Rationales: Gene-targeting ribozymes represent promising nucleic acid-based gene interference agents for therapeutic application. We previously used an in vitro selection procedure to engineer novel RNase P-based ribozyme variants with enhanced targeting activity. However, it has not been reported whether these ribozyme variants also exhibit improved activity in blocking gene expression in animals.Methods and Results: In this report, R388-AS, a new engineered ribozyme variant, was designed to target the mRNA of assemblin (AS) of murine cytomegalovirus (MCMV), which is essential for viral progeny production. Variant R338-AS cleaved AS mRNA sequence in vitro at least 200 times more efficiently than ribozyme M1-AS, which originated from the wild type RNase P catalytic RNA sequence. In cultured MCMV-infected cells, R338-AS exhibited better antiviral activity than M1-AS and decreased viral AS expression by 98-99% and virus production by 15,000 fold. In MCMV-infected mice, R388-AS was more active in inhibiting AS expression, blocking viral replication, and improving animal survival than M1-AS.Conclusions: Our results provide the first direct evidence that novel engineered RNase P ribozyme variants with more active catalytic activity in vitro are also more effective in inhibiting viral gene expression in animals. Moreover, our studies imply the potential of engineering novel RNase P ribozyme variants with unique mutations to improve ribozyme activity for therapeutic application.

OF THE THESIS CONTROLS ON BENTHIC MICROBIAL COMMUNITY STRUCTURE AND ASSEMBLY IN A KARSTIC COASTAL WETLAND by Nicholas Ogden Schulte Florida International University, 2016 Miami, Florida Professor Evelyn Gaiser, Major Professor The assembly mechanisms underlying microbial community abundance, biotic interactions, and diversity over space and time are unresolved, particularly in benthic microbial mats distributed along environmental gradients. Experimental enrichment of nutrient-limited microbial mats from the Florida Everglades along a nutrient subsidysalinity stress gradient stimulated autotrophic and heterotrophic metabolism, growth, and diversity independent of autotroph-heterotroph interactions across treatments and space. These results suggest spatial segregation of autotrophic and heterotrophic components within mats. Considering only the diatom component of Everglades mats over space and time, the subsidy-stress gradient controlled diatom compositional turnover at broad spatial scales while environmental and dispersal-based processes structured diatom communities at the regional scale and environmental processes independent of the environmental gradient at the temporal scale. These results indicate environmental gradients may not necessarily increase connectivity and dispersal across space, and temporal microbial diversity is driven at the local and regional scales by environmental heterogeneity in benthic microbial communities.

In ancient and medieval south India, from about the fifth century, the term nādu denoted a micro-region which was important as the basic unit of agricultural production. The agricultural community formed in the nādu was called nāttār or nāttavar, literally meaning the people of the nādu. Initially it was exclusively composed of the Vellāla peasantry, but from the eleventh century there began to appear in Tamil inscriptions the term periyanādu meaning “big nādu” to denote a supra-nādu assembly. In this paper we examine the meaning of the emergence of this and other similar supra-local and/or multi-community organizations.The Chola dynasty, which had ruled south India for about four hundred years, disappeared in the latter half of the thirteenth century. The succeeding Pandyan dynasty was put down in its turn by the invasion of the Delhi Sultan's army at the beginning of the fourteenth century. Therefore, the thirteenth and fourteenth centuries in south India were a period of political turmoil, during when, nevertheless, foreign trade was carried out vigorously in the Indian Ocean. Merchants and artisans joined peasants in the activities of the periyanādu, generating a new state and social formation that became explicitly visible in the fifteenth century under Vijayanagar rule.
