Potential role of oxidative stress in pathogenesis of benign prostatic hyperplasia in male dogs.

The pathogenesis of benign prostatic hyperplasia (BPH) in dogs is still not fully understood. Some studies suggest that oxidative stress may have a potential role in the pathogenesis of BPH in dogs. Age-related hormonal changes may activate a chronic inflammatory response in the prostate. This causes the generation of reactive oxygen species (ROS) and results in oxidative stress. Excessive production of ROS results in DNA damage and hyperplastic transformation of prostatic cells. The use of antioxidants for improvement of treatment outcomes for canine PBH has been discussed. Further research is needed on the importance of oxidative stress in the development of BPH in dogs and the usefulness of antioxidants in the supportive treatment of this condition.

Oxidative stress biomarkers in dogs with benign prostatic hyperplasia

BackgroundThe aim of this study was to evaluate total antioxidant capacity (TAC) and biomarkers of lipid and protein peroxidation in the blood serum of dogs with benign prostatic hyperplasia (BPH). The study was conducted on 36 intact male dogs of various breeds. The dogs were assigned to two groups: BPH group (n = 18) and non-affected group (n = 18). Blood samples were collected from the cephalic vein. The antioxidant status of the serum was assessed using TAC. The levels of bityrosine, formylkynurenine and SH-groups were used as protein peroxidation biomarkers and the level of radical cations of N,N-diethyl-paraphenylene diamine (RC-DEPPD) was used as a marker for lipid peroxidation. TAC and the concentrations of SH-groups and RC-DEPPD in the serum were determined spectrophotometrically, the concentrations of bityrosine and formylokynurenine, were determined using spectrofluorimetric methods.ResultsThe mean value of TAC in the serum was significantly lower (P = 0.01) in BPH dogs than in non-affected dogs (3.10 ± 0.56 vs 4.20 ± 1.60 μmol/g protein). Mean levels of protein and lipid oxidation biomarkers showed a trend towards oxidative imbalance, but there were no statistically significant differences between dogs with BPH and controls (P > 0.05).ConclusionIn conclusion, significantly lower serum TAC in dogs with BPH compared to non-affected dogs suggests a potential involvement of oxidative stress in the pathogenesis of BPH in dogs. More studies are needed to clarify the role of oxidative stress in the development of BPH in dogs.

Assessment of the utility of platelet indices to diagnose clinical benign prostatic hyperplasia in dogs.

Platelet indices changes in severely ill people and in dogs with inflammation are compatible findings. This study aimed to compare platelet indices between dogs with clinical benign prostatic hyperplasia (BPH) and healthy controls. Additionally, to determine whether there is a correlation between the relative prostatic size (S rel) and the platelet indices in BPH dogs. Thirty-five adult intact male dogs of different breeds were allocated to the experimental groups: dogs with clinical BPH (groups A; n = 24; median age of 6 years; the median weight of 8.50 kg) and healthy dogs (group B; n = 11; median age 5.50 years; the median weight of 7.00 kg) based on physical examination, clinical signs, and S rel detected by ultrasonographic findings. The individual prostatic volume (IPV) was divided by the expected prostatic volume (EPV) to determine the relative prostatic size in dogs over 4 years old. Platelet indices were compared between the two groups, and a correlation between S rel and these indices was calculated. The median S rel of dogs in group A was significantly higher (P = 0.001), and the mean plateletcrit (PCT) was significantly lower (P = 0.003) compared with those in group B. S rel showed a significant negative correlation with PLT and PCT (r = -0.388; P = 0.02 and r = -0.402; P = 0.01). Receiver operating characteristic (ROC) analysis showed PLT and PCT thresholds for estimating S rel > 1 with 75% and 87.5% sensitivity and 71.82 and 63.64% specificity. The findings of this study support the use of platelet indices like PLT and PCT to detect clinical BPH in dogs. However, more research is needed to confirm their utility in conjunction with other previously described diagnostic factors.

