Transforming growth factor (TGF)-β1 is a pro-fibrotic cytokine that affects extracellular matrix (ECM) deposition and fibroblast activity. 17β-Estradiol (E2), the dominant ovarian steroid during the follicular phase (FLP) of the estrous cycle, can also influence ECM remodeling and fibrosis, through prostaglandin (PG) synthesis. PGs have opposing roles in fibrosis, with PGE₂ showing anti-fibrotic effects and PGF₂α promoting fibrosis. Equine endometrosis, whose main pathological feature is fibrosis, is marked by chronic inflammation and ECM accumulation, and may involve mediators like TGF-β1, PGs, and E2. This study aimed to assess how TGF-β1, E2, and their combination affect PG synthase and receptors transcription (qPCR) and PG concentrations (ELISA) in equine endometrial explants during the FLP, after 24 and 48 h. Prostaglandin-endoperoxide synthase 2 (PTGS-2) mRNA was reduced with TGF-β1 and combination treatments at 24 h. Estradiol and combined treatments downregulated microsomal prostaglandin E synthase1 (PGES) mRNA at 24 h, while prostaglandin F synthase (PGFS) mRNA reduced with TGF-β1 at 24 h and with E2 at 48 h. The PGE₂ concentration was lower in TGF-β1 +E2 group than in controls and TGF-β1 alone at 48 h. In contrast, PGF₂α concentration increased with E2 at 24 h and TGF-β1 and TGF-β1 +E2 treatments at 48 h. Prostaglandin E receptor (EP)2 and 4 mRNA upregulated with the combination treatment, while prostaglandin F receptor (FP) mRNA decreased in all treated groups. These findings suggest that TGF-β1 and E2 interact to regulate PG pathways, with potential to drive fibrotic changes in the equine endometrium, by shifting the balance between anti- and pro-fibrotic mediators like PGE₂ and PGF₂α.

Raising of mitogenic and anti-apoptotic agents - such as insulin, insulin-like growth factor type 1, and estrogen - during obesity and diabetes mellitus (types 1 and 2) favors the endometrial cancer (EC) development. Metformin, commonly used for treating type 2 diabetes, and resveratrol, a natural polyphenol, can both decrease cancer cell proliferation by modulating the PI3K/Akt/mTOR pathway. We evaluate the effects of metformin and/or resveratrol in an in vitro model of human type 1 endometrioid EC. Ishikawa cells were treated with 0.1 to 50mM of metformin and/or 0.1 to 75μM of resveratrol from 24h to 72h. Analyses assessed cell viability, cytotoxicity, caspases activation, mitochondrial function, cellular death, cell cycle, and the PI3K/Akt/mTOR pathway gene expression. In-silico analysis was conducted using Cytoscape. Metformin induced mitochondrial swelling, caspase-mediated apoptosis, and cell cycle arrest. Resveratrol decreased mitochondrial mass, cytotoxicity, and induced cell cycle arrest. Combined treatment with the highest concentrations reduced mitochondrial activity, cytotoxicity, and caspase activation while maintaining apoptotic features and cell cycle arrest. Resveratrol attenuated the toxic effects of metformin but it could be inducing a caspase-independent cell death in co-treated cells. Although in-silico analysis suggested potential molecular targets and interconnected mechanisms, lower concentrations did not alter PI3K/Akt/mTOR gene expression.

Rational design of composite gels demands innovative strategies to enhance structural and functional properties. Dietary fibers (DFs) offer promising potential for reinforcing protein-polysaccharide networks, but their role in soy protein isolate (SPI)-chitosan (CS) binary systems is underexplored. A novel ternary composite gel from SPI, CS and DF was developed to investigate DF-mediated reinforcement mechanisms. The synergistic combination of microbial transglutaminase (MTGase) cross-linking and heat treatment significantly enhanced the water holding capacity (WHC) of SPI-CS-DF, achieving the highest 30.32% increase compared to single treatments. SPI-CS with 1 mg mL-1 citrus dietary fiber (CDF) under the combined treatment exhibited the highest WHC (40.82% increase versus SPI-CS). Furthermore, texture analysis revealed that the addition of 5 mg mL-1 apple dietary fiber (ADF) and 5 mg mL-1 CDF increased gel strength by 37.31% and 36.27%, respectively. However, the addition of oat dietary fiber (ODF) simultaneously reduced the gel strength and hardness. Fourier transform infrared spectroscopy, scanning electron microscopy and confocal laser scanning microscopy demonstrated that ADF and CDF promoted uniform protein networks with porous structures, whereas ODF disrupted matrix continuity. MTGase-treated gels showed higher amide I peak intensity, which exhibited stronger covalent cross-linking. Overall, DF type and concentration are critical to tailoring SPI-CS gel structure and performance. MTGase-heat treatment combined with appropriate DF addition offers an effective strategy to improve WHC and mechanical properties in composite gels. These results provide a theoretical foundation for designing high-performance gel systems with potential applications. © 2026 Society of Chemical Industry.

Campylobacter jejuni remains a major cause of foodborne illness worldwide, with poultry serving as the primary reservoir. In the absence of commercial vaccines or effective feed additives, probiotics and their byproducts (postbiotics) represent a promising and sustainable approach to reducing Campylobacter colonization in poultry. This study compared the efficacy of oral and cloacal administration of probiotic lactobacilli and their postbiotics in reducing Campylobacter colonization and modulating the cecal microbiome in broiler chickens. Day-old chicks were assigned to seven treatment groups that received either probiotics (live cells of four poultry-derived Lactobacillus strains: L. reuteri P43, L. acidophilus P42, L. animalis P38, and L. crispatus C25) or postbiotics (Lactobacillus supernatants) or their combination (whole cultures) orally or intracloacally, with a non-treated group serving as a control. Chickens were challenged with C. jejuni strain 81-176 at the second week of age, and cecal contents were collected at the fifth week for Campylobacter enumeration and microbiome profiling. The results revealed that both oral and cloacal administration of Lactobacillus cells significantly reduced Campylobacter cecal loads by 0.34 and 0.78 log₁₀, respectively, compared to the control. Significant differences in microbial richness and evenness were observed among treatment groups, with groups administered orally with probiotics, postbiotics, or their combination consistently showing higher alpha diversity indices than controls. NMDS ordination confirmed distinct community clustering among the treatment groups. Differential abundance analysis (MaAsLin2) further revealed that Ruminococcus was significantly enriched in the group receiving intracloacal postbiotic treatment, whereas the genus unclassified Firmicutes was more abundant in the group that received the combined probiotic-postbiotic treatment orally. Opportunistic genera, such as Escherichia-Shigella and Faecalicoccus, were significantly higher in the control group compared to all treated groups. Overall, while probiotics and postbiotics, whether given alone or together, modulated the gut microbial composition in Campylobacter-infected broilers, the administration of probiotic cells offered additional benefits by reducing Campylobacter colonization.
