP) values. These results are part of a continuous series of studies to analyze the glucosinolates of Brassica germplasms that are being conserved at RDA-Genebank. We aim to provide related results through a public platform accessible to everyone and thereby improve the distribution of Brassica germplasms.

The Distribution of Glucosinolates in Different Phenotypes of Lepidium peruvianum and Their Role as Acetyl- and Butyrylcholinesterase Inhibitors-In Silico and In Vitro Studies.

The aim of the study was to present the fingerprint of different Lepidium peruvianum tuber extracts showing glucosinolates-containing substances possibly playing an important role in preventinting dementia and other memory disorders. Different phenotypes of Lepidium peruvianum (Brassicaceae) tubers were analysed for their glucosinolate profile using a liquid chromatograph coupled with mass spectrometer (HPLC-ESI-QTOF-MS/MS platform). Qualitative analysis in 50% ethanolic extracts confirmed the presence of ten compounds: aliphatic, indolyl, and aromatic glucosinolates, with glucotropaeolin being the leading one, detected at levels between 0–1.57% depending on phenotype, size, processing, and collection site. The PCA analysis showed important variations in glucosinolate content between the samples and different ratios of the detected compounds. Applied in vitro activity tests confirmed inhibitory properties of extracts and single glucosinolates against acetylcholinesterase (AChE) (15.3–28.9% for the extracts and 55.95–57.60% for individual compounds) and butyrylcholinesterase (BuChE) (71.3–77.2% for the extracts and 36.2–39.9% for individual compounds). The molecular basis for the activity of glucosinolates was explained through molecular docking studies showing that the tested metabolites interacted with tryptophan and histidine residues of the enzymes, most likely blocking their active catalytic side. Based on the obtained results and described mechanism of action, it could be concluded that glucosinolates exhibit inhibitory properties against two cholinesterases present in the synaptic cleft, which indicates that selected phenotypes of L. peruvianum tubers cultivated under well-defined environmental and ecological conditions may present a valuable plant material to be considered for the development of therapeutic products with memory-stimulating properties.

Evaluation of Seasonal Variations in the Glucosinolate Content in Leaves and Roots of Four European Horseradish (Armoracia rusticana) Landraces

In comparison with other cruciferous vegetables, horseradish has rarely been the object of scientific research, and the knowledge about the composition, content and distribution of glucosinolates (GLS) in different organs of horseradish plants is limited. Therefore, the aim of this study was to evaluate changes in the GLS content in leaves and roots of four horseradish landraces during the growing season. The presence of 13 GLS was determined in the examined horseradish tissues, and glucoraphanin, glucoraphenin and napoleiferin were noted for the first time in the species. During the growing season, the content of individual GLS changed significantly. The rate and direction of these changes varied across the examined landraces and plant organs. In the leaves, between May and June, the content of sinigrin, the main GLS in all horseradish landraces, decreased in Bavarian (40%) and Hungarian (11%) horseradish, increased (22%) in Creamy horseradish, whereas in Danish horseradish, the difference was not significant. Despite the changes observed in the first two months, the highest content of sinigrin was noted in July in all horseradish landraces. During the growing season (August-October), the content of sinigrin fluctuated in the roots of Creamy and Danish landraces, reaching the highest level in October and September, respectively, whereas in the roots of Hungarian and Bavarian landraces, sinigrin concentrations continued to increase and peaked in October. Changes in the content of other, minor GLS during the growing season often differed from those noted in sinigrin levels.