Establishment of hormone-induced canine benign prostatic hyperplasia model: A prospective, controlled study

PurposeTo evaluate the feasibility of the establishment of a more efficient hormone-induced canine benign prostatic hyperplasia (BPH) model. MethodsThis prospective pilot study included a total of 16 male beagle dogs who underwent dihydrotestosterone and β-estradiol treatment after castration. They were randomly divided into three groups; eight beagles in group A with 1.0 ml hormone treatment, four beagles in group B with 0.8 ml hormone treatment, and four beagles in group C with 1.2 ml hormone treatment, each according to the table of random digit. The size of the prostate was measured using magnetic resonance imaging before and 4, 8, and 12 weeks after intramuscular injection of hormone drugs. Prostate size larger than 18 g was regarded as BPH in dogs. ResultsBeagle dogs in all three groups were successfully modeled for BPH. The experimental group A (n = 8) was successfully modeled after 4 weeks of 1.0 ml hormone treatment, while the control group B (n = 4) was successfully modeled after 8 weeks of 0.8 ml hormone treatment, and the control group C (n = 4) was successfully modeled after 12 weeks of 1.2 ml hormone treatment. ConclusionsAppropriate dose of hormone can significantly shorten the time of successful establishment of canine BPH model, and an increase in hormone dosage can inhibit prostatic hyperplasia.

MicroRNA and vascular endothelial growth factor (VEGF) as new useful markers in the diagnosis of benign prostatic hyperplasia in dogs

Numerous specific biomarkers with a prognostic and diagnostic value comparable to histopathological findings are now used for non-invasive diagnosis of prostate diseases in humans. Meanwhile, as far as dogs are concerned, the diagnosis of prostate disorders is based solely on clinical examination and ultrasound (USG). Therefore, the aim of the study was to assess the usefulness of two biomarkers, i.e. miRNA-129 and VEGF for the diagnosis of BPH in dogs. The study involved 40 dogs divided into three groups. Group I (n = 9) comprised healthy dogs up to the age of 5 years, Group II (n = 17) comprised dogs between the ages of 5–10 suffering from BPH as confirmed by the examination and Group III (n = 14) comprising dogs over 10 years of age, which also had BPH confirmed. The results demonstrated that dogs in group II and III exhibited a significant decrease in miRNA expression (P < 0.0001) and a significant increase in serum VEGF levels (P = 0.025) when compared to the dogs in group I. There was also a positive correlation between the prostate size and VEGF level. The findings led to the conclusion that the determination of miRNA-129 and VEGF can significantly contribute to the diagnosis of prostate disorders in dogs.

Computer-assisted sperm analysis and comparative diagnostic imaging of benign prostatic hyperplasia in dogs by ultrasound, X-ray and computed tomography

Early diagnosis of benign prostatic hyperplasia (BPH) in dogs using imaging methods has become increasingly prevalent in recent years. That allows accurate differentiation of BPH from other diseases, as well as timely prophylaxis and treatment. The aim of the research was to make a comparative analysis of BPH diagnosis by ultrasound, X-ray and computed tomography (CT) and computer-assisted sperm analysis (CASA) of ejaculates from sexually mature male dogs. Two hundred and fifty patients of different breeds, 80 of whom with previous clinical history and BPH-related complaints at 3 to 7 years of age, were followed. The results of echographic studies of dogs with clinical signs of BPH showed structural prostate changes. Focal inflammation, small or larger intra- or paraprostatic cysts (7%), neoplasia (3%), acute or chronic prostatitis (15%) and prostate abscesses (3%) were detected. The remaining 72% of patients demonstrated evidence of hypo -to hyper heterogenic parenchyma with moderate heterogeneity, which are characteristic of BPH. X-ray findings about prostate volume showed that 85% of the patients studied had an increase in prostate volume, enlarged soft tissue shadow in the caudal abdomen or cranially to the pelvis but caudally to the bladder. These data were found out in 13% more patients than those with ultrasound data for BHP. In this study, computed tomography demonstrated structural changes and precise prostate size in all 10 patients with suspected BPH. The CASA results showed that the percentage of static sperm was significantly higher in dogs with BPH than in healthy dogs. At the same time, there was a decline in the percentage of cells with progressive movement at a rapid and medium rate of movement in patients with BPH compared to those data in healthy dogs. These changes could have a negative effect on the fertilising potential of the sperm. The obtained results suggested that the application of CT and CASA for early diagnosis of BPH in dog breeders from rare and valuable breeds is advisable and useful for timely prevention and treatment of the disease.

