Abstract

A method is described for preserving coupling activity in isolated chloroplasts stored in the frozen state for periods up to 16 weeks. Pea chloroplasts stored in a high osmolarity medium supplemented with 50% (v/v) glycerol and 10 mg/ml bovine serum albumin show high rates of photophosphorylation, good coupling efficiencies, photosynthetic control, and modulation of the high energy state by adenine nucleotides.
