Isolation and Molecular Identification of Pathogenic Bacteria in Giant Gourami (<i>Osphronemus gouramy</i>) Cultivated in Tamansari and Singasari Villages, Banyumas Regency

Giant gourami (Osphronemus gouramy) is a freshwater fishery commodity with high economic value and is widely cultivated in Banyumas Regency. However, the emergence of diseases caused by pathogenic bacteria often becomes an obstacle in cultivation activities because it can reduce the survival rate and productivity of fish. This study aims to identify the types of pathogenic bacteria found in gouramy fish raised in Tamansari and Singasari Villages, Banyumas Regency. The research method was carried out by taking liver samples of gouramy fish which were then isolated on TSA media. Identification was carried out based on the morphological characteristics of bacterial colonies, the Gram KOH test, and molecular identification. The results showed that from both locations, several bacterial isolates were obtained with similar colony morphological characteristics, namely round (circular), small size, smooth edges, smooth surface, and cream color. The results of the Gram KOH test showed that all isolates were Gram-negative bacteria. Based on these characteristics, the isolates have similarities with the genus Acinetobacter which is often found as an opportunistic bacteria in freshwater fish. The results of molecular identification also showed that the bacteria obtained from the cultivated gouramy fish were Acinetobacter sp. This study provides a preliminary overview of the presence of opportunistic bacteria such as Acinetobacter sp., which have the potential to become pathogens in gourami when water quality conditions are unstable.

Study of the Presence of Streptococcosis in Tilapia Cultivation in Banyumas Regency

Tilapia is the most widely cultivated fish in Banyumas which is susceptible to bacterial diseases caused by the increasing number of disease cases in tilapia (Oreochromis niloticus) due to high cultivation intensification. One of the diseases that often attacks is streptococcosis caused by the bacteria Streptococcus sp., which can cause mass mortality and major economic losses for farmers. The purpose of this study was to identify pathogenic bacteria in farmed tilapia in Banyumas. The method used in this study was a purposive sampling method, namely by taking samples of tilapia that have characteristics of disease symptoms such as protruding eyes, damaged fins, and red wounds on the scales from three locations, namely Beji Village, Karangsalam Village, and Pasir Wetan Village. Bacterial samples were obtained from four organs, namely the brain, kidneys, eyes, and liver, then observed the morphology of bacterial colonies and Gram KOH tests were carried out. The results showed that bacterial colonies found in most organs were round to irregular in shape, white to yellowish in color, and Gram positive, which shows the characteristics of Streptococcus sp. bacteria. Based on these results, it can be concluded that Streptococcus sp. bacteria are still the main threat in tilapia cultivation in Banyumas, so it is necessary to take preventive measures through the implementation of good biosecurity, the use of immunostimulants, and the development of vaccines to maintain the sustainability of tilapia cultivation businesses, especially in Banyumas Regency.

Peran Kearifan Lokal Etnis Padang Pariaman dalam Mendukung Pembangunan Kabupaten Banyumas

This study aims to analyze the role of the local wisdom of the Padang Pariaman ethnic community in supporting the development of Banyumas Regency through social, economic, cultural, and institutional dimensions. A qualitative approach was employed using data collected through in-depth interviews with the Chairman of the Padang Pariaman Family Association (PKDP) Banyumas, participant observation of community activities, and review of regional policy documents and academic literature. The findings reveal that the local wisdom of the Padang Pariaman community—including katok ampe, raso pareso, a strong work ethic, kinship-based networks, and the adaptive principle of “where the earth is trodden, there the sky is upheld”—functions as cultural capital that strengthens social integration between migrants and the local Banyumas community. Economically, microenterprises and the culinary sector managed by Padang Pariaman migrants contribute significantly to job creation, income generation, and regional revenue. Socially, solidarity through the PKDP association is manifested in community-based activities such as fundraising, humanitarian support, and women’s empowerment initiatives. Culturally, the preservation of Minangkabau traditions in the public sphere enriches cultural diversity and supports the development of an inclusive regional identity. These findings indicate that the local wisdom of the Padang Pariaman diaspora serves not only as an ethnic marker but also as sustainable development capital that positively contributes to strengthening the creative economy, social cohesion, and multicultural character of Banyumas Regency.