Echotexture Analysis of Prostate Parenchyma for Detection of Benign Prostatic Hyperplasia in Dogs

Ultrasonography is one of the most common methods for the diagnosis of prostate disorders, such as benign prostatic hyperplasia (BPH), in dogs. Changes in the echotexture are one of the indicators used to diagnose prostate disorders. The purpose of this study was to investigate the changes occurred in the dogs’ prostate echotexture during the induction of BPH using image analysis. Twenty sexually mature male intact mixed-breed dogs were selected and divided randomly into control and BPH-induced groups. BPH was induced using testosterone and estrogen injections for 63 days. The ultrasound imaging of the dogs’ prostate was performed during the induction of BPH on days 0, 21, 42, and 63. The echotexture of the prostate parenchyma was analyzed using the Image J software. Then, the changes in the echotexture and its correlation and linear regression with the prostate volume and canine prostate specific esterase (CPSE) concentration were evaluated by statistical tests. The prostate parenchyma echotexture did not show any significant changes during the induction of BPH and in comparison with that of the control group. While prostate volume and CPSE concentration increased significantly, indicating that BPH was induced in the dogs. There was no significant correlation and linear regression between the prostate echotexture and prostate volume or between the CPSE concentration and prostate echotexture. According to the results, the alteration in the prostate parenchymal echotexture did not occur in the early stages of induced BPH, but significant changes occurred in the prostate volume and CPSE concentration during those early stages.

Comparison of Clinical Effectiveness of Deslorelin Acetate and Osaterone Acetate in Dogs with Benign Prostatic Hyperplasia.

Simple SummaryThe article compares the treatment efficacy and adverse effects of two drugs used for benign prostate hyperplasia (BPH) therapy in dogs: YpozaneTM (osaterone acetate) and SuprelorinTM (deslorelin acetate). YpozaneTM is a registered medication for this condition in dogs, whereas SuprelorinTM is registered for pharmacological castration in dogs. The clinical trial proved both drugs to be safe and effective in reducing BPH-related symptoms in dogs, and the noted adverse effects were only mild—mostly weight gain. With osaterone acetate the clinical improvement was noted sooner (from day 7 onwards) than with SuprelorinTM (from day 21 onwards), but it lasted shorter up to 24 weeks, while in the SuprelorinTM group, clinical effect remained stable until the end of the study (36 weeks). Both medications can be recommended for treatment of symptoms related to BPH in dogs, as none of the drugs had serious influence on the general health status and both provided substantial clinical improvement.This article presents the results of a randomized clinical trial, designed to compare the efficacy and therapeutic profiles of YpozaneTM (osaterone acetate—OA) or SuprelorinTM (deslorelin acetate—DA) in male dogs with clinical signs of benign prostate hyperplasia (BPH). Forty-five intact male dogs were used in the study. The Group I (negative control) included 10 healthy dogs, the Group II (positive control) included 10 dogs with confirmed BPH and no treatment, whereas Group III and IV consisted of dogs with BPH and treated either with DA (15 dogs) or OA (10 dogs). The clinical response, testosterone and estradiol levels, hematology, biochemistry, and adverse effects incidence were evaluated. Both OA and DA proved to be effective for BPH treatment in dogs, as they allowed for the clinical remission in all treated dogs. The complete alleviation of BPH symptoms was noticed sooner with the use of OA (in 80% of dogs from day 7) compared to DA (in 40% of dogs within the first 21 days). The recurrence of clinical signs related to BPH was observed from week 24 in dogs treated with OA, whereas no relapse was noticed in dogs treated with DA at the end of the 36 weeks of the observation period. In 5 dogs (33%) treated with DA, a flare-up effect (increase in the clinical signs associated with BPH) was noticed on day 7. Despite individual differences in the clinical action, both medications were effective and safe options for the treatment of symptoms related to BPH in dogs.

