The method described extracts high quality RNA from a clay-rich soil for which other published extraction protocols have failed. Transcript encoding the enzyme β-glucuronidase could be reliably detected by reverse transcriptase polymerase chain reaction in soil seeded with a genetically modified Rhizobium leguminosarum containing the uidA gene.
