Abstract

The specific growth rate was used as a parameter for tracing growth limiting substances. The exponential growth phase is very short in a normal hybridoma batch culture. The specific growth rate reaches its maximum early during culture, when the cell density is low, and then declines immediately after that. This is not due to accumulation of inhibitory metabolites such as ammonium and lactate or depletion of macro nutrients but of some other factors. When serum was added intermittently during the growth phase to a batch culture the specific growth rate was kept constant and the cells grew exponentially. Insulin was identified as the main component for keeping the specific growth rate constant.
