Mental abacus training affects high-level executive functions: Comparison of activation of the frontal pole

The role of executive function training in supporting child development has been increasingly studied. Executive function is largely related to the prefrontal cortex. The anterior portion of the prefrontal cortex, which is area 10 on the Brodmann map, is essential for the emergence of higher-order executive functions. Accumulating evidence indicates that mental abacus training, which is closely related to mathematics education, activates the prefrontal cortex. Based on these findings, it can be hypothesized that the mental abacus is valuable for training more advanced functions. Therefore, this study analyzed the activation of children’s brains with a focus on the frontal pole (Brodmann area 10). The results illustrated that mental abacus task more strongly activated the brain than piano task, the marshmallow test, or letter–number sequencing tasks. Thus, it was suggested that the mental abacus is valuable for training higher-level executive functions (i.e., frontal pole).

Cerebral blood flow-single-photon emission computed tomography and Brodmann mapping may facilitate the evaluation of clinical features of early-onset Alzheimer’s disease

A relatively large numbers of studies have reported on the clinical features, classifications, and impairment of cerebral blood flow (CBF) in cases with Alzheimer’s disease. However, only few reports have investigated the relationships between clinical tests and lesion areas in the brain with significantly reduced CBF, assessed using single-photon emission computed tomography (SPECT). This study aimed to assess the correlations between impaired Mini-Mental State Examination (MMSE) scores and reduced CBF using Brodmann area mapping and classify early-onset Alzheimer’s disease (EOAD). Thirty-one patients aged <65 years, with memory impairment, were examined using CBF-SPECT, and MMSE during the same period. Twenty patients were diagnosed with EOAD. We divided the patients into two groups: one with bilateral reduction of CBF in temporo-parietal region and the other with unilateral reduction. The bilateral group had significantly lower MMSE scores than the unilateral reduction group, but it had significantly less family history, significantly poor prognoses, and ten significantly reduced CBF in the Brodmann area (BA) with SPECT. In the bilateral group, there were large correlations between impaired MMSE and reduced CBF regions in the BA. These findings confirm that the classification of EOAD may pave the way to understand patients with EOAD.

A network approach to the topological organization of the Brodmann map.

Brain morphology is the result of functional factors associated with cortical areas, but it is also influenced by structural aspects due to physical and spatial constraints. Despite the noticeable advances in brain mapping, Brodmann's map is still used in many research fields that rely on macroscopic cortical features for practical or theoretical issues. Here, the topological relationships among the Brodmann areas were modeled according to the principles of network analysis, in order to provide a synthetic view of their spatial properties following a criterion of contiguity. The model evidences the importance of the parieto-temporal region in terms of biological burden and topological complexity. The retrosplenial region is particularly influenced by spatial constraints, and the cingulate cortex occupies a position that bridges the anterior and posterior topological blocks. Such spatial framework should be taken into account when dealing with brain morphology in both ontogeny and phylogeny.

The inferior frontal sulcus: Cortical segregation, molecular architecture and function

The inferior frontal sulcus is conceptualized as the landmark delineating ventro-from dorsolateral prefrontal cortex. Functional imaging studies report activations within the sulcus during tasks addressing cognitive control and verbal working memory, while their microstructural correlates are not well defined. Existing microstructural maps, e.g., Brodmann's map, do not distinguish separate areas within the sulcus. We identified six new areas in the inferior frontal sulcus and its junction to the precentral sulcus, ifs1-4, ifj1-ifj2, by combined cytoarchitectonic analysis and receptor autoradiography. A hierarchical cluster analysis of receptor densities of these and neighbouring prefrontal areas revealed that they form a distinct cluster within the prefrontal cortex. Major interhemispheric differences were found in both cyto- and receptorarchitecture. The function of cytoarchitectonically identified areas was explored by comparing probabilistic maps of the areas in stereotaxic space with their functions and co-activation patterns as analysed by means of a coordinate-based meta-analysis. We found a bilateral involvement in working memory, as well as a lateralization of different language-related processes to the left hemisphere, and of music processing and attention to the right-hemispheric areas. Particularly ifj2 might act as a functional hub between the networks. The cytoarchitectonic maps and receptor densities provide a powerful tool to further elucidate the function of these areas. The maps are available through the Human Brain Atlas of the Human Brain Project and serve in combination with the information on the cyto- and receptor architecture of the areas as a resource for brain models and simulations.

