Localization, identification, and action of galanin in the frog adrenal gland

The distribution of galanin-like immunoreactivity was studied in the adrenal gland of the frog Rana ridibunda using the indirect immunofluorescence technique. A dense network of varicose fibers immunoreactive to galanin was found in the adrenal tissue. A combination of HPLC analysis and RIA detection was used to characterize galanin-like immunoreactivity in frog adrenal gland extracts. The elution profile revealed the existence of a single form of galanin exhibiting the same retention time as synthetic frog galanin. The possible involvement of galanin in the regulation of corticosteroid secretion was investigated in vitro using a perifusion system for frog adrenal slices. For concentrations ranging from 10(-9) to 3 x 10(-6) M, synthetic frog galanin induced a dose-dependent inhibition of corticosterone and aldosterone release. Repeated pulses of galanin (10(-6) M), given at 90-min intervals, resulted in a reproducible inhibition of corticosteroid secretion without any apparent tachyphylaxis. During prolonged administration of galanin (10(-6) M), the steroidogenic effect of ACTH (10(-9) M) was significantly reduced. In contrast, galanin did not attenuate the stimulation of corticosteroid secretion induced by the angiotensin II analog [Sar1,Val5]angiotensin II. These results show the occurrence of galanin in fibers innervating the frog adrenal gland. The data also demonstrate that synthetic galanin inhibits spontaneous and ACTH-induced corticosteroid release. Taken together, these findings suggest that galanin, released by nerve fibers in the adrenal tissue, can act locally as a modulator of steroid secretion.

Extensive Distribution of Galanin-Immunoreactive Neuronal Systems in a Fresh Water Snail, Indoplanorbis exustus, and Partial Coexistence with Choline Acetyltransferase-Iike Immunoreactivity

Abstract Immunohistochemical techniques were employed to investigate the distribution of galanin-like immunoreactivity in the central nervous system (CNS) of the snail, Indoplanorbis exustus, and immunoreactivity with an antibody against choline acetyltransferase (CAT) in galanin-immunoreactive somata was also examined. Galanin-immunoreactive (Gal-IR) cells were observed in all ganglia. They were classified into large (50 μm), medium (21 μm), and small (13 μm)-sized neurons and small Gal-IR cells dominated over large and medium Gal-IR cells in number. Gal-IR cells were most abundant in the cerebral ganglion. Several immunoreactive somata were seen in the parietal, visceral, and pedal ganglia, and only a few Gal-IR cells in the pleural and buccal ganglia. Densely arranged Gal-IR fiber bundles were observed in the cerebro-pleural, parieto-pleural and bucco-cerebral connectives. The prepubertal ontogeny of galanin-like immunoreactivity in the male Meishan pig brain

Galanin (GAL) is a neuropeptide found in the mammalian brain and is involved in numerous functions including the control of feeding, growth and reproduction, and therefore may be an important peptide to study in agricultural species. We describe the immunohistochemical localization of GAL throughout prepubertal development in the Meishan pig, a Chinese breed known for its superior reproductive characteristics, but slow growth rate and abundant adipose tissue. Brains of animals from gestational day (g) 30, 50, 70, 90 and 110 and postnatal day (pn) 1, 10, 20 and 50 (duration of pregnancy averaged 114 days) were processed using a standard immunohistochemical technique utilizing a commercially available rabbit anti-porcine GAL antibody. Galanin-like immunoreactivity (GAL-IR) in cell bodies and fibers was evident in the brain at g30, primarily in the hypothalamus. Throughout prenatal development, cell bodies containing GAL-IR generally increased in number and distribution in the brain. During postnatal development, the number of cell bodies displaying GAL-IR decreased, particularly in hypothalamic areas. The distribution of GAL-IR in fibers became more widespread throughout gestational development, showing a pattern by pn1 that continued during later postnatal ages. The intensity of GAL-IR in fibers also increased throughout gestation. Some additional increases in immunoreactivity occurred postnatally, especially in the periventricular hypothalamus. The results of this study indicate that the distribution of GAL-IR in cell bodies and fibers in the Meishan pig brain was similar to that seen in other species, including the rat. These results support the hypothesis that GAL participates in the control of feeding, growth and reproduction in the pig.

