Role of Resveratrol in Endotoxin Induced Uveitis: Literature Review

Uveitis is defined by a range of diverse clinical manifestations of ocular disease and involves inflammation of the uveal tissue, which includes the iris, choroid and ciliary body. Uveitis is characterized by a variety of heterogeneous clinical manifestations of ocular diseases. Corticosteroid and immunosupressive medication are required for non-infectious uveitis since they can successfully control inflammation. This study investigates role of Resveratrol as a promosing treatment in endotoxin induced uveitis as a mimic human anterior uveitis. Resveratrol is acknowledged as an antioxidant polyphenol discovered in grape skin and red wine. Anti-inflammatory, anti-cancer, and antioxidant properties are only a few of Resveratrol many biological and medicinal potentials. Our study evaluated the role of Resveratrol in potential therapy of uveitis. Results demonstrate significant reduce of inflammatory cytokine and cells suggesting the potential therapeutic effect of Resveratrol.

Maximizing the Hesperidin Extraction Using Supercritical Carbon Dioxide and Ethanol: Theoretical Prediction and Experimental Results

Agroindustrial waste can be valorized towards the obtaining of several products such as pigments, proteins, fibers, and polyphenolic compounds with antioxidant capacity. Orange peel waste is a promising source of polyphenolic compounds such as hesperidin. However, conventional extraction techniques present some environmental limitations such as high solvent consumption and high wastewater generation. Supercritical fluid extraction (SFE) has emerged as a green extraction technique due to the low use of solvent. The aim of this study was to maximize the hesperidin extraction based on theoretical predictions of the operating conditions from empirical thermodynamic models using SFE with carbon dioxide (CO2) as a solvent. The theoretical conditions were validated experimentally on a semi-pilot scale. The extracts were evaluated in terms of hesperidin content, total polyphenol content, and antioxidant capacity. Thermodynamic prediction of the operating conditions showed that the ethanol used as a co-solvent promotes hesperidin extraction. The optimum operating conditions were 25 °C, 80 bar, and a volumetric co-solvent concentration of 10%. The validation of the operating conditions resulted in a final hesperidin concentration of 11.5 ± 0.03 g/kg of orange peel waste. The experimental results were 30.26-times higher using 10% vol of ethanol than the extraction of hesperidin with pure ethanol as a co-solvent. The total polyphenol content and antioxidant capacity resulted in 831.92 ± 40.01 mg Galic acid/100 g orange peel waste, 15.41± 0.91 EC50/mL, and 5.31 ± 0.67 µMolTrox/100 g orange peel. Finally, the prediction of operating conditions from empirical thermodynamic models such as the Peng–Robinson equation of state with some modifications (Stryjek Vera) for solid–gas equilibrium solubility calculations, allows for maximizing the content of the polyphenolic compounds using SFE.

Mitigation of Oxidized Oil Effects on Production Performances and Meat Quality of Broilers by Dietary Supplementation of Allicin

The objective of this study was to evaluate the effects of dietary oxidized oil and allicin (two different dietary sources) as natural antioxidants on the growth performance and meat quality of broilers. A total of 200 one-day-old Ross 308 broilers were randomly divided into four dietary groups (50 birds/group). The experimental groups (OO—oxidized oil; OOA—oxidized oil and allicin; OOG—oxidized oil and garlic leaves) differed from the control one by the presence of oxidized oil in their dietary structure (peroxide value 9.07 (OO, OOA and OOG groups) vs. 1.70 (C group) meq active oxygen/kg). The diets given to the experimental groups differed from each other by the presence of allicin (100 mg/kg inclusion rate as extract (OOA) and 0.5% as garlic leaf powder (OOG)). At the end of the experiment, six animals/group were slaughtered, meat samples (breast and thigh) were collected, and nutritional value was established. The results showed that the allicin included in the experimental diet did not influence the proximate composition of breast meat (crude protein, fat, ash, and dry matter). The fatty acid profile was determined for each group of samples; a significant decrease in omega 3 FAs was noticed between the C group and the E groups (3.27% vs. 1.46%, 1.60%, and 1.56%) in breast meat samples, and a corresponding increase was noticed in saturated fatty acid (SFA) concentrations. Health indices with implications for atheroma and thrombus formation and cholesterol level were negatively affected by the presence of oxidized oil in the experimental diets, but the allicin extract supplement appeared to mitigate its influence. A positive influence of the dietary supplement was noticed on antioxidant capacity and polyphenol concentrations determined in breast and thigh samples under allicin supplement influence. The results of the current study revealed that the use of low oxidized oil in broilers diets did not affect productive performance. The nutritional quality of meat (breast and thigh) was negatively influenced by the presence of oxidized oil, but allicin supplements (extract or garlic leaves) improved lipid quality indices and antioxidant potential.