Mapping the Human Brain: What Is the Next Frontier?

Abstract The structure and function of the human brain have long been the topic of exploration by scientists. To understand how the brain works, researchers have created various brain navigation maps, i.e. brain atlases, which are still being developed and improved. To clarify the trend of human brain mapping and to achieve better applications in brain science and diseases, this commentary briefly traces the history of human brain mapping and describes insights into the next stage of building brain atlases.

A detailed comparison of the cytoarchitectonic and myeloarchitectonic maps of the human neocortex produced by the Vogt-Vogt school.

The comprehensive research programme of the Vogt-Vogt (V-V) school, which was active during the period 1900-1970, included detailed cytoarchitectonic and myeloarchitectonic analyses of the human cerebral cortex, with the aim to integrate the data obtained into a map, showing a parcellation of the human cerebral cortex into fundamental structural and potentially functional units. The cytoarchitectonic V-V analyses yielded two maps of the human cerebral cortex, the famous map of Brodmann (Vergleichende Lokalisationslehre der Grosshirnrinde in ihren Prinzipien dargestellt auf Grund des Zellenbaues. Barth, Leipzig, 1909), Brodmann (in: Bruns P (ed) Neue deutsche Chirurgie, Enke, Stuttgart, 1914), and the less known, but more detailed map of Sarkisov et al. (Cytoarchitecture of the human cortex cerebri. Medgiz, Moscow, 1949). Sarkisov et al. used in their cytoarchitectonic parcellation of the cortex the same numbering scheme as Brodmann. They confirmed the presence of most of the areas delineated by the latter, but they subdivided several of these areas into two or more separate areas or subareas. Within the realm of the myeloarchitectonic V-V analyses, numerous meticulous studies of the cortex of individual cerebral lobes were carried out, but these were not united into a single map. Consequently, the envisioned integration of cytoarchitectonic and myeloarchitectonic data mentioned above was never realized. Some years ago, we (Nieuwenhuys et al. in Brain Struct Funct 220:2551-2573, 2015a, Nieuwenhuys et al. in Brain Struct Funct 220:3753-3755, 2015b) reanalyzed the V-V myeloarchitectonic data, and succeeded in constructing a complete myeloarchitectonic map of the human neocortex from these data. Because the data provided by the V-V school were derived from many different brains, a standard brain had to be introduced as a template to which all data available could be transferred. As such the MNI305 template was selected. Having made available now the cytoarchitectonic maps of Brodmann and Sarkisov et al. and the recently prepared myeloarchitectonic map, an attempt is made here to realize at last the original aim of the V-V school, viz. the preparation of a single, combined (cyto + myelo) architectonic map of the human cortex. To this end, the following three steps have been made. First, Brodmann's (BR) map, and the map of Sarkisov et al. (SA) were harmoniously transferred to the same template brain as the one used during the construction of our myeloarchitectonic map. Second, the standardized BR and our myeloarchitectonic (NI) map were compared, and the data contained within these maps were integrated into a single standardized combined BR-NI map (Fig.11). The standardized SA and NI maps were subjected to the same procedure (Fig.12). Finally, the standardized combined BR-NI and SA-NI maps were united into a single combined BR-SA-NI map (Fig.13). This map renders it possible to make direct comparisons between the results of the architectonic studies of the V-V school and current parcellations of the human neocortex.

Brodmann's map is one of the best known maps of the human cerebral cortex. However, Korbinian Brodmann's 1909 monograph, containing the first map, has not been widely read by neurologists. Following the publication of the first map, Brodmann reproduced the map in 1910, and again in 1914, with several important differences. The latter map is also characterized by the exclusion of Brodmann area numbers, "12-16, and 48-51;" contrastingry numbers "1-52" have been described in the animal brain. Here, we provide a detailed explanation of the versions of Brodmann's maps, and review his academic profile and works.