Localisation of mRNA encoding the protein precursor of galanin in the monkey hypothalamus and basal forebrain.

The hypothalamic and basal forebrain sites of synthesis of preprogalanin mRNA were identified in three adult monkeys (Macaca fascicularis) by in situ hybridisation performed with a radiolabelled cRNA probe transcribed from human preprogalanin cDNA. With stringent hybridisation conditions, the cRNA probe was hybridised to free-floating sections containing structures contiguous with the rostral hypothalamus through to the caudal limit of the hypothalamus as defined by the mammillary bodies. Specific hybridisation of the preprogalanin cRNA probe occurred throughout the hypothalamus but was particularly intense in the arcuate, paraventricular (parvicellular and magnocellular portions), and dorsomedial nuclei. Moderate hybridisation was found in the periventricular nucleus and scattered hybridisation in the medial preoptic nucleus. The medial preoptic area and the anterior and lateral hypothalamic areas showed moderate to intense hybridisation in scattered cells. A few cells in the tuberal portion and dorsal cap of the anterior portion of the supraoptic nucleus were labelled. Isolated cells were also labelled in the zona incerta. There was little labelling in the dorsal hypothalamic area but moderate labelling in the posterior hypothalamic area. Structures contiguous with the rostral hypothalamus including the diagonal band of Broca, bed nucleus of stria terminalis, substantia innominata, and basal nucleus of Meynert showed intense hybridisation. These data indicate a widespread distribution of preprogalanin mRNA in the monkey hypothalamus. A comparison with the previously reported distribution of preprogalanin mRNA in the rat, as well as with the distribution of galanin-like immunoreactivity in the rat and human, suggests some important species differences. Of particular interest were differences in the supraoptic, suprachiasmatic, and dorsomedial nuclei. The intense hybridisation throughout the paraventricular nucleus and in the rostral arcuate nucleus suggests that galanin may play a role in the regulation of both posterior and anterior pituitary function.

Distribution of galanin‐like immunoreactivity in the brain of Rana esculenta and Xenopus laevis

The immunocytochemical distribution of galanin-containing perikarya and nerve terminals in the brain of Rana esculenta and Xenopus laevis was determined with antisera directed toward either porcine or rat galanin. The pattern of galanin-like immunoreactivity appeared to be identical with antisera directed toward either target antigen. The distribution of galanin-like immunoreactivity was similar in Rana esculenta and Xenopus laevis except for the absence of a distinct laminar distribution of immunoreactivity in the optic tectum of Xenopus laevis. Galanin-containing perikarya were located in all major subdivisions of the brain except the metencephalon. In the telencephalon, immunoreactive perikarya were detected in the pars medialis of the amygdala and the preoptic area. In the diencephalon, immunoreactive perikarya were detected in the caudal half of the suprachiasmatic nucleus, the nucleus of the periventricular organ, the ventral hypothalamus, and the median eminence. In the mesencephalon, immunoreactive perikarya were detected near the midline of the rostroventral tegmentum, in the torus semicircularis and, occasionally, in lamina A and layer 6 of the optic tectum. In the myelencephalon, labelled perikarya were detected only in the caudal half of the nucleus of the solitary tract. Immunoreactive nerve fibers of varying density were observed in all subdivisions of the brain with the densest accumulations of fibers occurring in the pars lateralis of the amygdala and the preoptic area. Dense accumulations of nerve fibers were also found in the lateral septum, the medial forebrain bundle, the periventricular region of the diencephalon, the ventral hypothalamus, the median eminence, the mesencephalic central gray, the laminar nucleus of the torus semicircularis, several laminae of the optic tectum, the interpeduncular nucleus, the isthmic nucleus, the central gray of the rhombencephalon, and the dorsolateral caudal medulla. The extensive system of galanin-containing perikarya and nerve fibers in the brain of representatives of two families of anurans showed many similarities to the distribution of galanin-containing perikarya and nerve fibers previously described for the mammalian brain.

