Exploring product maturation, microbial communities and antibiotic resistance gene abundances during food waste and cattle manure co-composting

This study proposed combining food waste (FW) and cattle manure (CM) in composting to improve the product maturity. The findings suggested that the inclusion of CM effectively extended the thermophilic stage, facilitated the decomposition of cellulose, and enhanced the production of humus-like substances by enhancing beneficial microbial cooperation. Adding 40 % CW was optimal to reduce the nitrogen loss, increase the cellulose degradation rate to 22.07 %, increase germination index (GI) to 140 %, and reduce normalized antibiotic resistance gene (ARG) abundances. Adding CW could promote the transformation of protein-like compounds, thereby enhancing the humification process of organic substances. Structural equation modeling further verified that the temperature was the key factor affecting humification production, while the main driver for ARGs was physiochemical parameters. This study shows that co-composting of FW and CM offers the potential to promote humification, reduce ARG abundance, and improve fertilizer quality for utilization of both biowastes in the field.

Unlocking the potential of oat straw: Efficient pretreatment methods for enhanced glucose production

The excellent physical and chemical properties of deep eutectic solvents, especially the ability of some of them to dissolve biomass, make them broadly applicable in biomass pretreatment. In this study, oat straw was pretreated with deep eutectic solvents composed of choline chloride and an acid (formic, lactic, or oxalic acid). The highest reducing sugar yield was obtained for the formic acid/choline chloride mixture. Using a pretreatment temperature of 110 °C, a reaction time of 2 h, and a solid-liquid ratio of 1:20, the reducing sugar yield obtained by cellulase hydrolysis was 23.5%, the degradation rate of cellulose reached 76.9%, and hemicellulose was completely degraded. The pretreated oat straw was then analyzed by scanning electron microscopy, Fourier transform infrared spectroscopy, and X-ray diffraction. The changes in its crystallinity and lignin content compared to the untreated specimen were determined. A preliminary mechanism for the pretreatment of oat straw with formic acid/choline chloride was revealed. The study could provide an opportunity to expand the application in biochemicals or biological feed processes.

Removal efficiency and abundance of nitrogen cycling microorganism in three bio-matrix materials to treat swine wastewater.

A bio-matrix material (BMM) system is used to pretreat swine wastewater and reduce the nitrogen (N) concentration to the tolerance range of plants in constructed wetlands. In this study, rice straw (RS), wheat straw (WS), and corn stalk (CS) were applied to treat pollutants from swine wastewater, respectively. This one year-long field experiment make up for the lack of long-term experiments and mechanistic investigations of BMM. The pollutant removal efficiency, degradation process of crop straw, and the abundance of nitrogen cycling genes were determined in different BMM systems. The results showed that the removal efficiency of COD, TN, NH4+, and NO3- was the best in the initial 6months. Furthermore, RS and WS exhibited favorable annual removal efficiency of TN and NH4+, which were 32.81% and 32.99%, 35.3% and 34.97%, respectively. Moreover, the removal efficiency of COD was 30.81% in three BMM systems. Meanwhile, it was found that the dry matter (DM) degradation of crop straws was fast in the first 4-5months. The degradation rates of cellulose, hemicellulose, and lignin were 94.19%, 94.36%, and 87.32%, respectively, in 1year. The abundance of nitrogen cycling genes significantly increased by adding BMM, compared with CK (P < 0.05). This showed the abundance of the hzsB gene in RS was the highest, while nirK, nirS, AOA, and AOB were the highest in WS. The addition of RS and WS was better than that of CS in promoting the abundance of nitrogen cycling microorganisms. The results indicated that adding BMM could enhance the anaerobic ammonia oxidation, nitrification, and denitrification. This study not only extends our comprehension of BMM mechanisms in swine wastewater treatment but also serves as a guiding light for numerous farms in similar climate regions.

Construction of a Synthetic Microbial Community for Enzymatic Pretreatment of Wheat Straw for Biogas Production via Anaerobic Digestion.

