Beyond Domain-Specific Expertise: Neural Signatures of Face and Spatial Working Memory in Baduk (Go Game) Experts.

Recent advances of neuroimaging methodology and artificial intelligence have resulted in renewed interest in board games like chess and Baduk (called Go game in the West) and have provided clues as to the mechanisms behind the games. However, an interesting question that remains to be answered is whether the board game expertise as one of cognitive skills goes beyond just being good at the trained game and how it maps on networks associated with cognitive abilities that are not directly trained. To address this issue, we examined functional activity and connectivity in Baduk experts, compared to novices, while performing a visual n-back working memory (WM) task. We found that experts, compared to novices, had greater activation in superior parietal cortex during face WM, though there were no group differences in behavioral performances. Using a data-driven, whole-brain multivariate approach, we also found significant group differences in the multivariate pattern of connectivity in frontal pole and inferior parietal cortex, further showing greater connectivity between frontal and parietal regions and between frontal and temporal regions in experts. Our findings suggest that long-term trained Baduk experts have the reorganization of functional interactions between brain regions even for untrained cognitive ability.

Biomarkers for uranium risk assessment for the development of a molecular epidemiological protocol

We used a functional MRI paradigm involving conventional vs. unconventional views of objects to assess bottom-up vs. top-down visual processing in Parkinson's disease (PD) with normal cognition, PD with mild cognitive impairment (MCI), and MCI due to Alzheimer's disease (AD) as compared to healthy controls. We particularly aimed at determining whether the task discriminated between PD with and without MCI and between two MCI groups due to distinct pathologies (AD and PD).116 right-handed subjects (21 MCI due to AD; 16 PD with normal cognition; 24 PD with MCI; 55 healthy controls) performed a visual object-matching task in a T MR scanner. T statistic maps were computed to contrast task-based activation during unconventional vs. conventional view conditions. One-way ANOVAs and post hoc tests were performed to assess differences across and between groups.Both MCI groups performed worse than controls in the unconventional views condition and showed reduced activation of right anterior cingulate cortex and right superior parietal lobule (PD with MCI), and right middle and inferior frontal gyri (MCI due to AD). Neural responses in cortical areas within the ventral and dorsal visual pathway appeared to be preserved in both MCI groups. Receiver operating characteristic analysis of MRI contrast in the right superior parietal lobule distinguished PD with and without MCI with 87.50% sensitivity and 86.98% specificity.Impaired recognition of objects presented in unconventional orientations in MCI due to PD and AD was associated with decreased activation of frontoparietal regions, consistent with defective top-down regulation of visual processing. Aberrant activation of superior parietal cortex may serve as an early imaging biomarker of impending cognitive impairment in PD.

Activity in superior parietal cortex during training by observation predicts asymmetric learning levels across hands.

A dominant concept in motor cognition associates action observation with motor control. Previous studies have shown that passive action observation can result in significant performance gains in humans. Nevertheless, it is unclear whether the neural mechanism subserving such learning codes abstract aspects of the action (e.g. goal) or low level aspects such as effector identity. Eighteen healthy subjects learned to perform sequences of finger movements by passively observing right or left hand performing the same sequences in egocentric view. Using functional magnetic resonance imaging we show that during passive observation, activity in the superior parietal lobule (SPL) contralateral to the identity of the observed hand (right\\left), predicts subsequent performance gains in individual subjects. Behaviorally, left hand observation resulted in positively correlated performance gains of the two hands. Conversely right hand observation yielded negative correlation - individuals with high performance gains in one hand exhibited low gains in the other. Such behavioral asymmetry is reflected by activity in contralateral SPL during short-term training in the absence of overt physical practice and demonstrates the role of observed hand identity in learning. These results shed new light on the coding level in SPL and have implications for optimizing motor skill learning.

Association between striatal dopamine D2/D3 receptors and brain activation during visual attention: effects of sleep deprivation.

Sleep deprivation (SD) disrupts dopamine (DA) signaling and impairs attention. However, the interpretation of these concomitant effects requires a better understanding of dopamine’s role in attention processing. Here we test the hypotheses that D2/D3 receptors (D2/D3R) in dorsal and ventral striatum would distinctly regulate the activation of attention regions and that, by decreasing D2/D3, SD would disrupt these associations. We measured striatal D2/D3R using positron emission tomography with [11C]raclopride and brain activation to a visual attention (VA) task using 4-Tesla functional magnetic resonance imaging. Fourteen healthy men were studied during rested wakefulness and also during SD. Increased D2/D3R in striatum (caudate, putamen and ventral striatum) were linearly associated with higher thalamic activation. Subjects with higher D2/D3R in caudate relative to ventral striatum had higher activation in superior parietal cortex and ventral precuneus, and those with higher D2/D3R in putamen relative to ventral striatum had higher activation in anterior cingulate. SD impaired the association between striatal D2/D3R and VA-induced thalamic activation, which is essential for alertness. Findings suggest a robust DAergic modulation of cortical activation during the VA task, such that D2/D3R in dorsal striatum counterbalanced the stimulatory influence of D2/D3R in ventral striatum, which was not significantly disrupted by SD. In contrast, SD disrupted thalamic activation, which did not show counterbalanced DAergic modulation but a positive association with D2/D3R in both dorsal and ventral striatum. The counterbalanced dorsal versus ventral striatal DAergic modulation of VA activation mirrors similar findings during sensorimotor processing (Tomasi et al., 2015) suggesting a bidirectional influence in signaling between the dorsal caudate and putamen and the ventral striatum.