Characterization of dark chocolates based on polyphenolic profiles and antioxidant activity.

Dark chocolates were characterized according to geographical origin, cocoa variety, and cocoa content using the methylxanthine and polyphenolic composition and antioxidant activity as the data. The main study objective was to uncover sample patterns and identify possible markers of quality, variety, or origin to deal with authentication or fraud detection issues. In the study, a set of 26 dark chocolates from different varieties (e.g., Criollo, Forastero, and Trinitario) harvested in Africa, America, and Asia was analyzed. The optimized sample treatment consisted of defatting the chocolate (1g of sample with 5mL of cyclohexane for 15min, three times) and then extracting the analytes by sonication with methanol/water 60:40 (v:v) for 15min. The filtered extracts were analyzed by reversed-phase high-performance liquid chromatography with UV and spectrophotometric methods (Folin-Ciocalteu, ferric reducing antioxidant power, and aluminum methods) to determine individual phenolics and overall indexes of antioxidant and flavonoid content. Results from this chocolate set indicated that American samples are richer than African counterparts in alkaloids and phenolics (e.g., 1.7 vs. 1.1mgg-1 caffeine and 14.5 vs. 12.5mgg-1 total flavanols, respectively). Regarding cocoa varieties, Criollo cocoa was richer in bioactive compounds and antioxidant capacity (e.g., 16, 15, and 12mgg-1 total flavanols for Criollo, Forastero, and Trinitario, respectively). These results indicate that the analytes resulted in potential descriptors of varietal or geographical attributes.

Optimization of Microwave-Assisted Polyphenol Extraction and Antioxidant Activity from Papaya Peel Using Response Surface Methodology and Artificial Neural Network

The objective of the present study is to determine the optimal conditions of microwave-assisted extraction (MAE) such as microwave power (MWP), irradiation time (I-time), ethanol concentration (EtOH%), and solvent-to-solid ratio (S/S) for maximizing total polyphenol content (TPC) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity from papaya peel. Box-Behnken design (BBD) was used considering MWP 300–600 W, I-time 60-120 s, EtOH% 30-70%, and S/S 30-70 mL/g. Additionally, response surface methodology (RSM) and artificial neural networks (ANN) were employed for modelling, with cross-validation to enhance reliability. These models were combined with the desirability function (DF) and/or genetic algorithm (GA) optimization approaches. Maximizing TPC and DPPH activity while maintaining MWP, I-time, EtOH%, and S/S within their respective ranges using hybrid optimization approaches (RSM-DF: TPC = 1058 mgGAE/100g, and DPPH = 83%, RSM-GA: TPC = 1064 mgGAE/100g and DPPH = 79%, and ANN-GA: TPC = 1086 mgGAE/100g, and DPPH = 83%,) yielded consistent optimal results. An additional optimization approach (RSM-DF: TPC = 955 mgGAE/100g, and DPPH = 90%), aimed at minimizing MWP, I-time, EtOH%, and S/S while maximizing TPC and DPPH activity, showed a significant reduction of approximately 3 L in ethanol consumption for each 100 g of dry samples. The maximized TPC and DPPH activity in all four approaches showed a significant (p < 0.05) alignment with their corresponding validation tests, demonstrating the better performance of these modelling and optimization processes. In terms of assessing the effectiveness of microwave assistance, conventional solvent extraction was also performed, maintaining a 70 mL/g S/S ratio of 55% ethanol at 60°C for 2 h. This method resulted in lower values of TPC (857 ± 23 g GAE/100g) yield and DPPH activity (72 ± 5%) compared to the optimized condition of MAE.

Exploring the Correlation Between the Molecular Structure and Biological Activities of Metal-Phenolic Compound Complexes: Research and Description of the Role of Metal Ions in Improving the Antioxidant Activities of Phenolic Compounds.