Ultrasonographic Evaluation of the Effect of Nigella sativa Oil on Prostate Dimensions in Adult Iranian Mixed-Breed Dogs

Objective- The aim of the present study was to assess the effect of Nigella sativa oil on prostate dimensions in adult dogs. Design- Experimental study Animals- Five adult male dogs Procedures- In this research, 5 adult male Iranian mixed-breed dogs with a mean age of 7.30 ± 2.77 years and a mean weight of 27 ± 5.7 kg with clinical symptoms of prostatitis were selected. Sagittal and transverse ultrasound images were performed by using a 5 MHZ convex transducer. Results- In the obtained images desired parameters such as length, width, and height of the prostate gland were measured before and after administration of Nigella sativa. Results showed significant differences between all measured parameters before and after drug administration (p < 0.05).Conclusion and Clinical Relevance- This study confirms that Nigella sativa oil can reduce prostatic dimensions in adult male dogs. So, it can be used as a safe and effective drug for the treatment of prostate-related diseases such as benign prostatic hyperplasia in dogs.

Treatment of experimentally induced benign prostatic hyperplasia with Tadalafil and castration in dogs

New methods are being developed for the treatment of benign prostatic hyperplasia (BPH) in dogs. The aim of the present study was to evaluate the effects of Tadalafil on the treatment of experimentally induced BPH in dogs. Twenty-five adult intact male dogs were randomly divided into five groups (n = 5): normal group; dogs induced with BPH and treated with Tadalafil (5 mg/day p.o.); dogs which received Tadalafil (5 mg/day p.o.); dogs induced with BPH and treated with castration; and dogs induced with BPH. For 4 sequential weeks, the hematologic and prostate-specific factors (dihydrotestosterone (DHT), serum prostate-specific antigen (PSA), serum prostatic acid phosphatase (PAP), and canine prostatic specific esterase (CPSE)) were measured. Significant differences were observed in the level of PSA, CPSE, and PAP concentration between the normal vs. BPH-Tadalafil, BPH-castrated, and BPH groups. Treating BPH-induced dogs with Tadalafil or castration significantly declined the serum PSA, CPSE, and PAP levels compared to those of the untreated BPH-induced group. The treatment of normal dogs with Tadalafil did not affect prostate-specific biomarkers in comparison with normal dogs. In conclusion, and according to the prostatic indices, it could be stated that Tadalafil, compared with castration, could be used for the treatment of BPH in dogs.

Changes in the oxidative stress factors and inflammatory proteins following the treatment of BPH-induced dogs with an anti-proliferative agent called tadalafil.

Finding a medical treatment which can combat cell proliferation and relax smooth muscles in canine benign prostatic hyperplasia (BPH) appears to be imperative. This study aimed to evaluate the oxidative stress and inflammatory proteins following the treatment of dogs induced for BPH with an anti-proliferative agent called tadalafil. Twenty-five adult intact male dogs were randomly designated into five groups (n=5): Control group was not induced for BPH and not treated with tadalafil; dogs induced for BPH by testosterone enanthate and estradiol benzoate and treated with tadalafil (5mg/day P.O.); dogs which received tadalafil (5mg/day P.O.); dogs induced for BPH and treated with castration; and dogs induced for BPH. Oxidative stress factors (glutathione peroxidase [GPX], superoxide dismutase [SOD], catalase) and inflammatory proteins (haptoglobin, serum amyloid A [SAA], malondialdehyde [MDA]) were measured in the blood serum for four sequential weeks. Glutathione peroxidase and SOD serum levels declined in dogs in the BPH-induced group compared to those in the control group. Those levels diminished in BPH-induced castrated and tadalafil-treated groups. The changes in the GPX and SOD serum concentrations were not significant between the BPH-induced castrated group and BPH-induced tadalafil-treated group. Moreover, MDA concentration increased slightly in groups with BPH and groups which were castrated. Generally, however, there were no significant differences in the MDA serum concentrations between other groups. Haptoglobin and SAA concentrations increased in BPH-castrated group. Also, the differences in haptoglobin and SAA were not significant between the groups. Tadalafil could not control oxidative stress and inflammatory mediators which happened during BPH in dogs.