Galanin and neuropeptide Y in chromaffin granules from the guinea-pig

We have examined the subcellular distribution of galanin-like immunoreactivity, neuropeptide Y-like immunoreactivity and the catecholamines noradrenaline and adrenaline in the adrenal medulla from guinea-pigs. By differential centrifugation of the adrenal medulla homogenate the neuropeptides as well as the catecholamines sedimented in a 10,000 g pellet. This pellet was resuspended and further examined in discontinuous and continuous density gradients. In the discontinuous gradient the catecholamines peaked in the heavy bottom fraction, assumed to contain chromaffin granules. Galanin-like immunoreactivity and neuropeptide Y-like immunoreactivity were also enriched in this fraction. However, both neuropeptides showed high levels of sedimentable material also in a fraction of intermediate density. In the continuous density gradient, the sum of sedimentable and soluble catecholamines showed peak values in two fractions corresponding to 1.07 and 1.47 M sucrose, respectively. The NA peak in the denser fraction was more pronounced than the corresponding A peak. Galanin-like immunoreactivity showed only one peak, in the fraction corresponding to 1.07 M sucrose. The data suggest that galanin-like immunoreactivity and neuropeptide Y-like immunoreactivity are partly stored with catecholamines in chromaffin granules. However, galanin-like immunoreactivity and neuropeptide Y-like immunoreactivity was also found in fractions lighter than those containing the bulk of the catecholamines.

Distribution of 125I‐galanin binding sites, immunoreactive galanin, and its coexistence with 5‐hydroxytryptamine in the cat spinal cord: Biochemical, histochemical, and experimental studies at the light and electron microscopic level

The distribution of galanin-like immunoreactivity (GAL-LI) in the spinal cord of the cat was studied by use of indirect histochemistry and the peroxidase-antiperoxidase (PAP) technique. In the ventral horn GAL-immunoreactive (IR) axonal fibers and terminals were most frequent in the ventral part of the motor nucleus. The GAL-IR axons also contained 5-hydroxytryptamine (5-HT)-LI, and they disappeared after spinal cord transection. It was concluded that these GAL-IR fibers belong to the serotoninergic bublospinal pathway. In the medulla oblongata from normal cats, scattered GAL-IR cell bodies were encountered within the nucleus raphe obscurus and nucleus raphe pallidus. Electron microscopic observations revealed that the fine structure of the GAL-IR axonal boutons in the motor nucleus was similar to that of 5-HT-IR boutons with a varying number of immunoreactive large dense core vesicles. The postsynaptic element in all cases studied was a dendrite. A dense GAL-IR axonal plexus was found in the superficial laminae I-II of the dorsal horn. Coexistence was found between the GAL- and substance P-LI in fibers within the dorsal horn plexus. Spinal cord transection did not alter the pattern of GAL-LI in the dorsal horn, while the vast majority of GAL-IR axonal swellings disappeared following dorsal root sectioning. Electron microscopic observations in lamina II (substantia gelatinosa) revealed that the GAL-IR axonal terminals could be divided into two main groups. One with small to medium-sized axonal boutons formed synaptic contacts with both dendritic and axonal profiles. The other formed the central axon terminals of glomeruli, suggesting that GAL-LI may be present in C-type primary afferents. Numerous small GAL-IR cell bodies were encountered in laminae II and III. GAL-IR cell bodies were also observed in lamina X. The dorsal root ganglia contained a low but consistent number of small to medium-sized GAL-IR cell bodies, which all contained immunoreactive calcitonin gene-related peptide (CGRP). Following peripheral sciatic nerve transection, the number and the labeling intensity of GAL-IR cell bodies in the corresponding dorsal root ganglia showed a moderate increase. Radioimmunoassay revealed that the concentration of GAL-LI increased along the rostrocaudal axis of the normal spinal cord, and was about three times higher in the dorsal than in the ventral regions. The concentration in the dorsal root ganglia was intermediate to those seen in the corresponding dorsal and ventral cord regions.(ABSTRACT TRUNCATED AT 400 WORDS)