Longitudinal development of frontoparietal activity during feedback learning: Contributions of age, performance, working memory and cortical thickness

Feedback learning is a crucial skill for cognitive flexibility that continues to develop into adolescence, and is linked to neural activity within a frontoparietal network. Although it is well conceptualized that activity in the frontoparietal network changes during development, there is surprisingly little consensus about the direction of change. Using a longitudinal design (N=208, 8–27 years, two measurements in two years), we investigated developmental trajectories in frontoparietal activity during feedback learning. Our first aim was to test for linear and nonlinear developmental trajectories in dorsolateral prefrontal cortex (DLPFC), superior parietal cortex (SPC), supplementary motor area (SMA) and anterior cingulate cortex (ACC). Second, we tested which factors (task performance, working memory, cortical thickness) explained additional variance in time-related changes in activity besides age. Developmental patterns for activity in DLPFC and SPC were best characterized by a quadratic age function leveling off/peaking in late adolescence. There was a linear increase in SMA and a linear decrease with age in ACC activity. In addition to age, task performance explained variance in DLPFC and SPC activity, whereas cortical thickness explained variance in SMA activity. Together, these findings provide a novel perspective of linear and nonlinear developmental changes in the frontoparietal network during feedback learning.

Neural Activity in Superior Parietal Cortex during Rule-based Visual-motor Transformations

Cognition allows for the use of different rule-based sensorimotor strategies, but the neural underpinnings of such strategies are poorly understood. The purpose of this study was to compare neural activity in the superior parietal lobule during a standard (direct interaction) reaching task, with two nonstandard (gaze and reach spatially incongruent) reaching tasks requiring the integration of rule-based information. Specifically, these nonstandard tasks involved dissociating the planes of reach and vision or rotating visual feedback by 180°. Single unit activity, gaze, and reach trajectories were recorded from two female Macaca mulattas. In all three conditions, we observed a temporal discharge pattern at the population level reflecting early reach planning and on-line reach monitoring. In the plane-dissociated task, we found a significant overall attenuation in the discharge rate of cells from deep recording sites, relative to standard reaching. We also found that cells modulated by reach direction tended to be significantly tuned either during the standard or the plane-dissociated task but rarely during both. In the standard versus feedback reversal comparison, we observed some cells that shifted their preferred direction by 180° between conditions, reflecting maintenance of directional tuning with respect to the reach goal. Our findings suggest that the superior parietal lobule plays an important role in processing information about the nonstandard nature of a task, which, through reciprocal connections with precentral motor areas, contributes to the accurate transformation of incongruent sensory inputs into an appropriate motor output. Such processing is crucial for the integration of rule-based information into a motor act.

Neural events leading to and associated with detection of sounds under high processing load

The neural events that lead to successful or failed detection of suprathreshold sounds are not well established. In this experiment, event-related potentials (ERPs) and functional magnetic resonance imaging (fMRI) were recorded while participants performed two tasks: a primary difficult duration judgment task on a sequence of tones presented to one ear, and a secondary target detection task on an auditory oddball stream presented to the other ear. The paradigm was designed to elicit competition and variability in detection of auditory targets despite identical input. Successful detection of auditory targets was associated mainly with greater fMRI activity in superior parietal cortex and thalamus. In the ERPs, successful detection was linked with a larger fronto-central negativity at 200-400 ms, and a later centro-posterior positivity. Failure to detect targets was associated with greater fMRI signal in the default mode network, a significantly smaller electrical fronto-central negativity and no late positivity. These findings demonstrate that variability in auditory detection is related to modulation of activity in multimodal parietal and frontal networks active ∼ 200 ms after target onset. Results are consistent with a limited capacity and late selection view of attention.