Biological pretreatment is a viable method for enhancing biogas production from straw crops, with the improvement in lignocellulose degradation efficiency being a crucial factor in this process. Herein, a metagenomic approach was used to screen core microorganisms (Bacillus subtilis, Acinetobacter johnsonii, Trichoderma viride, and Aspergillus niger) possessing lignocellulose-degrading abilities among samples from three environments: pile retting wheat straw (WS), WS returned to soil, and forest soil. Subsequently, synthetic microbial communities were constructed for fermentation-enzyme production. The crude enzyme solution obtained was used to pretreat WS and was compared with two commercial enzymes. The synthetic microbial community enzyme-producing pretreatment (SMCEP) yielded the highest enzymatic digestion efficacy for WS, yielding cellulose, hemicellulose, and lignin degradation rates of 39.85, 36.99, and 19.21%, respectively. Furthermore, pretreatment of WS with an enzyme solution, followed by anaerobic digestion achieved satisfactory results. SMCEP displayed the highest cumulative biogas production at 801.16 mL/g TS, which was 38.79% higher than that observed for WS, 22.15% higher than that of solid-state commercial enzyme pretreatment and 25.41% higher than that of liquid commercial enzyme pretreatment. These results indicate that enzyme-pretreated WS can significantly enhance biogas production. This study represents a solution to the environmental burden and energy use of crop residues.

Synergistic effect of two bacterial strains promoting anaerobic digestion of rice straw to produce methane

A large amount of agricultural waste causes global environmental pollution. Biogas production by microbial pretreatment is an important way to utilize agricultural waste resources. In this study, Sporocytophaga CG-1 (A, cellulolytic strain) was co-cultured with Bacillus clausii HP-1 (B, non-cellulolytic strain) to analyze the effect of pretreatment of rice straw on methanogenic capacity of anaerobic digestion (AD). The results showed that weight loss rate of filter paper of co-culture combination is 53.38%, which is 29.37% higher than that of A. The synergistic effect of B on A can promote its degradation of cellulose. The cumulative methane production rate of the co-culture combination was the highest (93.04 mL/g VS substrate), which was significantly higher than that of A, B and the control group (82.38, 67.28 and 67.70 mL/g VS substrate). Auxiliary bacteria can improve cellulose degradation rate by promoting secondary product metabolism. These results provide data support for the application of co-culture strategies in the field of anaerobic digestion practices.

The impact of microbial inoculants on large-scale composting of straw and manure under natural low-temperature conditions

Understanding large-scale composting under natural conditions is essential for improving waste management and promoting sustainable agriculture. In this study, corn straw (400 tons) and pig manure (200 tons) were composted with microbial inoculants. The thermophilic phase of composting lasted for fourteen weeks, resulting in an alkaline final product. Microbial systems with low-temperature initiation and high-temperature fermentation played a crucial role in enhancing lignocellulose degradation and humic substances (HS) formation. Adding microbes, including Rhodanobacter, Pseudomonas, and Planococcus, showed a positive correlation with degradation rates of cellulose, hemicellulose, and lignin. Bacillus, Planococcus, and Acinetobacter were positively correlated with HS formation. Microorganisms facilitated efficient hydrolysis of lignocelluloses, providing humic precursors to accelerate composting humification through phenolic protein and Maillard pathways. This study provides significant insights into large-scale composting under natural conditions, contributing to the advancement of waste management strategies and the promotion of sustainable agriculture.

Fully Biodegradable Packaging Films for Fresh Food Storage Based on Oil-Infused Bacterial Cellulose.