Immunohistochemical mapping of galanin-like immunoreactivity in the brain of the dogfish Scyliorhinus canicula

The distribution of galanin-like immunoreactivity in the brain of the dogfish Scyliorhinus canicula was investigated using the indirect immunofluorescence technique. In the telencephalon, positive cells and fibers were located in the mid-caudal part of the area superficialis basalis, the n. septi caudoventralis and in the n. interstitialis commissurae anterioris. Most of the galanin-containing neurons observed in the hypothalamus were located in the magnocellular preoptic nucleus. Positive perikarya were also found in the n. lobi lateralis hypothalami and in the n. lateralis tuberis. A dense network of positive nerve processes was noted in the caudal part of the median eminence. In the dorso-caudal part of the diencephalon numerous immunoreactive neurons were seen in the recessus posterioris. A large bundle of galanin-containing fibers, which divided in two branches, was observed in the basal midbrain tegmentum. The widespread distribution of galanin-like material suggests that, in the dogfish, galanin may be involved in various brain functions including neuroendocrine regulations.

Distribution and Effect of Galanin on Gallbladder and Sphincter of Oddi Motility in the Pig

This study was designed to determine the occurrence and topographical distribution of galanin-like immunoreactivity (GAL-LI) in the porcine gallbladder and sphincter of Oddi and to investigate the pharmacologic effect of GAL on gallbladder and sphincter of Oddi motility. By radioimmunoassay the concentration of GAL-LI in the gallbladder was 2.75 ± 0.23, 9.73 ± 1.33 in the common bile duct and 5.10 ± 0.37 in the sphincter of Oddi (pmol/g ± SE). By immunohistochemistry GAL-LI was found exclusively in ganglionic cells and in nerve fibers among the smooth muscle bundles. Gallbladder and sphincter of Oddi pressures were recorded before and during 5-minute local intraarterial infusion of 4, 8, 19, 39, 78 and 194 ng GAL-Kg-1-min-1 in 12 anaesthetized pigs. GAL in doses ≥ 39 ng.kg-1.min-1 significantly reduced sphincter of Oddi phasic wave frequency (4.8 ± 0.4 vs. 2.1 ± 0.5; p = 0.004) and sphincter of Oddi motility index (70.2 ± 6.02 vs. 27.7 ± 8.3; p = 0.002) but did not affect gallbladder pressure. We conclude that the distribution of GAL-LI in the sphincter of Oddi and the effect that a pharmacologic dose of GAL has on sphincter of Oddi motor activity, suggests that GAL may be involved in the physiologic control of bile flow in the pig.

Distribution of galanin-like immunoreactivity in the pig, rat and human central nervous system

The distribution of galanin-like immunoreactivity in various regions of the central nervous system was assessed in three mammalian species, pig, rat, and human, by radioimmunoassay. Galanin concentrations were highest in the hypothalamus and pituitary region. In spinal cord, there was a rostrocaudal/dorsoventral gradient with highest levels observed in the sacral dorsal horn. Serial dilutions of porcine tissue extracts diluted parallel to the porcine standard curve, while the rat and human tissue extracts did not. In all tissues examined by high pressure liquid chromatography, the principal peak of immunoreactivity coeluted with the authentic porcine galanin standard and was decreased by trypsin cleavage. These results suggest a role for galanin in the central nervous system and support species differences in the structure of galanin.