We discussed and summarized the latest data from the global literature on the action of polyphenolic antioxidants and their metal complexes. The review also includes a summary of the outcomes of theoretical computations and our many years of experimental experience. We employed various methods, including spectroscopy (FT-IR, FT-Raman, NMR, UV/Vis), X-ray diffraction, thermal analysis, quantum calculations, and biological assays (DPPH, ABTS, FRAP, cytotoxicity, and genotoxicity tests). According to our research, the number and position of hydroxyl groups in aromatic rings, as well as the delocalization of electron charge and conjugated double bonds, have a major impact on the antioxidant effectiveness of the studied compounds. Another important factor is metal complexation, whereby high ionic potential metals (e.g., Fe(III), Cr(III), Cu(II)) enhance antioxidant properties by stabilizing electron charge, while the low ionic potential metals (e.g., Ag(I), Hg(II), Pb(II)) reduce efficacy by disrupting electron distribution. However, we observed no simple correlation between ionic potential and antioxidant capacity. This paper gives insights that will aid in identifying new, effective antioxidants, which are vital for nutrition and the prevention of neurodegenerative illnesses. Our results outline the connections between biological activity and molecular structure, offering a foundation for the methodical design of antioxidants. Our review also shows in detail how we use various complementary methods to assess the impact of metals on the electronic systems of ligands. This approach moves beyond the traditional "trial and error" method, allowing for the more efficient and rational development of future antioxidants.

Insights into the Therapeutic and Pharmacological Properties of Resveratrol as a Nutraceutical Antioxidant Polyphenol in Health Promotion and Disease Prevention.

Resveratrol (3, 5, 4'-trihydroxystilbene) is a polyphenolic derivative with herbal origin. It has attracted considerable attention in recent decades. Many studies have revealed the benefits of Resveratrol over several human disease models, including heart and neurological diseases, nephroprotective, immune regulation, antidiabetic, anti-obesity, age-related diseases, antiviral, and anticancer in experimental and clinical conditions. Recently, the antioxidant and anti-inflammatory activities of Resveratrol have been observed, and it has been shown that Resveratrol reduces inflammatory biomarkers, such as tissue degradation factor, cyclooxygenase 2, nitric oxide synthase, and interleukins. All of these activities appear to be dependent on its structural properties, such as the number and position of the hydroxyl group, which regulates oxidative stress, cell death, and inflammation. Resveratrol is well tolerated and safe even at higher pharmacological doses and desirably affects cardiovascular, neurological, and diabetic diseases. Consequently, it is plausible that Resveratrol can be regarded as a beneficial nutritional additive and a complementary drug, particularly for therapeutic applications. The present review provides an overview of currently available investigations on preventive and therapeutic characteristics and the main molecular mechanisms of Resveratrol and its potent derivatives in various diseases. Thus, this review would enhance knowledge and information about Resveratrol and encourage researchers worldwide to consider it as a pharmaceutical drug to struggle with future health crises against different human disorders.

Green tea (Camellia sinensis L.) potential to liver of hyperuricemia rats (Rattus norvegicus) fed with high purine diet

Uric acid is produced in tissues containing xanthine oxidases, like the small intestine and liver, as involved in the purine metabolism of adenine and guanine. High uric acid levels cause damage to cell membranes due to the lipid peroxidase chain reaction. Foods containing high purines will activate the xanthine oxidase (XOD), causing an increase in free radicals. Free radicals attack polyunsaturated fatty acids (PUFA) by forming lipid peroxides. PUFA molecules contain multiple double bonds, which make them susceptible to oxidation by reactive oxygen species (ROS), such as free radicals. PUFA will be broken into simple compounds such as pentane, ethane, and aldehydes, leading to malondialdehyde (MDA) formation. High MDA levels illustrate the process of cell membrane oxidation leading to cell membrane damage. Hyperuricemia is often treated using synthetic drugs such as allopurinol; however, it can cause side effects. Some people will choose medicinal plants with minimal side effects. Green tea (Camellia sinensis L.) contains many polyphenol antioxidants, especially flavonoids, which also have a strong antioxidant effect by exerting multiple mechanisms such as inhibiting several enzymes, including antiphospholipid peroxidase, free radical scavengers, metal bonds, and xanthine oxidase (XOD). Statistical analysis results proved that green tea with a dose of 600 mg/kg BW could decrease the level of MDA in the liver by 84.87% (p 0.01) and reduce the activity of XOD by 35.36% (p 0.01). Taken together, green tea can improve liver histopathology.