Fully biodegradable packaging materials are demanded to resolve the issue of plastic pollution. However, the fresh food storage performance of biodegradable materials is generally much lower than that of plastics due to their high permeability, microbial friendliness, and limited stretchability and transparency. Here a biodegradable packaging material is reported with high fresh food storage performance based on an oil-infused bacterial cellulose (OBC) porous film. The oil infusion significantly improved cellulose's food-keeping performance by reducing its gas permeability, increasing its stretchability and transparency, and enabling the active release of green vapor-phase preservative molecules, while maintaining its intrinsically high degradability. Strawberries stored in a container with the OBC lid at 23°C after 5 days exhibited a moldy rate of 0%, in contrast to the 100% moldy rate of those stored by poly(ethylene). Enhanced storage performance is also obtained on tomatoes, pork, and shrimp. The OBC film is naturally degraded after being buried in wet soil at 30°C for 9 days, identical to the degradation rate of bacterial cellulose. The liquid seal strategy broadly applies to different celluloses, providing a general option for developing cellulose-based biodegradable packaging materials.

Development of functional consortia for the pretreatment of compostable lignocellulosic waste: A simple and effective solution to a large-scale problem

Microorganisms drive the degradation of organic matter thanks to their enzymatic versatility. However, the structure of lignocellulose poses a great challenge for the microbiota inhabiting a compost pile. Our purpose was to increase the biodegradability of vegetable waste in the early stages of the composting process by applying a microbial consortium with lignocelllulolytic capacity. For this, a previous screening was performed among the culturable microbiota from different composting processes to find inoculants with ligninocellulolytic activity. Selected strains were applied as a pure culture and as a microbial consortium. The starting material was composed of tomato plant and pruning remains mixed in a ratio (50:50 v/v), whose humidity was adjusted to around 65%. To determine the ability of both treatments to activate the biodegradation of the mixtures, moisture, organic matter, ash, C/N ratio, 4-day cumulative respirometric index (AT4) and degradation rates of cellulose, hemicellulose and lignin were evaluated. Subsequently, a real composting process was developed in which the performance of the microbial consortium was compared with the composting process without inoculum (control). According to our tests, three microbial strains (Bacillus safensis, Bacillus licheniformis and Fusarium oxysporum) were selected. The results showed that the application of the bacteria strains at low doses (104 CFU g−1 on the complete residual material of the pile) resulted in higher rates of lignocelullose degradation after 10 days of treatment compared to that observed after application of the fungus in pure culture or untreated controls. The implementation of the strategy described in this work resulted in obtaining compost with better agronomic quality than the uninoculated controls. Therefore, the application of this consortium could be considered as an interesting tool for bioactivation of lignocellulosic waste prior to the composting process.

Microbial mechanisms involved in negative effects of amoxicillin and copper on humification during composting of dairy cattle manure

Livestock manure often contains various pollutants. The aim of this study was to investigate how adding amoxicillin (AMX), Cu, and both AMX and Cu (ACu) affected humification during composting and the microbial mechanisms involved. The cellulose degradation rates were 16.96%, 10.86%, and 9.01% lower, the humic acid contents were 18.71%, 12.89%, and 16.78% lower, and the humification degrees were 24.72%, 24.16%, and 15.73% lower for the AMX, Cu, and ACu treatments, respectively, than the control. Adding AMX and Cu separately or together inhibited humic acid formation and decreased the degree of humification, but the degree of humification was decreased less by ACu than by AMX or Cu separately. The ACu treatment decreased the number of core bacteria involved in humic acid formation and decreased carbohydrate and amino acid metabolism during the maturing period, and thereby delayed humic acid formation and humification. The results support composting manure containing AMX and Cu.

Comparative Transcriptome Analysis Explores the Mechanism of Angiosperm and Gymnosperm Deadwood Degradation by Fomes fomentarius.

In forest ecosystems, most of the soil organic matter is derived from trees, as deadwood lignocellulose and wood-decaying basidiomycetes are the most important decomposers of lignin and cellulose. Fomes fomentarius is one of the most common white-rot fungi colonizing angiosperm trees and is often found in birch deadwood but seldom in pine deadwood. To reveal the mechanism through which F. fomentarius selects angiosperms as its preferred host trees, birch and pinewood sticks were selected for culturing for two months. The weight loss, cellulose and lignin degradation rates, activities of degrading enzymes, and transcriptome analyses of two degradation models were compared and analyzed. The results showed that F. fomentarius-degraded birchwood with higher efficiency than pinewood. A GO enrichment analysis found that more upregulated genes related to the top 30 terms showed a molecular function related to degradation, and most genes belonged to the CAZymes family in F. fomentarius-degraded birchwood. However, pinewood degradation did not show these phenomena. A KEGG pathway analysis also indicated that, for the same pathway, more upregulated genes were involved in birchwood degradation caused by F. fomentarius than in pinewood degradation.