Stable state of serum inflammatory cytokines during induction of benign prostate hyperplasia in dogs

Objective: To monitor serum inflammatory cytokines during induction of benign prostate hyperplasia in dogs. Methods: This research was designed as a case-control study. There were 20 adult mixed-breed intact male dogs, which were divided into the normal group (n=10) and the benign prostate hyperplasia group (n=10). In the benign prostate hyperplasia group, benign prostate hyperplasia was induced by injection of testosterone (75.00 mg/dog, i.m.) and estrogen (0.75 mg/dog, i.m.) on day 0 (day of the first injection), day 21, day 42, and day 63. The doses of testosterone were doubled on days 21, 42, and 63. The normal group did not receive any injection. Blood sampling was performed from the jugular vein at days 0, 21, 42, and 63. The concentrations of interleukin-8, interleukin-10, and tumor necrosis factor-α (TNF-α) were assayed by enzyme-linked immunosorbent assay. Results: The levels of interleukin-8, interleukin-10 and TNF-α were not significantly different between the normal group and the benign prostate hyperplasia group. Also, concentrations of the pro-inflammatory cutokines were not significantly different between the normal group and the BPH group in each day of sampling. Conclusions: In spite of the induction of benign prostate hyperplasia, changes in the concentration of blood serum inflammatory cytokines were not significantly different with that of the normal group and between the days of induction of benign prostate hyperplasia during two months. It reveals that there is a stable state of serum inflammatory cytokines during induction of benign prostate hyperplasia in dogs.

Comparison of computed tomographic and cytological results in evaluation of normal prostate, prostatitis and benign prostatic hyperplasia in dogs.

Prostate gland can be structurally evaluated by computed tomography (CT) with taking advantages of tomographic feature and post-contrast parenchymal changes. The current examination initiated to determine association between computed tomographic and cytological results in evaluation of canine prostate. Thirty mature male dogs were included and under gone by both CT and fine needle sampling of prostate. The cytology and CT examination results showed 18/30 (60.00%) and 15/30 (50.00%) normal prostate, 5/30 (16.66%) and 4/30 (13.33%) prostatitis and 7/30 (23.33%) and 11/30 (36.66%) benign prostatic hyperplasia, respectively. Moderate agreement has been found between cytology and final diagnosis based on pre-contrast CT images, however fair agreement was existed between cytological diagnosis and final CT interpretation according to post-contrast and both pre- and post- contrast CT series. Additionally, the internal iliac lymph node length showed statistically significant difference in prostatitis compared to normal and benign hyperplastic prostates in this study. In conclusion, the fair and moderate associations between cytology and final diagnosis based on CT images should be considered and they can be used in further investigations and clinical examinations. Also, using internal iliac lymph node length to differentiate prostatitis with normal and benign hyperplastic prostates can be used efficiently in diagnosis to choose the best method of management and have a proper follow up and prognosis.

Canine prostate models in preclinical studies of minimally invasive interventions: part II, benign prostatic hyperplasia models.