Distribution of glucosinolates in camelina seed fractions by HPLC‐ESI‐MS/MS

High glucosinolate concentrations were detected and identified in camelina (Camelina sativa L. Crantz.) seed fractions using reversed phase high performance liquid chromatography‐electrospray ionization‐mass spectrometry (HPLC‐ESI‐MS) in multiple reaction monitoring mode. Total glucosinolate quantitation was performed using proton nuclear magnetic resonance spectrometry using N,N‐dimethylformamide as an internal standard. Individual glucosinolate concentrations were determined by analysis on extracted ion chromatograms generated by a MS detector. Distribution of glucosinolates in C. sativa seed fractions during pressing is reported.Practical applications: HPLC‐ESI‐MS and HPLC‐ESI‐MS/MS using monolithic HPLC columns were used to detect, quantify, and identify the three major glucosinolates in Camelina sativa L. and fractions of C. sativa produced by processing. Quantitative extracted ion MS chromatograms afforded excellent quantitation of individual glucosinolates. Uncovering different parameters influencing florivory in a specialist herbivore

1. It has become increasingly recognised that several herbivores switch from folivory (leaf-feeding) to florivory (flower-feeding) during larval development. Yet, it remains poorly understood which cues influence this behaviour, whether a switch to florivory is consistently shown on different hosts, and to what extent florivory could be hindered by plant traits. 2. Using the sawfly Athalia rosae and two Brassicaceae differing in architecture and surface structure, the cues that influence larval movement to the flowers were investigated. A broad set of behavioural assays was employed and physical and chemical plant traits potentially affecting the larvae were analysed. Furthermore, the consequences of folivory versus florivory on insect performance were studied. 3. The larvae preferred flowers over leaves. Consumption of particular flower parts correlated partly with measured plant traits such as glucosinolate distribution. Visual cues were of higher importance than volatile cues. The initial position of newly hatched larvae on plants influenced the probability of the larvae reaching the flowers during development. Trichomes and surface waxes hindered the larvae from moving upwards to the flowers. Larvae developed slower and gained less body mass when feeding on inflorescences of Brassica nigra than when feeding on leaves, in contrast to the patterns observed before on Sinapis alba, where florivory led to an improved performance. 4. This study demonstrates that florivory depends on various host plant traits. It reveals new insights into different parameters influencing this multifaceted phenomenon and into the expected impact on the ecology and fitness of both the attacking herbivores and the plants.

Glucosinolate profile and distribution among plant tissues and phenological stages of field-grown horseradish

Profile and distribution of glucosinolates (GLS) were detected in plant tissues of horseradish at different developmental stages: beginning of vegetative re-growth, flowering and silique formation. The GLS profile varied widely in the different tissues: we identified 17 GLS in roots and sprouts, one of which was not previously characterized in horseradish, i.e. the 2(S)-hydroxy-2-phenylethyl-GLS (glucobarbarin) and/or 2(R)-hydroxy-2-phenylethyl-GLS (epiglucobarbarin), 11 already found in the roots, including the putative 2-methylsulfonyl-oxo-ethyl-GLS, and 5 previously recognized only in the sprouts. Fifteen of those GLS were also identified in young and cauline leaves, 12 in the mature leaves and 13 in the inflorescences. No difference in GLS profile was observed in plant among the phenological stages. Differences in concentrations of GLS, quantified as desulfated, were found in plant. At the beginning of vegetative re-growth, sprouts while showing the same profile of the roots were much richer in GLS having the highest total GLS concentrations (117.5 and 7.7μmolg−1 dry weight in sprouts and roots, respectively). During flowering and silique forming stages, the roots still maintained lower amount of total GLS (7.4μmolg−1 of dry weight, on average) with respect to the epigeous tissues, in which mature and young leaves showed the highest total concentrations (70.5 and 73.8μmolg−1 of dry weight on average, respectively). Regardless of the phenological stages, the aliphatic GLS were always predominant in all tissues (95%) followed by indolic (2.6%) and benzenic (2.4%) GLS. Sinigrin contributed more than 90% of the total GLS concentration. Aliphatic GLS concentrations were much higher in the epigeous tissues, particularly in the mature and young leaves, while benzenic and indolic GLS concentrations were higher in the roots. Through the phenological stages, GLS concentration increased in young and mature leaves and decreased in cauline leaves and inflorescences, while it remained constant over time in roots.

