Abstract

An automated method for the determination of hemoglobin (Hb) in rat blood plasma by the CentrifiChem Analyzer was developed to assess the effect of hemolysis on routine blood chemistry. The procedure was based on the hemebenzidine reaction in the presence of hydrogen peroxide. The reproducibility of the assay was evaluated by the simultaneous analysis of 10 aliquots from each of three standard solutions which contained 150,300, and . 560 mg Hb/liter. Coefficients of variation (CV) of 1.0,1.4, and 4.4% were obtained for each sample, respectively. Variation between runs was examined by comparing the standard solution containing 581 mg/liter of Hb in 10 different runs. A CV of 3.0% was obtained. The test requires 30 minutes of preparation time and actual analysis time for 23 samples is only 50 seconds. Hb can be determined in as little as 5pI of plasma.