Estrogen Increases Galanin Immunoreactivity in Hyperplastic Prolactin-Secreting Cells in Fisher 344 Rats*

Galanin is a widely distributed regulatory peptide which modulates the pituitary secretion of PRL and GH. Estrogen administration strongly stimulates galanin gene expression in the rat anterior pituitary. In adult female Fischer 344 rats, estrogen also induces hyperplasia of lactotropes. We used immunocytochemical analysis to assess the effects of estrogen on galanin-like immunoreactivity (Gal-IR) in the rat pituitary and hypothalamus during sc diethylstilbestrol (DES) implantation and after its removal at 30 days. In the anterior pituitary, DES implantation increased the portion of Gal-IR-containing cells from less than 2% in the control rats to 18.3% after 3 days of DES and 36% after 30 days. These changes paralleled the lactotrope hyperplasia exhibited in response to DES exposure. Ten and 30 days after removal of the DES capsules, the percentage of Gal-IR-containing cells in the anterior pituitary decreased to 6.3% and 1.5%, respectively. Colocalization studies revealed that Gal-IR-containing cells were predominantly lactotropes. Immunoelectron microscopy demonstrated that Gal-IR was concentrated in the Golgi region of these hyperplastic lactotropes and suggests that little of the synthesized galanin is secreted. The distribution of Gal-IR in the hypothalamus, median eminence, and neurohypophysis was unaffected by DES treatment. These data demonstrate that galanin is synthesized by hyperplastic pituitary lactotropes of Fischer 344 rats and that peptide accumulation is dependent on the presence of circulating estrogens. In contrast, neuronal galanin synthesis in the hypothalamus does not appear to be regulated by estrogen.

Distribution of galanin-like peptide in various tissues of Necturus maculosus

Galanin is a biologically active peptide which has a wide pattern of distribution in the mammalian central and peripheral nervous systems. However, the distribution of galanin-like immunoreactivity in amphibian species has not been well elucidated. In the present study, biochemical and immunohistochemical techniques were used to determine the relative concentrations, biochemical nature, and cellular localization of galanin-like immunoreactivity in the brain, heart, urinary bladder, and small intestine of Necturus maculosus (common name: mudpuppy). The results of this study indicate that each of these types of tissue contain a galanin-like peptide which is similar to porcine galanin. Brain and heart concentrations of galanin-like immuno-reactivity were particularly high, although substantial amounts were also present in the small intestine and urinary bladder. Galanin immunoreactivity was observed in ascending fiber tracts of the brainstem and in fibers in the hypothalamus. In addition, galanin immunoreactivity was observed in autonomic neurons and processes in the heart, bladder, and small intestine. The pattern of distribution of galanin-like immunoreactivity in many tissues of this amphibian species is similar to the previously described mammalian pattern; however, galanin-immunoreactive innervation of cardiac tissue has not been reported in mammals. We suggest that galanin-like immunoreactivity in the heart may be more extensive in amphibian species than in mammals.

Distribution of galanin-like immunoreactivity in the human brain

The distribution of galanin-like immunoreactivity has been studied in complete coronal sections taken from 9 normal human brains (4 male, 5 female mean age 54, range 42–72 years). Galanin-immunoreactive cells were restricted largely to the basal nucleus of Meynert and to the supraoptic, ventromedial and posterior areas of the hypothalamus. Fibre staining was more widespread, seen throughout the hypothalamus and in the diagonal band, septum, amygdala, hippocampus and scattered throughout the cortex. This distribution may indicate a role for galanin in the modulation of hypothalamic and basal forebrain function in man.