The Influence of Altitude on the Polyphenols Content and Antioxidant Capacity of Northern Moroccan 'Dellahia' Prickly Pear Juice

Moroccan cactus exhibits high genetic variability with several cultivars. The 'Dellahia' prickly pear variety, prevalent in northern Morocco and noted for its green pulp, is among the least valued cactus varieties, primarily consumed fresh. This study aimed to assess the impact of altitude on total phenolic acids and flavonoid content (TPC and TFC) and the antioxidant activity of 'Dellahia' prickly pear juice from northern Morocco. Significant differences in TPC ranged from 91.29 to 130.45 mg GAE/Kg of juice from the Mestassa and Wahran sites (at 119 m and 482 m altitude, respectively). TFC also varied slightly, from 18.8 to 19.1 mg RE/Kg of juice. Variations in antioxidant activity were evident in both DPPH• and ABTS+ assays, with DPPH• inhibition percentages ranging from 8.85% to 19.14% and ABTS+ inhibition from 41.07% to 54.35%. However, the influence of altitude on these parameters was inconclusive, as samples from higher altitudes did not consistently yield lower or higher values. Other factors such as soil composition, sunlight, and farming practices may influence these results.

Seasonal Variation of Polyphenols and Pigments in Ginkgo (Ginkgo biloba L.) Leaves: Focus on 3',8″-Biflavones.

Ginkgo (Ginkgo biloba L.) is a widely recognized medicinal plant, often grown as an ornamental species in parks around the world. Its leaves change color from green in spring to yellow in autumn. In this study, we collected ginkgo leaves at seven developmental stages from May to November and measured chlorophylls, carotenoids, flavonoids, and antioxidant activity. The total polyphenol content showed a significant increase from May to November, rising from 15.15 ± 0.14 mg GAE g-1 dw to 45.18 ± 0.42 mg GAE g-1 dw. The total flavonoid content reached its peak in August at 5.87 ± 0.18 mg GAE g-1 dw. In contrast, the highest concentrations of total polyphenolic acids (4.13 ± 0.16 mg CAE g-1 dw) and antioxidant activity (306.95 ± 3.47 µmol TE g-1) were recorded in May. We specifically focused on a less-studied group of dimeric flavonoids or biflavonoids-3',8″-biflavones. We identified five 3',8″-biflavones (amentoflavone, bilobetin, ginkgetin, isoginkgetin, and sciadopitysin) throughout all developmental stages. Sciadopitysin was the most abundant biflavonoid, with its concentration rising from 614.71 ± 5.49 µg g-1 dw in May to 2642.82 ± 47.47 µg g-1 dw in November. Alongside sciadopitysin, the content of other biflavonoids (excluding amentoflavone) generally increased over the same period. This trend is further highlighted by the total biflavonoid content, which grew from 1448.97 ± 6.63 µg g-1 dw in May to 6071.67 ± 97.15 µg g-1 dw in November. We observed a negative correlation between biflavonoid and chlorophyll content, which may indicate their involvement in leaf senescence. However, this hypothesis warrants further investigation.

Ethyl gallate ameliorates diabetes-induced Alzheimer's disease-like phenotype in rats via activation of α7 nicotinic receptors and mitigation of oxidative stress

Cognitive decline, an important comorbidity of type 2 diabetes (T2D), is attributed to oxidative stress and impaired cholinergic signaling in the brain. The α7 nicotinic acetylcholine receptor (α7nAChR) is densely distributed in the hippocampus and cortex, and exerts neuroprotective and procognitive actions. Ethyl gallate (EG), a natural phenolic antioxidant compound, showed high in-silico binding affinity towards α7nAChR and brain penetrability. Therefore, the present study aimed to evaluate the involvement of α7nAChR in the potential of EG to ameliorate T2D-induced Alzheimer's disease-like condition. T2D was induced by intraperitoneal (i.p.) injection of streptozotocin (35 mg/kg) in rats on high-fat diet. Diabetic animals were treated with EG (10 and 20 mg/kg, i.p.) for four weeks, and their learning and memory performance was evaluated by the Morris water maze (MWM). Further, the brains were subjected to biochemical analysis of antioxidants like glutathione (GSH), superoxide dismutase (SOD), and catalase (CAT), and oxidative stress marker malonaldehyde (MDA). While diabetic rats showed a significant decline in cognitive performance in the MWM, a substantial improvement was noticed following EG treatment. Further, the diabetes-associated reductions in GSH, SOD, and CAT levels, along with increased MDA contents in the brain, were effectively restored by EG. Interestingly, pre-treatment with α7nAChR antagonist methyllycaconitine (1 mg/kg, i.p.) attenuated the effects of EG on behavioral and biochemical parameters. The results suggest that EG may augment cholinergic signaling in the brain via α7nAChR to mitigate oxidative stress, consequently alleviating T2D-associated dementia. Therefore, EG could be a potential candidate for addressing cognitive impairment comorbid with T2D.