Effect of in-situ mineralization of calcium carbonate on the aging resistance of Wikstroemia bast fiber

Abstract Paper cultural relics, as the carriers of human civilization, inevitably deteriorate during the long-term storage. The restoration of those damaged or aged paper cultural relics is an important part of cultural relics protection. How to obtain the fiber with high permanence property is the key to the repair paper used to cultural relic restoration. Herein, in order to obtain high permanence lignocellulosic fiber, nano-CaCO3 was introduced inside the Wikstroemia bast fiber by in-situ mineralization. Under accelerated aging condition, the microstructure, chemical structure, crystallinity, degree of polymerization, variation of pH, alkali reserve and mechanical strength of the fiber before and after mineralization were studied. The results showed that nano-CaCO3 had no obvious effect on the properties of paper prepared from those mineralized fibers, but it can effectively neutralize the acid generated inside the fiber, slow down the degradation rate of cellulose and improve the permanence of the paper during the aging process. It provides a new insight for the manufacture of repair paper for cultural relics restoration.

Responses of microorganisms to different wavelengths of light radiation during green waste composting

Light radiation can degrade recalcitrant materials like lignocelluloses in litter and serve as a physical condition to accelerate green waste (GW) decomposition, but few studies have considered the microbial effects of light wavelength on GW composting. This study innovatively investigated the effects of different wavelengths of light radiation, including full-spectrum, no blue light, no UV, no UV-A, no UV-B, and dark conditions, on accelerating the GW composting process. Especially, the study explored the dynamic changes in the degradation of lignocelluloses and evaluated the responses of microorganisms throughout the composting process under different light radiation wavelengths. No blue light (where radiation between 400 and 500 nm was blocked by the film) yielded the highest-quality compost within 40 days. In comparison to the dark (control), no blue light exhibited an elevated composting temperature (56.7 °C), an extended thermophilic phase (6 days), and increased degradation rates of lignin, cellulose, and hemicellulose by 13 %, 15 %, and 12 %, respectively. This study revealed that during the composting mesophilic phase, bacterial diversity performed best under no blue light, while fungal diversity excelled under full-spectrum. In the thermophilic phase, microbial diversity exhibited optimal performance under full-spectrum. During the cooling phase, bacterial diversity was highest under no blue light, and fungal diversity excelled under no UV-A. During the mesophilic and cooling phases, the bacterial ACE index for no blue light exceeded that of the other light radiation wavelengths, with values of 418 and 494, respectively. Under no blue light, the Shannon index of microorganisms remained within the range of 2.0–4.8, demonstrating superior performance. Meanwhile, the relative abundances of lignin-degrading microorganisms (Flavobacterium, Acaulium, and Acremoniu) under no blue light has increased, demonstrating improved microbial community structures. Therefore, no blue light radiation offered a novel approach to expedite GW composting.

Cellulase with Bacillus velezensis improves physicochemical characteristics, microbiota and metabolites of corn germ meal during two-stage co-fermentation.