Body Topography Parcellates Human Sensory and Motor Cortex.

The cytoarchitectonic map as proposed by Brodmann currently dominates models of human sensorimotor cortical structure, function, and plasticity. According to this model, primary motor cortex, area 4, and primary somatosensory cortex, area 3b, are homogenous areas, with the major division lying between the two. Accumulating empirical and theoretical evidence, however, has begun to question the validity of the Brodmann map for various cortical areas. Here, we combined in vivo cortical myelin mapping with functional connectivity analyses and topographic mapping techniques to reassess the validity of the Brodmann map in human primary sensorimotor cortex. We provide empirical evidence that area 4 and area 3b are not homogenous, but are subdivided into distinct cortical fields, each representing a major body part (the hand and the face). Myelin reductions at the hand–face borders are cortical layer-specific, and coincide with intrinsic functional connectivity borders as defined using large-scale resting state analyses. Our data extend the Brodmann model in human sensorimotor cortex and suggest that body parts are an important organizing principle, similar to the distinction between sensory and motor processing.

The association of post-stroke anhedonia with salivary cortisol levels and stroke lesion in hippocampal/parahippocampal region.

BackgroundAnhedonia constitutes a coherent construct, with neural correlates and negative clinical impact, independent of depression. However, little is known about the neural correlates of anhedonia in stroke patients. In this study, we investigated the association of post-stroke anhedonia with salivary cortisol levels and stroke location and volume.Patients and methodsA psychiatrist administered the Structured Clinical Interview for Diagnostic and Statistical Manual of Mental Disorders, Fourth Edition to identify anhedonia in 36 inpatients, without previous depression, consecutively admitted in a neurology clinic in the first month after a first-ever ischemic stroke. Salivary cortisol levels were assessed in the morning, evening, and after a dexamethasone suppression test. We used magnetic resonance imaging and a semi-automated brain morphometry method to assess stroke location, and the MRIcro program according to the Brodmann Map to calculate the lesion volume.ResultsPatients with anhedonia had significantly larger diurnal variation (P-value =0.017) and higher morning levels of salivary cortisol (1,671.9±604.0 ng/dL versus 1,103.9±821.9 ng/dL; P-value =0.022), and greater stroke lesions in the parahippocampal gyrus (Brodmann area 36) compared to those without anhedonia (10.14 voxels; standard deviation ±17.72 versus 0.86 voxels; standard deviation ±4.64; P-value =0.027). The volume of lesion in the parahippocampal gyrus (Brodmann area 36) was associated with diurnal variation of salivary cortisol levels (rho=0.845; P-value =0.034) only in anhedonic patients.ConclusionOur findings suggest that anhedonia in stroke patients is associated with the volume of stroke lesion in the parahippocampal gyrus and with dysfunction of the hypothalamic–pituitary–adrenal axis.

Cytoarchitectonical analysis and probabilistic mapping of two extrastriate areas of the human posterior fusiform gyrus

The human extrastriate visual cortex comprises numerous functionally defined areas, which are not identified in the widely used cytoarchitectonical map of Brodmann. The ventral part of the extrastriate cortex is particularly devoted to the identification of visual objects, faces and word forms. We analyzed the region immediately antero-lateral to hOc4v in serially sectioned (20 μm) and cell body-stained human brains using a quantitative observer-independent cytoarchitectonical approach to further identify the anatomical organization of the extrastriate cortex. Two novel cytoarchitectonical areas, FG1 and FG2, were identified on the posterior fusiform gyrus. The results of ten postmortem brains were then registered to their MRI volumes (acquired before histological processing), 3D reconstructed, and spatially normalized to the Montreal Neurological Institute reference brain. Finally, probabilistic maps were generated for each cytoarchitectonical area by superimposing the areas of the individual brains in the reference space. Comparison with recent functional imaging studies yielded that both areas are located within the object-related visual cortex. FG1 fills the gap between the retinotopically mapped area VO-1 and a posterior fusiform face patch. FG2 is probably the correlate of this face patch.