Pressurized Liquid Extraction of Antioxidant and α-Amylase-Inhibitory Compounds from Red Seaweed Using Water-Ethanol Mixtures.

Red seaweeds from the coastal shores of Ilo (Peru) are a natural source of high-value compounds beneficial to health due to their high antioxidant capacity. Thus, this work evaluated the effect of water-ethanol mixtures (0, 15, and 30%; v/v) at high temperatures (90, 120, and 150 °C) on the polyphenol content, antioxidant capacity, and polyphenols profile of red seaweed (Chondracanthus chamissoi) during a pressurized liquid extraction process, whose parameters were set at 10 atm, with a single cycle of extraction and a volume of 150%. An increase in temperature and ethanol had a positive effect on antioxidant compounds. Thus, the best processing conditions were established at 150 °C and 30% ethanol, allowing for the extraction of a high polyphenol content (2.04 mg GAE/g dw) and antioxidant capacity (IC50: 7.46 mg/mL, ORAC: 148.98 μmol TE/g dw). High ethanol concentrations (30%) effectively recovered phenolic acids, flavonols, and phlorotannins for the polyphenols profile. However, the use of pure water was more effective in recovering flavonols. Interestingly, using pure water as an extraction solvent at high temperatures allowed for a more significant inhibition of the α-amylase enzyme than water-ethanol mixtures under the same conditions. Finally, the results can be utilized for future industrial scaling and the potential utilization of extracts in developing diabetes treatments.

One or multiple chlorophyll molecules are employed in the reaction center of photosystem II's main electron donor (defined as P680). We have a poor understanding of how the reaction center facilitates water oxidation and the roles that mono‐ and/or multimeric chlorophyll groups play when P680 oxidizes a neighboring tyrosine in order to drive water oxidation at the oxygen evolving complex. We have prepared a dimeric MgII‐porphyrin complex [Mg2(BTPP)] (1, H4‐BTPP = 1,2‐bis‐(10,15,20‐triphenylporphyrin‐5‐yl)‐benzene) as a structural and functional mimic of the dimeric core of P680. 1 was oxidized by one‐electron to the corresponding π‐cation radical complex 2. The radical cation was characterized by UV‐Vis‐NIR, FT‐IR, and EPR spectroscopic techniques. 2 was shown to be reactive towards phenols to give the corresponding phenoxyl radicals, mimicking the reactivity of the P680 cation radical which oxidizes tyrosine to tyrosyl radical. Critically, the dimeric π‐cation radical showed markedly higher rates of proton coupled electron transfer oxidation (PCET) of phenols when compared to its monomeric counterpart [Mg(TPP)] (TPP = 5,10,15,20‐tetraphenylporphyrin). Our findings demonstrate that MgII‐porphyrin complexes are reliable mimics of photosynthetic PCET processes and suggest that photosynthetic reaction centers with multiple π‐conjugated complexes likely lower the barrier to PCET oxidation by π‐cation radical species.

Sustainable Valorisation of Coffee Waste as a Protein Source, Mycelium-Based Packaging Material and Renewable Energy Pellet.

