Abstract

During the development, multicellular organisms form the structure with a complex shape in the morphogenesis, in which invagination plays an important role. However, the mechanism of the invagination is not well understood because of the difficulty in observation in vivo. In this study, to observe the invagination in vitro by using the three-dimensional culture method of mES cells, we examined the culture conditions of the cells for differentiation into an optic cup and the observation method of the optic cup form. We revealed that the improvement of culture condition enhanced the differentiation of mES cells. In addition, we observed the optic cup made from mES cells by the two-photon excitation microscopy of Rx-GFP, and further research is need to observe the deep part of the optic cup form.