Microstructural Parcellation of the Human Cerebral Cortex – From Brodmann's Post-Mortem Map to in vivo Mapping with High-Field Magnetic Resonance Imaging

The year 2009 marked the 100th anniversary of the publication of the famous brain map of Korbinian Brodmann. Although a “classic” guide to microanatomical parcellation of the cerebral cortex, it is – from today's state-of-the-art neuroimaging perspective – problematic to use Brodmann's map as a structural guide to functional units in the cortex. In this article we discuss some of the reasons, especially the problematic compatibility of the “post-mortem world” of microstructural brain maps with the “in vivo world” of neuroimaging. We conclude with some prospects for the future of in vivo structural brain mapping: a new approach which has the enormous potential to make direct correlations between microstructure and function in living human brains: “in vivo Brodmann mapping” with high-field magnetic resonance imaging.

Centenary of Brodmann's map — conception and fate

Rarely in the history of neuroscience has a single illustration been as influential as the cytoarchitectonic map of the human brain published by Korbinian Brodmann in his monograph from 1909. The map presents the segregation of the cerebral cortex into 43 areas, as visible in cell body-stained histological sections. More importantly, Brodmann provided a comparative neuroanatomical approach and discussed ontogenetic and pathological aspects as well as structural-functional correlations. One hundred years later, a large number of neuroscientists still use Brodmann's map for localizing neuroimaging data obtained in the living human brain.

One Century of Brain Mapping Using Brodmann Areas*

100 years after their publication, Brodmann's maps of the cerebral cortex are universally used to locate neuropsychological functions. On the occasion of this jubilee the life and work of Korbinian Brodmann are reported. The core functions of each single Brodmann area are described and Brodmann's views on neuropsychological processes are depicted.

Anatomical organization of the auditory cortex.

The identification of areas that contribute to auditory processing in the human cerebral cortex has been the subject of sporadic investigation for more than one century. Several anatomical schemas have been advanced, but a standard model has not been adopted by researchers in the field. Most often, the results of functional imaging or electrophysiological studies involving auditory cortex are related to the cytoarchitectonic map of Brodmann (1909). Though useful as a guide and point of reference, this map has never been validated and appears to be incomplete. In recent years, renewed interest in the organization of auditory cortex has fostered numerous comparative studies in humans, nonhuman primates, and other mammalian species. From these efforts, common features of structural and functional organization have begun to emerge from which a working model of human auditory cortex can be derived. The results of those studies and the rudiments of the model are reviewed in this manuscript.

Twenty-first century brain banking. Processing brains for research: the Columbia University methods.

Carefully categorized postmortem human brains are crucial for research. The lack of generally accepted methods for processing human postmortem brains for research persists. Thus, brain banking is essential; however, it cannot be achieved at the cost of the teaching mission of the academic institution by routing brains away from residency programs, particularly when the autopsy rate is steadily decreasing. A consensus must be reached whereby a brain can be utilizable for diagnosis, research, and teaching. The best diagnostic categorization possible must be secured and the yield of samples for basic investigation maximized. This report focuses on integrated, novel methods currently applied at the New York Brain Bank, Columbia University, New York, which are designed to reach accurate neuropathological diagnosis, optimize the yield of samples, and process fresh-frozen samples suitable for a wide range of modern investigations. The brains donated for research are processed as soon as possible after death. The prosector must have a good command of the neuroanatomy, neuropathology, and the protocol. One half of each brain is immersed in formalin for performing the thorough neuropathologic evaluation, which is combined with the teaching task. The contralateral half is extensively dissected at the fresh state. The anatomical origin of each sample is recorded using the map of Brodmann for the cortical samples. The samples are frozen at −160°C, barcode labeled, and ready for immediate disbursement once categorized diagnostically. A rigorous organization of freezer space, coupled to an electronic tracking system with its attached software, fosters efficient access for retrieval within minutes of any specific frozen samples in storage. This report describes how this achievement is feasible with emphasis on the actual processing of brains donated for research.