This study investigates the valorization of spent coffee grounds (SCGs) through protein extraction and their application in mycelium-based packaging and renewable energy pellets. Three extraction methods-mechanical stirring, ultrasound-assisted, and CO2-assisted extraction-were applied to SCGs. CO2-assisted extraction yielded the highest protein content at 34.24%, followed by mechanical stirring (31.46%) and ultrasound-assisted extraction (28.51%). The total polyphenol content and antioxidant capacity were also highest in the CO2 extracts, suggesting that this method preserves bioactive compounds most effectively. After protein extraction, SCGs were tested as a component in mycelium-based packaging, with results showing an apparent density of 0.551 g/cm3 and compression resistance of 3.354 MPa, indicating its suitability for structural applications. The energy value of SCGs remained high, with a calorific value of 19,887 J/g DW, slightly decreasing after extraction but still sufficient for renewable energy production. These findings highlight the potential of SCGs as a multi-functional resource, contributing to sustainable solutions across various industries.

Optimization of Germination Conditions for Enriched γ-Aminobutyric Acid and Phenolic Compounds of Foxtail Millet Sprouts by Response Surface Methodology.

The optimum germination conditions for foxtail millet sprouts enriched with γ-aminobutyric acid (GABA) and antioxidant polyphenols were investigated. From single-factor experimental results, both the GABA level and total phenolic content (TPC) were more significantly affected by soaking temperature and time, and concentration of sucrose culture solution. Response surface methodology (RSE) was used to optimize the germination conditions of foxtail millet sprouts, where the interaction between soaking temperature and sucrose concentration exhibited a significant (p < 0.05) effect on TPC, and the interaction between soaking time and sucrose concentration displayed a significant (p < 0.05) effect on GABA content. The optimal germination conditions for TPC and GABA enrichment of foxtail millet sprouts were soaking at 31 °C for 4.5 h and germinating at 35 °C with 4.5 g/L sucrose solution for 5 days. Under the optimized conditions, the TPC and GABA content of foxtail millet sprouts were 926.53 milligrams of ferulic acid equivalents per 100 g dry weight (mg FAE/100 g DW) and 259.13 mg/kg, separately, with less difference from the predicted values of 929.44 mg FAE/100 g DW and 263.60 mg/kg, respectively. Collectively, all the individual phenolic compounds increased significantly (p < 0.05) by optimization, except for cis-p-coumaric acid and cis-ferulic acid in bound. The results provide a practical technology for suitable germination conditions to improve the health components of foxtail millet sprouts and increase their added value.

Combining Ability of Capsicum annuum Hybrid for Antioxidant Activities, Polyphenol Content, α-Glucosidase Inhibitory, Yield, and Yield Components.

Chili (Capsicum annuum) consumption is often suggested, and using functional food cultivars is the most effective strategy post COVID-19 pandemic. Controlling chili breeding activity is one of the most effective methods to produce new hybrid varieties. However, the general combining ability (GCA), specific combining ability (SCA), and heterotic effect of functional biochemicals (polyphenol content, antioxidant activities, and α-glucosidase inhibitory compounds) remain poorly known in C. annuum. This study aimed to estimate these parameters in C. annuum by using five different genotypes and their hybrid combinations based on growth characteristics, yield, yield components, and fruit functional biochemicals. The F1 and F1R progenies were obtained from crosses in a greenhouse with a full diallel mating design. Each parent used in this study had a GCA advantage for each characteristic. The hybrid combination of IPB074 × IPB005 and IPB435 × IPB367 displayed the best yield results. However, the results indicated the opposite regarding α-glucosidase inhibitory compounds. The heterotic effect of functional biochemicals was observed for traits related to genotypes, polyphenol content, antioxidant activity, α-glucosidase inhibitory compounds, and similar properties related to yield and yield components, indicating their use in hybrid chili production.

The Optimisation of Ultrasound-Assisted Extraction for the Polyphenols Content and Antioxidant Activity on Sanguisorba officinalis L. Aerial Parts Using Response Surface Methodology