This study aims to analyze the role of a multiparty cooperative model in developing pesantren-based halal tourism and its contribution to strengthening the local sharia economic ecosystem in Banyumas Regency, Central Java, Indonesia. This research employs a qualitative approach with a case study design. Data were collected through in-depth interviews, field observations, and document analysis involving pesantren leaders, cooperative managers, local communities, and tourism stakeholders. The data were analyzed using an interactive model, including data reduction, data display, and conclusion drawing, supported by triangulation techniques to ensure validity and reliability. The findings reveal that pesantren institutions play a strategic role as socio-religious and economic hubs in supporting halal tourism development. However, the absence of an integrated institutional framework limits the optimization of economic benefits. The study proposes a multiparty cooperative model that integrates pesantren, communities, government, and business actors. This model enhances coordination, promotes equitable resource distribution, and strengthens sectors such as halal culinary, religious tourism, and creative industries, contributing significantly to local economic resilience. This study is limited to a specific regional context, which may affect generalizability. However, it provides important implications for policymakers and practitioners in designing inclusive, community-based halal tourism strategies rooted in Islamic economic principles. This research offers a novel integrative model linking pesantren institutions, cooperative economics, and halal tourism within a unified framework. It contributes to the literature by demonstrating how Islamic social institutions can be operationalized to strengthen sustainable local sharia economies.

Strengthening Legal Certainty Through the Updating of Village Land Data to Support Land Registration

Introduction: This community service activity is motivated by the condition of land data in Pekuncen Village, Banyumas Regency, Central Java Province, which is obsolete both physically and substantively. This condition has resulted in disorderly village land administration that has led to legal cases involving village officials. Such disorder is caused by limitations in human resources in mastering appropriate technology.Purposes of The Devotion: The purpose of this community service is to provide appropriate technology transfer for updating village land data in order to achieve orderly village land administration and legal certainty.Method of The Devotion: The methods used include socialization, mentoring, and monitoring and evaluation of activity outcomes to support sustainability.Results Main Findings of the Devotion: The results of this community service demonstrate a significant impact on improving the capacity of village officials’ human resources through a series of activities that serve as solutions to the identified problems, namely: 1) enhanced understanding of village officials regarding valid documents as the basis for the history of land ownership; 2) improved mastery of computerized administration of juridical land data to ensure greater accuracy and effectiveness in addressing physical document issues of the Letter C Book, which is no longer feasible and contains many torn or missing pages; and 3) improved capacity in managing the administration of juridical land data renewal.

This study aims to classify rice-farming areas in Banyumas Regency based on differences in rice production characteristics between regions that have the potential to influence rice production planning and distribution. This study was conducted to support food security in line with Indonesia's national development vision for 2045. The data used included rice production volume and rice field area. Cluster analysis was performed using the K-Means method with the determination of the optimal number of clusters through the Elbow method, resulting in three regional clusters, namely low, medium, and high rice production clusters, which reflect the differences in rice production characteristics between subdistricts in Banyumas Regency. Rice production dynamics were modeled using the Verhulst growth model, which was solved numerically using the fourth-order Runge Kutta method, followed by the Adams-Bashforth-Moulton method to predict rice production. Predictions were made for 2025, 2030, and 2045 to illustrate rice production trends over several time periods. The main analysis of this study focused on the period up to 2030 because the model did not include several important external factors that could potentially affect rice production in the long term. The error analysis results showed that the error values in the predictions for the 2025 and 2030 periods were relatively small, indicating that the model was able to represent rice production dynamics well during those periods. Predictions up to 2045 are presented as a mathematical illustration of long-term trends and are not used as the main basis for error evaluation or drawing conclusions. The results of this study are expected to be used as preliminary considerations for local governments in formulating rice production plans, taking into account the different characteristics of regional clusters and the predicted rice production in Banyumas Regency.