Canine prostate is widely used as animal model in the preclinical evaluation of emerging therapeutic interventions. Spontaneous benign prostatic hyperplasia (BPH) is common in adult intact male dogs with two distinct pathological types: glandular and complex form of prostatic hyperplasia. The complex form of prostatic hyperplasia, usually occurring in older dogs, represents an ideal model because of its unique pathologic feature, including not only glandular hyperplasia but also an increase in prostate stromal components. The limited commercial availability of adult dogs with spontaneous BPH motivates experimentally induced BPH in young dogs. Hormone-induced canine BPH model has been well established with various hormonal treatment regimens and administration approaches. The goal of this review is to provide the veterinary background in spontaneous BPH in dogs, summarize the techniques in hormonal induction of canine BPH, and highlight the pathological and clinical limitations of the canine models that may lead to distinct therapeutic responses compared to clinical trials in humans.

Hemato-biochemical alterations and urinalysis in dogs suffering from benign prostatic hyperplasia.

Aim:The study was designed to evaluate the hemato-biochemical alterations, urinalysis along with histomorphological and histological changes of prostate glands in dogs affected with benign prostatic hyperplasia (BPH) in and around Bhubaneswar, Odisha, India.Materials and Methods:In toto, 445 dogs presented to the Teaching Veterinary Clinical Complex of the College of Veterinary Sciences and Animal Husbandry, one Government Veterinary Hospital and two pet clinics in and around Bhubaneswar screened for the presence of BPH. Most of the 57 dogs were 6 years and above as reported by the owners. Only 57 dogs found positive for BPH basing on the presence of typical clinical signs subjected for a detailed hemato-biochemical study. Most of the 57 dogs were 6 years and above as reported by the owners. Routine and microscopic urinalyses were done as per the routine procedure. Histomorphological evaluations of prostate glands were done through manual rectal palpation. Histological examinations of prostate tissue sections of two dead dogs were conducted with routine hematoxylin and eosin stain.Results:The study revealed about 12.8% (57/445) of dogs was suffering from BPH. Typical clinical signs - such as passing small thin tape-shaped feces, holding tail away from backward, tenesmus, and straining during urination and defecation - were seen in most of the cases. Urine samples of affected dogs were positive for glucose, occult blood, and protein. A significant decrease in lymphocytes and increase in eosinophil counts in dogs with BPH was recorded. Serum biochemical analysis showed a nonsignificant increase in creatinine and blood urea nitrogen with a significant decrease in total protein, albumin, globulin, A:G ratio. Histology of prostate glands collected during postmortem was characterized by fibrosis of prostate gland, and hyperplasia of the acinar epithelium.Conclusions:High rate of the prevalence of BPH in dogs poses an alarming condition which if diagnosed at an early stage can certainly prolong the longevity of the dogs.

Utilização da furosemida em bolus e em infusão contínua em cães e gatos: revisão de literatura

A furosemida é um potente diurético com ação no ramo espesso ascendente da alça de Henle. Em Medicina Veterinária é amplamente utilizada devido à rápida ação na remoção de fluidos intersticiais, média toxicidade e rápida excreção do organismo. É indicada no tratamento da retenção líquida inadequada, aumento da diurese, edema pulmonar, insuficiência cardíaca congestiva, síndrome nefrótica, doenças renais agudas e crônicas, ascites, edemas periféricos e pacientes irresponsivos a outros diuréticos ou anti-hipertensivos. A via de administração da furosemida endovenosa pode ser realizada sob a forma de bolus ou infusão contínua, com diferenças marcantes entre elas. Alterações hemodinâmicas, variações nas concentrações séricas e resistência diurética são desvantagens observadas na forma de bolus. Já sob a forma de infusão contínua, o aumento e a manutenção da diurese são vantagens observadas, mas com desequilíbrios hidroeletrolíticos mais evidenciados comparada à administração em bolus, necessitando de monitoramento constante. Atualmente, poucos são os estudos científicos na Medicina Veterinária que demonstram as diferenças específicas, com as indicações, benefícios ou efeitos deletérios orgânicos da utilização da furosemida intravenosa sob a forma de bolus ou infusão contínua. Assim, este trabalho busca promover uma atualização científica quanto às características farmacológicas, doses e efeitos da furosemida endovenosa nas diferentes formas de administração, visando a auxiliar a conduta terapêutica quando utilizada em cães e gatos.