Glucosinolate profiling of seeds and sprouts of B. oleracea varieties used for food

Consumption of plants of the species Brassica oleracea is related to a decreased incidence of certain cancer forms in humans, and this has been linked to the presence of glucosinolates in those vegetables. After ripe seeds, sprouts of some brassicaceous plants contain the highest concentration of these compounds and are therefore a good source of glucosinolates for chemoprotection. In the present experiments, the content and distribution of glucosinolates was determined in ripe seeds and sprouts (seedlings) of five varieties of B. oleracea (white cabbage, red cabbage, Savoy cabbage, broccoli and cauliflower) by high performance liquid chromatography. The type and concentration of individual glucosinolates varied according to variety of B. oleracea, plant parts in which they occurred and the sprouting period of the seed. Concentration of alkyl glucosinolates decreased whereas that of indol-3-ylmethylglucosinolates increased throughout the sprouting period. Roots had the highest glucosinolate concentration in four and seven day old sprouts whereas at both sprouting times, cotyledons had the highest concentration of alkylthio- and alkylsulphinylglucosinolates.

The interest in food with chemoprotective properties has been steadily increasing due to many epidemiological studies indicating the lower probability of acquiring some kinds of cancer (e.g. colon, prostate, breast, cervical) in populations, whose diet includes large quantities of Brassica vegetables. The biologically active compounds in Brassica vegetables are the breakdown products of glucosinolates, including isothiocyanates, nitriles, thiocyanates, indoles and oxazolidinethiones from which indoles and isothiocyanates in particular have been implicated to have anticarcinogenic properties. The object of the present study was to investigate glucosinolates distribution as well as their losses during blanching and cooking in edible parts of white cabbage, cauliflower, kohlrabi and collard. In all vegetable, only indole glucosinolates as precursors of anticarcinogenic compounds were identified, while glucoraphane was not detected. In cauliflower and collard, proportion of indole glucosinolates was more then 90%, followed by kohlrabi (>70%) and white cabbage (>50%). Analysis of glucosinolates losses during blanching and cooking showed substantial decrease in all vegetables. The major losses were observed for cauliflower, around 60%, for cooked vegetable, followed by white cabbage (55%), kohlrabi (53%) and collard (44%). The losses after blanching were approximately 15% less then in cooked vegetables.

Response of Drosophila to Wasabi Is Mediated by painless, the Fly Homolog of Mammalian TRPA1/ANKTM1

A number of repellent compounds produced by plants elicit a spicy or pungent sensation in mammals . In several cases, this has been found to occur through activation of ion channels in the transient receptor potential (TRP) family . We report that isothiocyanate (ITC), the pungent ingredient of wasabi, is a repellent to the insect Drosophila melanogaster, and that the painless gene, previously known to be required for larval nociception, is required for this avoidance behavior. A painless reporter gene is expressed in gustatory receptor neurons of the labial palpus, tarsus, and wing anterior margin, but not in olfactory receptor neurons, suggesting a gustatory role. Indeed, painless expression overlaps with a variety of gustatory-receptor gene reporters. Some, such as Gr66a, are known to be expressed in neurons that mediate gustatory repulsion . painless mutants are not taste blind; they show normal aversive gustatory behavior with salt and quinine and attractive responses to sugars and capsaicin. The painless gene is an evolutionary homolog of the mammalian "wasabi receptor" TRPA1/ANKTM1 , also thought to be involved in nociception. Our results suggest that the stinging sensation of isothiocyanate is caused by activation of an evolutionarily conserved molecular pathway that is also used for nociception.