Quality and safety evaluation of traditionally processed coconut crystal sugar in Cilongok, Banyumas

Abstract Coconut sugar is a powdered coconut sap derivative product that has high economic potential. In Cilongok District, Banyumas Regency, coconut sugar production is still produced traditionally without standardized procedures, resulting in inconsistencies in product quality, especially in the aspects of color, texture, and food safety. This study aims to evaluate the quality of coconut sugar produced by 11 local artisans through analysis of physicochemical, microbiological and organoleptic properties. The analysis showed that the moisture content of coconut sugar ranged from 1.10% to 3.38%, with a sweetness level (Brix) of 14.80%-31.00%. Color analysis using the CIELab system showed L values between 51.4-66.25 (bright), a values of 7.65-9.25 (tends to be reddish), and b values of 13.2-19.44 (tends to be yellowish). Organoleptic analysis that acceptance levels for color, aroma, and texture ranged from 3.30–5.40, 3.71–5.18, and 3.88–5.29, respectively (neutral to slightly liked categories). The Total Plate Count (TPC) test revealed that 6 out of 11 samples had too many colonies to count (TNTC), indicating high levels of microorganism contamination. Based on the evaluation results, only one sample did not meet the moisture content requirements according to the Indonesian National Standard (SNI 7388:2009), while all samples did not meet the microbiological safety standards. These findings suggest the need for the implementation of processing operational standards to improve quality consistency and safety of coconut sugar in the area.

The tourism industry in the Baturraden area can be said to be one of the most popular in Banyumas Regency. Tourism development around Baturraden has also changed along with the development of an educational tourism area, namely the Baturraden Botanical Garden. This study aims to examine and analyze the influence of attractiveness and price perception on revisit intention, using tourist satisfaction as a mediating variable. The research location is the Baturraden Botanical Garden, a tourist destination located in Banyumas Regency, Central Java. This research method uses a quantitative approach with a purposive sampling technique. The sample in this study is primary data obtained from questionnaires distributed to a sample of 140 respondents. The data analysis technique used in this study is SmartPLS v.4 software. The results of the study show that: 1) attractiveness has a positive effect on the intention to revisit, 2) price perception has a positive effect on the intention to revisit, 3) attractiveness has a positive effect on tourist satisfaction, 4) price perception has a positive effect on tourist satisfaction, 5) tourist satisfaction has a positive effect on the intention to revisit, 6) tourist satisfaction is able to mediate the effect of attractiveness on the intention to revisit, 7) tourist satisfaction is able to mediate the effect of price perception on the intention to revisit.

This study aims to analyze the influence of compensation, leadership style, work environment, and work motivation on employee job satisfaction at the Department of Population and Civil Registration of Banyumas Regency. This research employs a quantitative method with primary data collection through a 5-point Likert scale questionnaire. The sampling technique uses saturated sampling with a population of 91 employees, resulting in a sample of 90 employees, excluding the Head of Department due to their different structural role. Data analysis uses Partial Least Square-Structural Equation Modeling (PLS-SEM) with SmartPLS software version 3.0. The results indicate that compensation does not significantly influence employee job satisfaction, while leadership style, work environment, and work motivation have a positive and significant influence on employee job satisfaction. These findings contribute to the Human Resource Management literature by emphasizing the importance of leadership quality, work environment, and internal motivation as key factors shaping employee job satisfaction.

Evaluation of Ambient Air Quality at Integrated Waste Processing Facilities and Its Implications for Workers

Aim: This study evaluated air quality, worker exposure, and health risks at a 3R Integrated Waste Processing Facility in Banyumas Regency, Indonesia, where waste is combusted for extended periods. Methodology and results: Over five consecutive days, concentrations of PM2.5, PM10, CO2, CO, and respirable dust were measured during morning and afternoon shifts. Results revealed persistently hazardous particulate matter levels: PM2.5 exceeded World Health Organization (WHO) limits by 10–90 times, and PM10 by 5–44 times, with no significant difference between sampling periods (p > 0.05). Respirable dust and CO remained within safe thresholds, while CO2 was consistently elevated during morning startup operations. Risk Quotient (RQ) analysis identified CO2 as the only pollutant above the safety threshold (RQ = 1.184), correlating with worker-reported symptoms such as dizziness, fatigue, and difficulty concentrating. Intake calculations confirmed high chronic health risks, particularly for respiratory and cardiovascular disease. Conclusion, Significance, and Impact: Immediate interventions are required, including improved combustion efficiency, local exhaust ventilation, strict use of personal protective equipment, and regular worker health monitoring. Long-term strategies should focus on regulatory reform, advanced emission control technologies, and transitioning toward sustainable waste management through recycling, composting, and circular economy practices. These actions align with United Nations Sustainable Development Goals for health, sustainable cities, and climate action.