Distribution of galanin-like immunoreactivity in baboon brain

Galanin-like immunoreactivity (GLI) was measured in baboon brains using a recently developed radioimmunoassay. Concentrations were measured in 10 cortical regions, hippocampus and 20 subcortical regions. The highest concentrations were in the median eminence, followed by hypothalamus, locus ceruleus, periaqueductal grey, bed nucleus of the stria terminalis, septum, amygdala and substantia innominata. Substantial amounts were also measurable in the inferior olive, basal ganglia and thalamus with very low levels in cerebellum. In cerebral cortex, concentrations were lowest in occipital cortex and highest in dorsolateral frontal cortex. Hippocampal concentrations were higher than those in cerebral cortex. Concentrations of GLI in cerebral cortex were significantly correlated with choline acetyltransferase activity and substance P immunoreactivity but not with concentrations of somatostatin or neuropeptide Y. Approximately half the GLI coeluted with porcine standards while half corresponded to a lower molecular weight species on gel permeation chromatography. With reverse phase high performance liquid chromatography (HPLC) the majority of the immunoreactivity eluted just in front of the porcine standard with a smaller amount coeluting with the porcine standard. These results show a widespread distribution of GLI in primate brain and are in accord with previous immunocytochemical studies.

Galanin-like immunoreactivity in hippocampal afferents in the rat, with special reference to cholinergic and noradrenergic inputs

The distribution of galanin-like immunoreactivity in the rat hippocampal formation (hippo campus and dentate gyrus) was studied and its origins were determined using various lesioning techniques. Special reference was made to the known cholinergic and noradrenergic hippocampal inputs from the septum-basal forebrain complex and locus coeruleus, both of which have previously been shown to co-contain galanin-like immunoreactivity at the cell body level. Galanin-immunoreactive fibers in the hippocampal formation were of at least three different morphological types: (1) Fine, slender, faintly immunoreactive fibers were seen throughout the hippocampal formation. (2) A strongly fluorescent varicose fiber population was observed mainly in the strata radiatum and oriens of the ventral CA3 region (3) A population of fine, faint puncta was seen within the granule and pyramidal cell layers throughout the hippocampal formation. Knife cut lesions of the dorsal afferent pathways resulted in almost complete disappearance of all fiber types, except for the ventral fine fibers. Lesions of the fimbria affected mainly the coarse and punctate fiber types, while lesions of the supracallosal striae depleted mainly the fine fibers. Cuts anterior and ventral to the hippocampal formation caused a decrease in ventral fine fibers. Furthermore, lesions of the dorsal bundle caused an almost complete disappearance of the fine fibers in all regions of the hippocampal formation. Neurotoxin lesions of the diagonal band/septal complex resulted in decreases in faintly immunoreactive puncta within the granule cell layer and adjacent fine fibers. It is concluded that most fine galanin-positive fibers originate in the lower brain stem, presumably the locus coeruleus, and appear to reach the hippocampal formation primarily through the supracallosal striae and the ventral route. The fimbria seems to contain a large proportion of the fibers giving rise to the coarse strongly fluorescent innervation, which appears to originate rostral to the pons. The galanin-immunoreactive fibers originating in cholinergic somata of the diagonal band, medial septal nuclei, previously shown to project to the hippocampal formation, seem to give rise to faintly labeled puncta within the granule and pyramidal cell layers, and to a small proportion of the fine fibers bordering the cell layers, as revealed by immunohistochemistry using our antibody. The immunohistochemical localization of a galanin-like peptide(s) in cholinergic and noradrenergic hippocampal afferents provides a morphological basis upon which to propose a peptidergic modulation of the action of these transmitters on hippocampal functions, e.g. memory and learning.