Corn germ meal (CGM) is one of the major byproducts of corn starch extraction. Although CGM has rich fiber content, it lacks good protein content and amino acid balance, and therefore cannot be fully utilized as animal feed. In this study, we investigated the processing effect of cellulase synergized with Bacillus velezensis on the nutritional value of pretreated CGM (PCGM) in two-stage solid-state fermentation (SSF). High-throughput sequencing technology was used to explore the dynamic changes in microbial diversity. The results showed that compared with four combinations of B. velezensis + Lactiplantibacillus plantarum (PCGM-BL), cellulase + L. plantarum (PCGM-CL),control group (PCGM-CK), and cellulase + B. velezensis + L. plantarum (PCGM-BCL), the fourth combination of PCGM-BCL significantly improved the nutritional characteristics of PCGM. After two-stage SSF (48h), viable bacterial count and contents of crude protein (CP) and trichloroacetic acid-soluble protein (TCA-SP) all were increased in PCGM-BCL (p < 0.05), while the pH was reduced to 4.38 ± 0.02. In addition, compared with PCGM-BL, the cellulose degradation rate increased from 5.02 to 50.74%, increasing the amounts of short-chain fatty acids (216.61 ± 2.74 to 1727.55 ± 23.00µg/g) and total amino acids (18.60 to 21.02%) in PCGM-BCL. Furthermore, high-throughput sequencing analysis revealed significant dynamic changes in microbial diversity. In the first stage of PCGM-BCL fermentation, Bacillus was the dominant genus (99.87%), which after 24h of anaerobic fermentation changed to lactobacillus (37.45%). Kyoto Encylopaedia of Genes and Genomes (KEGG) metabolic pathway analysis revealed that the pathways related to the metabolism of carbohydrates, amino acids, cofactors, and vitamins accounted for more than 10% of the enriched pathways throughout the fermentation period. Concisely, we show that cellulase can effectively improve the nutritional value of PCGM when synergized with B. velezensis in two-stage SSF.

The Impact of Marasmius tricolor 310b on the Degradation of Cellulose in Rapeseed Straw Composting

Composting, a vital process for organic waste management, hinges on intricate microbial dynamics. The role of specific microbial inoculants, such as fungi, in shaping these dynamics remains an area of keen interest. This study aimed to elucidate the impact of the fungal inoculant Marasmius tricolor 310b on composting dynamics, with a focus on temperature variations and the degradation of lignocellulosic components. We conducted a comprehensive composting experiment with four treatments: pure rapeseed straw, rapeseed straw with Marasmius tricolor 310b, a combination of rapeseed straw and pig manure, and the combination with Marasmius tricolor 310b. Temperature dynamics, lignocellulosic degradation rates, and fungal community structures were meticulously analyzed. While Marasmius tricolor 310b did not significantly alter the temperature profile, its presence markedly enhanced the degradation rates of cellulose and lignin. With the introduction of Marasmius tricolor 310b, the degradation rate of cellulose in the rapeseed straw compost increased from 0.0319 (ST) to 0.0419 (STM). In the combined rapeseed straw and pig manure compost, this rate rose from 0.0422 (STPM) to 0.045 (STPMM). For hemicellulose, the degradation rate in the rapeseed straw compost increased from 0.0103 (ST) to 0.014 (STM). However, in the combined compost, it slightly decreased from 0.0146 (STPM) to 0.013 (STPMM). As for lignin, its degradation rate in the rapeseed straw compost saw an increase from 0.0024 (ST) to 0.00366 (STM). In the combined compost, the rate rose from 0.00302 (STPM) to 0.00402 (STPMM). Fungal community analyses revealed discernible shifts in structure and diversity upon the fungus’s introduction. Network analyses further highlighted a highly interconnected microbial community, suggesting its robustness against perturbations. Temporal dynamics underscored the intricate interplay of microbial interactions, substrate availability, and external inoculants. The introduction of Marasmius tricolor 310b profoundly influences composting dynamics, offering valuable insights into microbial community interactions and presenting potential avenues for optimizing composting strategies.

Characterization of Cellulose-Degrading Bacteria Isolated from Silkworm Excrement and Optimization of Its Cellulase Production.