Stunting is a chronic nutritional problem that reflects impaired linear growth resulting from the interaction of biological, social, and environmental factors since early life. The prevalence of stunting remains high, as indicated by national data showing rates of 24.4% in 2021 and 21.6% in 2022. This study aimed to analyze the determinants of stunting severity among children under five based on a history of exclusive breastfeeding and household socioeconomic status. This study employed an observational analytic design with a cross-sectional approach using secondary data obtained from medical records of children under five at Sokaraja I Primary Health Center, Banyumas Regency. The study sample consisted of 328 stunted children selected through total sampling. Stunting severity was classified into stunted and severely stunted based on height-for-age (HAZ) indicators according to WHO standards. Independent variables included a history of exclusive breastfeeding and household socioeconomic status. Data were analyzed using the Chi-square test with a significance level of p 0.05. The results showed a statistically significant association between a history of exclusive breastfeeding and stunting severity, in which children who did not receive exclusive breastfeeding were more likely to experience more severe stunting. In addition, household socioeconomic status was significantly associated with stunting severity, with a higher proportion of severe stunting observed among children from low-income families. In conclusion, stunting severity among children under five is associated with exclusive breastfeeding practices and household socioeconomic conditions, highlighting the need for comprehensive stunting prevention strategies through strengthened midwifery care and multisectoral approaches

PENGARUH BRISK WALKING EXERCISE TERHADAP PENURUNAN TEKANAN DARAH PADA PENDERITA HIPERTENSI DIWILAYAH KERJA PUSKESMAS KEMBARAN I

Hypertension is a prevalent health problem that increases the risk of cardiovascular complications. In addition to pharmacological therapy, non-pharmacological interventions such as brisk walking exercise play an important role in helping control blood pressure. This study aimed to analyze the effect of brisk walking exercise on blood pressure reduction among patients with hypertension in the working area of Puskesmas Kembaran 1, Banyumas Regency. This research employed a quasi-experimental design with a one-group pretest–posttest approach. A total of 32 respondents were selected using purposive sampling, consisting of hypertensive patients without complications and with stable conditions. The intervention consisted of brisk walking exercise performed for three consecutive days, 15 minutes per session. Blood pressure was measured 30 minutes before and after each intervention using a sphygmomanometer and stethoscope. Data analysis used the paired t-test for systolic pressure and the Wilcoxon test for diastolic pressure. The results showed that most respondents were late elderly (56.3%), female (62.5%), and had a family history of hypertension (65.6%). The mean blood pressure before the intervention was 149.47 mmHg (systolic) and 89.97 mmHg (diastolic). After the intervention, it decreased to 142.09 mmHg and 84.38 mmHg, respectively. Statistical tests indicated a significant reduction in systolic (7.38 mmHg) and diastolic pressure (5.59 mmHg) with p = 0.0001. The study concludes that brisk walking exercise has a significant effect on reducing blood pressure among hypertensive patients. This intervention is recommended as a simple, safe, and affordable strategy for hypertension management in primary health care settings

Banyumas Regency is the largest producer of cardamom in Central Java. One of the production centres is in Cilongok District, but in 2024 there was a decline due to drought and low selling prices, which reduced farmers' motivation to cultivate cardamom. This condition necessitates the adaptation of agricultural practices to manage the risks arising from climate change and other challenges, ensuring the sustainability of farming activities amid environmental and economic pressures. This study aimed to analyze the resilience level of cardamom farmers in response to climate change and declining market prices. A cross-sectional survey design was employed, and the sampling technique used was cluster sampling, involving 50 cardamom farmers in Cilongok District. Data were analyzed using descriptive statistics and the Livelihood Vulnerability Index (LVI) approach. Based on the analysis, the LVI score was (0.55), which indicates a moderate to high level of vulnerability. The exposure index was (0.51), reflecting considerable exposure to climatic variability. The sensitivity index was (0.44), indicating moderate impacts on health, food security, and water availability, with fluctuating cardamom prices worsening the condition. The adaptive capacity index scored the highest at (0.68), suggesting a low adaptive capacity among farmers, mainly due to advanced age, low education levels, and limited institutional support. These findings underscore the urgency of implementing adaptation strategies such as technical training, strengthening of farmer organizations, expanding market access, and policy interventions to reduce vulnerability and enhance the resilience of cardamom farmers to climate variability and price instability.