Computed Tomography of the Prostate Gland in Healthy Intact Dogs and Dogs with Benign Prostatic Hyperplasia.

To date, there is only scarce data on the evaluation of the prostate gland in dogs using computed tomography (CT). The aims of our study were to describe CT features of BPH in dogs and to determine the size of the prostate gland in healthy male dogs and dogs with benign prostatic hyperplasia (BPH) through CT. Additionally, we aimed to compare and establish the most useful parameters for CT measurements of the prostate in patients with BPH. The study population consisted of 20 healthy intact male dogs and 20 male intact dogs with confirmed BPH. Pre- and post-contrast CT studies were evaluated. The most common CT features in dogs with recognized BPH were symmetrical prostatomegaly and heterogeneity of the prostatic parenchyma. The mean prostatic density (D) was 56HU (±4.39) in pre-contrast CT images and 84HU (±8) in post-contrast images in dogs with BPH. The mean prostatic length (L) was 43.87 mm (±11), the mean width (W) amounted to 48.95 mm (±8.76) and the mean height (H) reached 44.9 mm (±9.48) in clinically affected patients. The mean ratios were: rL - 2,12 (±0.5); rW - 2.39 (±0.53) and rH - 2.16 (±0.39) in the BPH group. The prostate should be considered to be enlarged when rL exceeds 3.05; rW exceeds 3.38 and rH exceeds 2.94. Our findings indicated that CT is a useful tool in diagnosing prostate disorders, including BPH. The heterogeneity, density and ratios of prostatic length, width and height can be useful parameters in the diagnosis of BPH.

Transarterial prostatic embolization:initial experience in hormone-induced benign prostatic hyperplasia in dogs

Objective To evaluate the feasibility of transarterial prostate embolization for reducing the volume and its technical safety in hormone-induced dogs prostate hyperplasia. Methods Prostate hyperplasia was induced on 12 adult male dogs by administering testosterone propionate for 12 weeks.Then,the dogs were randomly assigned to transarterial prostatic embolization group (n=6) and control group (n=6).Twelve weeks after initiating the hormone treatment,6 animals in transarterial prostatic embolization group underwent prostatic arterial embolization with Embospheres 100—300μm in diameter.Based on MRI,the volume of prostate was measured for both groups on the three time points: before injection of hormone (BI),before the procedure of prostatic arterial embolization (BP) and 12 weeks after the procedure of prostatic arterial embolization (AP).Finally,the prostates and surrounding tissues were harvested for histopathology study.A statistical software package (SPSS version 19.0) was used for analysis.Measurement data were expressed as x± s.Samples paired t test was used to compare the prostatic volume between BP and AP.The t test was applied in the intergroup comparison.P<0.05 was considered to indicate a statistically significant difference. Result s The successful rate of BPH model establishment and superselective catheterization were 100%.No serious procedure-related complication was observed.After 12 weeks of hormone administration,the mean volume of prostate was 19.5±2.5 mm3in embolization group and 19.2±3.8 mm3in control group before prostatic arterial embolization,without statistical difference (t=0.14,P>0.05).At 12 weeks after prostatic arterial embolization,the mean volume of prostate was 6.1±1.4 mm3in embolization group and 17.8±3.5 mm3in control group with significant statistical difference (t=-7.6,P 0.05).Histopathology study revealed that embosphere particles had occluded partial arterioles of the prostate in the embolization group of dogs.Near the occluded arterioles,normal gland structure disappeared; while residual gland tissue appeared to be atrophied.In embolization group,there was no gross or microscopic change in the surrounding tissues including bladder,deferent duct and rectum.Extensive prostate glandular hyperplasia was observed in control group. Conclusions Transcatheter arterial embolization is feasible for reducing prostate volume without serious complication in hormone-induced prostate hyperplasia dogs and worthy of further investigation.