The aim of this study was to optimise ultrasound-assisted extraction (UAE) of the herb Sanguisorba officinalis L. in terms of the antioxidant activity (DPPH and FRAP method) and total polyphenol content (TPC). Optimisation was performed using the response surface methodology (RSM) with a third-degree (33) Central Composite Design (CCD) approach. The RSM was applied to obtain the optimal combination of (1) raw material content (2.25–7.5 g raw material/100 mL of solvent), (2) ethanol concentration (20–60% v/v), and (3) extraction time (1–15 min). The optimal conditions for the extraction of polyphenols and antioxidant potential were a raw material content of 7.5 g/100 mL of solvent (solid/solvent ratio 13.3 mL/g), an ethanol concentration of 47% v/v, and an extraction time of 10 min. At these optimal extraction parameters, the maximum extraction of polyphenols and antioxidant activity obtained experimentally was found to be very close to its predicted value and was 12.9 mmol Trolox/L (DPPH method), 19.4 mmol FeSO4/L (FRAP method), and 2.1 g GA/L (TPC). The mathematical model developed was found to fit with the experimental data on the antioxidant potential and polyphenol extraction. The n-octanol/water partition coefficient of the optimised extract was used to determine their lipophilicity. Our studies have shown that the optimised extract is highly hydrophilic (log P &lt; 0). Optimal parameters can be used for the industrial extraction of the S. officinalis herb for the needs of, among others, the pharmaceutical or cosmetic industry.

Characterization and monitoring of changes during lactation in the profile of multiple bioactive compounds of milk from grazing mares.

Mare milk has often been considered a food product with potential functional properties. However, the bioactive compound composition of mare milk, including vitamins and other minor bioactive compounds, as well as factors affecting this composition have scarcely been studied. Therefore, the present study aimed to characterize the changes during lactation in the content of water- and fat-soluble vitamins and total polyphenols, and the total antioxidant capacity of mare milk from semi-extensive farms. A total of 310 individual milk samples from 18 mares belonging to three commercial farms and 12 lactation times were analyzed. Ascorbic acid (vitamin C), thiamine (vitamin B1), riboflavin (vitamin B2), nicotinic acid and niacinamide (vitamins B3), pantothenic acid (vitamin B5), pyridoxal and pyridoxine (vitamins B6), folic acid (vitamin B9), cyanocobalamin (vitamin B12), tocopherols and tocotrienols (vitamin E) and retinol and retinyl esters (vitamin A) were quantified using liquid chromatography. Total polyphenols and antioxidant capacity assays were analyzed using spectrophotometry. The concentration of most bioactive compounds tended to decline as lactation progressed, with the exception of polyphenols and the total antioxidant capacity that oscillated during lactation. On the other hand, the effect of the different semi-extensive management of the farms was only significant for vitamin B3 content. To the best of our knowledge, the present study provides the most in-depth description of the vitamin profile of mare milk as well as new insights into polyphenol content and antioxidant capacity of mare milk. © 2024 The Author(s). Deciphering the mechanisms underlying the neuroprotective potential of kaempferol: a comprehensive investigation.

Neurodegenerative disorders are characterized by neuronal degradation, dysfunction, or death within the CNS. Oxidative and inflammatory stress play crucial roles in the pathogenesis of various neurodegenerative diseases. The interplay between these stressors and dysregulated cellular signaling pathways contributes to neurodegeneration. Downregulation of NRF-2 compromises antioxidant defense, exacerbating neuronal damage, while increased TLR-4/MAPK and TLR-4/NF-κB signaling promotes neuroinflammation. Excessive ROS production by NADPH oxidase leads to oxidative damage and neuronal apoptosis. The strategies targeting NRF-2, TLR-4-mediated inflammatory stress, and NADPH oxidase activity promise to mitigate neuronal damage and halt the progression of the disease. Kaempferol is a flavonoid polyphenol antioxidant found abundantly in various fruits and vegetables, including apples, grapes, tomatoes, and broccoli. It is widely found in medicinal plants including Equisetum spp., Sophora japonica, Ginkgo biloba, and Euphorbia pekinensis (Rupr.). A substantial body of in vitro and in vivo evidences have demonstrated the neuroprotective potential of kaempferol against neurodegenerative diseases such as Alzheimer's disease and Parkinson's disease. Kaempferol demonstrates multifaceted potential in mitigating neuroinflammation, apoptosis, and oxidative stress in different neurodegenerative diseases through the modulation of various pathways including NRF-2, NADPH oxidase, TLR-4/MAPK, and TLR-4/NF-κB. This review article was developed through a comprehensive analysis and interpretation of research published between 2009 and 2024, sourced from multiple scientific databases, including PubMed, Scopus, ScienceDirect, and Web of Science. This review aims to provide an in-depth overview of the neuroprotective effects of kaempferol, focusing on its underlying molecular mechanisms. A total of 24 research evidence were included to elucidate the molecular pathways by which kaempferol exerts its protective effects against neurodegenerative diseases.