Introduction: This study aims to examine the effects of financial inclusion, digital financial literacy, financial management, and locus of control on the performance of micro, small, and medium enterprises (MSMEs) in Banyumas Regency, using a Resource-Based Theory framework.Methods: The quantitative methodology of this study is based on primary data collected from 115 respondents, selected at random from a pool of MSME owners, via an online and offline survey.Results: The findings indicate that financial management and locus of control have a positive, significant effect on MSME performance, underscoring the importance of internal managerial capabilities and psychological resources. Conversely, financial inclusion and digital financial literacy do not show a significant impact on MSME performance, suggesting that access to financial services and digital financial tools has not yet been optimally utilized to enhance business performanceConclusion and suggestion: These results imply that financial access alone is insufficient without adequate managerial skills and strong internal controls. Therefore, MSME development programs are encouraged to focus on strengthening financial management practices and internal locus of control to improve MSME performance and sustainability. Micro, Small, and Medium Enterprises (MSMEs) play a vital role in strengthening the local economy of Sunyalangu Village, Karanglewas District, Banyumas Regency. However, most MSMEs still face limitations in utilizing digital technology, particularly in marketing, business administration, and transaction management. This community service activity, titled "Building Resilient MSMEs in the Digital Economy 5.0 Era," aims to improve digital literacy and technology-based business management skills among MSMEs. This program was implemented using a gradual training, mentoring, and counseling approach from October to November 2025. A total of 30 MSMEs participated in the activities, which included digital literacy counseling, digital marketing training through social media and marketplaces, and assistance in creating online store accounts and digital-based administration systems. The results of the activity showed that most participants successfully established active marketplace accounts and began implementing online marketing strategies, which have resulted in an expanded market reach and increased business operational efficiency. However, limited internet infrastructure and the availability of digital devices remain obstacles to optimizing technology utilization. Overall, this activity has had a positive impact on changing the business behavior of MSMEs and strengthening their business independence and resilience in the face of the digital economy's development.

This study examines the influence of social capital and human capital on theentrepreneurial intentions of former Indonesian migrant workers (PMI purna), withpsychological capital positioned as a mediating variable. Grounded in the Theory ofPlanned Behavior (TPB), the research analyzes how social networks, skills, workexperience, and psychological readiness shape entrepreneurial intentions amongreturnees. A descriptive quantitative approach was employed through thedistribution of Likert-scale questionnaires to 100 purposively selected respondentsfrom a popolation of 476 former Indonesian migrant workers in Cihonje Village,Banyumas Regency. The four variables, namely social capital, human capital,psychological capital, and entrepreneurial intention, were measured using validatesindicators and analiyzed with PLS-SEM.The finding indicate that social capital and human capital positively andsignificantly influence entrepreneurial intentions. Psychological capital alsodemonstrates a strong direct effect and serves as a partial mediator thatstrengthens the relationship between both forms of capital and entrepreneurialintention. The R-Square value of 0,803 reflects strong predictive power of themodel, while the value of 0,604 for psychological capital confirms its essentialmediating role. These results highlight the importance of social support,competence development, and psychological readiness in fostering entrepreneurialbehavior. The study recommends empowerment programs that integrate social,skill-bassed and psychological components to enchance the economicindependence of former Indonesian migrant workers.

This study focuses on the decline in durian production in Kemranjen subdistrict in 2021 and 2022. Bawor durian is a type of durian that is widely cultivated by farmers in Kemranjen subdistrict. The objectives of this study are: 1. To identify the factors influencing the production of Bawor durian farming 2. To determine the technical efficiency level of Bawor durian farming production 3. To identify the factors influencing the inefficiency of Bawor durian farming. The study was conducted in Alasmalang Village, Karangsalam Village, and Pageralang Village in Kemranjen Subdistrict, Banyumas Regency. Sampling was conducted using the census method. The sample size for this study was 62 farmers. The analysis method used stochastic frontier analysis. The results of the study indicate that the production factors significantly influencing durian bawor production are manure, NPK fertilizer, dolomite, pesticides, and the number of trees. The average technical efficiency value of Bawor durian farming is 0.71738845. The factors significantly influencing the inefficiency of Bawor durian farming are age and extension services. The results of this study are useful as a guideline for Bawor durian farmers in Kemranjen Subdistrict to achieve maximum results.
