Abstract

Thromboxane B 2 (TXB 2 ) and the thromboxane A 2 mimicking agent, (15S)-hydroxy-11α, 9α-(epoxy methano) prosta-5Z, 13E-dienoic acid increase the proliferation of B16 amelanotic melanoma cells in vitro . TXB 2 (25 μg ml) −I lowers basal cyclic AMP (cAMP) levels and blocks the rise in intracellular and released cAMP by B16a cells in response to PGE 1 (1 μg ml −1 ). Prostacyclin and thromboxane synthetase inibitors decrease tumor cell proliferation and inhibit DNA synthesis. We propose the hypothesis that tumor cell proliferation may be modulated positively and negatively by TXA 2 and PGI 2 . Furthermore, this regulation may center on the modulation of tumor cell cAMP levels.