An abundance of refractory cellulose is the key limiting factor restricting the resource utilization efficiency of silkworm (Bombyx mori) excrement via composting. Screening for cellulose-degrading bacteria is likely to provide high-quality strains for the safe and rapid decomposition of silkworm excrement. In this study, bacteria capable of degrading cellulose with a high efficiency were isolated from silkworm excrement and the conditions for cellulase production were optimized. The strains were preliminarily screened via sodium carboxymethyl cellulose culture and staining with Congo red, rescreened via a filter paper enzyme activity test, and identified via morphological observation, physiological and biochemical tests, and phylogenetic analysis of the 16S rDNA sequence. Enzyme activity assay was performed using the 3,5-dinitrosalicylic acid method. DC-11, a highly cellulolytic strain, was identified as Bacillus subtilis. The optimum temperature and pH of this strain were 55 °C and 6, respectively, and the filter paper enzyme activity (FPase), endoglucanase activity (CMCase), and exoglucanase activity (CXase) reached 15.40 U/mL, 11.91 U/mL, and 20.61 U/mL. In addition, the cellulose degradation rate of the treatment group treated with DC-11 was 39.57% in the bioaugmentation test, which was significantly higher than that of the control group without DC-11 (10.01%). Strain DC-11 was shown to be an acid-resistant and heat-resistant cellulose-degrading strain, with high cellulase activity. This strain can exert a bioaugmentation effect on cellulose degradation and has the potential for use in preparing microbial inocula that can be applied for the safe and rapid composting of silkworm excrement.

Effect of Thermophilic Microbial Agents on Crude Fiber Content, Carbohydrate-Active Enzyme Genes, and Microbial Communities during Chinese Medicine Residue Composting.

The objective of this study was to investigate the impact of thermophilic bacteria on crude fiber content, carbohydrate-active enzyme (CAZyme) genes, and associated microbial communities during Chinese medicine residues composting. The study examines changes over 15 days of composting with (T) and without (CK) thermophilic microbial agents. Results show that the group T compost temperature reached a maximum of 71.0 °C and remained above 70 °C for 2 days, while the group CK maximum temperature was only 60.9 °C. On Day 15, the seed germination index (GI) of group T reached 98.7%, while the group CK GI was only 56.7%. After composting, the degradation rates of cellulose, hemicellulose, and lignin in group T increased by 5.1, 22.5, and 18.5%, respectively, compared to those in group CK. Thermophilic microbial agents changed the microbial communities related to CAZymes, increasing unclassified_o_Myxococcales and Sphaerobacter abundance and reducing Acinetobacter and Sphingobacterium abundance. Thermophilic microbial agents also increased the abundance of the GT4, GT2_Glycos_transf_2, and AA3 gene families. These results show that thermophilic microbial agents can increase composting temperature, accelerate compost maturation, and promote crude fiber degradation. Therefore, they have broad application potential.

Novelty three stages for humification of sewage sludge during hyperthermophilic aerobic fermentation

Compared with conventional aerobic fermentation (CAF), there is limited knowledge of how hyperthermophilic aerobic fermentation (HAF) enhances the humification of sewage sludge. This study compared three novel stages of organic degradation, precursors, functional groups, bacterial community, and humus synthesis mechanism in HAF with CAF. The results showed that organic matter (OM) degraded rapidly, and 68% of the degradation could be completed of stage I in HAF. Compared with the initial stage, ammonium nitrogen (NH4+-N), water-soluble organic carbon, and water-soluble total nitrogen increased by 2.83 times, 40.5 times, and 33.5 times, respectively. Cellulose and hemicellulose decreased by 29.22% and 21.85%, respectively. These results suggested that temperature (>80 °C) and Bacillus dominated accelerate the humification process by rapidly improving OM degradation. Compared with the initial value of HAF, the maximum increment of reducing sugar at stage II was 297%, and the degradation rate of cellulose was effectively increased by 21.03% compared with that of CAF. The precursors such as reducing sugars and amino acids formed humus at stage II. The content of Aryl C increased significantly during the HAF process, the degree of polymerization of humus and the aromatization degree of HA and FA increased significantly, and complex organic macromolecular material polymers were formed at stage III. The sugar-amine condensation was the mechanism of humification in the sludge HAF process. This investigation provided three new stages of insights into the synthesis of humification during the HAF process and extended the current mechanism of humification in the HAF process.