Spatio-temporal brain dynamics in a combined stimulus–stimulus and stimulus–response conflict task

It is yet not well known whether different types of conflicts share common or rely on distinct brain mechanisms of conflict processing. We used a combined Flanker (stimulus–stimulus; S–S) and Simon (stimulus–response; S–R) conflict paradigm both in an fMRI and an EEG study. S–S conflicts induced stronger behavioral interference effects compared to S–R conflicts and the latter decayed with increasing response latencies. Besides some similar medial frontal activity across all conflict trials, which was, however, not statically consistent across trials, we especially found distinct activations depending on the type of conflict. S–S conflicts activated the anterior cingulate cortex and modulated the N2 and early P3 component with underlying source activity in inferior frontal cortex. S–R conflicts produced distinct activations in the posterior cingulate cortex and modulated the late P3b component with underlying source activity in superior parietal cortex. Double conflict trials containing both S–S and S–R conflicts revealed, first, distinct anterior frontal activity representing a meta-processing unit and, second, a sequential modulation of the N2 and the P3b component. The N2 modulation during double conflict trials was accompanied by increased source activity in the medial frontal gyrus (MeFG). In summary, S–S and S–R conflict processing mostly rely on distinct mechanisms of conflict processing and these conflicts differentially modulate the temporal stages of stimulus processing.

Experience-dependent alterations in conscious resting state activity following perceptuomotor learning

In monkeys and rats, neural activity patterns during learning are reactivated during subsequent periods of rest or sleep. According to the reactivation–consolidation account, this process underlies the consolidation of memories. Brain imaging studies have extended these findings to humans during sleep, but not yet, during rest. Here, we show that learning-related reactivation also occurs in humans during rest. During functional MRI-scanning, participants trained on a perceptuomotor task flanked by rest periods. During training, we found robust activity in the superior parietal cortex. During post-training rest, this same area reactivated. We also found a link between parietal reactivation and learning. Activity in superior parietal cortex was associated with learning during training, and a control group that did not train on the perceptuomotor task did not show any difference between the pre- and post-training rest blocks in this region. These findings indicate that, during rest, reactivation also occurs in humans. This process may contribute to consolidation of perceptuomotor memories.

Frontal control of attentional capture in visual search

Lavie and colleagues recently suggested that cognitive control functions that are mediated by frontal cortex provide goal-directed control of selective attention, serving to minimize interference by goal-irrelevant distractors. Here we provide new evidence for this claim from an attentional capture paradigm. An event-related fMRI experiment shows that the presence (vs. absence) of an irrelevant colour singleton distractor in a visual search task was not only associated with activity in superior parietal cortex, in line with a psychological attentional capture account, but was also associated with frontal cortex activity. Moreover, behavioural interference by the singleton was negatively correlated with frontal activity, suggesting that frontal cortex is involved in control of singleton interference. Behavioural tests confirmed that singleton interference depends on availability of cognitive control to the search task: Singleton interference was significantly increased by high working memory load. These results demonstrate the important role of frontal cognitive control of attention by working memory in minimizing distraction.

PET studies of parietal involvement in spatial attention: Comparison of different task types.

Five experiments are described that concern the mechanisms that direct attention to spatial and non-spatial features of a stimulus and the effects that attention has on the visual system's analysis of that stimulus. Shifts of attention from one spatial location to another activated the superior parietal lobe and this activation was fairly independent of the task performed on the attended object, the response made to the attended object, and whether the shift of attention was controlled endogenously or exogenously. Maintaining attention tonically on a location or a particular visual feature such as shape, colour or motion did not produce a superior parietal response. Tonic attention to a feature (colour, shape, motion) or location, however, did produce enhancements in the response of various regions that are probably specialized for processing the attended visual feature. The activation of superior parietal cortex during shifts of spatial attention as well as the activation of parietal-occipital cortex when attention is tonically maintained on a location suggest that the parietal cortex plays an important role in spatial computations.

Dissociation of Object and Spatial Vision in Human Extrastriate Cortex: Age-Related Changes in Activation of Regional Cerebral Blood Flow Measured with [15 O]Water and Positron Emission Tomography

We previously reported selective activation of regional cerebral blood flow (rCBF) in occipitotemporal cortex during a face matching task (object vision) and activation in superior parietal cortex during a dot-location matching task (spatial vision) in young subjects, The purpose of the present study was to determine the effects of aging on these extrastriate visual processing systems. Eleven young (mean age 27 ± 4 years) and nine old (mean age 72 ± 7 years) male subjects were studied. Positron emission tomographic scans were performed using a Scanditronix PC1024-7B tomograph and H2(15)O to measure rCBF. To locate brain areas that were activated by the visual tasks, pixel-by-pixel difference images were computed between images from a control task and images from the face and dot-location matching tasks. Both young and old subjects showed rCBF activation during face matching primarily in occipitotemporal cortex, and activation of superior parietal cortex during dot-location matching. Statistical comparisons of these activations showed that the old subjects had more activation of occipitotemporal cortex during the spatial task and more activation of superior parietal cortex during the object task than did the young subjects. These results show less functional separation of the dorsal and ventral visual pathways in older subjects, and may reflect an age-related reduction in the processing efficiency of these visual cortical areas.