Distribution of glucosinolates in ornamental cabbage and kale cultivars

Glucosinolate concentrations were measured in seven ornamental cabbage and six ornamental kale ( Brassica oleracea var. acephala) cultivars. Fifteen glucosinolates were detected in varying concentrations, but only four (progoitrin, sinigrin, glucoraphanin and glucobrassicin) were found in concentrations greater than 0.5 μmol g −1 dry weight. In general, total glucosinolates in ornamental cabbage were about 35% higher than in ornamental kale. Differences in total glucosinolates were also observed within each Brassica group. In cabbage, total glucosinolates were significantly higher in ‘Osaka Pink’, ‘Osaka Red’, ‘Pigeon Red’ and ‘Tokyo Pink’ than in ‘Osaka White’ or ‘Tokyo White’, while total glucosinolates in ‘Tokyo Red’ was not significantly different from the other five cultivars. In kale, total glucosinolate concentration was highest (17.16 μmol g −1 dry weight) in ‘Coral Prince’ and lowest (6.8 μmol g −1 dry weight) in ‘Sparrow White’. Ornamental cabbage cultivars contained higher glucoraphanin, followed by sinigrin, progoitrin and glucobrassicin, while ornamental kale cultivars contained about the same concentrations of glucoraphanin, sinigrin and glucobrassicin, except ‘Coral Prince’ which contained three- to nine-fold higher glucoraphanin than the other five kale cultivars. The presence of higher concentrations of glucoraphanin in ornamental cabbage and ornamental kale warrant further investigation into their potential use as breeding stocks to enhance the level of this important phytochemicals in edible cabbage and kale.

Distribution of Glucosinolates and Myrosinase Activity in Cruciferous Vegetables

A diet rich in curciferous vegetables has been associated with inhibition of chemically induced carcinogenesis in laboratory animals and humans. The underlying mechanism(s) for this inhibition is not completely clear; however, cruciferous vegetables are rich in sulfur-containing glucosides called gluco-sinolates. Myrosinase-mediated breakdown of glucosinolates has been shown to produce compounds with anti-cancer activity. About 100 forms of glucosinolates have been identified, mostly in the Cruciferae family. In this study, we have developed a unique data set of glucosinolate types and amounts in 51 broccoli, three cauliflower, five Brussels sprouts, five cabbage, and two kale lines. The types of glucosinolates and their concentrations were variable among the different genotypes and within each genotype. The dominant glucosinolates in broccoli were glucoraphanin, gluconapin, and glucobrassicin. Glucoraphanin concentration in broccoli ranged from 0.1 μmol·g–1 dry weight in EV6-1 to 21.7 μmol·g–1 dry weight in Brigadier. Concentrations of the other types of glucosinolates in broccoli also showed wide range of variability. In cabbage, Brussels sprouts, cauliflower, and kale, the dominant glucosinolates were sinigrin (7.8, 8.9, 9.3, and 10.3 μmol·g–1 dry weight, respectively) and glucobrassicin (0.9, 3.2, 1.3, and 1.2 μmol·g–1 dry weight, respectively). Brussels sprouts also has significant amounts of gluconapin (6.8 μmol·g–1 dry weight). Myrosinase activity, using sinigrin as substrate, was significantly higher in cabbage, Brussels sprouts, cauliflower, and kale than in broccoli. Wide variations in glucosinolates, glucosinolates content, and myrosinase activity among cruciferous genotypes and within each genotype suggest differences in the health-promoting properties of these vegetables.

Glucosinolate Content and Susceptibility for Insect Attack of Three Populations of Sinapis alba

Sinapis alba is less susceptible to damage by insect pests than Brassica napus. We investigated the composition and distribution of glucosinolates in different plant parts in three populations of S. alba; two populations selected for low-seed-glucosinolate content and one commercial cultivar. We have assessed the susceptibility of low-seed-glucosinolate content populations of S. alba to four insect pests, a flea beetle, a pollen beetle, and two species of aphids. Over 90% of the total glucosinolates in the cotyledons of the three populations of S. alba consisted of sinalbin. There was no difference in feeding damage by flea beetles on different S. alba populations at the cotyledon stage, nor was there a difference in sinalbin concentration of cotyledons. Total glucosinolate levels were highest in younger plant tissues. Sinalbin declined as a proportion of total glucosinolate content in later growth stages, especially in the “low” breeding lines. Reproduction by aphids was the same on all three populations despite differences in sinalbin content of the S. alba leaves at the growth stage tested. The specialist aphid, Brevicoryne brassicae, was found mainly on young tissues, while the generalist aphid, Myzus persicae, was found predominantly on older plant parts. There was no difference in oviposition by pollen beetles between the S. alba populations, despite the fact that on one population total glucosinolate concentration and the proportion of sinalbin found in the buds were much lower than on the other two populations.