WHITE-ROT FUNGAL PRETREATMENT OF WHEAT STRAW: EFFECT ON ENZYMATIC HYDROLYSIS OF CARBOHYDRATE POLYMERS

The aim of the study was the comparative analysis of degradation of wheat straw lignin by white-rot fungi and its implications on the efficiency of enzymatic hydrolysis of holocellulose. Peroxidases were found to be predominantly responsible for lignin degradation, even though high laccase activities were detected, especially in the initial stages of fungal culturing. The studied fungal species showed various ability to degrade lignin in wheat straw, which further affected the release of reducing sugars during enzymatic saccharification. The highest rate of lignin, hemicelluloses and cellulose degradation was noticed in the sample pretreated with Irpex lacteus. Among all the tested species, only Ganoderma resinaceum was found as a suitable lignin degrader, with 2-fold higher hydrolysis yield (51.1 ± 4.7%) than in the control. A key mechanism that enhances convertibility of carbohydrates is the selective lignin removal from biomass. Operating time, holocellulose loss and unpredictable fungal response to culturing conditions are the main challenges in fungal pretreatment of lignocellulosic feedstock.

Construction of a synthetic microbial community based on multiomics linkage technology and analysis of the mechanism of lignocellulose degradation

The efficient degradation of lignocellulose is a bottleneck for its integrated utilization. This research performed species analysis and made functional predictions in various ecosystems using multiomics coupling to construct a core synthetic microbial community with efficient lignocellulose degradation function. The synthetic microbial community was employed to degrade corn straw via solid-state fermentation. The degradation mechanisms were resolved using proteomics. The optimum culture conditions included 10% inoculum level (w/v), 4% nitrogen source ratio and a fermentation time of 23 d. Under these conditions, the degradation rates of cellulose, hemicellulose, and lignin were 34.91%, 45.94%, and 23.34%, respectively. Proteomic analysis revealed that lignin 1,4-β-xylanase, β-xylosidase and endo-1,4-β-xylanase were closely related to lignocellulose degradation. The metabolic pathways involved in lignocellulose degradation and the functional roles of eight strains were obtained. The synthesis of a microbial community via multiomics linkage technology can effectively decompose lignocellulose, which is useful for their further utilization.

Gut microbial communities and their potential roles in cellulose digestion and thermal adaptation of earthworms

Adaptations to temperature and food resources, which can be affected by gut microbiota, are two main adaptive strategies allowing soil fauna to survive in their habitats, especially for cold-blooded animals. Earthworms are often referred to as ecosystem engineers because they make up the biggest component of the animal biomass found in the soil. They are considered as an important indicator in the triangle of soil quality, health and functions. However, the roles of gut microbiota in the environmental adaptation of earthworms at a large scale remain obscure. We explored the gut bacterial communities and their functions in the environmental adaptation of two widespread earthworm species (Eisenia nordenskioldi Eisen and Drawida ghilarovi Gates) in Northeast China (1661 km). Based on our findings, the alpha diversity of gut bacterial communities decreased with the increase of latitude, and the gut bacterial community composition was shaped by both mean annual temperature (MAT) and cellulose. Actinobacteria, Proteobacteria, Firmicutes, and Planctomycetes, recognized as the predominant cellulose degraders, were keystone taxa driving gut bacterial interactions. Actinobacteria, Firmicutes, and Planctomycetes were influenced by MAT and cellulose, and had higher contributions to gut total cellulase activity. The optimal temperature for total cellulase in the gut of E. nordenskioldi (25-30 °C) was lower than that of D ghilarovi (40 °C). The gut microbiota-deleted earthworms had the lowest cellulose degradation rate (1.07 %). The cellulose was degraded faster by gut bacteria from the host they were derived, indicating the presence of home field advantage of cellulose decomposition. This study provides a foundation for understanding the biotic strategies adopted by earthworms when they enter a new habitat, with gut microbiota being central to food digestion and environmental adaptability.