Distribution of galaninlike immunoreactivity in the rat central nervous system

The localization of galanin (GAL) immunoreactive (IR) neuronal structures in the rat central nervous system has been investigated by using the indirect immunofluorescence technique. GAL-IR structures were seen in high concentrations in the hypothalamus, medulla oblongata, and spinal cord. Less extensive systems were detected in the telencephalon, thalamus, mesencephalon, and pons, while virtually no GAL-positive structures were seen in the olfactory bulb and cerebellum. Major populations of cell bodies staining for GAL-like material were seen in many areas. In the telencephalon somata were revealed in the bed nucleus of stria terminalis, in the nucleus of the diagonal band, medial septum, and in the medial aspects of the central amygdaloid nucleus, and in small numbers in cortical areas. The anterodorsal and periventricular nuclei of the thalamus contained positive cell bodies. In the hypothalamus GAL-IR somata were seen in the medial and lateral preoptic nuclei, arcuate nucleus, periventricular nucleus, in the dorsomedial nucleus, in the medial forebrain bundle area, in the tubular, caudal, accessory, supraoptic, and paraventricular magnocellular nuclei and lateral to the mammillary recess. The dorsal raphe nucleus hosted a large number of GAL-positive somata. Locus coeruleus of the pons contained a large number of GAL-IR perikarya. In the medulla oblongata positive somata were found in the caudal spinal trigeminal nucleus, the nucleus of the solitary tract, and in the ventral lateral area just rostral to area postrema. Small cell bodies were detected in the superficial layers of the dorsal horn of the spinal cord at all levels and in lamina X at lumbar levels. Analysis of GAL-positive fibers in the telencephalon revealed highly or medium-dense networks in the lateral septal nucleus, in the bed nucleus of stria terminalis, and in the central and medial amygdaloid nuclei. Positive fibers were found in the thalamus in and around the periventricular nucleus as well as in the lateral habenular nucleus and extending in a lateral, caudal direction from the third ventricle and fasciculus retroflexus to the lateral tip of the medial lemniscus. In the hypothalamus the external layer of the median eminence contained a very dense fiber network. Dense or medium-dense GAL-IR networks were detected in the periventricular nucleus, throughout the medial and lateral preoptic areas, in the medial forebrain bundle area, in the dorsomedial nucleus, and lateral to the mammillary recess. In the pons GAL-IR fibers were seen in the parabrachial nuclei, dorsal to the superior olive, and in the periaqueductal central gray.(ABSTRACT TRUNCATED AT 400 WORDS)

Distribution of galanin-like immunoreactivity in the gastro-intestinal tract of several mammalian species.

The distribution of galanin-immunoreactive (GAL-IR) neurons was mapped in detail in the gastro-intestinal tract of the rat, mouse, guinea-pig and pig by use of the indirect immunofluorescence technique. GAL-IR cell bodies were found in both the submucous and the myenteric plexus, with considerably higher numbers in the former ganglia. The largest number of GAL-IR perikarya was seen in the duodenal submucous plexus of the pig. With some (single) exceptions, GAL-IR cell somata were not observed in the myenteric plexus of the pig and guinea-pig, and in the submucous plexus of the esophagus and the stomach of the guinea-pig. GAL-IR fibers occurred in most parts of the gastro-intestinal tract. In the lamina propria a few non-varicose, weakly fluorescent fibers were noted in the mouse and rat, whereas in the pig and guinea-pig were large numbers of GAL-IR fibers with a varicose appearance was observed. These fibers were in all species most numerous in the distal portion of the intestinal tract. In the submucosa GAL-IR fibers were detected in all four species, and in the pig and guinea-pig some fibers surrounded blood vessels. A large number of GAL-IR fibers was generally seen in the circular smooth muscle layer, except in the guinea-pig, which only seemed to contain a few fibers. In the longitudinal muscle layer only single fibers could be detected. However, the gastric fundus region of the pig contained a moderate number of fibers in the longitudinally and obliquely oriented layers. In general, in the rat, mouse and pig, the submucous and myenteric plexus contained moderate or large numbers of GAL-IR fibers. In the guinea-pig, no or only single fibers were observed in the plexus of the upper gastro-intestinal tract and the rectum, while moderate numbers were seen in the ileum and colon. Thin adjacent sections stained for vasoactive intestinal polypeptide (VIP) and GAL revealed the coexistence of these two peptides in cell bodies of the myenteric plexus in the pig duodenum and guinea-pig colon. In these two species the GAL- and VIP-nerve fiber networks also exhibited marked similarities. However, in the rat and mouse VIP- and GAL-distribution patterns were in general different. The present findings indicate the presence of yet another neuropeptide or peptide family in the gastro-intestinal tract of several rodents and the pig.