Physiological role of glucosinolates in brassica napus. Concentration and distribution pattern of glucosinolates among plant organs during a complete life cycle

AbstractConcentrations and distribution of glucosinolates in various organs of rape (Brassica napus L) were examined during major stages of development. Fluctuations in the total content as well as in the individual levels of glucosinolates were investigated in double low Drakkar and high glucosinolate Chine 32 cultivars. The distribution pattern of aliphatic and aromatic glucosinolates between the various organs, as well as the variation in the concentrations of individual glucosinolates during plant development were quite similar between the two cultivars. Peculiar stages of accumulation during the vegetative growth period and the seed maturation period were identified. These stages were followed by periods of intense degradation during flowering, germination and early growth stages of seedlings. Fluctuations in the amount and structure of the glucosinolates in each of these periods suggest a storage function for these compounds which are required at specific stages of development. The involvement of some of these compounds in hormonal control of flowering is also discussed.

Distribution and chemotaxonomic significance of glucosinolates in certain Middle-Eastern cruciferae

Eighty-four collections of 51 species of Middle-eastern Cruciferae were analysed by paper and gas-liquid chromatography for seed glucosinolates. Thirty-two species were investigated for the first time. Thirty compounds were identified and three unknowns of limited occurrence were found, p-Hydroxybenzylglucosinolate was found in six species of various tribes. 4-Methylsulfinyl-3-butenyl, 3-methoxycarbonylpropyl, and p-rhamnopyranosyloxybenzyl glucosinolates, which are rare in the Cruciferae, were found in Sinapis aucheri two species of Erysimum and Thlaspi perfoliatum, respectively. The biosynthetic precursors of all but one of the 30 glucosinolates are protein amino acids and at least 20 glucosinolates are derived from methionine. The distribution of glucosinolates is very useful at the species and genus levels but may provide only minimal support to the tribal alliance of certain genera. It supports the removal of Sinapis aucheri from the genus, the recognition of Fibigia clypeata and F. macrocarpa as distinct species, and the alliance of Ochthodium with Euclidium and Arabis with Drabopsis.

Analytical Methodology for Determining Glucosinolate Composition and Content

Abstract New analytical techniques and instrumentation and increased knowledge of the diversity and distribution of glucosinolates, the diversity of their enzymatically released products, and factors influencing their release, have led to significant advances in methodology for analysis of glucosinolates over the past three decades. However, many of the methods have been developed for specific agricultural crops or commodities and their particular glucosinolate compositions. They can only be applied to certain types of plant material or can detect and quantitate only certain glucosinolates or their products. Other methods have been designed to meet specific research, regulatory, or quality control requirements. This had necessitated sacrifice of either speed, simplicity, accuracy, precision, or the ability to distinguish different glucosinolates or their products. This review examines the methods available for sample preparation, identification, and quantitation of glucosinolates in light of current knowledge of their diversity, distribution, and chemistry. Consideration is given to the suitability of methods for rapid screening or precise, discriminating measurement, and to the standardization of methodology and reporting of results.

4,5,6,7-tetrahydroxydecyl isothiocyanate derived from a glucosinolate in Capparis grandis

Root material of Capparis grandis, of Indian origin, contains a free isothiocyanate, identified as 4,5,6,7-tetrahydroxydecyl isothiocyanate 1 by spectroscopical and chemical means, yet without specification as to its stereochemistry. The presence of the parent glucosinolate, and three other, unidentified glucosinolates in C. grandis has been noted. The distribution of glucosinolates within the genus Capparis is reviewed and the biosynthetic origin of the individual compounds discussed.