Gene expression profiling of primate neocortex: molecular neuroanatomy of cortical areas

One hundred years have passed since Brodmann's mapping of the mammalian neocortex. Solely on the basis of morphological observations, he envisaged the conservation and differentiation of cortical areal structures across various species. We now know that neurochemical, connectional and functional heterogeneity of the neocortex accompanies the morphological divergence observed in such cytoarchitectonic studies. Nevertheless, we are yet far from fully understanding the biological significance of this cortical heterogeneity. In this article, we review our past works on the gene expression profiling of the postnatal primate cortical areas, by quantitative real-time polymerase chain reaction (PCR), DNA array, differential display PCR and in situ hybridization analyses. These studies revealed both the overall homogeneity of gene expression across different cortical areas and the presence of a small number of genes that show markedly area-specific expression patterns. In situ hybridization showed that, among such genes, occ1 and retinol-binding protein (RBP) mRNAs are selectively expressed in the neuronal populations that seem to be involved in distinct neural processing such as sensory reception (for occ1) and associative function (for RBP). Such a molecular neuroanatomical approach has the promise to provide an important link between structure and function of the cerebral cortex.

Cytoarchitectonic identification and probabilistic mapping of two distinct areas within the anterior ventral bank of the human intraparietal sulcus

Anatomical studies in the macaque cortex and functional imaging studies in humans have demonstrated the existence of different cortical areas within the intraparietal sulcus (IPS). Such functional segregation, however, does not correlate with presently available architectonic maps of the human brain. This is particularly true for the classical Brodmann map, which is still widely used as an anatomical reference in functional imaging studies. The aim of this cytoarchitectonic mapping study was to use previously defined algorithms to determine whether consistent regions and borders can be found within the cortex of the anterior IPS in a population of 10 post-mortem human brains. Two areas, the human intraparietal area 1 (hIP1) and the human intraparietal area 2 (hIP2), were delineated in serial histological sections of the anterior, lateral bank of the human IPS. The region hIP1 is located posterior and medial to hIP2, and the former is always within the depths of the IPS. The latter, on the other hand, sometimes reaches the free surface of the superior parietal lobule. The delineations were registered to standard reference space, and probabilistic maps were calculated, thereby quantifying the intersubject variability in location and extent of both areas. In the future, they can be a tool for analyzing structure-function relationships and a basis for determining degrees of homology in the IPS among anthropoid primates. We conclude that the human IPS has a more finely grained parcellation than shown in Brodmann's map.

Human Somatosensory Area 2: Observer-Independent Cytoarchitectonic Mapping, Interindividual Variability, and Population Map

We analyzed the topographical variability of human somatosensory area 2 in 10 postmortem brains. The brains were serially sectioned at 20 μm, and sections were stained for cell bodies. Area 2 was delineated with an observer-independent technique based on significant differences in the laminar densities of cell bodies. The sections were corrected with an MR scan of the same brain obtained before histological processing. Each brain's histological volume and representation of area 2 was subsequently reconstructed in 3-D. We found that the borders of area 2 are topographically variable. The rostral border lies between the convexity of the postcentral gyrus and some millimeters deep in the rostral wall of the postcentral sulcus. The caudal border lies between the fundus of the postcentral sulcus and some millimeters above it in the rostral wall. In contrast to Brodmann's map, area 2 does not extend onto the mesial cortical surface or into the intraparietal sulcus. When the postcentral sulcus is interrupted by a gyral bridge, area 2 crosses this bridge and is not separated into two segments. After cytoarchitectonic analysis, the histological volumes were warped to the reference brain of a computerized atlas and superimposed. A population map was generated in 3-D space, which describes how many brains have a representation of area 2 in a particular voxel. This microstructurally defined population map can be used to demonstrate activations of area 2 in functional imaging studies and therefore help to further understand the role of area 2 in somatosensory processing.
